Glycolipids are biosurfactants with a wide range of structural diversity. They are biodegradable, based on renewables, ecocompatible and exhibit high surface activity. Still, studies comparing glycolipids and conventional surfactants in terms of interfacial properties and foaming performance are lacking. Here, we compared interfacial and foaming properties of microbial and enzymatically synthesized glycolipids to those of the widely-used, conventional surfactant sodium dodecyl sulfate (SDS). The enzymatically produced sorbose monodecanoate, as well as microbially produced di-rhamno-di-lipids exhibited high foam stabilizing properties, similar to those of SDS. However, sophorolipid and mono-rhamno-di-lipids did not produce metastable foams. An appropriate selection of head and tail groups depending on the application of interest is therefore necessary. Then, glycolipids can serve as an ecofriendly and efficient alternative to petroleum-based surfactants, even at substantially lower concentrations than e.g. SDS. Moreover, the influence of three foaming gases on the foaming properties of the glycolipids was evaluated. Slightly higher foam stability and lower coarsening rates were determined for sorbose monodecanoate when using nitrogen as the foaming gas instead of air. Foams generated with carbon dioxide were not metastable, no matter which surfactant was used.

In angiosperms, the α/β hydrolase DWARF14 (D14), along with the F-box protein MORE AXILLARY GROWTH2 (MAX2), perceives strigolactones (SL) to regulate developmental processes. The key SL biosynthetic enzyme CAROTENOID CLEAVAGE DIOXYGENASE8 (CCD8) is present in the moss Physcomitrium patens, and PpCCD8-derived compounds regulate moss extension. The PpMAX2 homolog is not involved in the SL response, but 13 PpKAI2LIKE (PpKAI2L) genes homologous to the D14 ancestral paralog KARRIKIN INSENSITIVE2 (KAI2) encode candidate SL receptors. In Arabidopsis thaliana, AtKAI2 perceives karrikins and the elusive endogenous KAI2-Ligand (KL). Here, germination assays of the parasitic plant Phelipanche ramosa suggested that PpCCD8-derived compounds are likely noncanonical SLs. (+)-GR24 SL analog is a good mimic for PpCCD8-derived compounds in P. patens, while the effects of its enantiomer (−)-GR24, a KL mimic in angiosperms, are minimal. Interaction and binding assays of seven PpKAI2L proteins pointed to the stereoselectivity toward (−)-GR24 for a single clade of PpKAI2L (eu-KAI2). Enzyme assays highlighted the peculiar behavior of PpKAI2L-H. Phenotypic characterization of Ppkai2l mutants showed that eu-KAI2 genes are not involved in the perception of PpCCD8-derived compounds but act in a PpMAX2-dependent pathway. In contrast, mutations in PpKAI2L-G, and -J genes abolished the response to the (+)-GR24 enantiomer, suggesting that PpKAI2L-G, and -J proteins are receptors for moss SLs.

Strigolactones are a class of plant hormones involved in shoot branching, growth of symbiotic arbuscular mycorrhizal fungi, and germination of parasitic plant seeds. Assaying new molecules or compound exhibiting strigolactone-like activities is therefore important but unfortunately time-consuming and hard to implement because of the extremely low concentrations at which they are active. Seeds of parasite plants are natural integrator of these hormones since they can perceive molecule concentrations in the picomolar to nanomolar range stimulating their germination. Here we describe a simple and inexpensive method to evaluate the activity of these molecules by scoring the germination of parasitic plant seeds upon treatment with these molecules. Up to four molecules can be assayed from a single 96-well plate by this method. A comparison of SL-like bioactivities between molecules is done by determining the EC50 and the maximum percentage of germination.

In flowering plants, the α/β hydrolase DWARF14 (D14) perceives strigolactone (SL) hormones and interacts with the F-box protein MORE AXILLARY GROWTH2 (MAX2) to regulate developmental processes. Candidate receptors are among the 13PpKAI2LIKE-A to -Lgenes, homologous to the ancestralD14paralogKARRIKIN INSENSITIVE2 (KAI2).In Arabidopsis, AtKAI2 is the receptor for a still elusive endogenous KAI2-Ligand (KL). We show that inP. patens,among SL analogs, the (+)-GR24 enantiomer is a good mimic for PpCCD8-derived compounds, while the effects of the (-)-GR24 enantiomer, a KL mimic in flowering plants, are opposite. Interaction and binding assays of seven PpKAI2L proteins using pure enantiomers pinpoint at stereoselectivity towards (-)-GR24 for the (A-E) clade. Enzyme assays highlight strong hydrolytic activity of the PpKAI2L-H protein. Moss mutants for allPpKAI2Lgene subclades were obtained and tested for their response to both enantiomers. We show thatPpKAI2L-Ato-Egenes are not involved in PpCCD8-derived compound perception, but act in a PpMAX2-dependant pathway. In contrast, mutations inPpKAI2L-G, and-Jgenes abolish the response to (+)-GR24, suggesting that encoded proteins are receptors for PpCCD8-derived SLs.

Phelipanche ramosa is an obligate root-parasitic weed that threatens major crops in central Europe. In order to germinate, it must perceive various structurally divergent host-exuded signals, including isothiocyanates (ITCs) and strigolactones (SLs). However, the receptors involved are still uncharacterized. Here, we identify five putative SL receptors in P. ramosa and show that PrKAI2d3 is involved in the stimulation of seed germination. We demonstrate the high plasticity of PrKAI2d3, which allows it to interact with different chemicals, including ITCs. The SL perception mechanism of PrKAI2d3 is similar to that of endogenous SLs in non-parasitic plants. We provide evidence that PrKAI2d3 enzymatic activity confers hypersensitivity to SLs. Additionally, we demonstrate that methylbutenolide-OH binds PrKAI2d3 and stimulates P. ramosa germination with bioactivity comparable to that of ITCs. The formation of the haustorium in holoparasitic plants as demonstrated in Phelipanche ramosa is initiated through perception of a host-exudated cytokinin and a cytokinin signaling response., The heterotrophic lifestyle of parasitic plants relies on the development of the haustorium, a specific infectious organ required for attachment to host roots. While haustorium development is initiated upon chemodetection of host-derived molecules in hemiparasitic plants, the induction of haustorium formation remains largely unknown in holoparasitic species such as Phelipanche ramosa. This work demonstrates that the root exudates of the host plant Brassica napus contain allelochemicals displaying haustorium-inducing activity on P. ramosa germinating seeds, which increases the parasite aggressiveness. A de novo assembled transcriptome and microarray approach with P. ramosa during early haustorium formation upon treatment with B. napus root exudates allowed the identification of differentially expressed genes involved in hormone signaling. Bioassays using exogenous cytokinins and the specific cytokinin receptor inhibitor PI-55 showed that cytokinins induced haustorium formation and increased parasite aggressiveness. Root exudates triggered the expression of cytokinin-responsive genes during early haustorium development in germinated seeds, and bio-guided UPLC-ESI(+)-/MS/MS analysis showed that these exudates contain a cytokinin with dihydrozeatin characteristics. These results suggest that cytokinins constitutively exudated from host roots play a major role in haustorium formation and aggressiveness in P. ramosa.

International audience; We developed 20 microsatellite markers to genotype over 100 populations of the parasitic weed Phelipanche ramosa, which covers a wide host crop and geographic range. A representative core collection of 15 populations was also used in cross-infestation assays to study host preference during germination, attachment and shoot formation. We observed low genetic differentiation within most of the populations, but high genetic differentiation between populations partitioned into 3 genetic groups with different host preferences and geographic distributions. Genetic group 1 is detected exclusively in western France and on various host crops, notably winter oilseed rape (WOSR) and not hemp. Cross-infection assays confirmed its incompatibility with hemp and showed its preference for WOSR and tobacco in terms of germination and attachment success. The group 2 populations share a large geographic distribution in France and Europe, low germination success with WOSR and high germination success, attachment success and shoot formation with hemp, tobacco or tomato. The subclades 2a and 2b include most of the French populations in hemp crops in eastern France and in tobacco fields in several European countries respectively. The genetic analyses revealed the potential of the three groups to increase their geographic range in the future. Intermediate genetic groups showed higher intrapopulation diversity and represent potential stocks for new host race emergence. Those findings argue in favour of the existence of host races in P. ramosa and should be considered for appropriate management strategies, notably in breeding programmes for resistance against this parasitic weed.

BACKGROUND Some radiation-mutagenised chickpea mutants potentially resistant to the broomrape, Orobanche foetida Poir., were selected through field trials. The objectives of this work were to confirm resistance under artificial infestation, in pots and mini-rhizotron systems, and to determine the developmental stages of broomrape affected by resistance and the relevant resistance mechanisms induced by radiation mutagenesis. RESULTS Among 30 mutants tested for resistance to O. foetida, five shared strong resistance in both pot experiments and mini-rhizotron systems. Resistance was not complete, but the few individuals that escaped resistance displayed high disorders of shoot development. Results demonstrated a 2–3-fold decrease in stimulatory activity of root exudates towards broomrape seed germination in resistant mutants in comparison with non-irradiated control plants and susceptible mutants. Resistance was associated with an induction of broomrape necrosis early during infection. When infested, most of the resistant mutants shared enhanced levels of soluble phenolic contents, phenylalanine ammonia lyase activity, guaiacol peroxidase activity and polyphenol oxidase activity, in addition to glutathione and notably ascorbate peroxidase gene expression in roots. CONCLUSION Results confirmed enhanced resistance in chickpea radiation-mutagenised mutants, and demonstrated that resistance is based on alteration of root exudation, presumed cell-wall reinforcement and change in root oxidative status in response to infection. © 2016 Society of Chemical Industry

International audience; Sedimentary basins are, in many cases, the only preserved record of the geological processes active through geological time. These deposits preserve age, isotopic and geochemical signatures of the source rocks, providing insights into tectonic setting. In this context, we investigate the lithostratigraphy, provenance and tectonic environment of the Carrancas Group, a meta-sedimentary sequence characterised by a basal psammitic unit (Lower Unit) passing up into interbedded psammo-pelites (Intermediate Unit), which are overlain by a further psammitic unit (Upper Unit). This succession, along with the Canastra, Paranoá and Vazante groups (Brasília orogen), comprises a mature passive margin developed at the western-southern margin of the São Francisco craton during the Neoproterozoic. Detrital zircon U-Pb ages and Nd-Hf isotope data enable revised stratigraphic correlations across the basin, highlighting variations in the source of detritus. Deposition began after 0.92 Ga, with the sedimentation of the Lower Unit, with input of juvenile ∼2.1 Ga detritus (εNdt = −0.1 to +3.2) derived from the São Francisco craton. A marine transgression characterises the transition to the Intermediate Unit, which gradually covered over juvenile ∼2.1 Ga terrains and rendered them unavailable as possible source rocks for detrital zircons. Evolved Archaean-Palaeoproterozoic basement rocks, the Espinhaço Supergroup and evolved Neoproterozoic granitoids, all belonging to the São Francisco craton, acted as sources for the Intermediate and Upper units (U-Pb = 2.7 Ga to 0.9 Ga; εNdt = −10.4 to +7.1). The Goiás massif might also have acted as a source, providing juvenile Mesoproterozoic detritus. The sediments of the Upper Unit register a significant shift in source, with incoming of juvenile Mesoproterozoic and crustal 1.9 – 1.7 Ga detritus. This change in the ages of the detrital material may be related to exhumation of the Espinhaço Supergroup during the evolution of the passive continental margin basin. The whole-rock Nd TDM model ages become progressively younger from the base to the top of the sequence (Nd TDM = 2.6–1.8 Ga). The collisional events between Amazonian and São Francisco cratons and the Paranapanema block set the minimum age of deposition along this passive continental margin at around 0.67 Ga.

Karrikins stimulate Arabidopsis thaliana germination, whereas parasitic weeds of the Orobanchaceae family have evolved to respond to host-exuded compounds such as strigolactones, dehydrocostus lactone, and 2-phenylethyl isothiocyanate. In Phelipanche ramosa, strigolactone-induced germination was shown to require one of the CYP707A proteins involved in abscisic acid catabolism. Here, germination and gene expression were analysed to investigate the role of CYP707As in germination of both parasitic plants and Arabidopsis upon perception of germination stimulants, after using pharmacological inhibitors and Arabidopsis mutants disrupting germination signals. CYP707A genes were up-regulated upon treatment with effective germination stimulants in both parasitic plants and Arabidopsis. Obligate parasitic plants exhibited both intensified up-regulation of CYP707A genes and increased sensitivity to the CYP707A inhibitor abscinazole-E2B, whereas Arabidopsis cyp707a mutants still positively responded to germination stimulation. In Arabidopsis, CYP707A regulation required the canonical karrikin signalling pathway KAI2/MAX2/SMAX1 and the transcription factor WRKY33. Finally, CYP707As and WRKY33 also modulated Arabidopsis root architecture in response to the synthetic strigolactone rac-GR24, and wrky33-1 exhibited a shoot hyperbranched phenotype. This study suggests that the lack of host-independent germination in obligate parasites is associated with an exacerbated CYP707A induction and that CYP707As and WRKY33 are new players involved in a variety of strigolactone/karrikin responses.

Less than 25% of the volume of the juvenile continental crust preserved today is older than 3 Ga, there are no known rocks older than approximately 4 Ga, and yet a number of recent models of continental growth suggest that at least 60–80% of the present volume of the continental crust had been generated by 3 Ga. Such models require that large volumes of pre-3 Ga crust were destroyed and replaced by younger crust since the late Archaean. To address this issue, we evaluate the influence on the rock record of changing the rates of generation and destruction of the continental crust at different times in Earth's history. We adopted a box model approach in a numerical model constrained by the estimated volumes of continental crust at 3 Ga and the present day, and by the distribution of crust formation ages in the present-day crust. The data generated by the model suggest that new continental crust was generated continuously, but with a marked decrease in the net growth rate at approximately 3 Ga resulting in a temporary reduction in the volume of continental crust at that time. Destruction rates increased dramatically around 3 billion years ago, which may be linked to the widespread development of subduction zones. The volume of continental crust may have exceeded its present value by the mid/late Proterozoic. In this model, about 2.6–2.3 times of the present volume of continental crust has been generated since Earth's formation, and approximately 1.6–1.3 times of this volume has been destroyed and recycled back into the mantle. The successful synthesis of chiral amines from ketones using ω-transaminases has been shown in many cases in the last two decades. In contrast, the amination of β-keto acids is a special and relatively new challenge, as they decompose easily in aqueous solution. To avoid this, transamination of the more stable β-keto esters would be an interesting alternative. For this reason, ω-transaminases were tested in this study, which enabled the transamination of the β-keto ester substrate ethyl benzoylacetate. Therefore, a ω-transaminase library was screened using a coloring o-xylylenediamine assay. The ω-transaminase mutants 3FCR_4M and ATA117 11Rd show great potential for further engineering experiments aiming at the synthesis of chiral (S)- and (R)-β-phenylalanine esters. This alternative approach resulted in the conversion of 32% and 13% for the (S)- and (R)-enantiomer, respectively. Furthermore, the (S)-β-phenylalanine ethyl ester was isolated by performing a semi-preparative synthesis.

a b s t r a c t Mantle plumes have been increasingly studied in the past years, yet they remain not well understood. While geophysical approaches attempt to locate the depth of sources, petrological approaches characterize the nature of the materials that melt and geochemical approaches trace their history and origin. However, combined studies remain rare while they could provide strong constraints on the processes that give rise to mantle plumes. Here we present a new model that combines petrological and geochemical approaches to constrain the nature of source materials, their temperature of melting, their composition and their age. We focus on Gambier Island, Pitcairn chain in Polynesia because it has the advantage of combining two important features, (a) most lavas are tholeiites, a necessary feature to perform an accurate petrological modeling that can be compared to other plumes and (b) it belongs to the Pitcairn Chain, whose composition varies through time and samples today the extreme EM1-type source. We present geochemical analyses as well as Sr-Nd-Pb-Hf isotopes of lavas and high-precision olivine analyses. Using in situ olivine analyses and REE modeling, we constrain the composition of the mantle source (5% recycled oceanic crust - 95% peridotite), how both components melt (25-30% versus 0.5-1.5%) and we estimate the temperature of Gambier source at about 1400 ◦ C, a rather low temperature compared to Hawaii for example. We also constrain both the age and the nature of the recycled material using the isotopic data. They require the presence of about 3% sediment associated to recycled basalt in the pyroxenitic component and its age is about 1.5 Gyr. We also attribute the marked Nb-Ta positive anomalies and the elevated Ce/Pb ratios to the presence of recycled basalt in the mantle source. These features resemble typical HIMU lavas but the younger age of the recycled material together with the presence of sediment in the plume source explains the lower Pb isotopic ratios. The clear isotopic change between Gambier and Pitcairn Islands suggests that the plume source changed radically within 5 Ma. More generally, our new data and model suggest that the FOZO composition as sampled by Gambier lavas does not necessarily represent a large and rather primitive mantle source.
