133. Unusual tripodal ligands. Synthesis of 2,2-bis(diphenylphosphinomethyl)-1-phenylthiopropane and its group VI complexes

Abstract

A hybrid tripodal ligand containing phosphorus and sulfur atoms, 2,2-bis(diphenylphosphinomethyl)-1-phenylthiopropane (P2S), was synthesized and its metal carbonyl complexes (P2S)M(CO)4(M = Cr, Mo and W) were prepared. All synthetic intermediates and complexes have been characterized by spectroscopic methods and elemental analysis. The spectroscopic identification of complexes shows that the P2Sligand behaves as a bidentate ligand with two phosphorus atoms coordinating to the metal center. The structure was confirmed by a crystal structure study of (P2S)Mo(CO)4, which revealed an octahedral coordination around the molybdenum atom, and two cis-coordinated phosphorus donors. The conformation of the six-membered chelate ring is a twisted-boat and the possible reasons for this conformational arrangement are discussed.

A mononuclear complex (CO)3ReL2Cl (1a) (L = NH2CHMe2) and a dinuclear CO2complex (CO)3L2Re(μ,η1,η1-CO2)Re(CO)4L (2) are prepared from the reaction of a carbamoyl complex, (CO)4ReL(CONHCHMe2) with H2O in CHCl3. The coupling of a proposed intermediate (CO)4LReCOOH with 1ais a viable process for the formation of complex 2. The structures of (CO)3ReL2Cl (1b) (L= NH2CH2CH=CH2) and 2have been established by X-ray diffraction.

Abstract

Background: This study investigates the changes in human flu viruses in areas where unique animal flu viruses or epidemics were isolated in Taiwan and Kinmen islet. Materials and methods: Sentinel physicians obtained throat swabs from patients presenting with flu-like illness and two-step RT-PCR, using five sets of primers for matrix (M) and HA2, to detect all the animal influenza viruses and then typing/subtyping for human flu viruses. Questionnaires included travel history, animal contacts and occupations. Results: One flu B and eight flu A H3N2 isolates were identified in the flu season of 2002–2003, including 1, 2, 2 and 3 H3N2 in Taoyuan, Yilan, Tainan and Kinmen, respectively. Amino acid sequences of HA of human isolates revealed that Pro (P) at position 227 among most of the Kinmen isolates (2/3) replaced Ser (S) in the Taiwanese isolates, including one mutant from Gln (Q) to His (H) at position 156 of the HA gene for five out of eight H3N2 Taiwan isolates. Phylogenetic analysis demonstrated that all of the H3N2 viruses were of human origin (belonging to the lineage of A/Fujian/411/2002), with 80–90% homology in HA, NA and M gene segments of A/Swine/PingTung/Taiwan/199.2/02 (H3N2) and 75–77% homology in HA gene segment of A/Wild Bird/Taiwan/243/02(H3) and A/Mallard Duck/Taiwan/3.3/03 (H3). Conclusions: Residents in Kinmen islet frequently traveling from China need to monitor their flu A viruses that might be different from those obtained on the main Taiwan island. Abstract

Novel low-pathogenic avian influenza (LPAI) H5N2 viruses hit poultry farms in Taiwan in 2003, and evolved into highly pathogenic avian influenza (HPAI) viruses in 2010. These viruses are reassortant viruses containing HA and NA genes from American-lineage H5N2 and six internal genes from local H6N1 viruses. According to a serological survey, the Taiwan H5N2 viruses can cause asymptomatic infections in poultry workers. Therefore, a development of influenza H5N2 vaccines is desirable for pandemic preparation. In this study, we employed reverse genetics to generate a vaccine virus having HA and NA genes from A/Chicken/CY/A2628/2012 (E7, LPAI) and six internal genes from a Vero cell-adapted high-growth H5N1 vaccine virus (Vero-15). The reassortant H5N2 vaccine virus, E7-V15, presented high-growth efficiency in Vero cells (512 HAU, 107.6 TCID50/mL), and passed all tests for qualification of candidate vaccine viruses. In ferret immunization, two doses of inactivated whole virus antigens (3 μg of HA protein) adjuvanted with alum could induce robust antibody response (HI titre 113.14).
