Purpose: To examine the sleep of rugby league athletes the night before and following an evening match and to investigate the association between caffeine intake and sleep on the night of competition. Methods: On the night prior to, night of, and night after a professional rugby league match, 15 athletes had their sleep monitored using wrist activity monitors. Additionally, saliva samples were collected 60 minutes before and 30 minutes after the competition to assess salivary caffeine concentration. Differences in sleep across the 3 nights were examined using linear mixed models, and changes were assessed using effect size (ES). Pearson correlation (r) assessed the relationship between salivary caffeine levels and sleep indices. Results: On the night of competition, athletes went to bed later than the night before (P = .00002, ES = 1.84) and night after (P = .0003, ES = 1.49) competition. Consequently, their sleep duration was reduced on the night of competition compared with the previous night (P P = .001, ES = 1.53). Postcompetition salivary caffeine concentration was substantially elevated in athletes when compared with precompetition measures (P r = .48, P = .07), increased sleep latency (r = .45, P = .09), decreased sleep duration (r = −.30, P = .28), and reduced sleep efficiency (r = −.34, P = .22). Conclusions: These results demonstrate that evening competition results in sleep disturbance in rugby league athletes, and caffeine supplementation prior to and during competition leads to substantial increases in postcompetition salivary caffeine concentration.

Aluminum components used in aerospace structures are commonly coupled with stainless steel fasteners. These through-hole geometries on the aluminum substrate cause a concentrated stress field. The high-stresses at the fastener sites can preferentially initiate coating damage allowing for moisture ingress, which can lead to the formation of a galvanic couple between the aluminum alloy and the stainless steel fastener. Corrosion damage is known to cause early initiation of fatigue cracks, thus severely reducing the total life of the component. This work aims to understand the relative impact and interaction of fastener hole geometry-induced stress concentrations and corrosion damage on the fatigue crack initiation behavior and total fatigue life. Specifically, by imparting various levels of corrosion severities at different locations within the macro-scale stress field, the relative impact of each on the fatigue process can be determined. This work demonstrated a dominant role of the macro-scale stress field on the ability of corrosion morphologies to initiate fatigue cracks. Specifically, crack formation was found to preferentially occur at high-stress regions in lieu of forming at lower-stress regions, regardless of corrosion severity, and corrosion severity in the through-hole had a significant, but nonpredictive, correlation with the total fatigue life. Critically, the findings of this work will inform the means by which coatings are evaluated and will serve as a controlled validation of experiments for fracture mechanics modeling.

The ability of a rugby league player to express high levels of strength and power is crucial for success in competition. Although researchers have shown relationships between physical attributes and performance, there is a lack of information related to changes in strength and power across various phases of the season. The purpose of this review was to identify the magnitude of change in muscular strength and power during different phases of an elite rugby league season. Percentage change and effect size were calculated to evaluate the magnitude of changes in strength and power. Meta-analyses were conducted to provide pooled estimates and 95% confidence intervals. Twelve studies were included; six reported changes following pre-season training, two during the competition phase and four examined changes over multiple seasons. Pooled estimates indicated large increases in upper and lower body strength 0.81 [0.43-1.19] and 1.35 [0.79-1.91], respectively, following pre-season training. Studies incorporating predominantly heavier loads (p 80% 1 RM) in training reported greater increases in maximal strength than studies completing lighter loads. Four studies used a wave-like programming strategy to obtain large improvements in strength and power 0.91 [0.36-1.45] and 0.90 [0.22-1.57], over multiple seasons. The results of this review highlight the limited current evidence and provides a preliminary reference point for strength and conditioning coaches aiming to develop and maintain strength and power across various stages, and over multiple seasons. Importantly, the results also indicate that higher loads result in greater increases in strength than lower loads.

Objective To determine if recent evolutions in laboratory protocols, including the increased use of natural cycles and the use of a hyaluronan-containing transfer medium, affected the rate of monozygotic twin (MZT) pregnancies after single frozen embryo transfer (FET). Design Retrospective cohort study. Setting Urban university-based fertility center. Patient(s) Patients who underwent single FET between January 2016 and December 2018 resulting in an intrauterine pregnancy. Intervention(s) Transition to a transfer protocol with a hyaluronan-containing transfer medium in July 2017. Main Outcome Measure(s) Number of MZT pregnancies. Result(s) There were 1,619 cycles that met the inclusion criteria and 31 (1.9%) resulted in MZT pregnancies. A hyaluronan-containing transfer medium was used in 875 (54.1%) cycles. Programmed cycles were used for 1,385 (85.5%) FETs and 234 (14.5%) cycles were natural. The mean age at FET, oocyte age, endometrial echo thickness, inner cell mass grade, trophectoderm grade, expansion, and day of blastocyst vitrification were similar between the groups. The use of a hyaluronan-containing transfer medium resulted in fewer MZTs. After controlling potential confounders with a multivariate regression, the use of the hyaluronan-containing medium still resulted in fewer MZTs. Monozygotic twins were colinear with preimplantation genetic testing (PGT), so PGT was excluded as a variable in our regression. A regression of PGT only cycles showed that the use of the hyaluronan-containing medium was still associated with a reduction in MZT pregnancies. Conclusion(s) The use of a hyaluronan-containing transfer medium was associated with a lower rate of MZTs. Other clinical parameters, including cycle type, were not associated with changes in the number of MZTs. The use of PGT needs to be further investigated as a risk factor for MZTs.

Recent developments in the 3D printing of austenitic stainless steels have led to the need for standardization of electrochemical techniques used to assess the corrosion performance of these alloys. Currently, ASTM standards for austenitic stainless focus on assessing their resistance to different modes of corrosion such as pitting, crevice, and intergranular corrosion. Due to the complexity of the additive process, selective corrosion occurs in microstructural features such as cellular structures and melt pool boundaries. Standardized corrosion testing needs to incorporate these microstructural features. This study characterizes the corrosion behavior of laser powder bed fusion stainless steel in a variety of ASTM standards with special attention to melt pool boundary dissolution, cellular structures, and intergranular corrosion.

ONE OF THE ABUNDANT UNCANNY FEATURES of the Covid-19 lockdowns was the still photography and video footage that emerged of cities deserted, emptied of their ordinary human activities, in broad daylight. While commentary sometimes focused on the potentially positive impact on air quality and the renewed sense of a natural presence taking hold in the urban space – thereby underlining the challenges posed by ‘business as usual’ in a dynamic of permanent activity and expansion – there was also an awareness that here, revealed in an exceptional way, were the infrastructure and decor of the urban phenomena stripped of their human actors.1 The uncanniness of such a revelation was that it showed, at one and the same time, the enabling constructions of ongoing urbanity and interdependence (matters of acute collective awareness in the circumstances) together with an urban world as if after the end of human presence: a prefiguration of post-apocalypse; the ‘day after’ become the day underway.

The figure of Baudelaire could be argued to have been conscripted into an excessive amount of paradigmatic constructions over the years. He becomes the name, or cultural face, of a moving configuration of essential problems in discussions of artistic work and subjectivity, and of the ‘modern regime’ – separately and in conjunction. This article analyzes Baudelaire’s afterlife as a mythological one and examines how, across a selection of ‘moments’ over the past three-quarters of a century, the pre-eminence of that figure can come to obscure the traces of a lived – synchronous – process in the oeuvre. Our ability to reconnect with those traces, it seeks to suggest, is key to an understanding of the continued ability of the Baudelaire figure to address our contemporary scene. Moving from Bourdieu’s construction of Baudelaire as nomothète in his sociology of the literary field, it revisits rival co-optations by Jouve and Sartre, in the service of aesthetic and critical ethical accounts respectively, before examining instances of the interweaving of these strands in a brief survey of broadly contemporary work. This survey concludes with a more extended focus on work by contemporary poet Cédric Demangeot, and suggests that the ‘poetic margin’ is where Baudelaire’s powerfully integrated navigation of the adversities of life and of art continues to resonate with greatest urgency.

Purpose: The aim of this study was to map responses over 32 hours following high-load (HL) and moderate-load (ML) half-squat priming. Methods: Fifteen participants completed control, HL (87% 1RM), and ML (65% 1RM) activities in randomized, counterbalanced order. Countermovement jump (CMJ), squat jump (SJ), saliva testosterone, saliva cortisol, and perceptual measures were assessed before and 5 minutes, 8 hours, 24 hours, and 32 hours after each activity. Results are presented as percentage change from baseline and 95% confidence interval (CI). Cliff delta was used to determine threshold for group changes. Results: SJ height increased by 4.5% (CI = 2.2–6.8, Cliff delta = 0.20) 8 hours following HL. CMJ and SJ improved by 6.1% (CI = 2.1–7.8, Cliff delta = 0.27) and 6.5% (CI = 1.2–11.8, Cliff delta = 0.30), respectively, 32 hours after ML. No clear diurnal changes in CMJ or SJ occurred 8 hours following control; however, increases of 3.9% (CI = 2.9–9.2, Cliff delta = 0.26) and 4.5% (CI = 0.9–8.1, Cliff delta = 0.24), respectively, were observed after 32 hours. Although diurnal changes in saliva hormone concentration occurred (Cliff delta = 0.37–0.92), the influence of priming was unclear. Perceived “physical feeling” was greater 8 hours following HL (Cliff delta = 0.36) and 32 hours after ML and control (Cliff delta = 0.17–0.34). Conclusions: HL priming in the morning may result in small improvements in jump output and psychophysiological state in the afternoon. Similar improvements were observed in the afternoon the day after ML priming.

We studied the trends and various outcomes, including the readmission rates, health care utilization, and complications among living liver donors (LLDs) in the United States. We queried the National Database for data from 2010 to 2017 for all LLDs. The primary outcomes were 30-day and 90-day readmission rates. The secondary outcomes included health care use (length of stay [LOS], cost of care), index admission, and calendar-year mortality. Logistic regression models were fit for various outcomes. A total of 1316 LLDs underwent hepatectomy during the study period. The median donor age was 35.0 years (interquartile range, 27.4-43.6), and donors were predominantly women (54.2%). The trend of LLD surgeries remained stable at large medical centers (85.3%). The 30-day and 90-day readmission rates were low at 5% and 5.9%, respectively. Older age (50 years and older; 8%; confidence interval [CI], 0.6%-15.9%; P = 0.03) and hepatectomy at small to medium-sized hospitals were associated with increased index LOS (13.4%; 95% CI, 3.1%-24.7%; P = 0.01). Moreover, older age of donor (-11.3%; 95% CI, -20.3% to -1.4%; P = 0.03), Elixhauser score ≥3 (17%; 95% CI, 1.2%-35.3%; P = 0.03), and Medicaid insurance (24.5%; 95% CI, 1.2%-53.1%; P = 0.04) were also associated with increased cost. The overall rate of any complications during index admission was 42.8%. Male sex (odds ratio [OR], 1.63; 95% CI, 1.19-2.23) was an independent predictor of post-LLD complications. There was no index admission or calendar-year mortality reported during the study period. This is the largest national report of LLDs to date, showing that the trend of LLD surgeries is stable in the United States. With established safety, fewer complications, and less health care utilization, LLDs can be a potential source of continuation of liver transplantation in the context of changing liver allocation policies in the United States.

Background: The arterial pole of the heart is a hotspot for life-threatening forms of congenital heart defects (CHDs). Development of this cardiac region occurs by addition of Second Heart Field (SHF) progenitor cells to the embryonic outflow tract (OFT) and subsequently the base of the ascending aorta and pulmonary trunk. Understanding the cellular and genetic mechanisms driving arterial pole morphogenesis is essential to provide further insights into the cause of CHDs. Methods: A synergistic combination of bioinformatic analysis and mouse genetics as well as embryo and explant culture experiments were used to dissect the cross-regulatory transcriptional circuitry operating in future subaortic and subpulmonary OFT myocardium. Results: Here, we show that the lipid sensor PPARγ (peroxisome proliferator–activated receptor gamma) is expressed in future subpulmonary myocardium in the inferior wall of the OFT and that PPARγ signaling-related genes display regionalized OFT expression regulated by the transcription factor TBX1 (T-box transcription factor 1). Modulating PPARγ activity in ex vivo cultured embryos treated with a PPARγ agonist or antagonist or deleting Pparγ in cardiac progenitor cells using Mesp1-Cre reveals that Pparγ is required for addition of future subpulmonary myocardium and normal arterial pole development. Additionally, the non-canonical DLK1 (delta-like noncanonical Notch ligand 1)/NOTCH (Notch receptor 1)/HES1 (Hes family bHLH transcription factor 1) pathway negatively regulates Ppar γ in future subaortic myocardium in the superior OFT wall. Conclusions: Together these results identify Pparγ as a regulator of regional transcriptional identity in the developing heart, providing new insights into gene interactions involved in congenital heart defects.

Prognosticating is central to primary palliative care in neurology. Many neurologic diseases carry a high burden of troubling symptoms, and many individuals consider health states due to neurologic disease worse than death. Many patients and families report high levels of need for information at all disease stages, including information about prognosis. There are many barriers to communicating prognosis including prognostic uncertainty, lack of training and experience, fear of destroying hope, and not enough time. Developing the right mindset, tools, and skills can improve one's ability to formulate and communicate prognosis. Prognosticating is subject to many biases which can dramatically affect the quality of patient care; it is important for providers to recognize and reduce them. Patients and surrogates often do not hear what they are told, and even when they hear correctly, they form their own opinions. With practice and self-reflection, one can improve their prognostic skills, help patients and families create honest roadmaps of the future, and deliver high-quality person-centered care.

We sought to evaluate hospital outcomes of cirrhosis patients with nonvariceal upper gastrointestinal bleeding (NVUGIB).NVUGIB is common in patients with cirrhosis. However, national outcome studies of these patients are lacking.We utilized the 2014 Nationwide Readmission Database to evaluate NVUGIB in patients with cirrhosis, further stratified as no cirrhosis (NC), compensated cirrhosis (CC), or decompensated cirrhosis (DC). Validated International Classification of Diseases, Ninth Revision, Clinical Modification codes captured diagnoses and interventions. Outcomes included 30-day readmission rates, index admission mortality rates, health care utilization, and predictors of readmission and mortality using multivariable regression analysis.Overall, 13,701 patients with cirrhosis were admitted with NVUGIB. The 30-day readmission rate was 20.8%. Patients with CC were more likely to undergo an esophagogastroduodenoscopy (EGD) within 1 calendar day of admission (74.1%) than patients with DC (67.9%) or NC (69.4%). Patients with DC had longer hospitalizations (4.1 d) and higher costs of care ($11,834). The index admission mortality rate was higher in patients with DC (6.2%) than in patients with CC (1.7%, P0.001) or NC (1.4%, P0.001). Predictors of 30-day readmission included performing an EGD1 calendar day from admission (OR: 1.21; 95% CI, 1.00 to 1.46) and DC (OR: 1.78; 95% CI, 1.54 to 2.06). DC was a predictor of index admission mortality (OR: 3.68; 95% CI, 2.67 to 5.05).NVUGIB among patients with DC is associated with higher readmission rates, mortality rates, and health care utilization compared with patients with CC and NC. Early EGD is a modifiable variable associated with reduced readmission rates. Early identification of high-risk patients and adherence to guidelines may improve clinical outcomes.

Highly self-reactive T cells are censored from the repertoire by both central and peripheral tolerance mechanisms upon receipt of high-affinity TCR signals. Clonal deletion is considered a major driver of central tolerance; however, other mechanisms such as induction of regulatory T cells and functional impairment have been described. An understanding of the interplay between these different central tolerance mechanisms is still lacking. We previously showed that impaired clonal deletion to a model tissue-restricted antigen (TRA) did not compromise tolerance. In this study, we determined that murine T cells that failed clonal deletion were rendered functionally impaired in the thymus. PD-1 was induced in the thymus and was required to establish cell-intrinsic tolerance to TRA in CD8+thymocytes independently of clonal deletion. PD-1 signaling in developing thymocytes was sufficient for tolerance induction, however in some cases the absence of PD-L1-mediated signaling in the periphery resulted in broken tolerance at late time points. Finally, we showed that chronic exposure to high affinity antigen supported the long-term maintenance of tolerance. Taken together, our study identifies a critical role for PD-1 in establishing central tolerance in autoreactive T cells that escape clonal deletion. It also sheds light on potential mechanisms of action of anti-PD-1 pathway immune checkpoint blockade and the development of immune-related adverse events.Graphical Abstract

AA5456-H116 undergoes accelerated localized corrosion when in a galvanic couple with cupronickel alloy C70600 (CDA 706) during ASTM G85 Appendix 2 Wet Bottom (G85 WB) testing. Surface and subsurface damage of AA5456-H116 and the region of protection of the C70600 were investigated and quantified after 31 d (124 cycles). These results were compared to finite element modeling results investigating relative humidity (RH) and water layer (WL) thickness. The best fit between experimental results and the modeling results was found when modeling assumed that a 3,000 μm water layer was formed during the spray portion at the start of each G85 cycle, with thinner water layers present during the decreasing RH portion of the cycle, which led to intergranular corrosion attack that was primarily focused in the proximity of the C70600/AA5456-H116 interface, but extending along the AA5456-H116 surface for some distance. The high-water layer thickness is likely the result of the corrosion product formation that traps additional electrolyte than would be present on a clean surface.

Herein, a high-yielding cycloaddition reaction of β-ketoesters and azides to provide 1,2,3-triazoles is described. The reactions employing 2-unsubstituted β-ketoesters were found to provide 5-methyl-1,2,3-triazoles, whereas 2-alkyl-substituted β-ketoesters provided 5-hydroxy-1,2,3-triazoles (shown to be relatively acidic) in high yields and as single regioisomers. Several novel compounds were reported and characterized including long-chain 5-hydroxy-1,2,3-triazoles potentially bioisosteric to hydroxamic acids.

Multiple sclerosis (MS) is a disabling immune-mediated disease of the CNS and considered one of the health conditions with the highest annual spending growth paid by both public and private insurance.1 Health care spending in the United States for MS was reported as $13.9 billion in 2016.1 The estimated cumulated (over 10 years) prevalence of MS among adults in the United States in 2010 was approximately 727,000.2 In 2017, that number was substantially higher, approximately 914,000.2,3 The prevalence is higher among women, with a female: male ratio of 2.8.2 Improving high-quality care for these patients can lead to improved outcomes, reduced spending, and better quality of life.

Characterization of germinal center B and T cell responses yields critical insights into vaccine immunogenicity. Nonhuman primates are a key preclinical animal model for human vaccine development, allowing both lymph node (LN) and circulating immune responses to be longitudinally sampled for correlates of vaccine efficacy. However, patterns of vaccine Ag drainage via the lymphatics after i.m. immunization can be stochastic, driving uneven deposition between lymphoid sites and between individual LN within larger clusters. To improve the accurate isolation of Ag-exposed LN during biopsies and necropsies, we developed and validated a method for coformulating candidate vaccines with tattoo ink in both mice and pigtail macaques. This method allowed for direct visual identification of vaccine-draining LN and evaluation of relevant Ag-specific B and T cell responses by flow cytometry. This approach is a significant advancement in improving the assessment of vaccine-induced immunity in highly relevant nonhuman primate models.

American Heart Association/American Stroke Association guidelines recommend endovascular stroke therapy (EST) with recombinant tissue plasminogen activator (rt-PA) for eligible patients in acute ischemic stroke (AIS). Using the National Inpatient Sample database, we evaluated trends in treatment with rt-PA and EST for AIS and their outcomes.This is a cross-sectional observational study of patients with AIS admitted in US hospitals from 2012 to 2016. Patients were grouped into those who received rt-PA alone, EST alone, and rtPA+EST. Survey statistical procedures were performed. Multivariable regression analysis with pairwise comparisons of each treatment group with no treatment group was performed for discharge outcomes.The study included 2,290,520 patients with AIS with the mean age of 70.46 years. Treatment rates increased from 2012 to 2016 for rt-PA by 7% per year (5.86%-7.67%, odds ratio [OR] = 1.07, 95% confidence interval [CI]: 1.05-1.08) and EST by 38% per year (0.55%-1.75%, OR = 1.38, 95% CI: 1.31-1.45) but not rt-PA+EST (0.54%-0.57%, OR = 1.04, 95% CI: 0.99-1.08). The mean length of stay reduced from 2012 to 2016 for rt-PA (6.07-4.91 days, P0.0001) and rt-PA+EST (9.19-7.10 days, P = 0.0067) but not for EST (9.61-8.51 days, P = 0.5074). The odds of patients discharged home increased by 8%, 9%, and 15% among patients who received rt-PA alone, EST alone, and rt-PA+EST, respectively, compared with no treatment group.The utilization of rt-PA alone and EST alone increased but that of rt-PA+EST remained unchanged from 2012 to 2016 in the National Inpatient Sample.

Background and Purpose: Although most strokes present with mild symptoms, these have been poorly represented in clinical trials. The objective of this study is to describe multidimensional outcomes, identify predictors of worse outcomes, and explore the effect of thrombolysis in this population. Methods: This prospective observational study included patients with ischemic stroke or transient ischemic attack, a baseline National Institutes of Health Stroke Scale (NIHSS) score 0 to 5, presenting within 4.5 hours from symptom onset. The primary outcome was a 90-day modified Rankin Scale score of 0 to 1; secondary outcomes included good outcomes in the Barthel Index, Stroke Impact Scale-16, and European Quality of Life. Multivariable models were created to determine predictors of outcomes and the effect of alteplase. Results: A total of 1765 participants were included from 100 Get With The Guidelines-Stroke participating hospitals (age, 65±14; 42% women; final diagnosis of ischemic stroke, 90%; transient ischemic attack, 10%; 57% received alteplase). At 90 days, 37% were disabled and 25% not independent. Worse outcomes were noted for older individuals, women, non-Hispanic Blacks and Hispanics, Medicaid recipients, smokers, those with diabetes, atrial fibrillation, prior stroke, higher baseline NIHSS, visual field defects, and extremity weakness. Similar outcomes were noted for the alteplase-treated and untreated groups. Alteplase-treated patients were younger (64±13 versus 67±1.4) with higher NIHSS (2.9±1.4 versus 1.7±1.4). After adjusting for age, sex, race/ethnicity, and baseline NIHSS, we did not identify an effect of alteplase on the primary outcome but did find an association with Stroke Impact Scale-16 in the restricted sample of baseline NIHSS score 3–5. Few symptomatic intracerebral hemorrhages were recorded ( Conclusions: A large proportion of stroke patients presenting with low NIHSS have a disabled outcome. Baseline predictors of worse outcomes are described. An effect of alteplase on outcomes was not identified in the overall cohort, but a suggestion of efficacy was noted in the NIHSS 3–5 subgroup. The hallmarks of COVID-19 are higher pathogenicity and mortality in the elderly compared to children. Examining baseline SARS-CoV-2 cross-reactive immunological responses, induced by circulating human coronaviruses (hCoVs), is needed to understand such divergent clinical outcomes. Here we show analysis of coronavirus antibody responses of pre-pandemic healthy children (n = 89), adults (n = 98), elderly (n = 57), and COVID-19 patients (n = 50) by systems serology. Moderate levels of cross-reactive, but non-neutralizing, SARS-CoV-2 antibodies are detected in pre-pandemic healthy individuals. SARS-CoV-2 antigen-specific Fcγ receptor binding accurately distinguishes COVID-19 patients from healthy individuals, suggesting that SARS-CoV-2 infection induces qualitative changes to antibody Fc, enhancing Fcγ receptor engagement. Higher cross-reactive SARS-CoV-2 IgA and IgG are observed in healthy elderly, while healthy children display elevated SARS-CoV-2 IgM, suggesting that children have fewer hCoV exposures, resulting in less-experienced but more polyreactive humoral immunity. Age-dependent analysis of COVID-19 patients, confirms elevated class-switched antibodies in elderly, while children have stronger Fc responses which we demonstrate are functionally different. These insights will inform COVID-19 vaccination strategies, improved serological diagnostics and therapeutics., Antibody responses to SARS-CoV-2 are critical in the immune response to infection, but the potential cross-reactivity to other human corona viruses is poorly appreciated. Here the authors apply a systems based approach to characterise the antibody response in pre-pandemic cohorts and assess heterotypic reactivity to SARS-CoV-2.

Elevated atmospheric CO2 (eCa ) may benefit plants during drought by reducing stomatal conductance (gs ) but any 'water savings effect' could be neutralized by concurrent stimulation of leaf area. We investigated whether eCa enhanced water savings, thereby ameliorating the impact of drought on carbon and water relations in trees. We report leaf-level gas exchange and whole-plant and soil water relations during a short-term dry-down in two Eucalyptus species with contrasting drought tolerance. Plants had previously been established for 9 to 11 months in steady-state conditions of ambient atmospheric CO2 (aCa ) and eCa , with half of each treatment group exposed to sustained drought for 5 to 7 months. The lower stomatal conductance under eCa did not lead to soil moisture savings during the dry-down due to the counteractive effect of increased whole-plant leaf area. Nonetheless, eCa -grown plants maintained higher photosynthetic rates and leaf water potentials, making them less stressed during the dry-down, despite being larger. These effects were more pronounced in the xeric species than the mesic species, and in previously water-stressed plants. Our findings indicate that eCa may enhance plant performance during drought despite a lack of soil water savings, especially in species with more conservative growth and water-use strategies.

Highlights • Nutrient concentration thresholds based on pressure-response models are needed for management. • Nutrient thresholds were derived from relationships of phytoplankton and macrophytes against nutrients. • Very shallow eastern continental lakes do not demonstrate a strong response to nutrients. • Fish stocking is the dominant pressure in these highly eutrophic shallow lakes. • These understudied lakes challenge the dominant paradigm of lake assessment and management. Nutrient targets based on pressure-response models are essential for defining ambitions and managing eutrophication. However, the scale of biogeographical variation in these pressure-response relationships is poorly understood, which may hinder eutrophication management in regions where lake ecology is less intensively studied. In this study, we derive ecology-based nutrient targets for five major ecoregions of Europe: Northern, Central-Baltic, Alpine, Mediterranean and Eastern Continental. As a first step, we developed regressions between nutrient concentrations and ecological quality ratios (EQR) based on phytoplankton and macrophyte communities. Significant relationships were established for 13 major lake types; in most cases, these relationships were stronger for phosphorus than for nitrogen, and stronger for phytoplankton than for macrophytes. Using these regressions, we estimated the total phosphorus (TP) and total nitrogen (TN) concentrations at which lakes of different types are likely to achieve good ecological status. However, in the very shallow lakes of the Eastern Continental region, relations between nutrient and biological communities were weak or non-significant. This can be attributed to high nutrient concentrations (in the asymptotic zone of phosphorus-phytoplankton models) suggesting other factors (light, grazing) limit primary production. However, we also show that fish stocking is a major pressure on Eastern Continental lakes, negatively affecting ecological status: lakes with low fish stocking show low chlorophyll-a concentrations and good ecological status despite high nutrient levels, while the lakes with high fish stocking show high chlorophyll-a and low ecological status. This study highlights the need to better understand lakes in biogeographic regions that have been, for historical reasons, less studied. This, in turn, helps reveal factors that challenge the dominant paradigms of lake assessment and management.

Predators are among the most threatened animal groups globally, with prey declines contributing to their endangerment. However, assessments of the habitat conditions that influence the successful capture of different prey species are rare, especially for small, cryptic predators. Accordingly, most predator conservation plans are based on the relative importance of habitats inferred from coarse-scale studies that do not consider habitat features contributing to hunting success, which can vary among prey species. To address this limitation, we integrated high-resolution global positioning system tracking and nest video monitoring to characterize habitat features at prey capture locations during the nestling provisioning stage for the Spotted Owl (Strix occidentalis) a small, cryptic predator that has been at the center of a decades-long forest management conflict in western North America. When all prey species were considered collectively, males provisioning nests tended to capture prey: (1) in areas with more large-tree forest, (2) in areas with more medium trees/medium canopy forest, and (3) at edges between conifer and hardwood forests. However, when we considered the owl’s two key prey species separately, males captured woodrats (Neotoma fuscipes) and Humboldt flying squirrels (Glaucomys oregonensis) in areas with markedly different habitat features. Our study provides clarity for forest management in mixed-ownership landscapes because different prey species achieve high densities in different habitat types. Specifically, our results suggest that promoting large trees, increasing forest heterogeneity, and creating canopy gaps in forests with medium trees/high canopy cover could benefit Spotted Owls and their prey, which has the ancillary benefit of enhancing forest resilience. Combining high-resolution global positioning system tagging with video-based information on prey deliveries to breeding sites can strengthen conservation planning for small predators by more rigorously defining those habitat features that are associated with successful prey acquisition.

To evaluate toxicity, quality of life and PFS in patients with advanced ovarian cancer who underwent neoadjuvant chemotherapy (NAC) followed by CRS and HIPEC with carboplatin.Patients with stage IIIC or IVA epithelial ovarian cancer, who were not candidates for primary CRS, were enrolled in this phase two trial. Patients received 3-6 cycles of NAC with an IV carboplatin doublet followed by CRS with HIPEC (carboplatin 800 mg/mTwenty patients were enrolled. HIPEC was completed successfully in all twenty patients, and there was no peri-operative mortality. Twelve (70.6%) patients experienced a grade 3 or 4 toxicity; most commonly anemia (59%), thrombocytopenia (29%), and hypokalemia (24%). There was no significant change between the pre-operative and postoperative 6 weeks, 3 month, and 6 month FACT-O, NTX, and AD scores. Nine (45%) patients have experienced disease recurrence to date. The median progression free survival in this cohort is 11.2 months (2.5-23.7 months).The addition of HIPEC with carboplatin to interval CRS was well tolerated in patient population. Myelosuppression was the most common adverse event. CRS with HIPEC did not adversely impact these patients' QOL indices. The efficacy of this regimen should be further evaluated in a larger clinical trial.
