Both sporadic variably protease-sensitive prionopathy (VPSPr) and familial Creutzfeldt-Jakob disease linked to the prion protein (PrP) V180I mutation (fCJDV180I) have been found to share a unique pathological prion protein (PrPSc) that lacks the protease-resistant PrPSc glycosylated at residue 181 because two of four PrP glycoforms are apparently not converted into the PrPSc from their cellular PrP (PrPC). To investigate the seeding activity of these unique PrPSc molecules, we conducted in vitro prion conversion experiments using serial protein misfolding cyclic amplification (sPMCA) and real-time quaking-induced conversion (RT-QuIC) assays with different PrPC substrates. We observed that the seeding of PrPSc from VPSPr or fCJDV180I in the sPMCA reaction containing normal human or humanized transgenic (Tg) mouse brain homogenates generated PrPSc molecules that unexpectedly exhibited a dominant diglycosylated PrP isoform along with PrP monoglycosylated at residue 181. The efficiency of PrPSc amplification was significantly higher in non-CJDMM than in non-CJDVV human brain homogenate, whereas it was higher in normal TgVV than in TgMM mouse brain homogenate. PrPC from the mixture of normal TgMM and Tg mouse brain expressing PrPV180I mutation (Tg180) but not TgV180I alone was converted into PrPSc by seeding with the VPSPr or fCJDV180I. The RT-QuIC seeding activity of PrPSc from VPSPr and fCJDV180I was significantly lower than that of sCJD. Our results suggest that the formation of glycoform-selective prions may be associated with an unidentified factor in the affected brain and the glycoform-deficiency of PrPSc does not affect the glycoforms of in vitro newly amplified PrPSc. To solve the problem of difficult recovery, high toxicity, serious restraint on reaction pH of traditional catalyst in advanced oxidation processes based on sulfate radical (SO4-), nano-scaled α-Fe2O3 was chosen as novel catalyticmaterial. The effects of different influence factors on the degradation of acid orange (AO) II by activation of hydrogen peroxide with nano α-Fe2O3 were investigated. The results show that the removal rate of organic dyes AO II has positive correlation with oxidant and catalyst dosages, and the corresponding kobs value linearly increases with catalyst dosage, the degradation efficiency of AO II is inhibited in actual waters. The mineralization rate of AO II in α-Fe2O3/PMS system is relatively low, most of the removed AO II is degraded to intermediate products. Nano-scaled α-Fe2O3 is confirmed to be a recyclable catalyst with high catalytic performance and low toxcity. [ABSTRACT FROM AUTHOR]

This study used a Mendelian randomization (MR) approach to investigate the causal relationship between genetically predicted endometriosis (EMS) and breast cancer risk. A total of 122,977 cases and 105,974 controls were included in the analysis, with gene-level summary data obtained from the Breast Cancer Association Consortium (BCAC). An inverse variance-weighting approach was applied to assess the causal relationship between EMS and breast cancer risk, and weighted median and MR-Egger regression methods were used to evaluate pleiotropy. Results showed a causal relationship between EMS and a decreased risk of overall breast cancer (odds ratio [OR] 0.95; 95% CI 0.90–0.99, p = 0.02). Furthermore, EMS was associated with a lower risk for estrogen receptor (ER)-positive breast cancer in a subgroup analysis based on immunohistochemistry type (OR 0.91; 95% CI 0.86–0.97, p = 0.005). However, there was no causal association between ER-negative breast cancer and survival (OR 1.00; 95% CI 0.94–1.06, p = 0.89). Pleiotropy was not observed. These findings provide evidence of a relationship between EMS and reduced breast cancer risk in invasive breast cancer overall and specific tissue types, and support the results of a previous observational study. The extracellular electron transfer (EET) is regarded as one of the crucial factors that limit the application of the bioelectrochemical system (BES). In this study, two different solid-phase redox mediators (RMs), biochar (1.2 g/L, T-B) and humin (1.2 g/L, T-H) were used for boosting the microorganisms accessing the electrons required for 2,4,6-TCP dechlorination under weak electrical stimulation (−0.278 V vs. Standard hydrogen electrode). BES with dissolved RM anthraquinone-2,6-disulfonate (AQDS 0.5 mmol/L, T-A) was used as a comparison. The results showed that dechlorination of 2,4,6-TCP could be greatly accelerated by biochar (1.78 d−1) and humin (1.50 d−1) than AQDS (0.24 d−1) and no RM control (T-M, 0.27 d−1). Moreover, phenol became the predominant dechlorination product in T-H (78.5 %) and T-B (63.0 %) instead of 4-CP in T-M (67.1 %) and T-A (89.8 %). Pseudomonas , Sulfurospirillum , Desulfuromonas , Dehalobacter , Anaeromyxobacter , and Dechloromonas belonging to Proteobacteria or Firmicutes rather than Chloroflexi might be responsible for the dechlorination activity. Notably, different RMs tended to stimulate distinct electroactive bacteria. Pseudomonas was the most abundant microorganism in T-M (41.92 %) and T-A (17.24 %), while Rhodobacter was most prevalent in T-H (20.04 %) and Azonexus was predominant in T-B (48.48 %). This study is essential in advancing the understanding of EET in BES for microbial degradation of organohalide contaminants under weak electrical stimulation. Objective: Angiogenesis-associated genes (AAGs) play a critical role in cancer patient survival. However, there are insufficient reports on the prognostic value of AAGs in head and neck squamous cell carcinoma (HNSC). Therefore, this study aimed to investigate the correlation between AAG expression levels and survival in HNSC patients, explore the predictive value of signature genes and lay the groundwork for future in-depth research. Methods: Relevant data for HNSC were obtained from the databases. AAGs-associated signature genes linked to prognosis were screened to construct a predictive model. Further analysis was conducted to determine the functional correlation of the signature genes. Results: The signature genes (STC1, SERPINA5, APP, OLR1, and PDGFA) were used to construct prognostic models. Patients were divided into high-risk and low-risk groups based on the calculated risk scores. Survival analysis showed that patients in the high-risk group had a significantly lower overall survival than those in the low-risk group (P < 0.05). Therefore, this prognostic model was an independent prognostic factor for predicting HNSC. In addition, patients in the low-risk group were more sensitive to multiple anti-cancer drugs. Functional correlation analysis showed a good correlation between the characteristic genes and HNSC metastasis, invasion, and angiogenesis. Conclusion: This study established a new prognostic model for AAGs and may guide the selection of therapeutic agents for HNSC. These genes have important functions in the tumor microenvironment; it also provides a valuable resource for the future clinical trials investigating the relationship between HNSC and AAGs. We evaluate the Opportunity Zones provision of the Tax Cuts and Jobs Act, focusing on its targeting and effects on investment and resident outcomes. The policy allowed substantial discretion for state governors to designate Opportunity Zones that were not necessarily the most distressed, though we find that in aggregate their ultimate selections were still somewhat well-targeted. However, we show that the policy is insufficient to encourage investment with a significantly below-market rate of return and provides the largest tax benefits to investment that would have occurred regardless of the policy. Consistent with these features of the policy's design, a substantial amount of Opportunity Zone investment has been made, including in many lower-income areas. However, it appears that much of the investment would have occurred anyway, and the evidence to date mostly points to limited effects on resident wellbeing. [ABSTRACT FROM AUTHOR]
