110. Design of a questionnaire for detecting contact lens discomfort: the Contact Lens Discomfort Index

Abstract

Clinical relevance: Questionnaires can provide unique information regarding the status of a condition from the perspective of the patient. Contact lens (CL) discomfort (CLD) is a condition subjectively reported by CL users; thus, updated and reliable questionnaires are needed. Background: The objective of this study was to design a new questionnaire for identifying CLD, based on the currently established CLD definition considered in the Tear Film and Ocular Surface CLD Workshop. Methods: A survey to characterise CL wearers was created and administered. Some items of the survey were selected by a focus group and analysed using a Rasch model to develop a questionnaire to identify CLD. Unidimensionality was checked applying principal components analysis. To build a plausible CLD classification (symptomatic vs asymptomatic), an unsupervised cluster analysis was used. Gower’s general similarity coefficient, the partitioning around medoids algorithm, and the silhouette width were calculated. The reliability of the questionnaire was assessed using the intraclass correlation coefficient and Cohen’s kappa coefficient. A study was performed to compare this questionnaire with the Contact Lens Dry Eye Questionnaire (CLDEQ)-8. Results: The 31-item survey was completed by 1104 volunteers, and the posterior analysis resulted in a 9-item questionnaire, the Contact Lens Discomfort Index (CLDI). It showed a good fit with the model (mean square infit and outfit values range was 0.76–1.10), and good unidimensionality (eigenvalues ≤ 1.3). The CLDI score range was 0–18 and the cut-off score for identifying CLD was 8. The intraclass correlation coefficient was 0.88 (95% confidence interval, 0.75–0.94), and kappa was 0.67 (95% confidence interval, 0.41–0.93). Comparison of the CLDI with the CLDEQ-8 in a sample of 58 CL wearers showed that 70.7% were classified equally by both questionnaires. Conclusions: The CLDI is a well-structured instrument, with acceptable psychometric properties, making it a valid and reliable tool to measure CL-related symptoms.

Trabajo presentado en el XVI Congreso Nacional de Ciencias Hortícolas, celebrado en Córdoba del 17 al 22 de octubre de 2021, El hierro (Fe) es un micronutriente esencial para las plantas. En suelos calizos se encuentra en su forma oxidada (Fe 3+), presentando baja solubilidad y disponibilidad para las plantas. Bajo deficiencia de Fe, las plantas dicotiledóneas inducen diversas respuestas fisiológicas y morfológicas en sus raíces para facilitar así su captación y paliar los efectos que produce su deficiencia. Existen evidencias de que determinados microorganismos rizosféricos, como los que provocan la Respuesta Sistémica Inducida (ISR), pueden facilitar la nutrición férrica de las plantas. El objetivo de este trabajo ha sido, por una parte, estudiar el efecto de la cepa no patogénica de Fusarium oxysporum (FO12), posiblemente inductora de ISR, sobre el crecimiento y clorosis de plantas de tomate (Solanum lycopersicum Mill.), y sobre la colonización de raíces de pepino (Cucumis sativus L.). Por otra parte, estudiar el efecto de determinadas levaduras (Debaryomyces hansenii, Saccharomyces cerevisiae y Hansenula polymorpha) sobre diversas respuestas a la deficiencia de Fe en plantas de pepino. Los experimentos con tomate se han desarrollado en suelo calizo y los de pepino en cultivo hidropónico. Los resultados obtenidos con FO12 han mostrado su capacidad para colonizar endofiticamente las raíces de pepino y su efecto promotor del crecimiento, la concentración de clorofila y de Fe (en hojas) en plantas de tomate. Las diferentes levaduras utilizadas han causado inducción de la capacidad reductora de Fe 3+, de la acidificación de la rizosfera, y de la proliferación de pelos radicales en la zona subapical de las raíces de pepino. Estos resultados sugieren que, tanto FO12 como las diferentes levaduras utilizadas, tienen potencial como biofertilizantes de Fe., Plan Propio de la Universidad de Córdoba y Ministerio de Ciencia e Innovación.

112. Agrupamiento de poemas de autores suicidas y no suicidas usando K-means y enjambre de partículas

Abstract

Suicide is considered a public health issue, and its early detection and treatment may contribute to its prevention. Automatic detection of suicidal ideation indicators within texts can be a useful tool to prevent it. In this work a corpus was compiled, which consists of poems written by twelve different poets, where six of them committed suicide. Two vector representations were experimented on, one with the total number of words and another with words related to negative emotional concepts. The vectors were clustered using two algorithms: K-Means and a K-Means with Particle Swarm Optimization hybrid. The efficiency of the vector representations and the used algorithms were compared, obtaining as result that, through the hybrid algorithm and the negative emotional concepts vocabulary, the groups of poets with suicidal ideation and without it could be distinguished with an accuracy of 0.98. La detección automática de indicadores de ideación suicida en textos tiene posibilidad de ser una herramienta útil para su prevención. En este trabajo, se reunió un corpus formado por poemas de doce poetas distintos, de los cuales seis cometieron suicidio. Se experimentó con dos representaciones vectoriales, una por número total de palabras y otra por palabras relacionadas con conceptos emocionales negativos. Los vectores se agruparon utilizando dos algoritmos: K-means y un híbrido de K-means con Optimización por Enjambre de Partículas. 126. Reevaluating a non‐conventional procedure to microencapsulate beneficial lactobacilli: assessments on yield and bacterial viability under simulated technological and physiological conditions.

Abstract

BACKGROUND Maintaining viability of beneficial microorganisms applied to foods still constitutes an industrial challenge. Many microencapsulation methodologies have been studied to protect probiotic microorganisms and ensure their resistance from manufacturing through to consumption. However, in many Latin‐American countries such as Argentina there are still no marketed food products containing microencapsulated beneficial bacteria. The objectives of this work were: (i) to obtain microcapsules containing Lactobacillus fermentum L23 and L. rhamnosus L60 in a milk protein matrix; and (ii) to evaluate the viability of microencapsulated lactobacilli exposed to long‐term refrigerated storage, mid–high temperatures and simulated gastrointestinal conditions. RESULTS: The method of emulsification/rennet‐catalyzed gelation of milk proteins used in this study led to high encapsulation yields for both strains (98.2–99%). Microencapsulated lactobacilli remained viable for 120 days at 4 °C, while free lactobacilli gradually lost their viability under the same conditions. Microencapsulation increased the resistance of lactobacilli to mid–high temperatures, since they showed survival rates of 95–99.3% at 50 °C, and of 72.5–74.4% at 65 °C. Under simulated gastric conditions, the microencapsulated lactobacilli counts were higher than 8.5 log CFU mL−1 and showed survival rates between 96.61% and 97.74%. Furthermore, in the presence of bile (0.5–2% w/v) the survival of microencapsulated strains was higher than 96%. CONCLUSION: The microencapsulation process together with the matrix of milk proteins used in this study protected beneficial Lactobacillus strains against these first simulated technological and physiological conditions. These findings suggest that this microencapsulation method could contribute to secure optimal amounts of living lactobacilli cells able to reach the intestine. © 2021 Society of Chemical Industry. Abstract

Amaranthus palmeri S. Watson is probably the worst glyphosate-resistant (GR) weed worldwide. The EPSPS (5-enolpyruvylshikimate-3-phosphate-synthase) gene amplification has been reported as the major target-site-resistance (TSR) mechanism conferring resistance to glyphosate in this species. In this study, TSR and non-target-site-resistance (NTSR) mechanisms to glyphosate were characterized in a putative resistant A. palmeri population (GRP), harvested in a GR-soybean crop from Argentina. Glyphosate resistance was confirmed for the GRP population by dose-response assays. No evidence of TSR mechanisms as well as glyphosate metabolism was found in this population. Moreover, a susceptible population (GSP) that absorbed about 10% more herbicide than the GRP population was evaluated at different periods after treatment. The GSP population translocated about 20% more glyphosate to the remainder of the shoots and roots at 96 h after treatment than the control, while the GRP population retained 62% of herbicide in the treated leaves. This is the first case of glyphosate resistance in A. palmeri involving exclusively NTSR mechanisms. This research was funded by the MINECO-Spain (Grant AGL2016-78944-R).

Abstract

Abstract Background Animal studies have shown the reproductive toxicity of a number of heavy metals. Very few human observational studies have analyzed the relationship between male reproductive function and heavy metal concentrations in diverse biological fluids. Methods The current study assessed the associations between seminal and hormonal parameters and the concentration of the 3 most frequent heavy metal toxicants (lead, cadmium and mercury) in three different body fluids. Sixty one men attending infertility clinics that participated in a case-control study to explore the role of environmental toxins and lifestyles on male infertility were analyzed. Concentration of lead, cadmium and mercury were measured in blood and seminal plasma and whole blood using anodic stripping voltammetry and atomic absorption spectrophotometry. Serum samples were analyzed for follicle-stimulating hormone, luteinizing hormone and testosterone. Semen analyses were performed according to World Health Organization criteria. Mann-Whitney test and Spearman's rank correlations were used for unadjusted analyses. Multiple linear regression models were performed controlling for age, body mass index and number of cigarettes per day. Results There were no significant differences between cases and controls in the concentrations of heavy metals in any of the three body fluids. In multivariate analyses using all subjects no significant associations were found between serum hormone levels and metal concentrations. However there was a significant positive association between the percentage of immotile sperms and seminal plasma levels of lead and cadmium. Conclusions Our results suggest that the presence of lead and cadmium in the reproductive tract of men may be related to a moderate alteration of their seminal parameters.

Abstract

Abstract Background The ability of an intracellular pathogen to establish infection depends on the capacity of the organism to survive and replicate inside the host. Mycobacterium fortuitum is a bacteria that contains genes involved in the detoxification of the oxygen reactive species such as those produced by the host during the infection. In this work, we investigate the effects of hydrogen peroxide on the transcription and expression of these genes by developing a real time quantitative PCR technique (qRT-PCR) using the ribosomal promoter region (rrnA-P1) as reference product for quantification of the mRNA levels. Results M. fortuitum cultures were treated with different hydrogen peroxide concentrations (0.02 to 20 mM) during several periods of time (30 to 120 minutes). The activity of the enzymes KatGII and SodA, and the transcription of corresponding genes were evaluated. The transcriptional regulator furAII gene was also studied. The ribosomal promoter region rrnA-P1 was validated as referential product under the stress conditions checked by qRT-PCR. Minor changes were observed under the conditions tested except when bacteria were incubated in the presence of 20 mM hydrogen peroxide. Under those conditions, the levels of transcription of the three genes under study increased at 30 minutes of treatment. The viability of the bacteria was not influenced under the conditions tested. Conclusion In this work, we have quantified transcriptional responses to stress suggesting that, the opportunistic pathogen M. fortuitum is more resistant and differs in behaviour in the presence of hydrogen peroxide, when compared to the major pathogen Mycobacterium tuberculosis and the saprophyte Mycobacterium smegmatis. Besides, we demonstrate the mycobacterial non-coding region rrnA-P1 to be a suitable reference product in the analysis of qRT-PCR transcriptional data of M. fortuitum.

Abstract

Purpose: To identify alterations in neuropathic and inflammatory pain gene expression associated with contact lens (CL) wear and CL discomfort (CLD).Methods: Eight non-wearers, eight asymptomatic CL wearers (CLWs) and eight symptomatic CLWs were included. Conjunctival cells were collected by impression cytology and the mRNA expression levels of 85 genes were analyzed. Differentially expressed genes between non-wearers and CLWs and between asymptomatic and symptomatic CLWs were analyzed. An enrichment analysis was also performed.Results: Twelve genes were upregulated (including IL10, PDYN and PENK) and 28 downregulated (CCL2, IL1A, IL1B, IL2 and NGF) in CLWs (p ≤ 0.050). Eleven genes were upregulated (CCL2, IL1A, IL1B, IL2 and NGF) and nine downregulated (PDYN and PENK) in symptomatic CLWs (p ≤ 0.035). Enriched overrepresented terms were related to pain, neuronal transmission and inflammation.Conclusion: Contact lens wear might produce a desensitization-like mechanism responsible for comfortable CL wear.
