Carbapenem-resistant Salmonella has recently aroused increasing attention. In this study, a total of four sequence type 36 Salmonella enterica subsp. enterica serovar Typhimurium (S. Typhimurium) isolates were consecutively isolated from an 11-month-old female patient with a gastrointestinal infection, of which one was sensitive to carbapenems and three were resistant to carbapenems. Via antibiotic susceptibility testing, a carbapenemases screening test, plasmid conjugation experiments, Illumina short-reads, and PacBio HiFi sequencing, we found that all four S. Typhimurium isolates contained a blaCTX-M-14-positive IncI1 plasmid. One carbapenem-sensitive S. Typhimurium isolate then obtained an IncHI2 plasmid carrying blaNDM-1 and an IncP plasmid without any resistance genes during the disease progression. The blaNDM-1 gene was located on a new 30 kb multiple drug resistance region, which is flanked by IS26 and TnAs2, respectively. In addition, the ST_F0903R isolate contained eight tandem copies of the ISCR1 unit (ISCR1-dsbD-trpF-ble-blaNDM-1-ISAba125Δ1), but an increase in MICs to carbapenems was not observed. Our work further provided evidence of the rapid spread and amplification of blaNDM-1 through plasmid. Prompting the recognition of carbapenem-resistant Enterobacterales and the initiation of appropriate infection control measures are essential to avoid the spread of these organisms. Stable carbon isotopes (δ13C) and elemental stoichiometry characteristics are important ways to research the water and nutrient use strategies of plants. Investigating the variation patterns inof δ13C and the major nutrient elements in different organs of plants and the correlation among them can reveal the ecological strategies of desert plants in extreme arid environments. In this study, two typical desert plants, Alhagi sparsifolia and Karelinia caspia, were studied in the Tarim Basin. By analyzing the changes in δ13C, carbon (C), nitrogen (N), and phosphorus (P) and the ecological stoichiometry of their roots, stems, and leaves, the distribution patterns among different organs and their correlation with soil environmental factors were revealed. The results showed the following: (1) The δ13C of the two plants differed significantly among different organs (p < 0.01). The root and stem of Alhagi sparsifolia had significantly greater δ13C than the leave, while the δ13C of Karelinia caspia showed a root > stem > leaf gradient; (2) the C content in the leaves of the two plants was significantly lower than that of the root (p < 0.01), whereas the N content showed the opposite trend (p < 0.01); (3) the average N:P of Alhagi sparsifolia was >16.00, indicating it was mainly limited by P elements, while the average N:P of Karelinia caspia was <14.00, suggesting it was mainly limited by N elements; (4) in the root, stem and leave of Alhagi sparsifolia and Karelinia caspia, the N content and C:N and the P content and C:P showed a significantly negative correlation (p < 0.01), and δ13C was negatively correlated with C:P; (5) soil total phosphorus (TP) is an important soil environmental factor affecting δ13C and the nutrient elements in Alhagi sparsifolia and Karelinia caspia. This study demonstrates that Alhagi sparsifolia and Karelinia caspia are able to effectively coordinate and regulate their water, N, and P use strategies in response to environmental stress. Background: Duchenne muscular dystrophy (DMD) and Becker muscular dystrophy (BMD) are X-linked recessive neuromuscular diseases resulting from dystrophin (DMD) gene mutations. It has been known that the carrier of DMD mutations may also have symptoms of the disease. While de novo mutation is quite common in BMD/DMD patients, it is rarely reported in the female carriers. Methods: Two sporadic Chinese patients with progressive muscular dystrophy and their familial members were recruited. The targeted next-generation sequencing (NGS) and the multiplex ligation-dependent probe analysis (MLPA) were performed in the proband. Blood tests, electrocardiography, echocardiography, and electromyography were also evaluated. Results: Two novel mutations of DMD gene were identified, c.7318C>T (p.Q2440*) in the male proband and c.4983dupA (p.A1662Sfs*24) in the female carrier. The MLPA analysis did not detect any large rearrangements. The haplotype analysis indicated that the two mutations were derived from de novo mutagenesis. Conclusions: We identified two novel de novo mutations of DMD gene in two Chinese pedigrees, one of which caused a female patient with muscular dystrophy. The mutational analysis is important for DMD patients and carriers in the absence of a family history. The NGS can help detect the mutations in MLPA-negative patients.

Background: Paroxysmal kinesigenic dyskinesia (PKD) is the most common subtype of paroxysmal dyskinesias and is caused by mutations in PRRT2 gene. The majority of familial PKD was identified to harbor PRRT2 mutations. However, over two-third of sporadic PKD patients did not carry anyPRRT2 mutation, suggesting an existence of additional genetic mutations or possible misdiagnosis due to clinical overlap. Methods: A cohort of 28 Chinese patients clinically diagnosed with sporadic PKD and excluded PRRT2 mutations were recruited. Clinical features were evaluated, and all subjects were screened for MR-1, SLC2A1, and CLCN1 genes, which are the causative genes of paroxysmal nonkinesigenic dyskinesia (PNKD), paroxysmal exertion-induced dyskinesia, and myotonia congenita (MC), respectively. In addition, 200 genetically matched healthy individuals were recruited as controls. Results: A total of 16 genetic variants including 4 in MR-1 gene, 8 in SLC2A1 gene, and 4 in CLCN1 gene were detected. Among them, SLC2A1 c.363G>A mutation was detected in one case, and CLCN1 c.1205C>T mutation was detected in other two cases. Neither of them was found in 200 controls as well as 1000 Genomes database and ExAC database. Both mutations were predicted to be pathogenic by SIFT and PolyPhen2. The SLC2A1 c.363G>A mutation was novel. Conclusions: The phenotypic overlap may lead to the difficulty in distinguishing PKD from PNKD and MC. For those PRRT2- negative PKD cases, screening of SLC2A1 and CLCN1 genes are useful in confirming the diagnosis.

Clostridium perfringens causes necrotic enteritis (NE) after proliferation in the intestine of poultry, resulting in considerable losses to the poultry industry. This study aimed to investigate the impact of tannic acid on the antioxidant, immunity, and gut health of broilers with NE. In the experiment, 630 one-day-old Cobb500 male chicks were randomly divided into six treatment groups, with seven replicate cages and with fifteen birds in each cage. The treatment groups were as follows: control group (NC), challenged group (PC), and challenged NE chickens treated with 250, 500, 750, and 1000 mg/kg tannic acid (PTA1, PTA2, PTA3, and PTA4, respectively). To induce NE, coccidia vaccine and Clostridium perfringens were administered on day 19 and days 22–28, respectively. Indexes related to antioxidant, immune, and intestinal health were measured on days 28 and 35. During the infection period, we observed significant increases in fecal water content, D-LA, TNF-α, and malondialdehyde concentrations (p < 0.05). Conversely, significant decreases were noted in chyme pH and in T-AOC, IL-4, and IL-10 concentrations (p < 0.05). The addition of tannic acid exhibited a linear decrease in fecal water content and TNF-α concentration (p < 0.05). Furthermore, tannic acid supplementation resulted in a quadratic curve decrease in D-LA concentration and linear increases in T-AOC, IL-4, and IL-10 (p < 0.05). Cecal microbiological analysis revealed that Ruminococcaceae and Butyricimona were dominant in PTA3. In conclusion, the dietary addition of tannic acid may reduce the negative effects of NE by increasing antioxidant and anti-inflammatory capacity, improving the intestinal barrier, and regulating the intestinal flora. In this work, 3Mg(OH)2·MgCl2·8H2O (318MHCH) nanowires with ultra-high aspect ratios were first synthesized by a liquid method, and the effects of cetyltrimethylammonium bromide (CTAB) in highly concentrated salt solution were investigated. The structure and morphology of the synthesized 318MHCH nanowires were characterized. The as-prepared 318MHCH nanowires have a typical triclinic structure and relatively good crystallization properties. By introducing CTAB, the aspect ratio of 318MHCH nanowires will significantly increase from 500 to 4000. The high aspect ratio of 318MHCH nanowires is due to the increase in solution viscosity and the selective adsorption of CTAB molecules on the crystal surface. FTIR, Raman and XPS spectroscopy confirmed that the positively charged nitrogen-containing head of the CTAB molecule interacts with the surface of the 318MHCH crystal. Furthermore, since the (−1 0 1) plane has a lower electrostatic potential, the nitrogen-containing head of CTAB with a high potential is more readily adsorbed, which was confirmed by molecular dynamics simulations. Therefore, CTAB is selectively adsorbed on the side of 318MHCH nanowires, hindering the lateral growth and promoting the aspect ratio of 318MHCH nanowires.
