109. Seasonal abundance of horse flies ( Diptera, Tabanidae ) in the Chirpan eminences, Bulgaria

The aim of the present study was to explore the seasonal activity of tabanids on the territory of the Chirpan Eminences, in Stara Zagora region, Bulgaria. The study was carried out during the flight period (May - September, 2010 - 2011). A total of 1034 specimens of tabanids from 23 species and 8 genera were collected and identified. The horse flies began flying during the second half of May and its flight period lasted until mid - September. The peak of seasonal abundance was observed during the first half of July. The most abundant species in the horse flies assemblages on the Chirpan Eminences were Tabanus quatuоrnotatus (35.30%), Tabanus tergestinus (28.05%) and Philipomyia graeca (10.93%). The largest number of specimens of T. tergestinus was collected during the peak of seasonal abundance of horse flies on the territory of the Chirpan Eminences.

Low-dose once weekly methotrexate (MTX) is the first-line disease-modifying antirheumatic drug (DMARD) to treat rheumatoid arthritis and psoriasis. Although methotrexate is generally considered to have a good safety profile it can occasionally induce severe side effects such as pancytopenia, mucositis, disorders of kidney and liver. Oral mucositis should alert physicians to MTX toxicity. We report a 64-year-old woman with a severe drug reaction including disseminated shingles, oral mucositis and pancytopenia only three days after starting a therapeutic low-dose of MTX. Initially, mucositis and myelosuppression couldn’t be accounted for by the underlying disease. In taking the patient’s history, MTX intake 10 days ago was missed, the patient reporting only current medications. However, there was more to the skin rash than meets the eye. Only after further inquiry did the patient reveal the intake of 2 doses of MTX and the subsequent withdrawal of medication. Arriving at the correct diagnosis in difficult cases, as in the case presented, requires further evaluation, including repeat history taking and eliciting more details if diagnosis remains elusive.

116. Singing the nature - еthnobotanical knowledge in Bulgarian folk songs

Background: Bulgarian poetic folklore reflects an agrarian culture deeply connected with land and nature. However, traditional ecological knowledge (TEK) transmitted through Bulgarian folklore is scarcely assessed. Questions: What are the dimensions of the TEK related to plant diversity (native and introduced) that appear in Bulgarian folk songs and what is their potential as transmitters of ТЕК? Data description: The lyrics of 10,113 Bulgarian folk songs were excerpted from major academic collections and a set of unpublished songs. Study site and dates: Current study covers songs that have been documented since mid-19th century onwards in the present and former Bulgarian territories and in areas that have been inhabited by ethnic Bulgarians abroad. Methods: Common plant names and descriptions of plants and landscapes were used to detect botanical affiliations of the denoted plants. We focused on frequencies of plant representations and their functions associated with cultural, agricultural and food-processing practices. Results: A total of 146 plant taxa from 109 genera were mentioned in 47.3 % of the studied songs. Over 60 % of the most frequently mentioned taxa were archaeophytes while neophytes were represented by seven taxa, denoting everyday and spiritual importance of nature. Conclusions: Bulgarian folk songs presented wide range of nature-related information typically reported for ethnic groups outside Europe. Quantification of TEK preserved in documented poetic folklore and further assessment of performance of nature-rich folklore would allow development additional tools for evaluation of cultural significance of species, landscapes and ecosystems as well as for development of educational and inspirational materials.

117. Acquisition of renogenic competence in the early mouse embryo and embryonic stem cells

The acquisition of renogenic competence (the ability to give rise to kidney) during embryonic development is not yet fully understood. Clarifying the temporal and molecular aspects of this process is equally essential for understanding excretory system development and for devising methods for successful differentiation of embryonic stem cells (ESCs) to renal cells for disease modeling, toxicology screening and potential cell replacement therapies. In embryo development, the metanephric (permanent) kidney arises as a result of inductive interactions between two embryonic structures that arise in the intermediate mesoderm - the ureteric bud (UB, a diverticulum of the Wolffian duct) and the metanephric mesenchyme (MM). The UB develops into the collecting duct system and the MM undergoes an epithelial-to-mesenchymal transition to form the secretory units of the kidney - the nephrons. In this thesis, I used a tissue disaggregation-reaggregation method that allows the reconstruction of mouse organotypic kidney rudiments to place different embryonic cells in the environment of a developing kidney and assess their potential to integrate into kidney epithelia and differentiate to renal cells. First, the suitability of this method was evaluated and a quantitative assay for evaluating the numbers of test cells integrating in various renal compartments was developed. Second, the reaggregation method was used to characterise the renogenic potential of undifferentiated mouse ESCs, ESC-derived cells after Notch inhibition, and cells derived from the presumptive nephrogenic regions of embryos at various stages of development. ESCs are isolated from the inner cell mass of an embryo and have the potential to differentiate to any tissue of the body when injected into mouse blastocysts. Strategies have successfully been devised for ESC differentiation to many lineages, but very few studies reported any success with the differentiation of ESCs to a renal lineage. Undifferentiated ESCs showed a very good ability to form chimeric structures with developing kidney tubules (both nephrons and extending UBs). Nevertheless, the resulting structures were morphologically different from renal epithelia in most cases and integrated ESC-derived cells were not positive for several combinations of kidney markers. These results suggested that the influence of the niche was not sufficient for a successful ESC differentiation to renal cells. Treatment of ESC with an inhibitor of the Notch pathway to increase the proportion of mesodermal cells did not improve this outcome. On the basis of these results, it was speculated that the earliest lineage to which embryonic stem cells must be differentiated in order to become competent to make renal cells should first be identified. I addressed this by determining the developmental stage at which cells able to contribute to the formation of metanephric epithelia first appear in mouse embryo development. When mixed in embryonic kidney reaggregates labelled cells isolated from the nephrogenic regions of E9.5 embryos integrated into various renal compartments. These cells were seen in UBs, nephrons, glomeruli and the condensing mesenchyme. Marker expression studies showed that the exogenous E9.5 cells expressed an array of kidney markers - Pax2 in renal epithelia and the condensing mesenchyme, Wt1 in glomeruli and Six2 in the condensing mesenchyme. Furthermore, exogenous E9.5 cells also co-expressed Pax2/Wt1 in the condensing mesenchyme, Megalin/Ecadherin in the proximal tubule and Pax2/E-cadherin in renal epithelia. This provides evidence that challenges the existing model and suggests that some cells from the intermediate mesoderm at a stage where the metanephric blastema is yet formed are competent to contribute to kidney structures. Furthermore, experiments with E8.5 embryos showed that such a renocompetence could be acquired even before the specification of intermediate mesoderm. These findings contribute to our knowledge about kidney cell specification and provide valuable information to guide future attempts to develop an efficient method for deriving renal cells from ESCs. Furthermore, the reported ability of ESC-derived non-kidney cells to form chimeric structures with renal tubules provides a proof-of-principle that it might be possible to use exogenous types of cells for physiological support to injured kidney tubules, thus offering a possible novel approach for cell replacement therapies.

Objectives: The aim of this research is to examine the effects of fructose-drinking on the plasma levels of appetite-regulating hormones insulin, leptin and ghrelin in male and female rats. Methods: Mature Wistar rats were divided as follows: two control groups - male (CM) and female (CF); two fructose-drinking groups - male (FDM) and female (FDF), received 15% fructose solution. The experiment lasted 11 weeks. At the end, insulin, leptin and ghrelin levels as well as lipid and glucose profile were assessed. Results: Plasma concentrations of the examined hormones were elevated in fructose-drinking groups. However, in the FDM group only the leptin levels were significantly increased compared to the control. In the FDF group, all three appetite-regulating hormones showed the highest concentrations in comparison to the other groups. Conclusion: Sex hormones may affect the appetite-regulation signals and could be a factor contributing to degree of metabolic changes caused by long-term fructose overconsumption.

122. Extraction of Proteins and Other Intracellular Bioactive Compounds From Baker’s Yeasts by Pulsed Electric Field Treatment

Yeasts are rich source of proteins, antioxidants, vitamins, and other bioactive compounds. The main drawback in their utilization as valuable ingredients in functional foods and dietary supplements production is the thick, indigestible cell wall, as well as the high nucleic acid content. In this study, we evaluated the feasibility of pulsed electric field (PEF) treatment as an alternative method for extraction of proteins and other bioactive intracellular compounds from yeasts. Baker’s yeast water suspensions with different concentration (12.5–85 g dry cell weight per liter) were treated with monopolar rectangular pulses using a continuous flow system. The PEF energy required to achieve irreversible electropermeabilization was significantly reduced with the increase of the biomass concentration. Upon incubation of the permeabilized cells in water, only relatively small intracellular compounds were released. Release of 90% of the free amino acids and low molecular UV absorbing compounds, 80% of the glutathione, and ∼40% of the total phenol content was achieved about 2 h after pulsation and incubation of the suspensions at room temperature. At these conditions, the macromolecules (proteins and nucleic acids) were retained largely inside. Efficient protein release (∼90% from the total soluble protein) occurred only after dilution and incubation of the permeabilized cells in buffer with pH 8–9. Protein concentrates obtained by ultrafiltration (10 kDa cut off) had lower nucleic acid content (protein/nucleic acid ratio ∼100/4.5) in comparison with cell lysates obtained by mechanical disintegration. The obtained results allowed to conclude that PEF treatment can be used as an efficient alternative approach for production of yeast extracts with different composition, suitable for application in food, cosmetics and pharmaceutical industries.

123. First Survey of the Vascular and Cryptogam Flora on Bulgaria’s Ancient Mounds

This work represents the first study of the floristic diversity on Bulgaria’s ancient mounds. The objective of this research was to assess the importance of the mounds for the preservation of the native vascular and cryptogam flora. Our sampling design included 111 ancient mounds distributed throughout the country. We recorded a total of 1059 vascular plants, 58 bryophytes and 61 lichen taxa. Despite their small area, the mounds were shown to preserve nearly a quarter of the Bulgarian flora. The vegetation cover on the mounds included 61% perennials indicating a long-term persistence and stability. The majority (98%) of the established vascular plants were native species. Although the conservation significance of the vascular plant species were not common, we recorded 2 critically endangered, 9 endangered and 14 Balkan endemics during the present study. The lichen Arthopyrenia salicis was recorded for the first time in Bulgaria and a new locality of the rare bryophyte Ceratodon conicus was discovered. The established compositional difference between plots from the northern and southern slopes of the mounds (88.95%) is a testament to the high local habitat diversity. The prevalence of species characteristic for Festuco-Brometea suggests that the mounds preserve fragments of native grasslands and steppes. The variation in cover of agricultural and other human modified areas in the mounds’ immediate surroundings did not substantially affect their species richness. We argue that the ancient mounds should be taken into consideration in future green space planning.

Juvenile idiopathic arthritis (JIA) is the most common rheumatic disease in childhood, with prevalence of 16–150 cases per 100,000 children. The etiopathogenesis of JIA is a challenge incorporating a complex network with only 18% attributed to genetic factors. The remaining part should therefore be explained by non-hereditary factors. Given that around 70% of the immune cells are located in the gut, the potential role of the gut microbiota in the etiopathogenesis of JIA has been recently investigated. The aim of this review is to discuss the complexity of the link between gut microbiota and JIA, the different methods for identifying bacteria, the shape-up of the microbiota from birth to adulthood. The objectives are to discuss various pathways involved in this process: changes in the microbiota contents in healthy individuals and JIA patients, increased gut permeability, influence on T-cell differentiation and proliferation. Factors that have been associated with dysbiosis: diet, pathogens and drug use, are discussed. JIA is not a benign disease, it is a chronic disease and an important cause of short- and long-term disability-significant joint contractures, leg-length inequalities and uveitis, which can lead to impaired vision. It is known that at least one-third of children will have ongoing active disease into their adult years, and many will have some limitation in their daily life activities. A deeper understanding of the pathways by which disturbances in the microbiome may evolve to disease may open doors to the development of new treatment or prevention strategies in the future.

The electronic system histdict is designed as a tool for research, adequate presentation and popularization of a part of Bulgaria’s cultural and historical heritage: the Bulgarian language and its medieval literature. The article describes the various steps in the development of histdict. Attention is paid to each component of the resource: specialized Unicode fonts, electronic diachronic corpus, dictionary of Old Bulgarian, historical dictionary equipped with tools for writing and editing dictionary entries, grammatical dictionary, prototypical search engine, and virtual keyboard. The article also lays out the principles followed in the development of the diachronic grammatical dictionary of the Bulgarian language.

Rain-fed peatlands are dominated by peat mosses (Sphagnum sp.), which for their growth depend on nutrients, water and CO2 uptake from the atmosphere. As the isotopic composition of carbon (12,13C) and oxygen (16,18O) of these Sphagnum mosses are affected by environmental conditions, Sphagnum tissue accumulated in peat constitutes a potential long-term archive that can be used for climate reconstruction. However, there is inadequate understanding of how isotope values are influenced by environmental conditions, which restricts their current use as environmental and palaeoenvironmental indicators. Here we tested (i) to what extent C and O isotopic variation in living tissue of Sphagnum is species-specific and associated with local hydrological gradients, climatic gradients (evapotranspiration, temperature, precipitation) and elevation; (ii) whether the C isotopic signature can be a proxy for net primary productivity (NPP) of Sphagnum; and (iii) to what extent Sphagnum tissue δ18O tracks the δ18O isotope signature of precipitation. In total, we analysed 337 samples from 93 sites across North America and Eurasia using two important peat-forming Sphagnum species (S. magellanicum, S. fuscum) common to the Holarctic realm. There were differences in δ13C values between species. For S. magellanicum δ13C decreased with increasing height above the water table (HWT, R2 = 17 %) and was positively correlated to productivity (R2 = 7 %). Together these two variables explained 46 % of the between-site variation in δ13C values. For S. fuscum, productivity was the only significant predictor of δ13C but had low explanatory power (total R2 = 6 %). For δ18O values, approximately 90 % of the variation was found between sites. Globally modelled annual δ18O values in precipitation explained 69 % of the between-site variation in tissue δ18O. S. magellanicum showed lower δ18O enrichment than S. fuscum (−0.83 ‰ lower). Elevation and climatic variables were weak predictors of tissue δ18O values after controlling for δ18O values of the precipitation. To summarize, our study provides evidence for (a) good predictability of tissue δ18O values from modelled annual δ18O values in precipitation, and (b) the possibility of relating tissue δ13C values to HWT and NPP, but this appears to be species-dependent. These results suggest that isotope composition can be used on a large scale for climatic reconstructions but that such models should be species-specific.

Sommer vorm Balkon has received critical acclaim for its director’s specific brand of low-key and crowd-pleasing realism. Praised have been also the film’s stylistic austerity and peculiar post-unification mix of various generic elements. At the same time, Sommer vorm Balkon can be considered a part of the venerable tradition of earlier Berlin films written by Wolfgang Kohlhaase, a tradition that this latest work draws from and reinvents. Instead of looking at the film exclusively as an example of the work of two auteurs, this paper analyzes it as a case of creative intervention into the historical interplay of place and cinema. This collaboration between the representatives of two different generations of filmmakers (Dresen and Kohlhaase) consciously transforms this Berlin film into an archive of urban memories – buried memories from preceding decades, cinematic memories, as well as memories that are now in the making.

Carotenes have been identified as the most essential carotenoids in the human body, together with lycopene, lutein, and β-cryptoxanthin due to their beneficial antioxidant activities. Because of these properties, we have designed and conducted in 2018--2021 an experiment where various tomato accessions were metabolically profiled for their antioxidant content and other fruit parameters for further usage in crop improvement programs. Same genotypes were phenotyped and profiled with DNA markers to characterize their relative genetic distance. Aggregated data from metabolic and genetic profiling were complemented by the phenotypic data to select for the most promising cross (Pl. karotina x L1116) that was performed and developed into F1 and F2 generations. After phenotyping and genetic profiling of the 104 F2 individuals obtained from that cross, 24 individuals were selected for metabolic profiling for antioxidant contents. In relation to carotenoids accumulated in fully matured fruits the highest lycopene concentration detected in our study was 587.03 µg/g DW, while β-carotene reached 440.29 µg/g DW. Our results support the use of such integrative approach to accelerate tomato breeding when improved metabolic content of the fruits is aimed. [ABSTRACT FROM AUTHOR]

Introduction:MicroRNAs are small non-coding RNA molecules, which possess the potential to become biomarkers for several cardiac diseases. Investigations of microRNAs, associated with cardiac damage in children and young patients with beta-thalassemia major, would contribute to a better understanding of their role in commonly occurring cardiac complications, as well as examine the possibility of being used as diagnostic markers.Aim:The present study is aimed at investigating the expression of specific microRNAs associated with cardiac damage in children and young adults with beta-thalassemia major, as well as at comparing the results with the cardiac function in the examined subjects.Patients and Methods:The design of the study was prospective and included 50 subjects of whom 27 patients were with beta-thalassemia major (13 girls and 14 boys) at an average age of 15.14 years, and 23 healthy controls matched for sex and age. The average patient age was 15.14 years (between 5 and 24 years), and the average healthy control’s age was 16.1 years (between 6 and 23 years). From all of the subjects, the relative quantity of 5 microRNAs in blood serum was determined via RT-qPCR. A thorough echocardiographic examination was performed to evaluate cardiac function.Results:Five specific types of microRNAs, associated with acute cardiac damage, cardiac fibrosis, and remodeling, were studied. The results showed that hsa-miR-30a-5p is significantly downregulated, and hsa-miR-150-5p is upregulated in patients with beta-thalassemia major, in comparison to healthy controls.Conclusion:The upregulation of hsa-miR-30a-5p in patients with beta-thalassemia major could be a prognostic marker for a future cardiac injury that is not yet clinically manifested. The different expression pattern of hsa-miR-150-5p is more likely to be associated with the disease itself.

The purpose of this research paper is to detect, present and generalize all forms of information resonance of the book “Interior Chinatown“ by Charles Yu and on this basis to check up the hypothesis that the book was realized as a media. The research objectives include searching and presenting various forms of readers' feedback about the book, summarizing and visualizing data using diagrams, recapitulating the results by quantitative generalization and formulating an answer to the question whether the book has been implemented as a communication medium. The methodology includes information retrieval, information monitoring and bibliometric analysis. The summary of the results follows the algorithm for information resonance of the book in 8 phases, developed by M. Tsvetkova. Information monitoring covers the period from 2020 to 2023. The conclusions are likely to be useful for theoretical discussions about the difference between buying a book and reading a book. Kawasaki disease (KD) is a childhood vasculitis syndrome and the coronary arteries are the main target of the vascular damage. The outcome is formation of coronary lesions (CL) that develop in 20%–25% of untreated children. KD is the leading cause of myocardial infarction in infancy. Its consequences among young people are acute coronary syndrome and susceptibility to early atherosclerosis, if the disease were to remain unrecognized. We investigated the cardiac manifestations during the acute phase of the disease, the values of the fever, C-reactive protein (CRP) and erythrocyte sedimentation rate (ESR) and the therapeutic factors that created increased coronary risk in our cohort of patients. The study is retrospective (1993–2014). In the cohort (n = 107), 30.8% had coronary lesions, including 19.6% coronary aneurysms and 11.2% significant dilatations. The subject of the research in the framework of the article is the international and national practice of regulation and implementation of activities in the field of tourism in the Central Ecological zone of the Baikal Natural Territory. As part of the analysis of the impact of tourism activities on the ecosystem of the World Heritage site "Lake Baikal", based on the materials of the authorities, conclusions are formulated about the possibility of deterioration of the ecological state of Lake Baikal and the emergence of social tension in the event of an increase in the number of tourists on its shores. The content of guidelines on the organization of tourism at UNESCO World Heritage sites is disclosed, the most successful international practices of tourism management are correlated with the implementation of regional programs for the development and organization of sustainable tourism. The conclusion is formulated about the importance of collecting information about the tourist infrastructure, statistical data. The program documents defining the policy in the field of tourism in the Central ecological zone of the Baikal Natural Territory are analyzed, the conclusion is made that there is no unified strategy for the development of Baikal as a World Heritage site. The factor of significant influence and participation of the local population in the implementation of tourist activities is noted, the need for the involvement of the local population in the planning of tourist activities is indicated. The analysis of the Global Code of Ethics of Tourism, the Framework Convention on the Ethics of Tourism. The main content of the Global Initiative to Reduce Plastic Consumption in Tourism and recommendations for improving procurement activities in the tourism sector are revealed. The problem of compliance with moral and ethical requirements by subjects of relations in the field of tourism is noted as one of the underestimated problems. The study resulted in recommendations both on the modernization of the existing domestic regulatory framework and the development of new documents in the field of sustainable tourism.

- Abstract
The article is a continuation of the presentation of the results of the study of the current state and consideration in retrospect of the existing cases of exclusion of UNESCO World Heritage sites from the World Heritage List. Focusing on the general problems of conservation of World Heritage sites, the authors extrapolate the findings to the situation in the field of conservation of the World Natural Heritage site Lake Baikal in order to develop proposals to improve the situation in the field of ensuring its protection. In particular, the problem of insufficient certainty and clarity in establishing the boundaries of World Heritage sites is noted, in order to solve which a proposal is made to consolidate the regime of World Natural heritage sites in the law "On Specially Protected Natural Territories", with the mandatory formation of a unified administration and approval of the boundaries of such a protected area by an act of the Russian Government, taking into account the requirements of the World Heritage Convention and Guidelines for the application of the Convention when they change. The problem of preserving the appearance of cultural heritage is also considered, since the issue of aesthetic perception of the lake and its immediate surroundings is considered important from the point of view of fulfilling obligations under the Convention on the Protection of World Heritage (taking into account the fact that Baikal is recognized as a World Heritage by all four criteria for assessing outstanding universal value). The lack of representation of protection of natural beauty at the national level is noted, since the current legislation does not contain tools for the protection of Baikal landscapes. Based on the results of the analysis of the world practice of excluding World Heritage sites from the List, the authors conclude that the extrapolation of the main reasons for such exclusionof Lake Baikal as a World Natural Heritage site necessitates the state to develop and make appropriate adjustments to the legislation on the protection of Lake Baikal and to the practice of its application, regardless of the relevant UNESCO events.
