Objectives This study aimed to demonstrate potential translation of pre-clinical studies to a home-based exercise intervention in mediating inflammatory cytokine markers and tumor progression in men under active surveillance for prostate cancer. Methods A 2-arm randomized control parallel group design was used. The exercise intervention consisted of 24 weeks of an aerobic and resistance home-based exercise program and results were compared to a waitlist control group. Data were collected at baseline and end of study for eotaxin, interferon-γ (INF-γ), interleukin-12 (IL-12), interleukin-1α (IL-1α), interleukin-5 (IL-5), interleukin-6 (IL-6), tumor necrosis factor alpha (TNF-α), and vascular endothelial growth factor (VEGF), distanced walked during a 6-minute walk test (6MWT), body mass index, and health-related quality of life. Results Non-significant decreases were observed in all biomarkers, especially VEGF (pre: 125.16 ± 198.66, post: 80.29 ± 124.30, P = .06) and INF-γ (pre: 152.88 ± 312.71, post: 118.93 ± 158.79, P = .08), in the intervention group; only IL- α (pre: 332.15 ± 656.77, post: 255.12 ± 502.09, P = .20) decreased in the control group while all other biomarkers increased from baseline to end of study. A non-significant increase in 6MWT distance was observed in the intervention group, while a decrease was seen in the control group. Significant decreases in physical function, emotional wellbeing, and total composite scale on the FACIT-F were observed in the intervention group, possibly due to the isolation restrictions of COVID-19. Physical function on the SF-36 significantly increased in the control group. Conclusions Future studies with powered samples are needed to confirm the trends observed for inflammatory biomarkers and functional fitness.

Background: Skepticism among the public surrounding the COVID-19 vaccine is still prevalent despite vaccine-positive communication and many Americans having already received the vaccine. Side effects of the vaccine, as well as its expeditious research and development, are among the top concerns among those hesitant to receive the coronavirus vaccine. Moreover, there is additional concern regarding the association between comorbidities and severity of illness due to the coronavirus pandemic. Objective: We aimed to describe the pandemic- and vaccine-related concerns of South Texas residents who attended the UT Health San Antonio School of Nursing’s vaccine clinic with the goal of better understanding vaccine-related misconceptions and hesitancy for subsequent vaccination campaigns and boosters. Methods: An electronic survey accessible via a QR code on printed flyers was distributed throughout the waiting areas and post-vaccine observation rooms within the COVID-19 vaccine clinic at UT Health San Antonio School of Nursing from April 5 to 16, 2021. The survey contained a primary open-ended question designed to obtain information on concerns of the clinic attendees regarding the COVID-19 pandemic and COVID-19 vaccine. A thematic analysis was performed on the qualitative data to identify major themes to better understand concerns of vaccine clinic visitors. Results: During the 11-day period, 510 attendees received vaccinations through the vaccination clinic and completed the survey. Five areas of concern were identified by the 277 attendees: immunity, future vaccinations, vaccine symptoms and safety, protocol post-vaccination, and child vaccinations. Post-hoc sentiment analysis showed that responses were generally neutral or negative. Conclusion: This study provides a perspective regarding questions and concerns of South Texas residents regarding the COVID-19 pandemic, the vaccine, and their general health status within a vaccinated population. Vaccine recipients were found to still have questions even after receiving the vaccine, suggesting that eliminating uncertainty surrounding the COVID-19 vaccine is not necessary to motivate individuals to receive the vaccine. Instead, addressing concerns through public health messaging could be a useful strategy to address vaccine-related concerns and increase subsequent vaccine uptake in future vaccination campaigns and boosters.

Background: Due to associated comorbidities, the elderly population is more vulnerable to injuries with complications. This study was done to assess the severity of trauma and outcome of injuries among these patients presenting to the Emergency Department (ED). Materials and Methods: This was a retrospective cohort study. We included all patients aged more than 60 years, who presented to the ED with trauma in the year 2018. Details of the mode of trauma, severity of injuries and outcome were analysed. Results: During the study period a total of 7666 trauma patients presented to ED, among which 879 (879/7666: 11.4%) were in the geriatric age group. The mean age was 68.9 (SD: 7) years with 90.8% being young-old (60–79 years) and 9.2% being old-old (>80 years). Common modes of injuries were road traffic accidents (RTA) (64%) fall on level ground (FLG) (20%) and fall from height (FFH) (8%). Most of them had Injury Severity Score (ISS) and New Injury Severity Score (NISS) score of 0–7, i.e.: 62% and 51%, respectively. On multivariate logistic regression analysis RTA, FFH FLG and triage priority 1 patients were associated with trauma in the old-old as compared to the young–old age groups significantly. Independent risk factors associated with severe trauma were seen RTA, FLG and priority 1 patients. Forty-four per cent patients were managed by the ED team alone. Trauma speciality departments referred to included orthopaedics (47%), neurosurgery (22%), trauma surgery (14%), plastic surgery (8%) and hand reconstruction surgery (6%). About half of the population under study was discharged stable (44%), of which the majority belonged to the young-old category (44.1%). The in-hospital mortality rate was 0.5% (4/879). Conclusion: This study shows the gravity of multiple injuries sustained by the geriatric age group with RTA, FLG and FFH being the predominant causes of trauma. The NISS highlights the severity of injuries in the old and the frail.

Background: The Internet plays a very valuable role in knowledge, learning, and skills enhancement. There is scarce literature regarding the profile of internet addiction (IA) in patients of mixed anxiety and depressive disorders among developing countries. Aims: The aim of this study was to assess the profile of IA, anxiety, and depression in patients of mixed anxiety and depressive disorders. Methodology: This study was conducted at the Department of Psychiatry, MGM Medical College and Hospital, Navi Mumbai, India. Inclusion criteria comprised adult patients who were diagnosed with ICD 10 diagnostic criteria for mixed anxiety and depressive disorder, having access to the internet, and willing to participate in the study. Patients not having access to the internet and not willing to participate were excluded from the study. The patients were assessed using the IA test (IAT), Hamilton Depression Rating Scale (HAM-D), and Hamilton Anxiety Rating Scale (HAM-A). Informed consent and Institutional Ethics Committee Clearance was obtained. Data were analyzed biostatistically. Results: Our sample size of n = 60 comprised 60% (n = 36) males and 40% (n = 24) females. The mean age of the sample was 32.6 years. Majority of patients came from the semi-urban area. The mean HAM-D score of the sample was 18.96 and the mean score HAM-A score was 20.2. The mean score of IAT was 34.7. Conclusions: We conclude that the IA is prevalent in patients with mixed anxiety and depressive disorders and clinical rating scales can provide additional symptom profiles for anxiety and depression.

Introduction: Trauma to the temporomandibular joint (TMJ) may be associated with injury to its associated soft tissues along with the bony condyle. However, it is often neglected that even in the absence of fracture of the mandibular condyle, there may be damage to the associated soft tissues of the joint. The routine radiographic study does not reveal the soft-tissue injury around the joint and its extent. Such injuries may progress to temporomandibular joint disorders (TMDs) in the future. Materials and Methods: A prospective study was conducted among 50 patients diagnosed with anterior mandibular fractures without condylar fracture. The patients were assessed for soft-tissue injury around the TMJ using an arthroscopic examination by a single qualified operator. Patients were evaluated clinically and arthroscopic examination was performed to examine the TMJ soft tissues at the time of surgery for facial fracture reduction under general anesthesia. Results: Among the 50 patients, n = 37 (74%) patients had severe injury to the soft tissues surrounding the TMJ. Majority of the patients had arthroscopic evidence of soft-tissue injury around the joint. Conclusion: From this study, we observe that patients with mandibular fracture without condylar involvement should be evaluated for TMJ soft-tissue injuries and subjected to long-term follow-up to prevent TMDs in the late postoperative period.

Background: Procedural sedation and analgesia (PSA) in the emergency department (ED) is mainly used for wound irrigation, reduction of fractures, and wound closure. Ketamine is one of the most commonly used drugs for PSA in the ED. The study was conducted in the ED of a large tertiary care hospital in southern India to evaluate the adverse effects of Ketamine on PSA. Materials and Methods: This is a prospective observational study performed in the ED for 6 months (October 2019–March 2020) in 151 patients who required Ketamine for PSA. Titrated doses of Ketamine was administered in all patients; hemodynamic variables and adverse events were recorded at timed intervals. Results: During the study period, a total of 151 patients in the ED required PSA. The mean age of the study Cohort was 37 ± 15 years, and males accounted for 83%. All individuals obtained adequate sedation and pain relief. It was found that the incidence of adverse reactions to Ketamine was higher in young people (18- to 40-year-old), which was 63%. The most common adverse reaction in the study population was 39 cases of hypertension (44.8%), followed by vomiting in 25 cases (28.7%) and delusion in 6 cases (4%). There was no significant adverse effect in any patients which necessitated admission. Conclusion: Ketamine is a drug with good analgesic, sedative properties and has been shown to have a good safety profile with minimal adverse events for use as PSA in ED. Side effects were most common in the younger adult age group and hypertension was the most common side effect.

This paper reports a study of the effect of wafer tilt during dopant implantation on the performance of silicon PN phase shifter and traveling-wave Mach-Zehnder modulator. The PN phase shifter is designed and process simulated to include the effects of different fabrication processes in the device performance. The wafer tilt during implantation is varied from 0° to 3°, 5°, and 7°. The resulting crystal damage during dopant implantation to form the PN junction and the active concentration profile upon annealing, along with the formation of dopant–defect clusters, are discussed. Compared to 0° tilt, 7° tilt results in $1.58\times $ higher phase shift and better modulation efficiency. The overall phase shifter performance is improved using 5° wafer tilt for implantation resulting in $1.23\times $ lower absorption, $1.45\times $ better modulation efficiency, and $3.14\times $ higher 3 dB $RC$ modulation bandwidth for lumped-driven phase shifter. A traveling-wave electrode to enhance the modulation bandwidth is used, and the modulator performance for non-return-to-zero on-off-keying modulation with–2.5 V bias and 2.5 $\text{V}_{pp}$ drive signal across each arm is evaluated using a dual-arm push-pull drive. The sample with 5° tilt shows better traveling-wave and modulator high-speed characteristics compared to the other samples. Among the four samples with different wafer tilts, the sample with 0° tilt shows the worst phase shifter performance, and the sample with 3° tilt shows the worst modulator characteristics. The best overall performance is obtained for the sample with 5° tilt. Compared to the modulator with implantation at 0° tilt, the 5° tilted sample shows $2.3\times $ higher 6.4 dB electrical bandwidth and $1.36\times $ higher 3 dB electro-optic bandwidth at–2.5 V using a traveling-wave electrode with $1.48\times $ lower energy-per-bit for 5 km transmission at the KP4-forward-error-correction bit-error-rate threshold. The comparison of the effect of wafer tilt angles on various device metrics is presented and discussed.

Background and Objectives: Pedestrian injuries are a common mode of trauma presenting to the emergency department (ED). This study analyzes the profile, severity, and hospital outcome of these victims. Methods: We conducted a retrospective cross-sectional analysis of pedestrian trauma victims who presented to our ED from January 2017 to December 2018. Results: During the 2-year study period, our ED received 463 patients with pedestrian trauma. The mean age was 39 (standard deviation: 22.72) years, with a male (309/463: 66.6%) predominance. Based on their hemodynamic stability, majority were triaged as Priority 2 (279/463: 60.2%). Injuries included fractures (49.5%; n = 229), lacerations (46.4%; n = 215), abrasions: (38.9%; n = 179) and brain contusions (23.1%; n = 107). Extremities were the most injured (66.6%), followed by the scalp/head (39.9%) and face/neck injuries (20.3%) patients. A New Injury Severity Score of more than or equal to 14 was noted in 64 (13.9%) patients. Most of the victims required surgical (major/minor) intervention as part of their treatment which accounted for 63.9% (n = 296) patients. The ED mortality rate was 0.4% (n = 2), whereas the in-hospital mortality rate was 2.2% (n = 8). Conclusions: Middle-aged males were the leading demographic affected by most events taking place in the evening. Extremities were the most injured body part with fractures being the principal type of injury. A significant number of victims had injuries that were grievous and required hospital admission.

Poly[(μ4-3-carboxybenzenesulfonato)silver(I)], Ag(O3SC6H4CO2H) or [Ag(C7H5O5S)]n, has been found to undergo a reversible phase transition from monoclinic to triclinic between 160 and 150 K. The low-temperature triclinic structure (space group P\overline{1}) has been determined at 100 K. In contrast to the reported room temperature monoclinic structure, in which the nearly equivalent carboxylate C—O distances indicate that the acidic hydrogen is randomly distributed between the O atoms, at 100 K the C—O (protonated) and C=O (unprotonated) bonds are clearly resolved, resulting in the reduction in symmetry from C2/c to P\overline{1}.

Background: : Breast cancer being one of the most common cancers in women all around the world is a dominant cause of deaths occurring all around the globe. The available potent drugs for breast cancer show adverse effects and are found ineffective in patients. Furthermore, this acquired resistance that is vulnerable to cancer-related mutations and the resistance due to minor heterogeneous subpopulation may improve the treatment's inefficiency. This study aims to find suitable and effective bioactive compounds by screening numerous phytochemical molecules using a bioinformatic approach to develop an effective treatment for breast cancer. Methods: : In this study, the proteins that play a crucial role in breast cancer were used. The phytochemicals with known anticancer properties were scrutinized based on pathophysiological relevance, pharmacokinetic characteristics, and drug-like properties using the SwissADME web server. These phytochemicals were filtered through the process of docking, and the top five ligands for each protein were selected. Further, these shortlisted molecules' bioavailability and toxicity profiles were assessed using SwissADME and ADMETlab 2.0 web server, respectively. Results: : The six target receptors such as CTLA4, CDK8, EGFR, mTOR, p53R2 and PR were identified using a bibliographic study. Further, the screening of 68 phytochemicals resulted in 38 molecules that satisfied the Lipinski rule. Owing to the binding affinity scores of 38 molecules, the top 5 molecules with each receptor were chosen, resulting in the top 10 drug candidates. Out of these ten, only five molecules, such as Ursolic acid, Enterolactone, Parthenolide, Berberine, and its derivative Berberastine, were safe and non-toxic. Conclusion: : The screening of numerous phytochemicals against pivotal breast cancer target proteins led to the five most promising therapeutic candidates. We believe these findings will aid in the development of traditional medicine-based therapy methods and the identification of viable hits for future lead optimization in breast cancer medication development.

BackgroundIn breast cancer survivors, chemotherapy-induced muscle loss has been shown to be attenuated with structured resistance exercise. Creatine supplementation can increase bioenergetics in skeletal muscle, which helps to improve overall strength and endurance and reduce muscular fatigue. Therefore, we hypothesize that adding creatinine supplementation to exercise training will accelerate improvements in strength, endurance, and bioenergetics in breast cancer survivors. ObjectiveThe primary objective is to determine the effects of combining creatine supplementation with exercise on modulating strength and physical function in breast cancer survivors by comparing these effects to those of exercise alone. The secondary objectives are to determine if creatine supplementation and exercise can increase the intramuscular storage of creatine and improve body composition by comparing this intervention to exercise alone. MethodsWe aim to test our hypothesis by conducting an open-label randomized controlled trial of 30 breast cancer survivors who have completed chemotherapy within 6 months of enrollment. Eligible participants will be equally randomized (1:1) to either a creatine and exercise group or an exercise-only group for this 12-week intervention. Individuals who are randomized to receive creatine will be initially dosed at 20 g per day for 7 days to boost the availability of creatine systemically. Thereafter, the dose will be reduced to 5 g per day for maintenance throughout the duration of the 12-week protocol. All participants will engage in 3 center-based exercise sessions, which will involve completing 3 sets of 8 to 12 repetitions on chest press, leg press, seated row, shoulder press, leg extension, and leg curl machines. The primary outcomes will include changes in strength, body composition, and physical function in breast cancer survivors. The secondary outcomes will be intramuscular concentrations of creatine and adenosine triphosphate in the vastus lateralis, midthigh cross-sectional area, and quality of life. ResultsAs of October 2021, a total of 9 patients have been enrolled into the study. No unexpected adverse events have been reported. ConclusionsCreatine is being studied as a potential agent for improving strength, endurance, and bioenergetics in breast cancer survivors following chemotherapy. The findings from our trial may have future implications for supporting breast cancer survivors in reversing the muscle loss experienced during chemotherapy and improving their physical function and quality of life. Pyo-pneumopericardium or purulent pericarditis is a rare medical entity associated with high mortality. We hereby report a rare case of a 25-years old lady with pyo-pneumopericardium, aspirated pus culture from the pericardial cavity of which grew yeast (Candida species) like organism. This patient underwent a pericardiocentesis and was initiated on generic antibiotic treatment. However, despite the best possible medical management, she succumbed to her illness. This is a rare case report from India and an addition to the already available literature.

Introduction: The extent of periodontal tissue destruction can be assessed using pocket depth which also indicates the condition of the extent of the local inflammation. There are so many social stigmas associated with eunuchs community that these people do not come out in open and even if they come out in open, they often resort to various debilitating lifestyle habits that affect their general well-being and oral health in a negative sense. The objective of the present study is to assess the periodontal status among the eunuchs residing in Central Gujarat and to analyse the association between the periodontal status of eunuchs residing in Central Gujarat and various demographic variables. Methodology: The survey was conducted in Central Gujarat, and data were collected through primary source using the WHO Oral Health Assessment Form for Adults (2013) to assess the periodontal status and loss of attachment. Clinical examination was performed by using a plane mouth mirror and Community Periodontal Index Probe (WHO probe) (CPI) probe under adequate natural/artificial light. Results: Out of 384 eunuchs, the mean value of periodontal pocket depth among the eunuchs is 3.12 ± 0.89; clinical attachment loss is 2.88 ± 0.74. The difference in the periodontal pocket depth and clinical loss of attachment was statistically insignificant indicating that there is no difference in periodontal pocket depth and clinical loss of attachment across all the age groups and education status of eunuchs. Conclusion: The results of the current study showed poor periodontal status among eunuch population in Central Gujarat. The periodontal condition helps estimate the oral condition of this population. These estimates are important for the future planning of dental services among eunuch community.

Background: National efforts to reduce maternal mortality with respect to community services have primarily focused on upgrading transportation infrastructure and formalizing training for care providers. There is, however, a paucity of baseline data on the profile and outcomes of pregnant women presenting to the Emergency Department (ED) in India. Methods: This retrospective study enrolled all pregnant women presenting to a large tertiary medical care center in India, between November 2016 and November 2017. Results: There were 696 ED visits by pregnant women during the study period. The mean age was 26.85 (SD: 4.88) years. Pregnant women in the first trimester contributed to 50.8% of all visits, and 54% being multigravida. The most common presenting complaints were bleeding/spotting per vaginum (PV) (38.2%) and abdominal pain (37.6%) followed by fever (21.6%) and vomiting (21.5%). Obstetric causes contributed to 53.2% of the ED visits, while nonobstetric causes amounted to 43.2%. Over a third (39.7%) required hospital admission. Of these patients, 73% delivered in CMC with live births amounting to 62.3% while 3.5% ended in fetal deaths. The miscarriages rate was as high as 28%. More than half (51.1%) of the deliveries were by normal vaginal delivery. There were no maternal deaths during the time of admission. Conclusions: Our study sheds new light on the profile of emergency visits among pregnant patients and their relationship to the outcome of pregnancy. First trimester visits were most common with complaints of bleeding PV and abdominal pain. This could explain the high rate of miscarriages among this population.

Background: Violence-related injuries top the list as a cause of mortality in the 15–40 years' age group in India. In contrast to the West, the spectrum of assault injuries in Southeast Asian countries is different. Our main aim was to profile intentional injuries due to interpersonal violence treated in the emergency department (ED) and to describe the severity, pattern, etiology, and outcome in such patients. Materials and Methods: We conducted a case-series analysis of assault victims who presented to the ED of Christian Medical College and Hospital, Vellore, India, from January 2017 to December 2018. Data were retrieved electronically from the clinical workstation. Categorized variables were summarized using counts and percentages. Quantitative variables were summarized using mean and standard deviation (SD). Results: During the study period, a total of 381 patients with a mean age of 36.16 (SD: 13.9) years presented to the ED. Male (81.9%) predominance was noted among these victims. A majority of them, i.e., 257 (67%) victims, were assaulted by people that were known to them, of which 66 (17.3%) victims were reported as domestic violence. Blunt objects were used in most, i.e., 234 (61.4%) cases. A spike in the incidence of assault, in general, was noted in the month of September during the South Indian festival season. Approximately one-fourth (21.3%) of the victims required hospital admission. Overall, 15.74% of the victims had to undergo major surgical procedures. There were no mortalities recorded among these study participants. Conclusions: Violence and its consequences lead to severe injuries and levy a heavy burden on health care. There is an urgent need to address the social and emotional needs of adolescents and young adults who are most at risk of being the victims of assault.

This paper deals with two-unit cold standby system with switching device and proviso of rest. The system consists of two non-identical units which are connected in parallel redundancy. Initially, the first unit is in operative mode and second is kept in cold standby mode. There is also a proviso for rest for the first unit after a random time from its starting operation. For operation and repair, priority is always given to first unit. A switching device is used to shift the failed unit to repair mode. If the switching device is found non-operational then priority is given to it for repair provided that system is in functional mode. The effectiveness of the proposed system has been carried out by determining various reliability characteristics of the proposed model.

In Saccharomyces cerevisiae, the meiosis-specific axis proteins Hop1 and Red1 are present nonuniformly across the genome. In a previous study, the meiosis-specific VMA1-derived endonuclease (VDE) was used to examine Spo11-independent recombination in a recombination reporter inserted in a Hop1/Red1-enriched region (HIS4) and in a Hop1/Red1-poor region (URA3). VDE-initiated crossovers at HIS4 were mostly dependent on Mlh3, a component of the MutLγ meiotic recombination intermediate resolvase, while VDE-initiated crossovers at URA3 were mostly Mlh3-independent. These differences were abolished in the absence of the chromosome axis remodeler Pch2, and crossovers at both loci became partly Mlh3-dependent. To test the generality of these observations, we examined inserts at six additional loci that differed in terms of Hop1/Red1 enrichment, chromosome size, and distance from centromeres and telomeres. All six loci behaved similarly to URA3: the vast majority of VDE-initiated crossovers were Mlh3-independent. This indicates that, counter to previous suggestions, levels of meiotic chromosome axis protein enrichment alone do not determine which recombination pathway gives rise to crossovers during VDE-initiated meiotic recombination. In pch2∆ mutants, the fraction of VDE-induced crossovers that were Mlh3-dependent increased to levels previously observed for Spo11-initiated crossovers in pch2∆, indicating that Pch2-dependent processes play an important role in controlling the balance between MutLγ-dependent and MutLγ-independent crossovers.

The study investigated the levels of organochlorine pesticides (OCPs) in different types of vegetables grown in periurban area of National Capital Region (NCR), India. Vegetable sampling was carried out in winter and summer season of 2012. A total of 20 different OCPs were determined using gas chromatography (GC) assembled with electron capture detector (ECD). Obtained results showed that average levels of ∑OCP ranged from 83.8 ± 25.5 ng g in smooth gourd to 222.4 ± 90.0 ng g in cauliflower. The mean concentrations of different OCPs were observed in order of ∑HCH > ∑CHLs > drins > ∑endosulfan > ∑DDT in all vegetables except in brinjal and smooth gourd. Most of the OCP residues recorded in vegetable samples exceeded the maximum residue levels (MRLs) set by international and national regulatory agencies. Health risk assessment suggests that daily dietary OCP exposure via vegetable consumption was higher for children (mean value 4.25E−05) than adults (mean value 2.19E−05). The hazard quotient (HQ) and lifetime cancer risk (LCR) estimated from dietary exposure of these vegetables were above the acceptable limit and can be considered as a serious concern for Delhi population. Allergic response to clopidogrel is a significant and difficult clinical problem as it is one of the recommended drugs after coronary stenting to prevent thrombosis. We present the case of a 57-year-old man who underwent coronary revascularization and developed an urticarial rash after being started on dual antiplatelet medication (aspirin and clopidogrel). He was managed conservatively, to which he responded well and was discharged on prasugrel. This case study from India adds to the available literature and discusses the alternative drugs that could be used in patients with clopidogrel hypersensitivity.

Background: Herein, we propose a novel RCT study to collect preliminary data on the impact of a 24-week home-based exercise program that can improve prognosis, physical function, and quality of life (QoL) in men with prostate cancer (PCa). This study will provide data on the feasibility of conducting a home-based exercise study and pilot data on the impact of exercise on circulating concentrations of biomarkers reported in the literature to be beneficial for the prognostication of PCa. Methods/design: Thirty male patients, clinically-diagnosed with prostate cancer under active surveillance, will be recruited to participate in a 2-arm, 24-week home-based program. Random allocation to each arm - intervention, and control – will be performed in a 1:1 ratio. Participants assigned to the intervention group will perform 30 min of light-to-moderate intensity walking five days a week (40–60% heart rate reserve) and three sets of 15 repetitions of light callisthenic exercises (bodyweight squats, incline push-ups, and hip thrusts) 3 days a week. Participants randomized to the control group will maintain normal activity throughout the 24 weeks. Four visits occurring at baseline, 12-, 18-, and 24-weeks will be used to assess QoL, body composition, prognostic biomarker concentrations, and overall physical function. Primary endpoints include significant changes in prognostic biomarkers. Secondary endpoints include changes in quality of life, physical function and body composition. Discussion: This study should demonstrate preliminary evidence that a home-based exercise intervention can impact biomarkers of progression while improving quality of life, physical function and body composition. Results from this study have the potential to promote health and wellness while minimizing cancer progression in men with PCa.

Background and Aims: Pregnancy is a developmental period demanding special adjustments. Psychologically, pregnancy can be associated with positive preparedness in some and fear of childbirth or doubts of mothering role in others. Anxiety and depression during pregnancy can have negative implications on its outcome. The current study aims to provide insight into factors influencing maternal preparedness for motherhood. Methodology: Hundred first time pregnant women were enrolled after written informed consent and ethical approval. The Pregnancy Experiences Scale-Brief Version was used to study maternal rating of uplifts and hassles experiences specific to pregnancy, indicating affective valence toward the pregnancy. Pregnancy-Related Anxiety Questionnaire (PRAQ-R) helped to identify pregnancy-specific anxiety and depressive symptoms were screened using Whooley's Questions. Data were analyzed using Microsoft Excel 15.30 (170107). Results: The mean age of the women was 23.1 years. More than half of the pregnancies were unplanned, and the mean duration of pregnancy was 6.8 months. The positive experience was reported more than a negative experience. Negative valence increased in the presence of psychosocial stressors. On PRAQ-R, anxiety symptoms were reported by 52% of the patients, especially high on the domain of “fear of giving birth” and increased with trimester. Depressive symptoms were reported by 23% of the women. Conclusion: Advanced gestation, presence of stressors, past psychiatric illness, ANC complications, and the presence of anxiety/depression influence maternal preparedness for motherhood. There is a need of sensitizing the doctors, nurses, health-care workers, and relatives regarding screening, referral of unmet psychological needs of pregnant women to help new mothers to adjust with their motherhood positively.

ABSTRACT IMPACT: Knowledge of which aspects of social connectedness most strongly associate with caregiver health and health behaviors can inform intervention targets to improve caregiver health OBJECTIVES/GOALS: Stressed dementia caregivers are at risk of poor health. Social connectedness may reduce adverse health effects, yet it is unknown about which aspects relate most strongly to health. This is a barrier to intervention development. Our study identifies aspects of social connectedness most strongly associate with caregivers’ daily health behaviors. METHODS/STUDY POPULATION: Data. Enrolled spousal caregivers completed 14 consecutive days of online surveys. Measures. We examined multiple health behaviors each day, which included: 1) number of occurrences of 3 potential binge-eating behaviors (range 0 to 30), 2) whether participants engaged in at least 30 minutes of physical activity, and 3) perceived sleep quality, rated 1 (very bad) to 5 (very good). We also examined a count of health symptoms caregivers experienced (e.g., backache; range: 0 to 7). Measures of social connectedness included: spousal emotional support, perceived spousal appreciation, emotional support from any source, and loneliness. Analysis. We applied bivariate multi-level mixed effects models to examine the association between each aspect of social connectedness and health behaviors day-to-day. RESULTS/ANTICIPATED RESULTS: Since November 2020, 5 of N=40 participants were enrolled, of whom 3 had completed all diary surveys. Participants were women ages 59 to 73, and included 4 non-Hispanic white and 1 Hispanic caregivers. Data included 51 days of surveys (93% adherence). No differences in eating behaviors nor physical activity according to social connectedness were found. Emotional support from any source was positively associated with sleep quality (B= 0.20; SE=0.08; p-value 0.015). On days when caregivers indicated their spouse appreciated them ‘A lot’ versus ‘Not at all,’ sleep quality was marginally better (B=0.73, SE=0.42; p-value-0.08). Finally, days when caregivers felt lonely at least ‘Some of the time’ versus ‘Not at all’ were associated with experiencing more adverse health symptoms (B=1.54; SE=0.58; p-value

Statement of Problem: Various osteotomy modalities seem to have an impact on the primary and secondary stability of the dental implant. The available literature lacks the comparison of various available osteotomy modalities used for the dental implant placement and its effects on the initial surgical bone removal. Purpose: The purpose of this study is to compare and evaluate the osteotomy sites created using standard drill, bone trephine, and alveolar expanders for dental implant surgery. Materials and Methods: The study was done on ten goat hemimandibles. Three osteotomy sites were prepared at the inferior border of the mandible using standard drill, trephine, and alveolar expander in each hemimandibles and the sites were subjected to cone-beam computed tomography (CBCT). The CBCT images obtained were compared for the amount of cortical bone and bone marrow loss at osteotomy sites in different techniques. Results: The mean and standard deviation of loss of cortical bone with standard drills, trephines, and alveolar expanders was 3.62 ± 4.216 × 10−2, 3.6 ± 4.681 × 10−16 and 3.15 ± 7.071 × 10−2. At the middle-third region, the loss of marrow bone was 3.38 ± 7.88 × 10−2, 2.15 ± 8.498 × 10−2 and 0.03 ± 9.487 × 10−2, and at lower third region, it was 2.3 ± 4.714 × 10−2, 0.02 ± 6.325 × 10−2, and 0.0, respectively. Conclusion: CBCT images showed minimum bone loss with the use of alveolar expander which may be due to the lateral bone condensation rather the removal of the marrow. Trephine showed less marrow removal in comparison to the standard drill used for dental implant surgery.

Abstract Background There are currently 15 million Americans who provide over 80% of the care required by their family members with Alzheimer’s disease and other dementias. Yet care for caregivers continues to be fragmented and few evidence-based interventions have been translated into routine clinical care and therefore remain inaccessible to most family caregivers. To address this gap, the Caring for the Caregiver program is being developed at UT Health San Antonio, School of Nursing to improve support services and health outcomes for family caregivers. Our purpose is to describe the engagement process undertaken to assess caregiver and community needs and how findings are informing program development. Methods We are using a model of public engagement that consists of communication of information, collection of information from stakeholders, and collaboration where stakeholders are partners in an exchange of information to guide program activities. An assessment of the community was undertaken to identify resources/services for family caregivers. Subsequently, stakeholders were invited to a community-academic forum to discuss strategies to build on existing strengths for family caregiving and to identify gaps in care. Detailed notes were taken and all discussions were recorded and transcribed for analysis. Data were analyzed using thematic content analysis. Results We conducted site visits with 15 community agencies, interviewed 13 family caregivers, and attended community events including support groups and health and senior fairs. Fifty-three diverse stakeholders attended the community-academic forum. Participants identified existing assets within our community to support family caregivers. Consistent among groups was the need to increase awareness in our community about family caregivers. Themes identified from the discussion were: making the invisible visible, you don’t know what you don’t know, learning too late, and anticipating and preparing for the future. Conclusions Incorporating caregiver and community stakeholders was critical to ensure that the priorities of our community are addressed in a culturally responsive accessible program for family caregivers. The forum served as important mechanism to partner with the community and will be an annual event where we can continue to work with our stakeholders around needs for practice, education, and research.

In this paper, a graded-index strained Si1-xGex optical PN phase shifter with multiple strained layers has been proposed for high speed data modulation at 1550 nm with low device footprint. The quasi-vectorial finite difference method, non-local empirical pseudopotential method and the Payne-Lacey model has been used to analyze the proposed device. A phase shift of 122°/mm, VπLπ of 0.74 V.cm, and insertion loss of 4 dB at 5 V reverse bias with a 3-dB modulation bandwidth of 27.5 GHz has been obtained for a 500 nm x 250 nm cross-section rib waveguide with 50 nm slab. A 15°/mm increase in phase shift and 0.08 V.cm decrease in VπLπ has been observed when the junction is shifted by 60 nm towards the N side.

There has been recent research on the use of dexamethasone as an adjunct to local anaesthetics to enhance the block characteristics and improve post-operative pain outcomes. Numerous studies have shown that perineural dexamethasone improves post-operative analgesia, along with other clinical benefits. Intra-space pterygomandibular twin mix anaesthesia is a novel technique for inferior alveolar nerve block used for mandibular anaesthesia. Twin mix anaesthesia has its advantages in shortening the latency and prolonging the duration of the soft tissue anaesthesia, along with improving the quality of life in the post-operative period after mandibular oral surgical procedures. The concern regarding the use of perineural dexamethasone has been discussed.

We synthesized cavitands containing COOH moieties at the upper rim of the cavity and evaluated the interaction between these cavitands and the sarin nerve gas simulant, dimethylmethylphosphonate (DMMP) using surface plasmon resonance (SPR) spectroscopy. The carboxylic acid group on the cavitand is expected to form a hydrogen bond with the P=O group of the organophosphorus vapors. Films of these cavitands produced a rapid and reversible SPR response to low concentrations of DMMP. We observed concentration dependent sorption of the DMMP molecule into the COOH containing layer in the ppb to ppm range. Spin-cast films and Langmuir–Blodgett (LB) depositions of bilayer thick cavitand films produced identical SPR shifts upon exposure to DMMP. The sensitivity of the sensor was enhanced via LB deposition of multiple bilayers. Eight-layer-thick films of the COOH cavitand showed sensitivity to DMMP concentrations as low as 16 ppb. The orientation of the COOH group into or out of the cavity did not affect DMMP binding, but strongly influenced the water uptake. In both cases the molecular recognition event responsible for the DMMP uptake has been elucidated via crystal structure analyses of the complexes of COOH in/out cavitands with DMMP. Furthermore, we demonstrated that the COOH containing cavitand had an SPR sensitivity to DMMP higher than the standard fluoropolyol sensing layer, and that the cavitand layer was less prone to water vapor and alcohol interferences. Hence, cavitand layers containing a COOH moiety are promising for use as sensitive and specific sensors for nerve gas agents. [ABSTRACT FROM AUTHOR]

Purpose: Various floating and pulsatile drug delivery systems suffer from variations in the gastric transit time affecting the bioavailability of drugs. The objective of the study was to develop Pantoprazole Sodium (PAN) microballoons that may prolong the gastric residence time and could enhance the drug bioavailability. Methods: Microballoons were prepared using Eudragit®L100 by adopting emulsion solvent diffusion method with non-effervescent approach, in vitro studies were performed and the in vivo evaluation was carried out employing ethanol induced ulceration method. Optimization and validation were carried out through Design Expert® software. Results: The results demonstrate an increase in percentage yield, buoyancy, encapsulation efficacy and swelling. Particles were in the size range 80-100 µm following zero order release pattern. SEM study revealed their rough surface with spherical shape, internal cavity and porous walls. DSC thermo gram confirms the encapsulation of drug in amorphous form. Significant anti ulcer activity was observed for the prepared microballoons. The calculated ulcer index and protection were 0.20±0.05 and 97.43 % respectively for LRS-O (optimized formulation). Conclusion: This kind of pH dependent drug delivery may provide an efficient dosage regimen with enhanced patient compliance.

Abstract Candida albicans is an opportunistic fungal pathogen colonizing the oral cavity. C. albicans secreted aspartic protease Sap6 is important for virulence during oral candidiasis since it degrades host tissues to release nutrients and essential transition metals. We found that zinc specifically increased C. albicans autoaggregation induced by Sap6; and that Sap6 itself bound zinc ions. In silico analysis of Sap6 predicted four amyloidogenic regions that were synthesized as peptides (P1–P4). All peptides, as well as full length Sap6, demonstrated amyloid properties, and addition of zinc further increased amyloid formation. Disruption of amyloid regions by Congo red significantly reduced auotoaggregation. Deletion of C. albicans genes that control zinc acquisition in the ZAP1 regulon, including zinc transporters (Pra1 and Zrt1) and other zinc-regulated surface proteins, resulted in lower autoaggregation and reduction of surface binding of Sap6. Cells with high expression of PRA1 and ZRT1 also showed increased Sap6-mediated autoaggregation. C. albicans ∆sap6 deletion mutants failed to accumulate intracellular zinc comparable to ∆zap1, ∆zrt1, and ∆pra1 cells. Thus Sap6 is a multi-functional molecule containing amyloid regions that promotes autoaggregation and zinc uptake, and may serve as an additional system for the community acquisition of zinc.

The purpose of this study was to examine the effects of exercise in modulating biomarkers of sarcopenia in a treatment naïve transgenic adenocardinoma of the mouse prostate (TRAMP) model. Thirty TRAMP mice were randomized to either exercise (voluntary wheel running) or no-treatment control group for a period of 20 weeks. During necropsy, gastrocnemius muscles and prostate tumors were harvested and weighed. Gastrocnemius concentrations of myostatin, insulin-like growth factor (IGF)-1 and tumor necrosis factor (TNF)-α were quantified. Exercise mice had greater muscle mass than controls (p=0.04). Myostatin was significantly lower in the exercise group compared to controls (p=0.01). Exercise mice maintained forelimb grip force while control mice had a significaint decrease (p=0.01). Enteric fevers, caused by the Salmonella enterica serovars Typhi (ST), Paratyphi A (PA) and Paratyphi B (PB), are life-threatening illnesses exhibiting very similar clinical symptoms but with distinct epidemiologies, geographical distributions and susceptibilities to antimicrobial treatment. Nevertheless, the mechanisms by which the host recognizes pathogens with high levels of homology, such as these bacterial serovars, remain poorly understood. Using a three-dimensional organotypic model of the human intestinal mucosa and PA, PB, and ST, we observed significant differences in the secretion patterns of pro-inflammatory cytokines and chemokines elicited by these serovars. These cytokines/chemokines were likely to be co-regulated and influenced the function of epithelial cells, such as the production of IL-8. We also found differing levels of polymorphonuclear leukocyte (PMN) migration among various infection conditions that either included or excluded lymphocytes and macrophages (Mϕ), strongly suggesting feedback mechanisms among these cells. Blocking experiments showed that IL-1β, IL-6, IL-8, TNF-α and CCL3 cytokines were involved in the differential regulation of migration patterns. We conclude that the crosstalk among the lymphocytes, Mϕ, PMN and epithelial cells is cytokine/chemokine-dependent and bacterial-serotype specific, and plays a pivotal role in orchestrating the functional efficiency of the innate cells and migratory characteristics of the leukocytes.

Abstract Muscle loss is a debilitating side effect to prostate cancer (PCa) experienced by nearly 60% of men. The purpose of this study was to test the hypothesis that Nexrutine®, a bark extract from the Phellodendrum amurense, can protect against prostate cancer induced muscle loss in a similar manner as exercise, using the transgenic adenocarcinoma of mouse prostate (TRAMP) model. Forty‐five, 8‐ to 10‐week old TRAMP mice were randomized to either control, Nexrutine® (600 mg/kg pelleted in chow) or exercise (voluntary wheel running). Mice were serially sacrificed at weeks 4, 8, 12, and 20, at which time either the left or right gastrocnemius muscle was harvested, weighted, and frozen. Proteolysis inducing factor (PIF), ubiquitin, and NF‐κB concentrations were quantified using ELISA kits. Nexrutine® and exercise were equally able to protect TRAMP mice against PCa‐induced muscle loss (P = 0.04). Addition of manufacturing defects to computational models of 3D-printed lattice structures enable improved simulation accuracy. A computational model of a cellular structure based on finite element method (FEM) analysis, often starts from defect-free computer-aided design (CAD) geometries to generate discretised meshes. Such idealised CAD geometries neglect imperfections, which occur during the additive manufacturing process of lattice structures, resulting in model oversimplification. This research aims to incorporate manufacturing defects in the strut elements of a lattice structure, thereby enhancing predictive capabilities of models. In this work, a method of generating CAD AM representative strut models is proposed. The models are generated from micro-computer tomography (μCT) analysis of SLM fabricated struts. The proposed additive manufacturing (AM) representative strut FE model's axial stiffness and critical buckling load is compared to idealised- and μCT- based FE models, with significant error reduction over idealised strut models. The AM representative strut models are then used to generate full lattice FE models and compared with manufactured and idealised FE models. The AM representative FE lattice models show greater correlations toward experiment and more realistic deformation behaviours. Overall, the methodology used in this study demonstrates a novel approach to representing struts in FE models of AM fabricated lattice structures. Over the past decade, there is an increasing interest in alcohol based fuels around the world due to the ease of handling, blending and the potential of production using renewable resources. Butanol is one of the suitable alternative candidate for IC engines due to its lower heating value (32 MJ/kg) in comparison to other alcohol based fuels such as ethanol (26.8 MJ/kg) and methanol (20 MJ/kg). The present study was carried out to evaluate the performance and emission characteristics of a single cylinder carbureted engine fuelled with the following gasoline-butanol blends ranging from 5%, 10%,15% and 20% of butanol by volume. During the tests negligible reduction in the performance characteristics were observed for gasoline-butanol blends in comparison to conventional gasoline. The emission characteristics of gasoline-butanol blends were observed to be superior to that of conventional gasoline. In the overall picture, the performance of gasoline-butanol blends indicated promising results as a substitute to conventional gasoline.

The purpose of this investigation was to compare the antitumorigenic effects of the natural product Nexrutine to voluntary wheel running (VWR) in the transgenic adenocarcinoma of the mouse prostate (TRAMP) model. Forty-five, 10-week old TRAMP mice were randomized to either receive free access to the running wheel, Nexrutine pelleted into chow at 600 mg/kg or no treatment control. Mice were serially sacrificed at weeks 4, 8,12 and 20 weeks. Palpable tumors, body weight, food consumption and running wheel activity were monitored weekly. At necropsy, tumors and serum were harvested and stored for analysis. Serum was used to quantify circulating cytokines in 4 and 20 week time points. Nexrutine supplementation led to a 66% protection against high grade tumors. Exercise resulted in a 60% protection against high grade tumors. Both interventions reduced concentrations of IL-1α. Exercise also significantly lowered concentrations of eotaxin, IL-5, IL-12(p40) and VEGF. While there were no significant differences at baseline, exercise mice had significantly lower IL-5 and VEGF compared to control at the 20 week time point. Nexrutine also significantly reduced circulating IL-9 concentrations. No significant differences were observed when compared to the control group. Immunohistochemistry of tumor sections showed significantly lower expression of pAkt in Nexrutine fed mice with no visible differences for NFκB. In conclusion, both Nexrutine and exercise suppressed tumor growth. Though similar outcomes were seen in this comparative effectiveness study, the mechanisms by which exercise and Nexrutine exert this benefit may focus on different pathways.

Summary: Human oligodendrocyte progenitor cells (hOPCs) persist into adulthood as an abundant precursor population capable of division and differentiation. The transcriptional mechanisms that regulate hOPC homeostasis remain poorly defined. Herein, we identify paired related homeobox protein 1 (PRRX1) in primary PDGFαR+ hOPCs. We show that enforced PRRX1 expression results in reversible G1/0 arrest. While both PRRX1 splice variants reduce hOPC proliferation, only PRRX1a abrogates migration. hOPC engraftment into hypomyelinated shiverer/rag2 mouse brain is severely impaired by PRRX1a, characterized by reduced cell proliferation and migration. PRRX1 induces a gene expression signature characteristic of stem cell quiescence. Both IFN-γ and BMP signaling upregulate PRRX1 and induce quiescence. PRRX1 knockdown modulates IFN-γ-induced quiescence. In mouse brain, PRRX1 mRNA was detected in non-dividing OPCs and is upregulated in OPCs following demyelination. Together, these data identify PRRX1 as a regulator of quiescence in hOPCs and as a potential regulator of pathological quiescence. : Wang et al. investigate the role of transcription factor PRRX1 in human oligodendrocyte progenitor cells. The development of spiking neural network simulation software is a critical component enabling the modeling of neural systems and the development of biologically inspired algorithms. Existing software frameworks support a wide range of neural functionality, software abstraction levels, and hardware devices, yet are typically not suitable for rapid prototyping or application to problems in the domain of machine learning. In this paper, we describe a new Python package for the simulation of spiking neural networks, specifically geared toward machine learning and reinforcement learning. Our software, called BindsNET1, enables rapid building and simulation of spiking networks and features user-friendly, concise syntax. BindsNET is built on the PyTorch deep neural networks library, facilitating the implementation of spiking neural networks on fast CPU and GPU computational platforms. Moreover, the BindsNET framework can be adjusted to utilize other existing computing and hardware backends; e.g., TensorFlow and SpiNNaker. We provide an interface with the OpenAI gym library, allowing for training and evaluation of spiking networks on reinforcement learning environments. We argue that this package facilitates the use of spiking networks for large-scale machine learning problems and show some simple examples by using BindsNET in practice.

Biofilm formation is a common strategy utilized by bacterial pathogens to establish persistence in a host niche. Salmonella enterica serovar Typhi, the etiological agent of Typhoid fever, relies on biofilm formation in the gallbladder to chronically colonize asymptomatic carriers, allowing for transmission to uninfected individuals. S. enterica serovar Typhimurium utilizes biofilms to achieve persistence in human and animal hosts, an issue of both clinical and agricultural importance. Here, we identify a compound that selectively inhibits biofilm formation in both S. Typhi and S. Typhimurium serovars at early stages of biofilm development with an EC50 of 21.0 and 7.4 μM, respectively. We find that this compound, T315, also reduces biofilm formation in Acinetobacter baumannii, a nosocomial and opportunistic pathogen with rising antibiotic resistance. T315 treatment in conjunction with sub-MIC dosing of ciprofloxacin further reduces S. enterica biofilm formation, demonstrating the potential of such combination therapies for therapeutic development. Through synthesis of two biotin-labeled T315 probes and subsequent pull-down and proteomics analysis, we identified a T315 binding target: WrbA, a flavin mononucleotide-dependent NADH:quinone oxidoreductase. Using a S. Typhimurium strain lacking WrbA we demonstrate that this factor contributes to endogenous S. enterica biofilm formation processes and is required for full T315 anti-biofilm activity. We suggest WrbA as a promising target for further development of anti-biofilm agents in Salmonella, with potential for use against additional bacterial pathogens. The development of anti-biofilm therapeutics will be essential to combat chronic carriage of Typhoid fever and thus accomplish a meaningful reduction of global disease burden.

Summary: Plasticity of thalamocortical (TC) synapses is robust during early development and becomes limited in the adult brain. We previously reported that a short duration of deafening strengthens TC synapses in the primary visual cortex (V1) of adult mice. Here, we demonstrate that deafening restores NMDA receptor (NMDAR)-dependent long-term potentiation (LTP) of TC synapses onto principal neurons in V1 layer 4 (L4), which is accompanied by an increase in NMDAR function. In contrast, deafening did not recover long-term depression (LTD) at TC synapses. Potentiation of TC synapses by deafening is absent in parvalbumin-positive (PV+) interneurons, resulting in an increase in feedforward excitation to inhibition (E/I) ratio. Furthermore, we found that a brief duration of deafening adult mice recovers rapid ocular dominance plasticity (ODP) mainly by accelerating potentiation of the open-eye responses. Our results suggest that cross-modal sensory deprivation promotes adult cortical plasticity by specifically recovering TC-LTP and increasing the E/I ratio. : Plasticity of thalamocortical (TC) synapses is limited in adults. Rodríguez et al. demonstrate that a brief period of deafening adults recovers LTP at TC synapses in visual cortex and accelerates ocular dominance plasticity. These results suggest that cross-modal sensory deprivation may be an effective way to promote adult cortical plasticity. Salivary Histatin 5 (Hst 5) is an antimicrobial peptide that exhibits potent antifungal activity towards Candida albicans, the causative agent of oral candidiasis. However, it exhibits limited activity in vivo, largely due to inactivation by salivary components of both host and pathogen origin. Proteins secreted by C. albicans during infection such as secreted aspartyl proteases (Saps) and shed mucin Msb2 can reduce Hst 5 activity; and human salivary mucins, while suggested to protect Hst 5 from proteolytic degradation, can entrap peptides into mucin gels, thereby reducing bioavailability. We show here that Sap6 that is secreted during hyphal growth reduces Hst 5 activity, most likely a result of proteolytic degradation of Hst 5 since this effect is abrogated with heat inactivated Sap 6. We further show that just like C. albicans shedding Msb2, mammalian mucins, fetuin and porcine gut mucin (that is related to salivary mucins), also reduce Hst 5 activity. However, we identify mucin-like protein-induced changes in C. albicans cell morphology and aggregation patterns, suggesting that the effect of such proteins on Hst 5 cannot be interpreted independently of their effect on yeast cells.

Background and Objectives: The present study was undertaken to compare and evaluate the area increase of root canals with ProTaper, iRaCe and Revo-S systems using cone beam computed tomography for analysis. Materials and Methodology: Forty five extracted human mandibular premolars having single canal and straight root were collected. Teeth were randomly assigned to three groups (n=15). Samples were decoronized by maintaining root length at 14 mm. Pre-instrumentation cone beam computed tomography scan was done after stabilizing the samples on wax blocks. The working length was determined at 1 mm short from the apical foramen by using a ISO 15 K-file tip protruding at apical foramen. Preparation was carried out according to the manufacturer′s instructions. Finally, canals were instrumented upto 30/.06 apically for each group. After each instrumentation, root canals were irrigated with 2ml of 3% sodium hypochlorite solution followed by 2 ml of 17% EDTA solution. Final irrigation was done with 5ml of saline. Post instrumentation cone beam computed tomography scans of all samples in the 3 groups were acquired. Results: Mean percentage of area increase in different thirds of the canal was highest for ProTaper followed by i-RaCe and Revo-s system which was statistically significant. Interpretation and Conclusion: Root canal area increase was highest for ProTaper followed by i-Race and Revo-S systems.
