Error-tolerant likelihood-based match calling presents a promising technique to accurately identify recapture events in genetic mark–recapture studies by combining probabilities of latent genotypes and probabilities of observed genotypes, which may contain genotyping errors. Combined with clustering algorithms to group samples into sets of recaptures based upon pairwise match calls, these tools can be used to reconstruct accurate capture histories for mark–recapture modelling. Here, we assess the performance of a recently introduced error-tolerant likelihood-based match-calling model and sample clustering algorithm for genetic mark–recapture studies. We assessed both biallelic (i.e. single nucleotide polymorphisms; SNP) and multiallelic (i.e. microsatellite; MSAT) markers using a combination of simulation analyses and case study data on Pacific walrus (Odobenus rosmarus divergens) and fishers (Pekania pennanti). A novel two-stage clustering approach is demonstrated for genetic mark–recapture applications. First, repeat captures within a sampling occasion are identified. Subsequently, recaptures across sampling occasions are identified. The likelihood-based matching protocol performed well in simulation trials, demonstrating utility for use in a wide range of genetic mark–recapture studies. Moderately sized SNP (more than or equal to 64) and MSAT (10–15) panels produced accurate match calls for recaptures and accurate non-match calls for samples from closely related individuals in the face of low to moderate genotyping error. Furthermore, matching performance remained stable or increased as the number of genetic markers increased, genotyping error notwithstanding.

The data correspond to total integrated luminosities of up to 9.7 fb$^{−1}$ per experiment. The t-channel cross section is measured to be σ$_t$=2.25$_{-0.31}^{+0.29}$ pb. We also present the combinations of the two-dimensional measurements of the s- vs t-channel cross section. In addition, we give the combination of the s+t channel cross section measurement resulting in σ$_{s+t}$=3.30$_{-0.40}^{+0.52}$ pb, without assuming the standard model value for the ratio σ$_s$/σ$_t$. Abstract: Over the past decade, several new medications have been developed to treat type 2 diabetes mellitus. Large‐scale outcome trials have been performed with patients at high cardiovascular risk to assess the cardiovascular safety of these agents. These trials are changing the landscape of diabetes therapy with evidence beyond safety to cardiovascular benefits of sodium‐glucose cotransporter 2 (SGLT‐2) inhibitors and some glucagon‐like peptide‐1 receptor agonists. This review provides an overview of incretin‐based therapies and SGLT‐2 inhibitors with a particular focus on the results of published cardiovascular outcome trials, which have also provided unique opportunities to evaluate uncommon but potentially serious adverse events of these newer agents. The cardiovascular benefits of SGLT‐2 inhibitors and some glucagon‐like peptide‐1 receptor agonists suggest that they may be the preferred choice, usually as an add‐on to metformin, for patients with type 2 diabetes mellitus at high cardiovascular risk. Abstract

In fisheries management, understanding anthropogenic impacts on fish population genetic structure is essential because genetic diversity is a fundamental attribute contributing to a species' evolutionary capacity. An extended history of supplemental stocking has led to the introgression of genes from non-local, hatchery-reared brook trout ( Salvalinus fontinalis) into natural Adirondack populations in the state of New York. Managers have therefore gone to great lengths to protect known or suspected pristine 'heritage' populations, but the genetic integrity of most populations is unknown. We used 11 microsatellite loci to examine a putative, but as yet unconfirmed 'heritage' population in Dix Pond (Essex County, New York), in an effort to confirm its genetic uniqueness, quantify genetic diversity, and determine the geographic extent of the population. No spatial population structure was found within the Dix Pond/Elk Lake watershed, with minimal signs of introgression from historical stocking. The Dix/Elk population showed allelic richness, and effective population size comparable to the highest diversity heritage population among the four that we used for comparison. These patterns support continued heritage status for the Dix Pond population and recognition of the entire Dix-Elk watershed as habitat for this strain. We conclude this study by discussing how the genetic techniques employed here may help to inform future management decisions associated with the conservation and protection of imperiled populations throughout the globe. Abstract

Gulf of Mexico versus Atlantic populations of several coastal species in the southeastern United States are known to differ sharply in genetic composition, but most transitional zones have not previously been examined in detail. Here we employ molecular markers from mitochondrial and nuclear loci to characterize cytonuclear genetic associations at meso- and microgeographic scales along an eastern Florida transitional zone between genetically distinct Atlantic and Gulf populations of the American oyster, Crassostrea virginica. The single- and multilocus cytonuclear patterns display: (1) a cline extending along 340 km of the east Florida coastline; (2) a pronounced step in the cline centered at Cape Canaveral (shifts in allelic frequencies by 50-75% over a 20 km distance); (3) a close agreement of observed genotypic frequencies with Hardy-Weinberg expectations within locales; and (4) mild or nonexistent nuclear and cytonuclear disequilibria in most local population samples. These results imply: (1) considerable restrictions to interpopulational gene flow along the eastern Florida coastline; (2) within locales, free interbreeding (as opposed to mere population admixture) between Gulf and Atlantic forms of oysters; and (3) localized population recruitment in the transition zone localities. These findings demonstrate that marine organisms with high dispersal potential via long-lived pelagic larvae can nonetheless display pronounced spatial population genetic structure, and more generally they exemplify the utility of pronounced genetic transition zones for the study of population level processes.

Morbidity and mortality from diabetes and its complications are increasing in populations globally. Different ethnic groups have varying degrees of risk. The concept of ethnicity encompasses numerous factors relevant to health including genetics, socioeconomics and health behaviours. Ethnicity-related discordance in the glycaemic markers used to diagnose diabetes and to identify those at risk of diabetes has been reported. Furthermore, many ethnicity- and country-specific diabetes risk prediction models have been developed. This review provides a thorough discussion of the impact of ethnicity on how diabetes is detected and the evidence for and against ethnicity-specific approaches to diagnosis.

Abstract

The eastern oyster ( Crassostrea virginica Gmelin) is an economically and ecologically valuable marine bivalve occurring in the Gulf of Mexico. This study builds upon previous research that identified two divergent populations of eastern oysters in the western Gulf of Mexico. Allelic and genotypic patterns from 11 microsatellite markers were used to assess genetic structure and migration between the previously described oyster populations in Texas. The main findings are as follows: (1) there are two distinct populations ( FST = 0.392, P < 0.001) of oysters that overlap in the Corpus Christi/Aransas Bay estuarine complex in Texas, (2) the distribution of genotypes among individuals in the contact zone suggests limited hybridization between populations, (3) the variables of salinity, temperature, dissolved oxygen, turbidity and depth are not correlated with allele frequencies on reefs in the contact zone or when analyzed across Texas, and (4) there is little evidence of directional selection acting on the loci assayed here, although patterns at four markers suggested the influence of balancing selection based on outlier analyses. These results are consistent with long-term historical isolation between populations, followed by secondary contact. Recent hydrological changes in the area of secondary contact may be promoting migration in areas that were previously inhospitable to eastern oysters, and observed differences in the timing of spawning may limit hybridization between populations. Comparison of these findings with the results of an earlier study of oysters in Texas suggests that the secondary contact zone has shifted approximately 27 km north, in as little as a 23-year span. Abstract

Objective Emerging evidence suggests that psychosocial stress may influence weight gain. The relationship between stress and weight change and whether this was influenced by demographic and behavioral factors was explored. Design and Methods A total of 5,118 participants of AusDiab were prospectively followed from 2000 to 2005. The relationship between stress at baseline and BMI change was assessed using linear regression. Results Among those who maintained/gained weight, individuals with high levels of perceived stress at baseline experienced a 0.20 kg/m2 (95% CI: 0.07-0.33) greater mean change in BMI compared with those with low stress. Additionally, individuals who experienced 2 or ≥3 stressful life events had a 0.13 kg/m2 (0.00-0.26) and 0.26 kg/m2 (0.14-0.38) greater increase in BMI compared with people with none. These relationships differed by age, smoking, and baseline BMI. Further, those with multiple sources of stressors were at the greatest risk of weight gain. Conclusion Psychosocial stress, including both perceived stress and life events stress, was positively associated with weight gain but not weight loss. These associations varied by age, smoking, obesity, and multiple sources of stressors. Future treatment and interventions for overweight and obese people should consider the psychosocial factors that may influence weight gain. [ABSTRACT FROM AUTHOR]

Abstract

OBJECTIVE--To determine whether glucose-independent differences in HbA1c exist between people of African, South Asian, and Chinese ethnicities. RESEARCH DESIGN AND METHODS--Data from 6,701 people aged 19-78 years, without known diabetes, from Mauritius, and participating in the population-based Non-Communicable Disease Surveys of the main island and the island of Rodrigues were included. Participants were African (n = 1,219 from main island, n = 1,505 from Rodrigues), South Asian (n = 3,820), and Chinese (n = 157). Survey data included HbA1c, plasma glucose during oral glucose tolerance testing (OGTT), anthropometry, demographics, and medical and lifestyle history. RESULTS--Mean HbA1c, after adjustment for fasting and 2-h plasma glucose and other factors known to influence HbA1c, was higher in Africans from Rodrigues (6.1%) than in South Asians (5.7%, P < 0.001), Chinese (5.7%, P < 0.001), or Africans from the main island of Mauritius (5.7%, P < 0.001). The age-standardized prevalence of diabetes among Africans from Rodrigues differed substantially depending on the diagnostic criteria used [OGTT 7.9% (95% CI 5.8-10.0); HbA1c 17.3% (15.3-19.2)]. Changing diagnostic criteria resulted in no significant change in the prevalence of diabetes within the other ethnic groups. CONCLUSIONS--People of African ethnicity from Rodrigues have higher HbA1c than those of South Asian or African ethnicity from the main island of Mauritius for reasons not explained by plasma glucose during an OGTT or traditional factors known to affect glycemia. Abstract

Unexpectedly strong geographic structures in many cosmopolitan species of marine holoplankton challenge the traditional view of their unrestrained dispersal and presumably high gene flow. We investigated cryptic lineage diversity and comparative phylogeography of a common estuarine copepod, Acartia tonsa, on the US Atlantic coast, using mitochondrial (mtCOI) and nuclear (nITS) gene markers. Three broadly sympatric lineages ( F, S, X) were defined by genealogically concordant clades across both gene trees, strongly supporting recognition as reproductively isolated species. Limited dispersal seems to have had a major role in population differentiation of A. tonsa in general, with gene flow propensities rank ordered X > S > F. Geographic structure was found only at large scales (1000-2000 km) in X and S. Phylogeographic patterns in all three lineages were mostly concordant with previously recognized zoogeographic provinces but a large mid-Atlantic gap in the occurrence of lineage X, coupled with its presence in Europe, suggests possible nonindigenous origins. For lineage F, physiological adaptation to low-salinity environments is likely to have accentuated barriers to gene flow and allopatric differentiation at both regional and continental scales. Three allopatric F sublineages inferred a southern centre of origin and a stepwise northward diversification history at the continental scale. The most recently derived F sublineages, in the mid-Atlantic Bight, showed strong phylogeographic patterns at nITS albeit weaker at mtCOI. Applying a crustacean mtCOI molecular clock suggests that A. tonsa lineages diverged pre-Pleistocene but mid-Atlantic F lineage diversification may be post-Pleistocene. [ABSTRACT FROM AUTHOR]

Abstract

The degree of population structure within species often varies considerably among loci. This makes it difficult to determine whether observed variance reflects neutral-drift stochasticity or locus-specific selection at one or more loci. This uncertainty is exacerbated when evolutionary equilibrium cannot be assumed and/or admixture potentially inflates genomic variance. Thus, the value of a ‘genome scan’, where locus-specific summary statistics are compared with a simulated neutral distribution among loci, may be limited in secondary contact zones if the null distribution is sensitive to the timing of secondary contact. Of particular interest here is the wide variance previously observed in locus-specific patterns of population structure between Atlantic and Gulf of Mexico populations of eastern oyster, Crassostrea virginica. To test the robustness of an equilibrium null model, we compared among-locus distributions of FST simulated under migration-drift equilibrium and several nonequilibrium secondary contact histories. We then tested for evidence of divergent selection between two oyster populations on either side of a secondary contact zone using 215 amplified fragment length polymorphism (AFLP) loci. Constant-migration equilibrium and nonequilibrium secondary-contact simulations produced equivalent distributions of FST when anchored by the global mean FST observed in oysters, 0.0917. The 99th quantile of simulated neutral FST encompassed most of the variation among oyster loci. Three AFLP loci exhibited FST values higher than this threshold. Although no locus was significant after correcting for multiple tests, our results show in geographically clinal organisms: AFLPs can efficiently characterize the genomic distribution of FST; equilibrium models can be used to evaluate outliers; these procedures help focus research on genomic regions of interest. Abstract

Aboriginal and Torres Strait Islander women have high rates of gestational diabetes (GDM). The Pregnancy And Neonatal Diabetes Outcomes in Remote Australia (PANDORA) study is a prospective longitudinal cohort of women with type 2 diabetes (T2D), GDM or normoglycemia in pregnancy in the Northern Territory, Australia. In this analysis we report progression to prediabetes and T2D at 2.5 years [range 2.1, 3] postpartum in a subgroup of Aboriginal and Europid women with GDM and normoglycemia in pregnancy (n=337). Women with pre-existing T2D were excluded. Data were analysed using Fisher's exact tests. Among Aboriginal women with GDM we assessed predictions for progression using multivariate logistic regression. Aboriginal women with GDM (n=111) were younger than Europid women with GDM (n=104) (29 years (SD 5.9) vs. 32 (5.6) p<0.01), with similar first trimester BMI (28.9 kg/m2 (SD 7.2) vs. 28.5 (6.7) p=0.64). Of Aboriginal women with GDM, 24 (22%) progressed to T2D and 12 (11%) to prediabetes. Of Aboriginal women with normoglycemia (n=60), 1 (2%) progressed to T2D and 1 (2%) to prediabetes (p<0.01 for combined outcome vs. GDM women). Of Europid women with GDM, none progressed to T2D and 4 (4%) to prediabetes and of those with normoglycemia (n=62) none progressed to diabetes or prediabetes (p=0.09 vs. GDM women). Among Aboriginal women with GDM, factors associated with postpartum diabetes or prediabetes were age (OR 1.11, 1.03-1.2) and, after adjusting for age: severity of GDM (higher fasting plasma glucose (per 1 mmol/L OR 2.11, 1.23-3.62), use of insulin (OR 3.20, 1.35-7.6)) and higher first trimester BMI (per 3 kg/m2 OR 1.22, 1.01-1.47). Not smoking was protective (OR 0.35, 0.14-0.90), although any breastfeeding at 6-months postpartum was not (OR 0.95, 0.36-2.47). To our knowledge this is the only prospective study of Aboriginal and Torres Strait Islander women with GDM.
