What allows a planet to be both within a potentially habitable zone and sustain habitability over long geologic time? With the advent of exoplanetary astronomy and the ongoing discovery of terrestrial-type planets around other stars, our own solar system becomes a key testing ground for ideas about what factors control planetary evolution. Mars provides the solar system's longest record of the interplay of the physical and chemical processes relevant to habitability on an accessible rocky planet with an atmosphere and hydrosphere. Here we review current understanding and update the timeline of key processes in early Mars history. We then draw on knowledge of exoplanets and the other solar system terrestrial planets to identify six broad questions of high importance to the development and sustaining of habitability (unprioritized): (1) Is small planetary size fatal? (2) How do magnetic fields influence atmospheric evolution? (3) To what extent does starting composition dictate subsequent evolution, including redox processes and the availability of water and organics? (4) Does early impact bombardment have a net deleterious or beneficial influence? (5) How do planetary climates respond to stellar evolution, e.g., sustaining early liquid water in spite of a faint young Sun? (6) How important are the timescales of climate forcing and their dynamical drivers? Finally, we suggest crucial types of Mars measurements (unprioritized) to address these questions: (1) in situ petrology at multiple units/sites; (2) continued quantification of volatile reservoirs and new isotopic measurements of H, C, N, O, S, Cl, and noble gases in rocks that sample multiple stratigraphic sections; (3) radiometric age dating of units in stratigraphic sections and from key volcanic and impact units; (4) higher-resolution measurements of heat flux, subsurface structure, and magnetic field anomalies coupled with absolute age dating. Understanding the evolution of early Mars will feed forward to understanding the factors driving the divergent evolutionary paths of the Earth, Venus, and thousands of small rocky extrasolar planets yet to be discovered.

Abstract Rock shapes and heights around the InSight lander are examined to refine the degradation history of the 27 m‐diameter Homestead hollow. Results document decreasing average exposed rock height and increasing percentage of rocks where height comprises the short axis from outside to within the hollow and support prior models of ejecta deflation accompanied by hollow infilling. We estimate 0.3 m of deflation at the current rim that is realistic compared to rock relief, original ejecta thickness, and predicted aeolian contributions to infilling. We also find that shapes of embayed basalt rocks outside the hollow appear platy, bladed, and elongate in a triangular form factor plot, and more discoidal and bladed in an axes ratio plot. By contrast, expected shapes based on terrestrial studies of basalt rocks are mostly compact, compact platy, compact bladed, compact elongated, platy, bladed, and elongate in triangular form factor plots, and equant with lesser, but significant disc‐ and blade‐shaped rocks in axes ratio plots. We find addition of 10 cm to the heights of rocks near the hollow rim, to account for continued partial embedding in ejecta, yields the best match between observed and expected rock shapes. Exposure of small ejecta rocks in the hollow supports degradation rates of 10−4 m/Myr during most of hollow history. Results indicate that deflation from ejecta accompanied by downwind deposition in the hollow can account for the current degraded form of the crater. Our approach is a new tool for characterizing small crater degradation on regolith‐covered lava plains on Mars.

A time resolved experiment to investigate the ultrafast dynamics following an ion impact onto a solid surface requires an ultrashort ion pump pulse in combination with a properly synchronized and time resolved probe. In order to realize such an experiment, we have investigated a strategy to use femtosecond laser photoionization of atoms entrained in a pulsed supersonic jet for the production of sufficiently short ion pulses. While the generation of Ar ^q+ ions was targeted in previous work, it has in the meantime been demonstrated that argon is not suitable due to extensive cluster formation in the supersonic expansion. Here, we therefore present numerical simulations investigating the use of neon as a precursor gas and show the feasibility of pulses containing up to ∼1000 Ne ^+ ions at keV energies and picosecond duration. In the process, we demonstrate that space charge broadening can be significantly reduced by detuning the flight time focusing conditions of an ion bunching system. Moreover, the results show that a controlled variation of the buncher geometry and potentials permits the generation of picosecond pulses at variable ion energy between 1 and 5 keV.

Interval timing is a complex cognitive process that involves the estimation of time within the seconds-to-minutes range. This temporal processing depends on cortico-striatal interactions, as well as an optimal dopaminergic function. On the other hand, the circadian system controls physiological and behavioral functions with periods close to 24 hr. We have previously reported that short-time perception in mice is influenced by the circadian pacemaker, with dopamine signaling as a link between both temporal systems. In this work we evaluated the involvement of melatonin in the circadian modulation of interval timing, as well as the interaction between this hormone and dopamine levels in the striatum. We report that melatonin-depleted rats, by pinealectomy, present an impairment in their ability to estimate a short (24 s) target duration in the peak-interval procedure. Moreover, melatonin administration in drinking water restores interval timing precision in pinealectomized rats. We also show that circadian desynchronization causes a transient impairment in the timing task. In addition, melatonin administration affects interval timing only when rats are trained and tested during the night. Furthermore, we report that melatonin depletion increases striatal dopamine availability, which is reverted by external melatonin administration. Taken together, our findings add further support to the notion that the circadian system modulates interval timing, probably by using melatonin as an output to regulate dopaminergic functions in brain areas that are important for interval timing mechanisms.

Aneurysms pose life-threatening risks due to the dilatation of the arteries and carry a high risk of rupture. Despite continuous research efforts, there are still no satisfactory or clinically effective pharmaceutical treatments for this condition. Accelerated inflammatory processes during aneurysm development lead to increased levels of matrix metalloproteinases (MMPs) and destabilization of the vessel wall through the degradation of the structural components of the extracellular matrix (ECM), mainly collagen and elastin. Tissue inhibitors of metalloproteinases (TIMPs) directly regulate MMP activity and consequently inhibit ECM proteolysis. In this work, the synthesis of TIMP-1 protein was increased by the exogenous delivery of synthetic TIMP-1 encoding mRNA into aortic vessel tissue in an attempt to inhibit MMP-9. In vitro, TIMP-1 mRNA transfection resulted in significantly increased TIMP-1 protein expression in various cells. The functionality of the expressed protein was evaluated in an appropriate ex vivo aortic vessel model. Decreased MMP-9 activity was detected using in situ zymography 24 h and 48 h post microinjection of 5 µg TIMP-1 mRNA into the aortic vessel wall. These results suggest that TIMP-1 mRNA administration is a promising approach for the treatment of aneurysms. [ABSTRACT FROM AUTHOR]

Abstract The application of synthetic modified messenger RNA (mRNA) is a promising approach for the treatment of a variety of diseases and vaccination. In the past few years, different modifications of synthetic mRNA were applied to render the mRNA more stable and less immunogenic. However, the repeated application of synthetic mRNA still requires the suppression of immune activation to avoid cell death and to allow a sufficient production of exogenous proteins. Thus, the addition of type I interferon (IFN) inhibiting recombinant protein B18R is often required to avoid IFN response. In this study, the ability of B18R encoding mRNA to prevent the immune response of cells to the delivered synthetic mRNA was analyzed. The co-transfection of enhanced green fluorescent protein (eGFP) mRNA transfected fibroblasts with B18R encoding mRNA over 7-days resulted in comparable cell viability and eGFP protein expression as in the cells transfected with eGFP mRNA and incubated with B18R protein. Using qRT-PCR, significantly reduced expression of interferon-stimulated gene Mx1 was detected in the cells transfected with B18R mRNA and stimulated with IFNβ compared to the cells without B18R mRNA transfection. Thereby, it was demonstrated that the co-transfection of synthetic mRNA transfected cells with B18R encoding mRNA can reduce the IFN response-related cell death and thus, improve the protein expression.

Information about unsafe foods or feeds must be exchanged between European Union (EU) member states as quickly as possible. This is why the EU’s Rapid Alert System for Food and Feed (RASFF) exists. It helps to ensure that products that may be harmful to health do not enter the market or can be specifically withdrawn from the market. Different notifications are used depending on the risk and urgency. This article provides an overview of the 61 notifications in the RASFF between 2020 and 2022 on the Δ9-tetrahydrocannabinol (Δ9-THC) content in cannabidiol (CBD) oils and CBD food supplements. These products are available on the EU market despite the lack of novel food approval. Δ9-THC is a naturally occurring psychotropic compound extracted from the hemp plant Cannabis sativa that can have adverse effects on consumers (such as drowsiness, dizziness, tachycardia, or changes in blood pressure). In a previous German national survey, 23 of the 125 products tested (18%) exceeded the lowest observed adverse effect level (LOAEL) of Δ9-THC. In comparison, for products identified as a serious risk in the RASFF, the Δ9-THC concentrations were generally higher (up to 2410 mg/kg) and 14 of 34 products (41%) exceeded the LOAEL. Considering these data, a threshold of 500 mg/kg (0.05%) may be proposed to define a serious risk, as the LOAEL would not be exceeded in typical consumption scenarios below this level and serious risks, as well as narcotic effects in the product group of food supplements, could be excluded. This threshold could be used in the interim until the full toxicological assessment is available within the novel food approval procedure.
