The bile acid intermediate 3α,7α,12α-trihydroxy-5β-cholestanoic acid (THCA) is converted to cholic acid exclusively in peroxisomes by the oxidative cleavage of the side chain. To investigate the mechanism by which the biosynthetic intermediates of bile acids are transported into peroxisomes, we incubated THCA or its CoA ester (THC-CoA) with isolated intact rat liver peroxisomes and analyzed their oxidation products, cholic acid and 3α,7α,12α-trihydroxy-5β-cholest-24-enoic acid. The oxidation of both THCA and THC-CoA was dependent on incubation time and peroxisomal proteins, and was stimulated by ATP. THC-CoA was efficiently oxidized to cholic acid and 3α,7α,12α-trihydroxy-5β-cholest-24-enoic acid as compared with THCA, suggesting that THC-CoA is the preferred substrate for transport into peroxisomes. The oxidation of THC-CoA was significantly inhibited by sodium azide, verapamile, and N-ethylmaleimide. Furthermore, the stimulatory effect of ATP on the oxidation was not replaced by GTP or AMP. In addition, the ATP-dependent oxidation of THC-CoA was markedly inhibited by pretreatment of peroxisomes with proteinase K when peroxisomal matrix proteins were not degraded. These results suggest that an ATP-dependent transport system for THC-CoA exists on peroxisomal membranes. [ABSTRACT FROM PUBLISHER]

109. Uniaxial-Pressure Effects on Spin-Driven Lattice Distortions in Geometrically Frustrated Magnets CuFe1-xGaxO2($x=0, 0.035$)

- Abstract
The triangular lattice antiferromagnet CuFeO2(CFO) is a spin--lattice coupled system, whose magnetic phase transitions are accompanied by crystal lattice distortions to relieve the geometrical spin frustration. Recent neutron diffraction and magnetic susceptibility measurements on CFO have revealed that the application of uniaxial pressure (up to 100 MPa) in the triangular lattice plane shifts the magnetic phase transition temperatures to higher values [Nakajima et al.: J. Phys. Soc. Jpn. 81(2012) 094710]. In the present study, we have performed synchrotron radiation x-ray diffraction measurements on CFO and CuFe1-xGaxO2(CFGO) with $x=0.035$ under applied uniaxial pressure in order to directly observe the uniaxial-pressure effects on the lattice. As a result, we have revealed that the applied uniaxial pressure certainly affects the crystal structure so that the structural transition temperatures increase as well as the magnetic phase transition temperatures. We have also found that CFO exhibits a pronounced lattice instability in the vicinity of the phase transition from the paramagnetic phase with a trigonal structure to an incommensurate magnetic phase with a monoclinic structure. In addition, the present results for CFGO ($x=0.035$) have revealed that the lattice instability is suppressed by the substitution of a small amount of nonmagnetic Ga3+ions for the magnetic Fe3+ions. This indicates that the spin degree of freedom plays an important role in the lattice instability in this system.

110. Strong-Gravity Effect on Twinned Y1Ba2Cu3O7-x Single Crystal

- Abstract
Ultracentrifuge experiments were performed on the twinned Y1Ba2Cu3O7-x (Y123) single crystal at much lower temperatures than the melting point. Two layers structure with slightly different compositions was observed in the sample ultracentrifuged at 250℃(380,000 G), which might be due to the sedimentation of atoms. In the strong gravity layer, it was found that the Y123 phase disappeared, and unknown XRD peaks appeared. Decomposition occurred in the sample ultracentrifuged at 400℃.

- Abstract
A visible four-layers structure with anomalous nano-sturucture was formed from a homogeneous -phase Bi3Pb7 intermetallic compound under a strong gravitational field (1.02x106 G, 130°C, 100 hours). In the 4th layer (lowest-gravity region), pure Bi particles precipitate. In the 2nd 3rd layers, composition graded structures, where Pb content increased along the gravity direction, were formed. It was found that the very broad XRD peak appeared in the 2nd layer, which indicated that an amorphous structure was contained.

- Abstract
Hematologic abnormalities are the most common symptoms of Shwachman-Diamond syndrome (SDS). The causative gene for SDS is the Shwachman-Bodian-Diamond syndrome (SBDS) gene; however, the function of SBDS and pathogenesis of each condition in SDS are largely unknown. SBDS is known to be localized at mitotic spindles and stabilizes microtubules. Previously, we demonstrated that SBDS is ubiquitinated and subsequently degraded in the mitotic phase, thereby accelerating mitotic progression. In this study, we examined mitosis in a myeloid cell model of SDS (SDS cells). 4',6-Diamidino-2-phenylindole (DAPI)-stained chromosome observation and cell cycle analysis of nocodazole-synchronized cells revealed that the SDS cells have abnormally rapid mitosis. In addition, many lagging chromosomes and micronuclei were detected. Moreover, the phosphorylation of threonine tyrosine kinase, the crucial kinase of the spindle assembly checkpoint (SAC), was suppressed. Chromosomal instability caused by SAC dysfunction may cause a variety of clinical conditions, including hematologic tumors in patients with SDS.

115. Anomalous Superfluid Density in a Disordered Charge-Density-Wave Material: Pd-Intercalated ErTe_{3}.

- Abstract
We image local superfluid density in single crystals of Pd-intercalated ErTe_{3} below the superconducting critical temperature T_{c}, well below the onset temperature T_{CDW} of (disordered) charge-density-wave order. We find no detectable inhomogeneities on micron scales. We observe a rapid increase of the superfluid density below T_{c}, deviating from the behavior expected in a conventional Bardeen-Cooper-Schrieffer superconductor, and show that the temperature dependence is qualitatively consistent with a combination of quantum and thermal phase fluctuations.

- Abstract
Farnesoid X receptor (FXR) is a nuclear receptor that regulates the synthesis and enterohepatic circulation of bile acids (BAs). It also regulates lipid and carbohydrate metabolism, making FXR ligands potential therapeutic agents for systemic and/or hepatic metabolic disorders. We previously synthesized a series of FXR antagonists and showed that oral administration of FLG249 reduced the expression of several FXR target genes in the mouse ileum. Here, we investigated the effects of FLG249 on lipid metabolism in mice fed a high-fat diet (HFD). When FLG249 was administered for 4 weeks to HFD-induced obese mice, it altered the expression of genes related to BA metabolism, ceramide synthesis and fatty acid β-oxidation, improving lipid metabolism in the liver and ileum without decreasing body weight. These findings suggest that FLG249 has the potential to be a low toxicity pharmaceutical compound and likely acts as a nonsteroidal FXR antagonist to improve lipid metabolism disorders.

- Abstract
As a cellular bile acid sensor, farnesoid X receptor (FXR) participates in regulation of bile acid, lipid and glucose homeostasis, and liver protection. With respect to the bone metabolism, FXR positively regulates bone metabolism through both bone formation and resorption of the bone remodeling pathways. Some of FXR agonists possessing isoxazole moiety are undergoing clinical trials for the treatment of non-alcoholic steatohepatitis. To date, therefore, the activation of FXR leads to considerable interest in FXR as potential therapeutic targets. We have identified a series of nonsteroidal FXR agonists bearing N
1 -methyl benzimidazole and isoxazole moieties that are bridged with aromatic derivatives. They showed affinity to FXR, but also weak affinity toward the vitamin D receptor (VDR) that involves regulation of calcium and phosphate homeostasis and is activated by bile acids. The deployment of FXR agonists without activity against VDR as off-target is therefore crucial in the development of FXR ligands. Our efforts focusing on increasing the agonist properties towards FXR led to the discovery of 19, which activates FXR at and below nanomolar levels (EC50 = 26.5 ± 10.5 nM TR-FRET and 0.8 ± 0.2 nM luciferase, respectively) and functions as a FXR agonist: the affinity toward FXR over eight nuclear receptors, including VDR [IC50 (VDR) / EC50 (FXR) > 5000] and TGR5, effects FXR target genes, and activates bone morphogenetic protein-2-induced differentiation of mouse bone marrow-derived mesenchymal stem cell-like ST2 cells into osteoblast., (Copyright © 2020 Elsevier Ltd.
