Many bacterial pathogens use a type III secretion system (T3SS) to inject effector proteins into host cells. Here, Wimmi et al. show that the external pH regulates the assembly of T3SS cytosolic components in intestinal pathogens, thus preventing T3SS activity in the stomach and allowing T3SS reactivation in the intestine.

Abstract Background The accumulation of intracellular fat depots is a polygenic trait. Therefore, the extent of lipid storage in the individuals of a species covers a broad range and is determined by many genetic factors. Quantitative trait loci analysis can be used to identify those genetic differences between two strains of the same species that are responsible for the differences in a given phenotype. We used this method and complementary approaches to identify genes in the yeast Saccharomyces cerevisiae that are involved in neutral lipid storage. Results We selected two yeast strains, the laboratory strain BY4741 and the wine yeast AWRI1631, with a more than two-fold difference in neutral lipid content. After crossing, sporulation and germination, we used fluorescence activated cell sorting to isolate a subpopulation of cells with the highest neutral lipid content from the pool of segregants. Whole genome sequencing of this subpopulation and of the unsorted pool of segregants implicated several loci that are involved in lipid accumulation. Three of the identified genes, PIG1, PHO23 and RML2, were investigated in more detail. Deletions of these genes and the exchange of the alleles between the two parental strains confirmed that the encoded proteins contribute to neutral lipid storage in S. cerevisiae and that PIG1, PHO23 and RML2 are the major causative genes. Backcrossing of one of the segregants with the parental strains for seven generations revealed additional regions in the genomes of both strains with potential causative genes for the high lipid accumulation phenotype. Conclusions We identified several genes that contribute to the phenotype of lipid accumulation in an allele-specific manner. Surprisingly, no allelic variations of genes with known functions in lipid metabolism were found, indicating that the level of storage lipid accumulation is determined by many cellular processes that are not directly related to lipid metabolism.

Early number skills represent critical milestones in children's cognitive development and are shaped over years of interacting with quantities and numerals in various contexts. Several connectionist computational models have attempted to emulate how certain number concepts may be learned, represented, and processed in the brain. However, these models mainly used highly simplified inputs and focused on limited tasks. We expand on previous work in two directions: First, we train a model end‐to‐end on video demonstrations in a synthetic environment with multimodal visual and language inputs. Second, we use a more holistic dataset of 35 tasks, covering enumeration, set comparisons, symbolic digits, and seriation. The order in which the model acquires tasks reflects input length and variability, and the resulting trajectories mostly fit with findings from educational psychology. The trained model also displays symbolic and non‐symbolic size and distance effects. Using techniques from interpretability research, we investigate how our attention‐based model integrates cross‐modal representations and binds them into context‐specific associative networks to solve different tasks. We compare models trained with and without symbolic inputs and find that the purely non‐symbolic model employs more processing‐intensive strategies to determine set size. [ABSTRACT FROM AUTHOR]

Today, new therapies (e.g. extracorporeal membrane oxygenation (ECMO), protective ventilation methods and new forms of patient positioning) are available and applied regularly. What impact on the patient’s outcome does the thoracic trauma have today? Methods Prospective data collection of multiple injured patients in a level-I trauma center was performed between 2008 and 2014. Evaluationsmethoden für E-Learning, Forschungsmethoden zur Mediennutzung und zur Analyse von Medienkulturen sind Gegenstand dieses Jahrbuchs Medienpädagogik. Dabei reicht das Spektrum der Medien von Fernsehen über Video bis zum Internet. Die Auswahl dieses Themenschwerpunkts kommt dabei nicht von ungefähr. Zum einen hat wissenschaftliche Medienforschung bereits eine lange Tradition und ist in vielen Disziplinen etabliert. Forschungsmethoden lieferten der medienpädagogischen Praxis empirisch fundierte Ergebnisse und Instrumente. Forschungsdesigns und Methodologien wurden entwickelt, um Mediennutzungsverhalten und Medienwirkungen zu erklären, Evaluationsmethoden eingesetzt, um Lernprozesse zu beurteilen. Zum anderen gewinnen Qualitätssicherung und Leistungsmessungen im Bildungssystem generell immer mehr an Bedeutung. In den Ergebnissen aktueller Evaluationen zeigen sich Bestrebungen nach Qualität von Bildungs- und Lernprozessen. Empirische Forschung kommt die Aufgabe zu medienpädagogische Wirkungsgrade und Zielerreichung zu überprüfen und zu kontrollieren. Quantitative Forschungsmethoden ermöglichen es Mediennutzungsverhalten bestimmter Zielgruppen in Wechselwirkung mit Programmstrukturen und -inhalten zu ermitteln. Nutzungsmuster von Medienangeboten – vor allem bei Kindern und Jugendlichen – sind auch bei der Entwicklung medienpädagogischer Ansätze wichtig. Qualitative Medienforschung dient als Planungsinstrument für medienpädagogische Konzeptionen sowie für medienpädagogische Praxis und hilft bei der Entwicklung neuer, mehr als bisher an inhaltlichen Kriterien orientierter Angebote eingesetzt werden. Eine der zentralen Fragestellungen war und ist dabei, mit welchen kognitiven und ästhetischen Vermittlungsformen die Inhalte einer Sendung am besten kommuniziert werden können. Dies ist zugleich eine der Grundfragen im Hinblick auf den Medieneinsatz in pädagogisch orientierten Lernprozessen (vgl. Dichanz 1998).

Ein Jahrbuch, herausgegeben von einer wissenschaftlichen Vereinigung, ist nicht primär auf ein Thema zugeschnitten. Es will vor allem den Stand der erziehungswissenschaftlichen Disziplin der Medienpädagogik erkenn- und diskutierbar machen. Die zentralen Eckpfeiler in diesem Jahrbuch Medienpädagogik 4 sind Mediendidaktik, Mediennutzung und Medienhandeln. Ein Teil der Texte speist sich aus der Herbsttagung der Kommission Medienpädagogik der Deutschen Gesellschaft für Erziehungswissenschaft von 2002 an der Humboldt Universität Berlin und der Frühjahrstagung 2003 an der Universität Erlangen-Nürnberg. Deren Themenschwerpunkte waren in Berlin „Empirische Ansätze und Theoriefragen zu neuen Medien” und in Nürnberg die Auseinandersetzung mit „medialen Räumen für Bildung”. Der andere Teil der Beiträge kommt ohne vorausgehende Diskussion in der Kommission Medienpädagogik aus den Arbeitsfeldern der Kolleginnen und Kollegen. Diese Beiträge beschäftigen sich mit Zukunftsfeldern der Medienpädagogik, diskutieren den Umgang mit digitalen Medien aus sozialwissenschaftlicher Perspektive oder stellen neue empirische Ansätze vor.
