104. Molecular Dynamics Simulation of CO2 Hydrate Growth and Intermolecular Weak Interaction Analysis.

Abstract

The growth of carbon dioxide (CO2) hydrate in CO2 aqueous solution (box A), and in CO2 aqueous solution with structure I (sI) CO2 hydrate cell as seed (box B) under the same conditions of 275 K and 10 MPa, were simulated respectively. The total simulation time is 5000 ns. The nucleation process is very slow, so it is difficult to observe CO2 hydrate growth in box A without CO2 hydrate seed, CO2 just accumulates to form bubbles, while H2O still remains liquid, the F4 order parameter remains about –0.1. By contrast, beautiful CO2 hydrate in box B grows rapidly, CO2 is trapped in the cages formed by H2O molecules, F4 rises from 0.12 to 0.7 due to the formation of sI hydrate. The density of CO2 hydrate in box B is as high as 1.25 g/cm3, higher than that of seawater, which is beneficial to its permanent sealing on the seabed. In box B, in accordance with the independent gradient model based on Hirshfeld partition (IGMH) analysis, a H2O molecule forms four hydrogen bonds with surrounding H2O molecules, CO2 only has van der Waals interaction with surrounding water cages. In the actual large-scale CO2 storage process, especially for the rapid storage of CO2 in the form of hydrate, it is strongly recommended to pre-add CO2 hydrate crystals into the solution to achieve the purpose of rapid growth of CO2 hydrate. 120. Understanding the uniaxial-deformation behavior of stable and unstable CO2hydrates under squeezing and stretching conditions

Abstract

As an efficient method for lowering carbon emissions, sealing carbon dioxide (CO2) on the seabed with CO2hydrates has gained extensive interest from researchers in recent decades. Unfortunately, the underwater environment is not always quiet and peaceful, and crustal movement may impair the storage environment, making it unsuitable for storing CO2. To better understand how CO2hydrates change when subjected to external forces at the molecular level, we used the molecular dynamics method to simulate the behavior of stable and unstable CO2hydrates under squeezing and stretching conditions, respectively. According to the results, a significant amount of CO2is released alongside the transformation of a huge number of solid water molecules into liquid during the squeezing process. In the stretching process, the CO2hydrate can be pulled apart rapidly, with only a small area of the hydrate cage being damaged and some CO2escaping, and only the water molecules near the fracture surface changing from solid to liquid state. The three-body order parameter (F3) and the four-body order parameter (F4), water molecules number in different phase, and the hydrate cage number, all demonstrate the process of water molecules transitioning from solid hydrate form to liquid state. It can be concluded that, to avoid massive CO2emissions, it is necessary to anticipate geological changes and choose suitable hydrate storage sites in advance. In addition, new advancements in CO2conversion technology is necessary for safeguarding our planet.

Abstract

CBT-Cys click condensation reaction has a high second-order reaction rate constant and has found wide applicability in recent years., CBT-Cys click condensation reaction has a high second-order reaction rate constant and has found wide applicability in recent years. However, its reaction mechanism has not been experimentally validated and its application for identifying bioactive N-terminal Cys peptides in real clinical samples has not been reported. Herein, firstly, by employing induced nanoelectrospray ionization-mass spectrometry (InESI-MS) and a home-built micro-reactor, we successfully intercepted and structurally characterized the crucial intermediate in this click reaction for the first time. With the intermediate, the proposed mechanism of this reaction was corroborated. Moreover, we also applied this MS setup to monitor the reaction in real time and obtained the second-order reaction rate constants of this reaction at different pH values. After mechanistic study, we applied this click reaction for identifying bioactive N-terminal cysteine peptides in amniotic fluid (AF). Eight unique N-terminal Cys peptides in AF, three of which are located in the functional domain regions of their corresponding proteins, were identified with a false positive rate less than 1%. One of the three peptides was found able to inhibit the growth of uterine endometrial cancer HEC-1-B cells but not the endometrial normal cells via a typical apoptotic pathway. With its mechanism satisfactorily elucidated, the kinetic parameters obtained, as well as its application for fishing bioactive N-terminal Cys peptides from vast complex clinical samples, we anticipate that this CBT-Cys click reaction could be applied more widely for the facile isolation, site-specific identification, and quantification of N-terminal Cys-containing peptides in complex biological samples.

Abstract

Objectives: Whether the maternal vitamin D deficiency is associated with preeclampsia is still an argument. We aimed to assess the association between maternal serum 25-hydroxyvitamin D [25(OH)D] concentrations and risk of preeclampsia in a Chinese population and systematically evaluate published evidence on this association.Methods: We conducted a nested case-control study involving 122 pregnant women with preeclampsia and 488 pregnant women whose blood pressure was within the normal range (as controls). For further meta-analysis, 20 studies and our study were included for the final pooled analysis, involving 39,031 participants and 3305 preeclampsia cases with various ethnicities.Results: The results showed that 65.6% of women with preeclampsia had serum 25(OH)D concentrations <50.0 nmol L-1 compared with 55.3% of women in controls. The 25(OH)D concentrations were significantly lower in women with preeclampsia than controls [Median (IQR), women with preeclampsia versus controls: 43.3 (35.5, 55.2) versus 47.5 (37.6, 60.4) nmol L-1, p = .014]. For women with 25(OH)D concentrations <50.0 nmol L-1, they had a 65% increase in preeclampsia risk (95% CI = 1.02-2.69), as compared with women with 25(OH)D concentrations from 50.0 to 74.9 nmol L-1. Further, meta-analysis showed that low 25(OH)D concentrations were associated with a significantly increased risk of preeclampsia by 62% (pooled OR = 1.62, 95%CI = 1.36-1.94), and the risk effect of low 25(OH)D concentrations existed in most subgroups.Conclusions: Low 25(OH)D concentration in pregnancy was significantly associated with preeclampsia risk, and it may serve as biomarkers for the surveillance of high-risk pregnant women. Abstract

Breast cancer has become the most common cancer in women, and nontriple negative breast cancer (non‐TNBC) accounts for 80–90% of all invasive breast cancers. Early detection, diagnosis, and treatment are considered key to a successful cure. Conventionally, breast imaging and needle core biopsy are used for detection and monitoring. However, small variations in volume might be ignored in imaging, and traditional biopsies are spatially and temporally limited, leading to a significant delay in cancer detection and thus prompting renewed focus on early and accurate diagnosis. In this article, we investigated whether there is an accurate molecule in peripheral blood that can help diagnose breast cancer. Similar to microRNAs, tRNA‐derived fragments (tRFs) have been reported to be involved in many pathological processes in breast cancer, but whether they can serve as candidate biomarkers for breast cancer remains unclear. Using high‐throughput sequencing technology, we identified 4,021 differentially expressed tRFs in normal and breast cancer cell lines, and eight tRFs were selected to establish a signature as a predictive biomarker of non‐TNBC. Furthermore, quantitative reverse‐transcriptase polymerase chain reaction was performed to verify the expression of the signature and analyze the correlation between dysregulated tRFs and breast cancer. The results indicated that tDR‐7816, tDR‐5334, and tDR‐4733 might be promising biomarkers. Through further bioinformatics analysis, we predicted that tDR‐7816 influences the xenobiotic metabolic processes that support the oncogenesis of breast cancer. In summary, our results provide a rationale for using circulating tDR‐7816 expression as a novel potential biomarker for the diagnosis of patients with early non‐TNBC. [ABSTRACT FROM AUTHOR]
