To design and evaluate a novel oxyntomodulin (OXM) derivative with albumin-binding helix domain and dual GLP-1 receptor (GLP-1R) and glucagon receptor (GcgR) activation activity to achieve metabolize improvement on the diabetes-related complication.Mutation (D-Ser2) on OXM was performed and then different helix albumin-binding domains were fused to the mutated OXM via a thrombin-cleavable linker to generate seven fusion peptides, named LM01-LM07. Seven LM peptides were synthesized and screened via in vitro receptor activation test, albumin binding measurement and protease cleavage assay to select potent candidate peptide for further in vivo study. Moreover, acute and chronic efficacy studies were conducted to evaluate the efficacy of selected candidate using db/db mice.LM06, as selected OXM derivative, exhibited higher albumin-binding affinity, sustained-release efficiency and balanced activation activities on both GLP-1R and GcgR compared with other ones. Moreover, LM06 was demonstrated with improved hypoglycemic and insulinotropic abilities in receptor-deficient mice via activating GLP-1R. In addition, prolonged anti-diabetic efficacies of LM06 were demonstrated via hypoglycemic duration assay and OGTT in db/db mice. Further pharmacokinetic test of LM06 in both rats and monkeys identified improved half-life and other metabolic characteristics. Nevertheless, 8-week subcutaneously dosed LM06 in db/db mice achieved prominent efficacies on glucostasis, weight-lowering, pancreatic function and adipogenesis via activating GLP-1R and GcgR. Moreover, LM06 also could accelerate diabetic skin wound closure in combination with low-intensity ultrasound.LM06, as a long-acting dual GLP-1R/GcgR agonist, exerts potential as a once-weekly therapeutic candidate against diabetes-related complication in combination with low-intensity ultrasound.

With many hard efforts, the epidemic prevention and control work in China has borne successful, accelerating the gradual restoration of production, living order and routine medical work. However, there is increasing evidence that many patients with COVID-19 are asymptomatic, but they are potential transmitter of the virus. There are difficulties in screening for asymptomatic infections, which makes it more difficult for national prevention and control of this epidemic. Therefore, it is urgent to develop better screening and laboratory testing for asymptomatic infections with COVID-19 with high speed, sensitivity and specificity. This study aimed at investigating the effect of atorvastatin on the expression of TLR4 and NF-κB in brain tissue of stroke rats and its protective effect on brain.Sixty SD rats were selected. The control group (n=20) was raised without treatment. MCAO was used in the experimental group (n=20) and model group (n=20) for stroke modeling. The experimental group was treated with atorvastatin intragastric administration, while the model group received no treatment. The blood lipid level was detected by a full-automatic biochemical analyzer. The expression levels of TLR4 and NF-κB proteins were detected by Western-Blot (WB), and the mRNA expression of NF-κB and TLR4 in brain tissues was detected by real-time quantitative fluorescence (qtPCR). The neurological deficit score was performed.TC, TG, LDL-C in blood lipid indexes of the experimental group and the model group were remarkably higher than those of control group, while HDL-C was remarkably lower than that of control group (p0.05). After treatment, TC, TG, LDL-C in the experimental group gradually decreased, while HDL-C concentration gradually increased (p0.05). The expression of TLR4 and NF-κB protein in the experimental group was significantly lower than that in the model group (p0.01) but significantly higher than that in the control group (p0.01). The expression of TLR4 and NF-κB mRAN in the experimental group was also considerably lower than that in the model group (p0.01) but higher than that in the control group (p0.01). After atorvastatin treatment, the neurological deficit score of the experimental group was remarkably lower than that of the model group (p0.001).Atorvastatin can effectively reduce the expression of TLR4 and NF-κB in brain tissue of stroke rats, and has a certain protective effect on cerebral nerve function, which is expected to be the first choice for stroke treatment in the future.

OBJECTIVE: To explore the predictive value of carotid femoral artery pulse wave velocity (CF-PWV), carotid radial artery pulse wave velocity (CR-PWV), cardio-ankle vascular index (CAVI), and ankle brachial index (ABI) on coronary heart disease (CHD) and cerebral infarction (CI), and the preliminary validation of Beijing vascular health stratification (BVHS). METHODS: Subjects with at least 2 in-patient records were included into the study between 2010 and 2017 from Vascular Medicine Center of Peking University Shougang Hospital. Subjects with CHD or CI, and without data of vascular function at baseline were excluded. Eventually, 467 subjects free of CHD [cohort 1, mean age: (63.4±12.3) years, female 42.2%] and 658 subjects free of CI [cohort 2, mean age: (64.3±12.2) years, female 48.7%] at baseline were included. The first in-patient records were as the baseline data, the second in-patient records were as a following-up data. Cox proportional hazard regression was used to establish the predictive models of CHD or CI derived from BVHS by multivariable-adjusted analysis. RESULTS: The median follow-up time of cohort 1 and cohort 2 was 1.9 years and 2.1 years, respectively. During the follow-up, 164 first CHD events occurred in cohort 1 and 117 first CI events occurred in cohort 2. Four indicators were assessed as continuous variables simultaneously by multivariable-adjusted analysis. The senescence and degeneration of the intervertebral disc are closely related to the reduction of nucleus pulposus (NP) cells caused by apoptosis. TIR-domain-containing adapter-inducing interferon-β (TRIF) is an adapter for Toll-like receptors 3/4 (TLR3/4), which involves in cell apoptosis. The aim of this study is to detect the role of TRIF in the apoptotic progress of NP cells. The expression of collagen II, aggrecan, TLR3/4, and TRIF were analyzed in different degrees of degenerated human NP samples from patients. NP cells were isolated from mild degenerated tissues and cultured with IL-1β to accelerate the degradation, and treated with TLR3/4 protein. siRNA was used to silence TRIF gene expression, and TRLF-plasmid was used to upregulate TRLF gene expression. We used flow cytometry assay to analyze cell apoptosis. The expression of collagen II, aggrecan, TLF3/4, TRIF, caspase-8/3, MMP-13, TNF-α was determined by immunofluorescence, Western blot, or RT-PCR. That the expression of collagen II and aggrecan markedly decreased, but TLF3/4, TRIF, caspase-8/3, MMP-3, TNF-α, and IL-1β were increased in severely degenerated disc tissues. IL-1β treatment induced NP cell degeneration and TLF3/4, TRIF, caspase-8/3, MMP-3, TNF-α overexpression. TLF3/4 protein treatment promoted NP cell degeneration and apoptosis by upregulation of TRIF, caspase-8/3, MMP-3, and TNF-α. Furthermore, TRIF silencing reversed the negative effect of TLF3/4 overexpression, and TRIF overexpression played the same role in NP cell apoptosis. Based on these results, we believe that TRIF is activated in a degenerated intervertebral disc. TLF3/4 promotes NP cell apoptosis and inflammation through the TRLF adaptor. TRLF expression is positively related to the apoptosis and inflammation in NP cells. These results suggest a therapeutic potential of the TRIF in the treatment of disc degeneration.

Flagellin from bacteria elicits a proinflammatory immune response and may act as a vaccine adjuvant. In this study, we evaluated the adjuvant effect of the N-terminus of flagellin (residues 1&ndash, 99) when linked to an antigen (a truncated, conserved domain of lipoprotein E of Pasteurella multocida). Immunization of chickens with the antigen-adjuvant chimeric protein showed that the N-terminus of flagellin accelerated the antibody response and enhanced the cellular immunity (CD8+ T cell expansion). Stimulation of peripheral blood mononuclear cells from vaccinated chickens showed both TH1 (IFN-&gamma, and IL-12) and TH2 (IL-4)-type cytokine gene expressions. In a challenge test, the N-terminus of flagellin increased the survival rate to 75%, compared to 25% in the antigen-only group. In conclusion, our study found that the N-terminus of flagellin can increase the immune response and enhance vaccine protection.

This study aims to identify and describe social and psychosocial factors that may harm the health of low-income adults.DesignParticipatory qualitative research using a problem-posing method based on Freire’s pedagogy.SettingCommunity organisation in a low-income district in the metropolitan area of Montreal, in the province of Quebec, Canada.ParticipantsEight low-income adults using food bank services provided by the community organisation. Participants had to reside in the community organisation’s service area, have insufficient income to cover living expenses, be aged 18 years or older, and be able to participate in group sessions.MethodsEight weekly group sessions (average of 2.5 hours per session) were held using problem-posing method, which included activities such as presentation of representative stories or images and role-playing. Five additional sessions were held for thematic data analysis with participants.ResultsA main factor identified as harming the health of participants was the dissonance between perceived current circumstances and the ideal of circumstances conceived early in life. This dissonance follows a loss or a failure to achieve the ideal of circumstances and generates profound malaise. Other main factors identified were the current circumstances as a social trap and the lack of love and support from an early age.ConclusionsThis study identifies a new psychosocial factor, namely, the dissonance between perceived current circumstances and the ideal of circumstances conceived early in life. Larger studies should examine the influence of this factor on health.

Endoscopic submucosal dissection (ESD) has been established as a standard endoscopic method for treating esophageal superficial neoplasms, and it can be performed using a conventional or a tunneling method. The aim of the present study was to compare the safety and efficacy of tunneling ESD (t-ESD) and standard ESD (s-ESD) for treating large esophageal superficial neoplasms and to explore the risk factors for postoperative strictures.Fifty-five consecutive patients with large esophageal superficial neoplasms were treated by t-ESD or s-ESD. Demographics, lesion characteristics, procedure-related parameters, and follow-up results were retrospectively collected to compare the efficacy and safety of these procedures. Multivariate analyses were conducted to determine the potential risk factors for postoperative strictures.Of the 55 patients, 13 underwent t-ESD and 42 underwent s-ESD. The dissection speed of t-ESD was significantly faster than that of s-ESD (7.42±1.99 min/cm2 vs. 9.01±2.11 min/cm2, P0.05). En bloc resection was achieved in 98.2% (54/55) of the cases, while R0 resection was achieved in 92.7% (51/55). Curative resection was achieved in 78.2% (43/55) of the cases. Fourteen patients (25.5%) had postoperative strictures, which resolved with endoscopic dilation and/or stent insertion. Circumferential involvement of3/4 and lesion length of3 cm were independent risk factors for strictures.T-ESD is a safe and effective method for treating large esophageal superficial neoplasms with a faster dissection speed than s-ESD, but postoperative strictures may be encountered for lesions involving more than three-fourths of the circumference or longer than 3 cm.

Detrital peat (organic carbon-enriched deposit) with high arsenic (As) content is widely distributed in sediments where groundwater As contamination exists. Iron sulfides often persist in these sediments under anoxic conditions. However, the mechanisms and pathways of formation of iron sulfides and its potential contribution in controlling As mobility are still poorly understood. In this study, we examined three As-contaminated peat sediments from the Hetao Basin in China to gain better understanding of the complex interplay between iron sulfides formation and As mobility. We employed high-resolution spectroscopic techniques, including X-ray absorption spectroscopy and 57Fe Mossbauer spectroscopy, coupled with electron microscopy to determine the speciation of iron sulfides and the associated As in the peat sediments. Pyrite (FeS2) and metastable greigite (Fe3S4) persisted in peat as end-members of S and Fe diagenetic pathways. The Fe-rich phyllosilicates and decaying plant tissues provided the ideal micro-environments for pyrite and greigite nucleation. Pyrite formation most likely occurred via the polysulfides pathway in the surface water-sediments interface during early diagenetic process, while the relative enrichment of reactive Fe compared to sulfide possibly inhibited the transformation of greigite to pyrite in such Fe-rich sediments. Our results revealed that the peat sediments could act as a stable sink for As immobilization under steady groundwater anoxic conditions, with As content up to 250 mg/kg and large proportions (40 to 60 wt.% As) sequestered in pyrite and greigite. Pyrite crystallites had up to 1 wt.% As content through the replacement of the S-I sites. Greigite crystallites had a relatively constant As content ranging from ∼500 to ∼1400 mg/kg. Instead of being adsorbed or structurally incorporated, arsenic formed distinct arsenic sulfide phase in the greigite-enriched sediments, which was analogous to realgar. The transfer of As from iron sulfides to ferrihydrite temporarily retarded As release into groundwater under slightly oxic groundwater conditions. However, the reductive dissolution of ferrihydrite and potential subsequent As re-release could be a source of As in groundwater under disturbed redox conditions.

The field of asymmetric organocatalysis has attracted the attention of chemists due to the environmentally benign conditions, particularly for the synthesis of chiral molecules, bioactive compounds, natural products, and drugs. Besides the conventional approaches using a single organocatalyst in asymmetric reactions, dual catalysis with two organocatalysts has emerged as an important strategy for resolving existing challenging problems, including the synthesis of complex molecules, improvement of enantioselectivities, and the development of new catalytic mechanisms. In this review, selected recent examples of the combination of two organocatalysts are covered in detail. Moreover, future perspectives are also described.

Background and Aims: Nonalcoholic steatohepatitis (NASH) is a common cause of chronic liver disease. Clinical trials use the NASH Clinical Research Network (CRN) system for semiquantitative histological assessment of disease severity. Interobserver variability may hamper histological assessment, and diagnostic consensus is not always achieved. We evaluate a second harmonic generation/two-photon excitation fluorescence (SHG/TPEF) imaging-based tool to provide an automated quantitative assessment of histological features pertinent to NASH. Approach and Results: Images were acquired by SHG/TPEF from 219 nonalcoholic fatty liver disease (NAFLD)/NASH liver biopsy samples from seven centers in Asia and Europe. These were used to develop and validate qFIBS, a computational algorithm that quantifies key histological features of NASH. qFIBS was developed based on in silico analysis of selected signature parameters for four cardinal histopathological features, that is, fibrosis (qFibrosis), inflammation (qInflammation), hepatocyte ballooning (qBallooning), and steatosis (qSteatosis), treating each as a continuous rather than categorical variable. OBJECTIVE: Antibodies against carbamylated protein (anti-CarP) were found to be a promising marker to evaluate joint damage and disease activity in patients with rheumatoid arthritis (RA). However, whether anti-CarP antibodies were present in systemic lupus erythematosus (SLE) remained ambiguity. We have therefore undertaken this study to assess the levels of serum anti-CarP antibodies and to evaluate their clinical value in SLE. METHODS: Serum levels of antibodies against carbamylated-fibrinogen (anti-CarP) were measured by enzyme-linked immunosorbent assay (ELISA) in 105 SLE patients and 73 healthy controls. Other clinical and laboratory measurements of the SLE patients were collected from medical records. 1. Paired box (Pax7) gene is a member of the paired box family and plays a critical role in animal growth and muscle development. However, the molecular characterisation of the goose Pax7 gene is unknown. 2. The open-reading frame of goose Pax7 is composed of 1509 bp, which encodes a protein of 503 amino acids and shares high homology with Pax7 of other birds. 3. Ten single-nucleotide polymorphisms were identified in the genomic DNA sequence, 8 located in the intron region and two located in the exon region. 4. Association analysis showed the C122T locus was significantly associated with the body weight of Zhedong-White geese in week 4, 6, 8, 10 and 12. 5. It was concluded that the goose Pax7 gene may be an important candidate gene for goose growth traits and marker-assisted selection.

Four homoleptic copper(II) complexes, [Cu(Meophtpy)2](ClO4)2 (Meophtpy = 4′-(4-methoxylphenyl)- 2,2′:6′,2″-terpyridine) (I), [Cu(Meophtpy)2](ClO4)2 · 2H2O (II), [Cu2(m-Clphtpy)4](ClO4)4 (m-ClPhtpy = 4′-(3-chlorophenyl)-2,2′:6′,2″-terpyridine) (III), and [Cu2(m-ClPhtpy)4](ClO4)4 (IV) have been synthesized by hydrothermal methods and characterized by IR, elemental analysis and single crystal X-ray diffraction (CIF files CCDC nos. 963375 (I), 885457 (II), 963377 (III), and 963376 (IV)). Complex II is a polymorph of I and complex IV is a polymorph of III. All these complexes are obtained with 95% ethanol solution or 50% ethanol solution and the solvent control on the crystallization are obviously found. In all complexes, the face-to-face interactions between pyridyl rings or phenyl rings facilitate the construction of 3D network in the crystal in addition to hydrogen bonds. The fluorescence properties of these complexes have been investigated.

Previous studies have reported that high carbon contents in FeCoCrNiMn high-entropy alloys lead to carbides precipitating from the alloys. Typically, carbides are used to improve the strength of alloys but also lead to decreased ductility. However, the strength and ductility of alloys can be improved when carbides shape, size and distribution are carefully controlled. Therefore, a carbide-containing FeCoCrNiMn alloy with 2 at.-% carbon was prepared by arc melting, and its microstructure and mechanical properties were further tuned by cold rolling with subsequent annealing treatment. The yield strength and uniform elongation of the resultant alloy were excellent, reaching 581 MPa and 25%, respectively, due to the additive combination of various strengthening mechanisms, such as solid-solution hardening, grain-boundary hardening and precipitation hardening.

Aim The cardioprotective effects of sevoflurane postconditioning (SpostC) are eliminated under diabetic conditions, and the underlying mechanism for this phenomenon remains unclear. Many studies have demonstrated that the hypoxia-inducible factor-1 (HIF-1) signalling pathway in the myocardium is impaired under diabetic conditions. This study was to investigate whether deferoxamine (DFO)-induced activation of HIF-1 signalling pathway can restore the cardioprotective effects of SpostC in diabetic rats. Methods A model of myocardial ischaemia/reperfusion (I/R) injury was induced via ligation of the left anterior descending artery. SpostC was conducted by administering 1.0 MAC sevoflurane. After inducing the I/R injury, the following parameters were measured: myocardial infarct size, cardiac function, myocardial ultrastructure, mitochondrial respiratory function, respiratory chain enzyme activity, rate of reactive oxygen species (ROS) generation, and protein expression of HIF-1α, vascular endothelial growth factor (VEGF), cleaved caspase-3, Bcl-2 and Bax. Results After DFO activated HIF-1 in the impaired myocardium of diabetic rats, SpostC significantly upregulated the protein expression of HIF-1α and its downstream mediator VEGF. This improved myocardial mitochondrial respiratory function and respiratory chain enzyme activity and reduced ROS generation as well as the protein expression of cleaved caspase-3 and Bax. As a result, myocardial infarct size decreased, and cardiac function and mitochondrial ultrastructure improved. Conclusion This study demonstrates for the first time that abolishment of the cardioprotective effects of SpostC in diabetic rats is associated with impairment of the HIF-1 signalling pathway and that DFO can activate HIF-1 to restore these cardioprotective effects of SpostC in diabetic rats.

OBJECTIVE: To quantitatively analyze image quality of two sets of phantom (CatPhan504 and Cheese) Megavoltage computed tomography (MVCT) images acquired by Helical Tomotherapy with three scanning modes (Fine, Normal and Coarse), and to explore and validate a semi-automatic quality assurance procedure for MVCT images of Helical Tomotherapy. METHODS: On Helical Tomotherapy, CatPan504 and Cheese phantoms were scanned with three pitch levels (Fine, Normal, Coarse: 4 mm, 8 mm, 12 mm/circle) respectively. Pylinac, Matlab and Eclipse were used to calculate and compare spatial resolution, noise level and low contrast resolution of images obtained under three scanning modes respectively. The spatial resolution can be evaluated by the blurring of line-pair CT value in the images of CatPhan504’s CTP528 module. The noise level can be evaluated by the integral non-uniformity in the images of Cheese’s uniformity module. the low contrast resolution can be evaluated by contrast-to-noise ratio of both phantoms’ plug-in module, or visibility of the region of interest (Supra-Slice) in the images of CatPhan504’s CTP515 module. RESULTS: Analyses on CatPhan504’s line pair module(CTP528 module) showed that the first three line pairs(the gap size are 0.500 cm, 0.250 cm and 0.167 cm respectively) could be clearly observed but blurring began to occur from the fourth line pair(the gap size is 0.125 cm) under Coarse mode. Meanwhile, the first four line pairs were all observable under the Normal and Fine modes. Integral non-integrity index(the value negatively correlated with the noise level) were 0.155 7, 0.136 8 and 0.122 9 for Coarse, Normal and Fine modes respectively. None of the Supra-Slice in CatPhan504’s CTP515 module could be observed under three imaging modes. Low contrast contrast-to-noise ratio of Cheese phantom was similar under three modes and the insert visibility exhibited nearly linear growth with the increasing difference between CT average value of the insert material and background. CONCLUSION: Superiority and inferiority of three image modes in terms of the three image quality index was not consistent. Evaluation results above could provide reference for more rational decision on scanning modes selection of helical tomotherapy, which was based on image visualization demands in clinical practice. The proposed method could also provide guidance for similar image quality assessment and periodic quality assurance.

Nasopharyngeal carcinoma is the most common head and neck tumor in Southern China and Southeast Asia, presenting high rates of local invasion and early distant metastasis. Abnormally expressed miR-99a has been discovered in many tumors, and it is involved in nasopharyngeal carcinoma as well. This study aims to explore the molecular mechanisms of miR-99a and mTOR in regulating nasopharyngeal carcinoma.MiR-99a expression in nasopharyngeal carcinoma cells was detected by quantitative Real Time-Polymerase Chain Reaction (qRT-PCR). MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide) assay was performed for accessing cell proliferative capacity. Dual-Luciferase reporter gene assay was employed to verify the combination between miR-99a and mTOR.We found that miR-99a was downregulated while mTOR was upregulated in nasopharyngeal carcinoma cell lines CNE1 and SUNE1. Low expression of miR-99a or high expression of mTOR predicted poor prognosis of nasopharyngeal carcinoma. MiR-99a overexpression inhibited the proliferation of CNE1 and SUNE1 cells through targeting mTOR.We provided evidence that miR-99a inhibits NPC cell proliferative ability by inhibiting mTOR. The newly identified miR-99a/mTOR axis provides novel insight into the pathogenesis of NPC and represents a potential therapeutic target for NPC.

The China Seismo-Electromagnetic Satellite (CSES) aims to monitor space electromagnetic fields, ionospheric plasma, high-energy charged particles and other features of the global space environment. The high-energy particle package (HEPP), which can effectively detect the energy spectrum, flux and pitch angle distribution of space electrons and protons, and soft X-ray emission from solar flares, is one of the main payloads of CSES. In this study, we designed, developed and calibrated the high-energy particle package and launched it into orbit with CSES. HEPP consists of the high-energy detector (HEPP-H), the low-energy detector (HEPP-L), and the solar X-ray monitor (HEPP-X). The three sub-detectors mainly use silicon detector and crystal calorimeter detection technology. Before launching, we calibrated the three sub-detectors in detail by using radiation source and accelerator beam. All the three sub-detectors have good energy linearity. After launching into orbit, the space energetic particles and X-ray distribution detected by HEPP are consistent with expectations. The performance indices of the detector reach the advanced level of the same kind of detector in the world. HEPP has a wide energy detection range, good energy resolution and high angular resolution ability for electrons and protons. It will play an important role in the study of space particle response and space physics of seismic activity.

Japanese encephalitis (JE) is the important viral encephalitis in China. In the 1940s, JE was confirmed to be epidemic in China. In 1971, the annual incidence rate was 20.92/100 000. Since 2008, JE vaccine was included in the national Expanded Program of Immunization (EPI). In 2013, the incidence of Japanese encephalitis decreased to 0.16/100 000. JE virus is divided into five genotypes, and genotype 1, 3 and 5 JE virus was isolated in China. Genotype 1 JE virus was the mainly genotype currently circulated in China. In recent years, the characteristics of the population of JE have been changed to adult, especially in the northern provinces of China. JE prevention and control faces new challenges.流行性乙型脑炎（乙脑）是我国重要的病毒性脑炎，于20世纪40年代证实乙脑在中国流行，1971年的年发病率20.92/10万，2008年乙脑疫苗纳入国家免疫计划，2013年乙脑发病率降至0.16/10万。乙脑病毒分为5个基因型，中国有基因1、3和5型，目前主要以基因1型乙脑病毒流行为主。近年来，乙脑发病人群特征出现变化，在我国北方地区主要以成人乙脑为主。乙脑防控面临新的挑战。.
