The Palaeoproterozoic Ni–Cu sulphide deposits of the Pechenga Complex, Kola Peninsula, occur in the lower parts of ferropicritic intrusions emplaced into the phyllitic and tuffaceous sedimentary unit of the Pilgujärvi Zone. The intrusive rocks are comagmatic with extrusive ferropicrites of the overlying volcanic formation. Massive lavas and chilled margins from layered flows and intrusions contain <3–7 ng/g Pd and Pt and <0·02–2·0 ng/g Ir, Os and Ru with low Pd/Ir ratios of 5–11. The abundances of platinum group elements (PGE) correlate with each other and with chalcophile elements such as Cu and Ni, and indicate a compatible behaviour during crystallization of the parental magma. Compared with the PGE-depleted central zones of differentiated flows (spinifex and clinopyroxene cumulate zones) the olivine cumulate zones at the base contain elevated PGE abundances up to 10 ng/g Pd and Pt. A similar pattern is displayed in intrusive bodies, such as the Kammikivi sill and the Pilgujärvi intrusion. The olivine cumulates at the base of these bodies contain massive and disseminated Ni–Cu-sulphides with up to 2 μg/g Pd and Pt, but the PGE concentrations in the overlying clinopyroxenites and gabbroic rocks are in many cases below the detection limits. The metal distribution observed in samples closely representing liquid compositions suggests that the parental magma became sulphide saturated during the emplacement and depleted in chalcophile and siderophile metals as a result of fractional segregation of sulphide liquids. Relative sulphide liquid–silicate melt partition coefficients decrease in the order of Ir > Rh > Os > Ru > Pt = Pd > Cu. R-factors (silicate-sulphide mass ratio) are high and of the order of 104–105, and they indicate the segregation of only small amounts of sulphide liquid in the parental ferropicritic magma. In differentiated flows and intrusions the sulphide liquids segregated and accumulated at the base of these bodies, but because of a low silicate–sulphide mass ratio the sulphide liquids had a low PGE tenor and Pt/Ir and Cu/Ir ratios similar to the parental silicate melts. During cooling the sulphide liquid crystallized 40–50% of monosulphide solid solution (mss) and the residual sulphide liquid became enriched in Cu, Pt and Pd and depleted in Ir, Os and Ru. The Cu-rich sulphide liquid locally assimilated components of the surrounding S-rich sediments as suggested by the radiogenic Os isotopic composition of some sulphide ores (γOs > 100). Most of the massive and breccia ores represent mixtures of mss and residual Cu-rich sulphide liquid whereas chalcopyrite-rich veins formed when the Cu-rich sulphide liquids were squeezed out into the surrounding sediments. Enteropathogenic Escherichia coli (EPEC) causes severe diarrhea in young children. Upon infection, EPEC induces the assembly of highly organized pedestal-like actin structures in host epithelial cells. All the EPEC genes that are involved in inducing formation of actin pedestals are located in a unique 35 kbp chromosomal pathogenicity island, termed LEE. These genes include the sep genes that encode components of type III protein secretion system, and genes that encode proteins secreted by this system, the esp genes. This protein secretion system is activated upon contact with the host cell, resulting in increased secretion of Esp proteins. Some of these Esp proteins form the translocation apparatus while others are translocated into the cytoplasm of the host cell. Concerted activity of the LEE genes including the eae, esp and the sep genes is needed to trigger signal transduction in the host cell which results in formation of an actin pedestal. Abstract: The abundance of platinum group alloy (PGA) grains available from some alluvial deposits combined with their great chemical resistance to conventional acid attack generates the need for an alternative method of obtaining routine, rapid yet precise Os isotopic data. Laser ablation multi-collector ICP-MS (LA-MC-ICP-MS) is ideally suited for this purpose and has been applied, in a relatively limited extent, to Os-rich samples by previous workers (e.g., Hirata, T., Michinari Hattori, M., and Tsuyoshi Tanaka, T., 1998. The wide variety of PGA minerals yield a broad spectrum of Pt, Re and Os contents which translate into a wide spread in parent–daughter ratios that is an attractive proposition for geochronology using the Re–Os and Pt–Os decay schemes. Here we show that single PGA grains, can contain sufficient internal Re, Pt and Os elemental variability and coupled variations in parent/daughter and daughter isotope compositions that they present an opportunity to obtain ‘isochron’ ages from two different isotope decay systems. We present a rapid (40 s acquisition time), precise and accurate LA-MC-ICP-MS methodology suitable for applying Pt–Os and Re–Os geochronology approaches to single PGA grains, for use in dating chromitite deposits and identifying and dating multiple sources in alluvial PGA deposits. Because of isobaric interferences from Pt, PGA analyses by LA-MC-ICP-MS are best corrected for instrumental mass fractionation using the 189Os/188Os ratio. We demonstrate that, within our analytical uncertainties, mass bias effects for Os isotope ratios during LA-MC-ICP-MS are similar in nature and magnitude to mass bias variations observed during normal solution-mode analysis. While robust elemental interference corrections can be applied for Re and W on the Os mass spectrum during solution analyses (Nowell, G.M., Luguet, A., Pearson, D.A., Horstwood, M.S.A., 2008-this volume. Precise and accurate 186Os/188Os and 187Os/188Os measurements by Multi-Collector Plasma Ionisation Mass Spectrometry (MC-ICP-MS) part I: solution analyses. Chem Geol.) we show that the accurate correction for the Re interference on Os during laser ablation is problematic at 187Re/188Os ratios >∼0.5 which means that Re-corrected 187Os/188Os ratios for some PGAs and high Re/Os sulfides must be regarded as problematic. Our interfering element correction approach can also be used for the correction for Os on 190Pt for obtaining 190Pt/188Os ratios, removing the need for an independent Pt monitor isotope. Accurate and precise geochronology by LA-MC-ICP-MS can be further limited by poorly quantified fractionation of the parent/daughter elemental ratio during ablation. Although additional work is required to understand the nature and exact extent of inter-element fractionation of the Pt/Os and Re/Os ratios during laser ablation analysis of Pt- and Re-rich PGA grains, our best estimate is that fractionation of the Pt/Os during laser ablation is around 5% or less. We present multi-grain isochrons and single-grain internal ‘isochrons’ for the Re–Os and Pt–Os isotope systems in PGA grains from Lapland. In all cases, the Pt–Os isotope system gives significantly more precise ‘isochrons’ than the Re–Os system. We interpret this discordance between the two systems as being most likely due to problems inherent with Re interference corrections at elevated Re/Os ratios. We show that a suite of Pt-rich PGA grains from the Central Lapland greenstone belt yields a spectrum of internal Pt–Os isochron ages that coincides well with pulses of formation of chromitite-bearing ophiolitic material within this magmatic terrane and we take this to indicate the record of chromitite formation in different magmatic units. The single-grain Pt–Os and Re–Os geochronology technique shows great promise in the dating of chromitite deposits in ophiolites and in the discrimination of multiple source regions within alluvial PGA deposits. Gut microbes shape many aspects of organismal biology, yet how these key bacteria transmit among hosts in natural populations remains poorly understood. Recent work in mammals has emphasized either transmission through social contacts or indirect transmission through environmental contact, but the relative importance of different routes has not been directly assessed. Here we used a novel radio-frequency identification-based tracking system to collect long-term high-resolution data on social relationships, space use and microhabitat in a wild population of mice (Apodemus sylvaticus), while regularly characterizing their gut microbiota with 16S ribosomal RNA profiling. Through probabilistic modelling of the resulting data, we identify positive and statistically distinct signals of social and environmental transmission, captured by social networks and overlap in home ranges, respectively. Strikingly, microorganisms with distinct biological attributes drove these different transmission signals. While the social network effect on microbiota was driven by anaerobic bacteria, the effect of shared space was most influenced by aerotolerant spore-forming bacteria. These findings support the prediction that social contact is important for the transfer of microorganisms with low oxygen tolerance, while those that can tolerate oxygen or form spores may be able to transmit indirectly through the environment. Recent research on the water content of large igneous provinces (LIPs) has revealed that water has a significant impact on the formation of LIPs. However, most studies focus on the water content of mafic-ultramafic rocks, while relatively little attention has been paid to the water content of continental flood basalts (CFB), which form the major part of LIPs and are characterized by huge volumes (> 1 × 10 5 km 3 ) and short eruption times. Here, we determined water contents of clinopyroxene crystals from the Akesu diabase, which is co-genetic with flood basalts of the Tarim LIP in China. Based on these measurements, we obtained a water content of higher than 1.23 ± 0.49 wt.% for the parental magma to the Tarim CFB and a minimum water content of 1230 ± 490 ppm for the mantle source, thus indicating the presence of a hydrous mantle plume. Combined with previous studies, our results suggest that water plays a key role in the formation of the Tarim LIP. Additionally, the whole-rock compositions of the Akesu diabase indicate a contribution of pyroxenite in the mantle source. Age is a key parameter in population ecology, with a myriad of biological processes changing with age as organisms develop in early life then later senesce. As age is often hard to accurately measure with non-lethal methods, epigenetic methods of age estimation (epigenetic clocks) have become a popular tool in animal ecology and are often developed or calibrated using captive animals of known age. However, studies typically rely on invasive blood or tissue samples, which limit their application in more sensitive or elusive species. Moreover, few studies have directly assessed how methylation patterns and epigenetic age estimates compare across environmental contexts (e.g. captive or laboratory-based vs. wild animals). Here, we built a targeted epigenetic clock from laboratory house mice (strain C57BL/6, Mus musculus) using DNA from non-invasive faecal samples, and then used it to estimate age in a population of wild mice (Mus musculus domesticus) of unknown age. This laboratory mouse-derived epigenetic clock accurately predicted adult wild mice to be older than juveniles and showed that wild mice typically increased in epigenetic age over time, but with wide variation in epigenetic ageing rate among individuals. Our results also suggested that, for a given body mass, wild mice had higher methylation across targeted CpG sites than laboratory mice (and consistently higher epigenetic age estimates as a result), even among the smallest, juvenile mice. This suggests wild and laboratory mice may display different CpG methylation levels from very early in life and indicates caution is needed when developing epigenetic clocks on laboratory animals and applying them in the wild., (© 2024 The Authors. The Saiga are migratory antelopes inhabiting the grasslands of Eurasia. Over the last century, Saiga have been pushed to the brink of extinction by mass mortality events and intense poaching. Yet, despite the high profile of the Saiga as an animal of conservation concern, little is known of its biology. In particular, the gut microbiota of Saiga has not been studied, despite its potential importance in health. Here, we characterise the gut microbiota of Saiga from two geographically distinct populations in Kazakhstan and compare it with that of other antelope species. We identified a consistent gut microbial diversity and composition among individuals and across two Saiga populations during a year without die-offs, with over 85% of bacterial genera being common to both populations despite vast geographic separation. We further show that the Saiga gut microbiota resembled that of five other antelopes. The putative causative agent of Saiga mass die-offs, Pasteurella multocida, was not detected in the Saiga microbiota. Our findings provide the first description of the Saiga gut microbiota, generating a baseline for future work investigating the microbiota's role in health and mass die-offs, and supporting the conservation of this critically endangered species., (© 2023. Introduction: GBS may cause a devastating disease in newborns. In early onset disease of the newborn the bacteria are acquired from the colonized mother during delivery. We characterized type VII secretion system (T7SS), exporting small proteins of the WXG100 superfamily, in group B Streptococci (GBS) isolates from pregnant colonized women and newborns with early onset disease (EOD) to better understand T7SS contribution to virulence in these different clinical scenarios., Methods: GBS genomes [N=33, 17 EOD isolates (serotype III/ST17) and 16 colonizing isolates (12 serotype VI/ST1, one serotype VI/ST19, one serotype VI/ST6, and two serotype 3/ST19)] were analyzed for presence of T7SS genes and genes encoding WXG100 proteins. We also perform bioinformatic analysis. Galleria mellonella larvae were used to compare virulence between colonizing, EOD, and mutant EOD isolates. The EOD isolate number 118659 (III/ST17) was used for knocking out the essC gene encoding a membrane-bound ATPase, considered the driver of T7SS., Results: Most GBS T7SS loci encoded core component genes: essC, membrane-embedded proteins (essA; essB), modulators of T7SS activity (esaA; esaB; esaC) and effectors: [esxA (SAG1039); esxB (SAG1030)].Bioinformatic analysis indicated that based on sequence type (ST) the clinicalGBS isolates encode at least three distinct subtypes of T7SS machinery. In all ST1isolates we identified two copies of esxA gene (encoding putative WXG100proteins), when only 23.5% of the ST17 isolates harbored the esxA gene. Five ST17isolates encoded two copies of the essC gene. Orphaned WXG100 molecule(SAG0230), distinct from T7SS locus, were found in all tested strains, except inST17 strains where the locus was found in only 23.5% of the isolates. In ST6 andST19 isolates most of the structure T7SS genes were missing. EOD isolates demonstrated enhanced virulence in G. mellonella modelcompared to colonizing isolates. The 118659DessC strain was attenuated in itskilling ability, and the larvae were more effective in eradicating 118659DessC., Conclusions: We demonstrated that T7SS plays a role during infection. Group A streptococcus (GAS) is a Gram-positive human pathogen that causes invasive infections with mild to life-threatening severity, like toxic shock syndrome, rheumatic heart disease, and necrotizing fasciitis (NF). NF is characterized by a clinical presentation of widespread tissue destruction due to the rapid spread of GAS infection into fascial planes. Despite quick medical interventions, mortality from NF is high. The early onset of the disease is difficult to diagnose because of non-specific clinical symptoms. Moreover, the unavailability of an effective vaccine against GAS warrants a genuine need for alternative treatments against GAS NF. One endoplasmic reticulum stress signaling pathway (PERK pathway) gets triggered in the host upon GAS infection. Bacteria utilize asparagine release as an output of this pathway for its pathogenesis. We reported that the combination of sub-cutaneous (SC) and intraperitoneal (IP) administration of PERK pathway inhibitors (GSK2656157 and ISRIB) cures local as well as systemic GAS infection in a NF murine model, by reducing asparagine release at the infection site. Group A streptococcus (GAS) necrotizing fasciitis (NF) causes high morbidity and mortality despite prompt intravenous administration of antibiotics, surgical soft-tissue debridement, and supportive treatment in the intensive care unit. Since there is no effective vaccine against GAS infections, a comprehensive understanding of NF pathogenesis is required to design more efficient treatments. To increase our understanding of NF pathogenesis, we need a reliable animal model that mirrors, at least in part, the infectious process in humans. Group A streptococcus (GAS) is among the top 10 causes of mortality from an infectious disease, producing mild to invasive life-threatening manifestations. Necrotizing fasciitis (NF) is characterized by a rapid GAS spread into fascial planes followed by extensive tissue destruction. Despite prompt treatments of antibiotic administration and tissue debridement, mortality from NF is still high. Moreover, there is no effective vaccine against GAS, and early diagnosis of NF is problematic because its clinical presentations are not specific. Thus, there is a genuine need for effective treatments against GAS NF. Previously, we reported that GAS induces endoplasmic reticulum (ER) stress to gain asparagine from the host. Here, we demonstrate that GAS-mediated asparagine induction and release occur through the PERK-eIF2α-ATF4 branch of the unfolded protein response. Inhibitors of PERK or integrated stress response (ISR) blocked the formation and release of asparagine by infected mammalian cells, and exogenously added asparagine overcame this inhibition. Moreover, in a murine model of NF, we show that the inhibitors minimized mortality when mice were challenged with a lethal dose of GAS and reduced bacterial counts and lesion size when mice were challenged with a sublethal dose. Immunohistopathology studies demonstrated that PERK/ISR inhibitors protected mice by enabling neutrophil infiltration into GAS-infected fascia and reducing the pro-inflammatory response that causes tissue damage. Inhibitor treatment was also effective in mice when started at 12 hours after infection. Infectious diseases are one of the main grounds of death and disabilities in human beings globally. Lack of effective treatment and immunization for many deadly infectious diseases and emerging drug resistance in pathogens underlines the need to either develop new vaccines or sufficiently improve the effectiveness of currently available drugs and vaccines. In this review, we discuss the application of advanced tools like bioinformatics, genomics, proteomics and associated techniques for a rational vaccine design.

Group A Streptococcus (GAS) causes diverse human diseases, including life-threatening soft-tissue infections. It is accepted that the human antimicrobial peptide LL-37 protects the host by killing GAS. Here, we show that GAS extracellular protease ScpC N-terminally cleaves LL-37 into two fragments of 8 and 29 amino acids, preserving its bactericidal activity. At sub-bactericidal concentrations, the cleavage inhibits LL-37-mediated neutrophil chemotaxis, shortens neutrophil lifespan, and eliminates P2X7 and EGF receptors' activation. Mutations at the LL-37 cleavage site protect the peptide from ScpC-mediated splitting, maintaining all its functions. The mouse LL-37 ortholog CRAMP is neither cleaved by ScpC nor does it activate P2X7 or EGF receptors. Treating wild-type or CRAMP-null mice with sub-bactericidal concentrations of the non-cleavable LL-37 analogs promotes GAS clearance that is abolished by the administration of either P2X7 or EGF receptor antagonists. A curated murine oral microbiome database to be used as a reference for mouse-based studies has been constructed using a combination of bacterial culture, 16S rRNA gene amplicon, and whole-genome sequencing. The database comprises a collection of nearly full-length 16S rRNA gene sequences from cultured isolates and draft genomes from representative taxa collected from a range of sources, including specific-pathogen-free laboratory mice, wild Mus musculus domesticus mice, and formerly wild wood mouse Apodemus sylvaticus At present, it comprises 103 mouse oral taxa (MOT) spanning four phyla- Firmicutes , Proteobacteria , Actinobacteria , and Bacteroidetes -including 12 novel undescribed species-level taxa. The key observations from this study are (i) the low diversity and predominantly culturable nature of the laboratory mouse oral microbiome and (ii) the identification of three major murine-specific oral bacterial lineages, namely, Streptococcus danieliae (MOT10), Lactobacillus murinus (MOT93), and Gemella species 2 (MOT43), which is one of the novel, still-unnamed taxa. Of these, S. danieliae is of particular interest, since it is a major component of the oral microbiome from all strains of healthy and periodontally diseased laboratory mice, as well as being present in wild mice. It is expected that this well-characterized database should be a useful resource for in vitro experimentation and mouse model studies in the field of oral microbiology. IMPORTANCE Mouse model studies are frequently used in oral microbiome research, particularly to investigate diseases such as periodontitis and caries, as well as other related systemic diseases. We have reported here the details of the development of a curated reference database to characterize the oral microbial community in laboratory and some wild mice. The genomic information and findings reported here can help improve the outcomes and accuracy of host-microbe experimental studies that use murine models to understand health and disease.
