106. DeepHealth: A Self-Attention Based Method for Instant Intelligent Predictive Maintenance in Industrial Internet of Things

Abstract

With the rapid development of artificial intelligence and industrial Internet of Things (IIoT) technologies, intelligent predictive maintenance (IPdM) has received considerable attention from researchers and practitioners. To efficiently predict impending failures and mitigate unexpected downtime, while satisfying the instant maintenance demands of industrial facilities is very important for improving the production efficiency. In this article, a self-attention based “Perception and Prediction” framework, called DeepHealth, is proposed for the instant IPdM. Specifically, the framework is composed of two submodels (i.e., DH-1 and DH-2), which are respectively utilized to perform the health perception and sequence prediction. By operating the framework, the proposed models can predict the health conditions via predicting the future signal samples, thereby completing the instant IPdM. Considering the potential temporal correlation in time series, we deploy an enhanced attention mechanism to capture global dependencies from the vibration signals, and leverage the long- and short-term sequence prediction of sensor signals to support instant maintenance decision-making. On this basis, we conduct a destructive experiment based on the IIoT-enabled rotating machinery and construct a balanced industrial dataset for model evaluations. Extensive experiment results show that the proposed solution achieves good prediction accuracy for instant IPdM on the automatic washing equipment and Case Western Reserve University datasets.

Abstract

This protocol describes a method to assess relative changes in the level of global protein synthesis in the preimplantation embryo using the Click-iT (®) Plus OPP Protein Synthesis Assays. In this assay, O-propargyl-puromycin (OPP), an analog of puromycin, is efficiently incorporated into the nascent polypeptide of newly translated proteins in embryonic cells. OPP is fluorescently labeled with a photostable Alexa Fluor (TM) dye and detected with fluorescence microscopy. The intensity of the fluorescence is quantitatively analyzed. This is a fast, sensitive, and non-radioactive method for the detection of protein synthesis in early embryo development. It provides a tool for analyzing the temporal regulation of protein synthesis, as well as the effects of changes in the embryonic microenvironment, and pharmacological and genetic modulations of embryo development. Abstract

Background: Cancer metastasis is still a life threat to patients with colorectal cancer (CRC). Brain and muscle ARNT‐like protein 1 (BMAL1) is an important biological proteins that can regulate the behavior of cancer cells and their response to chemotherapy. However, the role of BMAL1 in the tumorigenic phenotype of CRC remains unclear. Here, we aim to investigate the functional role and mechanisms of BMAL1 in CRC. Methods: The mRNA expression of BMAL1 was studied using the Cancer Genome Atlas (TCGA) databases. The protein level in clinical tissues was confirmed by immunohistochemistry (IHC). The effects of BMAL1 on the epithelial‐to‐mesenchymal transition (EMT) and proliferation of CRC cell lines (including BMAL1 overexpressed or silencing cells) were studied by Transwell, wound healing, CCK‐8 and colony formation experiments. A series of experiments were conducted to demonstrate the mechanisms of BMAL1 regulating EMT and cancer proliferation in vitro and in vivo. Results: We found that BMAL1 expression was closely related to the poor prognosis of CRC. BMAL1 overexpression promoted cell proliferation and migration. Mechanistically, we found that BMAL1 may activate the epithelial‐to‐mesenchymal transition (EMT) pathway and induce the β‐catenin release further promotes the expression of oncogene c‐Myc and the migration of colorectal cells by activating MAPK pathway. However, BMAL1 silencing achieved the opposite effect. In addition, blocking MAPK‐signaling pathway with specific inhibitors of ERK1/2 and JNK can also downregulate the expressions of c‐Myc in vitro. Taken together, these results suggested that the BMAL1/ c‐Myc‐signaling pathway may regulate the metastasis of CRC through the JNK/ERK1/2 MAPK‐dependent pathway. Conclusions: Our study showed that BMAL1 promotes CRC metastasis through MAPK‐c‐Myc pathway. These results deepen our understanding of the relationship between BMAL1 and tumorigenic phenotypes, which may become a promising therapeutic target for BMAL1 overexpressing CRC. Abstract

As a key component of rotating machinery, the health perception of bearings is essential to ensure the safe and reliable operation of industrial equipment. In recent years, research on equipment health perception based on data-driven methods has received extensive attention. Overall, most studies focus on several public datasets to verify the effectiveness of their algorithms. However, the scale of these datasets cannot completely satisfy the representation learning of deep models. Therefore, this paper proposes a novel method, called CarNet, to obtain a more robust model and ensure that the model is sufficiently trained on a limited dataset. Specifically, it is composed of a data augmentation method named equitable sliding stride segmentation (ESSS) and a hybrid-stacked deep model (HSDM). The ESSS not only amplifies the scale of the original dataset but also enables newly generated data with both spatial and temporal correlations. The HSDM can, therefore, extract shallow spatial features and deep temporal information from the strongly correlated 2-dimensional (2-D) sensor array using a CNN and a bi-GRU, respectively. Moreover, the integrated attention mechanism contributes to focusing limited resources on informative areas. The effectiveness of CarNet is evaluated on the CWRU dataset, and an optimal diagnostic accuracy of 99.92% is achieved.

They were divided into subgroups based on age: ≤ 35 years and > 35 years. Differences in baseline characteristics, ovarian stimulation characteristics, endocrinological characteristics, and clinical outcome between the two groups were assessed. Statistical analyses were stratified by age.Results: In all women with DOR, PPOS+CC was associated with a lower percentage of women with profound pituitary suppression than standard PPOS (0.0% vs 18.6%, P < 0.001 and 1.3% vs 11.0%, P = 0.002). In young women with DOR, more high-quality cleavage-stage embryos were harvested (1.96 vs 1.38, P = 0.018) and a lower dosage of gonadotropin per oocyte retrieved was required (558.37 vs 909.82, P = 0.036) in PPOS+CC. In older women with DOR, PPOS+CC led to an increase in the incidence of luteinizing hormone (LH) surge levels above 10 IU/L on trigger day (12.7% vs 4.9%, P = 0.028) and a decrease in the rate of oocyte maturation (84.7% vs 89.9%, P = 0.034) compared to standard PPOS.Conclusion: Clomiphene citrate i

Abstract

With the continuous development of information technology and information management systems, they are widely and successfully used in practice. Health behavior and psychological monitoring systems are a breakthrough in the technological transformation of the therapeutic field. It is widely used in the field of healing and health care with its characteristics of convenience, quickness, accuracy, and timeliness. In this study, 466 valid samples were collected by questionnaires from students. The results show that effort expectation, community influence, and convenience have a positive effect on the health behaviors monitored by the health behavior and psychological monitoring systems of school students, and the three factors indirectly influence the behavioral intention through influencing the perceived value, learning willingness, and perceived trust and then affect the actual behavior. Based on the analysis results of this research, the authors provide research and management recommendations for health behavior and psychological monitoring systems operators and researchers in related fields.

148. Transcriptomic profiling of day 3 human embryos of poor quality reveals molecular links to divergent developmental trajectories

Abstract

Poor-quality (PQ) embryos are commonly formed in assisted reproduction and often discarded, but emerging evidence suggests that some can develop into healthy children. Here, we performed single-embryo transcriptome profiling of day 3 human embryos of both good quality (GQ) and PQ. Our analyses revealed that day 3 PQ embryos can be categorized into two distinct subgroups: genuine PQ (gPQ) and morphological PQ (mPQ) embryos. This was further supported by additional transcriptomic data of GQ and PQ embryos donated by the same individuals. gPQ embryos showed greater impairments in both RNA decay and zygotic genome activation (ZGA) compared to mPQ embryos. Blastocysts that emerged from PQ embryos, more likely from mPQ embryos, demonstrated transcriptomic similarity to normal blastocysts. We also identified genes highly upregulated in day 3 gPQ embryos. These molecular insights may improve embryo selection and offer valuable clues for enhancing embryo quality in the future.

149. Study on vertical permeability regularity and collapsibility of a large thickness loess foundation by in-situ testing

Abstract

In many regions in China there is a wide distribution of dead-weight collapsible loess that experience frequent disasters resulting in enormous losses to national property. Loess collapsibility is the leading cause of disasters in loess areas. Many international scholars are working collaboratively to avoid property losses and human casualties caused by the collapsible loess. Loess collapsibility is closely related to the vertical permeability regularity, so the vertical permeability regularity should be the first critical factor taken into account in studying loess collapsibility. In Lanzhou city, the in-situ soaking test is carried out on an unsaturated natural loess foundation with a thickness of 36.5m, the test pit diameter is 40m, TDR moisture probes are embedded, and the test lasts 282 days. During the test, the vertical migration of water is observed and the temporal and spatial variations of volumetric water content are considered. In the detailed research of the vertical permeability regularity, the relation between permeability regularity and collapsibility are discussed. For shallow layer soil, soil volumetric water content reaches saturation at wetting front, the water flows downward due to gravity and the infiltration rate is larger. For deep layer soil, the moisture will increase because of the upper soil permeability and its own suction, but, due to soil compression caused by the occurrence of the upper soil collapsibility, the infiltration rate is significantly reduced. The temporal and spatial variations of water volumetric content can be used to determine whether a loess collapsible deformation has occurred. The limit permeability depth in this test is 25m, and the limit permeability depth can also be served as the collapsibility evaluation depth of the unsaturated natural Malan loess foundation.
