Abstract

The established role of vitamin K in nutrition is as a cofactor in the post-translational conversion of specific glutamyl to [Gamma]-carboxyglutamyl (Gla) residues in a limited number of proteins. Administration of the vitamin K antagonist warfarin has previously been shown to decrease brain sulfatide concentrations and decrease brain galactocerebroside sulfotransferase (GST) activity in young mice. A dietary deficiency of vitamin K has now been shown to decrease (P < 0.01) brain sulfatide concentrations of 30-d-old mice significantly (by 21%). Male 21-d-old rats fed an excess of vitamin K for 7 or 14 d had 26 and 31% (P < 0.05) greater GST activity and 15 and 18% (P < 0.05) greater brain sulfatide concentrations, respectively, than controls fed a vitamin K-deficient diet. Male 21-d-old rats fed a diet containing 500 mg of phylloquinone/kg diet had an intermediate response and were vitamin K sufficient by normal criteria. The vitamin K response was observed when either phylloquinone or menaquinone-4 was fed as a source of the vitamin. These data suggest that in addition to its recognized role in Gla synthesis, vitamin K status is important in the maintenance of normal complex lipid sulfatide metabolism in young rats and mice. Abstract

To determine whether curcumin (Cur) can treat mice with experimentally-induced colitis by regulating follicular helper T cells (Tfh) and follicular regulatory T cells (Tfr) by inhibiting interleukin (IL)-21. In this study, 40 male C57BL/6 mice were randomly grouped into four groups, i.e., normal, trinitrobenzene sulfonic acid (TNBS), TNBS + curcumin, and TNBS + anti- IL-21. Mice with experimental colitis were induced by 100 mg/kg TNBS. The mice in the TNBS + Cur group were treated with 100 mg/kg curcumin for seven days, and mice in the TNBS + anti-IL-21 group were treated with anti-IL-21 (150 μg/mouse) once per week, intraperitoneally, starting on the second day after establishing the experimental colitis model. On day eight, the therapeutic effect of curcumin was evaluated by colon mucosa damage index (CMDI), histological examination, and disease activity index (DAI). Furthermore, the number of CD4 + CXCR5 + PD-1 + Tfh and CD4 + CXCR5 + FoxP3 + Tfr cells were measured by flow cytometry. The mRNA and protein expression of IL-21, Bcl-6, FOXP3, ICOS, and PD-1 in colonic mucosa was detected by reverse transcription polymerase chain reaction and the Western blot technique. Compared with the TNBS group, the DAI, CMDI, histological score, the number of CD4 + CXCR5 + PD-1 + Tfh cells, the expression of IL-21, Bcl-6, ICOS, and PD-1 were significantly decreased in the TNBS + curcumin group and TNBS + anti- IL-21 group; body weight, number of CD4 + CXCR5 + FoxP3 + Tfr cells, and the expression of FoxP3 were observably elevated in the TNBS + curcumin group (all P < 0.05). Curcumin may have a potential therapeutic effect on mice with colitis treated experimentally through regulation of the balance of Tfh and Tfr cells via inhibiting the synthesis of IL-21. Abstract

To overcome the dependence of the plasma extraction on siphon hydrophilicity in a centrifugal microfluidic chip and to ensure long-term stability and reliability of the microfluidic chip, a compressed air reservoir connected to the blood separated reservoir was proposed to separate the blood in blood separated reservoir at high rotating speeds. Through lowering the spinning speed, the plasma in the separated reservoir was pumped toward the CD-like chip center to release the stored air pressure produced by the rotation at high spinning speeds. Based on the thermodynamics of the isothermal gas, the pneumatic pumping method was modeled, and the model was confirmed by analyzing pumping positions and air compression versus spinning speeds. By using the layered Polymethyl Methacrylate(PMMA)as materials, the centrifugal microfluidic chips for plasma extraction were fabricated with a CO2 laser process technique and the volume of compressed air and the blood position at siphon were tested at different rotational speeds. Experimental results demonstrate that the achieved volume of compression is 8.7L and the siphon valve effectively inhibits whole blood to overflow the crest of siphon at 4 000 r/min speed. With lowering the spin speed to 1 000 r/min, the release of compressed air overcomes the centrifugal force and drives the plasma to flow past the crest of siphon. Then the plasma extraction is realized in quantitation.
