116. T Cells infiltrating diseased liver express ligands for the NKG2D stress surveillance system

Abstract

NK cells, which are highly enriched in the liver, are potent regulators of antiviral T cells and immunopathology in persistent viral infection. We investigated the role of the NKG2D axis in T cell/NK cell interactions in hepatitis B. Activated and hepatitis B virus (HBV)-specific T cells, particularly the CD4 fraction, expressed NKG2D ligands (NKG2DL), which were not found on T cells from healthy controls (p < 0.001). NKG2DL-expressing T cells were strikingly enriched within HBV-infected livers compared with the periphery or to healthy livers (p < 0.001). NKG2D+NK cells were also increased and preferentially activated in the HBV-infected liver (p < 0.001), in direct proportion to the percentage of MICA/B-expressing CD4 T cells colocated within freshly isolated liver tissue (p < 0.001). This suggests that NKG2DL induced on T cells within a diseased organ can calibrate NKG2D-dependent activation of local NK cells; furthermore, NKG2D blockade could rescue HBV-specific and MICA/B-expressing T cells from HBV-infected livers. To our knowledge, this is the first ex vivo demonstration that non-virally infected human T cells can express NKG2DL, with implications for stress surveillance by the large number of NKG2D-expressing NK cells sequestered in the liver.

Abstract

The anisotropic transport properties of K0.8Fe1.65Se2 crystal have been investigated with the angle θ (between the magnetic field H and c-axis) varied from 0° to 180°. It is found that the angle dependencies of the mixed state resistivities near Tc0 show a small anomalous peak on the resistive valley for H//ab, probably due to the kinked vortex structure with a lower pinning energy in the phase-separated crystal. In the normal state, the resistivities increase with increasing angles and take a maximum at H//ab, which may be related to the larger resistivity when carriers move along the c-axis driven by Lorentz force. Abstract

Background: Renal cell carcinoma forming a venous tumor thrombus (VTT) in the inferior vena cava (IVC) has a poor prognosis. Recent investigations have been focused on prognostic markers of survival. Thrombus consistency (TC) has been proposed to be of significant value but yet there are conflicting data. The aim of this study is to test the effect of IVC VTT consistency on cancer specific survival (CSS) in a multi-institutional cohort. Methods: The records of 413 patients collected by the International Renal Cell Carcinoma–Venous Thrombus Consortium were retrospectively analyzed. All patients underwent radical nephrectomy and tumor thrombectomy. Kaplan–Meier estimate and Cox regression analyses investigated the impact of TC on CSS in addition to established clinicopathological predictors. Results: VTT was solid in 225 patients and friable in 188 patients. Median CSS was 50 months in solid and 45 months in friable VTT. TC showed no significant association with metastatic spread, pT stage, perinephric fat invasion, and higher Fuhrman grade. Survival analysis and Cox regression rejected TC as prognostic marker for CSS. Conclusions: In the largest cohort published so far, TC seems not to be independently associated with survival in RCC patients and should therefore not be included in risk stratification models. J. Abstract

32nd European Photovoltaic Solar Energy Conference and Exhibition; 1226-1228, The effect of Ag incorporation into CZTSSe kesterite solar cell by a non-vacuum environmentalfriendly process has been studied. We first demonstrated the fabrication of (Ag, Cu)2ZnSn(S,Se)4 kesterite solar cells using chemical spray pyrolysis. Ag content in the ACZTSSe thin film can be controlled by precursor solution concentration. For kesterite with high Ag content, aggregation of grain and formation of second phases occurred at elevated temperature, but can be solved when being annealed at relatively low temperature. Enhancement of grain size at lower process temperature is fulfilled by Ag incorporation. With 20% Ag addition, efficiency of the kesterite increased from 5.3% to 6.8% with the improvement of VOC from 388 mV to 444 mV. Bandgap of ACZTSSe can be controlled from 1.06 to 1.2 eV with Ag content varied from 0~50% and the tailing energy was reduced by Ag.

Abstract

Multifunctional theranostic platforms, especially single component-based platforms, enable both cancer treatment and real-time imaging as well as enhance the efficiency of treatment. In this study, 50 nm Mo2C nanospheres were explored as a “one-for-all” theranostic agent. The light-harvesting of Mo2C covered the entire near infrared region, and NIR irradiation concurrently triggered hyperthermia and reactive oxygen species (ROS) production; thus, synergistic outcomes of photothermal and photodynamic therapy could be realized. Both in vitroand in vivoexperiments have confirmed the superiority of the synergistic phototherapy in killing cancer cells and removing solid tumors; moreover, Mo2C proposed herein has been proven to be applicable as a photoacoustic imaging and CT imaging contrast agent for in vivotumor depiction; furthermore, Mo2C demonstrates excellent biocompatibility, showing minimal hematotoxicity and tissue toxicity. A theoretical simulation performed by density functional theory revealed that the metallic character and the interband/intraband transition of Mo2C accounted for its broad photoabsorption. The antitumor mechanism of Mo2C was investigated on a solid tumor by B-mode ultrasonography (US) and magnetic resonance imaging (MRI), revealing a typical liquefactive necrosis process; hence, herein, the dual-imaging guided phototherapy was efficiently mediated by Mo2C.
