Martin Fransen werkte op het internationale onderzoekscentrum CERN aan de ontwikkeling van een nieuw type detector die met weinig materiaal in de baan van een deeltje toch een nauwkeurige meting kan doen. Hij onderzocht hoe nauwkeurig prototypes van deze nieuwe detector zijn en hoe deze nauwkeurigheid nog verder verbeterd kan worden. Dit is van belang, omdat wanneer een deeltje door een detector heen schiet, het materiaal van de detector de baan van het deeltje verstoort. Om de baan zo min mogelijk te verstoren moet een dergelijke detector licht gebouwd zijn. Om meer inzicht te krijgen in hoe de natuur werkt, zoekt en onderzoekt men de meest elementaire bouwstenen van materie en hun eigenschappen. Voor het onderzoek hiernaar worden (subatomaire) deeltjes met bijna de lichtsnelheid op elkaar geschoten. Een bekend voorbeeld van zulk onderzoek is de zoektocht naar het Higgsdeeltje op het CERN. Bij de botsing tussen twee deeltjes ontstaan nieuwe (subatomaire) deeltjes die informatie geven over de processen die plaatsvonden bij de botsing. Die processen wil men begrijpen. Om de botsing te reconstrueren, moet er gemeten worden waar welke deeltjes vrijgekomen zijn, hoeveel energie ze hadden, en de richting waarin ze gegaan zijn. Om dit te meten is er vraag naar detectoren waarmee trajecten van elektrisch geladen deeltjes bepaald kunnen worden. Tijdens het onderzoek ontstond er interesse voor de detector vanuit andere vakgebieden, bijvoorbeeld in de zoektocht naar donkere materie en bij protontherapie tegen tumoren.

103. The depression epidemic at work: the role of communication in reducing depression

- Abstract
Building on prior findings indicating that depressed people perceive communication differently than non-depressed people, it is hypothesized that organizational communication mitigates depressive symptoms and therefore prevents them from developing their impact on work outcomes. Data were collected in 2005 and 2007 in an international financial consultancy firm, using a web-based questionnaire (n=1,014). The hypothesis is tested with structural equation modeling (SEM) using AMOS. The bootstrapping results indeed showed a negative association between symptoms of depression and work outcomes that was moderated through organizational communication. Based on the present research, a way of decreasing the negative effects of depression on work outcomes is to tailor organizational communication.

- Abstract
In 2008 we published the first set of guidelines for standardizing research in autophagy. Since then, research on this topic has continued to accelerate, and many new scientists have entered the field. Our knowledge base and relevant new technologies have also been expanding. Accordingly, it is important to update these guidelines for monitoring autophagy in different organisms. Various reviews have described the range of assays that have been used for this purpose. Nevertheless, there continues to be confusion regarding acceptable methods to measure autophagy, especially in multicellular eukaryotes. A key point that needs to be emphasized is that there is a difference between measurements that monitor the numbers or volume of autophagic elements (e.g., autophagosomes or autolysosomes) at any stage of the autophagic process vs. those that measure flux through the autophagy pathway (i.e., the complete process); thus, a block in macroautophagy that results in autophagosome accumulation needs to be differentiated from stimuli that result in increased autophagic activity, defined as increased autophagy induction coupled with increased delivery to, and degradation within, lysosomes (in most higher eukaryotes and some protists such as Dictyostelium) or the vacuole (in plants and fungi). In other words, it is especially important that investigators new to the field understand that the appearance of more autophagosomes does not necessarily equate with more autophagy. In fact, in many cases, autophagosomes accumulate because of a block in trafficking to lysosomes without a concomitant change in autophagosome biogenesis, whereas an increase in autolysosomes may reflect a reduction in degradative activity. Here, we present a set of guidelines for the selection and interpretation of methods for use by investigators who aim to examine macroautophagy and related processes, as well as for reviewers who need to provide realistic and reasonable critiques of papers that are focused on these processes. These guidelines are not meant to be a formulaic set of rules, because the appropriate assays depend in part on the question being asked and the system being used. In addition, we emphasize that no individual assay is guaranteed to be the most appropriate one in every situation, and we strongly recommend the use of multiple assays to monitor autophagy. In these guidelines, we consider these various methods of assessing autophagy and what information can, or cannot, be obtained from them. Finally, by discussing the merits and limits of particular autophagy assays, we hope to encourage technical innovation in the field. © 2012 Landes Bioscience.

For organizations, it is valuable to know how employees’ performance can be guaranteed. The aim of the current study was to examine whether information provision may improve employee performance by enhancing communication climate, work engagement, and employee well-being. To this end, employees (N = 306) of ten different organisations completed an online survey. The results showed that a lack of task-related information negatively affected communication climate. A sufficient amount of information, on the other hand, contributed to a better performance, through communication climate and greater well-being. Although work engagement was not related to performance, a positive communication climate did predict more work engagement among employees. Information overload proved unrelated to the communication climate. The findings of the current study advance existing theory and may serve as a basis for organisations to adjust their information provision and organizational communication.

The experiments presented here examine how managers and executives could improve the effectiveness of their (negative) written communications by incorporating the concept of fit into their message framing. By applying regulatory focus theory, the authors suggest that outcome-based fit between the message and the recipient or within a message leads to more favorable organizational behavior. Experiment 1 showed that employees retain greater feelings of psychological empowerment when they receive a written refusal that is framed in terms that match (versus mismatch) their current regulatory focus. Experiment 2 demonstrated that employees are willing to work more additional hours when they receive a written communication in which the framing of the message and request match, compared to when there is a mismatch. Management implications for designing successful written managerial communications are discussed.

- Abstract
OBJECTIVE: To estimate the global burden of hip and knee osteoarthritis (OA) as part of the Global Burden of Disease 2010 study and to explore how the burden of hip and knee OA compares with other conditions. METHODS: Systematic reviews were conducted to source age-specific and sex-specific epidemiological data for hip and knee OA prevalence, incidence and mortality risk. The prevalence and incidence of symptomatic, radiographic and self-reported hip or knee OA were included. Three levels of severity were defined to derive disability weights (DWs) and severity distribution (proportion with mild, moderate and severe OA). The prevalence by country and region was multiplied by the severity distribution and the appropriate disability weight to calculate years of life lived with disability (YLDs). As there are no deaths directly attributed to OA, YLDs equate disability-adjusted life years (DALYs). RESULTS: Globally, of the 291 conditions, hip and knee OA was ranked as the 11th highest contributor to global disability and 38th highest in DALYs. The global age-standardised prevalence of knee OA was 3.8% (95% uncertainty interval (UI) 3.6% to 4.1%) and hip OA was 0.85% (95% UI 0.74% to 1.02%), with no discernible change from 1990 to 2010. Prevalence was higher in females than males. YLDs for hip and knee OA increased from 10.5 million in 1990 (0.42% of total DALYs) to 17.1 million in 2010 (0.69% of total DALYs). CONCLUSIONS: Hip and knee OA is one of the leading causes of global disability. Methodological issues within this study make it highly likely that the real burden of OA has been underestimated. With the aging and increasing obesity of the world's population, health professions need to prepare for a large increase in the demand for health services to treat hip and knee OA.

In the past seven years the export of white pangasius fillets grew fast. The culture method shifted to intensive production of striped catfish (Ca Tra) in deep ponds because this is more efficient than the pen and cage culture of Ca Basa. Today, striped catfish comprises more than 90 % of the culture. The increased production was achieved by producers investing in large ponds. The market chain is gearing towards vertical integration. Most farms keep fish at relatively high densities of 15 to 25 fish/m3 in ponds having a depth of up to 4m, and are advised to exchange daily 20 to 40% of the water. The sustainability of the sector is threatened due to the increased environmental pressure, and hampered by the growing cost of inputs and reduced farm-gate prices of the fish. The Environmental Impact Assessment (EIA) intends to identify measures for preventing or mitigating the environmental impacts of catfish culture in the Mekong Delta. The EIA was a seven-step process during which we interacted twice with part of the main stakeholders. To build trust among the stakeholders from the sector, we conducted the scoping and goal setting with them.

Item does not contain fulltext, We compared the analgesic effects of single-injection or continuous femoral nerve block (FNB) with intravenous patient controlled analgesia (PCA) opioids. Two hundred patients undergoing knee arthroplasty were randomized to one of the three regimens. Significant knee pain on movement at postoperative 24h was reduced with single-injection (OR 0.30; 95% CI 0.12 to 0.74; P=0.009) or continuous (OR 0.21; 95% CI 0.08 to 0.51; P=0.001) FNB, compared with PCA. Allocation to FNBs also resulted in significantly less opioid consumption, fewer episodes of nausea and vomiting, and achieved knee flexion 90 degrees earlier than allocation to PCA. Compared to single-injection FNB, patients with continuous FNB had lower pain scores on movement at 24h (mean difference -0.57; 95% CI -1.14 to -0.01; P=0.045), consumed less opioid, and had fewer incidences of nausea and vomiting. The analgesic efficacy of single-injection and continuous FNBs was superior to PCA in the immediate postoperative period; with continuous FNB providing better analgesia than single-injection FNB.

Background: Often affecting knee joints, osteoarthritis (OA) is the most common type of arthritis and by 2020 is predicted to become the fourth leading cause of disability globally. Without cure, medication management is symptomatic, mostly with simple analgesics such as acetaminophen and non-steroidal anti-inflammatory drugs (NSAIDs), and glucosamine sulfate. Adherence to arthritis medications is generally low. Intentional non-adherence, that is deliberate decision-making about the use of analgesics, occurs in OA patients. To date, a limited number of studies have explored medication-taking decisions in people with OA nor the extent to which individuals' trade off one treatment factor for another in their decision-making using quantitative techniques. This study aimed to estimate the relative influence of medication-related factors and respondent characteristics on decisions to continue medications among people with symptomatic OA. Methods. A discrete choice experiment (DCE) was conducted among participants attending end-of-study visits in the Long-term Evaluation of Glucosamine Sulfate (LEGS) study (ClinicalTrials.gov ID: NCT00513422). The paper-based survey was used to estimate the relative importance of seven medication specific factors (pain efficacy, mode of action, dose frequency, treatment schedule, side effects, prescription, and out-of-pocket costs) and respondent characteristics on decisions to continue medications. Results: 188 (response rate 37%) completed surveys were returned. Four of the seven medication factors (side effects, out-of-pocket costs, mode of action, treatment schedule) had a significant effect on the choice to continue medication; patient characteristics did not. Assuming equivalent pain efficacy and disease-modifying properties for glucosamine, the positive relative likelihood of continuing with sustained-release acetaminophen was equivalent to glucosamine. - Abstract
BACKGROUND: Non-fatal health outcomes from diseases and injuries are a crucial consideration in the promotion and monitoring of individual and population health. The Global Burden of Disease (GBD) studies done in 1990 and 2000 have been the only studies to quantify non-fatal health outcomes across an exhaustive set of disorders at the global and regional level. Neither effort quantified uncertainty in prevalence or years lived with disability (YLDs). METHODS: Of the 291 diseases and injuries in the GBD cause list, 289 cause disability. For 1160 sequelae of the 289 diseases and injuries, we undertook a systematic analysis of prevalence, incidence, remission, duration, and excess mortality. Sources included published studies, case notification, population-based cancer registries, other disease registries, antenatal clinic serosurveillance, hospital discharge data, ambulatory care data, household surveys, other surveys, and cohort studies. For most sequelae, we used a Bayesian meta-regression method, DisMod-MR, designed to address key limitations in descriptive epidemiological data, including missing data, inconsistency, and large methodological variation between data sources. For some disorders, we used natural history models, geospatial models, back-calculation models (models calculating incidence from population mortality rates and case fatality), or registration completeness models (models adjusting for incomplete registration with health-system access and other covariates). Disability weights for 220 unique health states were used to capture the severity of health loss. YLDs by cause at age, sex, country, and year levels were adjusted for comorbidity with simulation methods. We included uncertainty estimates at all stages of the analysis. FINDINGS: Global prevalence for all ages combined in 2010 across the 1160 sequelae ranged from fewer than one case per 1 million people to 350,000 cases per 1 million people. Blauwvintonijn is een overkoepelende naam voor twee tonijnsoorten: Noordelijke blauwvintonijn (met twee ondersoorten: Atlantische blauwvintonijn (Thunnus thynnus thynnus) en Pacific blauwvintonijn (Thunnus thynnus orientalis) en Zuidelijke blauwvintonijn (Thunnus maccoyii). Visserij op blauwvintonijn is wellicht de lucratiefste visserij met atronomische recordopbrengsten. Hoewel de populaties van deze soorten sinds de jaren zestig van de vorige eeuw afnemen, zal de vangst, gedreven door de hoge opbrengsten, door blijven gaan totdat er een dieptepunt bereikt wordt. In een poging het tij te keren wordt er wereldwijd veel moeite gedaan om blauwvintonijn in gevangenschap te kweken om zodoende een gesloten productiecyclus te behalen.

Het onderzoeksgebied is het terrein van GGZ Noord-Holland, Heiloo. Het gebied met een psychiatrische instelling is in de loop der tijd omsloten door bebouwd gebied. Het terrein wordt nu onderdeel van de wijk, is besloten door de GGZ. Het totale terrein beslaat ca. 25 ha en is landschappelijk ingericht, ook is er op het terrein een oude lanenstructuur aanwezig en een aantal monumentale panden. Het ligt in de bedoeling de attractiewaarde ervan verhogen. Opdat meer bewoners van Heiloo het terrein gaan gebruiken en het ook gaan zien als openbaar terrein.

- Abstract
In 2008 we published the first set of guidelines for standardizing research in autophagy. Since then, research on this topic has continued to accelerate, and many new scientists have entered the field. Our knowledge base and relevant new technologies have also been expanding. Accordingly, it is important to update these guidelines for monitoring autophagy in different organisms. Various reviews have described the range of assays that have been used for this purpose. Nevertheless, there continues to be confusion regarding acceptable methods to measure autophagy, especially in multicellular eukaryotes. A key point that needs to be emphasized is that there is a difference between measurements that monitor the numbers or volume of autophagic elements (e.g., autophagosomes or autolysosomes) at any stage of the autophagic process vs. those that measure flux through the autophagy pathway (i.e., the complete process); thus, a block in macroautophagy that results in autophagosome accumulation needs to be differentiated from stimuli that result in increased autophagic activity, defined as increased autophagy induction coupled with increased delivery to, and degradation within, lysosomes (in most higher eukaryotes and some protists such as Dictyostelium) or the vacuole (in plants and fungi). In other words, it is especially important that investigators new to the field understand that the appearance of more autophagosomes does not necessarily equate with more autophagy. In fact, in many cases, autophagosomes accumulate because of a block in trafficking to lysosomes without a concomitant change in autophagosome biogenesis, whereas an increase in autolysosomes may reflect a reduction in degradative activity. - Abstract
Worldwide, osteoarthritis (OA) is estimated to be the fourth leading cause of disability. Most of this disability burden is attributable to the involvement of the hips or the knees. OA is strongly associated with ageing and the Asian region is ageing rapidly. Further, OA has been associated with heavy physical occupational activity, a required livelihood for many people living in rural communities in developing countries. Unfortunately, joint replacement surgery, an effective intervention for people with severe OA involving the hips or knees, is inaccessible to most people in these regions. On the other hand, obesity, another major risk factor, may be less prevalent, although it is on the increase. Determining region-specific OA prevalence and risk factor profiles will provide important information for planning future cost-effective preventive strategies and health care services. An update of what is currently known about the prevalence of hip and knee OA from population-based studies conducted in the Asian region is presented in this review. Many of the recent studies have conducted comparisons between urban and rural areas and poor and affluent communities. The results of Asian-based studies evaluating risk factors from population-based cohorts or case-control studies, and the current evidence on OA morbidity burden in Asia is also outlined.

- Abstract
De beelden van de alles vernietigende tsunami en de daarop volgende problemen met de kernramp in Fukushima zullen nog bij iedereen op het netvlies gebrand staan. Het menselijke leed bij deze ramp was enorm; 15.511 bevestigde doden en 7.189 vermisten. Ook de schade aan dorpen en steden en de kustinfrastructuur was gigantisch. Omdat Japan een grote visserij en aquacultuursector heeft, die zich vooral in de kustgebieden heeft ontwikkeld, hebben ook deze sectoren aanzienlijke schade opgelopen.

To elucidate unknown mammalian peroxisomal enzymes and functions, we subjected M13 phage expressing fusions between the gene encoding protein VI and a rat liver cDNA library to an immunoaffinity selection process in vitro (biopanning) with the use of antibodies raised against peroxisomal subfractions. In an initial series of biopanning experiments, four different cDNA clones were obtained. These cDNA species encoded two previously identified peroxisomal enzymes, catalase and urate oxidase, and two novel proteins that contained a C-terminal peroxisomal targeting signal (PTS1). A primary structure analysis of these novel proteins revealed that one, ending in the tripeptide AKL, is homologous to the yeast peroxisomal 2,4-dienoyl-CoA reductase (EC 1.3.1.34; DCR), an enzyme required for the degradation of unsaturated fatty acids, and that the other, ending in the tripeptide SRL, is a putative member of the short-chain dehydrogenase/reductase (SDR) family, with three isoforms. Green fluorescent protein (GFP) fusions encoding GFP-DCR-AKL, GFP-DCR, GFP-SDR-SRL and GFP-SDR were expressed in mammalian cells. The analysis of the subcellular location of the recombinant fusion proteins confirmed the peroxisomal localization of GFP-DCR-AKL and GFP-SDR-SRL, as well as the functionality of the PTS1. That the AKL protein is indeed an NADPH-dependent DCR was demonstrated by showing DCR activity of the bacterially expressed protein. These results demonstrate at the molecular level that mammalian peroxisomes do indeed contain a DCR. In addition, the results presented here indicate that the protein VI display system is suitable for the isolation of rare cDNA clones from cDNA libraries and that this technology facilitates the identification of novel peroxisomal proteins.

In de afgelopen vier decennia is er steeds meer onderzoek gedaan naar de levensloop van producten (vaak onder de noemer"van wieg tot graf") en het totale effect ervan op het milieu. Dit zogenoemde Life Cycle Assessment (LCA) onderzoek richtte zich in eerste instantie vooral op niet-agrarische industrieën (zoals de autosector, elektrosector en de petrochemische industrie), maar sinds de jaren negentig werden de mogelijkheden en toepassingen ervan ook door de agrarische sector ontdekt. In dit eerste artikel wordt LCA verder uitgelegd voor gebruik in de aquacultuur. In het volgende deel worden twee voorbeelden van LCA in aquacultuur beschreven.

BACKGROUND: There is little scientific evidence to support the usual practice of providing outpatient rehabilitation to patients undergoing total knee replacement surgery (TKR) immediately after discharge from the orthopaedic ward. It is hypothesised that the lack of clinical benefit is due to the low exercise intensity tolerated at this time, with patients still recovering from the effects of major orthopaedic surgery. The aim of the proposed clinical trial is to investigate the clinical and cost effectiveness of a novel rehabilitation strategy, consisting of an initial home exercise programme followed, approximately six weeks later, by higher intensity outpatient exercise classes. METHODS/DESIGN: In this multicentre randomised controlled trial, 600 patients undergoing primary TKR will be recruited at the orthopaedic pre-admission clinic of 10 large public and private hospitals in Australia. There will be no change to the medical or rehabilitative care usually provided while the participant is admitted to the orthopaedic ward. After TKR, but prior to discharge from the orthopaedic ward, participants will be randomised to either the novel rehabilitation strategy or usual rehabilitative care as provided by the hospital or recommended by the orthopaedic surgeon. Outcomes assessments will be conducted at baseline (pre-admission clinic) and at 6 weeks, 6 months and 12 months following randomisation. The primary outcomes will be self-reported knee pain and physical function. Secondary outcomes include quality of life and objective measures of physical performance. Health economic data (health sector and community service utilisation, loss of productivity) will be recorded prospectively by participants in a patient diary. This patient cohort will also be followed-up annually for five years for knee pain, physical function and the need or actual incidence of further joint replacement surgery.
