Collagen-induced arthritis (CIA) mouse model is currently the most widely used and reliable autoimmune model to study rheumatoid arthritis. In this model, we used bovine type II collagen (CII) and complete Freund’s adjuvant (CFA) or incomplete Freund’s adjuvant (IFA) to form emulsifier, and mice were injected intradermal to induce autoimmune arthritis (CIA). In this model, we ground bovine collagen type II (CII) with complete Freund’s adjuvant (CFA) or incomplete Freund’s adjuvant (IFA) to form emulsifiers, and intradermal injection in mice induced autoimmune arthritis (CIA). This article describes the mouse, CFA strains, key emulsification, anesthesia, and injection immune techniques, as well as the incidence, date of onset, score, pathological results of arthritis. The total time for preparation of reagents and immunization of 20 mice was about 2-2.5 hours. In this protocol, we induced a high incidence of CIA with DBA/1J in genetically susceptible mice and assessed the severity and pathology of the disease, at the same time we found that CII also can induced enteritis, including ileitis and colitis. The initial symptoms of arthritis appear in the 24-26 days of the experiment, that is, 3-5 days after the second immunization, the peak period of inflammation was 30-36 days, the arthritis incidence about 90-100%, at the same time, the incidence of enteritis and arthritis were the same, small intestinal inflammation was more severe, but the duration was short; while the colonic inflammation was mild, and the duration was longer than enteritis, we named it collagen induced inflammations (CIIs).

Background: Infertility and fertility treatment exacerbate the intensity of stress that couples experience and negatively affects the patients’ quality of life. Stress is a crucial factor in reproductive health and is likely to be one of the psychological factors affecting ART success. The qualitative research method permitted an understanding of the patients’ experience during the diagnosis of and treatment for IVF and identified the sources of psychological stress. This understanding could urge medical workers, families, and society at large to provide targeted help to these patients.Methods: A qualitative research was conducted in the reproductive medicine center of our hospital. Women who received IVF treatment were randomly selected.Results: In total, 16 interviews were conducted with female patients undergoing IVF-ET between May and October 2019. The psychological experience of female patients undergoing IVF–ET can be summarized into four themes: (1) negative emotions due to infertility; (2) suffering under stress; (3) eager to obtain family and social support; (4) desire for medical knowledge related to infertility.Conclusion: Medical staff should fully understand the emotional state of the patient during the process of infertility treatment and careful attention should be paid to the patients’ psychological needs. According to the individual differences between the patients, we should provide positive factors of social support. Furthermore, corresponding health guidance and psychological support to reduce the psychological stress of patients and thus improve their chances of getting pregnant should be provided.

© 2019 Elsevier Ltd The failure of underground structure is always caused by local stress redistribution resulted from transient impact loads. An analytical model is established and solved by using the complex variable function method to illustrate the dynamic stress concentration around a shallow-buried cavity under transient P wave loads. The transient wave response is attained as an integration of steady-state wave response in circular frequency domain. By adopting a Butterworth filter, the jump points in the dynamic stress concentration factor (DSCF) curve which is not in line with the overall trend is filtered out to obtain more reasonable results. The convergence and accuracy of the solutions are verified and discussed in detail. DSCF distributions of high wavenumber incidences differ from that of low wavenumber incidences significantly in amplitudes and distribution patterns. The effects of the cavity depth, incidence angle and position of wave peak on DSCF distributions are illustrated.

Many cases ultimately acquire resistance to crizotinib. Resistance, including ALK-dominant or ALK non-dominant, mechanisms have been described. Transformation to small-cell lung cancer is rare. Herein, we report a 49-year-old man diagnosed with adenocarcinoma, who was negative for EGFR and ALK genes as detected by reverse transcription polymerase chain reaction, and was treated with crizotinib. A new biopsy showed a small-cell lung cancer after disease progression. Then, next-generation sequencing (NGS) was carried out and detected a TP53 gene mutation, an ALK rearrangement, and no loss of the retinoblastoma gene (RB). Although a regimen for small-cell lung cancer may be one treatment option, a heterogeneous tumor may exist at the time of diagnosis and manifest during the course of disease. Hepatocellular carcinoma (HCC) originating from liver cells is the third leading cause of cancer-related death. Parietin is an anthraquinone, isolated from some plants such as Rheum ribes L. Cytotoxicity and genotoxicity tests have an important role in the assessment of heritable and carcinogenic risks. In this study, it was aimed to investigate the cytotoxic and genotoxic effect of parietin on HepG2 hepatocellular carcinoma cells. HepG2 cells were cultured in appropriate culture medium. Then, different concentrations of parietin (final concentrations in the well to be 25-1000 μM) were added to the cells and they were allowed to incubate for 24-48 hours. Cell viability and genotoxic effect were determined by using XTT method and comet assay, respectively. The IC50 concentration of parietin was detected as 25 μM at the 48 hour in HepG2 cells. According to the comet assay, there was no statistically significant increase DNA tail length, DNA tail intensity and DNA tail moment in parietin treated cell groups compared to control groups (p>0.05). In this study, it has been shown that parietin has cytotoxic at low dose, but has not genotoxic effect in HepG2 cells, and as a result parietin was found to be useful in combination with other drugs in the treatment of HepG2 cells. Hybrid perovskites incorporating conjugated organic cations enable unusual charge carrier interactions among organic and inorganic structural components, but are difficult to prepare as films due to disparate component chemical/physical characteristics (e.g., solubility, thermal stability). Here we demonstrate that resonant infrared matrix-assisted pulsed laser evaporation (RIR-MAPLE) mitigates these challenges, enabling facile deposition of lead-halide-based perovskite films incorporating variable-length oligothiophene cations. Density functional theory (DFT) predicts suitable organic and inorganic moieties that form quantum-well-like structures with targeted luminescence or exciton separation/quenching. RIR-MAPLE-deposited films enable confirmation of these predictions by optical measurements, which further display excited state behavior transcending traditional quantum-well models-i.e., with appropriate selection of specially synthesized organic/inorganic moieties, intercomponent carrier transfer efficiently converts excitons from singlet to triplet states in organics for which intersystem crossing cannot ordinarily compete with recombination. These observations demonstrate the uniquely versatile excited-state behavior in hybrid perovskite quantum wells, and the value of integrating DFT, organic synthesis, RIR-MAPLE and spectroscopy for screening/preparing rationally devised complex structures.
