Abstract

We constructed a sensor for the determination of Fe2+ and/or Fe3+ ions that consists of a polyaniline layer as an ion-to-electron transducer; on top of it, chelating molecules are deposited (which can selectively chelate specific ions) and protected with a non-biofouling poly(2-methyl-2-oxazoline)s layer. We have shown that our potentiometric sensing layers show a rapid response to the presence of Fe2+ or Fe3+ ions, do not experience interference with other ions (such as Cu2+), and work in a biological environment in the presence of bovine serum albumin (as a model serum protein). These NPs may selectively release their cargo, in this case paclitaxel (PTX), at the high levels of ROS present in the intracellular environment of inflamed tissues and most tumors.Purpose: The main objective was to determine anti-tumor efficacy of PTX-loaded ROS-sensitive NPs and to examine whether macrophage infiltration had any impact on treatment efficacy.Methods: NPs were synthesized and their characteristics in the presence of H2O2 were demonstrated. Both confocal microscopy as well as flow cytometry approaches were used to determine degradation of ROS-sensitive NPs. HeLa cells were cultured in vitro and used to establish tumor xenografts in nude mice. In vivo experiments were performed to understand toxicity, biodistribution and anti-tumor efficacy of the NPs. Moreover, we performed immunohistochemistry on tumor sections to study infiltration of M1 and M2 subsets of macrophages.Results: We demonstrated that PTX delivered in NPs containing a ROS-sensitive polymer exhibits a better anti-tumor efficacy than PTX in NPs containing ROS-non-sensitive polymer, free PTX or Abraxane® (nab-PTX). The biodistribution revealed that ROS-sensitive NPs exhibit retention in liver, spleen and lungs, suggesting a potential to target cancer metastasizing to these organs. Finally, we demonstrated a correlation between infiltrated macrophage subsets and treatment efficacy, possibly contributing to the efficient anti-tumor effects.Conclusion: Treatment with ROS-sensitive NPs containing PTX gave an improved therapeutic effect in HeLa xenografts than their counterpart, free PTX or nab-PTX. Abstract

The diploma thesis analyses the impact of online behaviour on a real life. It is divided into a theoretical (chapters 1 to 3) and practical part (chapters 4 to 7). The first chapter introduces the current trends and generally the topic of social networks, the internet, online communication and relationships. The second chapter describes main forms of cyber antisocial behaviour. The third chapter is focused on the process of socialization and growing up from the point of view of developmental psychology. It also shows individual mental and behavioural frames of the cyber reality which penetrates the real world. The practical part deals in four chapters with different forms of a research which aim was to verify outlined hypotheses and facts mentioned in the theoretical part. The selected methods are quantitative research among high school students distributed through questionnaires, the qualitative interviews among high school teachers and the quantitative analysis of the educational journal. The fourth chapter describes theoretically the methodology of the research. The last three chapters document each part of the research, partially verify the outlined hypothesis and interpret the results.

136. Biological characterization of a novel hybrid copolymer carrier system based on glycogen.

Abstract

The effective drug delivery systems for cancer treatment are currently on high demand. In this paper, biological behavior of the novel hybrid copolymers based on polysaccharide glycogen were characterized. The copolymers were modified by fluorescent dyes for flow cytometry, confocal microscopy, and in vivo fluorescence imaging. Moreover, the effect of oxazoline grafts on degradation rate was examined. Intracellular localization, cytotoxicity, and internalization route of the modified copolymers were examined on HepG2 cell line. Biodistribution of copolymers was addressed by in vivo fluorescence imaging in C57BL/6 mice. Our results indicate biocompatibility, biodegradability, and non-toxicity of the glycogen-based hybrid copolymers. Copolymers were endocyted into the cytoplasm, most probably via caveolae-mediated endocytosis. Higher content of oxazoline in polymers slowed down cellular uptake. No strong colocalization of the glycogen-based probe with lysosomes was observed; thus, it seems that the modified externally administered glycogen is degraded in the same way as an endogenous glycogen. In vivo experiment showed relatively fast biodistribution and biodegradation. In conclusion, this novel nanoprobe offers unique chemical and biological attributes for its use as a novel drug delivery system that might serve as an efficient carrier for cancer therapeutics with multimodal imaging properties. Abstract

The influence of hydrophobicity on formation, stability, and size of pH-responsive methacryloylated oligopeptide-based polymer nanoparticles has been studied by dynamic light scattering (DLS), transmission electron microscopy (Cryo-TEM), and NMR. Different polyanions/surfactant systems have been studied at constant polymer concentration and within a broad range of surfactant concentrations. The two newly synthesized pH-sensitive hydrophobic polyanions, poly(N(ω)-methacryloyl glycyl-L-leucine) and poly(N(ω)-methacryloyl glycyl-L-phenylalanyl-L-leucinyl-glycine), and three nonionic surfactants (Brij97, Brij98, and Brij700) have been investigated. The surfactants were different in the length of hydrophilic poly(ethylene oxide) (PEO) chain. In surfactant-free solution at basic pH, the polyanions form hydrophobic domains. In the presence of a surfactant, our results prove the complex formation at high pH between the nonionic surfactant and the polyelectrolyte; a pearl-necklace structure is formed. At low pH below critical pH (pH(tr)), reversible nanoscale structures occur in solutions for all systems. The detailed mechanism of the formation of pH-sensitive nanoparticles from polymer-surfactant complex with varying pH is established. Our results suggest that the polymer hydrophobicity is of primary importance in pretransitional behavior of the complex. Once preliminary nanoparticle nuclei are formed, the hydrophobicity of the polymer plays a minor role on further behavior of formed nanostructures. The subsequent transformation of nanoparticles is determined by the surfactant hydrophilicity, the length of hydrophilic tail that prevents further aggregation due to steric repulsions.
