Mycoplasma suis is a member of the group of uncultivable haemoplasmas which colonise erythrocytes of a wide range of vertebrates. Adhesion to erythrocytes is the crucial step in the unique haemoplasma life cycle. Due to the lack of a cultivation system, no adhesion structures have been identified so far. In order to determine potential adhesion molecules of M. suis, we screened genomic M. suis libraries. The protein MSG1 with glyceraldehyde-3-phosphate dehydrogenase (GAPDH) similarity was identified. The encoding gene msg1 is 1011bp in size. The overall homology of the deduced amino acid sequence to GAPDHs of other pathogenic mycoplasmas ranged from 52.6% to 54.5%. Recombinant MSG1 expressed in Escherichia coli exhibited GAPDH activity. Immunoblot and immunoelectron microscopy analyses using antibodies against rMSG1 verified the membrane and surface localisation of native MSG1 in M. suis. Furthermore, we showed that rMSG1 binds to erythrocyte lysate in a dose-dependent manner. E. coli transformants which express MSG1 on their surface acquire the ability to adhere to porcine erythrocytes. This adhesion could be specifically and significantly inhibited by rMSG1 and antibodies to MSG1. In conclusion, our studies indicate that the membrane-associated MSG1 represents the first putative adhesion protein identified in the group of haemoplasmas.

The antigenic structures of the haemotrophic Mycoplasma suis, an epicellular parasite of porcine erythrocytes, are largely unknown due to its unculturability. In this study, serological proteome and mass spectrometry analyses allowed the characterization of M. suis proteins targeted by the porcine antibody response: two proteins with characteristics of heat shock proteins, two proteins with characteristics of glycolytic enzymes, a RNA helicase- and an actin-like protein. The DnaK-like protein of M. suis (HspA1) was further analysed genetically and functionally. Its encoding gene (M. suis a1 gene) is 1.830 bp in size and corresponds to a 67 kDa protein. Immunoelectron microscopy verified the surface accessibility of HspA1 in M. suis. Recombinant HspA1 expressed in Escherichia coli demonstrated ATPase activity and antigenicity in experimentally infected pigs. In conclusion, this first identification and recombinant expression of an antigenic protein of M. suis provides the basis for the development of vaccines and new in vitro diagnostic assays.

According to the current German animal welfare law, male piglets may be surgical castrated without anaesthesia up to four weeks of life. This surgical procedure is painful during and also after the operation, for newborn animals as well as for adults. This study was aimed to investigate the impact of preoperative application of analgesics (Meloxicam) on the postoperative castration - pain of four to six days old male piglets. In this investigation all animals were randomly distributed in three groups:the first one was only immobilized but had no surgery, the second one was castrated without analgesics, and the third group was castrated after application of Meloxicam. Blood samples were taken immediately before immobilization, castration or application of the analgesic as well as one, four and 28 hours afterwards to determine Cortisol-concentration in the blood serum and, via this stress-marker, to indirectly evaluate the postoperative und possible intraoperative castration-pain. As a result all piglets castrated without preoperative application of Meloxicam showed significantly increased Cortisol-concentration one and four hours after castration. In contrast, piglets castrated with analgesics resulted in no significant increase during the entire experiment.

According to the applicable animal welfare legislation, the surgical castration of pigs is allowed up until the age of 4 weeks, without anaesthesia. According to the european guideline (2001/93/EG) it is only permitted in the first week after birth. The investigation should show, whether the castration of young piglets takes a milder course and to what extent stress reactions occur in different age groups. The healing process of castration wounds in piglets that were castrated at the age of four days progressed more rapidly and with less complications than those piglets that were castrated at 28 days of age. The catecholamine levels in younger piglets rose significantly after the operation, while these levels virtually stayed the same in piglets castrated at 28 days. As alternatives to castration without anaesthesia, several methods are in discussion: (1) Castration under general anaesthesia can only be practiced by a veterinary surgeon. The sole use of Azaperon and Ketamine has insufficient pain sedating effect. Isofluran anaesthesia is apparatively extravagant. (2) CO2-Anaesthesia in piglets leads to high strain. (3) Castration under local anaesthesia must be practiced by a veterinary surgeon. The application and the pain after the castration are not taken into consideration. (4) Jung boar fattening up until the slaughtering weight of 80 kg is not transformable, because of boar taste. (5) The breeding of slaughter pigs with little boar taint is not yet transformable. (6) Sperm sexing is not and will not, in the near future, be mature for practice. (7) Immunocastration is an active immunisation against GnRH. The immunological elimination of GnRH suppresses the development of sex hormones, such as testosterone, as well as the substance responsible for boar taint, Androstenone. To consider is the acceptance of the consumer. The preparation has the same effect in humans as it has in swine. "Self injections" have the same effect in humans as it has in swine. "Self injections" are therefore risky.

Porcine circovirus type 2 (PCV2) seems to cause reproductive failure in sows not only in experimental studies. A retrospective study was made with a total of 252 aborted fetuses, mummified fetuses, stillborn and nonviable neonatal piglets to determine the presence of PCV2, porcine parvovirus (PPV) and porcine respiratory and reproductive syndrome virus (PRRSV) by PCR. PCV2 was found in all stages of gestation in 27.1 percent of samples examined. A statistically significant association could be shown between the detection of PCV2 and PRRSV. However, no significant association was seen between the detection of PCV2 and PPV and between PPV and PRRSV.

A chlamydial vaccine efficacy trial with assessment of the clinical acceptability and serum antibody responses was performed in breeding sows. A BGM cell culture derived vaccine containing 10(8)/ml formalin-inactivated purified elementary bodies (Eb.) in sterile 0.15 M saline was prepared from Chlamydophila (Ch.) abortus strain OCHL03/99 which has been isolated in the herd from a sample of vaginal discharge. Vaccination was performed as a randomised trial with parallel treatment of a vaccinated group (25 sows) and non-vaccinated control group (20 sows). Sows received two 2.0-ml doses of vaccine intramuscularly at a three week interval. Control sows were dosed with sterile 0.15 M saline, accordingly. Serological response to vaccination was measured by ELISA with a total of 204 blood serum samples (114 from the vaccine group; 90 from the control group) using crude chlamydial LPS as the antigen. Compared to the control group, vaccinated sows showed a marked primary and secondary IgG serum antibody response following the two vaccinations. Antibody levels peaked between week 7 and 14 after priming vaccination, declined incrementally until week 27 but remained significantly higher than the corresponding sham-immune control levels and the prevaccination values of the vaccine group (p < 0.05). Western blot analysis of solubilized whole Eb. of Ch. abortus, Ch. pecorum, and Chlamydia (C.) suis with pre- and postvaccination sera confirmed that vaccination induced an antibody response preferentially against a range of 13 chlamydial antigens including the 40 kDa MOMP of Ch. abortus. Clinical side effects consisting of a transient mild local inflammatory reaction at the site of injection were observed in approx. 30% of vaccinated sows. These results provide the basis for further clinical evaluation of the Ch. abortus vaccine to protect sows from chlamydia-induced reproductive disorders.

An efficient method of control of porcine eperythrozoonosis (PE) caused by Mycoplasma suis is eradication of infection by detection and removal of infected carrier animals. At present, only a few tests are available for the diagnosis of these latent M. suis infections in pigs. The objective of this study was to develop a PCR assay based on novel DNA sequences for the identification of M. suis-infected pigs. A 1.8 kb EcoRI DNA fragment of the M. suis genome was isolated from the blood of pigs experimentally infected with M. suis. Specificity of the DNA fragment was confirmed by DNA sequence analysis and PCR using primers directed against sequences contained in the 1.8 kb fragment. PCR products of 782 bp in size were amplified only from M. suis particles prepared from the blood of experimentally infected pigs but not from any controls, comprising blood from gnotobiotic piglets and a panel of bacteria including other porcine mycoplasmas. PCR results were confirmed by dot blot hybridisation. The applicability of the PCR assay to diagnose M. suis infections in pigs was evaluated by investigating blood samples from 10 symptomatic pigs with clinical signs typical of porcine eperythrozoonosis and blood samples from 10 healthy pigs. The M. suis-specific PCR product was amplified from all samples taken at episodes of acute disease as well as from samples taken during the latent stage of infection, thus demonstrating the suitability of the PCR assay for detecting latent infected carrier animals.

FMD in pigs is primarily a foot disease. It is dominated by rather painful formation of vesicles in the epidermis of the feet (coronary band, interdigital clefts, bulbs) combined with severe lameness. Complications will be seen as detachment of the hoof and secondary infection of disrupted aphthae which may cause purulent arthritis of the pedal joint. Concerning differential diagnosis are to be considered all cases of acute lameness, vesicle-formation as typical lesions of exudative epidermites of pemphigus in the course of infections with S. hyicus or S. aureus as well as degenerative changes after selenium-intoxication, specially near the coronary band, sometimes linked with exungulation.

The reservoirs and the modes of transmission of the most frequent microsporidial species in humans, Enterocytozoon bieneusi, are still unknown. We have examined fecal samples of 26 humans and 350 animals from 37 species to find 18 samples containing this parasite from humans, cats, pigs, cattle, and a llama. Genotypic characterization of the internal transcribed spacer of the rRNA gene resulted in 14 different genotypes, 6 of them previously undescribed. Phylogenetic analysis revealed the lack of a transmission barrier between E. bieneusi from humans and animals (cats, pigs, and cattle). Thus, E. bieneusi appears to be a zoonotic pathogen.

60 pigs representing all age groups (suckling pigs, weaner pigs, hogs, gilts and sows; thereof 37 females, 2 males, 21 castrated males) were examined by ultrasound of the liver and by ultrasound guided gallbladder puncture. The visibility of the liver and gallbladder was strongly influenced by the size of the animals. The thickness of the abdominal and thoracic walls in older animals proved to be a highly ultrasound-absorptive medium, which limited the ability to assess the underlying tissue structures. As a result, gallbladder puncture of these animals was possible only with a certain degree of technical difficulty. The gallbladder puncture procedure itself posed little risk to the animal. The primary risk resulted from the general anesthesia required. Autopsy showed no pathological findings due to gallbladder puncture with the exception of minimal, rapidly healing, local infectious processes. The bile proved to be a suitable medium of the detection of mycotoxins and their metabolites. Selective accumulation of these toxins in bile provide a more reliable diagnostic tool than the standard mycotoxicological tests of feed.

Over 650 pigs died within a couple hours in a fattening unit with approximately 3,000 fattening spaces. The pigs showed vomiting, dyspnea, kyphosis, sunken flanks, diarrhea, and polyuria. Another striking symptom of the pigs, besides the apathy, was the aphonia, due to the calcification of the vocal cords. An acute vitamin D3-intoxication was found to be the cause. The pathologic findings, especially the histologic detection of calcification processes of the soft tissues, lead to the suspect of an intoxication with a vitamin D-like substance. Between 39,000 and 196,000 IU/kg of vitamin D3 have been detected in a ready-to-use food mix. 8.8 million IU/kg of crystaline vitamin D3 were found in an open whey bag. An explanation how vitamin D came into the bag could not be clarified to this point.

In a field trial, the development of antibodies of a combined vaccine against the porcine parvovirus (PPV) as well as against swine erysipelas was compared with corresponding mono vaccines. Furthermore, these vaccines were used in different vaccination schedules. The tests were carried out on 109 gilts in three closed farms. In all gilts, a basic immunization repeated twice was carried out at the age of six months and at intervals of three weeks. The revaccination was carried out four months after the basic immunization with half of the animals, and six months after the basic immunization with the remaining gilts. Between the combined vaccine and the mono vaccine no significant differences in the development of antibodies against PPV could be found according to different vaccination schedules. The gilts having been vaccinated with the mono vaccine and boostered six months later showed significantly higher antibody titers against Erysipelothrix rhusiopathiae. Between the remaining vaccination groups no significant difference in the development of the antibodies against swine erysipelas could be found. On only one farm, a continuous decrease of antibody titers against PPV in case of altogether 238 non-vaccinated piglets until the sixth month of life could be observed. On the two other farms, an increase of antibody titers against PPV could be found at different points of time, which indicates an infection of the piglets. Between the individual vaccination groups no significant antibody titers against PPV could be measured in milk tests. With regard to the number of piglets born alive per litter, the number of piglets born dead per litter and the number of mummies, a significant difference could neither be found between the vaccination groups 1-4.

Epizootiological, clinical, bacteriological and haematological studies were carried out to assess the effectiveness of the recently developed cephalosporin preparation Cefquinome in the treatment of the puerperal septicaemia and toxaemia syndrome. Cefquinome was administered at three different doses (1, 2 and 4 mg/kg BW) to 188 sows with feverish puerperal illness. Amoxicillin (7 mg/kg BW) was used as a control drug. In 41% of cases endometritis was a monoinfection whereas in 70% of mammary infections mixed infections were diagnosed. Results showed that for therapy of puerperal septicaemia and toxaemia Cefquinome at doses of 2 mg/kg BW and 4 mg/kg BW is clearly more effective than the control drug Amoxicillin and Cefquinome at its lowest dose of 1 mg/kg BW.

In wild boars kept in a paddock of approximately 1.7 square miles Brucella suis biotype 2 was isolated and Aujeszky's disease was diagnosed by serological tests. To clear the paddock, the wild boars were lured by offered food and caught using specially built smaller pens. They were then transported to a location far off their original surroundings and euthanized. Of a total of 297 wild boars kept in the paddock, 71% were caught and 28% shot. Three wild boars (= 1%) were found trapped. The described method could also be used in case of epidemics to catch wild boars living outside existing paddocks. In order to avoid killing the boars which stayed in separate quarantine enclosures, a darting gun was used to anaesthetize the animals with tiletamine/zolazepam. Achieved sedation levels were deep enough to allow for blood samples to be taken from the jugular vein without any problems, i.e. without any defence reactions of the boars. The origin of infection could not be identified.

In a three years old sow, neurologic signs resulting from lesions of some cranial nerves and similar to the symptoms typical of porcine pituitary abscesses were described. A generalized leukosis with tumor cell infiltration occurring in numerous organs e.g. in the pituitary gland was diagnosed in this animal. The neurologic and pathogenetic differences between the described case and the porcine pituitary abscess syndrome were discussed.

Since the beginning of 1997 the use of the hypnotic drug metomidate (Hypnodil) in swine is not longer allowed. This ban caused a substantial therapeutic deficit for anesthesia in swine. The ban is based on provisions of the European Council Directive 2377/90 for fixing maximum residue limits (MRL) of veterinary drugs in animal-derived food and due to the fact that the pharmaceutical industry has not submitted any application for determining an MRL for metomidate. According to the regulations of the German drug law only those substances can be used as alternatives which are authorized for use in swine or another food-producing animal species. Registrations for use in swine exist for the barbiturates thiamylal and narcobarbital, for ketamine and for the neuroleptics azaperone, acepromazine and propionylpromazine. In the exceptional case of therapeutic emergency levomethadone or xylazine can be used. The administration of propofol, thiopentone, opioids (except levomethadone) or benzodiazepines (except brotizolam) to swine is not allowed since these drugs are authorized only for use in humans or companion animals. At present the most appropriate alternative for anesthesia in swine is thiamylal after premedication of azaperone and ketamine inducing general anesthesia with sufficient tolerance at low risks. In current studies general anesthesia with tolerance could further be produced by combinations of levomethadone, ketamine and azaperone or xylazine. Combined use of levomethadone with benzodiazepines appeared to be not suitable in swine.

CSF samples were taken from 50 healthy pigs of both sexes and cytologically and biochemically examined. With the aid of regression calculations it was investigated, whether deep freezing of CSF samples influences the laboratory results. The parameters glucose, urea, creatinine, total protein, magnesium, anorganic phosphate, AST and AP were in good agreement with the values of the original samples exhibiting only slight variations. Minor age relationship was only seen with respect to the two enzymes LDH and HBDH. A linear relationship was found between values in CSF and in serum of the parameters urea, creatinine, sodium and potassium. CSF examination should include cell count, Pandy's test and bacteriological investigation as well as the determination of the levels of the enzymes LDH and HBDH. The activity of the enzymes AST and ALT increases along with the severity of the tissue damage (meningitis). The specific gravity increases similarly to the total protein level in cases of chronic compression of the spinal cord, and Pandy's test becomes positive. However, CSF glucose levels are partly reduced in animals with central nervous symptoms. The levels of the enzymes AP and CK are often increased in central nervous disturbances, but do not admit drawing a conclusion on the degree of damage.

In this study, the protective effect of specific egg yolk antibodies on diarrhea caused by Escherichia coli K88 (F4) was investigated with 179 weaning piglets in a double-blind field trial. The piglets were divided into three groups. The antibody group received egg powder with specific antibodies to E. coli K88, K99, 987P, and rotavirus, while one control group was fed with egg powder without specific antibodies and a second control group received no egg powder at all. The piglets were fed ad libitum. The egg powder was offered in a 5% feed ration. Compared with the control groups, the piglets of the antibody group showed significant differences (P < 0.05, chi 2-test) in the parameters rate of diarrhea (17.2%) (antibody group) to 60.7% (control egg group) or 56.7% (control group without egg powder), severity of symptoms (5.2-39.3% or 26.7%) and frequency of additional treatments (8.6-55.7% or 51.7%). Because of the additional treatments, no difference could be found in body-weight gain. In control groups, three piglets died, while no pigs died in the antibody group.

Paresis of the hindlimb in piglets after intramuscular injection of various therapeutics in the caudal thigh muscles were observed. The injection was followed by inflammatory changes of the muscle tissue at the injection site. The tibial and fibular nerve were enclosed in the alterations and showed a distinct to complete loss of nerve fibers resulting in clinical signs of paresis. The affected hindlimb was dragged on the dorsal digital surface. The piglet showed an extended consecutive swelling and ulceration below the hock and loss of the claws. These findings strongly demonstrate the necessity of a correct intramuscular injection in piglets, which should be done in the lateral neck muscles.

Acute myeloid leukemia is a rare finding in swine. A case of acute promyelocytic leukemia is described by light microscopy, enzyme- and immunohistochemistry as well as electron microscopy. Tumour cells can be demonstrated in bone marrow, lymph nodes, liver, spleen, kidneys, small intestine and myocardium. The tumorous neutrophil precursor cells are positive for naphthol-AS-D-chloroacetate esterase and lysozyme. They show ultrastructurally numerous granules as typical for promyelocytes. Virus particles of the oncorna-c-type can be demonstrated within the tumour cells.

We report on the outbreak of dermatomycosis caused by Microsporum canis at a bavarian pig breeding and fattening farm. Clinical signs were shown mainly by younger pigs. Microsporum canis could be identified in skin scrapings of pigs, cats, and the farmer's daughter by culture and fluorescence microscopy. Questions of epizootiology, diagnostics and therapy are discussed.

The effects of immunosuppressive treatment on natural-killer-cell activity and the manifestation of eperythrozoonosis in swine were investigated. After infection with Eperythrozoon suis (E. suis), German Landrace piglets received daily intravenous (i.v.) applications of cyclophosphamide (CY, Endoxan) on 6 consecutive days. A combined immunosuppressive treatment using azathioprine (Imurek), a single application of dexamethasone (Devan), and two injections of anti-asialo GM1 serum (1:50) was performed with a piglet over a period of 9 days. Leucocyte counts, differential blood-cell count, and microhaematocrit and rectal temperature were controlled. Direct microscopic observation of E. suis organisms was performed in Giemsastained blood smears. Immunosuppressive treatment resulted in relative lymphocytosis and transient agranulocytosis. CY and azathioprine therapy caused a temporary increase of E. suis organisms in red blood cells. Natural-killer-(NK) cell activity mediated by nylon-wool non-adherent peripheral-blood mononuclear leucocytes (NAD-PBML) was recorded during and after immunosuppressive treatment of pigs infected with E. suis. Cytotoxic activity of NK cells was consistently reduced after two CY injections and further decreased to 70-100% of the initial NK-mediated tumor cell lysis. NAD-PBML of two E. suis infected animals were monitored for cytotoxicity during CY immunosuppressive treatment and were compared to Percoll density separated NK effector cells derived from the same blood sample. Enhancement of NK-cell activity by density separation of NAD-PBML was not longer possible on days 3 and 4 during CY treatment, indicating a decrease of NK cells in peripheral blood. In vitro, only a high dose of CY (4 microgram/ml) suppressed cytolytic activity of NAD-PBML. Short-term treatment with immunosuppressive agents did not replace splenectomy in inducing clinically apparent eperythrozoonosis.

The application of sonography at pigs has hitherto been confined to gynecological questions. In this examination applicabilities of the ultrasonic method are examined from the internal point of view. The sonographical normal picture of organ structures and the value of this method as aid for diagnosis of clinically manifest changes are shown. A 5MHz linear-scanner and a 3.5 MHz convex-scanner (76 degrees) turned out to be suitable. 112 patients of the Clinic of Internal Medicine II and additional 35 healthy pigs were examined. Preliminary examinations were made with isolated organs, which were scanned in a water bath. The sonographical findings of pathological changes could be secured by the postmortem findings. Within the scope of the main examinations, manners of representation and sonographical appearance of liver, gall-bladder, kidneys, urinary-bladder, stomach, lungs, muscular system, spine and transponder were elaborated. The standardized measuring of liver and kidney proved statistically secured dependencies among the measuring data and were significantly related with the body-weight. The ultrasonic guided puncture of gall-urinary bladder and of liquor cerebrospinalis could be carried out successfully.

Epizootic examinations were made on 796 newborn piglets to test the consequences of the resection of a piglet's canine and lateral incisor teeth on its first day of life. As a comparative examination the resection was carried out with side-cutter pliers or with the teeth grinder PIGMATIC 110. A third group of piglets was not treated. Histological, radiographic and bacteriological examinations were made of 10 piglets--in each case with piglets with the resection with side-cutter pliers or with the teeth grinder. 48% of ground teeth reacted with an inflammation of the pulp. The clipping of the crown of teeth with side-cutter pliers caused pulpitis in 92% of the teeth. Inflammation of the gingiva could almost only be seen around clipped teeth. Splinters only occurred with teeth which had been clipped with side-cutter pliers. Gingivitis and pulpitis extended along the splinters to the bottom of the root. Bites among the litter mates were more frequent in the group of piglets whose teeth had not been treated than in the group of piglets with resected teeth. Compared to the grinding or the clipping of needle teeth, bites to the sow's udder were much more frequent if the resection had been omitted. During the whole period of examination the mortality of piglets was at its lowest in the group of piglets with clipped teeth. The development of grinding instruments for the resection of needle teeth presents a method which reduces the negative consequences of the conventional resection with side-cutter pliers and shows clearly the advantages of resection. In terms of animal welfare, teeth resection is an amputation and therefore every case requires veterinary justification.

The efficiency of magnesium-aspartate-hydrochloride (MAH) in prophylaxis of porcine stress-induced myopathy was investigated. For the examination a conveyor belt was used. MAH was administered orally or intramuscularly. Clinical investigations, blood specimen collections and ECG were carried out before and after running. Stress-induced tachypnoea and tachycardia and hyperthermia were reduced after both forms of administration of MAH. The enzyme activity of CK, LDH and alpha-HBDH increased slighter after oral and i. m. application. The increase of serum glucose concentration as well as serum lactate level were reduced after oral and i. m. substitution with MAH. In the ECG apart from the relative diastolic period all time values were significantly prolonged after MAH-substitution. The results indicate that several stress-induced changes of clinical and metabolic parameters can be reduced by administration of MAH.

The immersible culture medium MICUR-BT used in human medicine for the detection of antimicrobial substances in the urine was tested for its suitability in pigs. In in-vitro tests, 15 pure antibiotic substances, 2 pure substance mixtures as well as 5 veterinary injection preparations with no direct antimicrobial effect were checked. The agar perforation test was used as a reference procedure. In a total of 65 pigs the detectability of 9 pure antibiotic and chemotherapeutic agents, 2 pure substance mixtures as well as 5 veterinary drugs with no direct antimicrobial effect was tested after oral or parenteral application. In the in-vivo tests the MICUR-BT test turned out to be more reliable than the agar perforation test. In 76.9% of the untreated pigs below 30 kg b.w. the MICUR-BT-test showed an antimicrobial effect. Such effects were reduced by 38.3% in the reference procedure.

Osmotic fragility of erythrocytes was tested in weaned pigs experimentally infected with Eperythrozoon (E.) suis. Acute eperythrozoonosis of splenectomized pigs led to an increase of osmotic fragility. It is supposed that E. suis infection causes a structural change in erythrocyte membrane. Possible mechanisms of this cell membrane injury are discussed.

A light and electron microscopic study was carried out on pigs experimentally infected with Eperythrozoon (E.) suis during the acute and chronic phase of infection. During the acute phase Eperythrozoon organisms can be recognised by light microscopy as bright to dark orange points or circles. In contrast, in the chronically infected pigs E. suis appear as bright yellow or green small dots on the edge of the erythrocytes or in the plasma. By scanning electron microscopy, immature, juvenile and mature forms of E. suis could be seen on the same erythrocyte during the acute phase. In chronically infected pigs ring-shaped, rounded resting-stage forms, measuring about 1 micron in diameter, can be observed.

Eperythrozoon infection can be diagnosed clinically and hematologically only during acute attacks. In the febrile phase it is possible to detect the eperythrozoon organisms in the blood smear. The best method for staining is that by acridine-orange and examination by fluorescence microscopy. Serological identification (IHA, KBR, ELISA) is suitable for herd diagnosis only, because of the undulating level of the titre. In the individual case only positive results are regarded as significant. The best method to diagnose a latent infection of eperythrozoon suis is to splenectomize a suspected pig. The infection with eperythrozoon suis is a herd problem, therefore an examination should be made whenever there are pale pigs with fever, or a reduction of efficiency or an increased susceptibility to other infections.

Following on from clinical observations which point to severe metabolic disturbances in association with acute Eperythrozoon (E.) suis infection, the parameters of acid-base balance (pO2, pCO2, pH, actual bicarbonate, standard bicarbonate, base excess) as well as the glucose-, lactate- and pyruvate levels, were measured in venous blood during the course of eperythrozoonotic infection. Glucose consumption was investigated in in vitro experiments with differing numbers of pathogens. Acute E. suis infection is accompanied by a severe acidosis and hypoglycaemia. In vitro experiments showed that a rapid breakdown of glucose follows in E. suis infected blood. No significant reduction in glucose concentration was established in control blood in a comparable time period. The results give rise to the assumption that E. suis is capable of independent glucose breakdown. Both the increase in lactate concentration (metabolic component) and a disturbance of pulmonary gaseous exchange (respiratory component) are regarded as the cause of the acidosis.

Serological studies by means of a CF-Test and Elisa were undertaken for evaluation of antibody titers against Eperythrozoon suis infection in swine. The animals were partly splenectomized and underwent artificial infection. It could be shown that the splenectomized animals showed higher and longer lasting antibody titers than the non splenectomized swine. It is stated, that the above mentioned tests can be used in diagnosis on a herd basis but not on a single animal basis.

The influence of latent and of splenectomy-induced clinically manifest Eperythrozoon suis infection on the following parameters of the carbohydrate metabolism and the acid-base status was tested in venous blood of German Landrace pigs: Levels of glucose, lactic and pyruvic acid, blood-pH, base excess, actual bicarbonate concentration, standard bicarbonate concentration, pCO2, pO2. The latent E. suis infection resulted in a consistent decrease of blood glucose level. 23 days after infection, blood glucose was reduced by 25% of the initial value. The other parameters were not changed by latent E. suis infection. Acute Eperythrozoonosis induced severe hypoglycaemia (means Gluc, = 39.7 mg/dl and blood acidosis (means pH = 7.13). In vitro experiments showed that break-down of glucose in E. suis infected blood occurs very rapidly. There was no significant reduction of the glucose concentration in control blood that had been treated accordingly. There was an increase of lactic acid (means = 62.7 mg/dl), pyruvic acid (means = 1.86 mg/dl), and pCO2 (means = 82.1 mm Hg). The concentrations of actual bicarbonate (means = 24.8 mmol/l) and standard bicarbonate (means = 20.9 mmol/l) were lowered, and there was a negative base excess (means = -3.56 mmol/l). The ratio of lactic and pyruvic acid changed from 11:1 to 30:1. It seems likely that E. suis itself is able to metabolize glucose. Acidosis is considered to result from both the increase of lactic acid (metabolic component) and an impairment of pulmonary gas exchange (respiratory component).

Investigations were carried out on the influence of latent and clinically manifest Eperythrozoon suis infection upon haemostasis in swine. The study was carried out with 14 German Landrace pigs. Latent eperythrozoonosis was induced in 7 animals by experimental infection. Splenectomy of these 7 animals and 2 spontaneously infected pigs led to clinical manifestation of eperythrozoonosis. Five clinically healthy pigs were splenectomized and served as controls. In healthy pigs splenectomy was followed by a transient rise in fibrinogen and platelet count. Latent infection with Eperythrozoon suis did not cause an impairment of haemostasis. Acute eperythrozoonosis was associated with increased haemorrhagic tendency considered to be a consequence of intravascular coagulation and subsequent consumption coagulopathy. There was a prolongation of partial thromboplastin time and prothrombin time (Quick) and a decrease of platelet count. Thrombelastography showed prolongation of reaction and clot building time and a short-term decrease of maximum amplitude. Deviation from normal values was proportional to the number of red blood cells infected with Eperythrozoon suis. Anti-rickettsial therapy led to quick normalization of haemostasis. Various aspects of the cause and the consequences of the haemostatic defect are discussed with special regard to the underlying disease.

Ten splenectomized and non-splenectomized pigs were experimentally infected with E. suis bearing red blood cells in order to determine the antibody response. All animals were monitored for antibody titer by indirect hemagglutination over a period of 80-290 days postinfection. Latent E. suis infection only yielded a detectable antibody titer in one pig. Acutely infected pigs had a titer ranging up to 1:640. Maximum antibody response lasted only 2 months and dropped below the level of detection of our assay within 2 to 3 months. At this time, the clinical symptoms could reappear and antibodies were again detectable. However, no booster effect was observed with this second outbreak. We also determined the antibody frequency in 138 pigs from 16 herds in Southern Germany. Pigs from only 4 out of 6 clinically positive herds had antibody titer against E. suis. 20 out of 78 pigs of the clinical positive herds demonstrated a detectable E. suis antibody titer. In 10 herds that were asymptomatic and presumed uninfected all 80 pigs were serologically negative for E. suis.

For clinical interests it is advisable to subdivide cases of swine poisoning in such as caused by food, drugs and environmental poisonings. This division gives pointers to aetiologic connections and special measures necessary for the clearing of the processes. With food poisoning mycotoxicoses play an evermore important role, whereas poisonings by trace elements are on the decrease. Sodium chloride poisoning often results primarily from insufficient water supply. With environmental poisonings carbon monoxide and cyanamide intoxication are presented. Poisonings caused by drugs are mainly the result of an overdose, of segregation in food or of non-licensed drugs. A relatively unknown swine poisoning by a drug against coccidiosis--licensed for poultry--is described.

In older gravid sows in the last stage of gestation or after pariturition lateroflexion of the urinary bladder into the space between the vagina and the pelvic wall occasionally occurs. The reposition of the displaced urinary bladder by exerting external pressure either on the vulva or the perineum is usually unsuccessful. Only after the bladder has been emptied by a catheter spontaneous reposition does occur. In two cases displacement of the urinary bladder occurred immediately after normal pariturition. In both cases relapses were observed after the removal of the implanted balloon catheter. Both sows had the urinary balloon catheter reinserted and it was left in place until the weaning of the piglets. There were no complications during the whole lactation period. Both sows reared their piglets and could then be slaughtered.

In a feeding trial lasting for months Halothane-positive fattening pigs were put on a diet containing different concentrations of vitamin E and selenium. After a Myostress application vitamin E and selenium were additionally injected in order to examine stress resistance (CK-test) and regenerative capacity (histological examination of Musculus longissimus dorsi) of skeletal muscle. The study revealed that neither high dietary levels of vitamin E/selenium nor a vitamin E/selenium treatment applied after stress are capable of preventing stress related myopathy nor can they influence its healing up favourably.

Zearalenone is one of the most frequent, naturally occurring mycotoxins in feedstuffs: 13.9% of 710 samples analyzed contained zearalenone in concentrations of 1.0 to 1,725 micrograms/kg. Trial feeding with female pigs demonstrated that a zearalenone concentration of 250 micrograms/kg feed produced distinct redness and swelling of the vulva, slight swelling of the mamma and numerous vesicular follicles and some cystic follicles on the ovaria. The symptoms on the external genital tract were not observed when the zearalenone concentration in the feedstuff, was 50 micrograms/kg but the section also established that vesicular follicles on the ovaria of these animals were more frequent than on the ovaria of the control animal.

Pigs are not only anaesthetized in case of operations but often also if diagnostic and therapeutic procedures are necessary. Choice of the anaesthetic and way of application depend on the size of the patient and the indication. The only important local anaesthesia in pigs is epidural anaesthesia. It is indicated in all longer lasting painful procedures in caudal regions. The technique of this anaesthesia is described. From the possible methods of general anaesthesias, in practice injection anaesthesia has been generally accepted for several reasons. Together with barbiturate anaesthesia neurolept-analgesia with Azaperone--Metomidate has become more and more important, because with this form of anaesthesia the technically easy intra-abdominal application is possible in pigs under 15 kg of body weight.
