101. Lack of consensus on an aging biology paradigm? A global survey reveals an agreement to disagree, and the need for an interdisciplinary framework

Abstract

At a recent symposium on aging biology, a debate was held as to whether or not we know what biological aging is. Most of the participants were struck not only by the lack of consensus on this core question, but also on many basic tenets of the field. Accordingly, we undertook a systematic survey of our 71 participants on key questions that were raised during the debate and symposium, eliciting 37 responses. The results confirmed the impression from the symposium: there is marked disagreement on the most fundamental questions in the field, and little consensus on anything other than the heterogeneous nature of aging processes. Areas of major disagreement included what participants viewed as the essence of aging, when it begins, whether aging is programmed or not, whether we currently have a good understanding of aging mechanisms, whether aging is or will be quantifiable, whether aging will be treatable, and whether many non-aging species exist. These disagreements lay bare the urgent need for a more unified and cross-disciplinary paradigm in the biology of aging that will clarify both areas of agreement and disagreement, allowing research to proceed more efficiently. We suggest directions to encourage the emergence of such a paradigm.

Abstract

In a recent paper, Trofino Neto et al. have shown that one can formulate an LQ problem in the presence of uncertainties in such a way that the state and input weighting matrices can be derived as the solution of a nonlinear optimization problem. Here it is shown that for linear systems with jumping parameters, the introduction of uncertainties in the state and input matrices can still be carried out and a control law derived as the solution of a similar, but more complex, problem. Moreover, both of the nonlinear optimization problems can be formulated as linear matrix inequality (LMI) problems, i.e., convex optimization problems. Hence, if an optimal solution exists, it can easily be obtained by means of some available LMI software packages. The existence of such a solution is proved under a restrictive assumption. Abstract

This note is concerned with the adaptive stabilizing control problem for jump-Markov systems. A new control law is proposed based on a simple set of algebraic sufficient conditions. It is shown that this approach can be used for a class of stochastic models for which the more restrictive solution proposed by Caines and Zhang does not apply.

Abstract

International audience; The present paper describes a numerical study of force fluctuations experienced by a boundary wall subjected to a granular flow on two distinct systems, namely a lid-driven cavity and an immersed wall system. Though the two systems exhibit different time-averaged dynamics, the force fluctuations experienced by the boundary wall show robust features in terms of the shapes of the probability density distributions and auto-correlation functions, under a wide range of boundary confinement pressure and shearing velocity imparted to the granular flow at the top of the system. This study identifies the key link between the grain-wall force fluctuations and the μ(I)–rheology while moving from quasistatic to inertial regime.

Abstract

The International Design Study for the Neutrino Factory (the IDS-NF) was established by the community at the ninth International Workshop on Neutrino Factories, super-beams, and beta- beams which was held in Okayama in August 2007. The IDS-NF mandate is to deliver the Reference Design Report (RDR) for the facility on the timescale of 2012/13. In addition, the mandate for the study [3] requires an Interim Design Report to be delivered midway through the project as a step on the way to the RDR. This document, the IDR, has two functions: it marks the point in the IDS-NF at which the emphasis turns to the engineering studies required to deliver the RDR and it documents baseline concepts for the accelerator complex, the neutrino detectors, and the instrumentation systems. The IDS-NF is, in essence, a site-independent study. Example sites, CERN, FNAL, and RAL, have been identified to allow site-specific issues to be addressed in the cost analysis that will be presented in the RDR. The choice of example sites should not be interpreted as implying a preferred choice of site for the facility.

Abstract

International audience; Discrete element simulations are used to model a two-dimensional gravity-free granular sample, which is trapped between two smooth sidewalls and one bottom rough wall while being subject to a constant shearing velocity at the top under a given confinement pressure. This system, inspired by conventional fluid mechanics, is called a granular lid-driven cavity. Attention is firstly paid to the time-averaged dynamics of the grains once a steady-state is reached. Strong spatial heterogeneities associated with a large-scale vortex formed within the whole volume of the lid-driven cavity are observed. The mean steady force on the sidewall facing the shearing velocity is then investigated in detail for different cavity lengths, shearing velocities and confinement pressures at the top. The ratio of the force on the latter wall to the top confinement pressure force is not constant but depends on both the shearing velocity and the confinement pressure. Above a critical value of the cavity length relative to the wall height and over a wide range of both shearing velocity and top confinement pressure, all data merge into a one-to-one relation between the mean force scaled by the top pressure force and the macroscopic inertial number of the lid-driven cavity. This result reveals the key role played by the inertial rheology of the granular material in the granular force transmission.

Abstract

Although liver transplantation is a widely accepted treatment for hepatocellular carcinoma (HCC), much controversy remains and there is no generally accepted set of guidelines. An international consensus conference was held on Dec 2–4, 2010, in Zurich, Switzerland, with the aim of reviewing current practice regarding liver transplantation in patients with HCC and to develop internationally accepted statements and guidelines. The format of the conference was based on the Danish model. 19 working groups of experts prepared evidence-based reviews according to the Oxford classification, and drafted recommendations answering 19 specific questions. An independent jury of nine members was appointed to review these submissions and make final recommendations, after debates with the experts and audience at the conference. This report presents the final 37 statements and recommendations, covering assessment of candidates for liver transplantation, criteria for listing in cirrhotic and non-cirrhotic patients, role of tumour downstaging, management of patients on the waiting list, role of living donation, and post-transplant management.

Abstract

International audience; Impact forces from full-scale granular flows on civil engineering structures exhibit huge fluctuations mainly caused by the co-existence inside the granular medium between a quasi-static zone, in direct contact with the structure, and a surrounding inertial zone. The aim of the current paper is to describe a time-averaged study and to show first results about those force fluctuations with the help of discrete numerical simulations on a two dimensional granular lid-driven cavity. A small-scale granular sample is trapped between one bottom wall, two side walls and one upper wall shearing the sample at constant speed under a given confinement pressure. We conducted a systematic study of the time-averaged force on the walls under different velocities, pressures and lengths of the sample relative to its thickness. Our study evidences a critical length above which the results are not influenced by the length and we observe a recirculation zone whose center mainly depends on the shearing velocity. Despite the strong inhomogeneities of pressure distribution, the steady forces experienced by the walls are governed by the macroscopic inertial number. We also provide a preliminary analysis of the force fluctuations.

Abstract

The main goal of this paper is to study the infinite-horizon long run average continuous-time optimal control problem of piecewise deterministic Markov processes (PDMPs) with the control acting continuously on the jump intensity λ and on the transition measure Q of the process. We provide conditions for the existence of a solution to an integro-differential optimality inequality, the so called Hamilton-Jacobi-Bellman (HJB) equation, and for the existence of a deterministic stationary optimal policy. These results are obtained by using the so-called vanishing discount approach, under some continuity and compactness assumptions on the parameters of the problem, as well as some non-explosive conditions for the process. Abstract

We report the first measurement of the neutrino-oxygen neutral-current quasielastic (NCQE) cross section. It is obtained by observing nuclear deexcitation γ rays which follow neutrino-oxygen interactions at the Super-Kamiokande water Cherenkov detector. We use T2K data corresponding to 3.01 × 10²⁰ protons on target. By selecting only events during the T2K beam window and with well-reconstructed vertices in the fiducial volume, the large background rate from natural radioactivity is dramatically reduced. We observe 43 events in the 4–30 MeV reconstructed energy window, compared with an expectation of 51.0, which includes an estimated 16.2 background events. The background is primarily nonquasielastic neutral-current interactions and has only 1.2 events from natural radioactivity. The flux-averaged NCQE cross section we measure is 1.55 × 10⁻³⁸ cm² with a 68% confidence interval of (1.22, 2.20) × 10⁻³⁸ cm² at a median neutrino energy of 630 MeV, compared with the theoretical prediction of 2.01 × 10⁻³⁸ cm².

Abstract

Influence of surface area and age Fish are the main fauna compartment concerned with protection measures and management into Marine Protected Areas (MPA). Surveys were conducted in 8 MPAs by various sampling methods (visual surveys by SCUBA diving, anaesthetic catch, poisoning, fishing nets or fishing lines): Port-Cros (France), Lavezzi and Scandola (Corse, France), Monaco, Medes Islands (Spain), Tremiti, Torre Guaceto and Porto Cesareo (Italy). These areas presented similar biotopes (rocky substrata, Posidonia oceanica beds and sandy bottoms) but different ages (1963, Port-Cros to 1997, Porto Cesareo) and sizes (418 ha at Medes Islands to 16600 at Porto Cesareo). Tremiti and Porto Cesareo have a weak protection compared to the other MPAs considered. The first step of this work was to update surveys already published by including new observations and unpublished data. Surveys were then compared using two diversity indexes, Average Taxonomic Distinctness (AvTD) and Variation in Taxonomic Distinctness (VarTD). For each MPA, these indexes have been compared to theoretical values for the Mediterranean Sea. This theoretical value (331 species belonging to 94 families) was calculated from the total number of species reported in the Mediterranean (FishBase) without the lessepsian species, species inhabiting deep waters (more than 100 m depth), and sub-species. A total of 215 (Port-Cros), 198 (Scandola), 191 Lavezzi, 136 (Medes), 108 (Monaco), 105 (Torre Guaceto), 75 (Tremiti and Porto Cesareo) species have been reported in the different MPAs. Number of species found in Port-Cros, Scandola and Lavezzi corresponded to the diversity expected for Mediterranean fish samples: i.e. the diversity expected from a sample of n species (215, 198 or 191 species respectively) drawn at random from the total list (331 species). VarTD of all the well protected zones corresponded also to the Mediterranean expected values. Therefore, well protected areas allowed preservation of taxonomic relationships between species. Spearman non parametric correlations showed that species richness and AvTD were correlated to the age of MPA and not with its size. In conclusion, the implementation of new MPAs is discussed regarding these relationships: the size of the protected area is not as important as an efficient surveillance and a long term protection to ensure a high fish species diversity.

Abstract

Due to the industrial needs, one of the key issues nowadays is to develop numerical tools that are able to predict the leakage rate through a cracked structure. This paper presents a validation of a numerical modelling of leakage rate through a mortar specimen in a splitting test versus experimental results. The mechanical state of the material is described by means of an enhanced non-local damage model which takes into account the stress state and provides a realistic damage field at failure. Two continuous approaches based on permeability-damage law are considered to study the coupling between the mechanical state of the material and the permeability. The first one lies on coupling the permeability of a crack with the mechanical state by means of the damage variable, and the second one is based on the regularized equivalent strain. Results show that the first approach isn't acceptable in case of discontinuous macrocracking while the second one predict well the permeability in the same case.
