Abstract

This study examined the effect of pibutidine hydrochloride, a novel histamine H2 receptor antagonist (IT-066), on nitric oxide (NO) production in gastric mucosa by measuring nitrite and nitrate contents. IT-066 (0.3-3 mg/kg, p.o.) and NO donors, sodium nitroprusside (0.3 mg/kg, s.c.) and NOR3 (0.1 mg/kg, s.c.), increased the NO contents in gastric mucosa. Cimetidine (100 mg/kg, p.o.), ranitidine (30 mg/kg, p.o.) and famotidine (10 mg/kg, p.o.) did not significantly affect the NO production. Pretreatment with NG-nitro-L-arginine methyl ester (3 mg/kg, i.v.), a NO synthase inhibitor, decreased the level of IT-066 (3 mg/kg, p.o.)-stimulated NO production. These results suggest that IT-066 stimulated gastric mucosal NO production, and that endogenous NO may be implicated in the pharmacological effect of IT-066.

Abstract

C-type natriuretic peptide (CNP) is a potent stimulator of long bone and vertebral development via endochondral ossification. In the present study, we investigated the effects of CNP on craniofacial skeletogenesis, which consists of both endochondral and membranous ossification. Morphometric analyses of crania from CNP knockout and transgenic mice revealed that CNP stimulates longitudinal growth along the cranial length, but does not regulate cranial width. CNP markedly increased the length of spheno-occipital synchondrosis in fetal murine organ cultures, and the thickness of cultured murine chondrocytes from the spheno-occipital synchondrosis or nasal septum, resulting in the stimulation of longitudinal cranial growth. Mandibular growth includes endochondral and membranous ossification; although CNP stimulated endochondral bone growth of condylar cartilage in cultured fetal murine mandibles, differences in the lengths of the lower jaw between CNP knockout or transgenic mice and wild-type mice were smaller than those observed for the lengths of the upper jaw. These results indicate that CNP primarily stimulates endochondral ossification in the craniofacial region and is crucial for midfacial skeletogenesis. [ABSTRACT FROM PUBLISHER]

Abstract

Abstract: Results are presented of resistivity change measurements made on a model Fe–Cr–Ni–Ti–C austenitic alloy irradiated in the Fast Flux Test Facility to doses ranging from 1.87 to 67.8dpa. Two different electrical resistivity measurement systems were developed to overcome problems associated with examination of small microscopy specimens in order to investigate small changes in resistivity arising from voids and other radiation-induced microstructural features. A correlation is shown between resistivity changes arising primarily from void swelling. However, contributions arising probably from radiation-induced redistribution and perhaps precipitation of carbon and titanium can dominate the resistivity at low dpa levels. Abstract

Abstract: Neutron-induced microstructural evolution in response to long term irradiation at very low dose rates was studied for a Russian low-nickel austenitic stainless steel designated Х18Н9 that is analogous to AISI 304. The irradiated samples were obtained from an out-of-core support column for the pressure vessel of the BN-600 fast reactor with doses ranging from 1.7 to 20.5dpa generated at 3.8×10−9 to 4.3×10−8 dpa/s. The irradiation temperatures were in a very narrow range of 370–375°С. Microstructural observation showed that in addition to voids and dislocations, an unexpectedly high density of small G-phase precipitates was formed that are not usually observed at higher dpa rates in this temperature range. A similar behavior was observed in a Western stainless steel, namely AISI 304 stainless steel, irradiated at similar temperatures and somewhat higher dpa rates in the EBR-II fast reactor, indicating that irradiation at low dpa rates for many years can lead to a different precipitate microstructure and therefore different associated changes in matrix composition than are generated at higher dpa rates. The contribution of such radiation-induced precipitation to changes in electrical resistivity was measured in the Х18Н9 specimens and was shown to cause significant deviation from predictions based only on void swelling. Abstract

Checkpoint protein Chk1 has been identified as an Hsp90 client. Treatment with 100 nM geldanamycin (GM) for 24 h markedly reduced the Chk1 amount in Jurkat and ML-1 leukemia cell lines. Because Chk1 plays a central role in G2 checkpoint, we added GM to G2-arrested Jurkat and HL-60 cells pretreated with 50 nM doxorubicin for 24 h. GM slowly released both cell lines from doxorubicin-induced G2 arrest into G1 phase. GM also abrogated ICRF-193-induced decatenation G2 checkpoint in Jurkat and HL-60 cells. Western blot analysis showed that addition of GM attenuates doxorubicin- and ICRF-193-induced Chk1 phosphorylation at Ser345. GM, however, failed to abrogate G2 arrest in p53-positive ML-1 cells maybe due to the p21 induction. GM released HeLa cells from doxorubicin-induced G2 arrest but trapped them at M phase. Flow cytometric analysis showed that addition of GM converted doxorubicin-induced necrosis into apoptosis in Jurkat cells. Colony assay indicated that although GM has a weak cytotoxic effect as a single agent, it dramatically intensifies the cytotoxicity of doxorubicin and ICRF-193 in Jurkat and HL-60 cells.
