Abstract

Using resting-state functional magnetic resonance imaging, we test the hypothesis that subjects with post-traumatic stress disorder (PTSD) are characterized by reduced temporal variability of brain connectivity compared to matched healthy controls. Specifically, we test whether PTSD is characterized by elevated static connectivity, coupled with decreased temporal variability of those connections, with the latter providing greater sensitivity toward the pathology than the former. Static functional connectivity (FC; nondirectional zero-lag correlation) and static effective connectivity (EC; directional time-lagged relationships) were obtained over the entire brain using conventional models. Dynamic FC and dynamic EC were estimated by letting the conventional models to vary as a function of time. Statistical separation and discriminability of these metrics between the groups and their ability to accurately predict the diagnostic label of a novel subject were ascertained using separate support vector machine classifiers. Our findings support our hypothesis that PTSD subjects have stronger static connectivity, but reduced temporal variability of connectivity. Further, machine learning classification accuracy obtained with dynamic FC and dynamic EC was significantly higher than that obtained with static FC and static EC, respectively. Furthermore, results also indicate that the ease with which brain regions engage or disengage with other regions may be more sensitive to underlying pathology than the strength with which they are engaged. Abstract

Parkinson's disease (PD) is a heterogeneous neurodegenerative disorder with variable clinicopathologic phenotypes and underlying neuropathologic mechanisms. Each clinical phenotype has a unique set of motor symptoms. Tremor is the most frequent initial motor symptom of PD and is the most difficult symptom to treat. The dentate nucleus (DN) is a deep iron-rich nucleus in the cerebellum and may be involved in PD tremor. In this study, we test the hypothesis that DN iron may be elevated in tremor-dominant PD patients using quantitative susceptibility mapping. Forty-three patients with PD [19 tremor dominant (TD)/24 akinetic rigidity (AR) dominant] and 48 healthy gender- and age-matched controls were recruited. Multi-echo gradient echo data were collected for each subject on a 3.0-T MR system. Inter-group susceptibility differences in the bilateral DN were investigated and correlations of clinical features with susceptibility were also examined. In contrast with the AR-dominant group, the TD group was found to have increased susceptibility in the bilateral DN when compared with healthy controls. In addition, susceptibility was positively correlated with tremor score in drug-naive PD patients. These findings indicate that iron load within the DN may make an important contribution to motor phenotypes in PD. Moreover, our results suggest that TD and AR-dominant phenotypes of PD can be differentiated on the basis of the susceptibility of the DN, at least at the group level. 117. High-order numerical method and error analysis based on a mixed scheme for fourth-order problem in a ball.

Abstract

It is widely recognized that numerical computation in high-dimensional problems poses a significant challenge, particularly for intricate surface geometries, such as spherical and cylindrical domains. Therefore, in this paper, we present a high-precision numerical method based on a mixed scheme for solving fourth-order problems in a ball. The core concept behind this algorithm is to convert the original problem into a second-order coupled system by introducing an auxiliary Laplace equation. Subsequently, the second-order coupled system is disassembled into a sequence of one-dimensional decoupled second-order problems through spherical harmonic function expansion and variable separation. Building on this foundation, we formulated their variational formulations and discretizations, and demonstrated the uniqueness of weak and approximate solutions, as well as provided an error estimate between them. Furthermore, we have extended the algorithm to accommodate general variable coefficients. Abstract

Unreliable feature extraction and matching in handcrafted features undermine the performance of visual SLAM in complex real-world scenarios. While learned local features, leveraging CNNs, demonstrate proficiency in capturing high-level information and excel in matching benchmarks, they encounter challenges in continuous motion scenes, resulting in poor generalization and impacting loop detection accuracy. To address these issues, we present DK-SLAM, a monocular visual SLAM system with adaptive deep local features. MAML optimizes the training of these features, and we introduce a coarse-to-fine feature tracking approach. Initially, a direct method approximates the relative pose between consecutive frames, followed by a feature matching method for refined pose estimation. To counter cumulative positioning errors, a novel online learning binary feature-based online loop closure module identifies loop nodes within a sequence. Abstract

The presence of neurofibrillary tangles containing hyper‐phosphorylated tau is a characteristic of Alzheimer's disease (AD) pathology. The positron emission tomography (PET) radioligand sensitive to tau neurofibrillary tangles (18F‐AV1451) also binds with iron. This off‐target binding effect may be enhanced in older adults on the AD spectrum, particularly those with amyloid‐positive biomarkers. Here, we examined group differences in 18F‐AV1451 PET after controlling for iron‐sensitive measures from magnetic resonance imaging (MRI) and its relationships to tissue microstructure and cognition in 40 amyloid beta positive (Aβ+) individuals, 20 amyloid beta negative (Aβ‐) with MCI and 31 Aβ‐ control participants. After controlling for iron, increased 18F‐AV1451 PET uptake was found in the temporal lobe and hippocampus of Aβ+ participants compared to Aβ‐ MCI and control participants. Within the Aβ+ group, significant correlations were seen between 18F‐AV1451 PET uptake and tissue microstructure and these correlations remained significant after controlling for iron. These findings indicate that off‐target binding of iron to the 18F‐AV1451 ligand may not affect its sensitivity to Aβ status or cognition in early‐stage AD. [ABSTRACT FROM AUTHOR]

139. Mitofusin2 Promotes β Cell Maturation from Mouse Embryonic Stem Cells via Sirt3/Idh2 Activation

Abstract

β cell dysfunction is the leading cause of diabetes. Adult β cells have matured glucose-stimulated insulin secretion (GSIS), whereas fetal and neonatal β cells are insensitive to glucose and are functionally immature. However, how β cells mature and acquire robust GSIS is not fully understood. Here, we explored the potential regulatory proteins of β cell maturation process and the capacity for GSIS. Combined with the data from public databases, we found that the gene expression of Mitofusin2 (Mfn2) showed an increasing trend from mouse neonatal β cells to mature β cells. Moreover, its protein expression increased during mouse embryonic pancreas development and β cell differentiation from mouse embryonic stem cells. Knocking down Mfn2 reduced Urocortin3 (Ucn3) expression, GSIS, and ATP production in induced β cells, while overexpressing it had the opposite effect. However, neither Mfn2 knockdown nor overexpression affected the differentiation rate of insulin-positive cells. In immature and mature β cells, Mfn2 and its correlated genes were enriched in tricarboxylic acid (TCA) cycle-related pathways. The expressions of Sirtuin 3 (Sirt3) and isocitrate dehydrogenase 2 (NADP+) and mitochondrial (Idh2) were Mfn2-regulated during β cell differentiation. Inhibiting Idh2 or Sirt3 reduced cellular ATP content and insulin secretion levels that increased by Mfn2 overexpression. Thus, Mfn2 modulated the induced β cell GSIS by influencing the TCA cycle through Sirt3/Idh2 activation. We demonstrated that Mfn2 promoted embryonic stem cell-derived β cell maturation via the Sirt3/Idh2 pathway, providing new insights into β cell development. Our data contribute to understanding diabetes pathogenesis and offer potential new targets for β cell regeneration therapies.
