Abstract

This paper assesses the damage to high-set rectangular-plan houses with low-pitch gable roofs (built in the 1960 and 70s in the northern parts of Australia) to wind speeds experienced in tropical cyclones. The study estimates the likely failure mode and percentage of failure for a representative proportion of houses with increasing wind speed. Structural reliability concepts are used to determine the levels of damage. The wind load and the component connection strengths are treated as random variables with log-normal distributions. These variables are derived from experiments, structural analysis, damage investigations and experience. This study also incorporates progressive failures and considers the interdependency between the structural components in the house, when estimating the types and percentages of the Overall failures in the population of these houses. The progressively increasing percentage of houses being subjected to high internal pressures resulting from damage to the envelope is considered. Results from this study also compare favourably with levels of damage and related modes of failure for high-set houses observed in post-cyclone damage surveys. [ABSTRACT FROM AUTHOR]

Abstract

The immunosuppressive compounds FK-506, cyclosporin A (CsA) and rapamycin inhibit both the human and mouse mixed lymphocyte reactions (MLR) with IC50s of 2-5 × 10-10 M for FK-506 and rapamycin and 10-8 M for CsA. FK-506 and CsA were also potent inhibitors of A23187/PMA- stimulated IL-2 production by Jurkat and HuT-78 cells but had no effect on the response of mouse CTLL cells to IL-2. IC50 values for inhibition of IL-2 production closely matched those for inhibition of the M LR and both drugs were active only during the first 4-6 hr following stimulation. In contrast, rapamycin was a poor inhibitor of 1L-2 production, although it inhibited cellular responses to IL-2. The IC50 values for these two activities indicated that neither alone accounted for rapamycin inhibition of the MLR. FK-506 and CsA affected IL-2 gene transcription in Jurkat cells by the same mechanism. Both inhibited the appearance of the transcription factor, NFAT, whereas rapamycin did not. The appearance of another transcription factor, NFKβ, was unaffected by all three drugs. The effects of FK-506 and CsA on IL-2 gene expression, therefore, are similar even though the two drugs act through distinct cytosolic receptors. [ABSTRACT FROM AUTHOR]

Abstract

The mini-circle is a transposable element which is present in Streptomyces coelicolor A3(2) in both free circular and chromosomally integrated linear forms. The nucleotide sequences of the mini-circle and its preferred site of integration in the Streptomyces lividans TK64 chromosome were determined. Three putative open reading frames were identified in the mini-circle sequence. The mini-circle does not appear to cause a target site duplication on transposition and does not have perfect terminal inverted repeats. The observed site-specificity of the mini-circle is not mediated by extensive homology between the element and the chromosomal integration site. Transposition of the mini-circle into the S. lividans chromosome was demonstrated and found to be some two orders of magnitude less efficient than integration of the circular form of the element, suggesting that the circular form of the mini-circle might be a normal intermediate in the transposition process. Abstract

The expression of the placental proteins human chorionic gonadotrophin alpha (HCG alpha), beta (HCG beta) and human placental lactogen (HPL) was examined in trophoblast from human normal pregnancy and early embryonic failure (EEF) using Northern blot analysis. Trophoblast from EEF expressed significantly depressed levels of HCG alpha (P < 0.05) and HCG beta (P < 0.01) when compared to normal pregnancy. Levels of expression of HPL in EEF were not significantly different to those from normal pregnancy. The gestational ages of the two groups were not significantly different. Immunocytochemistry on paraffin-fixed tissue sections supported these data and showed that mRNA levels reflected protein production of HCG within the tissue. It appears from these data that the depressed level of HCG found in the serum of women with early pregnancy failure is not solely a consequence of diminished placentation in these pregnancies, or placental necrosis, but that the genes of the alpha and beta HCG subunits are down-regulated. This study further suggests that there are two subsets of women with early pregnancy failure; those with apparently normal levels of placental proteins, and those with severely depressed levels. Abstract

The 12C + 16O decay of 28Si states populated in the 12C(20Ne, 28Si*) reaction has been studied at beam energies of 160 and 110 MeV. Evidence for 28Si excited states is observed for the excitation energy range of 25–45 MeV. Angular correlation analysis shows that the spins of these states lie in the range of 10–16 &barh;. These measurements further indicate that these breakup states are associated with the decay of a 28Si nucleus with a large deformation.

133. A Simple Model for the Diffuse Double Layer Based on a Generalized Mean Spherical Approximation

Abstract

A semiempirical method of including the effects of ionic size in a simple analytical model of the diffuse double layer is described. Guided by earlier work in the hypernetted-chain approximation, the ion−wall correlation functions are modified from their Gouy−Chapman approximations by addition of two new parameters. The first is a constant related to the osmotic pressure and thus to the volume of the ions. The second is related to the effect that ion size has on the thickness of the ionic atmosphere in the electrolyte system, and depends on the electrical field at the wall. The exact form of the potential-dependent correction is found using Monte Carlo estimates of the potential drop across the diffuse layer φd. In this way, an analytical expression for the potential drop is derived and used to estimate φd as a function of electrolyte concentration and charge density on the wall for 1−1 electrolytes assuming that the component ions have a diameter of 425 pm.

135. Pax3 is required for cardiac neural crest migration in the mouse: evidence from the splotch (Sp2H) mutant.

Abstract

Neural crest cells originating in the occipital region of the avian embryo are known to play a vital role in formation of the septum of the cardiac outflow tract and to contribute cells to the aortic arches, thymus, thyroid and parathyroids. This 'cardiac' neural crest sub-population is assumed to exist in mammals, but without direct evidence. In this paper we demonstrate, using RT-PCR and in situ hybridisation, that Pax3 expression can serve as a marker of cardiac neural crest cells in the mouse embryo. Cells of this lineage were traced from the occipital neural tube, via branchial arches 3, 4 and 6, into the aortic sac and aorto-pulmonary outflow tract. Confirmation that these Pax3-positive cells are indeed cardiac neural crest is provided by experiments in which hearts were deprived of a source of colonising neural crest, by organ culture in vitro, with consequent lack of up-regulation of Pax3. Occipital neural crest cell outgrowths in vitro were also shown to express Pax3. Mutation of Pax3, as occurs in the splotch (Sp2H) mouse, results in development of conotruncal heart defects including persistent truncus arteriosus. Homozygotes also exhibit defects of the aortic arches, thymus, thyroid and parathyroids. Pax3-positive neural crest cells were found to emigrate from the occipital neural tube of Sp2H/Sp2H embryos in a relatively normal fashion, but there was a marked deficiency or absence of neural crest cells traversing branchial arches 3, 4 and 6, and entering the cardiac outflow tract. This decreased expression of Pax3 in Sp2H/Sp2H embryos was not due to down-regulation of Pax3 in neural crest cells, as use of independent neural crest markers, Hoxa-3, CrabpI, Prx1, Prx2 and c-met also revealed a deficiency of migrating cardiac neural crest cells in homozygous embryos. This work demonstrates the essential role of the cardiac neural crest in formation of the heart and great vessels in the mouse and, furthermore, shows that Pax3 function is required for the cardiac neural crest to complete its migration to the developing heart.

The mean spherical approximation integral equation is solved for a pair of large charged hard spheres in an ion-dipole mixture. In this work, the effect of the molecular nature of the solvent is taken into account approximately. Extra terms, not included in the DLVO theory, are found and are significant. The effect of dipole alignment near the macroions is repulsive at large separation but can be attractive at short separations.

139. Steroid Hormones and the Metabolism of Oxygen by the Uterus of Pregnant Sheep after Ovariectomy

Abstract

Effects of ovariectomy on maternal hormones, on uterine blood flow, and on O2metabolism of sheep late in gestation were studied. Ovariectomy was associated with lower concentrations of estrone, estradiol, and progestagens in femoral arterial and uterine venous blood and with less variation in the concentration of estrogens, variation which normally is related to day of pregnancy. Birth weight, neonatal weight-gain, and rates of both uterine blood flow and O2consumption apparently were unaffected, but statistically significant differences were observed in the arterial O2content and in the fraction of O2extracted by the uterus. Histologic examination of ovaries during the last half of gestation revealed follicles developing and at least one healthy corpus luteum in every pair of ovaries. This evidence suggests that the ovaries are active, that they contribute to maternal hormonal concentrations, and that they have a measureable effect on patterns of O2metabolism by the mother.

Abstract

Radiation safety and protection is a concern of all users of radiodiagnostic equipment. A retrospective dosimetry study of current technical factors used in full spine and sectional radiography at a chiropractic test facility were compared to established entrance skin exposure limits and averages. The chiropractic filtration and longer source-image receptor distances result in lower patient exposure when compared with sectional views of the same anatomic area. Both techniques studied represented a remarkable reduction in comparison to Ontario averages and Federal limits.
