Further, genetically predicted higher level of MCP1 was significantly associated with higher risk of CAD (OR for 1-SD increase: 1.05, 95% CIs: 1.02– 1.08, P value: 0.002) in MR analysis. Sensitivity analyses were also conducted to validate the main findings, and we also did not detect any directional pleiotropy effects using the MR Egger intercept test (P=0.831).Conclusion: To sum up, our study suggested that increased CAD risk was associated with a predisposition to higher level of MCP1. Additional insight into the contribution of MCP1 to the occurrence of CAD is still needed.

Abstract

Objective Real-time reverse transcription-polymerase chain reaction is the gold standard for the diagnosis of COVID-19, but it is necessary to utilize other tests to determine the burden of the disease and the spread of the outbreak such as IgG-, IgM-, and IgA-based antibody detection using enzyme-linked immunosorbent assay (ELISA). Materials and Methods We developed an indirect ELISA assay to quantitatively measure the amount of COVID-19 IgG, IgM, and IgA antibodies present in patient serum, dried blood, and plasma. Results The population cutoff values for positivity were determined by receiver operating characteristic curves to be 1.23 U/mL, 23.09 U/mL, and 6.36 U/mL for IgG, IgM, and IgA, respectively. After albumin subtraction, the specificity remained >98% and the sensitivity was 95.72%, 83.47%, and 82.60%, respectively, for IgG, IgM, and IgA antibodies to the combined spike subunit 1 receptor binding domain and N proteins in serum. Plasma and dried blood spot specimens were also validated on this assay. Conclusion This assay may be used for determining the seroprevalence of SARS-CoV-2 in a population exposed to the virus or in vaccinated individuals. [ABSTRACT FROM AUTHOR]

Here, we explored novel targets for use in future treatments. Previous studies showed the sialic acid-binding Ig-like lectin (Siglec) family to have a specific role in immunosuppression. We aimed to study the characteristics and immune function of Siglec family members.Methods: Transcriptome data from 1024 glioma samples and 1551 glioma single cells were used in our study. Clinical and molecular pathology information was also included. Statistical, bioinformatical methods, and single-cell sequencing analysis were applied to investigate the role of Siglec family members.Results: Siglecs-5, −7, −9, and −16 showed a significant correlation with immunosuppression in glioma. They are typically expressed in higher grade, IDH-wildtype, and mesenchymal subtype gliomas. Siglec-5, −7, and −9 had a similar immune function to TIM-3, while Siglec-16 was similar to PD-L1, suppressing tumor immunity via different mechanisms. Joint use of Siglec-inhibitors and immune checkpoint inhibitors could prolong the survival of glioma patients.Conclusion: Siglec-5, −7, −9, and −16 suppressed tumor immunity in different ways. 114. In situ measurement of [Cl.sup.-] concentrations and pH at the reinforcing steel/concrete interface by combination sensors

Abstract

This paper presents an in situ, nondestructive method of monitoring [Cl.sup.-] concentrations and pH values at the steel/ concrete interface. The Ag/AgCl electrodes prepared by the electrochemical anodization and the Ir/Ir[O.sub.2] electrodes prepared by thermal oxidation in carbonate served as [Cl.sup.-] concentration and pH sensors, respectively. The potentiometric response of the Ag/AgCl electrode to the logarithm of [Cl.sup.-] concentrations ranging from 1 x [10.sup.-4] to 2 M in saturated Ca[(OH).sub.2] solution simulating the inner electrolytic medium of concrete shows good linearity. The Ir/Ir[O.sub.2] electrode also exhibits an ideal Nernstian response in the range of pH 1-14. The Ag/AgCl and Ir/ Ir[O.sub.2] electrodes were combined into a multiplex [Cl.sup.-]/pH sensor, and the sensor was embedded in concrete close to the steel/concrete interface to realize an in situ and long-term measurement of [Cl.sup.-] concentrations and pH values. The results indicate that the combined sensor is robust and sensitive enough to in situ measure [Cl.sup.-] concentrations and pH quantitatively at the steel/concrete interface, which is of indispensable importance to the study of corrosion and protection of the steel in concrete.

135. Developing a periodontal disease antibody array for the prediction of severe periodontal disease using machine learning classifiers.

Abstract

Background: The aim of this study was to simultaneously and quantitatively assess the expression levels of 20 periodontal disease-related proteins in gingival crevicular fluid (GCF) from normal controls (NOR) and severe periodontitis (SP) patients with an antibody array.Methods: Antibodies against 20 periodontal disease-related proteins were spotted onto a glass slide to create a periodontal disease antibody array (PDD). The array was then incubated with GCF samples collected from 25 NOR and 25 SP patients. Differentially expressed proteins between NOR and SP patients were then used to build receiver operator characteristic (ROC) curves and compare five classification models, including support vector machine, random forest, k nearest neighbor, linear discriminant analysis, and Classification and Regression Trees.Results: Seven proteins (C-reactive protein, interleukin [IL]-1α, interleukin-1β, interleukin-8, matrix metalloproteinase-13, osteoprotegerin, and osteoactivin) were significantly upregulated in SP patients compared with NOR, while receptor activator of nuclear factor-kappa was significantly downregulated. The highest diagnostic accuracy using a ROC curve was observed for IL-1β with an area under the curve of 0.984. Five of the proteins (IL-1β, IL-8, MMP-13, osteoprotegerin, and osteoactivin) were identified as important features for classification. Linear discriminant analysis had the highest classification accuracy across the five classification models that were tested.Conclusion: This study highlights the potential of antibody arrays to diagnose periodontal disease.
