Fruit softening is a crucial factor that controls shelf life and commercial value. Pectate lyase (PL) has a major role in strawberry fruit softening. However, the PL gene family in strawberry has not been comprehensively analyzed. In this study, 65 FaPL genes were identified in the octoploid strawberry genome. Subcellular localization prediction indicated that FaPLs are mostly localized to the extracellular and cytoplasmic spaces. Duplication event analysis suggested that FaPL gene family expansion is mainly driven by whole genome or segmental duplication. The FaPL family members were classified into six groups according to the phylogenetic analysis. Among them, FaPL1, 3, 5, 20, 25, 42, and 57 had gradually increased expressions during strawberry fruit development and ripening and higher expression levels in the fruits with less firmness than that in firmer fruit. This result suggested that these members are involved in strawberry softening. Furthermore, overexpression of FaPL1 significantly reduced the fruit firmness, ascorbic acid (AsA), and malondialdehyde (MDA) content but obviously increased the anthocyanins, soluble proteins, and titratable acidity (TA), while it had no apparent effects on flavonoids, phenolics, and soluble sugar content. Background: Patients with early endometrial carcinoma (EC) have a good prognosis, but it is difficult to distinguish from endometrial polyps (EPs). Purpose: To develop and assess magnetic resonance imaging (MRI)-based radiomics models for discriminating Stage I EC from EP in a multicenter setting. Material and Methods: Patients with Stage I EC (n = 202) and EP (n = 99) who underwent preoperative MRI scans were collected in three centers (seven devices). The images from devices 1–3 were utilized for training and validation, and the images from devices 4–7 were utilized for testing, leading to three models. They were evaluated by the area under the receiver operating characteristic curve (AUC) and metrics including accuracy, sensitivity, and specificity. Two radiologists evaluated the endometrial lesions and compared them with the three models. Results: The AUCs of device 1, 2_ada, device 1, 3_ada, and device 2, 3_ada for discriminating Stage I EC from EP were 0.951, 0.912, and 0.896 for the training set, 0.755, 0.928, and 1.000 for the validation set, and 0.883, 0.956, and 0.878 for the external validation set, respectively. The specificity of the three models was higher, but the accuracy and sensitivity were lower than those of radiologists. Conclusion: Our MRI-based models showed good potential in differentiating Stage I EC from EP and had been validated in multiple centers. Verticillium wilt is a fungal disease in upland cotton and exerts a significant effect on growth and potential productivity. This disease is mainly caused by V. dahliae Kleb. Ethylene response factor (ERF) is one of the superfamilies of transcription factors that is involved in the development and environmental adaption of crops. A total of 30 ERF.B4 group members were detected in upland cotton and divided into 6 subgroups. Gene structures, conserved motifs, and domain analysis revealed that members in each subgroup are highly conserved. Further, the 30 GhERF.B4 group members were distributed on 18 chromosomes, and 36 gene synteny relationships were found among them. GhERF.B4 genes were ubiquitously expressed in various tissues and developmental stages of cotton. Amongst them, GhERF.B4-15D was predominantly expressed in roots, and its expression was induced by V. dahliae infection. In addition, GhERF.B4-15D responded to methyl jasmonate (MeJA), methyl salicylate (MeSA), and ethylene (ET) phytohormones. It was also found that the V. dahliae resistance was enhanced due to overexpression of GhERF.B4-15D in Arabidopsis thaliana. On the contrary, interference of GhERF.B4-15D by virus-induced gene silencing (VIGS) technology decreased the V. dahliae resistance level in upland cotton. The subcellular localization experiment showed that GhERF.B4-15D was located in the nucleus. Yeast two-hybrid (Y2H) and luciferase complementation (LUC) approaches demonstrated that GhERF.B4-15D interacted with GhDREB1B. Additionally, the V. dahliae resistance was significantly decreased in GhDREB1B knockdowns. Our results showed that GhERF.B4-15D plays a role during V. dahliae infection in cotton. SWEETs (sugars will eventually be exported transporters) play a vital role in longer-distance sugar transportation, and thus control carbon flow and energy metabolism in plants. SWEET genes have been identified in various plant species, but their functions in fruit development remain uncharacterized. Here, we isolated 15 putative PsSWEETs from the Prunus salicina genome. For further analysis, comprehensive bioinformatics methods were applied to determine the gene structure, chromosome distribution, phylogeny, cis-acting regulatory elements, and expression profiles of PsSWEETs. qRT-PCR analysis suggested that these SWEETs might have diverse functions in the development of plum fruit. The relative expression levels of PsSWEET1 and PsSWEET9 were obviously higher in ripened fruit than the ones in other developmental stages, suggesting their possible roles in the transport and accumulation of sugars in plum fruit. Positive correlations were found between the expression level of PsSWEET3/10/13 and the content of sucrose, and the expression level of PsSWEET2 and the content of fructose, respectively, during the development of 'Furongli' fruit, suggesting their possible roles in the accumulation of sucrose and fructose. The current study investigated the initial genomic characterization and expression patterns of the SWEET gene family in plum, which could provide a foundation for the further understanding of the functional analysis of the SWEET gene family. Background: Cytokinin oxidase/dehydrogenase (CKX) plays a vital role in response to abiotic stress through modulating the antioxidant enzyme activities. Nevertheless, the biological function of the CKX gene family has yet to be reported in cotton. Result: In this study, a total of 27 GhCKXs were identified by the genome-wide investigation and distributed across 18 chromosomes. Phylogenetic tree analysis revealed that CKX genes were clustered into four clades, and most gene expansions originated from segmental duplications. The CKXs gene structure and motif analysis displayed remarkably well conserved among the four groups. Moreover, the cis-acting elements related to the abiotic stress, hormones, and light response were identified within the promoter regions of GhCKXs. Transcriptome data and RT-qPCR showed that GhCKX genes demonstrated higher expression levels in various tissues and were involved in cotton's abiotic stress and phytohormone response. The protein-protein interaction network indicates that the CKX family probably participated in redox regulation, including oxidoreduction or ATP levels, to mediate plant growth and development. Functionally identified via virus-induced gene silencing (VIGS) found that the GhCKX14 gene improved drought resistance by modulating the antioxidant-related activitie. Conclusions: In this study, the CKX gene family members were analyzed by bioinformatics, and validates the response of GhCKX gene to various phytohormone treatment and abiotic stresses. Cotton bacterial blight (CBB), caused by the pathogen Xanthomonas citri subsp. malvacearum (Xcm), can inflict significant damage to cotton (Gossypium hirsutum L.) production. Previously, we identified and mapped the broad‐spectrum CBB‐resistant locus BB‐13 on the long arm of chromosome D02 using array‐based single nucleotide polymorphisms (SNPs). In the current study, linked SNPs were converted into easily assayable Kompetitive Allele‐Specific PCR (KASP) markers to enable efficient detection and marker‐assisted selection of alleles at the BB‐13 locus. The KASP marker's efficiency in detecting the BB‐13 resistant gene was validated using an Upland cotton diversity panel of 72 accessions phenotyped with Xcm race 18. The KASP marker NCBB‐KASP4, derived from the CottonSNP63K array‐based marker i25755Gh that is closely associated with BB‐13, predicted the CBB response phenotypes with an error rate of 4.17% in the diversity panel. Additionally, two independent biparental recombinant inbred line populations segregating for resistance to Xcm race 18 were used for KASP marker validation and to test their utility in detecting the presence of the BB‐13 locus in the resistant accession CABD3CABCH‐1‐89. NCBB‐KASP4, validated across breeding populations and broad germplasm, is a reliable KASP marker for detection and testing of BB‐13 locus in cotton. Further, diagnostic array‐based SNP marker i25755Gh's allele pattern and the potential CBB response is described for 875 Gossypium accessions. These SNP‐based phenotypic predictions for 875 accessions along with disease response phenotypes to Xcm race 18 for 253 accessions provide a reference for CBB resistance in diverse cotton germplasm in the United States. As unmanned vehicles become smaller and more autonomous, it is becoming feasible to use them in large groups with comparatively few human operators. Design and analysis of such distributed systems are complicated by the many interactions among agents and phenomena of human behavior. In particular, human susceptibility to fatigue and cognitive overload can introduce errors and uncertainty into the system. In this paper, we demonstrate how advanced computational tools can help to overcome these engineering difficulties by optimizing multirobot, multioperator surveillance systems for cost, speed, accuracy, and stealth according to diverse user preferences in multiple case studies. The tool developed is a graphical user interface that returns the optimal number and mix of diverse agent types as a function of the user's trade‐off preferences. System performance prediction relies on a multiagent simulation with submodels for human operators, fixed‐wing unmanned aerial vehicles (UAVs), quadrotor UAVs, and flapping wing UAVs combined in different numbers. Pseudomonas aeruginosa (P.a.) infection accounts for nearly 20% of all cases of hospital acquired pneumonia with mortality rates >30%. P.a. infection induces a robust inflammatory response, which ideally enhances bacterial clearance. Unfortunately, excessive inflammation can also have negative effects, and often leads to cardiac dysfunction with associated morbidity and mortality. However, it remains unclear how P.a. lung infection causes cardiac dysfunction. Using a murine pneumonia model, we found that P.a. infection of the lungs led to severe cardiac left ventricular dysfunction and electrical abnormalities. More specifically, we found that neutrophil recruitment and release of S100A8/A9 in the lungs activates the TLR4/RAGE signaling pathways, which in turn enhance systemic inflammation and subsequent cardiac dysfunction. Paradoxically, global deletion of S100A8/A9 did not improve but aggravated cardiac dysfunction and mortality likely due to uncontrolled bacterial burden in the lungs and heart. Our results indicate that P.a. infection induced release of S100A8/9 is double-edged, providing increased risk for cardiac dysfunction yet limiting P.a. growth. Author summary: Over 5 million people develop pneumonia annually in the United States and is the 8th leading cause of death worldwide. Surprisingly, up to 30% of patients admitted to the hospital for community acquired pneumonia develop cardiovascular complications but the mechanisms are not clearly understood. Pseudomonas aeruginosa infection is very common in hospital settings and accounts for up to 20% of all cases of hospital-acquired pneumonia with a mortality rate of ~30%. P. aeruginosa infection and severe lung and systemic inflammation increases levels of S100A8/9 (calprotectin) in the blood stream. Systemic inflammation or dissemination of pathogens into heart tissue can induce cardiac inflammation, cardiomyocyte death and cardiac dysfunction. We found that P. aeruginosa infection increases infiltration of inflammatory immune cells in the heart, leading to arrhythmia and left ventricular dysfunction. Our study provides evidence that S100A8/9 provides protection against bacterial infection yet when its production is uncontrolled it activates host immune responses leading to severe cardiovascular complications. Analyses of Illumina-based high-throughput sequencing data generated during characterization of the cotton leafroll dwarf virus population in Mississippi (2020–2022) consistently yielded contigs varying in size (most frequently from 4 to 7 kb) with identical nucleotide content and sharing similarities with reverse transcriptases (RTases) encoded by extant plant pararetroviruses (family Caulimoviridiae). Initial data prompted an in-depth study involving molecular and bioinformatic approaches to characterize the nature and origins of these caulimovirid-like sequences. As a result, here, we report on endogenous viral elements (EVEs) related to extant members of the family Caulimoviridae, integrated into a genome of upland cotton (Gossypium hirsutum), for which we propose the provisional name "endogenous cotton pararetroviral elements" (eCPRVE). Our investigations pinpointed a ~15 kbp-long locus on the A04 chromosome consisting of head-to-head orientated tandem copies located on positive- and negative-sense DNA strands (eCPRVE+ and eCPRVE-). Sequences of the eCPRVE+ comprised nearly complete and slightly decayed genome information, including ORFs coding for the viral movement protein (MP), coat protein (CP), RTase, and transactivator/viroplasm protein (TA). Phylogenetic analyses of major viral proteins suggest that the eCPRVE+ may have been initially derived from a genome of a cognate virus belonging to a putative new genus within the family. Unexpectedly, an identical 15 kb-long locus composed of two eCPRVE copies was also detected in a newly recognized species G. ekmanianum, shedding some light on the relatively recent evolution within the cotton family. The auxin receptor (TIR1/AFBs) family encodes the F-box protein subunit, which is involved in the formation of the E3 ubiquitin ligase SCFTIR1/AFBs complex, a key component of the auxin signaling pathway. However, there are few studies on the auxin receptor family in apple (Malus × domestica). In this study, eight MdAFBs were identified, and phylogenetic analysis showed that they were classified into four groups and distributed on eight chromosomes. Herein, a comprehensive analysis of the MdAFB gene family was conducted to identify cis-acting elements, gene structures, protein structures, aligned sequences, conserved motifs, conserved amino acids, and the proteineprotein interaction network. The results of yeast two-hybrid assays showed that MdAFB1 interacted with three auxin repressor proteins. The results of qRT-PCR showed that MdAFB1 responded to osmotic and salt stress. The overexpression of MdAFB1 increased osmotic and salt resistance in apple calli, and the ectopic expression of MdAFB1 enhanced osmotic and salt tolerance in Arabidopsis. Cytokinins are members of a group of phytohormones involved in various growth and developmental processes in plants. Isopentenyl transferase (IPT) is the rate-limiting enzyme in catalyzing the biosynthesis of cytokinins. In this study, to understand the role of IPT family in cold resistance, 78 IPT candidates were identified and characterized in nine Rosaceae genomes. The expansion of IPT families in the Rosaceae primarily occurred through segmental duplication rather than tandem duplication. In general, purifying selection controlled the evolution of IPT families in the Rosaceae, with IPT3 and IPT5 homologs as the primary drivers of evolution. Cis-elements, which are involved in the responses to many environmental stresses or phytohormone signals, were identified in the promoters of MdIPT members. This was consistent with the trends of expression of the MdIPT genes in apple (Malus domestica) calli. MdIPT5b was also found to exhibit multiple responses to phytohormones and stress signals. The ectopic expression of MdIPT5b resulted in an increase in cold resistance in transformed apple calli and tomato (Solanum lycopersicum) plantlets. The redox balance was partially stabilized through the accumulation of proline under cold stress. However, the ascorbateeglutathione cycle cannot be stabilized in the cold. All physiological and biochemical assays are preformed in spectrophotometer. These results showed that regulating the expression of IPT genes for moderate cytokinin improvement could enhance the accumulation of proline to stabilize the osmotic and redox balances to improve resistance to cold stress. Cognitive function in humans depends on the complex and interplay between multiple body systems, including the hypothalamic-pituitary-adrenal (HPA) axis. The gut microbiota, which vastly outnumbers human cells and has a genetic potential that exceeds that of the human genome, plays a crucial role in this interplay. The microbiota-gut-brain (MGB) axis is a bidirectional signalling pathway that operates through neural, endocrine, immune, and metabolic pathways. One of the major neuroendocrine systems responding to stress is the HPA axis which produces glucocorticoids such as cortisol in humans and corticosterone in rodents. Appropriate concentrations of cortisol are essential for normal neurodevelopment and function, as well as cognitive processes such as learning and memory, and studies have shown that microbes modulate the HPA axis throughout life. Stress can significantly impact the MGB axis via the HPA axis and other pathways. Animal research has advanced our understanding of these mechanisms and pathways, leading to a paradigm shift in conceptual thinking about the influence of the microbiota on human health and disease. Preclinical and human trials are currently underway to determine how these animal models translate to humans. In this review article, we summarize the current knowledge of the relationship between the gut microbiota, HPA axis, and cognition, and provide an overview of the main findings and conclusions in this broad field. This study investigates the cognitive levels of tugboat captains, key players during port manoeuvres, which are a crucial part of sea navigation. The objective is to reveal the main neurophysiological findings related to the measurements of the tugboat captains' brain activity during both rest and actual manoeuvring performance and to investigate the relationship between this brain activity and situational awareness. The study employed an experimental research method. Brain waves of the tugboat captains were recorded using the Emotiv X EEG device and the Emotiv Pro v2.0 program during real port manoeuvres which is a part of the sea navigation. Situational awareness levels were tried to be determined by analysing the obtained values in the Brain Products Vision Analyzer 2.1 software. The study examined the cognitive states of four tugboat captains during 16 manoeuvres and resting, focusing on the Fast Fourier Transform (FFT)/Band power graph values. Within the scope of the research non-parametric Friedman Test with five variables [Resting State (RS), 1st Manoeuvre Group, 2nd Manoeuvre Group, 3rd Manoeuvre Group, and 4th Manoeuvre Group] to analyse the FFT values of the participants. Post-hoc comparisons were calculated using Bonferroni correction. The results showed no difference in alpha wave power, while delta, theta, beta, and gamma power decreased. The primary outcome of the research indicates that professional tugboat captains, who were the participants of the study, exhibited consistent situational awareness levels both during manoeuvring and resting moments. Despite innovative advances in anti-infective therapies and vaccine development technologies, community-acquired pneumonia (CAP) remains the most persistent cause of infection-related mortality globally. Confronting the ongoing threat posed by Streptococcus pneumoniae (the pneumococcus), the most common bacterial cause of CAP, particularly to the non-immune elderly, remains challenging due to the propensity of the elderly to develop invasive pneumococcal disease (IPD), together with the predilection of the pathogen for the heart. The resultant development of often fatal cardiovascular events (CVEs), particularly during the first seven days of acute infection, is now recognized as a relatively common complication of IPD. The current review represents an update on the prevalence and types of CVEs associated with acute bacterial CAP, particularly IPD. In addition, it is focused on recent insights into the involvement of the pneumococcal pore-forming toxin, pneumolysin (Ply), in subverting host immune defenses, particularly the protective functions of the alveolar macrophage during early-stage disease. This, in turn, enables extra-pulmonary dissemination of the pathogen, leading to cardiac invasion, cardiotoxicity and myocardial dysfunction. The review concludes with an overview of the current status of macrolide antibiotics in the treatment of bacterial CAP in general, as well as severe pneumococcal CAP, including a consideration of the mechanisms by which these agents inhibit the production of Ply by macrolide-resistant strains of the pathogen. With the Internet of Things (IoT) development, there is an increasing demand for multi-service scheduling for Mobile Edge Computing (MEC). We propose using polling for scheduling in edge computing to accommodate multi-service scheduling methods better. Given the complexity of asymmetric polling systems, we have used an information-theoretic approach to analyse the model. Firstly, we propose an asymmetric two-level scheduling approach with priority based on a polling scheduling approach. Secondly, the mathematical model of the system in the continuous time state is established by using the embedded Markov chain theory and the probability-generating function. By solving for the probability-generating function's first-order partial and second-order partial derivatives, we calculate the exact expressions of the average queue length, the average polling period, and the average delay with an approximate analysis of periodic query way. Finally, we design a simulation experiment to verify that our derived parameters are correct. One emerging problem for onco-gynecologists is the incidence of premenopausal patients under 40 years of age diagnosed with stage I Endometrial Cancer (EC) who want to preserve their fertility. Our review aims to define a primary risk assessment that can help fertility experts and onco-gynecologists tailor personalized treatment and fertility-preserving strategies for fertile patients wishing to have children. We confirm that risk factors such as myometrial invasion and The International Federation of Gynecology and Obstetrics (FIGO) staging should be integrated into the novel molecular classification provided by The Cancer Genome Atlas (TCGA). We also corroborate the influence of classical risk factors such as obesity, Polycystic ovarian syndrome (PCOS), and diabetes mellitus to assess fertility outcomes. The fertility preservation options are inadequately discussed with women with a diagnosis of gynecological cancer. A multidisciplinary team of gynecologists, oncologists, and fertility specialists could increase patient satisfaction and improve fertility outcomes. The incidence and death rates of endometrial cancer are rising globally. International guidelines recommend radical hysterectomy and bilateral salpingo-oophorectomy as the standard of care for this cancer; however, fertility-sparing alternatives should be tailored to motivated women of reproductive age, establishing an appropriate cost–benefit balance between childbearing desire and cancer risk. New molecular classifications such as that of TCGA provide a robust supplementary risk assessment tool that can tailor the treatment options to the patient's needs, curtail over- and under-treatment, and contribute to the spread of fertility-preserving strategies. In recent years, the development of deep learning technology has significantly benefited agriculture in domains such as smart and precision farming. Deep learning models require a large amount of high-quality training data. However, collecting and managing large amounts of guaranteed-quality data is a critical issue. To meet these requirements, this study proposes a scalable plant disease information collection and management system (PlantInfoCMS). The proposed PlantInfoCMS consists of data collection, annotation, data inspection, and dashboard modules to generate accurate and high-quality pest and disease image datasets for learning purposes. Additionally, the system provides various statistical functions allowing users to easily check the progress of each task, making management highly efficient. Currently, PlantInfoCMS handles data on 32 types of crops and 185 types of pests and diseases, and stores and manages 301,667 original and 195,124 labeled images. The PlantInfoCMS proposed in this study is expected to significantly contribute to the diagnosis of crop pests and diseases by providing high-quality AI images for learning about and facilitating the management of crop pests and diseases. Rickettsial agents are a diverse group of alpha-proteobacteria within the order Rickettsiales, which possesses two families with human pathogens, Rickettsiaceae and Anaplasmataceae. These obligate intracellular bacteria are most frequently transmitted by arthropod vectors, a first step in the pathogens' avoidance of host cell defenses. Considerable study of the immune responses to infection and those that result in protective immunity have been conducted. Less study has focused on the initial events and mechanism by which these bacteria avoid the innate immune responses of the hosts to survive within and propagate from host cells. By evaluating the major mechanisms of evading innate immunity, a range of similarities among these bacteria become apparent, including mechanisms to escape initial destruction in phagolysosomes of professional phagocytes, those that dampen the responses of innate immune cells or subvert signaling and recognition pathways related to apoptosis, autophagy, proinflammatory responses, and mechanisms by which these microbes attach to and enter cells or those molecules that trigger the host responses. At birth, mammals experience a massive colonization by microorganisms. We previously reported that newborn mice gestated and born germ-free (GF) have increased microglial labeling and alterations in developmental neuronal cell death in the hippocampus and hypothalamus, as well as greater forebrain volume and body weight when compared to conventionally colonized (CC) mice. To test whether these effects are solely due to differences in postnatal microbial exposure, or instead may be programmed in utero, we cross-fostered GF newborns immediately after birth to CC dams (GF!CC) and compared them to offspring fostered within the same microbiota status (CC!CC, GF!GF). Because key developmental events (including microglial colonization and neuronal cell death) shape the brain during the first postnatal week, we collected brains on postnatal day (P) 7. To track gut bacterial colonization, colonic content was also collected and subjected to 16S rRNA qPCR and Illumina sequencing. In the brains of GF!GF mice, we replicated most of the effects seen previously in GF mice. Interestingly, the GF brain phenotype persisted in GF!CC offspring for almost all measures. In contrast, total bacterial load did not differ between the CC!CC and GF!CC groups on P7, and bacterial community composition was also very similar, with a few exceptions. Hormones act as master ripening regulators. In non-climacteric fruit, ABA plays a key role in ripening. Recently, we confirmed in Fragaria chiloensis fruit that in response to ABA treatment the fruit induces ripening-associated changes such as softening and color development. In consequence of these phenotypic changes, transcriptional variations associated with cell wall disassembly and anthocyanins biosynthesis were reported. As ABA stimulates the ripening of F. chiloensis fruit, the molecular network involved in ABA metabolism was analyzed. Therefore, the expression level of genes involved in ABA biosynthesis and ABA perception was quantified during the development of the fruit. Four NCED/CCDs and six PYR/PYLs family members were identified in F. chiloensis. Bioinformatics analyses confirmed the existence of key domains related to functional properties. Through RT-qPCR analyses, the level of transcripts was quantified. FcNCED1 codifies a protein that displays crucial functional domains, and the level of transcripts increases as the fruit develops and ripens, in parallel with the increment in ABA. In addition, FcPYL4 codifies for a functional ABA receptor, and its expression follows an incremental pattern during ripening. Legumes comprise one of the world's largest, most diverse, and economically important plant families, known for their nutritional and medicinal benefits. Legumes are susceptible to a wide range of diseases, similar to other agricultural crops. Diseases have a considerable impact on the production of legume crop species, resulting in large yield losses worldwide. Due to continuous interactions between plants and their pathogens in the environment and the evolution of new pathogens under high selection pressure; disease resistant genes emerge in plant cultivars in the field against those pathogens or disease. Thus, disease resistant genes play critical roles in plant resistance responses, and their discovery and subsequent use in breeding programmes aid in reducing yield loss. The genomic era, with its high-throughput and low-cost genomic tools, has revolutionised our understanding of the complex interactions between legumes and pathogens, resulting in the identification of several critical participants in both the resistant and susceptible relationships. However, a substantial amount of existing information about numerous legume species has been disseminated as text or is preserved across fractions in different databases, posing a challenge for researchers. As a result, the range, scope, and complexity of these resources pose challenges to those who manage and use them. Therefore, there is an urgent need to develop tools and a single conjugate database to manage genetic information for the world's plant genetic resources, allowing for the rapid incorporation of essential resistance genes into breeding strategies. Here, developed the first comprehensive database of disease resistance genes named as LDRGDb - LEGUMES DISEASE RESISTANCE GENES DATABASE comprises 10 legumes [Pigeon pea (Cajanus cajan), Chickpea (Cicer arietinum), Soybean (Glycine max), Lentil (Lens culinaris), Alfalfa (Medicago sativa), Barrelclover (Medicago truncatula), Common bean (Phaseolus vulgaris), Pea (Pisum sativum), Faba bean (Vicia faba), and Cowpea (Vigna unguiculata)]. In cotton, most agronomic traits are controlled by polygenes. In this study, 110 F6 recombinant inbred lines (RILs), derived from Upland cotton cross NC05AZ06 x NC11‐2100, were used to develop a linkage map and to identify quantitative trait loci (QTL) for six fiber quality traits, three yield traits, and plant height. These RILs were genotyped using the CottonSNP63K array and phenotyped for fiber quality, yield traits, and plant height in 2‐year field trials. Analysis of variance revealed significant (p < 0.05) differences among RILs for all traits studied, and the heritability estimates were moderate (30%–60%) to high (> 60%). Both positive and negative correlations were observed for fiber quality and yield traits. A total of 3,774 polymorphic SNP markers were used to develop a genetic map with an average marker density of 1.54 SNP/cM. Thirty QTL for fiber quality traits, yield traits, and plant height were detected on 15 different chromosomes, explaining 6.80%–20.02% of the phenotypic variance (PVE). Of these, 14 were major QTL (PVE > 10%), and three major QTL were detected in both years. Candidate gene analysis in the major QTL detected in both years and plant height QTL with PVE of 20.02% revealed five putative genes for fiber quality traits and one putative gene for plant height. The linkage map and identified QTL along with the candidate genes in the study could serve as additional breeding resources for Upland cotton genetic improvement. Introduction: Despite epidemiological associations between community acquired pneumonia (CAP) and myocardial infarction, mechanisms that modify cardiovascular disease during CAP are not well defined. In particular, largely due to a lack of relevant experimental models, the effect of pneumonia on atherosclerotic plaques is unclear. We describe the development of a murine model of the commonest cause of CAP, Streptococcus pneumoniae pneumonia, on a background of established atherosclerosis. We go on to use our model to investigate the effects of pneumococcal pneumonia on atherosclerosis. Methods: C57BL/6J and ApoE-/- mice were fed a high fat diet to promote atherosclerotic plaque formation. Mice were then infected with a range of S. pneumoniae serotypes (1, 4 or 14) with the aim of establishing a model to study atherosclerotic plaque evolution after pneumonia and bacteremia. Laser capture microdissection of plaque macrophages enabled transcriptomic analysis. Results: Intratracheal instillation of S. pneumoniae in mice fed a cholate containing diet resulted in low survival rates following infection, suggestive of increased susceptibility to severe infection. Optimization steps resulted in a final model of male ApoE-/- mice fed a Western diet then infected by intranasal instillation of serotype 4 (TIGR4) S. pneumoniae followed by antibiotic administration. This protocol resulted in high rates of bacteremia (88.9%) and survival (88.5%). Pneumonia resulted in increased aortic sinus plaque macrophage content 2 weeks post pneumonia but not at 8 weeks, and no difference in plaque burden or other plaque vulnerability markers were found at either time point. Microarray and qPCR analysis of plaque macrophages identified downregulation of two E3 ubiquitin ligases, Huwe1 and Itch, following pneumonia. Treatment with atorvastatin failed to alter plaque macrophage content or other plaque features. Discussion: Without antibiotics, ApoE-/- mice fed a high fat diet were highly susceptible to mortality following S. pneumoniae infection. The major infection associated change in plaque morphology was an early increase in plaque macrophages. Conservative management of endometrial cancer (CMEC) is viable for women with early-stage disease wishing to preserve fertility, but there is poor knowledge regarding clinicians' attitudes towards treatment or guidelines adherence. This 55-item survey study investigated CMEC-related experience, practice and attitudes among clinically active Swedish gynecologists and gynecological oncologists, focusing on reproductive eligibility criteria. The survey consisted of a general and two specific subsets, selectively delivered to clinicians active in infertility (subset A) and endometrial cancer (subset B) care. Answers from 218 clinicians were included. More than half agreed on CMEC whereas only 5% explicitly disagreed. The majority supported a fertility work-up to substantiate reasonable chances to pregnancy and live birth. Most disagreed about CMEC in case of previous unsuccessful fertility treatments, while more than 1/3 disagreed about CMEC in known fertility problems, recurrent miscarriages or previous children. Over 50% of respondents in subset A (n = 107) found it applicable with fertility investigations such as ovarian reserve testing or, in case of male partner, semen analysis. Respondents in subset B (n = 165) agreed on items based on existing recommendations regarding the oncological management of CMEC, including the use of continuous progestins, hysteroscopic resection of macroscopic lesions, control biopsy with curettage or hysteroscopy after 6 months of treatment, pursuing pregnancy as soon as possible after complete response, and performing a hysterectomy once live birth is achieved. While many clinicians were familiar with CMEC, the overall experience is limited. Background: Upland cotton (Gossypium hirsutum L.) is the most economically important species in the cotton genus (Gossypium spp.). Enhancing the cotton yield is a major goal in cotton breeding programs. Lint percentage (LP) and boll weight (BW) are the two most important components of cotton lint yield. The identification of stable and effective quantitative trait loci (QTLs) will aid the molecular breeding of cotton cultivars with high yield. Results: Genotyping by target sequencing (GBTS) and genome-wide association study (GWAS) with 3VmrMLM were used to identify LP and BW related QTLs from two recombinant inbred line (RIL) populations derived from high lint yield and fiber quality lines (ZR014121, CCRI60 and EZ60). The average call rate of a single locus was 94.35%, and the average call rate of an individual was 92.10% in GBTS. A total of 100 QTLs were identified; 22 of them were overlapping with the reported QTLs, and 78 were novel QTLs. Of the 100 QTLs, 51 QTLs were for LP, and they explained 0.29–9.96% of the phenotypic variation; 49 QTLs were for BW, and they explained 0.41–6.31% of the phenotypic variation. One QTL (qBW-E-A10-1, qBW-C-A10-1) was identified in both populations. Six key QTLs were identified in multiple-environments; three were for LP, and three were for BW. A total of 108 candidate genes were identified in the regions of the six key QTLs. Several candidate genes were positively related to the developments of LP and BW, such as genes involved in gene transcription, protein synthesis, calcium signaling, carbon metabolism, and biosynthesis of secondary metabolites. Seven major candidate genes were predicted to form a co-expression network. Six significantly highly expressed candidate genes of the six QTLs after anthesis were the key genes regulating LP and BW and affecting cotton yield formation. Conclusions: A total of 100 stable QTLs for LP and BW in upland cotton were identified in this study; these QTLs could be used in cotton molecular breeding programs. The error negativity/error-related negativity (Ne/ERN) is one of the most well-studied event-related potential (ERP) components in the electroencephalography (EEG) literature. Peaking about 50 ms after the commission of an error, the Ne/ERN is a negative deflection in the ERP waveform that is thought to reflect error processing in the brain. While its relationships to trait constructs such as anxiety are well-documented, there is still little known about how the Ne/ERN may subsequently influence task-related behavior. In other words, does the occurrence of the Ne/ERN trigger any sort of error corrective process, or any other behavioral adaptation to avoid errors? Several theories have emerged to explain how the Ne/ERN may implement or affect behavior on a task, but evidence supporting each has been mixed. In the following manuscript, we review these theories, and then systematically discuss the reasons that there may be discrepancies in the literature. We review both the inherent biological factors of the neural regions that underlie error-processing in the brain, and some of the researcher-induced factors in analytic and experimental choices that may be exacerbating these discrepancies. Membrane ion channels of the calcium homeostasis modulator (CALHM) family promote cell–cell crosstalk at neuronal synapses via ATP release, where ATP acts as a neurotransmitter. CALHM6, the only CALHM highly expressed in immune cells, has been linked to the induction of natural killer (NK) cell anti‐tumour activity. However, its mechanism of action and broader functions in the immune system remain unclear. Here, we generated Calhm6−/− mice and report that CALHM6 is important for the regulation of the early innate control of Listeria monocytogenes infection in vivo. We find that CALHM6 is upregulated in macrophages by pathogen‐derived signals and that it relocates from the intracellular compartment to the macrophage‐NK cell synapse, facilitating ATP release and controlling the kinetics of NK cell activation. Anti‐inflammatory cytokines terminate CALHM6 expression. CALHM6 forms an ion channel when expressed in the plasma membrane of Xenopus oocytes, where channel opening is controlled by a conserved acidic residue, E119. In mammalian cells, CALHM6 is localised to intracellular compartments. Our results contribute to the understanding of neurotransmitter‐like signal exchange between immune cells that fine‐tunes the timing of innate immune responses. MdPG1 encoding polygalacturonase in apple (Malus × domestica) is a key gene associated with fruit firmness and texture variations among apple cultivars. However, the causative variants of MdPG1 are still not known. In this study, we identified a SNPA/C variant within an ERF-binding element located in the promoter region of MdPG1. The promoter containing the ERF-binding element with SNPA, rather than the SNPC, could be strongly bound and activated by MdCBF2, a member of the AP2/ERF transcription factor family, as determined by yeast-one-hybrid and dual-luciferase reporter assays. We also demonstrated that the presence of a novel long non-coding RNA, lncRNAPG1, in the promoter of MdPG1 was a causative variant. lncRNAPG1 was specifically expressed in fruit tissues postharvest. lncRNAPG1 could reduce promoter activity when it was fused to the promoter of MdPG1 and a tobacco gene encoding Mg-chelatase H subunit (NtCHLH) in transgenic tobacco cells but could not reduce promoter activity when it was supplied in a separate gene construct, indicating a cis-regulatory effect. Rose rosette disease (RRD), caused by the rose rosette emaravirus (RRV), is a major viral disease in roses (Rosa sp.) that threatens the rose industry. Recent studies have revealed quantitative trait loci (QTL) for reduced susceptibility to RRD in the linkage groups (LGs) 1, 5, 6, and 7 in tetraploid populations and the LGs 1, 3, 5, and 6 in diploid populations. In this study, we seek to better localize and understand the relationship between QTL identified in both diploid and tetraploid populations. We do so by remapping the populations found in these studies and performing a meta-analysis. This analysis reveals that the peaks and intervals for QTL using diploid and tetraploid populations co-localized on LG 1, suggesting that these are the same QTL. The same was seen on LG 3. Three meta-QTL were identified on LG 5, and two were discovered on LG 6. The meta-QTL on LG 1, MetaRRD1.1, had a confidence interval (CI) of 10.53 cM. On LG 3, MetaRRD3.1 had a CI of 5.94 cM. MetaRRD5.1 had a CI of 17.37 cM, MetaRRD5.2 had a CI of 4.33 cM, and MetaRRD5.3 had a CI of 21.95 cM. For LG 6, MetaRRD6.1 and MetaRRD6.2 had CIs of 9.81 and 8.81 cM, respectively. The analysis also led to the identification of potential disease resistance genes, with a primary interest in genes localized in meta-QTL intervals on LG 5 as this LG was found to explain the greatest proportion of phenotypic variance for RRD resistance. Purpureocillium lilacinum is a biocontrol Ascomycota fungus against various plant pathogens. In the present study, the efficacy of P. lilacinum was evaluated against a root-knot nematode, Meloidogyne incognita that infects eggplants. We performed an in vitro experiment in which the direct effects of P. lilacinum on the second-stage juvenile survival and egg hatching of M. incognita were tested at different exposure times. The results showed that P. lilacinum significantly reduced the rates of egg hatching and juvenile survival in a dose-dependent manner. Microscopic observation demonstrated that P. lilacinum directly penetrated the eggs and contacted the juveniles, indicating how P. lilacinum parasitizes M. incognita. We also performed a pot assay in which soil-grown eggplants were treated with P. lilacinum followed by inoculation with M. incognita. The results indicated that P. lilacinum effectively reduced the nematode population and the number of galls in plant roots. Interestingly, application of P. lilacinum resulted in significant enhancements in plant growth and biomass, even under nematode infection, while it improved plant photosynthetic pigments, i.e., chlorophyll and carotenoids. Taken together, our study suggested that P. lilacinum can be used as a plant growth-promoting fungus and a biological nematicide for disease management of root-knot nematodes in eggplants. The Prunus genus encompasses a group of economically important and closely related crops, sharing an essentially common genome and, thereby, a high level of conserved and transferable microsatellite (SSR) loci. In Southern Italy, many of the local and/or neglected varieties are abandoned and at risk of extinction due to the high degree of urbanization and agricultural intensification, despite their value as genetic resources for crop improvement. This research aimed to genetically and morphologically characterize the traditional apricot (P. armenica) and peach (P. persica) germplasms collected in old family orchards. Most of the official descriptor categories were scored, thus revealing a rather high level of phenotypic variation in both collections. Genetic data allowed the discovery of diversity masked by morphological traits. Genotyping in 15 and 18 SSRs, eight of which were transferable across both species, showed an average polymorphic informativeness (PIC) of 0.44 and 0.59 for apricot and peach, respectively, and a total of 70 and 144 alleles. A reliable identification of each genotype was achieved, and the presence of possible mislabeling and/or erroneous denominations was solved. These results are encouraging for the valorization of the still poorly explored Italian Prunus germplasm, with significant economic consequences for bioresource conservation and management. Subtilases (SBTs), which belong to the serine peptidases, control plant development by regulating cell wall properties and the activity of extracellular signaling molecules, and affect all stages of the life cycle, such as seed development and germination, and responses to biotic and abiotic environments. In this study, 146 Gossypium hirsutum, 138 Gossypium barbadense, 89 Gossypium arboreum and 84 Gossypium raimondii SBTs were identified and divided into six subfamilies. Cotton SBTs are unevenly distributed on chromosomes. Synteny analysis showed that the members of SBT1 and SBT4 were expanded in cotton compared to Arabidopsis thaliana. Co-expression network analysis showed that six Gossypium arboreum SBT gene family members were in a network, among which five SBT1 genes and their Gossypium hirsutum and Arabidopsis thaliana direct homologues were down-regulated by salt treatment, indicating that the co-expression network might share conserved functions. Through co-expression network and annotation analysis, these SBTs may be involved in the biological processes of auxin transport, ABA signal transduction, cell wall repair and root tissue development. In summary, this study provides valuable information for the study of SBT genes in cotton and excavates SBT genes in response to salt stress, which provides ideas for cotton breeding for salinity resistance. Penicillin-binding proteins (PBPs) are considered essential for bacterial peptidoglycan biosynthesis and cell wall assembly. Clavibacter michiganensis is a representative Gram-positive bacterial species that causes bacterial canker in tomato. pbpC plays a significant role in maintaining cell morphological characteristics and stress responses in C. michiganensis. The current study demonstrated that the deletion of pbpC commonly enhances bacterial pathogenicity in C. michiganensis and revealed the mechanisms through which this occurs. The expression of interrelated virulence genes, including celA, xysA, xysB, and pelA, were significantly upregulated in △pbpC mutants. Compared with those in wild-type strains, exoenzyme activities, the formation of biofilm, and the production of exopolysaccharides (EPS) were significantly increased in △pbpC mutants. It is noteworthy that EPS were responsible for the enhancement in bacterial pathogenicity, with the degree of necrotic tomato stem cankers intensifying with the injection of a gradient of EPS from C. michiganensis. These findings highlight new insights into the role of pbpC affecting bacterial pathogenicity, with an emphasis on EPS, advancing the current understanding of phytopathogenic infection strategies for Gram-positive bacteria. Sea Island cotton is the best quality tetraploid cultivated cotton in the world, in terms of fiber quality. Glyphosate is a widely used herbicide in cotton production, and the improper use of herbicides has led to pollen abortion in sea island cotton and, consequently, to a dramatic decrease in yield; however, the mechanism remains unclear. In this study, different concentrations (0, 3.75, 7.5, 15, and 30 g/L) of glyphosate were applied to CP4-EPSPS transgenic sea island cotton Xinchang 5 in 2021 and 2022 at Korla, with 15 g/L glyphosate chosen as the suitable concentration. By comparing the paraffin sections of 2–24 mm anthers in the 15 g/L glyphosate treatment group with those in the water control group, we showed that the key period of anther abortion after glyphosate treatment was the formation and development of tetrads, which corresponded to 8–9 mm buds. Transcriptome sequencing analysis of the treated and control anthers revealed a significant enrichment of differentially expressed genes in phytohormone-related pathways, in particular abscisic acid response and regulation pathways. Additionally, after treatment with 15 g/L of glyphosate, there was a significant increase in the amount of abscisic acid in the anthers in the 8–9 mm buds. Further analysis of the differential expression of abscisic acid response and regulatory genes, an abscisic acid response gene GbTCP14 (Gbar_A11G003090) was identified, which was significantly upregulated in buds with 15 g/L glyphosate treatment than the control, and it could be a key candidate gene for the subsequent research involving male sterility induced by glyphosate in sea island cotton. Onion (Allium cepa L.) bacterial diseases can cause catastrophic crop losses. Chemical and biological control products are not curative and have mixed or limited efficacy at preventing onion bacterial diseases. Cultural management strategies, such as appropriate irrigation and nitrogen (N) management practices, can reduce the risk for bacterial infection and disease progression. This review synthesizes available literature on cultural management strategies to mitigate onion bacterial diseases and identifies knowledge gaps, promising strategies, and limitations. This synthesis revealed that high N fertilizer application rates, the use of overhead irrigation, and late‐season irrigation or rainfall, especially during field curing, can increase bacterial disease pressure. These factors contribute to conducive conditions for bacteria to colonize the foliage and neck of onion plants. Other in‐season strategies, such as reducing the total amount of irrigation or adjusting application rates of nutrients other than N, have not limited losses to bacterial bulb rots while maintaining desired yields. At the end of the growing season, appropriate timing of lifting bulbs (undercutting to sever the roots) and other practices that speed field curing can reduce the risk of bacterial bulb rots. However, it is less clear if topping (removing foliage) before or after curing influences bacterial bulb rots. Conflicting results reported for some cultural management strategies indicate that local conditions, such as climate and soil types, must be considered for optimizing practices to manage onion bacterial diseases. Advancing our scientific understanding and grower awareness of strategies that reduce onion bacterial diseases is a cost‐effective opportunity to avoid losses. Soil salinization–alkalization is a major hindrance to agricultural development globally. Tea crabapple is widely used in China. However, little remains known regarding the molecular mechanisms used to withstand mixed saline–alkali stress (MSAS). Herein, we exposed tea crabapple seedlings to MSAS, and RNA-seq was performed for the transcriptome analysis of roots. Between 43.26 million and 43.37 million clean reads were thus obtained. In comparison with the control group (day 0), 2931, 2335, and 3746 genes were differentially expressed at day 1, day 3, and day 6 of MSAS exposure, respectively, and 1022 genes were common in the three comparison groups. On functional annotation, we observed that numerous differentially expressed genes were involved in "global and overview maps"; "carbohydrate metabolism"; "folding, sorting, and degradation"; "biosynthesis of other secondary metabolites"; "environmental adaptation"; and "signal transduction." Heat shock proteins, cytochrome P450s, disease-resistant proteins, non-specific lipid-transfer proteins, pectate lyase, and beta-glucosidases were also induced in response to MSAS, in addition to nitrogen, phosphorus, and potassium absorption and metabolism-related genes. Transcription factor-coding genes appear to regulate the response of tea crabapple roots to MSAS by participating in, for example, plant hormone signal transduction and heat shock response. We also performed quantitative real-time PCR to validate the expression of six differentially expressed genes. Simple Summary: TIR-NBS-LRR (TNL) is a disease resistance gene family that responds to biotic stress in many plants, but the systematic analysis of this gene family and the expression response to biotic stress have rarely been reported in roses. In the present study, 96 intact TNL gene family members were identified by bioinformatics in Rosa chinensis, and analyzed from the perspectives of evolutionary relationships, conserved structures, expression regulation, collinear relationships, and expression patterns. Some of the TNL genes responded to hormones and fungal disease: RcTNL23 demonstrated strong responses to three hormones and three pathogens. In addition, some TNL genes responded significantly to the black spot pathogen that we isolated, and different members may be involved in different stages of disease defense. In conclusion, we found that the TNL gene family is involved in the response to fungal disease and may function as a disease resistance gene in the rose. The present study lays a theoretical foundation for the functional study of TNL genes and the mining of disease resistance gene in roses, which will inform the selection and breeding of disease-resistant varieties. Roses, which are one of the world's most important ornamental plants, are often damaged by pathogens, resulting in serious economic losses. As a subclass of the disease resistance gene family of plant nucleotide-binding oligomerization domain (NOD)-like receptors, TIR-NBS-LRR (TNL) genes play a vital role in identifying pathogen effectors and activating defense responses. However, a systematic analysis of the TNL gene family is rarely reported in roses. Herein, 96 intact TNL genes were identified in Rosa chinensis. Their phylogenies, physicochemical characteristics, gene structures, conserved domains and motifs, promoter cis-elements, microRNA binding sites, and intra- and interspecific collinearity relationships were analyzed. An expression analysis using transcriptome data revealed that RcTNL genes were dominantly expressed in leaves. Some RcTNL genes responded to gibberellin, jasmonic acid, salicylic acid, Botrytis cinerea, Podosphaera pannosa, and Marssonina rosae (M. rosae); the RcTNL23 gene responded significantly to three hormones and three pathogens, and exhibited an upregulated expression. Furthermore, the black spot pathogen was identified as M. rosae. After inoculating rose leaves, an expression pattern analysis of the RcTNL genes suggested that they act during different periods of pathogen infection. Background: Given the crucial role of gut microbiota in animal and human health, studies on modulating the intestinal microbiome for therapeutic purposes have grasped a significant attention, of which the role of fecal microbiota transplantation (FMT) has been emphasized. Methods: In the current study, we evaluated the effect of FMT on gut functions in Escherichia coli (E. coli) infection by using mice model. Moreover, we also investigated the subsequently dependent variables of infection, i.e., body weight, mortality, intestinal histopathology, and the expression changes in tight junction proteins (TJPs). Results: The FMT effectively decreased weight loss and mortality to a certain extent with the restoration of intestinal villi that resulted in high histological scores for jejunum tissue damage (p < 0.05). The effect of FMT on alleviating the reduction of intestinal TJPs was also proved by immunohistochemistry analysis and mRNA expression levels. Moreover, the abundance of health-threatening bacteria, belonging to phylum Proteobacteria, family Enterobacteriaceae and Tannerellaceae, genus Escherichia-Shigella, Sphingomonas, Collinsella, etc., were significantly increased, whereas beneficial bacteria, belonging to phylum Firmicutes, family Lactobacillaceae, genus Lactobacillus were decreased in the gut of infected mice. Furthermore, we sought to investigate the association of clinical symptoms with FMT treatment with modulation in gut microbiota. According to beta diversity, the microbial community of gut microbiota results reflected the similarities between non-infected and FMT groups. The improvement of the intestinal microbiota in FMT group was characterized by the significant high level of beneficial microorganisms with the synergistic decrease of Escherichia-Shigella, Acinetobacter, and other taxa. Conclusion: The findings suggest a beneficial host-microbiome correlation following fecal microbiota transplanatation for controlling gut infections and pathogens-associated diseases. Endometrial cancer (EC) is a common gynecological cancer that endangers women health. Although substantial progresses of EC management have been achieved in recent years, the incidence of EC still remains high. Obesity has been a common phenomenon worldwide that increases the risk of EC. However, the mechanism associating obesity and EC has not been fully understood. Metabolic reprogramming as a remarkable characteristic of EC is currently emerging. As the primary factor of metabolic syndrome, obesity promotes insulin resistance, hyperinsulinemia and hyperglycaemia. This metabolic disorder remodels systemic status, which increases EC risk and is related with poor prognosis. Glucose metabolism in EC cells is complex and mediated by glycolysis and mitochondria to ensure energy requirement. Factors that affect glucose metabolism may have an impact on EC initiation and progression. In this study, we review the glucose metabolic reprogramming of EC not only systemic metabolism but also inherent tumor cell metabolism. In particular, the role of glucose metabolic regulation in malignant properties of EC will be focused.
