Here, we present a large (n = 107,207) genome-wide association study (GWAS) of general cognitive ability (“g”), further enhanced by combining results with a large-scale GWAS of educational attainment. We identified 70 independent genomic loci associated with general cognitive ability. Results showed significant enrichment for genes causing Mendelian disorders with an intellectual disability phenotype. Competitive pathway analysis implicated the biological processes of neurogenesis and synaptic regulation, as well as the gene targets of two pharmacologic agents: cinnarizine, a T-type calcium channel blocker, and LY97241, a potassium channel inhibitor. Transcriptome-wide and epigenome-wide analysis revealed that the implicated loci were enriched for genes expressed across all brain regions (most strongly in the cerebellum). Enrichment was exclusive to genes expressed in neurons but not oligodendrocytes or astrocytes. Finally, we report genetic correlations between cognitive ability and disparate phenotypes including psychiatric disorders, several autoimmune disorders, longevity, and maternal age at first birth.

In response to immune pressure, influenza viruses evolve, producing drifted variants capable of escaping immune recognition. One strategy for inducing a broad-spectrum immune response capable of recognizing multiple antigenically diverse strains is to target conserved proteins or protein domains. To that end, we assessed the efficacy and immunogenicity of mRNA vaccines encoding either the conserved stem domain of a group 1 hemagglutinin (HA), a group 2 nucleoprotein (NP), or a combination of the two antigens in mice, as well as evaluated immunogenicity in naïve and influenza seropositive nonhuman primates (NHPs). HA stem-immunized animals developed a robust anti-stem antibody binding titer, and serum antibodies recognized antigenically distinct group 1 HA proteins. These antibodies showed little to no neutralizing activity in vitro but were active in an assay measuring induction of antibody-dependent cellular cytotoxicity. HA-directed cell-mediated immunity was weak following HA stem mRNA vaccination; however, robust CD4 and CD8 T cell responses were detected in both mice and NHPs after immunization with mRNA vaccines encoding NP. Both HA stem and NP mRNA vaccines partially protected mice from morbidity following lethal influenza virus challenge, and superior efficacy against two different H1N1 strains was observed when the antigens were combined. In vivo T cell depletion suggested that anti-NP cell-mediated immunity contributed to protection in the mouse model. Taken together, these data show that mRNA vaccines encoding conserved influenza antigens, like HA stem and NP in combination, induce broadly reactive humoral responses as well as cell-mediated immunity in mice and NHPs, providing protection against homologous and heterologous influenza infection in mice.

105. Ductile tearing of aluminium plates: experiments and modelling

This paper presents an experimental and numerical study on quasi-static ductile tearing of thin plates of the aluminium alloy AA6016 in three tempers. Depending on the temper, the main fracture mechanism in the plate tearing tests changes from grain boundary failure to coalescence of voids nucleated at the constituent particles. The experiments are complemented by nonlinear finite element simulations using an enriched Gurson-TvergaardNeedleman (GTN) model to describe the material response. The onset of accelerated void growth is initiated either by incipient material softening (named the softening model) or by the occurrence of strain localization (named the localization model). It was found that strain localization takes place at a critical porosity f(c), which depends on the current hydrostatic and deviatoric stress states. While the failure strain depends on the stress path, the critical porosity appears to be path independent. A third method is proposed (named the f(c)( T, L) model), where a critical porosity surface f(c) = f(c) (T, L) is used to determine when accelerated void growth starts. The surface is generated beforehand by solving for strain localization under proportional stress states defined by the stress triaxiality T and the Lode parameter L. By comparing the simulations to the experiments, it was found that the localization model performed well for a wide range of stress states. The softening model does not portray dependence on the Lode parameter and is therefore less versatile. Human cytomegalovirus (HCMV) is a ubiquitous pathogen that can cause disability in newborns and serious clinical diseases in immunocompromised patients. HCMV has a large genome with enormous coding potential; its viral particles are equipped with complicated glycoprotein complexes and can infect a wide range of human cells. Although multiple host cellular receptors interacting with viral glycoproteins have been reported, the mechanism of HCMV infection remains a mystery. Here we report identification of adipocyte plasma membrane-associated protein (APMAP) as a novel modulator active in the early stage of HCMV infection. APMAP is necessary for HCMV infection in both epithelial cells and fibroblasts; knockdown of APMAP expression significantly reduced HCMV infection of these cells. Interestingly, ectopic expression of human APMAP in cells refractory to HCMV infection, such as canine MDCK and murine NIH/3T3 cells, promoted HCMV infection. Furthermore, reduction in viral immediate early (IE) gene transcription at 6 h post infection and delayed nucleus translocation of tegument delivered pp65 at 4 h post infection were detected in APMAP-deficient cells but not in the wildtype cells. These results suggest that APMAP plays a role in the early stage of HCMV infection. Results from biochemical studies of APMAP and HCMV proteins suggest that APMAP could participate in HCMV infection through interaction with gH/gL containing glycoprotein complexes at low pH and mediate nucleus translocation of tegument pp65. Taken together, our results suggest that APMAP functions as a modulator promoting HCMV infection in multiple cell types and is an important player in the complex HCMV infection mechanism.

R01MH116269 and R01MH121455 [to DMR]), NIGMS of the National Institutes of Health (Grant No. T32GM007347 [to JK]), and the Brain & Behavior Research Foundation (NARSAD Young Investigator Award No. 29551 [to NM])., BACKGROUND: Suicide is a leading cause of death worldwide, and nonfatal suicide attempts, which occur far more frequently, are a major source of disability and social and economic burden. Both have substantial genetic etiology, which is partially shared and partially distinct from that of related psychiatric disorders. METHODS: We conducted a genome-wide association study (GWAS) of 29,782 suicide attempt (SA) cases and 519,961 controls in the International Suicide Genetics Consortium (ISGC). The GWAS of SA was conditioned on psychiatric disorders using GWAS summary statistics via multitrait-based conditional and joint analysis, to remove genetic effects on SA mediated by psychiatric disorders. We investigated the shared and divergent genetic architectures of SA, psychiatric disorders, and other known risk factors. RESULTS: Two loci reached genome-wide significance for SA: the major histocompatibility complex and an intergenic locus on chromosome 7, the latter of which remained associated with SA after conditioning on psychiatric disorders and replicated in an independent cohort from the Million Veteran Program. This locus has been implicated in risk-taking behavior, smoking, and insomnia. SA showed strong genetic correlation with psychiatric disorders, particularly major depression, and also with smoking, pain, risk-taking behavior, sleep disturbances, lower educational attainment, reproductive traits, lower socioeconomic status, and poorer general health. After conditioning on psychiatric disorders, the genetic correlations between SA and psychiatric disorders decreased, whereas those with nonpsychiatric traits remained largely unchanged. Special Issue: The ENIGMA Consortium: the first 10 years., Delineating the association of age and cortical thickness in healthy individuals is critical given the association of cortical thickness with cognition and behavior. Previous research has shown that robust estimates of the association between age and brain morphometry require large-scale studies. In response, we used cross-sectional data from 17,075 individuals aged 3–90 years from the Enhancing Neuroimaging Genetics through Meta-Analysis (ENIGMA) Consortium to infer age-related changes in cortical thickness. We used fractional polynomial (FP) regression to quantify the association between age and cortical thickness, and we computed normalized growth centiles using the parametric Lambda, Mu, and Sigma method. Interindividual variability was estimated using meta-analysis and one-way analysis of variance. For most regions, their highest cortical thickness value was observed in childhood. Age and cortical thickness showed a negative association; the slope was steeper up to the third decade of life and more gradual thereafter; notable exceptions to this general pattern were entorhinal, temporopolar, and anterior cingulate cortices. Interindividual variability was largest in temporal and frontal regions across the lifespan. Age and its FP combinations explained up to 59% variance in cortical thickness. 113. Algebraisk tenkning blant norske grunnskolelærerstudenter

- Abstract
Algebra har i flere tiår vært en betydelig årsak til nedgang i matematikkprestasjoner i norsk skole, og manglende kompetanse i emnet kan knyttes til frafall i yrkesutdanninger (Grønmo et al., 2010; NOKUT, 2008). Til tross for algebraens viktige betydning i samfunnet viser både lokale og globale undersøkelser at dette er et utfordrende område, både for elever og lærerstudenter (Grønmo & Onstad, 2012; NOKUT, 2023). Forskning viser videre at elevenes forståelse i matematikk i stor grad avhenger av lærernes kunnskap, noe som understreker viktigheten av å undersøke lærerstudenters ferdigheter i algebraisk tenkning. På bakgrunn av dette er formålet med denne kvalitative studien å undersøke algebraisk tenkning blant norske lærerstudenter ved den 5-årige grunnskolelærerutdanningen for 5.-10. trinn. Studien undersøker spesielt studentenes forståelse av sammenhengen mellom størrelser i arbeid med representasjoner, som er sentral del av algebra. Vi gjennomførte en analyse av 170 eksamensbesvarelser fra Nasjonalt organ for kvalitet i utdanningen (NOKUT) sin deleksamen i algebra, gjennomført av lærerstudenter ved den norske grunnskolelærerutdanningen for 5.-10. trinn. Det ble gjennomført en innholdsanalyse for å identifisere interessante funn omhandlende studentenes algebraiske tenkning. Deretter ble det utført en semiotisk analyse av studentbesvarelsene for å undersøke lærerstudentenes forståelse for sammenhenger mellom størrelser. For å se etter tegn til forståelse hos studentene ble hovedsakelig Duvals (2006) rammeverk benyttet, som utdyper at det å gjenkjenne matematiske objekter gjennom ulike representasjoner er essensielt for å snakke om forståelse. Resultatene fra studiet indikerte at lærerstudentene mestret direkte representasjonsoverganger som i mindre grad krevde å se sammenhenger. Imidlertid møtte studentene utfordringer knyttet til indirekte representasjonsoverganger, som i større grad krevde en kognitiv tolkningsprosess av relasjoner i representasjonene. I disse transformasjonene mellom representasjonene, kom ikke studentenes forståelse for begrepene funksjoner og variablers frem i stor grad. Resultatene synliggjør betydningen av en systematisk forbedring av grunnskolelærerutdanning i matematikk, med økt fokus på relasjoner. Lærernes fag- og pedagogiske kunnskap er viktige faktorer for elevers prestasjoner og forståelse i algebra, og dette er dermed et avgjørende aspekt for å bedre elevenes algebraforståelse.

In the realm of advertising, strategic placement and presentation of advertisements are crucial for attracting potential clients. Media companies employ various tactics, such as visually appealing features and vibrant colors, to capture the attention of consumers. However, achieving this objective is not always straightforward, as some advertising strategies may be perceived as irrelevant or disturbing by recipients. This Master's thesis aims to explore the relationship between audience interaction and the perception of advertisements on media platforms, with the overarching goal of enhancing advertising effectiveness and addressing ethical concerns associated with targeted advertising. To delve into this topic comprehensively, this study utilizes real-time data provided by Amedia, one of the largest media companies in Norway. Through an extensive analysis of this real-world data, the research aims to explore the correlation between audience interaction and the perception of advertisements on media platforms. This investigation involves the extraction of relevant features from advertisement images, leading to the creation of a new dataset. Concurrently, predictive machine learning models are developed to gain insights into effective advertising strategies for media companies, with a focus on personalization. Furthermore, a comprehensive user study is conducted to gain insights into user behavior within media platform advertisements. By uncovering the interplay between visual features, user behavior, and advertising effectiveness, this research contributes to improving personalized advertising strategies in the context of media companies. 115. Antibiotikaresistens - En global helsetrussel

Bakgrunn: Antibiotikaresistens er et økende globalt problem og en trussel mot folkehelsen (WHO, 2021) og ved å neglisjere problemet vil det kunne bli forskjell på liv og død for veldig mange pasienter i fremtiden. Sykepleiere har et daglig ansvar for administrering av antibiotika og har mesteparten av pasientkontakten. Dette gjør at hver og en sykepleier står i en posisjon til å kunne utgjøre en forskjell. Problemstilling: Hvordan oppleves og beskrives sykepleiers arbeid i forebyggingen av antibiotikaresistens? Hensikt: Å skildre sykepleiernes arbeid i forebyggingen av antibiotikaresistens. Metode: En litteraturstudie med bruk av tre kvalitative og to kvantitative vitenskapelige artikler og annen relevant litteratur for å besvare problemstillingen. Resultater: Resultatene viser at sykepleierne har variert kunnskap, men er bevisste på sitt ansvar knyttet til antibiotikaresistens. Sykepleierne hadde et ønske om å tilegne seg mer kunnskap gjennom bedre tilrettelegging av trening og undervisning på arbeidsplassen. Resultatene viste også at sykepleierne sto overfor praktiske utfordringer som lå utenfor deres kontroll.

- Abstract
Denne masteroppgåva er ein kvalitativ studie som tek for seg djupneintervju med fem lærarar, fire naturfagslærarar og ein samfunnsfaglærar. Problemstillinga er kva styrar seksualitetsundervisinga på mellomtrinnet? For å svare på denne problemstillinga har eg tatt for meg tre forskingsspørsmål; kva forståing har lærarar for seksualitetsundervising, kva styrar lærarane sine didaktiske val og korleis brukar lærarane ulike læringsressursar. I datamaterialet kom det fram at lærarane har ulike måtar å undervise på. Alle lærarane har ein elevsentrert orientering, men dei har ulik grad av fagorientering. Nokon er meir opptatt av kva faget og læreplanen fokuserer på, medan andre ser på alternative element som viktigare. Gjennom dette fokuset på læreplanen kom det fram at nokon av lærarane tek i bruk bolkundervising. Det blir då lagt vekt på pubertet på 5. trinn, og anten ingenting eller om seksuell åtferd på 7. trinn. Den heilskaplege tilnærminga legg bakgrunnen for drøftinga. Eg har sett på korleis lærarane stiller seg til seksualitetsundervising, opp i mot kva den heilskaplege tilnærminga legg vekt på. Den vektlegg eit heilskapleg fokus som skal famne alle aspekt ved seksualitet, kjønn og kropp; dette for å kunne gi barn best moglege liv (Støle-Nilsen, 2022, s. 18). Fagfornyinga peikar mot ein meir heilskapleg tilnærming, samt eit tverrfagleg samarbeid om seksualitetsundervising. Dette fokuset ser ein òg gjennom Snakk om det!-strategien frå regjeringa, som fokuserte på «en helhetlig innsats for å styrke hele befolkningens seksuelle helse» (Helse- og omsorgsdepartementet, 2016, s. 7). Noko av det som var avgjerande for korleis lærarane underviste i seksualitet var læraren si kompetanse, nemleg kva syn dei har på seksualitetsundervising og kva erfaring dei sit med. Dei lærarane som hadde mindre fagorientering, viste seg å prioritere temaet i stor grad når tematikken dukka opp naturleg i klasserommet, anten det var ein situasjon dei måtte ta tak i eller spontane spørsmål frå elevane. I hovudsak brukar lærarane digitale ressursar. Dei lærebøkene dei har tilgjengelege på skulen er frå LK06. Dei brukar digitale ressursar fordi disse har oppdatert informasjon, og ein kan variere mellom ulike ressursar i større grad, gjennom tekst, bilete, video, individuelle og elevaktive oppgåver. Dei lærarane som er sterkare fagorientert hadde meir fokus på bolkundervisinga og kva tema læreplanen legg vekt på. Ein faktor her er òg at den eldre læreboka har vore styrande for kva tematikk som blir lagt vekt på. Sjølv om den eldre læreboka ikkje blir aktivt brukt i klasserommet, har den bana veg for når og kva tema som dukkar opp i seksualitetsundervisinga. This master’s thesis is a qualitative study with in-depth interviews with five different teachers; four science teachers and one social studies teacher. The thesis research question is: What affects sexuality education in upper primary school? To answer this question, there are three research questions: What understanding do teachers have of sexuality education, what guides the teachers’ didactic choices, and lastly, how do the teachers use different learning resources? The analysis method consists of the first step in the collective qualitative (Eggebø, 2020), and before continuing with a thematic analysis by Braun og Clarke (2022). From the data material, it became apparent that teachers have different ways of teaching. All of the teachers have a student-centered orientation but with varying degrees of subject orientation. Some are more concerned with what the subject and the curriculum focus on, while others view different elements are more important. Through this focus on the curriculum, it emerged that some of the teachers use block teaching. Emphasis is then placed on puberty in the 5th grade and either nothing or sexual behavior in the 7th grade. The holistic approach provides the background for the discussion. I have observed how the teachers approach sexuality education, compared to what the holistic approach emphasizes. The holistic approach emphasizes a holistic focus that should embrace all aspects related to sexuality, gender and the body, to give children the best possible life (Støle-Nilsen, 2022, s. 18). The new curriculum, Fagfornyinga, aims for a more holistic approach as well as interdisciplinary collaboration on sexuality education. This is also the focus of the «Snakk om det!»-strategy from the government, which focused on «a comprehensive effort to strengthen the entire population’s sexual health» (Helse- og omsorgsdepartementet, 2016, s. 7). A crucial part of how teachers taught sexuality was the teacher’s competence. This is due to how they regard sexuality education and what experiences they have with the matter. The teachers who were less subject-oriented prioritized the sexuality education topic to a larger extent when the topic was brought up naturally in the classroom, whether it was a situation they had to deal with or spontaneous questions from the students. For the most part, the teachers used digital resources. The textbooks that are available at their schools are from earlier curricula. Digital resources are thus used due to the updated information, which further allows for a greater variation in teaching. Text, images, video, as well as both individual and student-active tasks are available through digital resources. The teachers who are more subject-oriented had a larger focus on block teaching, and which topics the curriculum emphasized. A factor here is also that the older textbooks have been crucial for which topics are emphasized. Although the older textbooks are not actively used in the classroom, it has paved the way for when, and which, topic appears in sexuality education.

Masteroppgave i klinisk sykepleie, Høgskulen på Vestlandet, campus Bergen Bakgrunn: Infeksjonsforebygging er en sentral oppgave for operasjonssykepleieren. Fordi pasienten er svært utsatt og sårbar, kreves det hygienisk miljø med et langt høyere standard enn ellers i helsevesenet. Fra praksis i vår operasjonsutdanning erfarte vi at det kirurgiske teamets holdninger til de hygieniske retningslinjer som omhandler påkledning inn i operasjonsstuen var svært ulike, noe som har fått oss til å undre oss over om alle i det kirurgiske team kjenner til de strenge hygieniske standardene som gjelder på operasjonsavdelingen. Dette har motivert oss til å få et større innblikk i det kirurgiske teamets holdninger til hygieneregimet på operasjonsstuen. Hensikt: Hensikten med studien er å undersøke hvilke holdninger det kirurgiske team har til de strenge hygieniske retningslinjene i operasjonsstuen, med spesielt fokus på påkledning før man går inn i operasjonsavdelingen. Vi er interessert i å undersøke om ringer, armbåndsur, armbånd, smykker og øredobber fjernes, hvilken hodebekledning som benyttes, om riktig bruk av skotøy brukes og om den grønne operasjonsuniformen faktisk blir brukt på riktig måte. Metode: Metoden er kvalitativ med en induktiv tilnærming, designet er det kvalitative forsknings intervjue. Det ble gjennomført 6 intervjuer med operasjonssykepleiere på et sykehus i Vest-Norge. Systematisk tekstkondensering ble benyttet som analyse metode. Resultat: Resultatene viser at det er både høy standard og strenge retningslinjer ved ortopediske inngrep, men standarden blir til tross for det ikke opprettholdt hos enkelte av teammedlemmene. Det kommer også fram at ut ifra en operasjonssykepleiers perspektiv er det overraskende gode og positive holdninger til de hygieniske retningslinjene i ortopediske avdelinger. Konklusjon: Standarden er god, men det vil alltid være forbedringspotensialer selv hos de aller beste. Det må også være slik at alle har ansvar for å gi beskjed når det oppstår brudd på retningslinjene. Men viktigst av alt er det ledernes ansvar Implementeringen av endringer i operasjonsavdelingen med hensyn til overholdelse av de hygieniske retningslinjene hviler hovedsakelig på nettopp lederen, da det er deres ansvar å sikre at slike endringer finner sted. Ledernes rolle innebærer å innføre og sikre gjennomføringen av tiltak som fremmer etterlevelsen av de hygieniske retningslinjene på operasjonsavdelingen. We propose a deep-learning framework to detect and categorize oil spills in synthetic aperture radar (SAR) images at a large scale. Through a carefully designed neural network model for image segmentation trained on an extensive dataset, we obtain state-of-the-art performance in oil spill detection, achieving results that are comparable to results produced by human operators. We also introduce a classification task, which is novel in the context of oil spill detection in SAR. Specifically, after being detected, each oil spill is also classified according to different categories of its shape and texture characteristics. The classification results provide valuable insights for improving the design of services for oil spill monitoring by world-leading providers. Finally, we present our operational pipeline and a visualization tool for large-scale data, which allows detection and analysis of the historical occurrence of oil spills worldwide.

Some evolutionary psychologists have hypothesized that animals have priority in human attention. That is, they should be detected and selected more efficiently than other types of objects, especially man-made ones. Such a priority mechanism should automatically deploy more attentional resources and dynamic monitoring toward animal stimuli than nonanimals. Consequently, we postulated that variations of the multiple object or identity tracking and multiple event monitoring tasks should be particularly suitable paradigms for addressing the animate monitoring hypothesis, given their dynamic properties and dependency on divided attention. We used images of animals and artifacts and found neither a substantial sign of improvement in tracking the positions associated with animal stimuli nor a significant distracting effect of animals. We also failed to observe a significant prioritization in orders of response for positions associated with animals. While we observed an advantage for animals in event monitoring, this appeared to be dependent on properties of the task, as confirmed in further experiments. Moreover, we observed a small but inconclusive advantage for animals in identity accuracy. Thus, under certain conditions, some bias toward animals could be observed, but the evidence was weak and inconclusive. To conclude, effect sizes were generally small and not conclusively in favor of the expected attentional bias for animals. We found moderate to strong evidence that images of animals do not improve positional tracking, do not act as more effective distractors, are not selected prior to artifacts in the response phase, and are not easier to monitor for changes in size.

Background: Brain functional connectivity (FC) in multiple sclerosis (MS) is abnormal compared to healthy controls (HCs). More longitudinal studies in MS are needed to evaluate whether FC stability is clinically relevant. Objective: To compare functional magnetic resonance imaging (fMRI)-based FC between MS and HC, and to determine the relationship between longitudinal FC changes and structural brain damage, cognitive performance and physical disability. Methods: T1-weighted MPRAGE and resting-state fMRI (1.5T) were acquired from 70 relapsing-remitting MS patients and 94 matched HC at baseline (mean months since diagnosis 14.0 ± 11) and from 60 MS patients after 5 years. Independent component analysis and network modelling were used to measure longitudinal FC stability and cross-sectional comparisons with HC. Linear mixed models, adjusted for age and sex, were used to calculate correlations. Results: At baseline, patients with MS showed FC abnormalities both within networks and in single connections compared to HC. Longitudinal analyses revealed functional stability and no significant relationships with clinical disability, cognitive performance, lesion or brain volume. Conclusion: FC abnormalities occur already at the first decade of MS, yet we found no relevant clinical correlations for these network deviations. Future large-scale longitudinal fMRI studies across a range of MS subtypes and outcomes are required.

Respiratory syncytial virus (RSV) is a leading cause of serious lower respiratory tract disease in young children and older adults throughout the world. Prevention of severe RSV disease through active immunization is optimal but no RSV vaccine has been licensed so far. Immune mechanisms of protection against RSV infection in humans have not been fully established, thus a comprehensive characterization of virus-specific immune responses in a relevant animal model will be beneficial in defining correlates of protection. In this study, we infected juvenile naive AGMs with RSV A2 strain and longitudinally assessed virus-specific humoral and cellular immune responses in both peripheral blood and the respiratory tract. RSV viral loads at nasopharyngeal surfaces and in the lung peaked at around day 5 following infection, and then largely resolved by day 10. Low levels of neutralizing antibody titers were detected in serum, with similar kinetics as RSV fusion (F) protein-binding IgG antibodies. RSV infection induced CD8+, but very little CD4+, T lymphocyte responses in peripheral blood. Virus-specific CD8+ T cell frequencies were ~10 fold higher in bronchoaveolar lavage (BAL) compared to peripheral blood and exhibited effector memory (CD95+CD28-) / tissue resident memory (CD69+CD103+) T (TRM) cell phenotypes. The kinetics of virus-specific CD8+ T cells emerging in peripheral blood and BAL correlated with declining viral titers, suggesting that virus-specific cellular responses contribute to the clearance of RSV infection. RSV-experienced AGMs were protected from subsequent exposure to RSV infection. Additional studies are underway to understand protective correlates in these seropositive monkeys.

- Abstract
Mechanisms underpinning neurotypical age-related variations in cortical thickness in the human brain remain insufficiently specified. Here we used cell-specific marker genes, followed by gene ontology and enrichment analyses, to quantify the association between gene-expression levels and inter-regional age-related variations in neurotypical cortical thinning using multicohort neuroimaging data from 14,248 individuals ages 4-89 years. We found that early-life (60 years) were associated with distinct patterns of association between inter-regional profiles of cortical thickness and expression profiles of markers genes for CA1 and S1 pyramidal cells, astrocytes, and microglia. Gene ontology and enrichment analyses indicated each of the three life-stages was associated with different biological processes and cellular components; these related to synaptic modeling in early life, neurotransmission in mid-life, and neurodegeneration in late-life. These findings provide mechanistic insights on age-related cortical thinning during typical development and ageing.

However, the normative studies available are constrained by small sample sizes, restricted age coverage and significant methodological variability. These limitations introduce inconsistencies and may obscure or distort the lifespan trajectories of brain morphometry. In response, we capitalized on the resources of the Enhancing Neuroimaging Genetics through Meta-Analysis (ENIGMA) Consortium to examine age-related trajectories inferred from cross-sectional measures of the ventricles, the basal ganglia (caudate, putamen, pallidum, and nucleus accumbens), the thalamus, hippocampus and amygdala using magnetic resonance imaging data obtained from 18,605 individuals aged 3-90 years. All subcortical structure volumes were at their maximum value early in life. The volume of the basal ganglia showed a monotonic negative association with age thereafter; there was no significant association between age and the volumes of the thalamus, amygdala and the hippocampus (with some degree of decline in thalamus) until the sixth decade of life after which they also showed a steep negative association with age. The lateral ventricles showed continuous enlargement throughout the lifespan. Age was positively associated with inter-individual variability in the hippocampus and amygdala and the lateral ventricles. These results were robust to potential confounders and could be used to examine the functional significance of deviations from typical age-related morphometric patterns.

Contains fulltext : 245396.pdf (Publisher’s version ) (Open Access), Delineating the association of age and cortical thickness in healthy individuals is critical given the association of cortical thickness with cognition and behavior. Previous research has shown that robust estimates of the association between age and brain morphometry require large-scale studies. In response, we used cross-sectional data from 17,075 individuals aged 3-90 years from the Enhancing Neuroimaging Genetics through Meta-Analysis (ENIGMA) Consortium to infer age-related changes in cortical thickness. We used fractional polynomial (FP) regression to quantify the association between age and cortical thickness, and we computed normalized growth centiles using the parametric Lambda, Mu, and Sigma method. Interindividual variability was estimated using meta-analysis and one-way analysis of variance. For most regions, their highest cortical thickness value was observed in childhood. Age and cortical thickness showed a negative association; the slope was steeper up to the third decade of life and more gradual thereafter; notable exceptions to this general pattern were entorhinal, temporopolar, and anterior cingulate cortices. Interindividual variability was largest in temporal and frontal regions across the lifespan. Age and its FP combinations explained up to 59% variance in cortical thickness. These results may form the basis of further investigation on normative deviation in cortical thickness and its significance for behavioral and cognitive outcomes.

The Enhancing NeuroImaging Genetics through Meta-Analysis copy number variant (ENIGMA-CNV) and 22q11.2 Deletion Syndrome Working Groups (22q-ENIGMA WGs) were created to gain insight into the involvement of genetic factors in human brain development and related cognitive, psychiatric and behavioral manifestations. To that end, the ENIGMA-CNV WG has collated CNV and magnetic resonance imaging (MRI) data from ~49,000 individuals across 38 global research sites, yielding one of the largest studies to date on the effects of CNVs on brain structures in the general population. The 22q-ENIGMA WG includes 12 international research centers that assessed over 533 individuals with a confirmed 22q11.2 deletion syndrome, 40 with 22q11.2 duplications, and 333 typically developing controls, creating the largest-ever 22q11.2 CNV neuroimaging data set. In this review, we outline the ENIGMA infrastructure and procedures for multi-site analysis of CNVs and MRI data. So far, ENIGMA has identified effects of the 22q11.2, 16p11.2 distal, 15q11.2, and 1q21.1 distal CNVs on subcortical and cortical brain structures. Each CNV is associated with differences in cognitive, neurodevelopmental and neuropsychiatric traits, with characteristic patterns of brain structural abnormalities. Evidence of gene-dosage effects on distinct brain regions also emerged, providing further insight into genotype–phenotype relationships. Taken together, these results offer a more comprehensive picture of molecular mechanisms involved in typical and atypical brain development. This “genotype-first” approach also contributes to our understanding of the etiopathogenesis of brain disorders. Finally, we outline future directions to better understand effects of CNVs on brain structure and behavior.

- Abstract
Human brain structure changes throughout the lifespan. Altered brain growth or rates of decline are implicated in a vast range of psychiatric, developmental and neurodegenerative diseases. In this study, we identified common genetic variants that affect rates of brain growth or atrophy in what is, to our knowledge, the first genome-wide association meta-analysis of changes in brain morphology across the lifespan. Longitudinal magnetic resonance imaging data from 15,640 individuals were used to compute rates of change for 15 brain structures. The most robustly identified genes GPR139, DACH1 and APOE are associated with metabolic processes. We demonstrate global genetic overlap with depression, schizophrenia, cognitive functioning, insomnia, height, body mass index and smoking. Gene set findings implicate both early brain development and neurodegenerative processes in the rates of brain changes. Identifying variants involved in structural brain changes may help to determine biological pathways underlying optimal and dysfunctional brain development and aging.

Contains fulltext : 251344.pdf (Publisher’s version ) (Open Access), The discrepancy between chronological age and the apparent age of the brain based on neuroimaging data - the brain age delta - has emerged as a reliable marker of brain health. With an increasing wealth of data, approaches to tackle heterogeneity in data acquisition are vital. To this end, we compiled raw structural magnetic resonance images into one of the largest and most diverse datasets assembled (n=53542), and trained convolutional neural networks (CNNs) to predict age. We achieved state-of-the-art performance on unseen data from unknown scanners (n=2553), and showed that higher brain age delta is associated with diabetes, alcohol intake and smoking. Using transfer learning, the intermediate representations learned by our model complemented and partly outperformed brain age delta in predicting common brain disorders. Our work shows we can achieve generalizable and biologically plausible brain age predictions using CNNs trained on heterogeneous datasets, and transfer them to clinical use cases.

- Abstract
Delineating the association of age and cortical thickness in healthy individuals is critical given the association of cortical thickness with cognition and behavior. Previous research has shown that robust estimates of the association between age and brain morphometry require large-scale studies. In response, we used cross-sectional data from 17,075 individuals aged 3–90 years from the Enhancing Neuroimaging Genetics through Meta-Analysis (ENIGMA) Consortium to infer age-related changes in cortical thickness. We used fractional polynomial (FP) regression to quantify the association between age and cortical thickness, and we computed normalized growth centiles using the parametric Lambda, Mu, and Sigma method. Interindividual variability was estimated using meta-analysis and one-way analysis of variance. For most regions, their highest cortical thickness value was observed in childhood. Age and cortical thickness showed a negative association; the slope was steeper up to the third decade of life and more gradual thereafter; notable exceptions to this general pattern were entorhinal, temporopolar, and anterior cingulate cortices. Interindividual variability was largest in temporal and frontal regions across the lifespan. Age and its FP combinations explained up to 59% variance in cortical thickness. These results may form the basis of further investigation on normative deviation in cortical thickness and its significance for behavioral and cognitive outcomes.

- Abstract
Age has a major effect on brain volume. However, the normative studies available are constrained by small sample sizes, restricted age coverage and significant methodological variability. These limitations introduce inconsistencies and may obscure or distort the lifespan trajectories of brain morphometry. In response, we capitalized on the resources of the Enhancing Neuroimaging Genetics through Meta-Analysis (ENIGMA) Consortium to examine age-related trajectories inferred from cross-sectional measures of the ventricles, the basal ganglia (caudate, putamen, pallidum, and nucleus accumbens), the thalamus, hippocampus and amygdala using magnetic resonance imaging data obtained from 18,605 individuals aged 3-90 years. All subcortical structure volumes were at their maximum value early in life. The volume of the basal ganglia showed a monotonic negative association with age thereafter; there was no significant association between age and the volumes of the thalamus, amygdala and the hippocampus (with some degree of decline in thalamus) until the sixth decade of life after which they also showed a steep negative association with age. The lateral ventricles showed continuous enlargement throughout the lifespan. Age was positively associated with inter-individual variability in the hippocampus and amygdala and the lateral ventricles. These results were robust to potential confounders and could be used to examine the functional significance of deviations from typical age-related morphometric patterns.

- Abstract
Delineating the association of age and cortical thickness in healthy individuals is critical given the association of cortical thickness with cognition and behavior. Previous research has shown that robust estimates of the association between age and brain morphometry require large-scale studies. In response, we used cross-sectional data from 17,075 individuals aged 3–90 years from the Enhancing Neuroimaging Genetics through Meta-Analysis (ENIGMA) Consortium to infer age-related changes in cortical thickness. We used fractional polynomial (FP) regression to quantify the association between age and cortical thickness, and we computed normalized growth centiles using the parametric Lambda, Mu, and Sigma method. Interindividual variability was estimated using meta-analysis and one-way analysis of variance. For most regions, their highest cortical thickness value was observed in childhood. Age and cortical thickness showed a negative association; the slope was steeper up to the third decade of life and more gradual thereafter; notable exceptions to this general pattern were entorhinal, temporopolar, and anterior cingulate cortices. Interindividual variability was largest in temporal and frontal regions across the lifespan. Age and its FP combinations explained up to 59% variance in cortical thickness. These results may form the basis of further investigation on normative deviation in cortical thickness and its significance for behavioral and cognitive outcomes.

Age has a major effect on brain volume. However, the normative studies available are constrained by small sample sizes, restricted age coverage and significant methodological variability. These limitations introduce inconsistencies and may obscure or distort the lifespan trajectories of brain morphometry. In response, we capitalized on the resources of the Enhancing Neuroimaging Genetics through Meta-Analysis (ENIGMA) Consortium to examine age-related trajectories inferred from cross-sectional measures of the ventricles, the basal ganglia (caudate, putamen, pallidum, and nucleus accumbens), the thalamus, hippocampus and amygdala using magnetic resonance imaging data obtained from 18,605 individuals aged 3–90 years. All subcortical structure volumes were at their maximum value early in life. The volume of the basal ganglia showed a monotonic negative association with age thereafter; there was no significant association between age and the volumes of the thalamus, amygdala and the hippocampus (with some degree of decline in thalamus) until the sixth decade of life after which they also showed a steep negative association with age. The lateral ventricles showed continuous enlargement throughout the lifespan. Age was positively associated with inter-individual variability in the hippocampus and amygdala and the lateral ventricles. These results were robust to potential confounders and could be used to examine the functional significance of deviations from typical age-related morphometric patterns.
