The secretory output of cell-cell interfaces must be tightly controlled in space and time to ensure functional efficacy. This is particularly true for the cytotoxic immune synapse (IS), the stereotyped junction formed between a cytotoxic lymphocyte and the infected or transformed target cell it aims to destroy1. Cytotoxic lymphocytes kill their targets by channeling a mixture of granzyme proteases and the pore forming protein perforin directly into the IS2, 3. The synaptic secretion of these toxic molecules constrains their deleterious effects to the target cell alone, thereby protecting innocent bystander cells in the surrounding tissue from collateral damage. Despite the importance of this process for immune specificity, the molecular and cellular mechanisms that establish secretory sites within the IS remain poorly understood. Here, we identified an essential role for integrin mechanotransduction in cytotoxic secretion using a combination of single cell biophysical measurements, ligand micropatterning, and functional assays. Upon ligand-binding, the αLβ2integrin LFA-1 functioned as a spatial cue, attracting lytic granules containing perforin and granzyme and inducing their fusion at closely adjacent sites within the synaptic membrane. LFA-1 molecules were subjected to pulling forces within these secretory domains, and genetic or pharmacological suppression of these forces abrogated cytotoxicity. We conclude that lymphocytes employ an integrin-dependent mechanical checkpoint to enhance both the potency and the security of their cytotoxic output.

The material damping of rotors causes instability by introducing tangential force to whirl orbit. Literature shows that the improvement of stability range is achieved using fibre-reinforced composites or layered rotors which are light yet strong and exhibit better damping properties. In this study, a multi-layered viscoelastic propeller shaft mounted on three journal bearings is considered. Higher order finite-element model is developed after utilizing operator-based constitutive relationship, which includes various asymmetries from several sources like gyroscopic effect, internal damping, and journal bearing support. A large number of states of the higher order model, as well as the presence of asymmetry, make the system complicated for post-processing. Modified system equivalent reduction expansion process (SEREP) is applied for model reduction that uses both right and left eigenvectors of the original propeller-shaft system. Different numerical results are compared to test the effectiveness of reduced model. Though SEREP is found in the literature, its application over most realistic and higher order asymmetric system is completely new.

Purpose This year, Benchmarking: An International Journal (BIJ) is celebrating its 25th anniversary. The purpose of this paper is to show the historical connections and professional impact of the journal over the years. This is accomplished through a detailed bibliometric analysis (BA) of its content from 1994 to 2018. This paper also identifies leading trends that have affected BIJ during this time frame. Design/methodology/approach In this paper, the authors employ BA on data retrieved from the Scopus database. The authors used various indicators such as the most productive authors, institutions and countries, as well as the most cited papers, citing journals and articles most cited by BIJ between 1994 and 2018. Additionally, the authors have used the Gephi, VOSviewer software to map journals, keywords and institutions with bibliographic coupling and co-citation analysis. Findings The journal has improved its impact, productivity and reputation in the period under review. Articles published in BIJ have been cited in other journals more than 15,000 times. The most prominent themes among the articles published in BIJ are benchmarking, supply chain management and performance improvement. Significant contributions have come from authors and institutions around the globe. Co-authorships, though global, have centered around India, the USA and the UK. There is also a thematic similarity between articles from India and Thailand, and the USA and Germany, indicating high academic collaboration or prevalence of similar issues. Practical implications This paper provides evidence of the impact of BIJ, and in so doing, it helps authors to decide on the appropriateness of this publication for their research. The reader will also understand leading trends occurring in BIJ over the history of the journal. Originality/value This is the first paper that traces the progress of BIJ covering a time span of 25 years.

Environmental conditions can influence biological characteristics, such as phenology and body size, with important consequences for organismal fitness. Examining these fitness consequences under natural conditions through genetic pedigree reconstruction offers a lens into potential population responses to changing environments. Over 3 years (2013–2015), we introduced adult alewife (Alosa pseudoharengus), anadromous, iteroparous clupeids, into one Massachusetts (USA) lake to complete the first detailed examination of this species’ mating system and assess relationships among body size, reproductive timing, and seasonal reproductive success. We reconstructed pedigrees using 15 microsatellites and genotypes from all possible parents and samples of naturally produced offspring within 4 months of hatching. In each of the 3 study years, spawning adults had multiple mates and spawned multiple times. Larger females that arrived and were translocated earlier had higher reproductive success. Declining body size and altered migration timing, through an influence on reproductive success, can influence population vital rates and productivity over time.

Low-frequency noise attenuation and normalisation are fundamental signal processing (SP) methods for surface electromyography (sEMG), but are absent, or not consistently applied, in equine biomechanics. The purpose of this study was to examine the effect of different band-pass filtering and normalisation conventions on sensitivity for identifying differences in sEMG amplitude-related measures, calculated from leading (LdH) and trailing hindlimb (TrH) during canter, where between-limb differences in vertical loading are known. sEMG and 3D-kinematic data were collected from the right Biceps Femoris in 10 horses during both canter leads. Peak hip and stifle joint angle and angular velocity were calculated during stance to verify between-limb biomechanical differences. Four SP methods, with and without normalisation and high-pass filtering, were applied to raw sEMG data. Methods 1 (M1) to 4 (M4) included DC-offset removal and full-wave rectification. Method 2 (M2) included additional normalisation relative to maximum sEMG across all strides. Method 3 (M3) included additional high-pass filtering (Butterworth 4th order, 40 Hz cut-off), for artefact attenuation. M4 included the addition of high-pass filtering and normalisation. Integrated EMG (iEMG) and average rectified value (ARV) were calculated using processed sEMG data from M1 – M4, with stride duration as the temporal domain. sEMG parameters, within M1 – M4, and kinematic parameters were grouped by LdH and TrH and compared using repeated measures ANOVA. Significant between-limb differences for hip and stifle joint kinematics were found, indicating functional differences in hindlimb movement. Regulated water level fluctuations alter the physical, chemical, and biological environments in lakes. However, few studies have measured the effects of repeated annual winter drawdowns on freshwater mussel populations (Bivalvia: Unionida), and it is unknown whether drawdowns permanently constrain mussel populations to deeper depths or are resilient to the annual disturbance. We quantified mussel densities during normal water levels and their mortality after the initiation of drawdown in lakes with annual winter (December–March) water level drawdown regimes that have existed for several decades. We used systematic quadrat sampling and visual snorkel surveys to estimate the number of live mussels at the surface and buried, and shell length in six drawdown and three control lakes at exposed (0.5‐m) and unexposed (1‐m) depths during winter drawdown. We also estimated mortality of mussels along the exposed lakebed for nine drawdown lakes. Study lakes were dominated by Elliptio complanata and Pyganodon cataracta. During normal water levels, mussel densities were much lower in drawdown lakes than control lakes at 0.5‐m depths. When mussels were present in drawdown lakes at 0.5‐m depths, they were often burrowed in the substrate and smaller than buried mussels in control lakes. At unexposed 1‐m depths, mussel densities were primarily explained by geographic region (western versus central Massachusetts), and found higher densities in drawdown lakes compared to control lakes. Mussel mortality after a single drawdown event in nine drawdown lakes was not correlated with drawdown magnitude or rate. Concordance of shell length‐frequency distributions between dead and buried living mussels in drawdown lakes suggests annual mortality of colonising individuals, which are typically small and presumably of young cohorts. Annual winter drawdowns still limit densities of E. complanata and P. cataracta in shallow‐water habitats in the following autumn (September–October) after 4–5 months of potential recovery, and hence constrain their distribution in lakes exposed to drawdowns. The development of sustainable water level management practices will be essential to minimise impacts to native mussel populations.

TrackMate is an automated tracking software used to analyze bioimages and distributed as a Fiji plugin. Here we introduce a new version of TrackMate rewritten to improve performance and usability, and integrating several popular machine and deep learning algorithms to improve versatility. We illustrate how these new components can be used to efficiently track objects from brightfield and fluorescence microscopy images across a wide range of bio-imaging experiments.

We agree with Che et al. (1) that understanding how mobile genetic elements (MGEs) spread antimicrobial resistance (AMR) is important. However, decades of research have already characterized a diverse toolbox of MGEs involved in the emergence of AMR, including conjugative plasmids and insertion sequences (ISs), and their interactions. “Mobile” AMR generally arises following rare capture of a chromosomal gene by a particular “intracellularly mobile” MGE, e.g., an IS or a transposon (Tn), and translocation to “intercellularly mobile” MGEs [plasmids, phage, integrative elements (2, 3)]. Tools for systematic analysis of AMR gene−MGE interactions in mounting sequence data are needed, but incorporating existing biological knowledge into their design is vital, as are representative benchmarking datasets and recognition of data biases and resulting limitations. Che et al. screened bacterial plasmid sequences for markers to classify them as nonmobilizable (no relaxase), mobilizable (relaxase), or conjugative (relaxase+). Relying on the absence of a predicted feature is risky, and Plascad’s poor sensitivity in many species was not recognized, due to a taxonomically inadequate benchmark dataset. Summary The parasite Cryptosporidium invades and replicates in intestinal epithelial cells and is a leading cause of diarrheal disease and early childhood mortality. The molecular mechanisms that underlie infection and pathogenesis are largely unknown. Here, we delineate the events of host cell invasion and uncover a mechanism unique to Cryptosporidium. We developed a screen to identify parasite effectors, finding the injection of multiple parasite proteins into the host from the rhoptry organelle. These factors are targeted to diverse locations within the host cell and its interface with the parasite. One identified effector, rhoptry protein 1 (ROP1), accumulates in the terminal web of enterocytes through direct interaction with the host protein LIM domain only 7 (LMO7) an organizer of epithelial cell polarity and cell-cell adhesion. Genetic ablation of LMO7 or ROP1 in mice or parasites, respectively, impacts parasite burden in vivo in opposite ways. Taken together, these data provide molecular insight into how Cryptosporidium manipulates its intestinal host niche.

The fate of technogenic sulfur released from a sour gas plant in Alberta (Canada) in the surrounding pedosphere and hydrosphere was assessed by chemical and isotopic techniques. The key to the successful use of stable sulfur isotopes in the case study was that sulfur emissions from the gas plant have δ34S-values near +22 %0, while δ34S-values near 0 %o found in several soil, groundwater, and surface water samples were assumed to represent preindustrial background values. Sulfur isotope analyses for total soil sulfur and groundwater sulfate indicated clearly increasing admixture of technogenic S with proximity to the emission source. In some groundwater and surface water samples, however, evidence for bacterial sulfate reduction was found. Since both the admixture of industrial sulfur and dissimilatory sulfate reduction cause increased δ34S-values for aqueous sulfate, it can be difficult to delineate the relative importance of both processes without using other chemical parameters.

Different measures of antimicrobial are used to ensure proper usage, as part of structured antimicrobial stewardship programs.The main objective of this study is to describe the consumption of antibiotics in a mother-child teaching hospital using the AWaRe classification over a period of 15 years.This descriptive and retrospective study included all the data on antibiotic consumption of patients admitted to mother-child University Hospital from April 1st 2005 to March 31st, 2021. Of the 180 international non-proprietary names of antibiotics proposed by the World Health Organization (WHO), 54 were used. The number of days of treatment (DOT) per 1000 patient-days (PD) was calculated per year, per patientele (e.g. surgery, pediatrics, oncology, intensive care, neonatal intensive care, nursery, psychiatry, rehabilitation and obstetrics and gynecology) and globally. The data was classified per the WHO AWaRe classification.A total of 10,489 DOT/1000PD were used globally in 15 years, corresponding to 53.9% of the "Access" group, 45.2% of the "Watch" group and 0.9% of the "Reserve" group. A reduction in the use of antibiotics was noted. The ratios of antibiotic consumptions in DOT/1000PD of 2020-2021 to 2005-2006 were 0.84 globally (1713/5653), 0.79 for the "Access" group (286/361), 0.89 for the "Watch" group (270/302) and 0.88 for the "Reserve" group (2.1/2.4).The AWaRe classification was used to describe the consumption of antibiotics in a mother-child center. Our consumption compared favorably with international data and is in line with the WHO targets. This position may be linked to the presence of a structured anti-biogovernance program that has been in place for several years within our hospital. More work is needed to more precisely identify the antibiotics that should benefit from better supervision.

PurposeThis study aimed to evaluate a novel communication system designed to translate surface electromyographic (sEMG) signals from articulatory muscles into speech using a personalized, digital voice. The system was evaluated for word recognition, prosodic classification, and listener perception of synthesized speech.MethodsEMG signals were recorded from the face and neck as speakers with (n = 4) and without (n = 4) laryngectomy subvocally recited (silently mouthed) a speech corpus comprising 750 phrases (150 phrases with variable phrase-level stress). Corpus tokens were then translated into speech via personalized voice synthesis (n = 8 synthetic voices) and compared against phrases produced by each speaker when using their typical mode of communication (n = 4 natural voices,n = 4 electrolaryngeal [EL] voices). Naïve listeners (n = 12) evaluated synthetic, natural, and EL speech for acceptability and intelligibility in a visual sort-and-rate task, as well as phrasal stress discriminability via a classification mechanism.ResultsRecorded sEMG signals were processed to translate sEMG muscle activity into lexical content and categorize variations in phrase-level stress, achieving a mean accuracy of 96.3% (SD = 3.10%) and 91.2% (SD = 4.46%), respectively. Synthetic speech was significantly higher in acceptability and intelligibility than EL speech, also leading to greater phrasal stress classification accuracy, whereas natural speech was rated as the most acceptable and intelligible, with the greatest phrasal stress classification accuracy.ConclusionThis proof-of-concept study establishes the feasibility of using subvocal sEMG-based alternative communication not only for lexical recognition but also for prosodic communication in healthy individuals, as well as those living with vocal impairments and residual articulatory function.Supplemental Materialhttps://doi.org/10.23641/asha.14558481

Robertson et al. show that unlike T cells responding to conventional chemokines, T cells responding to the lipid chemoattractant S1P utilize a bleb-based mode of motility. This response involves myosin-induced increases in intracellular pressure and is directed by the cortical cytoskeletal proteins ezrin and moesin., Ezrin, radixin, and moesin (ERM) family proteins regulate cytoskeletal responses by tethering the plasma membrane to the underlying actin cortex. Mutations in ERM proteins lead to severe combined immunodeficiency, but the function of these proteins in T cells remains poorly defined. Using mice in which T cells lack all ERM proteins, we demonstrate a selective role for these proteins in facilitating S1P-dependent egress from lymphoid organs. ERM-deficient T cells display defective S1P-induced migration in vitro, despite normal responses to standard protein chemokines. Analysis of these defects revealed that S1P promotes a fundamentally different mode of migration than chemokines, characterized by intracellular pressurization and bleb-based motility. ERM proteins facilitate this process, controlling directional migration by limiting blebbing to the leading edge. We propose that the distinct modes of motility induced by S1P and chemokines are specialized to allow T cell migration across lymphatic barriers and through tissue stroma, respectively.

Selection and training practices for jumping horses have not yet been validated using objective performance analyses. This study aimed to quantify the differences and relationships between movement and muscle activation strategies in horses with varying jump technique to identify objective jumping performance indicators. Surface electromyography (sEMG) and three-dimensional kinematic data were collected from horses executing a submaximal jump. Kinematic variables were calculated based on equestrian-derived performance indicators relating to impulsion, engagement and joint articulation. Horses were grouped using an objective performance indicator—center of mass (CM) elevation during jump suspension (ZCM). Between-group differences in kinematic variables and muscle activation timings, calculated from sEMG data, were analyzed using one-way ANOVA. Statistical parametric mapping (SPM) evaluated between-group differences in time and amplitude-normalized sEMG waveforms. Relationships between movement and muscle activation were evaluated using Pearson correlation coefficients. Horses with the greatest ZCM displayed significantly (p &lt, 0.05) shorter gluteal contractions at take-off, which were significantly correlated (p &lt, 0.05) with a faster approach and more rapid hindlimb shortening and CM vertical displacement and velocity, as well as shorter hindlimb stance duration at take-off. Findings provide objective support for prioritizing equestrian-derived performance indicators related to the generation of engagement, impulsion and hindlimb muscle power when selecting or training jumping horses.

A rod-shaped, motile anaerobic bacterium, designated CCRI-22567T, was isolated from a vaginal sample of a woman diagnosed with bacterial vaginosis and subjected to a polyphasic taxonomic study. The novel strain was capable of growth at 30–42 °C (optimum, 42 °C), at pH 5.5–8.5 (optimum, pH 7.0–7.5) and in the presence of 0–1.5 % (w/v) NaCl (optimally at 0.5 % NaCl). The phylogenetic trees based on 16S rRNA gene sequences showed that strain CCRI-22567T forms a distinct evolutionary lineage independent of other taxa in the family Peptostreptococcaceae . Strain CCRI-22567T exhibited 90.1 % 16S rRNA gene sequence similarity to Peptoanaerobacter stomatis ACC19aT and 89.7 % to Eubacterium yurii subsp. schtitka ATCC 43716. The three closest organisms with an available whole genome were compared to strain CCRI-22567T for genomic relatedness assessment. The genomic average nucleotide identities (OrthoANIu) obtained with Peptoanaerobacter stomatis ACC19aT, Eubacterium yurii subsp. margaretiae ATCC 43715 and Filifactor alocis ATCC 35896T were 71.8, 70.3 and 69.6 %, respectively. Strain CCRI-22567T contained C18 : 1 ω9c and C18 : 1 ω9c DMA as the major fatty acids. The DNA G+C content of strain CCRI-22567T based on its genome sequence was 33.8 mol%. On the basis of the phylogenetic, chemotaxonomic and other phenotypic properties, strain CCRI-22567T is considered to represent a new genus and species within the family Peptostreptococcaceae , for which the name Criibacterium bergeronii gen. nov., sp. nov., is proposed. The type strain of Criibacterium bergeronii is CCRI-22567T (=LMG 31278T=DSM 107614T=CCUG 72594T).

Event Horizon Telescope (EHT) observations at 230 GHz have now imaged polarized emission around the supermassive black hole in M87 on event-horizon scales. This polarized synchrotron radiation probes the structure of magnetic fields and the plasma properties near the black hole. Here we compare the resolved polarization structure observed by the EHT, along with simultaneous unresolved observations with the Atacama Large Millimeter/submillimeter Array, to expectations from theoretical models. The low fractional linear polarization in the resolved image suggests that the polarization is scrambled on scales smaller than the EHT beam, which we attribute to Faraday rotation internal to the emission region. We estimate the average density n e ∼ 104-7 cm-3, magnetic field strength B ∼ 1-30 G, and electron temperature T e ∼ (1-12) 1010 K of the radiating plasma in a simple one-zone emission model. We show that the net azimuthal linear polarization pattern may result from organized, poloidal magnetic fields in the emission region. In a quantitative comparison with a large library of simulated polarimetric images from general relativistic magnetohydrodynamic (GRMHD) simulations, we identify a subset of physical models that can explain critical features of the polarimetric EHT observations while producing a relativistic jet of sufficient power. The consistent GRMHD models are all of magnetically arrested accretion disks, where near-horizon magnetic fields are dynamically important. We use the models to infer a mass accretion rate onto the black hole in M87 of (3-20) 10-4 M o˙ yr-1. © 2021. The Author(s). Published by the American Astronomical Society..

In 2017, the Event Horizon Telescope (EHT) Collaboration succeeded in capturing the first direct image of the center of the M87 galaxy. The asymmetric ring morphology and size are consistent with theoretical expectations for a weakly accreting supermassive black hole of mass ∼6.5 × 109 M o˙. The EHTC also partnered with several international facilities in space and on the ground, to arrange an extensive, quasi-simultaneous multi-wavelength campaign. This Letter presents the results and analysis of this campaign, as well as the multi-wavelength data as a legacy data repository. We captured M87 in a historically low state, and the core flux dominates over HST-1 at high energies, making it possible to combine core flux constraints with the more spatially precise very long baseline interferometry data. We present the most complete simultaneous multi-wavelength spectrum of the active nucleus to date, and discuss the complexity and caveats of combining data from different spatial scales into one broadband spectrum. We apply two heuristic, isotropic leptonic single-zone models to provide insight into the basic source properties, but conclude that a structured jet is necessary to explain M87's spectrum. We can exclude that the simultaneous γ-ray emission is produced via inverse Compton emission in the same region producing the EHT mm-band emission, and further conclude that the γ-rays can only be produced in the inner jets (inward of HST-1) if there are strongly particle-dominated regions. Our understanding of strong gravity near supermassive compact objects has recently improved thanks to the measurements made by the Event Horizon Telescope (EHT). We use here the M87∗ shadow size to infer constraints on the physical charges of a large variety of nonrotating or rotating black holes. For example, we show that the quality of the measurements is already sufficient to rule out that M87∗ is a highly charged dilaton black hole. Similarly, when considering black holes with two physical and independent charges, we are able to exclude considerable regions of the space of parameters for the doubly-charged dilaton and the Sen black holes. © 2021 authors. Very-long-baseline interferometry (VLBI) observations of active galactic nuclei at millimetre wavelengths have the power to reveal the launching and initial collimation region of extragalactic radio jets, down to 10–100 gravitational radii (rg ≡ GM/c2) scales in nearby sources1. Centaurus A is the closest radio-loud source to Earth2. It bridges the gap in mass and accretion rate between the supermassive black holes (SMBHs) in Messier 87 and our Galactic Centre. A large southern declination of −43° has, however, prevented VLBI imaging of Centaurus A below a wavelength of 1 cm thus far. Here we show the millimetre VLBI image of the source, which we obtained with the Event Horizon Telescope at 228 GHz. Compared with previous observations3, we image the jet of Centaurus A at a tenfold higher frequency and sixteen times sharper resolution and thereby probe sub-lightday structures. We reveal a highly collimated, asymmetrically edge-brightened jet as well as the fainter counterjet. We find that the source structure of Centaurus A resembles the jet in Messier 87 on ~500 rg scales remarkably well. Furthermore, we identify the location of Centaurus A’s SMBH with respect to its resolved jet core at a wavelength of 1.3 mm and conclude that the source’s event horizon shadow4 should be visible at terahertz frequencies. This location further supports the universal scale invariance of black holes over a wide range of masses5,6. © 2021, The Author(s).

Emulsions are thermodynamically unstable systems, since they will separate to reduce the interfacial area between the oil phase and the water phase, as a function of time. As a metastable system, surfactant molecules, amphiphilic polymers or solid particles must be present before a stable emulsion system is formed. These components of an emulsion system are called emulsifiers. The relative balance of the hydrophilic and lipophilic properties of these emulsifiers is known to be the most important parameter dictating the emulsion type, whether an oil-in-water (o/w) or water-in-oil (w/o) emulsion. Irrespective of the emulsion type formed, demulsification is a costly exercise in the oil and gas industry. This chapter describes the fundamental role played by surface active agents (surfactants) as integral components of a chemical demulsifier.

We present the results from a full polarization study carried out with the Atacama Large Millimeter/submillimeter Array (ALMA) during the first Very Long Baseline Interferometry (VLBI) campaign, which was conducted in 2017 April in the λ3 mm and λ1.3 mm bands, in concert with the Global mm-VLBI Array (GMVA) and the Event Horizon Telescope (EHT), respectively. We determine the polarization and Faraday properties of all VLBI targets, including Sgr A∗, M87, and a dozen radio-loud active galactic nuclei (AGNs), in the two bands at several epochs in a time window of 10 days. We detect high linear polarization fractions (2%-15%) and large rotation measures (RM > 103.3-105.5 rad m-2), confirming the trends of previous AGN studies at millimeter wavelengths. We find that blazars are more strongly polarized than other AGNs in the sample, while exhibiting (on average) order-of-magnitude lower RM values, consistent with the AGN viewing angle unification scheme. For Sgr A∗ we report a mean RM of (-4.2 0.3) 105 rad m-2 at 1.3 mm, consistent with measurements over the past decade and, for the first time, an RM of (-2.1 0.1) 105 rad m-2 at 3 mm, suggesting that about half of the Faraday rotation at 1.3 mm may occur between the 3 mm photosphere and the 1.3 mm source. We also report the first unambiguous measurement of RM toward the M87 nucleus at millimeter wavelengths, which undergoes significant changes in magnitude and sign reversals on a one year timescale, spanning the range from -1.2 to 0.3 105 rad m-2 at 3 mm and -4.1 to 1.5 105 rad m-2 at 1.3 mm. Given this time variability, we argue that, unlike the case of Sgr A∗, the RM in M87 does not provide an accurate estimate of the mass accretion rate onto the black hole. We put forward a two-component model, comprised of a variable compact region and a static extended region, that can simultaneously explain the polarimetric properties observed by both the EHT (on horizon scales) and ALMA (which observes the combined emission from both components). These measurements provide critical constraints for the calibration, analysis, and interpretation of simultaneously obtained VLBI data with the EHT and GMVA. © 2021. The American Astronomical Society.
