115. HIF-1α effects on angiogenic potential in human small cell lung carcinoma

Abstract

Abstract Background Hypoxia-inducible factor-1 alpha (HIF-1α) maybe an important regulatory factor for angiogenesis of small cell lung cancer (SCLC). Our study aimed to investigate the effect of HIF-1α on angiogenic potential of SCLC including two points: One is the effect of HIF-1α on the angiogenesis of SCLC in vivo. The other is the regulation of angiogenic genes by HIF-1α in vitro and in vivo. Methods In vivo we used an alternative method to study the effect of HIF-1a on angiogenic potential of SCLC by buliding NCI-H446 cell transplantation tumor on the chick embryo chorioallantoic membrane (CAM) surface. In vitro we used microarray to screen out the angiogenic genes regulated by HIF-1a and tested their expression level in CAM transplantation tumor by RT-PCR and Western-blot analysis. Results In vivo angiogenic response surrounding the SCLC transplantation tumors in chick embryo chorioallantoic membrane (CAM) was promoted after exogenous HIF-1α transduction (p < 0.05). In vitro the changes of angiogenic genes expression induced by HIF-1α in NCI-H446 cells were analyzed by cDNA microarray experiments. HIF-1α upregulated the expression of angiogenic genes VEGF-A, TNFAIP6, PDGFC, FN1, MMP28, MMP14 to 6.76-, 6.69-, 2.26-, 2.31-, 4.39-, 2.97- fold respectively and glycolytic genes GLUT1, GLUT2 to2.98-, 3.74- fold respectively. In addition, the expression of these angiogenic factors were also upregulated by HIF-1α in the transplantion tumors in CAM as RT-PCR and Western-blot analysis indicated. Conclusions These results indicated that HIF-1α may enhance the angiogenic potential of SCLC by regulating some angiogenic genes such as VEGF-A, MMP28 etc. Therefore, HIF-1α may be a potential target for the gene targeted therapy of SCLC.

116. Master Synchronization in Physical-Layer Communications of Wireless Sensor Networks

Abstract

Synchronization in physical layer of wireless sensor networks is critical in restricting complexity of tag node and power consumption. Considering the master-to-tag communication (i.e., receiving signal from a master or anchor node by tag nodes), we propose a scheme on the basis of the principles of feedback control, to transfer the signal acquisition functionality from the tag receivers to the master nodes in a cluster. Furthermore, the algorithm of timing acquisition and phase adjustment do work in the master transmitter, and the tag nodes just need feedback results of the phase detection. The tag nodes do not require complicated clock or phase adjustment circuit any more or estimation in synchronization either. Thus, this master synchronization method reduces the complexity of tag nodes and power consumption. Due to the large random time delay in the wireless feedback loop, there exists the problem of stability and convergence in the acquisition. We analyze it and present a feasible scheme for the proposed master synchronization. In order to reduce acquisition time and cost in feedback, a two-step master acquisition algorithm is proposed. The acquisition performance under nonideal channel is analyzed, and further verified by simulations.

131. Endometrial regenerative cells: A novel stem cell population

Abstract

Abstract Angiogenesis is a critical component of the proliferative endometrial phase of the menstrual cycle. Thus, we hypothesized that a stem cell-like population exist and can be isolated from menstrual blood. Mononuclear cells collected from the menstrual blood contained a subpopulation of adherent cells which could be maintained in tissue culture for >68 doublings and retained expression of the markers CD9, CD29, CD41a, CD44, CD59, CD73, CD90 and CD105, without karyotypic abnormalities. Proliferative rate of the cells was significantly higher than control umbilical cord derived mesenchymal stem cells, with doubling occurring every 19.4 hours. These cells, which we termed "Endometrial Regenerative Cells" (ERC) were capable of differentiating into 9 lineages: cardiomyocytic, respiratory epithelial, neurocytic, myocytic, endothelial, pancreatic, hepatic, adipocytic, and osteogenic. Additionally, ERC produced MMP3, MMP10, GM-CSF, angiopoietin-2 and PDGF-BB at 10–100,000 fold higher levels than two control cord blood derived mesenchymal stem cell lines. Given the ease of extraction and pluripotency of this cell population, we propose ERC as a novel alternative to current stem cells sources.

137. Umbrella: Enabling ISPs to Offer Readily Deployable and Privacy-Preserving DDoS Prevention Services

Abstract

Defending against distributed denial of service (DDoS) attacks in the Internet is a fundamental problem. However, recent industrial interviews with over 100 security experts from more than ten industry segments indicate that DDoS problems have not been fully addressed. The reasons are twofold. On one hand, many academic proposals that are provably secure witness little real-world deployment. On the other hand, the operation model for existing DDoS-prevention service providers (e.g., Cloudflare, Akamai) is privacy invasive for large organizations (e.g., government). In this paper, we present Umbrella, a new DDoS defense mechanism enabling Internet Service Providers (ISPs) to offer readily deployable and privacy-preserving DDoS prevention services to their customers. At its core, Umbrella develops a multi-layered defense architecture to defend against a wide spectrum of DDoS attacks. In particular, the flood throttling layer stops amplification-based DDoS attacks; the congestion resolving layer, aiming to prevent sophisticated attacks that cannot be easily filtered, enforces congestion accountability to ensure that legitimate flows are guaranteed to receive their fair shares regardless of attackers' strategies; and finally the userspecific layer allows DDoS victims to enforce self-desired traffic control policies that best satisfy their business requirements. Based on Linux implementation, we demonstrate that Umbrella is capable to deal with large scale attacks involving millions of attack flows, meanwhile imposing negligible packet processing overhead. Further, our physical testbed experiments and large scale simulations prove that Umbrella is effective to mitigate various DDoS attacks.
