Objective To investigate the current status of nurses' practice environment in a grade-A tertiary integrated traditional Chinese and Western medicine hospital. Methods Use the "Nurse Practice Environment Evaluation Scale" to conduct a questionnaire survey on 520 nurses in the hospital. Results The average score was 77. 92(77. 92±10. 81) points. The item with highest score was ‘systematic training program for newly recruited nurses’, and the item with lowest score was ‘nurse participation in the decision making of hospital affairs ’. Conclusion Hospital managers need to provide nurses with more learning opportunities for communication and a platform for participating in hospital affairs decision-making, fully mobilize the enthusiasm of nurses to work, and further build a healthy nurse practice environment. In order to achieve a better balance between speed and accuracy in the process of clothing landmark detection, based on the human pose estimation network YOLOv8s-Pose, a T-shirt landmark detection method named YOLO-T-Shirt is proposed, which utilizes a cascade architecture and fused geometric information. Firstly, inspired by the CFNet architecture, the cascade fusion network design architecture is introduced into YOLOv8s-Pose, with a redesign of the original feature extraction and feature fusion architecture to better integrate multi-scale features, so as to have good robustness to changes in clothing size and shape. Secondly, the native OKS loss function is optimized, and an efficient landmark similarity loss function EOKS (Efficient Object Keypoint Similarity) that integrates integrating geometric information of area, width, height and distance of the center point of the frame is proposed to improve the accuracy of landmark detection. The proposed method achieves a prediction accuracy of 0.760 in the landmark detection task of the T-shirt category in the DeepFashion2 dataset, which is close to the accuracy of 0.765 of the current clothing landmark detection algorithm with the highest accuracy, while the inference speed is more than 9 times faster. In the paper, the possibility of two complete conversions is investigated, one is from the two‐photon hyperentangled Knill–Laflamme–Milburn (KLM) state to hyperentangled Bell states, the other is from three‐photon KLM state to the hyperentangled Greenberger–Horne–Zeilinger states, for photonic systems hyper‐encoded in polarization and spatial degrees of freedom assisted by error‐heralded quantum units (EHQUs). The system inhomogeneity and/or the imperfect photon scattering often restrict the performance of effective conversions of two types of hyperentangled states, and result in the reductions of the fidelity and the efficiency in tangible quantum information technologies. The polarization EHQU and spatial EHQU are two critical tools for actualizing two hyperentangled conversions in a heralded way, as the errors stemming from the system inhomogeneity and practically imperfect scattering are converted into efficacious responses of the detectors in our protocols. Thus, the fidelities of these conversion cases keep unchanged and have close to unity fidelities, which deepen the understanding of the properties of hyperentanglement. [ABSTRACT FROM AUTHOR]

Single-cell DNA sequencing (scDNA-seq) has been an effective means to unscramble intra-tumor heterogeneity, while joint inference of tumor clones and their respective copy number profiles remains a challenging task due to the noisy nature of scDNA-seq data. We introduce a new bioinformatics method called CoT for deciphering clonal copy number substructure. The backbone of CoT is a Copy number Transformer autoencoder that leverages multi-head attention mechanism to explore correlations between different genomic regions, and thus capture global features to create latent embeddings for the cells. CoT makes it convenient to first infer cell subpopulations based on the learned embeddings, and then estimate single-cell copy numbers through joint analysis of read counts data for the cells belonging to the same cluster. This exploitation of clonal substructure information in copy number analysis helps to alleviate the effect of read counts non-uniformity, and yield robust estimations of the tumor copy numbers. Performance evaluation on synthetic and real datasets showcases that CoT outperforms the state of the arts, and is highly useful for deciphering clonal copy number substructure. Background and AimsEndothelial senescence is an important risk factor leading to atherosclerosis. The mechanism of quercetin against endothelial senescence is worth exploring.MethodsQuercetin (20 mg/kg/d) was administered to ApoE-/- mice intragastrically to evaluate the effectiveness of quercetin on atherosclerotic lesion in vivo. In vitro, human aortic endothelial cells (HAECs) were used to assess the effect of quercetin on cellular senescence induced by oxidized low-density lipoprotein (ox-LDL). Transcriptome microarray and quantitative RT-PCR was conducted to study the pharmacological targets of quercetin.ResultsApoE-/- mice demonstrated obvious lipid deposition in arterial lumina, high level of serum sIcam-1 and IL-6, and high density of Vcam-1 and lower density of Sirt1 in aorta. Quercetin administration decreased lipid deposition in arterial lumina, serum sIcam-1, and IL-6 and Vcam-1 in aorta, while increased the density of Sirt1 in aorta of ApoE-/- mice. In vitro, quercetin (0.3, 1, or 3 μmol/L) decreased the expression of senescence-associated β-galactosidase and improved cell morphology of HAECs. And quercetin decreased the cellular apoptosis and increased mitochondrial membrane potential (ΔΨm) in dose-dependent manner, and decreased ROS generation simultaneously. Transcriptome microarray suggested 254 differentially expressed (DE) mRNAs (110 mRNAs were upregulated and 144 mRNAs were downregulated) in HAECs after quercetin treatment (fold change > 1.5, P < 0 .05, Que vs Ox-LDL). GO and KEGG analysis indicated nitrogen metabolism, ECM-receptor interaction, complement, and coagulation cascades, p53 and mTOR signaling pathway were involved in the pharmacological mechanisms of quercetin against ox-LDL.ConclusionsQuercetin alleviated atherosclerotic lesion both in vivo and in vitro.
