Animals that continually live in deep sea habitats face unique challenges and require adaptive specializations solutions in order to locate and identify food, predators, and conspecifics. The Ampullae of Lorenzini are specialized electroreceptors used by chondrichthyans for important biological functions. Ampullary organs of the ghost shark Chimaera monstrosa, a deep-sea species commonly captured as by-catch in the bottom trawl fishery, are here described for the first time using macroscopic, ultrastructural and histological approaches. The number of ampullary pores in C. monstrosa is about 700, distributed into the whole cephalic section of C. monstrosa, and organized in12 pore clusters and they are arranged into different configurations and form a distinct morphological pattern for this species, showing some anatomical peculiarities never described before in others cartilaginous fishes and may constitute an evolutionary adaptation of this ancient chondrichthyan species to the extreme environmental conditions of its deep sea niche. This article questions the contours of marine spatial planning and its application to non-humans while also paying attention to the role of the latter in structuring marine spaces and activities. Through two case studies focusing on sharks-oriented ecotourism in Fiji and sharks risk management in New Caledonia, we discuss how the 'right place' of sharks and humans at sea is negotiated, defined and enforced. Comparing the practices deployed to control sharks and humans' behaviours and whereabouts, we argue that sharks are increasingly incorporated into marine spatial planning although with distinct patterns of sharks–humans relations. Multiple paternity (MP) in the brown smooth‐hound shark (Mustelus henlei) was assessed in 15 litters (15 mothers and 97 embryos) collected in the northern Gulf of California of which 86.7% were sired by more than one male (i.e., from 2 to 4 sires). When taken together with results from previous studies, this record indicates that there is regional variation in MP in M. henlei in the northeastern Pacific. This pattern is associated with variations in the reproductive traits of each population (e.g., female size and litter size). In the Gulf of California, the results of a generalized linear model (GLZ) indicated that the litters of larger females had a higher probability of MP compared to those of smaller females. E2f4 is a multifunctional transcription factor that is essential for many cellular processes. Although the role of E2f4 during cell cycle progression has been investigated in great detail, less is known about E2f4 during embryonic development. Here, we investigated the role of E2f4 during zebrafish development. Zebrafish e2f4 mutants displayed ectopic otolith formation due to abnormal ciliary beating in the otic vesicle. The beating defects of motile cilia were caused by abnormal expression of ciliary motility genes. The expression of two genes, lrrc23 and ccdc151, were significantly decreased in the absence of E2f4. In addition to that, e2f4 mutants also displayed growth retardation both in the body length and body weight and mostly died at around 6 months old. Although food intake was normal in the mutants, we found that the microvilli of the intestinal epithelia were significantly affected in the mutants. Finally, the intestinal epithelia of e2f4 mutants also displayed reduced cell proliferation, together with an increased level of cell apoptosis. Our data suggested a tissue‐specific role of E2f4 during zebrafish development, which is distinct from the traditional views of this protein as a transcription repressor. [ABSTRACT FROM AUTHOR]

Secreted proteins of the Noggin family serve as pivotal regulators of early development and cell differentiation in all multicellular animals, including vertebrates. Noggin1 was identified first among all Noggins. Moreover, it was described as the first known embryonic inducer specifically secreted by the Spemann organizer and capable of inducing a secondary body axis when expressed ectopically. In the classical default model of neural induction, Noggin1 is presented as an antagonist of BMP signalling, playing a role as a neural inducer. Additionally, Noggin1 is involved in the dorsalization of embryonic mesoderm and later controls the differentiation of various tissues, including muscles, bones, and neural crest derivatives. Hitherto, noggin1 was found in all studied vertebrates. Here, we report the loss of noggin1 in elasmobranchs (sharks, rays and skates), which is a unique case among vertebrates. noggin2 and noggin4 retained in this group and studied in the embryos of the grey bamboo shark Chiloscyllium griseum revealed similarities in expression patterns and functional properties with their orthologues described in other vertebrates. The loss of noggin1 in elasmobranchs may be associated with histological features of the formation of their unique internal cartilaginous skeleton, although additional research is required to establish functional connections between these events., (© 2024. Pain is one of the most frequent and persistent side effects of breast cancer treatment. Besides pain, breast cancer survivors (BCS) are prone to experience a myriad of other signs and symptoms related to altered somatosensory function, including for example, hypoesthesia, allodynia, and hyperalgesia, both at the local site of cancer and in remote body parts. Different breast cancer treatments can have a direct effect on somatosensory functioning, resulting in a wide range of these signs and symptoms. To our knowledge, currently no comprehensive overview exists on altered somatosensory functioning and resulting signs and symptoms in BCS with persistent pain. Investigating altered somatosensory functioning in this population could provide more insights in the underpinning pathophysiological mechanisms and consequently improve prevention and treatment in the future. Therefore, in this paper, first, normal somatosensory functioning is described. Second, quantitative sensory testing is presented as the recommend method to evaluate somatosensory functioning. Third, existing evidence on altered somatosensory functioning in BCS with persistent pain is summarized. Altered somatosensory functioning related to the most common cancer treatment modalities, including surgery and radiotherapy, hormone therapy, and chemotherapy are discussed. In addition, evidence on the presence of nociplastic pain as pain resulting from altered somatosensory functioning without evidence for nociception and/or neuropathy in BCS is summarized. Evolution of the vertebrate urinary system occurs in response to numerous selective pressures, which have been incompletely characterized. Developing research into urinary evolution led to the occurrence of clinical applications and insights in paediatric urology, reproductive medicine, urolithiasis and other domains. Each nephron segment and urinary organ has functions that can be contextualized within an evolutionary framework. For example, the structure and function of the glomerulus and proximal tubule are highly conserved, enabling blood cells and proteins to be retained, and facilitating the elimination of oceanic Ca + and Mg + . Urea emerged as an osmotic mediator during evolution, as cells of large organisms required increased precision in the internal regulation of salinity and solutes. As the first vertebrates moved from water to land, acid-base regulation was shifted from gills to skin and kidneys in amphibians. In reptiles and birds, solute regulation no longer occurred through the skin but through nasal salt glands and post-renally, within the cloaca and the rectum. In placental mammals, nasal salt glands are absent and the rectum and urinary tracts became separate, which limited post-renal urine concentration and led to the necessity of a kidney capable of high urine concentration. For many years, tremendous effort has been dedicated to developing new industrial tuna fisheries, while their adverse impacts on threatened marine species have received relatively little attention. In tuna fisheries, bycatch is the major anthropogenic threat to marine megafauna in general, particularly sharks. Research on the development of gear technology for bycatch reduction and potential mitigation measures helped tuna Regional Fisheries Management Organizations adopt bycatch reduction management measures. After reviewing past research on the development of mitigation measures for pelagic longline and tropical purse seine fisheries based on pelagic species' behaviours, we describe promising new approaches integrating recent technological breakthroughs. New innovations include autonomous underwater vehicles carrying cameras along with miniaturized sensors, aerial drones, computer simulation of fishing gear geometry, environmental DNA assays, computer visualizations and deep learning. The successful application of such tools and methods promises to improve our understanding of factors that influence capture, escape and stress of caught species. Moreover, results emerging from recent ethological research explaining the power of social connection and learning in the "fish world" such as social learning from congeners, habituation to deterrents, and how past fishery interactions affect responses to fishing gear should be taken into account when developing technical mitigation measures. Scutellariae and Coptidis compose a classical drug pair applied in clinical practice to dispel heat, dryness, and dampness, and they are also precipitation compatible drug pairs. With modern technology, Scutellaria-Coptis is mostly prepared by decocting its components separately, while in the traditional method, it is predominantly prepared as a combined decoction. The present study investigated the effects and mechanisms of separate and combined application of Scutellaria-Coptis decoction on ulcerative colitis (UC) in mice induced by the administration of dextran sulfate sodium (DSS). Changes in body weight, colon length, and Disease Activity Index scores were also evaluated. Hematoxylin and eosin staining and other methods were used to evaluate the overall condition of animals in each group. Intestinal microflora was analyzed using 16S rRNA sequencing, while colon inflammation and antioxidant capacity were evaluated based on the levels of interleukin-6 (IL-6), IL-10, IL-1β, tumor necrosis factor-α, superoxide dismutase, malondialdehyde, and reduced glutathione. The results revealed that Scutellaria-Coptis significantly relieved colon inflammation in mice, and the combined decoction of Scutellaria-Coptis exerted a significant effect on UC. Notably, the protective effect of Scutellaria-Coptis against colon inflammation was weakened when the antibiotic mixture was partially consumed by the gut microbiota. The results of 16S rRNA sequencing showed that the group treated with combined decoction of Scutellaria-Coptis exhibited a higher intestinal microbial diversity and intestinal flora composition than the separated decoction group. Treatment of mice with UC by administering Scutellaria-Coptis decoction through intestinal flora removal (ABX) and fecal microbial transplantation (FMT) was closely associated with intestinal flora composition. In conclusion, Scutellaria-Coptis can relieve UC with an excellent effect especially when taken as a combined decoction, alleviating colon inflammation incurred by intestinal microbes to a certain extent. Although morphological abnormalities in several rays and skate species around the American continents have frequently mentioned, their numbers are unknown. The present work record morphological abnormalities in four Urotrygonidae species. Two anophthalmic specimens were detected (Urotrygon microphthalmum and Urobatis halleri). Two individuals lacked caudal fins (Urobatis maculatus and Urotrygon chilensis). Two round rays showed incomplete fusion of the pectoral fin to the head (U. microphthalmum and U. chilensis). Vertebral compression and fusion were found in a 6‐year‐old female Urotrygon rogersi. In addition, 118 abnormal batomorph specimens were gathered from the available bibliography, spanning the last six decades (1959–2021). Amblyraja doellojuradoi was the species with the highest number of abnormalities (18). The most common anomaly was an incomplete fusion of the pectoral fin with the head. Since 2010, at least 30 anomalous batomorphs have been recorded every 5 years. Sixty‐nine abnormal specimens occurred in the Northern Hemisphere (1.00–60.00 N). The Cortezian (Pacific) and Southeastern Brazil (Atlantic) marine ecoregions stood out with the highest number of these specimens. Mexico recorded 58 anomalous specimens, followed by Brazil (n = 36). Macroautophagy, the degradation and recycling of cytosolic components in the lysosome, is an important cellular mechanism. It is a membrane-mediated process that is linked to vesicular trafficking events. The sorting nexin (SNX) protein family controls the sorting of a large array of cargoes, and various SNXs impact autophagy. To improve our understanding of their functions in vivo, we screened all Drosophila SNXs using inducible RNA interference in the fat body. Significantly, depletion of Snazarus (Snz) led to decreased autophagic flux. Interestingly, we observed altered distribution of Vamp7-positive vesicles with Snz depletion, and the roles of Snz were conserved in human cells. SNX25, the closest human ortholog to Snz, regulates both VAMP8 endocytosis and lipid metabolism. Through knockout-rescue experiments, we demonstrate that these activities are dependent on specific SNX25 domains and that the autophagic defects seen upon SNX25 loss can be rescued by ethanolamine addition. We also demonstrate the presence of differentially spliced forms of SNX14 and SNX25 in cancer cells. This work identifies a conserved role for Snz/SNX25 as a regulator of autophagic flux and reveals differential isoform expression between paralogs. [ABSTRACT FROM AUTHOR]

Isolated teeth belonging to the genus Ptychodus Agassiz, 1834 (Chondrichthyes; Elasmobranchii) from the Upper Cretaceous of the Ryazan and Moscow Oblast regions (European Russia) are described and discussed in detail herein. The taxonomic composition of the Ptychodus assemblage from the Ryazan region is very diverse including the first records of the cuspidate species P. altior and P. anonymus, which thus is largely consistent with those from other contemporaneous European localities. Ptychodus ubiquitously inhabited epicontinental seas of Europe during most of the Cretaceous with the most diverse assemblages coming from southern England, northern Italy, Belgium, and European Russia. Additionally, the material documented here from the Cenomanian of Varavinsky ravine area (Moscow Oblast) represents the northernmost occurrence of Ptychodus hitherto reported from Europe. It is evident that the Late Cretaceous shallow seas of the Russian platform represented a crucial pathway for the dispersal of Ptychodus from the European peri-Tethys to the eastern margins of the Neo-Tethyan Ocean. The Albian–Campanian records of Ptychodus from Europe indicate that its dominance in the peri-Tethys persisted for most of its evolutionary history. A local temperature drop across most of the European shallow seas probably contributed to the narrowing of its geographic range in the peri-Tethyan seas towards the end of the Mesozoic Era. The fossil remains of Ptychodus documented herein are accordingly of utmost importance for better understanding the taxonomic composition of Russian fossil ichthyofaunas and also inform about the dispersal of Ptychodus towards western and eastern peri-Tethyan seas during the Late Cretaceous. Chondrichthyan fishes are ecologically and economically important, yet many are overfished or at elevated risk of extinction. Researchers report a desire to perform policy‐relevant science that can generate data in support of effective conservation and management plans, but also report a lack of clarity about how to most effectively to do that. To address this gap, we created a list of research and policy priorities for chondrichthyan species of conservation concern in US waters using a modified expert solicitation horizon scan approach. Thirty‐five policy‐relevant research priorities and twenty‐seven policy priorities are presented here, a list which can help to guide scientists and conservationists to maximize the effectiveness of their research and policy advocacy. Despite our critical dependence on aquatic wildlife, we lack a complete understanding of the drivers of population stability and structure for most fish species. Social network analysis has been increasingly used to investigate animal societies as it explicitly links individual decision-making to population-level processes and demography. While the study of social structure is of great ecological interest, it is also potentially important for species of economic value or of conservation concern. To date however, there has been little focus on how social processes are likely to influence the conservation of fish populations. Here we identify applications for how a social network approach can help address broad fish conservation themes such as population structure, biological invasions or fisheries management. We discuss the burgeoning opportunities offered and challenges still faced by current technologies to integrate social network approaches within fish conservation. The study of animal societies has been increasingly explored with social network analysis. Avasimibe (Ava) is an acetyl-CoA acetyltransferase 1 (ACAT1) specific inhibitor and an established medicine for atherosclerosis, owing to its excellent and safe anti-inflammation effects in humans. However, its efficacy in asthma has not yet been reported. We first administered varying concentrations of avasimibe to house dust mite (HDM)-induced asthmatic mice; results showed that 20 mg/kg avasimibe most significantly reduced IL-4 and IL-5 production in bronchoalveolar lavage fluid (BALF) and total IgE in serum, and the avasimibe treatment also exhibited lower mucus secretion, decreased goblet and basal cells but increased ciliated cells compared to the HDM group. And the redistribution of adherens junction (AJ) proteins induced by HDM was far more less upon avasimibe administration. However, avasimibe did not reduce the cholesterol ester ratio in lung tissues or intracellular cholesterol ester, which is avasimibe's main effect. Further analysis confirmed that avasimibe impaired epithelial basal cell proliferation independent of regulating cholesterol metabolism and we analyzed datasets using the Gene Expression Omnibus (GEO) database and then found that the KRT5 gene (basal cell marker) expression is correlated with the β-catenin gene. Moreover, we found that β-catenin localized in cytomembrane upon avasimibe treatment. Avasimibe also reduced β-catenin phosphorylation in the cytoplasm and inactivated the Wnt/β-catenin signaling pathway induced by HDMs, thereby alleviating the airway epithelial barrier disruption. Taken together, these findings indicated that avasimibe has potential as a new therapeutic option for allergic asthma. [ABSTRACT FROM AUTHOR]

The majority of intestinal in vitro screening models use cell lines that do not reflect the complexity of the human intestinal tract and hence often fail to accurately predict intestinal drug absorption. Tissue explants have intact intestinal architecture and cell type diversity, but show short viability in static conditions. Here, we present a medium throughput microphysiological system, Intestinal Explant Barrier Chip (IEBC), that creates a dynamic microfluidic microenvironment and prolongs tissue viability. Using a snap fit mechanism, we successfully incorporated human and porcine colon tissue explants and studied tissue functionality, integrity and viability for 24 hours. With a proper distinction of transcellular over paracellular transport (ratio >2), tissue functionality was good at early and late timepoints. Low leakage of FITC–dextran and preserved intracellular lactate dehydrogenase levels indicate maintained tissue integrity and viability, respectively. From a selection of low to high permeability drugs, 6 out of 7 properly ranked according to their fraction absorbed. Estrogen-related receptor (ERR) is an orphan nuclear receptor that was first discovered in animals, and play an important role in metabolism, development, and reproduction. Despite extensive research on the function of ERR, its transcriptional regulation mechanism remains unclear. In this study, we obtained the upstream region of Bombyx mori ERR (BmERR) and confirmed the promoter activity of this region. Interestingly, we found that 10 and 50 nM 20-hydroxyecdysone (20E) up-regulated the transcriptional activity of BmERR promoter. In addition, eight putative ecdysone response elements (EcREs) were predicted in the upstream sequence of BmERR. Based on their positions, the upstream sequence of BmERR was truncated into different fragments. Finally, an EcRE-like sequence (5′-AGTGCAGTAAACTGT-3′) was identified. Electrophoretic mobility shift assay (EMSA) and cell transfection experiments confirmed that this motif specifically binds to the complex formed between ecdysone receptor (BmEcR) and the ultraspiracle (BmUSP), a key complex in the 20E signaling pathway. Interference of BmERR or BmEcR mRNA in the embryonic cells of Bombyx mori significantly affected the expression of BmEcR and BmUSP. Overall, these results suggested that an EcRE element was identified from BmERR, and this will help understanding the detailed regulatory mechanism of ERR in insects. [ABSTRACT FROM AUTHOR]

Here we describe a new elasmobranch assemblage consisting of isolated dental material from the Aquitanian near-shore marine deposits of the Shuwayr and Warak formations at Sharbithat, in eastern Sultanate of Oman. The faunal composition clearly indicates affinities to other early Miocene elasmobranch-bearing localities worldwide. This assemblage is predominantly composed of large and common pelagic sharks as well as teeth attributable to a new species of fantail stingray, Taeniurops tosii, as old as the oldest undisputable fossil records of Taeniurops. The study of this fossil assemblage presented here improves the knowledge of the ancient elasmobranchs that frequented the eastern Arabian coasts during the closure of the Neotethys and the birth of the Arabian Sea. A new species of softnose skate, Bathyraja chapmani n. sp., is described from two female specimens collected in deepwater off Lima, Peru (-11.933°, -78.15°), in the southeastern Pacific, at 1714 m deep, in a fishery for Patagonian toothfish. The new species is separated from other southeastern Pacific Bathyraja species by having the same color and same irregular spotted pattern on both the dorsal and ventral surfaces, as well as the form and arrangement of thorns and denticles and several meristic characters. Bathyraja chapmani n. sp. has a uniform black-to-chocolate or dark-plum-brown background coloration with an irregular pattern of whitish to darkcolored scattered spots. Its closest congener, Bathyraja aguja, also from Peru, has two symmetrically arranged, large, white spots on the dorsal surface at the pectoral-fin bases, symmetrically arranged white spots along the disc margins, and a ventral surface lacking any spots or patterning. Featured Application: Full-length cathepsin B is sorted to endo-lysosomes of colorectal carcinoma cells, while N-terminally truncated cathepsin B accumulates in aggresomes. Cathepsin B is an endo-lysosomal cysteine protease. However, its increased expression and altered localization to the extracellular space, to mitochondria, or to the nucleus has been linked to tumor progression. In the present study, we show enhanced levels of cathepsin B in adenocarcinoma tissue in comparison to adjacent normal colon. Additionally, cathepsin B was observed in the nuclear compartment of mucosal cells in adenocarcinoma tissue samples and in the nuclei of the colorectal carcinoma cell line HCT116. Accordingly, a distinct 40-kDa form of cathepsin B was detected in HCT116 cells, which is proposed to represent a specific form lacking the signal peptide and parts of the propeptide. Trafficking studies with an EGFP-tagged N-terminally truncated form, mimicking the 40-kDa form, demonstrated accumulation in aggresome-like inclusion bodies, while EGFP-tagged full-length cathepsin B revealed regular sorting to endo-lysosomes. We conclude that the identity of nuclear cathepsin B in colorectal adenocarcinoma (in situ) and in carcinoma cells (in vitro) cannot be attributed to either full-length or 40-kDa N-terminally truncated cathepsin B forms. Hence, future studies are needed to demonstrate which form/s of cathepsin B may be sorted to the nuclei of colorectal carcinoma cells, and whether redundant regulation of related cathepsin expression occurs. Interactions of the extracellular matrix (ECM) and cellular receptors constitute one of the crucial pathways involved in colorectal cancer progression and metastasis. With the use of bioinformatics analysis, we comprehensively evaluated the prognostic information concentrated in the genes from this pathway. First, we constructed a ECM–receptor regulatory network by integrating the transcription factor (TF) and 5'-isomiR interaction databases with mRNA/miRNA-seq data from The Cancer Genome Atlas Colon Adenocarcinoma (TCGA-COAD). Notably, one-third of interactions mediated by 5'-isomiRs was represented by noncanonical isomiRs (isomiRs, whose 5'-end sequence did not match with the canonical miRBase version). Then, exhaustive search-based feature selection was used to fit prognostic signatures composed of nodes from the network for overall survival prediction. Two reliable prognostic signatures were identified and validated on the independent The Cancer Genome Atlas Rectum Adenocarcinoma (TCGA-READ) cohort. The first signature was made up by six genes, directly involved in ECM–receptor interaction: AGRN, DAG1, FN1, ITGA5, THBS3, and TNC (concordance index 0.61, logrank test p = 0.0164, 3-years ROC AUC = 0.68). The second hybrid signature was composed of three regulators: hsa-miR-32-5p, NR1H2, and SNAI1 (concordance index 0.64, logrank test p = 0.0229, 3-years ROC AUC = 0.71). While hsa-miR-32-5p exclusively regulated ECM-related genes (COL1A2 and ITGA5), NR1H2 and SNAI1 also targeted other pathways (adhesion, cell cycle, and cell division). Concordant distributions of the respective risk scores across four stages of colorectal cancer and adjacent normal mucosa additionally confirmed reliability of the models. [ABSTRACT FROM AUTHOR]

Wood turtles (Glyptemys insculpta) are considered rare or threatened throughout their range. Populations in Iowa occur at the western periphery of the species' range and may be particularly vulnerable to extirpation due to low population density, geographic isolation with restricted opportunities for gene flow, a small range, and a high percentage of land that has been converted for agriculture. To improve our understanding of the ecological needs of the species and to provide targeted conservation of required habitat, determining home range size, site fidelity, and movement patterns is needed. We conducted a 2-yr radiotelemetry study on wood turtles in Iowa to provide baseline data on movement patterns (including estimated total distance moved in an active season and mean daily movement) and home range size and site fidelity between years of survey. Home range size of adult male and female wood turtles differed significantly for 100%, 95%, and 50% minimum convex polygon home range, with male mean home range being nearly 3 times the mean size of female home range. Stream home range length also differed significantly between males and females, with males utilizing a larger portion of lotic habitat. Both sexes showed a high degree of site fidelity to annual home ranges but not to specific overwintering locations. Our study provides important data on home range size, degree of site fidelity, and movement patterns of wood turtles from an isolated population in Iowa at the southwest periphery of the species' range. These data will inform conservation agencies on relevant habitat protection and management strategies of riparian areas that are necessary for the continued survival and protection of the species in the state. The purpose of this study was to determine the optimal imaging protocol for contrast‐enhanced computed tomography (CECT) using micro‐CT (μ‐CT) for the posterior cardinal vein (PCV), dorsal aorta (DA), hepatic portal vein (HPV), kidney, liver, cephalic arteries (CAs), and gills of Cloudy Catsharks Scyliorhinus torazame. Additionally, we examined the availability of CECT screening for the coelomic organs. Different doses of iopamidol (100, 300, 500, and 700 mg iodine [mgI]/kg) were administered intravenously for 20 s in six sharks. The CT scans from the pectoral girdle to the pelvic girdle were performed at 0–600 s after administration. Contrast‐enhanced CT imaging of the CAs, gills, and coelomic organs was examined. Assessment of the signal enhancement value revealed that the PCV was easily visualized with all contrast doses at 25 s. The CAs, gills, and DA were visible at a slightly higher dose (CAs and gills: 200 mgI/kg at 40 s; DA: 300 mgI/kg at 50 s). The HPV was obvious at a dose of at least 500 mgI/kg after a 150‐s delay. The parenchyma of the kidney had a contrast effect at 300 mgI/kg, 150 s after the contrast effect of the renal portal system disappeared. The liver, which stores a lot of lipids, had poor overall contrast enhancement that was optimized at the highest dose of 700 mgI/kg. Contrast‐enhanced CT screening at 700 mgI/kg and 150 s is likely to obtain the optimal imaging of the reproductive organs, such as the ovary, oviducal gland, uterus, and testis. Colorectal cancer (CRC) is the second cause of cancer-related death in both sexes worldwide. As pre-menopausal women are less likely to develop CRC compared to age-matched men, a protective role for estrogens has been hypothesized. Indeed, two isoforms of nuclear estrogen receptors (ER) have been described: ERβ and ERα. While the binding of 17beta-estradiol to ERβ activates antiapoptotic pathways, the interaction with ERα activates caspase-3, inducing apoptosis. In this regard, several pieces of evidence show that ERα tends to be under-regulated in advanced adenomas and CRC, with an opposite trend for ERβ. Furthermore, ERα stimulation slows adenomatous polyp growth and modulates relevant CRC pathways. Based on such considerations, dietary modulation of ER is promising, particularly in subjects with genetic predisposition for CRC. This review presents a synthesis of shark bioluminescence knowledge. Up to date, bioluminescent sharks are found only in Squaliformes, and specifically in Etmopteridae, Dalatiidae and Somniosidae families. The state-of-the-art knowledge about the evolution, ecological functions, histological structure, the associated squamation and physiological control of the photogenic organs of these elusive deep-sea sharks is presented. Special focus is given to their unique and singular hormonal luminescence control mechanism. In this context, the implication of the photophore-associated extraocular photoreception—which complements the visual adaptations of bioluminescent sharks to perceive residual downwelling light and luminescence in dim light environment—in the hormonally based luminescence control is depicted in detail. Similarities and differences between shark families are highlighted and support the hypothesis of an evolutionary unique ancestral appearance of luminescence in elasmobranchs. Data from the Integrated Tracking of Aquatic Animals in the Gulf of Mexico (iTAG) network, and sister networks, were used to evaluate fish movements in the Florida Keys—an extensive reef fish ecosystem just north of Cuba connecting the Atlantic Ocean and Gulf of Mexico. We analysed ~2 million detections for 23 species, ranging from reef fish such as Nassau grouper (Epinephelus striatus, Serranidae) to migratory apex predators such as white sharks (Carcharodon carcharias, Lamnidae). To facilitate comparisons across species, we used an eco‐evolutionary movement strategy framework that identified measurable movement traits and their proximate and ultimate drivers. Detectability was species‐specific and quantified with a detection potential index. Life stages detected in the study area varied by species and residency varied with life stage. Four annual movement types were identified as follows: high site‐fidelity residents, range residents, seasonal migrants and general migrants. The endangered smalltooth sawfish (Pristis pectinata, Pristidae), a seasonal migrant, exhibited the greatest within‐ecosystem connectivity. Site attachment, stopover and deep‐water migration behaviours differed between individuals, species and annual movement types. All apex predators were migratory. General migrants were significantly larger than fish in the other movement types, a life‐history and movement trait combination that is common but not exclusive, as many small pelagics also migrate. Most teleosts exhibited movements associated with spawning. As concerns grow over habitat and biodiversity loss, multispecies movescapes, such as presented here, are expected to play an increasingly important role in informing ecosystem‐based and non‐extractive fisheries management strategies. The Oligocene (Rupelian) Byram Formation (Vicksburg Group) in Alabama, USA, is divided into three members, including (in ascending order) the Glendon Limestone, unnamed marl, and the Bucatunna Clay. The Oligocene marine units in Alabama have been historically under-investigated, but bulk samples recently obtained from Glendon Limestone Member exposures at site AWa-9 in Washington County yielded 20 unequivocal elasmobranch and teleost taxa. This surprisingly diverse paleofauna, based on isolated teeth, bones and otoliths, includes the new taxon, Gobiosoma? axsmithi sp. nov., as well as "Aetomylaeus" sp., Albula sp., Aplodinotus gemma Koken, 1888, Ariosoma nonsector Nolf and Stringer, 2003, Balistidae indet., Citharichthys sp., Myliobatoidei indet., Diretmus? sp., Hemipristis sp., Negaprion aff. N. gilmorei (Leriche, 1942), Pachyscyllium sp., Paralbula sp., Physogaleus sp., Preophidion meyeri (Koken, 1888), Sciaenapseu-doradians (Dante and Frizzell in Frizzell and Dante, 1965), Sciaenops? sp., Sparus? elegantulus Koken, 1888, Sphyraena sp., and Syacium sp. Additional remains were recovered but could not be identified beyond undetermined Elasmobranchii or Teleostei. All these taxa represent first occurrences within the Glendon Limestone Member in Alabama, and the "Aetomylaeus" sp., Pachyscyllium sp., Paralbula sp., and Sciaenops? sp. specimens represent the first occurrences of each in the Oligocene of the Gulf Coastal Plain of the USA. We also report the first record of Oligocene Paralbula Blake, 1940 teeth, and the first occurrence of an Oligocene member of the Balistidae in the Western Hemisphere. This marine vertebrate assemblage indicates that the Glendon Limestone Member at site AWa-9 represented a subtropical to temperate, middle shelf paleoenviron-ment with a paleowater depth interpreted as 30-100 m. Biodiversity is vital to the welfare and survival of humans, but public support for conservation of most animal species is appallingly limited. Vertebrates make up less than 5% of the world's documented animal species, but are viewed far more sympathetically than invertebrates. This is because humans are empathetic with the appearance and behaviour of many of them, particularly the charismatic superstars like pandas and tigers that currently are the mainstays of biodiversity fundraising. Conversely, just as such attractive icons are effective ambassadors of biodiversity conservation, so certain detested and sometimes dangerous vertebrate pests have greatly compromised the public image of biodiversity. Some of these species, admittedly, are responsible for significant damage to health and economic welfare. Nevertheless, this paper shows that all play important ecological roles, they have compensating economic values, their harm has often been exaggerated, and their very negative public images are undeserved. This first installment deals with the most reviled 'lower' vertebrate species: sharks (representing fish); frogs and toads (representing amphibians); snakes (representing reptiles); and vultures (representing birds). The mechanisms underlying polyandry and female mate choice in certain taxonomic groups remain widely debated. In elasmobranchs, several species have shown varying rates of polyandry based on genetic studies of multiple paternity (MP). We investigated MP in the finetooth shark, Carcharhinus isodon, in order to directly test the encounter rate hypothesis (ERH), which predicts that MP is a result of the frequency of encounters between mature conspecifics during the breeding season, and should therefore increase when more time is available for copulation and sperm storage. Female finetooth sharks in the northern Gulf of Mexico (GoM) have been found to reproduce with both annual periodicity and biennial periodicity, while finetooth sharks from the northwestern Atlantic Ocean have only been found to reproduce biennially, allowing us to compare mating opportunity to frequency of MP. Our results show high rates of MP with no significant difference in frequency between females in the GoM (83.0%) and Atlantic (88.2%, p =.8718) and varying but nonsignificant rates of MP between females in the GoM reproducing annually (93.0%) and biennially (76.6%, p =.2760). While the ERH is not supported by this study, it remains possible that reproductive periodicity and other physiological factors play a role in determining rates of MP in elasmobranchs, with potential benefits to individuals and populations. The brown‐banded bamboo shark, Chiloscyllium punctatum, is the most common shark caught in coastal commercial fisheries throughout Southeast Asia, yet there is a lack of the life‐history information necessary for reliable stock assessments. The authors estimated growth rates and age at maturity using analysis of growth bands in vertebral centra. They trialled four different techniques to enhance the visibility and improve identification of the putative annual growth bands necessary for age estimation. The authors found that the burn method on whole vertebral centra provided the most readable and consistent results for age analysis. The logistic model was chosen as the best‐fit growth model for age estimation of 330 individual C. punctatum from Indonesia. Several age verification methods, including marginal increment ratio and length‐frequency analysis, were performed with the support of age validation through the use of calcein‐labelled vertebrae from two sharks maintained in captivity. This study found that C. punctatum from Indonesian waters is a fast‐growing species that can grow up to 18 cm year−1, reach an estimated maximum total length of 1 m, mature at c. 6.5 years and live for up to 14 years. In the present work, we examined the morphology, dimensions, cytochemical staining reactions and ultrastructure of blood cells from three freshwater stingray species, Potamotrygon wallacei, Potamotrygon motoro and Paratrygon aiereba, living in the waters of the middle Rio Negro basin (Barcelos, Amazonas, Brazil). We identified erythrocytes, erythroblasts, thrombocytes and four types of leukocytes (basophils, heterophils, lymphocytes and monocytes) in the blood of these stingray species. In all the freshwater stingray species studied, the shapes and dimensions of these cells were similar to those of marine elasmobranchs. Positive PAS staining occurred in heterophils and thrombocytes, and weak staining occurred in lymphocytes and monocytes, while metachromasia only occurred in basophils. Positive Sudan Black B staining was observed in thrombocytes and lymphocytes, and weak staining occurred in heterophils. Basophils and heterophils were the only cells with positive bromophenol blue staining, while no peroxidase staining was observed in any of the four leukocyte types. This is the first study to establish the dimensions and cytochemical staining profiles of blood cells in Amazonian stingray species. Because these elasmobranch species are exported as ornamental fish to countries worldwide, this study can contribute to establishing standards for blood constituents that may be helpful in assessing the health and welfare of these fish in artificial systems. Mustelus mustelus from the Langebaan Marine Protected Area (LMPA) has been largely protected from fishing activities for the past 30 years; therefore, its biology represents those of an unfished stock. The reproductive biology, diet, growth and maturity of M. mustelus was studied in the Langebaan Lagoon region in the south-western Cape Coast of South Africa between 2007 and 2009. In total, 217 sharks (females: 381-1734 mm total length (TL); males: 467-1267 mm TL) were examined. Sharks in the Bay represented the largest specimens among six regions, globally. Female parturition, ovulating and mating occurred between November and December after a gestation period of 10-11 months. No ontogenetic shift in diet was evident, with the diet predominantly consisting of the following three species of crustaceans; Hymenosoma orbiculare, Upogebia africana and Callichirus kraussi. The von Bertalanffy growth parameters for combined sexes were L∞ = 1594 mm TL, k = 0.15 year-1, t0 = -2.01 year (n = 95). Possible age over-estimation was investigated using micro-computed tomography, which highlighted the existence of false check marks. This study indicated that the LMPA represents a pupping, nursery and feeding ground as well as a refuge area for the largest and oldest recorded smoothhound sharks globally, emphasising its importance in spatial conservation of this species. Stock-wide trends in fish relative abundance are challenging to obtain when a single, comprehensive survey is unavailable, and multiple, spatially, and/or temporally fragmented surveys must be relied upon instead. Indices of abundance from multiple surveys frequently have differing trends, resulting in obscured true abundance patterns of the resource. We use an age-structured simulation model of two coastal shark species in the southeast United States to explore the performance of dynamic factor analysis (DFA) for reconciling multiple indices of abundance that are in conflict. Survey-specific time-variation in catchability was induced to generate conflicting indices of abundance. Key simulation sensitivities included survey variability, abundance pattern in the resource, and missing years of survey data. We caution against using DFA when there is no contrast in the underlying stock abundance or when trends in catchability in all surveys result in no survey that is representative of stock abundance. When multiple representative surveys were available, DFA proved useful across species in estimating stock-wide trends from conflicting survey indices with different selectivities, catchabilities, variances, and, to a lesser extent, with missing data. Our results suggest that resolving contrasting patterns among multiple time-series of relative abundance can improve understanding of the temporal trend in stock abundance. [ABSTRACT FROM AUTHOR]

Dramatic declines in reef shark populations have been documented worldwide in response to human activities. Marine Protected Areas (MPAs) offer a useful mechanism to protect these species and their roles in coral reef ecosystems. The effectiveness of MPAs notably relies on compliance together with sufficient size to encompass animal home range. Here, we measured home range of 147 grey reef sharks, Carcharhinus amblyrhynchos, using acoustic telemetry in New Caledonia. The distribution of home range was then compared to local MPA sizes. We report a home range of 12 km2 of reef for the species with strong differences between adult males (21 km2), adult females (4.4 km2) and juveniles (6.2 km2 for males, 2.7 km2 for females). Whereas local historic MPA size seemed adequate to protect reef shark home range in general, these were clearly too small when considering adult males only, which is consistent with the reported failure of MPAs to protect sharks in New Caledonia. Fortunately, the recent implementation of several orders of magnitude larger MPAs in New Caledonia and abroad show that recent Indo-Pacific MPAs are now sufficiently large to protect the home ranges of this species, including males, across its geographical range. A suite of blood chemistry parameters (including acid–base indicators and plasma electrolytes) was serially measured in blacktip sharks (Carcharhinus limbatus), captured via rod-and-reel, to gain a more thorough understanding of the physiological stress response to recreational capture. Sharks were caught both from the shore and from fishing vessels and experienced varying degrees of air exposure during handling. While all captured sharks exhibited a metabolic acidosis during the fight on the line (increasing lactate and decreasing pH and bicarbonate), the observed acidosis was compounded by a respiratory component (increasing pCO2) in sharks removed from the water during handling. Vessel-caught sharks handled in the water exhibited significantly greater increases in lactate and glucose (0.73 ± 0.21 mmoll−1 min−1 and 0.81 ± 1.07 mg dl−1 min−1, respectively) than sharks handled out of water (0.21 ± 0.17 mmoll−1 min−1 and −0.32 ± 1.05 mg dl−1 min−1; p  <   0.001 and p  <   0.05, respectively). These findings provide insights into how differences in recreational capture methods and air exposure can mediate the origin and magnitude of capture-related stress and highlight the importance of considering both sampling time (time from capture to phlebotomy) and sampling location (in water vs. out) in studies conducted on capture-related stress. The present study deals with length-weight relationship of Scoliodon laticaudus from Dakshina Kannada and Udupi coast during September 2015 to May 2017. The regression co-efficient of female was found to be higher than male. From this observation it may be opnied that female gained more weight with increase in length compared to male. The corresponding equations for both the sexes were derived during September 2015 - May 2016 the corresponding equations were; W=0.00986 L2.7674 for male, W=0.0084 L2.8129 for female and pooled data was obtained at W=0.00914L2.7802. Likewise, during September 2016 to May 2017 the equations obtained for male W= 0.02138 L2.5826, for female W = 0.01972 L2.7249 and pooled value W= 0.02055 L2.6585. Analysis of the covariance indicated that there is no significant difference in the length-weight relationship between male and female. The data on seasonal variation in condition factor (Kn) of both male and female during both the years of study showed that the values were more or less similar in both the sexes, indicating almost identical metabolic activity in male and female which may be attributed to gonadal development and feeding intensity. Uterine milk is secreted in the uterus for embryo nutrition in several elasmobranch species and may contribute to rapid embryonic growth, but the details of its composition and its functions are poorly understood. In this study, to explore the roles of uterine milk for embryos, its components throughout the gestational period were analysed in detail. Uterine milk was collected from pregnant red stingrays (Hemitrygon akajei) in the early, middle and late gestational periods, respectively (n= 3 for each period). The crude composition, constituent proteins and fatty acids in the milk were analysed. The uterine milk was rich in proteins throughout the gestational period, whereas lipids dramatically increased in the middle period and reduced slightly towards the late period. Some proteins potentially associated with nutrition, cartilage growth and embryonic immunity were found. Several enzymes related to central metabolism were also detected. The constituent fatty acids in the middle and late periods were similar to those in the egg yolks of elasmobranchs, except for C18:2, which was rich only in the uterine milk. The most abundant fatty acid in the milk was C16:1, which could function as a lipokine to promote lipid metabolism in the embryo. This study's data suggest that uterine milk may be secreted in addition to the egg yolk in elasmobranchs to support rapid and healthy embryonic growth. [ABSTRACT FROM AUTHOR]

The brown-banded bamboo shark, Chiloscyllium punctatum, is one of the most common sharks caught in coastal fisheries across Southeast Asia; however, its small size and low economic value typically exclude it from the attention of fisheries managers. As part of an assessment of a potential role for this shark in improving the sustainability of coastal shark fisheries in Indonesia, we investigated its reproductive biology from waters around the Seribu Islands on the basis of monthly sampling from a fish landing in Tanjung Pasir, Banten. Females have fully functional uteri, but asymmetric ovaries, with only the right ovary being functional and the left ovary being atrophied. The species is reproductively active throughout the year, with the optimum period for males to copulate being from June to August, and the optimum period for egg-laying in females from July to January on the basis of their physiological condition. Females mature at a larger size than do males, 704- and 653-mm total length respectively. To allow a continuous recruitment flow to the population, and to facilitate compliance and monitoring, we recommend that fishers release all brown-banded bamboo sharks greater than 700 mm in total length. Summary: The objective of this study was to determinate the influence of carrier addition and spray drying temperatures (160 or 200°C) on physicochemical properties of spray‐dried carrot juice powder. As carriers, maltodextrin and gum Arabic in different proportions were used. In powders were determined the following: moisture content, water activity, apparent density, size of particles, colour and carotenoids content. The content of carotenoids in powders prepared from solutions with juice to carrier ratio 2:3 was 324.3–406.6 mg/kg d.m., while in powders with a J:C = 3:2 values ranged from 576.7 to 637.7 mg/kg d.m. The highest content of carotenoids and the most yellow colour were observed in powders with 1:3 gum Arabic to maltodextrin ratio. Powders with the highest amount of gum Arabic have the highest diameter and the lowest water activity. High inlet temperature caused brighter colour of powders, higher dry matter content, lower water activity and apparent density of powders. [ABSTRACT FROM AUTHOR]

Several shark species produce egg cases as protective casings in which their embryos develop. These casings are composed of multiple layers of collagen and are extremely durable, making them prone to fossilisation. Here we document Palaeoxyris (Spirangium) ‒ fossil shark egg cases from Lower Jurassic successions of southern Sweden. We present high-resolution 3D images of Palaeoxyris based on microfocus X-ray computed tomography (μCT) of seven specimens, including fossils that were embedded within a sandstone matrix. Our examination of the internal structure of the egg cases revealed the possible remnants of a yolk and foetus in one specimen. The cases were most likely produced by hybodont sharks, as outlined in previous studies, and the occurrence of hybodont teeth from Lower Jurassic successions of Sweden support this. Palynological analysis of the matrix from one of the specimens hosting Palaeoxyris, indicates an early Hettangian age. The high percentage of spores (c. 60%) reveals that the egg cases were laid during the Transitional Spore Spike Interval following the end-Triassic mass extinction. The egg cases are found in conjunction with fossil horsetails; with the broader palynological and sedimentological evidence, this suggests an estuarine depositional setting, and potentially indicates that newborn sharks were living in habitats comparable to modern mangroves, as is often the case today. Sorting nexins (SNXs) are a diverse group of cytoplasmic‐ and membrane‐associated phosphoinositide‐binding proteins containing the PX domain proteins. The function of SNX proteins in regulating intracellular protein trafficking consists of endocytosis, endosomal sorting, and endosomal signaling. Dysfunctions of SNX proteins are demonstrated to be involved in several cancerous/neoplastic diseases. Here, we review the accumulated evidence of the molecular structure and biological function of SNX proteins and discuss the regulatory role of SNX proteins in distinct cancerous/neoplastic diseases. Understanding mating systems is a pillar of behavioural ecology, placing the complex interactions between females and males into a reproductive context. The field of multiple paternity, the phenomenon whereby many sires contribute to an individual litter, has traditionally viewed females as passive players in a male–male competitive framework. With the emergence of feminist perspectives in ecological fields, novel alternative mechanisms and evolutionary theories across invertebrate and vertebrate taxa recognize females are active stakeholders in the reproductive process. Despite their evolutionary significance, ecological diversity and myriad reproductive modes, elasmobranch (sharks, skates and rays) research lags behind other fields regarding complex biological processes, such as multiple paternity which is often ascribed to convenience polyandry. Here, we layout hypotheses and resynthesize multiple paternity literature from a female and life history perspective to highlight how alternative mechanisms influence the predominance of multiple paternity across elasmobranchs. We draw upon parallels in other invertebrate and vertebrate taxa to demonstrate how female elasmobranchs can influence multiple paternity outcomes that benefit their reproductive success. Our article challenges dogma that has resulted from years of dismissing the female perspective as important and provides a framework for future advancement using more holistic approaches to studying mating systems. Intestinal epithelial cells (IECs) are the first to encounter luminal antigens and play an active role in intestinal immune responses. We previously reported that β‐glucans, major fungal cell‐wall glycans, induced chemokine secretion by IEC lines in a Dectin‐1‐ and Syk‐dependent manner. Here, we show that in contrast to β‐glucans, stimulation of IEC lines with Candida albicans and Saccharomyces cerevisiae did not induce secretion of any of the proinflammatory cytokines IL‐8, CCL2, CXCL1, and GM‐CSF. Commensal fungi and β‐glucans activated Syk and ERK in IEC lines. However, only β‐glucans activated p38, JNK, and the transcription factors NF‐κB p65 and c‐JUN, which were necessary for cytokine secretion. Furthermore, costimulation of IEC lines with β‐glucans and C. albicans yielded decreased cytokine secretion compared to stimulation with β‐glucans alone. Finally, ex vivo stimulation of human colonic mucosal explants with zymosan and C. albicans, leads to epithelial Syk and ERK phosphorylation, implying recognition of fungi and similar initial signaling pathways as in IEC lines. Lack of cytokine secretion in response to commensal fungi may reflect IECs' response to fungal glycans, other than β‐glucans, that contribute to mucosal tolerance. Our study sought to analyze patterns and overlap in prey items consumed by elasmobranch species in southern Brazil through a complex network analysis. We hypothesized that species with the same habitat use would show greater trophic niche overlap and lower diet specialization than species with different habitat use. We conducted a literature survey of the species recorded in southern Brazil and analyzed publications with quantitative data on the diet of each species. We found records of 103 species in the region, of which 58 had their diets analyzed and had quantitative data to contribute to this analysis. The results suggest nested diet patterns in this community. Nestedness may arise for different reasons, and suggests that species might be balancing their high feeding niche overlap through compensatory mechanisms. There are no prior records of such a pattern in elasmobranch communities, which brings to light important information about their feeding niche. Data on prey consumption and temporal and spatial patterns of elasmobranch feeding may yield further insight into how these species are interacting in the environment and assist the investigation of the processes that shaped the current community's trophic structure. A wide array of technologies are available for gaining insight into the movement of wild aquatic animals. Although acoustic telemetry can lack the fine‐scale spatial resolution of some satellite tracking technologies, the substantially longer battery life can yield important long‐term data on individual behavior and movement for low per‐unit cost. Typically, however, receiver arrays are designed to maximize spatial coverage at the cost of positional accuracy leading to potentially longer detection gaps as individuals move out of range between monitored locations. This is particularly true when these technologies are deployed to monitor species in hard‐to‐access locations.Here, we develop a novel approach to analyzing acoustic telemetry data, using the timing and duration of gaps between animal detections to infer different behaviors. Using the durations between detections at the same and different receiver locations (i.e., detection gaps), we classify behaviors into "restricted" or potential wider "out‐of‐range" movements synonymous with longer distance dispersal. We apply this method to investigate spatial and temporal segregation of inferred movement patterns in two sympatric species of reef shark within a large, remote, marine protected area (MPA). Response variables were generated using network analysis, and drivers of these movements were identified using generalized linear mixed models and multimodel inference.Species, diel period, and season were significant predictors of "out‐of‐range" movements. Silvertip sharks were overall more likely to undertake "out‐of‐range" movements, compared with gray reef sharks, indicating spatial segregation, and corroborating previous stable isotope work between these two species. High individual variability in "out‐of‐range" movements in both species was also identified.We present a novel gap analysis of telemetry data to help infer differential movement and space use patterns where acoustic coverage is imperfect and other tracking methods are impractical at scale. In remote locations, inference may be the best available tool and this approach shows that acoustic telemetry gap analysis can be used for comparative studies in fish ecology, or combined with other research techniques to better understand functional mechanisms driving behavior.
