105. SVM-Based Optimization on the Number of Data Streams for Massive MIMO Systems

Abstract

For millimeter-wave (mmWave) massive multiple-input and multiple-output (MIMO) systems, hardware cost and power consumption can be reduced via hybrid precoding, while keeping almost the same spectral efficiency (SE) as the fully digital precoding scheme. However, for different hardware configuration and channel environments, system SE varies with the number of transmitted data streams at the transmitter, and hence this should be determined primarily before hybrid precoding design. In this article, for single-user massive MIMO systems, a regression model based on the support vector machine technique is proposed, through which the optimal number of data streams can be obtained automatically based on the propagation characteristics of wireless channels, the number of antennas, and RF chains. To validate the correctness of the proposed model, simulation results are presented under various parameter settings.

108. H 2 O 2 /DEM-Promoted Maft Promoter Demethylation Drives Nrf2/ARE Activation in Zebrafish.

Abstract

The Nrf2/ARE signaling pathway is a cell survival response pathway in response to environmental stresses. The Nrf2/ARE signaling pathway can be activated by stimulating cysteine residues at different positions in the Keap1. However, the epigenetic mechanisms of the Nrf2/ARE pathway under different stimuli are still poorly understood. In this study, we found that both hydrogen peroxide (H2O2) and Diethyl Maleate (DEM) activated the Nrf2/ARE signaling pathway at 120 hpf in zebrafish. H2O2 regulated the demethylation of the maft promoter by inhibiting the expression of methyltransferase. This promotes the mRNA expression of the Nrf2 binding factor maft, thereby promoting the downstream antioxidant genes. The methylation of the Nrf2/ARE signaling pathway was not significantly regulated by DEM. However, under oxidative stress, the methyltransferase inhibitors (decitabine and azacitidine) demethylated the promoter region of maft. It activated the expression of the maft, further improving the Nrf2/ARE signal pathway. At last, antioxidant target genes were activated. It was shown that H2O2 and DEM cooperated with methyltransferase inhibitors, providing an important reference for the treatment of oxidative stress-related diseases and breaking new ground for the study of the mechanism of methyltransferase inhibitors in the process of tumor chemotherapy. 111. Double-hexahistidine tag with high-affinity binding for protein immobilization, purification, and detection on Ni-Nitrilotriacetic acid surfaces

Abstract

There is a particular need in protein analysis and purification for specific, functional, and generic methods of protein immobilization on solid supports. Here we describe a double-hexahistidine ([His.sub.6]) tag sequence, comprising two hexahistidines separated by an 11-amino acid spacer, which shows at least 1 order of magnitude stronger binding to Ni-NTA-modified surfaces than a conventional single-[His.sub.6] tag or two single-[His.sub.6] tags at N- and C-termini. Using, as a model, tagged versions of green fluorescent protein (GFP), stable and tight binding of the double-[His.sub.6] tag/Ni-NTA interaction was demonstrated by competitive elution from Ni-NTA agarose beads, surface plasmon resonance on a Ni-NTA chip, and ELISA in Ni-NTA microwell plates. Protein purification by Ni-NTA chromatography was improved by a 6-8-fold increase in imidazole concentration required for elution, while the dissociation rate of double-[His.sub.6] GFP from Ni NTA chips in SPR (BIAcore) was 10 times slower than for single-[His.sub.6]-tagged proteins. ELISA assays and protein microarrays constructed with double-[His.sub.6] GFP demonstrated greater detection sensitivity with anti-His antibodies and Ni-NTA conjugates. Moreover, the double-[His.sub.6] tag could serve simultaneously both for protein immobilization and for detection on surfaces. The double-[His.sub.6] peptide has the potential to be a universal tag for protein immobilization and detection on arrays and single-step purification of proteins from crude mixtures.

137. Si-Disordering in MgAl2O4-Spinel at High P-T Conditions, with Implication to Si-Mg Disorder in Mg2SiO4-Ringwoodite

Abstract

A series of Si-bearing MgAl2O4-spinels were synthesized at 1500-1650 °C and 3–6 GPa. These spinels had SiO2 contents up to ~1.03 wt%, and showed a substitution mechanism of Si4+ + Mg2+ = 2Al3+. Unpolarized Raman spectra were collected from polished single grains, and displayed a set of well-defined Raman peaks at ~610, 823, 856 and 968 cm-1 which had not been observed before. Aided with the Raman features of natural Si-free MgAl2O4-spinel, synthetic Si-free MgAl2O4-spinel, natural low quartz, synthetic coesite, synthetic stishovite and synthetic forsterite, we infer that these Raman peaks should belong to the SiO4 groups. The relations between the Raman intensities and SiO2 contents of the Si-bearing MgAl2O4-spinels suggest that at some P-T conditions some Si must adopt the M-site. Unlike the SiO4 groups with very intense Raman signals, the SiO6 groups are largely Raman-inactive. We have further found that the Si cations primarily appear on the T-site at P-T conditions ≤ ~3–4 GPa and 1500 °C, but attain a random distribution between the T-site and M-site at P-T conditions ≥ ~5–6 GPa and 1630–1650 °C. This Si-disordering process observed for the Si-bearing MgAl2O4-spinels hints that similar Si-disordering might happen to the (Mg,Fe)2SiO4-spinels (ringwoodite), the major phase in the lower part of the mantle transition zone of the Earth and the index mineral for the very strong shock stage experienced by extraterrestrial materials. The likely consequences have been explored.
