Eucalyptus plantation forests in southern China provide not only the economic value of producing timber, but also the ecological value service of absorbing carbon dioxide and releasing oxygen. Based on the theory of spatial colonial modeling, this paper proposes a new method for 3D reconstruction of tree terrestrial LiDAR point clouds for determining the aboveground carbon stock of eucalyptus monocotyledons, which consists of the main steps of tree branch and trunk separation, skeleton extraction and optimization, 3D reconstruction, and carbon stock calculation. The main trunk and branches of the tree point clouds are separated using a layer-by-layer judgment and clustering method, which avoids errors in judgment caused by sagging branches. By optimizing and adjusting the skeleton to remove small redundant branches, the near-parallel branches belonging to the same tree branch are fused. The missing parts of the skeleton point clouds were complemented using the cardinal curve interpolation algorithm, and finally a real 3D structural model was generated based on the complemented and smoothed tree skeleton expansion. The bidirectional Hausdoff distance, average Hausdoff distance, and F distance were used as evaluation indexes, which were reduced by 0.7453 m, 0.0028 m, and 0.0011 m, respectively, and the improved spatial colonization algorithm enhanced the accuracy of the reconstructed tree 3D structural model. To verify the accuracy of our method to determine the carbon stock and its related parameters, we cut down 41 eucalyptus trees and destructively sampled the measurement data as reference values. The R2 of the linear fit between the reconstructed single-tree aboveground carbon stock estimates and the reference values was 0.96 with a CV(RMSE) of 16.23%, the R2 of the linear fit between the trunk volume estimates and the reference values was 0.94 with a CV(RMSE) of 19.00%, and the R2 of the linear fit between the branch volume estimates and the reference values was 0.95 with a CV(RMSE) of 38.84%. In this paper, a new method for reconstructing eucalyptus carbon stocks based on TLS point clouds is proposed, which can provide decision support for forest management and administration, forest carbon sink trading, and emission reduction policy formulation.

This research paper systematically investigates the combined influence of fly ash, cementitious capillary crystalline waterproofing (CCCW) materials, and polypropylene fibers on the mechanical properties and impermeability of concrete through comprehensive orthogonal tests. Microscopic morphological changes in the concrete induced by different composite materials are examined via scanning electron microscopy (SEM) and X-ray diffraction (XRD) testing. The objective is to facilitate a beneficial synergetic interaction among these materials to develop highly permeable, crack-resistant concrete. Key findings of this study are: (1) The study unveils the impact of the concentration of three additive materials on the concrete’s compressive strength, tensile strength, and penetration height, thereby outlining their significant influence on the mechanical properties and impermeability of the concrete; (2) An integrated scoring method determined the optimal composite dosage of three materials: 15% fly ash, 2% CCCW, and polypropylene fibers at 1.5 kg/m3. This combination increased the concrete’s compressive strength by 12.5%, tensile strength by 48.4%, and decreased the average permeability height by 63.6%; (3) The collective introduction of these three materials notably augments the hydration reaction of the cement, resulting in denser concrete microstructure, enhanced bonding between fibers and matrix, and improved concrete strength and durability.

Abstract Influenza A virus infection causes a series of diseases, but the factors associated with disease severity are not fully understood. Disruption of the endothelial glycocalyx contributes to acute lung injury in sepsis, but has not been well studied in H1N1 influenza. We aim to determine whether the plasma glycocalyx components levels are predictive of disease severity in H1N1 influenza. This prospective observational study included 53 patients with influenza A (H1N1) during the influenza season, and 30 healthy controls in our hospital. Patients were grouped by severity and survival. We collected clinical data and blood samples at admission. Inflammatory factors (tumor necrosis factor-α, interleukin-6, interleukin-10) and endothelial glycocalyx components (syndecan-1, hyaluronan, heparan sulfate) were measured. The plasma levels of syndecan-1, hyaluronan, and heparan sulfate were significantly higher in patients with severe influenza A (H1N1) than in mild cases. Syndecan-1 and hyaluronan were positively correlated with disease severity, which was indicated by the APACHE II and SOFA scores and lactate levels, and negatively correlated with albumin levels. At a cutoff point ≥ 173.9 ng/mL, syndecan-1 had a 81.3% sensitivity and 70.3% specificity for predicting of 28-day mortality. Kaplan–Meier analysis demonstrated a strong association between syndecan-1 levels and 28-day mortality (log-rank 11.04, P = 0.001). Elevated plasma levels of syndecan-1 has a potential role in systemic organ dysfunction and may be indicative of disease severity in patients with influenza A (H1N1).

A new land surface clustering algorithm is developed to retrieve soil moisture (SM) using the Global Navigation Satellite System reflectometry (GNSS-R) technique. Data from the BuFeng-1 (BF-1) twin satellites A/B, a pilot mission for the Chinese GNSS-R constellation, is used for SM retrieval. The core concept of the algorithm is to cluster global land areas into different types according to the land properties and calculate the SM type by type, based on the linear relationship between equivalent specular reflectivity and SM. The global comparison between the results and SM product from the Soil Moisture Active Passive mission shows the correlation coefficient (R) is 0.82, and unbiased root mean square error (ubRMSE) is 0.070 cm3·cm−3. The results also show good agreement compared with in situ SM measurements with the mean ubRMSE of 0.036 cm3·cm−3. This study proves that the global SM can be retrieved successfully from the BF-1 mission with the land surface clustering algorithm. By taking full advantage of the similarity of land surface physical properties in different regions, the algorithm provides a practical approach for global SM retrieval using spaceborne GNSS-R data.

Tetrodotoxin (TTX) is a lethal neurotoxin produced by the endosymbiotic bacteria in the gut of puffer fish. Currently, there is no effective and economical method to detect TTX, so it is very interesting to develop low-cost and high-sensitivity detection methods by using nucleic-acid aptamers as the recognition molecules. However, traditional SELEX screening of aptamers for targeting small molecules such as TTX is labor-intensive, and usually the success rate is low. Here, we employed a strategy of “repurposing old aptamers for new uses” to develop high-affinity aptamers for TTX. To this end, we first collected thermally stable DNA aptamers and predicted their affinities for TTX by molecular docking; then, we identified high-affinity candidates and verified them by microscale thermophoresis (MST) experiments. In this way, two thermally stable aptamers (Tv-51 and AI-57) were found to possess nanomolar affinities for TTX. Moreover, we performed spontaneous binding simulations to reveal their binding mechanisms to TTX and thereby identified the key bases for the binding. Guided by these, two variants (Tv-46 and AI-52) with higher affinities and specificities were subsequently engineered and confirmed by the MST experiments. So, this study not only provides potential recognition molecules for the technology developments of TTX detection, but also demonstrates an effective repurposing approach to the discovery of high-affinity aptamers for new target molecules.

BackgroundThe purpose of this study was to develop a new prognostic model for osteosarcoma based on cuproptosis-mitochondrion genes.Materials and methodsThe data of osteosarcoma were obtained from TARGET database. By using Cox regression and LASSO regression analysis, a novel risk score was constructed based on cuproptosis-mitochondrion genes. Kaplan-Meier, ROC curve and independent prognostic analyses were performed to validate the risk score in GSE21257 dataset. Then, a predictive nomogram was constructed and further validated by calibration plot, C-index and ROC curve. Based on the risk score, all patients were divided into high-risk and low-risk group. GO and KEGG enrichment, immune correlation and drug sensitivity analyses were performed between groups. Real-time quantitative PCR verified the expression of cuproptosis-mitochondrion prognostic model genes in osteosarcoma. And we explored the function of FDX1 in osteosarcoma by western blotting, CCK8, colony formation assay, wound healing assay and transwell assays.ResultsA total of six cuproptosis-mitochondrion genes (FDX1, COX11, MFN2, TOMM20, NDUFB9 and ATP6V1E1) were identified. A novel risk score and associated prognostic nomogram were constructed with high clinical application value. Strong differences in function enrichment and tumor immune microenvironment were shown between groups. Besides, the correlation of cuproptosis-mitochondrion genes and drug sensitivity were revealed to search for potential therapeutic target. The expression of FDX1, COX11, MFN2, TOMM20 and NDUFB9 at mRNA level was elevated in osteosarcoma cells compared with normal osteoblast hFOB1.19. The mRNA expression level of ATP6V1E1 was decreased in osteosarcoma. Compared with hFOB1.19, western blotting revealed that the expression of FDX1 was significantly elevated in osteosarcoma cells. Functional experiments indicated that FDX1 mainly promoted the migration of osteosarcoma rather than proliferation.ConclusionsWe developed a novel prognostic model of osteosarcoma based on cuproptosis-mitochondrion genes, which provided great guidance in survival prediction and individualized treatment decision making for patients with osteosarcoma.

Osteoarthritis (OA) has been proven as the second primary cause of pain and disability in the elderly population, impact patients both physically and mentally, as well as imposing a heavy burden on the global healthcare system. Current treatment methods, whether conservative or surgical, that aim at relieving symptoms can not delay or reverse the degenerative process in the structure. Scientists and clinicians are facing a revolution in OA treatment strategies. The emergence of exosomes brings hope for OA treatment based on pathology, which is attributed to its full potential in protecting chondrocytes from excessive death, alleviating inflammation, maintaining cartilage matrix metabolism, and regulating angiogenesis and subchondral bone remodeling. Therefore, we summarized the recent studies of exosomes in OA, aiming to comprehensively understand the functions and mechanisms of exosomes in OA treatment, which may provide direction and theoretical support for formulating therapeutic strategies in the future.

In this paper, the performance of a high-pressure grouting material and consolidation body, the key parameters of tunnel surrounding rock reinforcement, and the field reinforcement effect are compared and studied. The results show that 1) the compressive strength, elastic modulus, and seepage resistance of the consolidation body are related to the water–cement ratio. The permeability of the consolidation body increases with increasing water–cement ratio under the constant head; under the condition of a certain water–cement ratio, the strength increases with an increase in cement content. To meet the construction requirements and the slurry stability, a small water–cement ratio is appropriate. 2) Slurry pressure, rotary spray speed, and lifting speed on the single pile static load bearing capacity have a significant impact. Under the same static load conditions, the greater the slurry pressure, the lower the cumulative settlement value of the single pile, the cumulative settlement value increases less, and the amount of change in rebound is the same; with the increase in the rotational speed of the rotary spray, the lower the cumulative settlement value of the single pile, and the rate of change of the cumulative settlement value of the single pile decreases; and with the increase in the lifting speed, the cumulative settlement value of the single pile increases and the rate of change of the cumulative settlement value decreases. 3) The pile body cement soil is more uniform, has high strength, and is brittle, and the stratum where the slurry part is located is a loose powder clay layer and sandy layer. 4) After high-pressure jet grouting pile reinforcement, the top surrounding rock is more stable after the excavation of an ultra-shallow buried tunnel, the fissures between the surrounding rocks are filled with cement soil consolidation, and the integrity is improved. Certain piles also play a certain role in stopping water; high-pressure jet grouting pile reinforcement to improve the integrity of the surrounding rock played a role in consolidating the weak soil layer above the tunnel vault, creating conditions for the safe excavation of the shallow buried section of the tunnel.

The skin contributes critically to health via its role as a barrier tissue against a multitude of external pathogens. The barrier function of the skin largely depends on the uppermost epidermal layer which is reinforced by skin barrier immunity. The integrity and effectiveness of skin barrier immunity strongly depends on the close interplay and communication between immune cells and the skin environment. Skin-associated adipocytes have been recognized to play a significant role in modulating skin immune responses and infection by secreting cytokines, adipokines, and antimicrobial peptides. This review summarizes the recent understanding of the interactions between skin-associated adipocytes and other skin cells in maintaining the integrity and effectiveness of skin barrier immunity.

To study the risk and control countermeasures of the TBM tunnel construction adjacent to the operational railway tunnel, based on the TBM tunnel project of Chongqing Rail Transit Line 5, this paper first evaluates the quality health degree of the operational tunnel lining (OTL) structure according to the on-site structural inspection. Then, the displacement, internal force, and proximity influence scope influenced by the metro TBM tunnel construction are studied using numerical simulation. Finally, the corresponding control countermeasures are proposed. The results show that: (1) The adjacent construction of the upper TBM tunnel will lead to the uplift deformation trend of the lower operational tunnel, and the uplift deformation of the vault is greater than that of the ballast bed. The influence scope is roughly a parallelogram, with the long axis parallel to the operational tunnel and the short axis parallel to the new TBM subway tunnel. (2) TBM tunnelling over the operational tunnel will cause the transformation of the mechanical mode of the OTL structure from the small eccentric compression mode to the large eccentric compression mode. The OTL structure between the left and right lines of TBM is unfavorable. (3) The longitudinal curve of the bending moment and axial force of the OTL fluctuates greatly within the influence range. The bending moment and axial force are reduced in operational tunnel construction joints. Based on field evaluation and numerical analysis, this paper puts forward some risk control countermeasures, such as TBM tunnelling parameters control, pea-gravel backfilling, backfill grouting, and bottom grouting, which can effectively solve the risk of the operational tunnel structure in the adjacent construction. This study has important reference value for risk control and safety assessment of tunnel in complex adjacent tunnel construction.

Several engineering practices have shown that the excavation of shallow-buried tunnels beneath major roads, as well as the selection of appropriate engineering measures and construction methods, has a significant impact on road surface settlement. Therefore, field monitoring and numerical simulation are adopted in this study to analyze the effect of the cross diaphragm (CRD) excavation method on surface settlement for the under-construction Yüan 1 railroad tunnel. The findings show that during the excavation of the four divisions of the CRD excavation method for shallow-buried tunnels, the amount of surface settlement caused by the excavation of part 1 accounts for 40% of the total surface settlement, followed by the excavation of part 3, accounting for 30% of the total surface settlement, and the difference between the excavation of parts 2 and 4 is insignificant, with part 2 slightly larger than part 4. The main influence of the CRD method on surface settlement for shallow-buried tunnels is 0.64–0.86 times the cavity diameter from the tunnel median, within which the final surface settlement caused by excavation is within the same horizontal range, and beyond which the surface settlement is prone to dramatically decline. By applying advanced grouting and adjusting the construction method of CRD based on the monitoring data, the effect of the CRD excavation method on surface settlement can be controlled.

The Sichuan-Tibet Railway is faced with risks of active faults, unfavorable geological conditions, high in-situ stress levels, and a high potential for Traditional tunnel seismic measures that can no longer guarantee the safety of tunnel structures in high-intensity earthquake areas. The new shock absorption capacity layer with resistance-limiting is proposed in this paper and the resistance-limiting shock absorption principle. To study the safety of the new anti-shock lining structure, a large-scale quasi-static model test was carried out based on the improved static-dynamic coupling shear force. The test analyzes the response rules of strain, structural internal force, and lining failure form of tunnel lining, taking into consideration three influencing factors including layer layout position, layout density, and tunnel buried depth. The results showed that: 1) Under the action of low-cycle reciprocating shear strain, the failure mode of the shallow buried tunnel is mainly the tensile failure of the arch waist and the failure of the arch waist and the side wall, and the failure mode of the deeply buried tunnel is mainly the compression failure of the arch waist; 2) Compared with the case of no shock absorption layer, the lining tensile stress on the inner side of the arch waist position is significantly reduced. Under the high shear strain (0.20%), the increased tensile stress trend was slowed down on the inner side walls; 3) The main working area of the resistance-limiting layer can effectively improve the lining stress distribution underground vibration and protects the tensile damage on the arch waist to the side wall. The width and depth of the crack are weakened and the seismic bearing capacity of the width and depth of the crack is weakened and the seismic bearing capacity of the lining structure is greatly strengthened; 4) The lining cracks without the resistance limiting layer develop violently, and the crack at the arch waist runs The lining structure failure mode of the resistance limiting layer arranged at the arch waist is similar to The lining structure failure mode of the resistance limiting layer.

Low-lethality ultrasound technology has received more and more attention in regulating microorganisms of fermentation industry. Herein, two representative Ames tester strains TA97a and TA98 as model organisms were used to explore the effects of ultrasound on insertion-deletion (InDel) polymorphisms of microbial DNA and its underlying mechanisms. Results revealed that a promotion was observed in the reversion mutation of TA98 upon sonication. Sequencing results from 1752 TA98 revertants showed that there was a total of 127 InDels, of which the InDels unique to ultrasound were 36 more than that of the control. Compared with the control, ultrasound-mediated InDels of DNA displayed additional −29 bp deletion and +7 ∼ +43 bp insertions of direct repeat sequences. Combined with the analysis of transcriptomics and prediction of secondary structure of single-stranded DNA from InDels core region (No. 832 ∼ 915 bp) in hisD3052 gene of TA98 strain, ultrasound-mediated “thermal breathing” mechanism was proposed based on the formation of DNA hairpin structure with micro-homologous sequence. This finding implied that low-intensity ultrasound is expected to be developed a new low-lethal mutagenic technology for continuous mutagenesis.

Abstract Background Lung cancer is one of the most lethal malignant tumors that endangers human health. Lung adenocarcinoma (LUAD) has increased dramatically in recent decades, accounting for nearly 40% of all lung cancer cases. Increasing evidence points to the importance of the competitive endogenous RNA (ceRNA) intrinsic mechanism in various human cancers. However, behavioral characteristics of the ceRNA network in lung adenocarcinoma need further study. Methods Groups based on SLC2A1 expression were used in this study to identify associated ceRNA networks and potential prognostic markers in lung adenocarcinoma. The Cancer Genome Atlas (TCGA) database was used to obtain the patients' lncRNA, miRNA, and mRNA expression profiles, as well as clinical data. Informatics techniques were used to investigate the effect of hub genes on prognosis. The Cox regression analyses were performed to evaluate the prognostic effect of hub genes. The methylation, GSEA, and immune infiltration analyses were utilized to explore the potential mechanisms of the hub gene. The CCK-8, transwell, and colony formation assays were performed to detect the proliferation and invasion of lung cancer cells. Results We eventually identified the ITGB1-DT/ARNTL2 axis as an independent fact may promote lung adenocarcinoma progression. Furthermore, methylation analysis revealed that hypo-methylation may cause the dysregulated ITGB1-DT/ARNTL2 axis, and immune infiltration analysis revealed that the ITGB1-DT/ARNTL2 axis may affect the immune microenvironment and the progression of lung adenocarcinoma. The CCK-8, transwell, and colonu formation assays suggested that ITGB1-DT/ARNTL2 promotes the progression of lung adenocarcinoma. And hsa-miR-30b-3p reversed the ITGB1/ARNTL2-mediated oncogenic processes. Conclusion Our study identified the ITGB1-DT/ARNTL2 axis as a novel prognostic biomarker affects the prognosis of lung adenocarcinoma.

Abstract Background CD8+ T cells play a critical role in the innate antitumour immune response. Recently, CD8+ T cell dysfunction has been verified in various malignant cancers, including non-small cell lung cancer (NSCLC). However, the molecular biological mechanisms of CD8+ T cell dysfunction in human NSCLC are still unclear. Methods The expression of circular ubiquitin-specific protease-7 (circUSP7) in NSCLC tissues, exosomes, and cell lines was detected by quantitative real-time polymerase chain reaction (qRT-PCR). Exosomes were isolated from the culture medium of NSCLC cells and the plasma of NSCLC patients using an ultracentrifugation method and the ExoQuick Exosome Precipitation Solution kit. The exosomes were then characterized by transmission electronic microscopy (TEM), NanoSight and western blotting. The role of circUSP7 in CD8+ T cell dysfunction was assessed by enzyme-linked immunosorbent assay (ELISA). In vivo circular RNA (circRNA) precipitation (circRIP), RNA immunoprecipitation (RIP), and luciferase reporter assays were performed to explore the molecular mechanisms of circUSP7 in CD8+ T cells. In a retrospective study, the clinical characteristics and prognostic significance of circUSP7 in NSCLC tissues were determined. Results The expression levels of circUSP7 were higher in human NSCLC tissues than in matched adjacent nontumour tissues. Increased levels of circUSP7 indicate poor clinical prognosis and CD8+ T cell dysfunction in patients with NSCLC. The circUSP7 found in NSCLC patient plasma is predominantly secreted by NSCLC cells in an exosomal manner, and circUSP7 inhibits IFN-γ, TNF-α, Granzyme-B and Perforin secretion by CD8+ T cells. Furthermore, circUSP7 inhibits CD8+ T cell function by upregulating the expression of Src homology region 2 (SH2)-containing protein tyrosine phosphatase 2 (SHP2) via sponging miR-934. Finally, we show that circUSP7 may promote resistance to anti-PD1 immunotherapy in NSCLC patients. Conclusions Exosomal circUSP7 is predominantly secreted by NSCLC cells and contributes to immunosuppression by promoting CD8+ T cell dysfunction in NSCLC. CircUSP7 induces resistance to anti-PD1 immunotherapy, providing a potential therapeutic strategy for NSCLC patients.

Abstract Vehicle lane-changing on urban roads is the most common traffic behavior, in which the driver changes the direction or increases the speed of the vehicle by changing its trajectory. However, in high-density traffic flow, when a vehicle changes lanes, a series of vehicles following the target vehicle in the target lane will be delayed. In this study, DJI Phantom 4 drones were used to vertically record the traffic on a road section. Tracker software was then used to extract vehicle information from the video taken by the drones, including the vehicle operating speeds, etc. SPSS 22 and Origin analysis software were then employed to analyze the correlations between different vehicle operating parameters. It was found that the operating speed of the first vehicle following the target vehicle in the target lane is related to the speeds and positions of both the target vehicle and the vehicle preceding it. Under the condition of high-density traffic flow, when the target vehicle is inserted into the target lane, the speed of the vehicles following the target vehicle in the target lane will change. To model this process, the corresponding Sine and DoseResp models were constructed. By calculating the delays of vehicles following the target vehicle in the target lane, it was concluded that the overall delay of the fleet is 3.9–9.5 s.

Diabetes mellitus is the most common metabolic disease associated with impaired wound healing. Recently, Schwann cells (SCs), the glia of the peripheral nervous system, have been suggested to accelerate normal skin wound healing. However, the roles of SCs in diabetic wound healing are not fully understood. In this study, Full-thickness wounds were made in the dorsal skin of C57/B6 mice and db/db (diabetic) mice. Tissue samples were collected at different time points, and immunohistochemical and immunofluorescence analyses were performed to detect markers of de-differentiated SCs, including myelin basic protein, Sox 10, p75, c-Jun, and Ki67. In addition, in vitro experiments were performed using rat SC (RSC96) and murine fibroblast (L929) cell lines to examine the effects of high glucose conditions (50 mM) on the de-differentiation of SCs and the paracrine effects of SCs on myofibroblast formation. Here, we found that, compared with that in normal mice, wound healing was delayed and SCs failed to rapidly activate a repair program after skin wound injury in diabetic mice. Furthermore, we found that SCs from diabetic mice displayed functional impairments in cell de-differentiation, cell-cycle re-entry, and cell migration. In vitro, hyperglycemia impaired RSC 96 cell de-differentiation, cell-cycle re-entry, and cell migration, as well as their paracrine effects on myofibroblast formation, including the secretion of TGF-β and Timp1. These results suggest that delayed wound healing in diabetes is due in part to a diminished SC repair response and attenuated paracrine effects on myofibroblast formation.

ABSTRACT The rapid development of methicillin-resistant Staphylococcus aureus (MRSA) drug resistance and the formation of biofilms seriously challenge the clinical application of classic antibiotics. Extracts of the traditional herb Chenopodium ambrosioides L. were found to have strong antibiofilm activity against MRSA, but their mechanism of action remains poorly understood. This study was designed to investigate the antibacterial and antibiofilm activities against MRSA of flavonoids identified from C. ambrosioides L. in combination with classic antibiotics, including ceftazidime, erythromycin, levofloxacin, penicillin G, and vancomycin. Liquid chromatography-mass spectrometry (LC-MS) was used to analyze the nonvolatile chemical compositions. Reverse transcription (RT)-PCR was used to investigate potential multitargets of flavonoids based on global transcriptional responses of virulence and antibiotic resistance. A synergistic antibacterial and biofilm-inhibiting activity of the alcoholic extract of the ear of C. ambrosioides L. in combination with penicillin G was observed against MRSA, which proved to be closely related to the interaction of the main components of kaempferol rhamnosides with quercetin. In regard to the mechanism, the increased sensitivity of MRSA to penicillin G was shown to be related to the downregulation of penicillinase with SarA as a potential drug target, while the antibiofilm activity was mainly related to downregulation of various virulence factors involved in the initial and mature stages of biofilm development, with SarA and/or σB as drug targets. This study provides a theoretical basis for further exploration of the medicinal activity of kaempferol rhamnosides and quercetin and their application in combination with penicillin G against MRSA biofilm infection. IMPORTANCE In this study, the synergistic antibacterial and antibiofilm effects of the traditional herb C. ambrosioides L. and the classic antibiotic penicillin G on MRSA provide a potential strategy to deal with the rapid development of MRSA antibiotic resistance. This study also provides a theoretical basis for further optimizing the combined effect of kaempferol rhamnosides, quercetin, and penicillin G and exploring anti-MRSA biofilm infection research with SarA and σB as drug targets.

The stability of the surrounding rock analysis and evaluation during tunnel construction is the basis of tunnel construction risk control. In this paper, we focus on the stability of a large-scale transportation tunnel complex during its construction in a densely-populated urban area. The tunnel complex includes seven shallow-buried tunnels with large cross-sections. In order to gain insight into the excavation influence of the different tunnels, stability analyses were first carried out using FLAC3D numerical simulation. Results showed that the tunnels were subjected to heave and crown settlement induced by adjacent excavation. Also, stress concentrated in the rock blocks connecting different tunnels. Subsequently, a bench-scale model test was performed to understand the failure of the rock blocks and to examine the accuracy of the numerical simulation. The test results agreed well with the numerical simulation. Based on the numerical and test results, the mechanism of the rock blocks failure was explained and construction risk control technology to stabilize the rock blocks was proposed. The construction risk analyses revealed: 1) tunnels are subjected to significant heave due to the excavation of tunnels located above; 2) the stability of the rock blocks is the paramount determinant for stabilizing the whole tunnel complex; 3) ensuring rock blocks to be in a state of triaxial stress is conducive to its stability and hence the stability of multiple tunnels.

Litter decomposition is the main process that affects nutrient cycling and carbon budgets in mixed forests. However, knowledge of the response of the soil microbial processes to the mixed-litter decomposition of fresh leaf, semi-decomposed leaf and fine root is limited. Thus, a laboratory microcosm experiment was performed to explore the mixed-litter effects of fresh leaf, semi-decomposed leaf and fine root on the soil enzyme activity and microbial community in an evergreen broadleaf karst forest in Southwest China. Fresh leaf litter, semi-decomposed litter and fine root in the Parakmeria nitida and Dayaoshania cotinifolia forests, which are unique protective species and dominant species in the evergreen broadleaf forest, were decomposed alone and in all possible combinations, respectively. Our results showed that the mass loss of fresh leaf litter in three mixed-litter treatment was significantly higher than that in two mixed-litter treatment in the P. nitida and D. cotinifolia forests. Mass loss of fine root in the single litter treatment was significantly lower in the P. nitida forest and higher in the D. cotinifolia forest compared to that in the other litter treatments. There were insignificant differences in the activities of β-glucosidase (BG) and leucine aminopeptidase (LAP) between control and mixed-litter treatment in the P. nitida forest and between control and single litter treatment in the D. cotinifolia forest. The N-acetyl-β-D-glucosaminidase (NAG) activity was significantly increased by the single litter decomposition of fresh leaf and fine root and three mixed-litter decomposition in the P. nitida and D. cotinifolia forests. The activity of acid phospomonoesterase (AP) in the decomposition of fresh leaf litter was lower in the P. nitida forest and higher in the D. cotinifolia forest compared to that in control. The most dominant soil bacteria were Proteobacteria in the P. nitida forest and were Actinobacteria and Proteobacteria in the D. cotinifolia forest. Shannon, Chao1, ACE and PD indexes in the mixed-litter decomposition of fresh leaf and semi-decomposition litter were higher than that in control in P. nitida forest. There were insignificant differences in observed species and indexes of Chao1, ACE and PD between litter treatments in the D. cotinifolia forest. Richness of mixed-litter significantly affected mass loss, soil enzyme activity and microbial diversity in the P. nitida forest. Litter N concentration and the presence of fresh leaf litter were significantly correlated with the mass loss and soil enzyme activity in the P. nitida and D. cotinifolia forests. These results indicated that the presence of fresh leaf litter showed a non-negligible influence on mixed-litter decomposition and soil enzyme activity, which might be partly explained by litter initial quality in the P. nitida and D. cotinifolia forests.

Merozoite invasion of the erythrocytes in humans is a key step in the pathogenesis of malaria. The proteins involved in the merozoite invasion could be potential targets for the development of malaria vaccines. Novel viral-vector-based malaria vaccine regimens developed are currently under clinical trials. Vesicular stomatitis virus (VSV) is a single-stranded negative-strand RNA virus widely used as a vector for virus or cancer vaccines. Whether the VSV-based malarial vaccine is more effective than conventional vaccines based on proteins involved in parasitic invasion is still unclear. In this study, we have used the reverse genetics system to construct recombinant VSVs (rVSVs) expressing apical membrane protein 1 (AMA1), rhoptry neck protein 2 (RON2), and reticulocyte-binding protein homolog 5 (RH5), which are required for Plasmodium falciparum invasion. Our results showed that VSV-based viral vaccines significantly increased Plasmodium-specific IgG levels and lymphocyte proliferation. Also, VSV-PyAMA1 and VSV-PyRON2sp prime-boost regimens could significantly increase the levels of IL-2 and IFN-γ-producing by CD4+ and CD8+ T cells and suppress invasion in vitro. The rVSV prime-protein boost regimen significantly increase Plasmodium antigen-specific IgG levels in the serum of mice compared to the homologous rVSV prime-boost. Furthermore, the protective efficacy of rVSV prime protein boost immunization in the mice challenged with P. yoelii 17XL was better compared to traditional antigen immunization. Together, our results show that VSV vector is a novel strategy for malarial vaccine development and preventing the parasitic diseases.

Autologous fat grafting is becoming increasingly common worldly. However, the long-term retention of fat grafting is still unpredictable due to the inevitable fibrosis arising during tissue repair. Fibrosis may be regulated by T-cell immune responses that are influenced by adipose-derived stem cells (ASCs). Therefore, we hypothesized that overly abundant ASCs might promote fibrosis by promoting T-cell immune responses to adipose tissue. We performed 0.3 ml fat grafts with 104/ml, 106/ml and 108/ml ASCs and control group in C57 BL/6 mice in vivo. We observed retention, fibrosis, T-cell immunity, and macrophage infiltration over 12 weeks. Besides, CD4+ T-helper 1 (Th1) cells and T-helper 2 (Th2) cells were co-cultured with ASCs or ASCs conditioned media (ASCs-CM) in vitro. We detected the ratio of Th2%/Th1%. Results showed that the retention rate was higher in 104 group, while even lower in 108 group with significantly increased inflammation and fibrosis than control group at week 12 in vivo. There was no significance between control group and 106 group. Also, 108 group increased the infiltration of M2 macrophages, CD4+ T-cells and Th2/Th1 ratio. In vitro, the ratio of Th2%/Th1% induced by ASCs-transwell group was higher than ASCs-CM group and showed concentration-dependent. Accordingly, high concentrations of ASCs in adipose tissue can promote Th1–Th2 shifting, and excessive Th2 cells might promote the persistence of M2 macrophages and increase the level of fibrosis which lead to a decrease in the long-term retention of fat grafts. Also, we found ASCs promoted Th1–Th2 shifting in vitro.

Given the background of electricity market, the electricity consumption optimization can reduce consumers’ energy cost, and also help to reduce the peak load and improve the accommodation level of clean energy. Aiming at solving the problem of consumers’ insufficient management of their own electricity consumption, a multi-objective optimization model of electricity consumption is proposed considering the correlation of equipment electricity consumption. First, the cross-correlation coefficient is adopted to identify the consumption correlation among various electricity equipment based on the historical load data of consumers, and the significance of the consumption correlation is analyzed by silhouette index. Next, a multi-objective optimization model is constructed and the optimal electricity consumption strategy is obtained to maximize the consumer-side economy and comfort benefits. Finally, the results of case studies on a Zhejiang stir-fried tea enterprise in China show that the proposed model is feasible and effective.

Introduction: The ambiguity of the drug target is one of the major factors restricting the development of traditional Chinese medicine (TCMs) and its bioactive constituents. The characteristics of “multiple components, multiple targets and multiple pathways” of TCMs make the research of drug targets extremely difficult. With the emergence of new theories, there are increasing technologies and strategies that can be used for the drug targets research of TCMs. In this paper, we summarize several techniques and methods applied to the study of TCM targets. Methods: Through consulting a large number of literature, research and summary, and finally summarized the application direction of the technical method, advantages and limitations. Results: The methods and techniques including computer aided drug design, network pharmacology, phage display, affinity fishing, drug affinity responsive target stability and cellular thermal shift assay were summarized, and their application directions, advantages and disadvantages were discussed. At the same time, a large number of application examples were given to provide reference for the research of TCM targets.

Background: There is an increasing demand for minimally invasive aortic valve replacement. This study aimed to investigate the safety and feasibility of minimally invasive aortic valve replacement through a right parasternal second intercostal transverse incision. Methods: This was a retrospective study, and we collected information from 111 patients who underwent isolated aortic valve replacement surgery performed by the same surgeon from January 2018 to December 2019. According to the operative approach, the patients were divided into a sternotomy aortic valve replacement (SAVR) group (n = 62) and a minimally invasive aortic valve replacement (Mini-AVR) group (n = 49). We compared the intraoperative and postoperative data between the two groups. Result: There was no difference in preoperative data between the Mini-AVR and SAVR. The postoperative ventilator-assisted time, CSICU time and postoperative hospital stay of the Mini-AVR were shorter than those of the SAVR [(15.45 ± 5.75) VS (18.51 ± 6.71) h; (1.77 ± 0.31) VS (2.04 ± 0.63) d; (8.69 ± 2.75) VS (10.77 ± 2.94) d], and the difference was statistically significant (P

ABSTRACT Treatment of osteomyelitis is still challenging, as conventional antibiotic therapy is limited by the emergence of resistant strains and the formation of biofilms. Sonoantimicrobial chemotherapy (SACT) is a novel therapy of low-frequency and low-intensity ultrasound (LFLIU) combined with a sonosensitizer. Therefore, in our study, a sonosensitizer named emodin (EM) was proposed to be combined with LFLIU to relieve acute osteomyelitis caused by methicillin-resistant Staphylococcus aureus (MRSA) through antibacterial and antibiofilm effects. The efficiencies of different intensities of ultrasound, including single (S-LFLIU, 15 min) and multiple ultrasound (M-LFLIU, 3 times for 5 min at 4-h intervals), against bacteria and biofilms were compared, contributing to developing the best treatment regimen. Our results demonstrated that EM plus S-LFLIU or M-LFLIU (EM+S-LFLIU or EM+M-LFLIU) had significant combined bactericidal and antibiofilm effects, with EM+M-LFLIU in particular exhibiting superior antibiofilm performance. Furthermore, it was suggested that EM+M-LFLIU could produce a large amount of reactive oxygen species (ROS), destroy the integrity of the bacterial membrane and cell wall, and downregulate the expression of genes involved in oxidative stress, membrane wall synthesis, and bacterial virulence, as well as that of other related genes (agrB, pbp3, sgtB, gmk, zwf, and msrA). In vivo studies, micro-computed tomography (micro-CT), hematoxylin and eosin (H&E) staining, enzyme-linked immunosorbent assay (ELISA), and bacterial quantification of bone tissue indicated that EM+M-LFLIU could also relieve osteomyelitis due to MRSA infection. Our work proffers an original approach to bacterial osteomyelitis treatment that weakens drug-resistant bacteria and suppresses and degrades biofilm formation through SACT, which may provide new prospects for clinical treatment. IMPORTANCE Antibiotic therapy is the first choice for clinical treatment of osteomyelitis, but the formation of bacterial biofilms and the emergence of many drug-resistant strains also create an urgent need to find an alternative treatment to effectively eliminate the infection. Recently, LFLIU has come to be considered a safe and promising method of debridement and antibacterial therapy. In this study, we found that ultrasound and EM have a significant combined antibacterial effect in vivo and in vitro, which may play an antibacterial role by stimulating the production of ROS, destroying the bacterial cell wall, and inhibiting the expression of related genes. Our study expands the body of knowledge on the antibacterial effect of drugs—specifically emodin (EM)—through combined physiotherapy. If successfully integrated into clinical practice, these methods may reduce the burden of high concentrations of drugs needed to treat bacterial biofilms and avoid the growing resistance of bacteria to antibiotics.

Sorghum is an important food crop with high salt tolerance. Therefore, studying the salt tolerance mechanism of sorghum has great significance for understanding the salt tolerance mechanism of C4 plants. In this study, two sorghum species, LRNK1 (salt-tolerant (ST)) and LR2381 (salt-sensitive (SS)), were treated with 180 mM NaCl salt solution, and their physiological indicators were measured. Transcriptomic and metabolomic analyses were performed by Illumina sequencing and liquid chromatography-mass spectrometry (LC-MS) technology, respectively. The results demonstrated that the plant height, leaf area, and chlorophyll contents in LRNK1 were significantly higher than in LR2381. Functional analysis of differently expressed genes (DEGs) demonstrated that plant hormone signal transduction (GO:0015473), carbohydrate catabolic processes (GO:0016052), and photosynthesis (GO:0015979) were the main pathways to respond to salt stress in sorghum. The genes of the two varieties showed different expression patterns under salt stress conditions. The metabolomic data revealed different profiles of salicylic acid and betaine between LRNK1 and LR2381, which mediated the salt tolerance of sorghum. In conclusion, LRNK1 sorghum responds to salt stress via a variety of biological processes, including energy reserve, the accumulation of salicylic acid and betaine, and improving the activity of salt stress-related pathways. These discoveries provide new insights into the salt tolerance mechanism of sorghum and will contribute to sorghum breeding.

Purpose: The purpose of this study was to explore the oncogenic role of small nuclear ribonucleoprotein polypeptides B and B1 (SNRPB) in human tumors.Materials and methods: Study cases were acquired from The Cancer Genome Atlas database, the Gene Expression Omnibus database, The Human Protein Atlas, and the Clinical Proteomic Tumor Analysis Consortium. We then used the R package and several online tools to analyze and visualize the role of SNRPB across tumors.Results: We found that the expression of SNRPB was significantly increased in 28 of 33 tumors, and higher expression was observed in late pathological and TNM stages. Significantly decreased levels of SNRPB promoter methylation were observed in 12 tumors. SNRPB was found to be a risk factor for decreased overall survival in 10 tumors (p < 0.05), a risk factor for decreased disease-specific survival in 8 tumors (p < 0.05), and a risk factor for decreased progression-free interval in 7 tumors (p < 0.05). The PPI network of SNRPB and the top 100 coexpressed genes revealed that CDK1, CDC6, AURKB, CCNB1, CCNA2, and CDC45 were the most closely interacting genes across tumors. The GO and KEGG enrichment analyses revealed that SNRPB and the above genes were mainly enriched with respect to functions in cell cycle-related genetic material replication, assembly, and distribution. SNRPB was significantly associated with immune cell infiltration and the expression of immunomodulation-related genes in several but not all tumors.Conclusion and limitations: The expression of SNRPB was significantly elevated in almost all tumors, and the decreased promoter methylation level may contribute to the elevated expression of SNRPB. SNRPB may facilitate the progression of pathological and TNM stages and is a risk factor for unfavorable prognosis across tumors. However, our research was based on data obtained from public databases, without further validation of our findings at the cellular and animal levels. Therefore, further studies are needed to clarify the oncogenic mechanism of SNRPB and its potential as a therapeutic target.

China has promised to peak carbon emission before 2030 and to achieve carbon neutrality before 2060 (i.e., “double carbon” goal). Under this background, the emission reduction transformation path of China's electric power industry is studied in this paper. First, several boundary conditions (i.e., assumptions) of electric power structure transformation (i.e., the costs of power generations, the costs of energy storage systems, the developments of carbon sinks, the emission factors, and the quotas of carbon sinks) are given considering the whole society electricity consumption in the future. Second, a transformation path optimization model is established aim to minimize the total cost in the electric power industry. Then, according to the optimization results, the transformation predictions for the power industry under the ''30·60 scenario'' (i.e., the scenario that can achieve carbon peak and carbon neutrality before 2030 and 2060) are analyzed in detail, and are compared with the ones of ''2 °C scenario'' and ''1.5 °C scenario'' defined by IPCC. Furthermore, the influence of different carbon prices on the transformation path is also analyzed. It can be concluded from the simulation results that the ''30·60 scenario'' is a scenario between ''2 °C scenario'' and ''1.5 °C scenario'', and carbon emission can be reduced rapidly under the guidance of high carbon prices.

The control based on dynamic model could improve the dynamic performance of manipulators and obtain better control effects than the control based on kinematic model. As manipulators are complex online multivariable systems, there are various uncertainties in different environments and working conditions. Accurate dynamic parameters are difficult to obtain in practical engineering applications. In this article, a controller is designed by combining the precise control torques obtained by the analytical dynamics method with the compensation control torques obtained by sliding mode method for trajectory tracking of the manipulator with bounded uncertainty. Precise control torques obtained from the Udwadia–Kalaba modeling method could be applied for the control of the ideal manipulator tracking the desired trajectory. Compensation control torques obtained from the sliding mode concept and the Lyapunov stability theory could be applied to compensate the uncertainties of parameters and external disturbances, thus enhancing the robustness of the system. By combining precise control torques with compensation control torques, the end point of the manipulator with uncertainty could track the end of the ideal manipulator and then track the desired trajectory. The simulation results of the three-link manipulator with uncertainty show that the control method can make the controlled target approach the desired trajectory in relatively small torque ranges and obtain high stability accuracy, and the chattering is effectively reduced at the stage of approaching the sliding mode surface.

Slope cutting is becoming more common in engineering construction to obtain a large floor area. Slope cutting disrupts a slope’s inherent stability, causing instability and sliding. To solve the problem of geological disasters caused by artificial slope cutting, Cangxi County is used as an example to analyze the cutting slope and the height of different slope structures in the county, to study the disaster mode and disaster factors caused by slope cutting, and to analyze the influence of slope cutting height and slope cutting gradient on the disaster process. This study demonstrates that the stability of cutting on different types of slopes is weakened to different degrees, with mixed rocky-soil slopes having the strongest resistance to weakening, thick-soil slopes the second strongest, and thin-soil slopes the weakest. Slope cutting is the main factor of disaster, and rainwater infiltration promotes landslide formation. Numerical simulation is employed to determine the proximities of the slope cutting height and slope cutting gradient, and 75% of the critical value is taken as the proposed threshold value of slope cutting. The recommended cut slope threshold value corresponds to the real stable cut slope gradient on site, and the research findings can be used to guide engineering cut slope decisions in Cangxi County and other red-layer areas.

Current wound dressing based on hydrogel offers a promising way to accelerate the healing process, yet great challenges remain in the development of a highly integrated and efficient platform with the combination of therapeutic biomolecules and stem cells. Herein, a natural hydrogel wound dressing from egg white can be conveniently obtained by feasible physical crosslinking, the prepared hydrogel dressing features interconnected microporous channels, direct 3D printing, cytocompatibility, and intrinsic biomolecules to advance cell behavior. The 3D printed egg white hydrogels promote the adhesion and proliferation of adipose-derived stem cells (ASCs) without obvious cytotoxicity. In addition, this integrated hydrogel platform accompanied with adipose-derived stem cells accelerates wound healing through the enhancement of fibroblast proliferation, angiogenesis, and collagen rearrangement in the wound bed. The egg white hydrogel provides an effective wound caring product possessing low cost, easy availability along with ready manufacturing, and advanced therapeutic effect, which may be extended for the management of chronic or other complicated wounds.

This study analyzes the relationship between haze pollution and industrial agglomeration using particulate matter (PM2.5) concentration data from 269 prefecture-level cities in China between 2007 and 2016. We calculate the industrial agglomeration level using the location entropy method with industrial output as the core variable. We find that the industrial agglomeration effect is adversely affected by increased haze pollution, proving a negative correlation between them. We also compare the effect of haze pollution on industrial agglomeration before and after the 18th National Congress. The comparison shows that the effect of haze pollution on industrial agglomeration during 2012 to 2016 was significantly less than that during 2007 to 2011. Therefore, the industrial agglomeration effect can be strengthened by reducing environmental pollution through vigorous environmental governance efforts. These findings provide empirical evidence for governments’ continued implementation of current environmental protection policies.

Fibrosis is defined as the abnormal and excessive deposition of extracellular matrix (ECM) components, which leads to tissue or organ dysfunction and failure. However, the pathological mechanisms underlying fibrosis remain unclear. The inflammatory response induced by tissue injury is closely associated with tissue fibrosis. Recently, an increasing number of studies have linked necroptosis to inflammation and fibrosis. Necroptosis is a type of preprogrammed death caused by death receptors, interferons, Toll-like receptors, intracellular RNA and DNA sensors, and other mediators. These activate receptor-interacting protein kinase (RIPK) 1, which recruits and phosphorylates RIPK3. RIPK3 then phosphorylates a mixed lineage kinase domain-like protein and causes its oligomerization, leading to rapid plasma membrane permeabilization, the release of cellular contents, and exposure of damage-associated molecular patterns (DAMPs). DAMPs, as inflammatory mediators, are involved in the loss of balance between extensive inflammation and tissue regeneration, leading to remodeling, the hallmark of fibrosis. In this review, we discuss the role of necroptotic DAMPs in tissue fibrosis and highlight the inflammatory responses induced by DAMPs in tissue ECM remodeling. By summarizing the existing literature on this topic, we underscore the gaps in the current research, providing a framework for future investigations into the relationship among necroptosis, DAMPs, and fibrosis, as well as a reference for later transformation into clinical treatment.

Photothermal therapy operated in the second near-infrared (NIR-II, 1000–1700 nm) window and fluorescence imaging in the NIR-IIb (1500–1700 nm) region have become the most promising techniques in phototheranostics. Their combination enables simultaneous high-resolution optical imaging and deep-penetrating phototherapy, which is essential for high-performance phototheranostics. Herein, carboxyl-functionalized small organic photothermal molecules (Se-TC) and multi-layered NIR-IIb emissive rare-earth-doped nanoparticles (NaYF4:Yb,Er,Ce@NaYF4:Yb,Nd@NaYF4, RENP) were rationally designed and successfully synthesized. Then, high-performance hybrid phototheranostic nanoagents (Se-TC@RENP@F) were easily constructed through the coordination between Se-TC and RENP and followed by subsequent F127 encapsulation. The carboxyl groups of Se-TC can offer strong binding affinity towards rare-earth-doped nanoparticles, which help improving the stability of Se-TC@RENP@F. The multilayered structure of RENP largely enhance the NIR-IIb emission under 808 nm excitation. The obtained Se-TC@RENP@F exhibited high 1064 nm absorption (extinction coefficient: 24.7 L g−1 cm−1), large photothermal conversion efficiency (PCE, 36.9%), good NIR-IIb emission (peak: 1545 nm), as well as great photostability. Upon 1064 nm laser irradiation, high hyperthermia can be achieved to kill tumor cells efficiently. In addition, based on the excellent NIR-IIb emission of Se-TC@RENP@F, in vivo angiography and tumor detection can be realized. This work provides a distinguished paradigm for NIR-IIb-imaging-guided NIR-II photothermal therapy and establishes an artful strategy for high-performance phototheranostics.

Abstract Background Fat grafting has been regarded as a promising approach for regenerative therapy. Given the rapidly aging population, better understanding of the effect of age on fat graft outcomes and the underlying mechanisms is urgently needed. Methods C57/BL6 mice [old (O, 18–20-month-old) and young (Y, 4-month-old)] were randomized to four fat graft groups [old-to-old (O-O), young-to-young (Y-Y), old-to-young (O-Y), and young-to-old (Y-O)]. Detailed cellular events before and after grafting were investigated by histological staining, RNA sequencing, and real-time polymerase chain reaction. The adipogenic differentiation potential of adipose-derived mesenchymal stem cells (AD-MSCs) from old or young donors was investigated in vitro. Additionally, adipogenesis of AD-MSCs derived from old recipients was evaluated in the culture supernatant of old or young donor fat tissue. Results After 12 weeks, the volume of fat grafts did not significantly differ between the O-O and O-Y groups or between the Y-Y and Y-O groups, but was significantly smaller in the O-O group than in the Y-O group and in the O-Y group than in the Y-Y group. Compared with fat tissue from young mice, senescence-associated secretory phenotype (SASP) factors were upregulated in fat tissue from old mice. Compared with the Y-O group, adipogenesis markers were downregulated in the O-O group, while SASP factors including interleukin (IL)-6, tumor necrosis factor-α, and IL-1β were upregulated. In vitro, AD-MSCs from old donors showed impaired adipogenesis compared with AD-MSCs from young donors. Additionally, compared with the culture supernatant of young donor fat tissue, the culture supernatant of old donor fat tissue significantly decreased adipogenesis of AD-MSCs derived from old recipients, which might be attributable to increased levels of SASP factors. Conclusions Age has detrimental effects on fat graft outcomes by suppressing adipogenesis of AD-MSCs and upregulating expression of SASP factors, and fat graft outcomes are more dependent on donor age than on recipient age. Thus, rejuvenating fat grafts from old donors or banking younger adipose tissue for later use may be potential approaches to improve fat graft outcomes in older adults.

Purpose: Septic cardiomyopathy (SCM) is an important world public health problem with high morbidity and mortality. It is necessary to identify SCM biomarkers at the genetic level to identify new therapeutic targets and strategies.Method: DEGs in SCM were identified by comprehensive bioinformatics analysis of microarray datasets (GSE53007 and GSE79962) downloaded from the GEO database. Subsequently, bioinformatics analysis was used to conduct an in-depth exploration of DEGs, including GO and KEGG pathway enrichment analysis, PPI network construction, and key gene identification. The top ten Hub genes were identified, and then the SCM model was constructed by treating HL-1 cells and AC16 cells with LPS, and these top ten Hub genes were examined using qPCR.Result: STAT3, SOCS3, CCL2, IL1R2, JUNB, S100A9, OSMR, ZFP36, and HAMP were significantly elevated in the established SCM cells model.Conclusion: After bioinformatics analysis and experimental verification, it was demonstrated that STAT3, SOCS3, CCL2, IL1R2, JUNB, S100A9, OSMR, ZFP36, and HAMP might play important roles in SCM.

Acellular adipose matrix (AAM) has received increasing attention for soft tissue reconstruction, due to its abundant source, high long-term retention rate and in vivo adipogenic induction ability. However, the current decellularization methods inevitably affect native extracellular matrix (ECM) properties, and the residual antigens can trigger adverse immune reactions after transplantation. The behavior of host inflammatory cells mainly decides the regeneration of AAM after transplantation. In this review, recent knowledge of inflammatory cells for acellular matrix regeneration will be discussed. These advancements will inform further development of AAM products with better properties.

BackgroundAutologous fat transfer is common in breast augmentationor reconstruction. However, AFG recipient site in the breast for fat grafting has not been carefully investigated.MethodsForty female patients requiring breast augmentation with fat grafting were randomly assigned into two groups. The retromammary group received 2/3 fat into the retromammary space and the other 1/3 into the subcutaneous and retropectoral planes. The retropectoral group received 2/3 fat into the retropectoral plane and the other 1/3 into the subcutaneous and retromammary planes. The fat grafting result at 6 months was assessed by 3D laser surface scanning and then ultrasound. Any complications were recorded during follow-up. Samples from a patient who underwent fat grafting for 6 months was obtained and histological examination was conducted.ResultsNo significant difference in the retention rate after 6 months was observed between the two groups (retromammary group: 35.9% ± 6.6; retropectoral group: 39.3% ± 5.1, p = 0.1076). The retromammary grouphad a higher incidence of oil cyst formation than the retropectoral group. Histological examination showed that there were more oil cysts and mac2 positive macrophage infiltration in the fat cells in retromammary group, while retropectoral group had more small-size adipocytes.ConclusionAlthough fat grafting into the retropectoral plane did not provide a superior fat graft retention rate, it did lower the incidence of complications. The retropectoral space show great potential to become a favorable recipient site.

Ulcerative colitis (UC) is characterized by chronic relapsing and persistent inflammation that mainly affects the colon. The purpose of this study was to investigate the therapeutic effect of Agaricus blazei Murrill polysaccharides (ABP) or selenium-enriched Agaricus blazei Murrill polysaccharides (ABP-Se) on UC. UC mouse model was established by 2.5% dextran sulfate sodium (DSS). Markers for colonic barrier functions, cytokines and oxidative stress were measured. The experimental results showed that ABP and ABP-Se significantly alleviated UC-related symptoms, including weight loss, colon shorten, and disease activity index scores. Meanwhile, ABP and ABP-Se significantly promoted tight junction proteins and mucus content, and decreased inflammation and oxidative stress. Moreover, ABP-Se exhibited a better anti-oxidative ability compared to ABP. The experimental results suggest that ABP and ABP-Se relieve DSS-induced colitis via reducing inflammatory cytokines and oxidative stress, and preventing colonic barrier damage. This study suggests a promising therapeutic effect of mushrooms-derived polysaccharides on UC.

In the process of electric energy substitution, many industries have gradually replaced the traditional energy consumption equipment with new electrical ones. To avoid the disadvantage of one-sidedness in equipment evaluation and provide a more comprehensive reference for enterprises to replace equipment, a two-dimensional evaluation model composed of static and dynamic rating model is proposed in this paper based on combined weighting and rating algorithm, considering the comprehensive performance in both the inherent properties and the actual power consumption features of electrical equipment. The static and dynamic parameters related are firstly extracted, and then the weights of parameters are determined by the analytic hierarchy process (AHP) subjective weighting algorithm and principal component analysis (PCA) objective weighting algorithm. To compare the performance of different equipment more intuitively, the rank sum ratio (RSR) and technology for order preference by similarity to an ideal solution (TOPSIS) are used to rate the static and dynamic scores, respectively. Case studies for different types of electrical tea dryers in Anji, Zhejiang Province, China, show that the proposed evaluation model is more comprehensive and the rating result is more reasonable, which helps the enterprises have a more comprehensive understanding of the electrical equipment performance.

Background: The underlying pathogenesis of patients with salt-sensitive hypertension expressing higher blood pressure and severer renal damage remains uncertain. Methods: We recruited 329 subjects, 131 in salt-sensitive (SS) group, 148 in nonsalt-sensitive (NSS) group, and 50 healthy people in normal group and tested their renal function, 24-h ambulatory blood pressure, and growth factor series. Results: The SS group showed worse renal function with lower estimated glomerular filtration rate and higher urinary microalbumin, α-microglobulin, urinary protein Cr ratio, and urinary immunoglobulin. Most indicators in 24-h ambulatory blood pressure of the SS group were significantly enhanced than the NSS group, indicating their higher blood pressure. The significantly elevated growth factors in the SS group were AR, BMP-5, EG-VEGF, GH, HGF, IGFBP-2, IGFBP-3, IGFBP-6, MCSFR, NT-4, PDGF-AA, SCF, SCFR, VEGFR2, VEGFR3, and VEGF-D, compared to other 2 groups or one of them. PI3K-AKT pathway was activated in the SS group. Conclusions: Differences in growth factors and pathways may account for the manifestations of the SS group. Activated PI3K-AKT pathway with higher IGFBP-3 and GH can lead to renal damage. Higher MCSFR in the SS group indicates that high blood pressure and severe kidney damage may be associated with the activation of the immune system. EG-VEGF, VEGFR2, VEGFR3, and VEGF-D can also explain the elevated blood pressure due to the dilated lymphatic system which drains excess sodium and water back into circulation. The SS group presented higher AR and HGF which may worsen renal function by regulating cell proliferation and tumor formation. However, due to the potential low awareness rate of hypertension at the very beginning, we cannot ensure the exact occurrence order of blood pressure, renal damage, and salt sensitivity. Therefore, further studies which can track data from the onset of hypertension are needed.

Abstract Background Cancer-secreted exosomal miRNAs regulates the biological processes of many tumours. The serum level of exosomal miR-106b-5p is significantly increased in melanoma patients. However, the role and molecular mechanisms of exosomal miR-106b-5p in melanoma remains unclear. Methods Quantitative real-time polymerase chain reaction (qRT-PCR) was used to detect the expression of miR-106b-5p and EphA4 in melanoma tissues. Transmission electron microscopy (TEM) and western blotting were used to identify exosome. QRT-qPCR and Cy3-labelled miR-106b-5p were used to demonstrated the transmission of melanoma cell-secreted exosomal miR-106b-5p. Western blotting, Immunofluorescence, adhesion, transwell and scratch wound assay were used to explore the role of exosomal miR-106b-5p in melanocytes. Luciferase reporter assays and RNA-Chromatin Immunoprecipitation (ChIP) assay were used to confirm whether erythropoietin-producing hepatocellular carcinoma receptor A4 (EphA4) was a direct target of miR-106b-5p. Results We found that miR-106b-5p levels were increased in melanoma tissue, and high miR-106b-5p expression is an independent risk factor for the overall survival of patients with melanoma. miR-106b-5p is enriched in melanoma cell-secreted exosomes and transferred to melanocytes. Exosomal miR-106b-5p promotes the epithelial-to-mesenchymal transition (EMT), migration, invasion and adhesion of melanocytes. Exosomal miR-106b-5p exerted its role by targeting EphA4 to activate the ERK pathway. We demonstrated that exosomal miR-106b-5p promoted melanoma metastasis in vivo through pulmonary metastasis assay. Conclusions Thus, melanoma cell-secreted exosomal miR-106b-5p may serve as a diagnostic indicator and potential therapeutic target in melanoma patients.

Accurate stratospheric temperature observations are crucial for weather forecasts and climate change studies. This paper discusses a precise measurement method for the stratospheric temperature profile using narrow bands with ultra-high spectral resolution from nadir-viewing satellites. First, the CO2 absorption band around 15 μm is selected as the major sounding source by the calculation and analysis of the temperature Jacobian and the atmospheric molecular spectra. Next, the influence of spectral resolution, spectral range and instrumental noise on the sounding capability is analyzed, and the sounding feasibility of the single spectral band and multiple spectral bands is discussed under the condition that the spaceborne long-wave infrared space heterodyne spectrometer (SHS) is selected as suggested sounder onboard the satellite. Finally, the optimal joint-sounding scheme of narrow bands is proposed. The temperature retrieval and validation show that the joint-sounding of two discontinuous narrow bands can realize the high precision measurement of the stratospheric temperature profile for the given spectral resolution, spectral range, and instrumental noise. When the sounder adopts two narrow bands (the regions of 666.87–676.44 cm−1 and 683.58–693.15 cm−1) and a spectral resolution of 0.03 cm−1, the retrieval accuracy (RMSE) is about 0.9 K over a pressure range of 200 to 0.7 hPa (11.5–50 km). This study will provide technical preparation for high-precision and low-cost satellite sounder design for stratospheric temperature observations.
