Diagnostic evaluations and early interventions of patients with bipolar disorder (BD) rely on clinical evaluations. Smartphones have been proposed to facilitate continuous and fine-grained self-monitoring of symptoms. The present study aimed to (1) validate daily smartphone-based self-monitored mood, activity, and sleep, against validated questionnaires and clinical ratings in young patients with newly diagnosed BD, unaffected relatives (UR), and healthy controls persons (HC).

The effect of monetary reward amount and test food type as factors of triangle test accuracy and assessment time for untrained panelists was explored. Monetary compensation is commonly used to reward panelists for their time and effort. While studies have documented that paying panelists can influence hedonic ratings, research on its possible influence on triangle test accuracy or time to assess products is lacking. Relatedly, some studies suggest that assessment time influences accuracy. Furthermore, little research has been conducted on the effect of the test food itself, and its general likeability, on biasing panelist accuracy or assessment time. Pairs of two liked foods – chocolate chip cookies and cheddar cheese, and two not-as-liked-foods – green olives and lima beans, were tested. In addition to correlating the two response variables with the main effects, interactions of overall expected hedonic liking of the test foods, panelist age, gender, time of day of the test, and day of the week the test were analyzed. Results indicate monetary compensation amount did not influence panelist accuracy or assessment time. However, accuracy increased significantly with longer assessment times; the effect of the “liked” vs. “not-as-well-liked” food categorizations were inconclusive. Expected hedonic liking, gender, age, and time of day of the test, were significant, suggesting that test accuracy and assessment time are likely influenced by multiple intrinsic and extrinsic factors. Overall results suggest that the use of broad demographics of untrained consumers for triangle tests results in data not consistently or strongly biased by payment amount or food type.

Pathogens in low water activity foods are an important emerging food safety concern due to notable survival and thermotolerance. Limited but growing data are publicly available to compare the thermotolerance of Listeria monocytogenes, Salmonella spp., and Enterococcus faecium NRRL B-2354 (a Salmonella surrogate). The aims of this study were to determine and assess bacterial thermal inactivation rates of two pathogens and a well-established surrogate across three different low-moisture food matrices containing different levels of fat using a dry inoculation method. Three low-moisture foods (0.11 aw, 45% fat peanut butter; 0.20 aw, 27% fat powder infant formula; and 0.45 aw, peanut butter > wheat flour for the vast majority of the comparable D-values. While Salmonella is the primary pathogen of concern in low-moisture foods due to survival and outbreaks, these results indicate L. monocytogenes can exhibit similar thermotolerances in low-water activity food matrices including peanut butter and powder infant formula.

Many Mexican women are deficient in folic acid. Fortification of the corn tortilla could be an effective way to help increase the folic acid levels among the Mexican population. Previous studies have shown significant folic acid losses in the masa dough as it is held before baking. This loss in folic acid could be owing to utilization by lactic acid bacteria naturally present in the masa. The objective of this study was to determine the effect of bacteria native to corn masa on the folic acid content in masa. Bacteria in dough samples from six mills in Guadalajara, Mexico, were isolated and identified. Bacterial isolates were inoculated into sterile fortified corn masa flour, which was converted to masa and held at 56°C for 0, 3, and 6 h, replicating the conditions of freshly milled masa as held before baking. Salmonella is a common pathogen which has been the cause of foodborne illness outbreaks implicating a variety of commodities, including low-moisture foods such as flour. Salmonella costs more than any other pathogen in the United States in terms of health care expenses and time of lost work. Heat treatment can be used to reduce Salmonella and other pathogens in flour to safe levels. However, in low-moisture foods, process times must be increased to achieve adequate lethality, possibly resulting in changes in the flour's functionality such as changes in the gluten quality, vitamin content, and the level of starch gelatinization. There is a need to determine the minimal heat treatment required to achieve desired lethality in flour and other low-moisture foods, with the goal of retaining the flour's functionality. Currently there is no published data about a nonpathogenic bacterial surrogate for Salmonella in flour. In this study, a surrogate, which closely matches the thermal death rate of Salmonella in flour, has been isolated. The surrogate was identified following an evaluation of thermal death curves of ten different strains of bacteria isolated from heat-treated flour and two nonpathogenic surrogates used in other commodities. Flour samples were inoculated with Salmonella or one of the twelve bacterial isolates, and then subjected to heat (70, 75, and 80 °C) for 12-60 min. This research was conducted to evaluate the effectiveness of a concentrated buffered vinegar product (CBV) and a simple buffered vinegar product (BV) for controlling Clostridium perfringens outgrowth during extended cooling times of ready-to-eat roast turkey and roast beef. Whole turkey breasts and beef inside rounds were injected with a typical brine and then ground and mixed with CBV (0.0, 2.01, 2.70, and 3.30% [w/w]) or BV (0.0, 1.75, 2.25, and 3.75% [w/w]) and a three-strain C. perfringens spore cocktail to a detectable level of ca. 2 to 3 log CFU/g. The meat was divided into 10-g portions, vacuum packaged, and stored frozen until tested. The turkey and beef were cooked in a programmable water bath to 71.6°C (160.8°F) in 5 h and to 57.2°C (135°F) in 6 h, respectively. The cooked turkey and beef were then cooled exponentially from 48.9 to 12.8°C (120 and 55°F) in 6, 9, 12, 15, and 18 h for the five cooling treatments. The cooling continued until the temperature reached 4.4°C (40°F). C. perfringens counts were determined at 54.4°C (130°F) and 4.4°C. CBV at 2.01% effectively limited C. perfringens growth in turkey to ≤1 log CFU/g with up to a 9-h cooling treatment, and 2.70 and 3.30% solutions were effective with up to the 18-h cooling treatment. BV had an inhibitory effect on C. perfringens outgrowth in beef but did not limit growth to ≤1 log CFU/g at any concentration tested for any of the cooling treatments.

Spatial control of G-protein-coupled receptor (GPCR) signaling, which is used by cells to translate complex information into distinct downstream responses, is achieved by using plasma membrane (PM) and endocytic-derived signaling pathways. The roles of the endomembrane in regulating such pleiotropic signaling via multiple G-protein pathways remain unknown. Here, we investigated the effects of disease-causing mutations of the adaptor protein-2 σ subunit (AP2σ) on signaling by the class C GPCR calcium-sensing receptor (CaSR). These AP2σ mutations increase CaSR PM expression yet paradoxically reduce CaSR signaling. Hypercalcemia-associated AP2σ mutations reduced CaSR signaling via Gαq/11 and Gαi/o pathways. The mutations also delayed CaSR internalization due to prolonged residency time of CaSR in clathrin structures that impaired or abolished endosomal signaling, which was predominantly mediated by Gαq/11. Thus, compartmental bias for CaSR-mediated Gαq/11 endomembrane signaling provides a mechanistic basis for multidimensional GPCR signaling.

In this work, we demonstrate simultaneous phase-contrast imaging (PCI) and X-ray diffractionfrom shock compressed matter at the Matter in Extreme Conditions endstation, at the LinacCoherent Light Source (LCLS). We utilize the chromaticity from compound refractive X-ray lensesto focus the 24.6 keV 3rd order undulator harmonic of the LCLS to a spot size of 5lm on target toperform X-ray diffraction. Simultaneous PCI from the 8.2 keV fundamental X-ray beam is used tovisualize and measure the transient properties of the shock wave over a 500lm field of view.Furthermore, we demonstrate the ability to extend the reciprocal space measurements by 5 Angstroem, rel-ative to the fundamental X-ray energy, by utilizing X-ray diffraction from the 3rd harmonic of theLCLS.
