104. Temporal expression profiles of organic anion transport proteins in placenta and fetal liver of the rat

Abstract

Physiological cholestasis linked to immature hepatobiliary transport systems for organic anions occurs in rat and human neonates. In utero, the placenta facilitates vectorial transfer of certain fetal-derived solutes to the maternal circulation for elimination. We compared the ontogenesis of organic anion transporters in the placenta and the fetal liver of the rat to assess their relative abundance throughout gestation and to determine whether the placenta compensates for the late maturation of transporters in the developing liver. The mRNA of members of the organic anion transporting polypeptide (Oatp) superfamily, the multidrug resistance protein (Mrp) family, one organic anion transporter (OAT), and the bile acid carriers [Na.sup.+]-taurocholate cotransporting polypeptide (Ntcp) and bile salt export: pump (Bsep) was quantified by real-time PCR. The most abundant placental transporters were Oatp4a 1, whose mRNA increased 10-fold during gestation, and Mrp 1. Mrp1 immunolocalized predominantly to epithelial cells of the endoplacental yolk sac, suggesting an excretory role that sequesters fetal-derived solutes in the yolk sac cavity, and faintly to the basal syncytiotrophoblast surface. The mRNA levels of Oatp2bl, Mrp3, and Bsep in the placenta exceeded those in the fetal liver until day 20 of gestation, suggesting that the fetus relies on placental clearance of substrates when expression in the developing liver is low. Mrp3 immunolocalized to the epithelium of the endoplacental yolk sac and less abundantly in the labyrinth zone and endothelium of the maternal arteries. Abstract

For $(n+1)$-webs by curves in an ambiant $n$-dimensional manifold, we first define a generalization of the well known Blaschke curvature of the dimension two, which vanishes iff the web has the maximum possible rank which is one. But, contrary to the dimension two where all 3-webs of rank one are locally isomorphic, we prove that there are infinitely many classes of isomorphism for germs of 4-webs by curves of rank one in the dimension three: we provide a procedure for building all of them, and give examples of invariants of these classes allowing in particular to distinguish the so-called quadrilateral webs among them.

Abstract

We describe sclerochronological analyses of tadpole codling (Salilota australis) vertebrae as a proxy for the seasonal timing of fishing activities at one Middle Holocene (6500 BP) and three Late–Middle Holocene (3500–2500 years BP) archaeological sites located in Seno Otway and in the central zone of the Strait of Magellan, Chile. These data provide an approximation of the population characteristics of the species in the past by estimating the relationship between size and age of the archaeological fish and comparing their growth rate to those of modern specimens captured monthly over one year. The results showed that the size at age was significantly larger in archaeological samples than in the modern reference collection. Our results indicate that fishing activities took place throughout the year at the Late Holocene sites, with a special emphasis on the cold season, while the Middle Holocene site showed a tendency to catch tadpole codling during the warm season. Abstract

BACKGROUND AND AIMS: Robust data on hepatocellular carcinoma (HCC) incidence among HIV/hepatitis B virus (HBV)-coinfected individuals on antiretroviral therapy (ART) are needed to inform HCC screening strategies. We aimed to evaluate the incidence and risk factors of HCC among HIV/HBV-coinfected individuals on tenofovir disoproxil fumarate (TDF)-containing ART in a large multi-cohort study. METHODS: We included all HIV-infected adults with a positive hepatitis B surface antigen followed in one of four prospective European cohorts. The primary outcome was the occurrence of HCC. Demographic and clinical information was retrieved from routinely collected data, and liver cirrhosis was defined according to results from liver biopsy or non-invasive measurements. Multivariable Poisson regression was used to assess HCC risk factors. RESULTS: 3,625 HIV/HBV-coinfected patients, of whom 72% have initiated TDF-containing ART were included. Over 32,673 patient-years (py), 60 individuals (1.7%) developed an HCC. The incidence of HCC remained stable over time among individuals on TDF, whereas it increased steadily among those not on TDF. Among individuals on TDF, the incidence of HCC was 5.9 per 1,000 py (95% confidence interval [CI] 3.60-9.10) in cirrhotics and 1.17 per 1,000 py (0.56-2.14) among non-cirrhotics. Age at initiation of TDF (adjusted incidence rate ratio per 10 years increase: 2.2, 95% CI 1.6-3.0) and the presence of liver cirrhosis (4.5, 2.3-8.9) were predictors of HCC. Among non-cirrhotic individuals, the incidence of HCC was above the commonly used screening threshold of 2 cases per 1,000 py only in patients aged >46 years at TDF initiation. CONCLUSIONS: Whereas the incidence of HCC was high in cirrhotic HIV/HBV-coinfected individuals, it remained below the HCC screening threshold in patients initiating TDF without cirrhosis at an age
