103. Comparison of Lorentzian and Gaussian based approaches for laser speckle imaging of blood flow dynamics

Abstract

Since blood flow is tightly coupled to the health status of biological tissue, several instruments have been developed to monitor blood flow and perfusion dynamics. One such instrument is laser speckle imaging (LSI). The objective of this work is to evaluate an LSI instrument employing two statistically based approaches to calculate the speckle flow index (SFI). To study the relation between SFI and the actual flow rate for the two statistical approaches, speckle images were acquired from a 0.5% blood filled tube embedded within a 5 mm thick agar gel. A syringe based infusion pump was used to inject the blood at flow rates between 0 and 5 mm/sec. We found a linear relationship between SFI and actual flow rate for both the Gaussian and Lorentzian based approaches. With the Gaussian based approach, the SFI dynamic range was up to six times larger than with the Lorentzian based approach. The Gaussian based approach is a good alternative for computation of SFI using LSI.

Abstract

We simulate temperature depth profiling in human skin using pulsed photothermal radiometry (PPTR). By taking into account blackbody emission characteristics, spectral variation of human skin IR absorption coefficient, detectivity of available radiation detectors, and shot noise, we compute realistic PPTR signals for a test temperature profile, representing a subsurface vascular lesion. Analysis of the reconstructed temperature profiles enables a performance comparison of quantum IR detectors utilizing different spectral acquisition bands. The results suggest that HgCdTe detector used in 6-10 μxm spectral band performs better than an InSb detector used at 4-5 μm. © EDP Sciences.

Abstract

Background and Objectives: Pulsed dye laser (PDL) irradiation is the standard clinical treatment for vascular lesions. However, PDL treatment of port wine stain birthmarks (PWS) is variable and unpredictable. Photodynamic therapy (PDT) using benzoporphyrin derivative monoacid ring A (BPD) and yellow light may induce substantial vascular effects and potentially offer a more effective treatment. In this study, we utilize a rodent dorsal skinfold model to evaluate the vascular effects of BPD-PDT at 576 nm as compared to PDL. Study Design/Materials and Methods: A dorsal skinfold window was created on the backs of female Sprague-Dawley rats, allowing epidermal and subdermal irradiation and subdermal imaging. One mg/kg BPD was administered intravenously via a jugular venous catheter. Study groups were: control (no BPD, no light), PDL (585 nm, τ p 1.5 ms, 10 J/cm 2), and PDT (BPD + continuous wave irradiation (CW) at 576nm, τ p 16 min, 96 J/cm 2). Vessels were imaged and assessed for damage using laser speckle imaging (LSI) before, immediately after, and 18 hours post-intervention. Results: Epidermal irradiation was accomplished without blistering, scabbing or ulceration. PDL and PDT resulted in similar reductions in vascular perfusion 18 hours post-intervention (34.6% and 33.4%, respectively). Conclusions: BPD-PDT can achieve safe and selective vascular effects and may offer an alternative therapeutic option for treatment of hypervascular skin lesions including PWS birthmarks.

108. Measuring the effects of topically applied skin optical clearing agents and modeling the effects and consequences for laser therapies

Abstract

Human skin prepared with an optical clearing agent manifests reduced scattering as a result of de-hydration and refractive index matching. This has potentially large effects for laser therapies of several skin lesions such as port wine stain, hair removal and tattoo removal. With most topically applied clearing agents the clearing effect is limited because they penetrate poorly through the intact superficial skin layer (stratum corneum). Agent application modi other than topically are impractical and have limited the success of optical clearing in laser dermatology. In recent reports, however, a mixture of lipofylic and hydrofylic agents was shown to successfully penetrate through the intact stratum corneum layer which has raised new interest in this field. Immediately after application, the optical clearing effect is superficial and, as the agent diffuses through the skin, reduced scattering is manifested in deeper skin layers. For practical purposes as well as to maximize therapeutic success, it is important to quantify the reduced scattering as well as the trans-cutaneous transport dynamics of the agent. We determined the time and tissue depth resolved effects of optically cleared skin by inserting a microscopic reflector array in the skin. Depth dependent light intensity was measured by quantifying the signal of the reflector array with optical coherence tomography. A 1-dimensional mass diffusion model was used to estimate a trans-cutaneous transport diffusion constant for the clearing agent mixture. The results are used in Monte Carlo modeling to determine the optimal time of laser treatment after topical application of the optical clearing agent.

Abstract

Novel multi-wavelength switching filters based on cascaded long-period fiber grating (LPG) and polarization-maintaining fiber (PMF) in a Sagnac loop configuration are proposed and experimentally demonstrated. While the multi-channel spectrum of PMF Sagnac loop filter is controlled by the polarization state in the loop, the polarization-independence of the LPG filter is used to drop one channel. The polarization-independence of the cascaded LPG filter is also used for the fixed periodic loss spectrum to drop multiple channels in the loop. Although the polarization state of the optical wave filter is switched by the electro-optic modulator in the loop, the device is independent of the polarization state of the input light due to the bi-directional polarization compensation of the Sagnac loop interferometer. This device can be used to achieve all-channel gating and odd/even-channel switching with fine channel selectivity and high speed response.

Abstract

Selection of infrared (IR) detector is a key consideration in designing an experimental setup for temperature depth profiling using pulsed photothermal radiometry (PPTR). In addition to common detector characteristics, such as the spectral response, detector noise, and response speed, application-specific details must be taken into account to ensure optimal system performance. When comparing detectors with different spectral responses, blackbody emission characteristics must be considered in terms of influence on radiometric signal amplitude, as well as on background shot noise. In PPTR, optical penetration depth of the sample in the acquisition spectral band is also an important factor, affecting the stability of the temperature profile reconstruction. Moreover, due to spectral variation of IR absorption coefficient in watery tissues, the acquisition band must be appropriately narrowed to ensure the validity of the customary approximation of a constant absorption coefficient value in signal analysis. This reduces the signal-to-noise ratio, adversely affecting the stability and quality of the temperature profile reconstruction. We present a performance analysis of PPTR depth profiling in human skin using commercially available IR detectors (InSb, HgCdTe), operating in different spectral bands. A measurement simulation example, involving a simple, hyper-Gaussian temperature profile, and realistic noise levels, illustrates their expected performance.

Abstract

The relationship between laser pulse duration (τ p) and stress confinement time (τ sr) was studied using acoustic-based measurements of material absorption coefficient (μ a). A numerical model was developed to simulate stress signals. Experimental photoacoustic measurements at several values of τ were performed to estimate μ a of water. A numerical deconvolution technique was implemented to overcome the limitation of conventional photoacoustics, resulting in up to a 30% improvement in photoacoustic-based estimates of the sample μ a.

Abstract

Previous studies suggest that optimal port wine stain (PWS) laser treatment parameters require knowledge of skin characteristics such as blood vessel size, depth, and distribution. Effective and rapid imaging modalities are not widely available. In the present study, photothermal tomography (PTT) images of an in vivo hamster window model and human PWS skin were obtained and analyzed. Subtherapeutic laser light pulses at 585 and 600 nm were applied to skin surface and image sequences acquired with an infrared camera. A nonnegatively constrained conjugate gradient algorithm was used to reconstruct a PTT image of the initial temperature distribution immediately following pulsed laser irradiation. Vessel dimensions determined from PTT images of hamster window model skin compared well with those measured directly using video microscopy. PTT images of human PWS skin contained vessels with estimated diameters of 200-250 μm over a 250-320 μm depth range. Use of dual wavelength excitation (DWE) analysis allowed for imaging of shallow vessels.

Abstract

In therapy of port wine stain (PWS) birthmarks using pulsed green or yellow lasers, non-specific absorption by epidermal melanin reduces the amount of incident radiation that reaches the target PWS blood vessels. The related epidermal heating can induce blistering, dyspigmentation, or scarring, which limits the applicable radiant exposure, thus adversely affecting the efficacy of treatment in many patients. Our objective was to assess temperature depth profiles induced in PWS skin by a novel Nd:YAG laser emitting simultaneously at 1064 and 532 nm. The results should help determine safe radiant exposures for use in future clinical trials. The underlying hypothesis is that the added 1064 nm radiation may lead to a higher temperature increase in PWS relative to the epidermis, in comparison with a customary KTP/Nd:YAG laser system for vascular treatments (emitting at 532 nm only). The laser induced temperature profiles were determined in vivo using pulsed photothermal radiometry. A PWS test site was irradiated with a sub-therapeutic laser pulse and the transient change of the infrared radiant emission was recorded by a fast infrared camera. The laser-induced temperature profiles were reconstructed by solving the thermal-radiative inverse problem using an iterative minimization algorithm.

Abstract

The purpose of this study was to investigate the dynamics of laser light attenuation during cryogen spray cooling (CSC). Two detection schemes were used to approximate collimated and diffuse light transmittance measurements of continuous-wave (λ = 594 nm) and pulsed (λ = 585 nm) laser light during application of short (20-100 ms duration) cryogen spurts on a glass substrate. High-speed video images were also obtained during CSC. Collimated light transmittance varied considerably during CSC. Comparison of collimated and total transmitted light detection indicated that the diffuse component was substantial. Light attenuation occurred despite transparency of the liquid cryogen layer. Light scattering by cryogen results in a diverging laser beam incident on the skin surface. Since specular reflectance at the cryogen-skin interface may differ for diffuse light, further study of light scattering during CSC is warranted. Due to the differences in optical properties of glass and skin, experiments on skin need to be performed to extrapolate our results to the clinical scenario. For dermatologic procedures such as laser port wine stain and vascular lesion removal, hair removal, and nonablative skin rejuvenation, recommended τd are 10-80 ms. This range of τd appears to be appropriate, although more studies are required to arrive at a definite conclusion. © 2002 SPIE.

Abstract

Differential phase optical low coherence reflectometry (OLCR) was used to detect sub-wavelength displacements in the infrared-sensitive thoracic pit organ of Melanophila acuminata (Coleoptera: Buprestidae) upon absorption of infrared radiation at 3.39 microm. The displacement had more complex morphology but similar amplitude ( approximately 100 nm at 1 W cm(-2)) when compared to the displacement measured from the exocuticle in an alternate region on the beetle's body. In addition, a simplified finite difference model was developed to predict the temperature distribution and resultant thermal expansion in the pit organ tissue. The experimental and model results were interpreted to help clarify the mechanism by which the sensilla in the pit organ convert infrared radiation to neural signals. The results of this paper are discussed in relation to the photo-thermal-mechanical transduction hypothesis. This is the first experimental examination of the transduction mechanism in Melanophila acuminata.

Abstract

We designed a photoacoustic probe to determine the depths of epidermal melanin layer and port wine stain (PWS) in human skin. We present experimental data showing limitations in its use. We developed acrylamide tissue phantoms with optical properties matched to those of human PWS skin. We determined the limit in resolving epidermal and PWS layers. Additionally, we determined the maximum epidermal melanin concentration that allowed threshold detection of the PWS layer.
