Abstract

The ethyl acetate-soluble fraction from a MeOH extract of the roots of Ferula variagave six new sesquiterpene lactones ( 1– 6) and five known sesquiterpenes ( 7– 11). Their structures were established on the basis of spectroscopic evidence. The cytotoxic activities of 1– 11were evaluated against selected human cancer cell lines. Compound 4showed significant selective cytotoxicity against multidrug-resistant cancer cells (KB-C2). The cytotoxicities of compounds 1, 3, 5, 8, and 11against KB-C2 cells were enhanced in the presence of nontoxic concentrations of colchicine.

Abstract

ABSTRACTA novel potent trypanocidal diterpene, komaroviquinone, was reduced by Trypanosoma cruziold yellow enzyme (TcOYE) to its semiquinone radical. The reductase activity in trypanosome lysates was completely immunoabsorbed by anti-TcOYE antibody. Since TcOYE is expressed throughout the T. cruzilife cycle, komaroviquinone is an interesting candidate for developing new antichagasic drugs.

Abstract

AbstractA suspension cell culture was established from Aquilaria sinensis, a timber species producing fragrant woody material called “Agarwood.” Methyl jasmonate was added to this culture in order to induce production of fragrant compounds. Cells were harvested after seven days of incubation and subjected to solid phase micro-extraction to produce an extract that was analyzed by GC/MS. Three peaks appeared on GC and were identifi ed as α-guaiene, γ-guaiene (= 1(10),11-guaiadiene), and α-humulene based on their mass fragmentation patterns and 13C-NMR data. The production of the guaiene derivatives and α-humulene showed different kinetics, which may mean they are the products of different induced biosynthetic enzymes.

Abstract

A considerable proportion of crude drugs is used for dermatological purposes and infectious diseases in thailand. The authors' primary concern was to examine their medicinal activity and to isolate active principles, if any, from them. Methanol extracts of 24 drugs were examined for antimicrobial activities against Escherichia coli, Staphyl-ococcus aureus, Candida albicans and Trichophyton mentagrophytes. Abstract

AbstractA new type of oil from Perilla frutescens, the piperitenone type, was discovered in Thailand. GC/MS analysis of steam-distilled oil showed that it contained piperitenone (36.3%) and limonene (23.7%). Crossing experiments between the piperitenone-type and the principal types of perilla suggested the presence of one dominant gene regarding the regulation of oil types between the piperitenone-type and the perillaldehyde-type.

148. Effect ofPerilla frutescenson nitric oxide production and DNA synthesis in cultured murine vascular smooth muscle cells

Abstract

The effects of perilla (Perilla frutescens,Labiatae) on murine cultured vascular smooth muscle cells (VSMC) were investigated. The water extract of perilla leaves induced nitric oxide (NO) production of VSMC and this effect was synergistically augmented when combined with interferon (IFN)‐γ or tumour necrosis factor (TNF)‐α, while the perilla extract significantly inhibited NO production induced by IFN‐γ combined with lipopolysaccharide (LPS). Northern blot analysis revealed that these effects of the perilla extract paralleled mRNA expression of inducible nitric oxide synthase. However, the perilla extract significantly inhibited platelet derived growth factor (PDGF) or TNF‐α‐induced VSMC proliferation measured as DNA synthesis. The inhibitory effect of the perilla extract on TNF‐α‐induced VSMC proliferation was significantly suppressed by NG‐monomethyl‐L‐arginine, a non‐specific nitric synthase inhibitor, suggesting that this effect was partially mediated by NO production as an autocrine/paracrine factor. The present findings suggest that perilla would be useful for the prevention of vascular diseases such as arteriosclerosis. Copyright © 2002 John Wiley & Sons, Ltd.

Abstract

Aerial parts of Daphne oleoidesSchreber ssp. oleoides(Thymelaeaceae) are used to treat rheumatoid arthritis and lumbago in Turkish folk medicine. In order to evaluate folkloric utilization, in vitro inhibitory effects of the ethyl acetate extract and fractions obtained from this extract on interleukin 1 (IL-1α, IL-1β) and tumour necrosis factor (TNF-α) biosynthesis were studied. Through chemical isolation techniques and activity-guided fractionation process, seventeen compounds were isolated and their structures were elucidated (numbered 1–17). Diterpenoids genkwadaphnin (3) and 1,2-dehydrodaphnetoxin (6) and a coumarin derivative daphnetin (9) showed potent inhibitory activity and were found to be the main active ingredients. Furthermore, gnidilatin (4), gnidilatin-20 palmitate (5), genkwadaphnin-20-palmitate (7) and gnidicin-20-palmitate (8), having diterpenoid structure, and eudesmine (12), wikstromol (13) and matairesinol (14), having lignan structure, were determined to possess moderate inhibitory activity and may have a contributory role in the effect of the remedy.

Abstract

AbstractThe essential oil of Perilla setoyensisG. Honda, a newly discovered wild perilla in Japan, was examined by GC and GC/MS. The principal volatile constituent of the leaves was revealed to be a novel monoterpene, shisofuran, which was fully characterized by spectroscopic means. Nonetheless, shisofuran (18.3–196%) was labile and decomposed during steam distillation, leaving 1-octen-3-ol (16.2–21.8%) and β-caryophyllene (18.3–19.6%) as the major constituents of the oil.
