Le projet PROTÉGER vise à promouvoir les techniques de génie végétal pour préserver l’extrême biodiversité des milieux rivulaires de Guadeloupe, tout en protégeant la population et leurs biens, des risques encourus lors des crues de rivière et des évènements cycloniques. La première phase du projet a consisté à décrire les ripisylves de l’archipel, afin d’en connaitre les modèles naturels et de pouvoir s’en inspirer pour développer des solutions fondées sur la nature. Sur les 270 berges échantillonnées, 302 espèces dont 231 indigènes ont été rencontrées, soit 10 % de la flore locale, reflétant la nature très forestière de l’archipel et son exceptionnelle diversité. L’analyse de ces relevés a permis de définir 12 types de ripisylves, et de documenter 80 espèces potentiellement utilisables en génie végétal. L’étude présente 30 d'entre elles sous forme de fiches individuelles disponibles sur le site internet du projet avec leur description, leur phénologie, leur statut de protection, leur écologie, leur usage et leur condition d’utilisation. Au-delà de la typologie et des fiches spécifiques, ce travail a aussi montré l’état alarmant des ripisylves de Guadeloupe, notamment sur la partie aval des cours d‘eau, avec des habitats dégradés par la pollution et la présence d’espèces exotiques envahissantes.

Engineered protein therapeutics offer advantages, including strong target affinity, selectivity and low toxicity, but like natural proteins can be susceptible to proteolytic degradation, thereby limiting their effectiveness. A compelling therapeutic target is mesotrypsin, a protease up-regulated with tumour progression, associated with poor prognosis, and implicated in tumour growth and progression of many cancers. However, with its unique capability for cleavage and inactivation of proteinaceous inhibitors, mesotrypsin presents a formidable challenge to the development of biological inhibitors. We used a powerful yeast display platform for directed evolution, employing a novel multi-modal library screening strategy, to engineer the human amyloid precursor protein Kunitz protease inhibitor domain (APPI) simultaneously for increased proteolytic stability, stronger binding affinity and improved selectivity for mesotrypsin inhibition. We identified a triple mutant APPIM17G/I18F/F34V, with a mesotrypsin inhibition constant (Ki) of 89 pM, as the strongest mesotrypsin inhibitor yet reported; this variant displays 1459-fold improved affinity, up to 350 000-fold greater specificity and 83-fold improved proteolytic stability compared with wild-type APPI. We demonstrated that APPIM17G/I18F/F34V acts as a functional inhibitor in cell-based models of mesotrypsin-dependent prostate cancer cellular invasiveness. Additionally, by solving the crystal structure of the APPIM17G/I18F/F34V-mesotrypsin complex, we obtained new insights into the structural and mechanistic basis for improved binding and proteolytic resistance. Our study identifies a promising mesotrypsin inhibitor as a starting point for development of anticancer protein therapeutics and establishes proof-of-principle for a novel library screening approach that will be widely applicable for simultaneously evolving proteolytic stability in tandem with desired functionality for diverse protein scaffolds.

Exclusionary policies are practiced widely in schools despite being associated with extremely poor outcomes for culturally and linguistically diverse students, particularly African American males with and without disabilities. This article discusses zero tolerance policies, the related research questioning their basic assumptions, and the negative effects on students in special education and the larger society. Behavioral and academic interventions also are discussed relative to evidence of effectiveness, potential outcomes, and culturally responsive applications.

In a recent paper, Jones et al. (2020a) claimed that we recommended the use of mowing for the “landscape management of invasive knotweeds” in an article we published earlier this year (i.e. Martin et al. 2020), a recommendation with which they strongly disagreed. Since we never made such a recommendation and since we think that, in order to successfully control invasions by Japanese knotweed s.l. taxa (Reynoutria spp.; syn. Fallopia spp.), stakeholders need to acknowledge the general complexity of the management of invasive clonal plants, we would like to (i) clarify the intentions of our initial article and (ii) respectfully discuss some of the statements made by Daniel Jones and his colleagues regarding mowing and knotweed management in general. Although we agree with Jones et al. that some ill-advised management decisions can lead to “cures worse than the disease”, our concern is that the seemingly one-sided argumentation used by these authors may mislead managers into thinking that a unique control option is sufficient to tackle knotweed invasions in every situation or at any given spatial scale, when it is generally admitted that management decisions should account for context-dependency (Wittenberg and Cock 2001; Pyšek and Richardson 2010; Kettenring and Adams 2011).

Les berges de cours d’eau sont particulièrement affectées par les invasions biologiques qui peuvent menacer la bodiversité et le fonctionnement de ces milieux. En s'appuyant sur les résultats d'une étude portant sur plusieurs types d'aménagements de berges de rivières, cette note permet de mieux comprendre l’intérêt du génie végétal pour diminuer les risques de prolifération des espèces invasives sur les berges de rivière.

Transitions entre les milieux aquatiques et terrestres, corridors écologiques pour le déplacement de nombreuses espèces, refuge de biodiversité... les ripisylves ont un rôle majeur pour la connectivité écologique des territoires. Cette note apporte des éléments pour mieux comprendre l'intérêt de préserver ces milieux et favoriser une meilleure prise en compte de la connectivité écologique dans l’aménagement et la gestion des berges de cours d’eau.

Many of the most invasive plant species in the world can propagate clonally, suggesting clonality offers advantages that facilitate invasion. Gaining insights into the clonal growth dynamics of invasive plants should thus improve understanding of the mechanisms of their dominance, resilience and expansion. Belonging to the shortlist of the most problematic terrestrial invaders, Reynoutria japonica var. japonica Houtt. (Japanese knotweed) has colonized all five continents, likely facilitated by its impressive ability to propagate vegetatively. However, its clonal growth patterns are surprisingly understudied; we still do not know how individuals respond to key environmental conditions, including light availability and disturbance. To contribute to filling this knowledge gap, we designed a mesocosm experiment to observe the morphological variation in R. japonica growth in homogeneous or heterogeneous conditions of light stress (shade) and disturbance (mowing). Rhizome fragments were planted in the middle of large pots between two habitat patches that consisted of either one or a combination of the following three environmental conditions: full light without mowing, full light with frequent mowing, or shade without mowing. At the end of the experiment, biomass and traits related to clonal growth (spacer and rhizome lengths, number of rhizome branches, and number of ramets) were measured. After 14 months, all individuals had survived, even those frequently mowed or growing under heavy shade. We showed that R. japonica adopts a ‘phalanx’ growth form when growing in full light and a ‘guerrilla’ form when entirely shaded. The former is characteristic of a space-occupancy strategy while the latter is more associated with a foraging strategy. In heterogeneous conditions, we also showed that clones seemed to invest preferentially more in favorable habitat patches rather than in unfavorable ones (mowed or shaded), possibly exhibiting an escape strategy. These observations could improve the management of this species, specifically by illustrating how aggressive early control measures must be, by highlighting the importance of repeated mowing of entire stands, as this plant appears to compensate readily to partial mowing, and by informing on its potential responses towards the restoration of a cover of competitive native plants.

ObjectiveTo determine the initial effectiveness of a novel, pediatric office-based intervention in motivating mothers to seek further assessment of positive depression screens.MethodsIn this pilot randomized controlled trial, English-speaking mothers (n = 104) with positive 2-question depression screens and presenting with children 0 to 12 years old for well-child care to a general pediatric training clinic received interventions from a trained research assistant. The Motivating Our Mothers (MOM) intervention included office-based written and verbal targeted depression education and motivational messages encouraging further depression assessment and a semistructured telephone booster delivered 2 days later. The control intervention included nontargeted written and verbal messages and 2 days later, an attention control telephone survey. Both groups received a list of depression care resources. The primary outcome was the proportion of mothers in each group who reported trying to contact any of 6 types of resources to discuss the positive screen at 2 weeks after intervention (ClinicalTrials.gov NCT01453790).ResultsDespite 6 contact attempts, 10 MOM and 9 control mothers were lost to follow-up. More mothers in the MOM intervention tried to contact a resource compared to control (73.8% vs 53.5%, difference 20.3%, 95% confidence interval for difference -0.1 to 38.5, P = .052).ConclusionsMothers receiving the MOM intervention made more attempts to contact a resource for follow-up of positive depression screens. If found effective in larger studies, MOM may prove a promising approach for motivating depression screen-positive mothers identified in general pediatric settings within and beyond the postpartum period to seek further depression assessment and support.

Fleuve fortement aménagé, le Rhône fait l'objet depuis plus de vingt ans de nombreux travaux de restauration écologique et hydraulique, accompagnés d'un programme scientifique de suivi et d'évaluation des effets des actions sur les milieux et la biodiversité. Dans ce contexte, d'anciens ouvrages de protection des berges ont été démantelés avec comme objectifs le développement d'espèces végétales liées au fleuve et la création de milieux nouveaux pour favoriser une dynamique latérale du Rhône. Dans cet article, les chercheurs nous présentent les premiers résultats prometteurs du suivi des sites restaurés et proposent des actions de restauration et de gestion pour le futur.

Abstract Background Arthropods comprise the largest and most diverse phylum on Earth and play vital roles in nearly every ecosystem. Their diversity stems in part from variations on a conserved body plan, resulting from and recorded in adaptive changes in the genome. Dissection of the genomic record of sequence change enables broad questions regarding genome evolution to be addressed, even across hyper-diverse taxa within arthropods. Results Using 76 whole genome sequences representing 21 orders spanning more than 500 million years of arthropod evolution, we document changes in gene and protein domain content and provide temporal and phylogenetic context for interpreting these innovations. We identify many novel gene families that arose early in the evolution of arthropods and during the diversification of insects into modern orders. We reveal unexpected variation in patterns of DNA methylation across arthropods and examples of gene family and protein domain evolution coincident with the appearance of notable phenotypic and physiological adaptations such as flight, metamorphosis, sociality, and chemoperception. Conclusions These analyses demonstrate how large-scale comparative genomics can provide broad new insights into the genotype to phenotype map and generate testable hypotheses about the evolution of animal diversity.

BackgroundCareful management of symptoms, particularly sleep and mood disturbances, may assist women in discontinuing hormone therapy (HT). We sought to describe characteristics associated with successful HT cessation in women who attempted to discontinue estrogen pills/patches with or without progestin.MethodsWe invited 2,328 women, aged 45-70, enrolled January 1, 2005, to May 31, 2006, at Group Health in Washington State and Harvard Vanguard Medical Associates in Massachusetts, to participate in a telephone survey about HT practices. For the sample, we selected 2,090 women with estrogen dispensings (pharmacy data) during the study period, 200 women without HT dispensing after January 2005, and 240 women with no estrogen dispensings; 1,358 (58.3%) completed the survey. These analyses are based on survey responses.ResultsAmong 802 women who attempted HT discontinuation, the mean age was 50 years, 93% were postmenopausal, 90% were white, 30% had had a hysterectomy, and 75% experienced hot flashes after discontinuation. Those who did not succeed had greater trouble sleeping (74% vs. 57%) and mood disturbances (51% vs. 34%) than those who succeeded. In multivariable analyses, factors associated with successful discontinuation included doctor advice (odds ratio [OR] 2.62, 95% confidence interval [CI] 1.68-4.08), lack of symptom improvement (OR 4.21, CI 1.50-12.17), vaginal bleeding (OR 5.96, CI 1.44-24.6), and learning to cope with symptoms (OR 3.36, CI 2.21-5.11). Factors associated with unsuccessful HT discontinuation included trouble sleeping (OR 0.40, CI 0.26-0.61) and mood swings or depression (OR 0.63, CI 0.42-0.92).ConclusionsDoctor advice is strongly associated with successful HT discontinuation. Symptom management, particularly sleep and mood disturbances, may help women discontinue HT.

Marked differences in the prevalence of allergy between urban and rural environments have been well established. Certain microorganisms encountered in and around farming and rural surroundings can 'train' and modulate the human immune system to protect against certain infectious and non-communicable diseases, including allergies. Research from South Africa has indicated that drinking fermented cow's milk from a young age may protect against allergy even in a urban population. Lactic acid bacteria are important role-players in allergy protection and these organisms are abundant in fermented food products. The questions arise: Can people living in urban areas, at least to some extent, 'replicate' the allergy protective environment of rural people or at least compensate for a lack of exposure to protective organisms? And can they do so safely? In this review we focus on fermented milk and other fermented foods and elaborate on the allergy-protective microbiota favoured by fermentation. [ABSTRACT FROM AUTHOR]

Abstract Background Patients with mental illness are frequently treated in primary care, where Primary Care Providers (PCPs) report feeling ill-equipped to manage their care. Team-based models of care improve outcomes for patients with mental illness, but multiple barriers limit adoption. Barriers include practical issues and psychosocial factors associated with the reorganization of care. Practice facilitation can improve implementation, but does not directly address the psychosocial factors or gaps in PCP skills in managing mental illness. To address these gaps, we developed Relational Team Development (RELATED). Methods RELATED is an implementation strategy combining practice facilitation and psychology clinical supervision methodologies to improve implementation of team-based care. It includes PCP-level clinical coaching and a team-level practice change activity. We performed a preliminary assessment of RELATED with a convergent parallel mixed method study in 2 primary care clinics in an urban Federally Qualified Health Center in Southwest, USA, 2017-2018. Study participants included PCPs, clinic staff, and patient representatives. Clinic staff and patients were recruited for the practice change activity only. Primary outcomes were feasibility and acceptability. Feasibility was assessed as ease of recruitment and implementation. Acceptability was measured in surveys of PCPs and staff and focus groups. We conducted semi-structured focus groups with 3 participant groups in each clinic: PCPs; staff and patients; and leadership. Secondary outcomes were change in pre- post- intervention PCP self-efficacy in mental illness management and team-based care. We conducted qualitative observations to better understand clinic climate. Results We recruited 18 PCPs, 17 staff members, and 3 patient representatives. We ended recruitment early due to over recruitment. Both clinics developed and implemented practice change activities. The mean acceptability score was 3.7 (SD=0.3) on a 4-point Likert scale. Les techniques de génie végétal sont de plus en plus mobilisées pour restaurer les milieux érodés ou dégradés comme les talus ou les berges des rivières. Parmi les pratiques utilisées, on peut citer l’ensemencement qui consiste à répandre sur le sol des mélanges de graines d’espèces le plus souvent herbacées. En s'appuyant sur le suivi de dix-huit chantiers de restauration de berge, cet article s'intéresse à la réussite des ensemencements réalisés en fonction de différents paramètres : recours à des semences certifiées, poids des graines et présence relative des espèces spontanées.

Abstract Background Team-based models of care are efficacious in improving outcomes for patients with mental and physical illnesses. However, primary care clinics have been slow to adopt these models. We used iterative stakeholder engagement to develop an intervention to improve the implementation of team-based care for this complex population. Methods We developed the initial framework for Relational Team Development (RELATED) from a qualitative study of Primary Care Providers’ (PCPs’) experiences treating mental illness and a literature review of practice facilitation and psychology clinical supervision. Subsequently, we surveyed 900 Colorado PCPs to identify factors associated with PCP self-efficacy in management of mental illness and team-based care. We then conducted two focus groups for feedback on RELATED. Lastly, we convened an expert panel to refine the intervention. Results We developed RELATED, a two-part intervention delivered by a practice facilitator with a background in clinical psychology. The facilitator observes PCPs during patient visits and provides individualized coaching. Next, the facilitator guides the primary care team through a practice change activity with a focus on relational team dynamics. Conclusion The iterative development of RELATED using stakeholder engagement offers a model for the development of interventions tailored to the needs of these stakeholders. Les techniques de génie végétal pour la stabilisation des berges de cours d'eau restent encore peu utilisées au Québec. Alternatives au génie civil, elles offrent pourtant de nombreux avantages structurels, environnementaux et récréatifs. Alors comment expliquer cette inertie et favoriser le développement de ces techniques ? Cet article fait un point sur le contexte spécifique québécois, apporte des connaissances sur les ouvrages de génie végétal existants et présente des premiers résultats d'expérimentations.
