Abstract

The aminoglycoside phosphotransferase (APH) gene of B. circulans has been cloned in E. coli (CC1105) using the vector pBR322 and has been found to confer antibiotic resistance. The 2.7 kb Sal I fragment containing the APH gene was ligated into the plasmid SLP1.2 and transformed into S. lividans 66 (CL1023), where it was also able to confer antibiotic resistance. When the phosphotransferase activity was assayed in crude extracts of B. circulans, E. coli CC1105 and S. lividans CL1023, using a variety of aminoglycosides as phosphate acceptors, a similar spectrum of activity was found in each strain. Using the methods of Sanger and Messing, the sequence of the 2.7 kb Sal I fragment was determined. An examination of the DNA sequence allowed the protein sequence to be deduced. When the deduced protein sequence was compared with those from S. fradiae, TN5 and TN903, significant homology was found, indicating that all the phosphotransferases may have a common origin. An examination of the sequence either side of the coding region allowed the identification of possible promotors, a ribosome binding site and a terminator. One of the promotors was similar to the σ³⁷ promotors of B. subtilis, showing that the B. circulans APH gene may be under developmental control.

109. Design and development of an adaptive external bone fracture fixation system

Abstract

External fixation is a surgical treatment primarily utilised for long bone fracture stabilisation. External fixation, through either pin or wire insertion, is done by constraining bone fragments and providing support to the injury via external scaffolding built across the fracture., but it can also be used to solve both traumatic and congenital bony deformities. Certain devices, particularly the Ilizarov Ring Fixator, can employ external bone fixation to create a biomechanical environment to gradually correct deformities (comprised of: translation, rotation and angulation). A typical application for deformity correction is the fixation of lower leg fractures, particularly tibial fractures, which have been recognised as the most common incident in long bone fractures. External ring fixators have become more developed; manufactured from sophisticated materials; or designed to incorporate computational support, to achieve accurate correction, however these factors have created limitations regarding their accessibility, complexity and ease in application. In addition, standard systems are not as versatile or correctively exact as required to prove their cost of use, creating reluctance as well as added bias towards the more developed devices. Threedimensional and multi-planar deformity correction has become major factors for current devices, yet the feasibility to use such expensive and complex devices may not be beneficial for all parties. External Fixation Systems are considered operationally expensive. Standard systems still utilise expensive and cumbersome setups, while developed devices require computational consultation and extensive training. With such complex procedural actions required to facilitate multi-planar correction, most devices utilise computational support, which in turn minimizes the clinician’s control. The current study aims to design a light-weight Adaptive External Bone Fracture Fixation System that can offer definite treatment and full clinical control over the injury. The system is to be able to stabilise and offer correction of planar bony deformities via controlled shape change. The functional verification of the device was limited to (according to the scope) stress testing. The proposed device consists of hinge systems capable of allowing for full assembly expansion to permit quick installation for various injury structures or states. In addition, the design possesses longitudinal elements that can offer both rapid and finite lengthening (with lock-and-switch) to offer both rapid and gradual system shape change, improving the control over the injury fixation. The device stress testing had revealed limited capabilities in providing enough scaffolding stability for a certain directional stress condition. To determine the quality of its structural integrity, the device was loaded under direct compressive and tensile load. The strain generated was measured and analysed using a Load-Deformation Curve. The device could support tension close to [3.5 kN], equivalent to standard models, whilst unable to support compression for loads close to [1.2 kN]. The conclusive points that were made had detailed that it was limited by its structural integrity, however the design was evaluated as functionally versatile as and should be further developed. Future recommendations proposed include the addition of constrained joints; improved locking capabilities; implementation of failure modes for hinges and lastly improved structural integrity by using sophisticated materials to further validate the skeletal structure of the fixation system.

Abstract

We have developed a rapid and simple methodology to locate yeast genes within cloned inserts, obtain partial sequence information, and construct chromosomal disruptions of these genes. This methodology has been used to study a nuclear gene from the yeast S. douglasii (a close relative of S. cerevisiae), which is essential for the excision of the mitochondrial intron aI1 of S. douglasii (the first intron in the gene encoding subunit I of cytochrome oxidase), an intron which is not present in the mitochondrial genome of S. cerevisiae. We have shown that this gene is the homologue of the S. cerevisiae MRS1 gene, which is essential for the excision of the mitochondrial introns bI3 and aI5 beta of S. cerevisiae, but is unable to assure the excision of the intron aI1 from the coxI gene of S. douglasii. The two genes are very similar, with only 13% nucleotide substitutions in the coding region, transitions being 2.5 times more frequent than transvertions. At the protein level there are 86% identical residues and 7% conservative substitutions. The divergence of the MRS1 genes of S. cerevisiae and S. douglasii, and the concomitant changes in the structure of their mitochondrial genomes is an interesting example of the co-evolution of nuclear and mitochondrial genomes.

Abstract

Background: Preclinical data suggest some cannabinoids may exert antitumour effects against glioblastoma (GBM). Safety and preliminary efficacy of nabiximols oromucosal cannabinoid spray plus dose-intense temozolomide (DIT) was evaluated in patients with first recurrence of GBM.Methods: Part 1 was open-label and Part 2 was randomised, double-blind, and placebo-controlled. Both required individualised dose escalation. Patients received nabiximols (Part 1, n = 6; Part 2, n = 12) or placebo (Part 2 only, n = 9); maximum of 12 sprays/day with DIT for up to 12 months. Safety, efficacy, and temozolomide (TMZ) pharmacokinetics (PK) were monitored.Results: The most common treatment-emergent adverse events (TEAEs; both parts) were vomiting, dizziness, fatigue, nausea and headache. Most patients experienced TEAEs that were grade 2 or 3 (CTCAE). In Part 2, 33% of both nabiximols- and placebo-treated patients were progression-free at 6 months. Survival at 1 year was 83% for nabiximols- and 44% for placebo-treated patients (p = 0.042), although two patients died within the first 40 days of enrolment in the placebo arm. There were no apparent effects of nabiximols on TMZ PK.Conclusions: With personalised dosing, nabiximols had acceptable safety and tolerability with no drug-drug interaction identified. 135. Unit bar architecture in a highly‐variable fluvial discharge regime: Examples from the Burdekin River, Australia.

Abstract

Unit bars are relatively large bedforms that develop in rivers over a wide range of climatic regimes. Unit bars formed within the highly‐variable discharge Burdekin River in Queensland, Australia, were examined over three field campaigns between 2015 and 2017. These bars had complex internal structures, dominated by co‐sets of cross‐stratified and planar‐stratified sets. The cross‐stratified sets tended to down‐climb. The development of complex internal structures was primarily a result of three processes: (i) superimposed bedforms reworking the unit bar avalanche face; (ii) variable discharge triggering reactivation surfaces; and (iii) changes in bar growth direction induced by stage change. Internal structures varied along the length and across the width of unit bars. For the former, down‐climbing cross‐stratified sets tended to pass into single planar cross‐stratified deposits at the downstream end of emergent bars; such variation related to changes in fluvial conditions whilst bars were active. A hierarchy of six categories of fluvial unsteadiness is proposed, with these discussed in relation to their effects on unit bar (and dune) internal structure. Across‐deposit variation was caused by changes in superimposed bedform and bar character along bar crests; such changes related to the three‐dimensionality of the channel and bar geometry when bars were active. Variation in internal structure is likely to be more pronounced in unit bar deposits than in smaller bedform (for example, dune) deposits formed in the same river. This is because smaller bedforms are more easily washed out or modified by changing discharge conditions and their smaller dimensions restrict the variation in flow conditions that occur over their width. In regimes where unit bar deposits are well‐preserved, their architectural variability is a potential aid to their identification. 144. Waiting for Homeownership: Assessing the Future of Homeownership

Abstract

The decade-long decline in the homeownership rate in the United States has generated substantial discussion over its future path. In the face of continued uncertainty, this Article seeks to assess what we know and do not know about the sources of the decline and the likely trajectory of the homeownership rate in coming years. The analyses use the Annual Social and Economic Supplement (ASEC) of the Current Population Survey for 1985 to 2015 to examine the determinants of changes in the homeownership rate, using shift-share analyses to measure the extent to which changing demographics explain the observed changes. The results show that demographic trends—aging of the population, increasing racial/ethnic diversity, delayed marriage and childbirth, and related factors—explain only a small portion of the housing market’s boom and bust. Instead, the homeownership rate’s rise and fall have been due to broader changes in the economy, credit conditions, and housing markets. This Article then presents homeownership projections for 2015 to 2035, describing three scenarios that define a range of homeownership outcomes. The low and high scenarios presented in this Article produce a range for the national homeownership rate of 60.7% to 64.8% by 2035. The analyses describe the implications of each scenario for growth in the number of homeowner households, as well as the distributional implications of lower versus higher homeownership rates for homeownership outcomes by age, race/ethnicity, and family type.
