105. Posterior corneal stability after small incision lenticule extraction with different optical zones

Abstract

Background: To assess the changes in the posterior corneal surface following small incision lenticule extraction (SMILE) with different optical zones. Methods: In this retrospective study, 106 eyes of 106 patients who underwent SMILE were recruited 3 years after the procedure. Eyes were divided into two groups according to the size of the surgical optical zone: group A (52 eyes, ≤6.2 mm) and group B (54 eyes, ≥6.5 mm). Posterior central elevation (PCE) and 12 other points at 45°, 135°, 225° and 315° with distances of 1 mm, 2 mm and 3 mm from the centre were recorded from Pentacam. Results: No iatrogenic keratectasia was identified, and eyes in the two groups showed comparable visual results. The overall trend in posterior corneal elevation changes was consistent for both groups. PCE decreased significantly from 1.33 ± 2.32 to 0.75 ± 2.41 in group A (P= 0.024) and from 0.87 ± 2.61 to 0.06 ± 2.74 in group B (P= 0.003). All points in the central 2 mm region in both groups were reduced postoperatively. In the 4 mm and 6 mm corneal annulus, almost all points at 225°and 315° showed backward displacement, with the most prominent change occurring at 315° in the 6 mm annulus (P< 0.001), indicating no forward protrusion in the inferior area. Conclusions: No forward protrusion in the posterior corneal surface was observed 3 years after SMILE with different optical zones. Comprehensive preoperative measurements are essential for ensuring corneal stability and avoiding iatrogenic keratectasia.

Abstract

The reliability of energy systems is strongly influenced by the prevailing climate conditions. With the increasing prevalence of renewable energy sources, the interdependence between energy and climate systems has become even stronger. This study examines the impact of different spatial resolutions in climate modeling on energy grid reliability assessment, with the Texas interconnection between 2033 and 2043 serving as a case study. Our results indicate that while lower-resolution climate simulations can provide a rough estimate of system reliability, higher-resolution simulations can identify more frequent extreme events during the summer months and produce more precise assessments of the timing and severity of the worst blackout event.

Abstract

Since its first demonstration, stimulated Raman scattering (SRS) microscopy has become a powerful chemical imaging tool that shows promise in numerous biological and biomedical applications. The spectroscopic capability of SRS enables identification and tracking of specific molecules or classes of molecules, often without labeling. SRS microscopy also has the hallmark advantage of signal strength that is directly proportional to molecular concentration, allowing for in situ quantitative analysis of chemical composition of heterogeneous samples with submicron spatial resolution and subminute temporal resolution. However, it is important to recognize that quantification through SRS microscopy requires assumptions regarding both system and sample. Such assumptions are often taken axiomatically, which may lead to erroneous conclusions without proper validation. In this review, we focus on the tacitly accepted, yet complex, quantitative aspect of SRS microscopy. We discuss the various approaches to quantitative analysis, examples of such approaches, challenges in different systems, and potential solutions. Through our examination of published literature, we conclude that a scrupulous approach to experimental design can further expand the powerful and incisive quantitative capabilities of SRS microscopy.

124. Channel Semantic Enhancement-Based Emotional Recognition Method Using SCLE-2D-CNN

Abstract

The existing EEG emotion classification methods have some problems, such as insufficient emotion representation and lack of targeted channel enhancement module due to feature redundancy. To this end, a novel EEG emotion recognition method (SCLE-2D-CNN) combining scaled convolutional layer (SCLs), enhanced channel module and two-dimensional convolutional neural network (2D-CNN) is proposed. Firstly, the time-frequency features of multi-channel EEG emotional signals were extracted by stacking scl layer by layer. Secondly, channel enhancement module is used to reassign different importance to all EEG physical channels. Finally, 2D-CNN was used to obtain deep local spatiotemporal features and complete emotion classification. The experimental results show that the accuracy of SEED data set and F1 are 98.09% and 97.00%, respectively, and the binary classification accuracy of DEAP data set is 98.06% and 96.83%, respectively, which are superior to other comparison methods. The proposed method has a certain application prospect in the recognition of human mental state.

Abstract

Influenza A virus (IAV) hijacks the endocytosis of host cells in the early stages of infection. It is known that microfilaments and microtubules play a vital role in viral infections. However, the potential involvement of intermediate filaments (IFs) and their mechanism in IAV infection are unclear. In this study, we combined quantum dot-based single-virus tracking (QSVT) with conventional biological methods to quantitatively analyze the effects of vimentin IFs on the early stages of IAV infection. The results showed that the knockdown of vimentin did not affect the internalization and motion dynamics of IAV but impacted the transport of IAV to late endosomes. Statistical analysis revealed that the knockdown of vimentin caused more than 20% IAV to remain in the early endosomes after rapid movement than in normal cells and reduced the efficiency of subsequent membrane fusion and genome release. These results suggested that vimentin facilitates IAV infection by participating in the maturation of endosomes. Our study expands the understanding of the mechanism of endocytosis of IAV and fills the gap in the knowledge on vimentin IFs. [ABSTRACT FROM AUTHOR]

Abstract

The dorsal raphe nucleus (DRN) has previously been proved to be involved in the regulation of the sleep–wake behavior. DRN contains several neuron types, such as 5-HTergic and GABAergic neurons. GABAergic neurons, which are the second largest cell subtype in the DRN, participate in a variety of neurophysiological functions. However, their role in sleep–wake regulation and the underlying neural circuitry remains unclear. Herein, we used fiber photometry and synchronous electroencephalogram (EEG)/electromyography (EMG) recording to demonstrate that DRN GABAergic neurons exhibit high activities during wakefulness and low activities during NREM sleep. Short-term optogenetic activation of DRN GABAergic neurons reduced the latency of NREM-to-wake transition and increased the probability of wakefulness, while long-term optogenetic activation of these neurons significantly increased the amount of wakefulness. Chemogenetic activation of DRN GABAergic neurons increased wakefulness for almost 2 h and maintained long-lasting arousal. In addition, inhibition of DRN GABAergic neurons with chemogenetics caused a reduction in the amount of wakefulness. Finally, similar to the effects of activating the soma of DRN GABAergic neurons, optogenetic stimulation of their terminals in the ventral tegmental area (VTA) induced instant arousal and promoted wakefulness. Taken together, our results illustrated that DRN GABAergic neurons are vital to the induction and maintenance of wakefulness, which promote wakefulness through the GABAergic DRN-VTA pathway.
