Abstract

We present angular diameter distance measurements obtained by locating the baryon acoustic oscillations (BAO) scale in the distribution of galaxies selected from the first year of Dark Energy Survey data. We consider a sample of over 1.3 million galaxies distributed over a footprint of 1336 deg2 with 0.6 < |$z$| photo < 1 and a typical redshift uncertainty of 0.03(1 + |$z$|⁠). This sample was selected, as fully described in a companion paper, using a colour/magnitude selection that optimizes trade-offs between number density and redshift uncertainty. We investigate the BAO signal in the projected clustering using three conventions, the angular separation, the comoving transverse separation, and spherical harmonics. Further, we compare results obtained from template-based and machine-learning photometric redshift determinations. We use 1800 simulations that approximate our sample in order to produce covariance matrices and allow us to validate our distance scale measurement methodology. We measure the angular diameter distance, D A, at the effective redshift of our sample divided by the true physical scale of the BAO feature, r d. We obtain close to a 4 per cent distance measurement of D A(⁠|$z$| eff = 0.81)/ r d = 10.75 ± 0.43. These results are consistent with the flat Λ cold dark matter concordance cosmological model supported by numerous other recent experimental results. [ABSTRACT FROM AUTHOR]

Abstract

In the study of visual perception, the use of computerized commercial programs for the presentation of stimuli, usually configured to admit a response for each stimulus presented, is common. However, the study of multistable perception requires the presentation of a stimulus that remains unchanged, although the perception of it can vary in two or more configurations during its observation, which demands that the program admit responses for each change in perception of the same stimulus. PercepFiguras is a system developed with C++ programming language that allows: a) present images and receive multiple responses to the same stimulus, b) synchronize the responses with an electroencephalographic recording system by sending TTL pulses, c) generate a behavioral file, d) generate a file with the total responses and averages of the latencies for each type of figure. In the pilot study with a task of continuous presentation of an ambiguous figure to alternating with two variants of less ambiguity, we observe greater frequency of perceptual and less perceptual stability to the ambiguous figure with respect to the less ambiguous figures, which proves the usefulness of the program. Abstract

La Artritis Reumatoide es una enfermedad de etiología multifactorial, que involucra la presencia de factores genéticos, ambientales, inmunológicos y hormonales. En los últimos años se ha establecido una asociación entre la predisposición a padecer esta enfermedad y la existencia de determinados haplotipos del HLA clase II. El objetivo fundamental de este trabajo es la caracterización del polimorfismo de las moléculas HLA-DQB1 y HLA-DRB de un grupo de pacientes cubanos con Artritis Reumatoide, además analizar una posible correlación entre los niveles de varias citocinas proinflamatorias en un grupo de estos pacientes, con los alelos HLA tipo II genotipados, el sexo y el tiempo de diagnosticada la enfermedad. El estudio se llevo a cabo en 50 pacientes cubanos con el diagnóstico de Artritis Reumatoide y un grupo control, compuesto de 211 donantes sanos. Los haplotipos HLA-DQ y HLA-DRB1 fueron determinados a través de la reacción en cadena de la polimerasa. La cuantificación de las citocinas se realizó empleando inmuno-ensayos comerciales. Los resultados obtenidos en este análisis indican que los alelos con un radio de la razón de probabilidades superior a 2 fueron para el caso de la molécula HLA-DQB1: HLADQB1*03 y *06, y para la molécula HLA-DRB1 los alelos: *01, *04, *09 y *10. Encontramos además que los niveles de la citocina interferón gamma están significativamente aumentados en los pacientes con menos tiempo de diagnosticada la enfermedad. Este trabajo constituye el primer reporte de caracterización de las moléculas HLA tipo II, a través de técnicas moleculares, en pacientes cubanos con Artritis Reumatoide.

Abstract

Some procedures for characterising the mechanical properties of femur diaphysis are reviewed here. We have used the three point bending test to measure the relative rupture modulus of ancient healthy human tissues (1250, 800, 614, and 185 years BP) as well as recent bones. The maximum resistance to fracture was measured applying a force (by a wedge) over the femoral inner surface. The maximum rupture strength was about 150 MPa for recent bone and decreased as the antiquity increased. The typical anisotropy that is observed in this kind of tissues is due to the anisotropical orientation of fibres as well as the textured orientation of the apatite crystals over the collagen fibres. Therefore we found that ancient bones show less fracture strength probably due to an abiotic crystal growth phenomenon during the diagenesis process. By LVSEM analysis we have found that in recent samples the fracture surface is irregular due to the crosslinking interactions between the collagen molecules, in comparison with the ancient samples, where a smooth surface is clearly appreciated as the antiquity of the sample increases. The results reported here strongly suggest that these composites should contain a fibrillar phase as a matrix constituted mainly by a natural polymer (i.e. collagen, cellulose, etc.). Abstract

The EGF promotes inhibition of cell proliferation in vitro and in vivo models depending on its concentration, application schema and the type of tumor cells on which it acts. Our research hypothesis was based on the fact that the EGF varies the expression of genes involved in a negative regulation of tumor cell lines proliferation carrying high levels of its receptor (EGFR). Our objectives were, to obtain information about the effect of EGF on tumor cell proliferation in vitro and in vivo models and, know the gene expression patterns of a group of genes involved in cancer signaling pathways and EGFR. The results showed that EGF at nanomolar concentrations inhibits the tumor cells proliferation bearing high levels of EGFR and, promotes the survival of treated animals, establishing a direct relationship between the inhibition of cell proliferation, high concentrations of EGF and, high amount of EGFR in the cells. The differential gene expression profile showed a variation in a group of genes which exert a powerful control over the cell cycle progression, gene transcription and apoptosis. It was concluded that the inhibition of tumor cell proliferation by the action of EGF is due to activation of molecular mechanisms controlling cell cycle progression. This work won the Annual Award of the Cuban Academy of Sciences in 2012.

Abstract

We study the effect of magnification in the Dark Energy Survey Year 3 analysis of galaxy clustering and galaxy-galaxy lensing, using two different lens samples: a sample of Luminous red galaxies, redMaGiC, and a sample with a redshift-dependent magnitude limit, MagLim. We account for the effect of magnification on both the flux and size selection of galaxies, accounting for systematic effects using the Balrog image simulations. We estimate the impact of magnification on the galaxy clustering and galaxy-galaxy lensing cosmology analysis, finding it to be a significant systematic for the MagLim sample. We show cosmological constraints from the galaxy clustering auto-correlation and galaxy-galaxy lensing signal with different magnifications priors, finding broad consistency in cosmological parameters in $\Lambda$CDM and $w$CDM. However, when magnification bias amplitude is allowed to be free, we find the two-point correlations functions prefer a different amplitude to the fiducial input derived from the image simulations. We validate the magnification analysis by comparing the cross-clustering between lens bins with the prediction from the baseline analysis, which uses only the auto-correlation of the lens bins, indicating systematics other than magnification may be the cause of the discrepancy. Abstract

The existence of multiple subclasses of type Ia supernovae (SNeIa) has been the subject of great debate in the last decade. One major challenge inevitably met when trying to infer the existence of one or more subclasses is the time consuming, and subjective, process of subclass definition. In this work, we show how machine learning tools facilitate the automatic discovery of sub-populations of SNIa; to that end we introduce the DRACULA Python package (Dimensionality Reduction And Clustering for Unsupervised Learning in Astronomy). Our approach is divided in three steps: (i) Transfer Learning, which takes advantage of all available spectra (even those without an epoch estimate) as an information source, (ii) dimensionality reduction through Deep Learning and (iii) unsupervised learning (clustering) using K-Means. Results match a previously suggested classification scheme, showing that the proposed method is able to grasp the main spectral features behind the definition of such subclasses. Moreover, our methodology is capable of automatically identifying a hierarchical structure of spectral features. This allows the confirmation of the velocity of lines as a first order effect in the determination of SNIa sub-classes, followed by 91bg-like events. In this context, SNIa spectra are described by a space of 4 dimensions + 1 for the time evolution of objects. We interpreted this as evidence that the progenitor system and the explosion mechanism should be described by a small number of initial physical parameters. Given the expected data deluge in the forthcoming years, our proposed approach is essential to allow a quick and statistically coherent identification of subclasses (and outliers). Abstract

We present cosmological constraints from the analysis of two-point correlation functions between galaxy positions and galaxy lensing measured in Dark Energy Survey (DES) Year 3 data and measurements of cosmic microwave background (CMB) lensing from the South Pole Telescope (SPT) and Planck. When jointly analyzing the DES-only two-point functions and the DES cross-correlations with SPT+Planck CMB lensing, we find $\Omega_{\rm m} = 0.344\pm 0.030$ and $S_8 \equiv \sigma_8 (\Omega_{\rm m}/0.3)^{0.5} = 0.773\pm 0.016$, assuming $\Lambda$CDM. When additionally combining with measurements of the CMB lensing autospectrum, we find $\Omega_{\rm m} = 0.306^{+0.018}_{-0.021}$ and $S_8 = 0.792\pm 0.012$. The high signal-to-noise of the CMB lensing cross-correlations enables several powerful consistency tests of these results, including comparisons with constraints derived from cross-correlations only, and comparisons designed to test the robustness of the galaxy lensing and clustering measurements from DES. Applying these tests to our measurements, we find no evidence of significant biases in the baseline cosmological constraints from the DES-only analyses or from the joint analyses with CMB lensing cross-correlations. However, the CMB lensing cross-correlations suggest possible problems with the correlation function measurements using alternative lens galaxy samples, in particular the redMaGiC galaxies and high-redshift MagLim galaxies, consistent with the findings of previous studies. Abstract

Joint analyses of cross-correlations between measurements of galaxy positions, galaxy lensing, and lensing of the cosmic microwave background (CMB) offer powerful constraints on the large-scale structure of the Universe. In a forthcoming analysis, we will present cosmological constraints from the analysis of such cross-correlations measured using Year 3 data from the Dark Energy Survey (DES), and CMB data from the South Pole Telescope (SPT) and Planck. Here we present two key ingredients of this analysis: (1) an improved CMB lensing map in the SPT-SZ survey footprint and (2) the analysis methodology that will be used to extract cosmological information from the cross-correlation measurements. Relative to previous lensing maps made from the same CMB observations, we have implemented techniques to remove contamination from the thermal Sunyaev Zel'dovich effect, enabling the extraction of cosmological information from smaller angular scales of the cross-correlation measurements than in previous analyses with DES Year 1 data. We describe our model for the cross-correlations between these maps and DES data, and validate our modeling choices to demonstrate the robustness of our analysis. We then forecast the expected cosmological constraints from the galaxy survey-CMB lensing auto and cross-correlations. We find that the galaxy-CMB lensing and galaxy shear-CMB lensing correlations will on their own provide a pffiffiffiffiffiffiffiffiffiffiffiffiffifficonstraint on S8 1/4 sigma 8 omega m=0.3 at the few percent level, providing a powerful consistency check for the DES-only constraints. We explore scenarios where external priors on shear calibration are removed, finding that the joint analysis of CMB lensing cross-correlations can provide constraints on the shear calibration amplitude at the 5% to 10% level.

Abstract

Recent cosmological analyses with large-scale structure and weak lensing measurements, usually referred to as 3$\times$2pt, had to discard a lot of signal-to-noise from small scales due to our inability to accurately model non-linearities and baryonic effects. Galaxy-galaxy lensing, or the position-shear correlation between lens and source galaxies, is one of the three two-point correlation functions that are included in such analyses, usually estimated with the mean tangential shear. However, tangential shear measurements at a given angular scale $\theta$ or physical scale $R$ carry information from all scales below that, forcing the scale cuts applied in real data to be significantly larger than the scale at which theoretical uncertainties become problematic. Recently there have been a few independent efforts that aim to mitigate the non-locality of the galaxy-galaxy lensing signal. Here we perform a comparison of the different methods, including the Y-transformation, the Point-Mass marginalization methodology and the Annular Differential Surface Density statistic. We do the comparison at the cosmological constraints level in a combined galaxy clustering and galaxy-galaxy lensing analysis. We find that all the estimators yield equivalent cosmological results assuming a simulated Rubin Observatory Legacy Survey of Space and Time (LSST) Year 1 like setup and also when applied to DES Y3 data. With the LSST Y1 setup, we find that the mitigation schemes yield $\sim$1.3 times more constraining $S_8$ results than applying larger scale cuts without using any mitigation scheme., Comment: 11+4 pages, 4+4 figures. Abstract

Cross-correlations of galaxy positions and galaxy shears with maps of gravitational lensing of the cosmic microwave background (CMB) are sensitive to the distribution of large-scale structure in the Universe. Such cross-correlations are also expected to be immune to some of the systematic effects that complicate correlation measurements internal to galaxy surveys. We present measurements and modeling of the cross-correlations between galaxy positions and galaxy lensing measured in the first three years of data from the Dark Energy Survey with CMB lensing maps derived from a combination of data from the 2500 deg$^2$ SPT-SZ survey conducted with the South Pole Telescope and full-sky data from the Planck satellite. The CMB lensing maps used in this analysis have been constructed in a way that minimizes biases from the thermal Sunyaev Zel'dovich effect, making them well suited for cross-correlation studies. The total signal-to-noise of the cross-correlation measurements is 23.9 (25.7) when using a choice of angular scales optimized for a linear (nonlinear) galaxy bias model. We use the cross-correlation measurements to obtain constraints on cosmological parameters. For our fiducial galaxy sample, which consist of four bins of magnitude-selected galaxies, we find constraints of $\Omega_{m} = 0.272^{+0.032}_{-0.052}$ and $S_{8} \equiv \sigma_8 \sqrt{\Omega_{m}/0.3}= 0.736^{+0.032}_{-0.028}$ ($\Omega_{m} = 0.245^{+0.026}_{-0.044}$ and $S_{8} = 0.734^{+0.035}_{-0.028}$) when assuming linear (nonlinear) galaxy bias in our modeling. Considering only the cross-correlation of galaxy shear with CMB lensing, we find $\Omega_{m} = 0.270^{+0.043}_{-0.061}$ and $S_{8} = 0.740^{+0.034}_{-0.029}$. Abstract

The search for neutrinoless double beta decay of nuclei is believed to be one of the most promising means to search for new physics. Observation of this very rare nuclear process, which violates Lepton Number conservation, would imply the neutrino sector has a Majorana mass component and may also provide an explanation for the universe matter-antimatter asymmetry of the universe. In the case where a heavy intermediate particle is exchanged in this process, QCD contributions from short range interactions become relevant and the calculation of matrix elements with four-quark operators becomes necessary. In these proceedings we will discuss our current progress in the calculation of these four-quark operators from LQCD.

Abstract

Rosell, A., et al. (DES Collaboration), In this paper, we present and validate the galaxy sample used for the analysis of the baryon acoustic oscillation (BAO) signal in the Dark Energy Survey (DES) Y3 data. The definition is based on a colour and redshift-dependent magnitude cut optimized to select galaxies at redshifts higher than 0.5, while ensuring a high-quality determination. The sample covers ~4100 deg2 to a depth of i = 22.3 (AB) at 10s. It contains 7031 993 galaxies in the redshift range from z = 0.6 to 1.1, with a mean effective redshift of 0.835. Redshifts are estimated with the machine learning algorithm DNF, and are validated using the VIPERS PDR2 sample. We find a mean redshift bias of zbias~0.01 and a mean uncertainty, in units of 1 + z, of σ68~0.03. We evaluate the galaxy population of the sample, showing it is mostly built upon Elliptical to Sbc types. Furthermore, we find a low level of stellar contamination of ≤ 4 per cent. We present the method used to mitigate the effect of spurious clustering coming from observing conditions and other large-scale systematics.We apply it to the BAO sample and calculate weights that are used to get a robust estimate of the galaxy clustering signal. This paper is one of a series dedicated to the analysis of the BAO signal in DES Y3. Abstract

We perform a cosmic shear analysis in harmonic space using the first year of data collected by the Dark Energy Survey (DES-Y1). We measure the cosmic weak lensing shear power spectra using the Metacalibration catalogue and perform a likelihood analysis within the framework of CosmoSIS. We set scale cuts based on baryonic effects contamination and model redshift and shear calibration uncertainties as well as intrinsic alignments. We adopt as fiducial covariance matrix an analytical computation accounting for the mask geometry in the Gaussian term, including non-Gaussian contributions. A suite of 1200 lognormal simulations is used to validate the harmonic space pipeline and the covariance matrix. We perform a series of stress tests to gauge the robustness of the harmonic space analysis. Finally, we use the DES-Y1 pipeline in configuration space to perform a similar likelihood analysis and compare both results, demonstrating their compatibility in estimating the cosmological parameters $S_8$, $\sigma_8$ and $\Omega_m$. The methods implemented and validated in this paper will allow us to perform a consistent harmonic space analysis in the upcoming DES data., Comment: 16 pages, 11 figures. (DES Collaboration), We perform an analysis in harmonic space of the Dark Energy Survey Year 1 Data (DES-Y1) galaxy clustering photometric data using products obtained for the real-space analysis. We test our pipeline with a suite of lognormal simulations, which are used to validate scale cuts in harmonic space as well as to provide a covariance matrix that takes into account the DES-Y1 mask. We then apply this pipeline to DES-Y1 data taking into account survey property maps derived for the real-space analysis. We compare with real-space DES-Y1 results obtained from a similar pipeline. We show that the harmonic space analysis we develop yields results that are compatible with the real-space analysis for the bias parameters. Abstract

Recent cosmic shear studies have reported discrepancies of up to $1\sigma$ on the parameter ${S_{8}=\sigma_{8}\sqrt{\Omega_{\rm m}/0.3}}$ between the analysis of shear power spectra and two-point correlation functions, derived from the same shear catalogs. It is not a priori clear whether the measured discrepancies are consistent with statistical fluctuations. In this paper, we investigate this issue in the context of the forthcoming analyses from the third year data of the Dark Energy Survey (DES-Y3). We analyze DES-Y3 mock catalogs from Gaussian simulations with a fast and accurate importance sampling pipeline. We show that the methodology for determining matching scale cuts in harmonic and real space is the key factor that contributes to the scatter between constraints derived from the two statistics. We compare the published scales cuts of the KiDS, Subaru-HSC and DES surveys, and find that the correlation coefficients of posterior means range from over 80% for our proposed cuts, down to 10% for cuts used in the literature. We then study the interaction between scale cuts and systematic uncertainties arising from multiple sources: non-linear power spectrum, baryonic feedback, intrinsic alignments, uncertainties in the point-spread function, and redshift distributions. We find that, given DES-Y3 characteristics and proposed cuts, these uncertainties affect the two statistics similarly; the differential biases are below a third of the statistical uncertainty, with the largest biases arising from intrinsic alignment and baryonic feedback. Friedrich, O., et al. DES Collaboration, We describe and test the fiducial covariance matrix model for the combined two-point function analysis of the Dark Energy Survey Year 3 (DES-Y3) data set. Using a variety of new ansatzes for covariance modelling and testing, we validate the assumptions and approximations of this model. These include the assumption of Gaussian likelihood, the trispectrum contribution to the covariance, the impact of evaluating the model at a wrong set of parameters, the impact of masking and survey geometry, deviations from Poissonian shot noise, galaxy weighting schemes, and other sub-dominant effects. We find that our covariance model is robust and that its approximations have little impact on goodness of fit and parameter estimation. The largest impact on best-fitting figure-of-merit arises from the so-called fsky approximation for dealing with finite survey area, which on average increases the χ2 between maximum posterior model and measurement by 3.7 per cent (Δχ2 ≈ 18.9). Standard methods to go beyond this approximation fail for DES-Y3, but we derive an approximate scheme to deal with these features. For parameter estimation, our ignorance of the exact parameters at which to evaluate our covariance model causes the dominant effect. We find that it increases the scatter of maximum posterior values for Ωm and σ8 by about 3 per cent and for the dark energy equation-of-state parameter by about 5 per cent⁠., The DES data management system is supported by the National Science Foundation under grant numbers AST-1138766 and AST-1536171. 138. Comparative Genomic Analyses of Colletotrichum lindemuthianum Pathotypes with Different Virulence Levels and Lifestyles.

Abstract

Colletotrichum lindemuthianum is the most frequent pathogenic fungus of the common bean Phaseolus vulgaris . This filamentous fungus employs a hemibiotrophic nutrition/infection strategy, which is characteristic of many Colletotrichum species. Due to host-pathogen coevolution, C. lindemuthianum includes pathotypes with a diversity of virulence against differential common bean varieties. In this study, we performed comparative genomic analyses on three pathotypes with different virulence levels and a non-pathogenic pathotype, isolated from different geographical areas in Mexico. Our results revealed large genomes with high transposable element contents that have undergone expansions, generating intraspecific diversity. All the pathotypes exhibited a similar number of clusters of orthologous genes (COGs) and Gene Ontology (GO) terms. TFomes contain families that are typical in fungal genomes; however, they show different contents between pathotypes, mainly in transcription factors with the fungal-specific TF and Zn2Cys6 domains. Peptidase families mainly contain abundant serine peptidases, metallopeptidases, and cysteine peptidases. In the secretomes, the number of genes differed between the pathotypes, with a high percentage of candidate effectors. Both the virulence gene and CAZyme gene content for each pathotype was abundant and diverse, and the latter was enriched in hemicellulolytic enzymes. We provide new insights into the nature of intraspecific diversity among C. lindemuthianum pathotypes and the origin of their ability to rapidly adapt to genetic changes in its host and environmental conditions.

Abstract

Antibiotic resistance has emerged as a global threat to public health, generating a growing interest in investigating the presence of antibiotic-resistant bacteria in environments influenced by anthropogenic activities. Wastewater treatment plants in hospital serve as significant reservoirs of antimicrobial-resistant bacteria, where a favorable environment is established, promoting the proliferation and transfer of resistance genes among different bacterial species. In our study, we isolated a total of 243 strains from 5 hospital wastewater sites in Mexico, belonging to 21 distinct Gram-negative bacterial species. The presence of β-lactamase was detected in 46.9% (114/243) of the isolates, which belonging to the Enterobacteriaceae family. We identified a total of 169 β-lactamase genes; bla TEM in 33.1%, bla CTX-M in 25.4%, bla KPC in 25.4%, bla NDM 8.8%, bla SHV in 5.3%, and bla OXA-48 in 1.1% distributed in 12 different bacteria species. Among the 114 of the isolates, 50.8% were found to harbor at least one carbapenemase and were discharged into the environment. The carbapenemase bla KPC was found in six Citrobacter spp. and E. coli , while bla NDM was detected in two distinct Enterobacter spp. and E. coli . Notably, bla NDM-1 was identified in a 110 Kb IncFII conjugative plasmid in E. cloacae , E. xiangfangensis, and E. coli within the same hospital wastewater. In conclusion, hospital wastewater showed the presence of Enterobacteriaceae carrying a high frequency of carbapenemase bla KPC and bla NDM . We propose that hospital wastewater serves as reservoirs for resistance mechanism within bacterial communities and creates an optimal environment for the exchange of this resistance mechanism among different bacterial strains., Importance: The significance of this study lies in its findings regarding the prevalence and diversity of antibiotic-resistant bacteria and genes identified in hospital wastewater in Mexico. The research underscores the urgent need for enhanced surveillance and prevention strategies to tackle the escalating challenge of antibiotic resistance, particularly evident through the elevated frequencies of carbapenemase genes such as bla KPC and bla NDM within the Enterobacteriaceae family. Moreover, the identification of these resistance genes on conjugative plasmids highlights the potential for widespread transmission via horizontal gene transfer. 140. Differential Carbon Catabolite Repression and Hemicellulolytic Ability among Pathotypes of Colletotrichum lindemuthianum against Natural Plant Substrates.

Abstract

Colletotrichum lindemuthianum is a phytopathogenic fungus that causes anthracnose in common beans ( Phaseolus vulgaris ) and presents a great diversity of pathotypes with different levels of virulence against bean varieties worldwide. The purpose of this study was to establish whether pathotypic diversity is associated with differences in the mycelial growth and secretion of plant-cell-wall-degrading enzymes (PCWDEs). We evaluated growth, hemicellulase and cellulase activity, and PCWDE secretion in four pathotypes of C. lindemuthianum in cultures with glucose, bean hypocotyls and green beans of P. vulgaris , and water hyacinth ( Eichhornia crassipes ). The results showed differences in the mycelial growth, hemicellulolytic activity, and PCWDE secretion among the pathotypes. Glucose was not the preferred carbon source for the best mycelial growth in all pathotypes, each of which showed a unique PCWDE secretion profile, indicating different levels of carbon catabolite regulation (CCR). The pathotypes showed a high differential hemicellulolytic capacity to degrade host and water hyacinth tissues, suggesting CCR by pentoses and that there are differences in the absorption and metabolism of different monosaccharides and/or disaccharides. We propose that different levels of CCR could optimize growth in different host tissues and could allow for consortium behavior in interactions with bean crops.

Abstract

Providencia rettgeri, belonging to the genus Providencia, had gained significant interest due to its increasing prevalence as a common pathogen responsible for healthcare-associated infections in hospitals. P. rettgeri isolates producing carbapenemases have been reported to reduce the efficiency of carbapenems in clinical antimicrobial therapy. However, coexistence with other resistance determinants is rarely reported. The goal of this study was the molecular characterization of carbapenemase-producing Providencia spp. clinical isolates. Among 23 Providencia spp. resistant to imipenem, 21 were positive to bla NDM-1 ; one positive to bla NDM-1 and bla OXA-58 like ; and one isolate co-producing bla IMP-27 , bla OXA-24/40 like , and bla OXA-58 like were identified. We observed a low clonal relationship, and the incompatibility groups Col3M and ColRNAI were identified in the plasmid harboring bla NDM-1 . We report for the first time a P. rettgeri strain co-producing bla IMP-27 , bla OXA-24-like , and bla OXA-58 like . Abstract

The spread of ESBL-producing Escherichia coli has constantly increased in both clinical and community infections. Actually, the main ESBL reported is the CTX-M family, which is widely disseminated between the Enterobacteriaceae family. The epidemiology of the CTX-M family shows the CTX-M-15 variant dominating worldwide, followed by CTX-M-14 and CTX-M-27. The specific ESBL-producing E. coli clones included mainly the sequence types ST131, ST405, and ST648. In this report, we present the molecular characterization of ESBL-producing E. coli clinical isolates from eight hospitals in Mexico. From a collection of 66 isolates, 39 (59%) were identified as bla CTX-M-14 and bla CTX-M-27 belonging to the group CTX-M-9. We identified 25 (38%) isolates, producing bla CTX-M-28 belonging to the group CTX-M-1. bla CTX-M-2 and bla TEM-55 were identified in one isolate, respectively. Fourteen isolates (21%) were positive for bla CTX-M-14 (13%) and bla CTX-M-28 (7.3%) that were selected for further analyses; the antimicrobial susceptibility showed resistance to ampicillin (> 256 µg/mL), cefotaxime (> 256 µg/mL), cefepime (> 64 µg/mL), and ceftazidime (16 µg/mL). The ResFinder analysis showed the presence of the antimicrobial resistance genes aacA4, aadA5, aac(3)lla, sul1, dfrA17, tet(A), cmlA1, and bla TEM-1B . PlasmidFinder analysis identified in all the isolates the replicons IncFIB, which were confirmed by PCR replicon typing. The MLST analysis identified isolates belonging to ST131, ST167, ST405, and ST648. The ISEcp1B genetic element was found at 250 pb upstream of bla CTX-M-14 and flanked by the IS903 genetic element at 35 pb downstream. The IS1380-like element ISEc9 family transposase was identified at 250 pb upstream of bla CTX-M-14 and flanked downstream by the IS5/IS1182 at 80 pb. Our study highlights the significant prevalence of CTX-M-14 and CTX-M-28 enzymes as the second-most common ESBL-producing E. coli among isolates in Mexican hospitals. The identification of specific sequence types in different regions provides valuable insights into the correlation between ESBL and E. coli strains. This contribution to understanding their epidemiology and potential transmission routes is crucial for developing effective strategies to mitigate the spread of ESBL-producing E. coli in healthcare settings., (© 2023. Abstract

Antimicrobial resistance is a major global public health problem, with fluoroquinolone-resistant strains of Escherichia coli posing a significant threat. This study examines the genetic characterization of ESBL-producing E. coli isolates in Mexican hospitals, which are resistant to both cephalosporins and fluoroquinolones. A total of 23 ESBL-producing E. coli isolates were found to be positive for the qepA gene, which confers resistance to fluoroquinolones. These isolates exhibited drug resistance phenotypes and belonged to specific sequence types and phylogenetic groups. The genetic context of the qepA gene was identified in a novel genetic context flanked by IS26 sequences. Mating experiments showed the co-transfer of qepA1 and chrA determinants alongside bla CTX-M-15 genes, emphasizing the potential for these genetic structures to spread among Enterobacterales. The emergence of multidrug-resistant Gram-negative bacteria carrying these resistance genes is a significant clinical concern for public healthcare systems., (© 2023. Abstract

Plasmids play a fundamental role in the evolution of bacteria by allowing them to adapt to different environments and acquire, through horizontal transfer, genes that confer resistance to different classes of antibiotics. Using the available in vitro and in silico plasmid typing systems, we analyzed a set of isolates and public genomes of K. variicola to study its plasmid diversity. The resistome, the plasmid multilocus sequence typing (pMLST), and molecular epidemiology using the MLST system were also studied. A high frequency of IncF plasmids from human isolates but lower frequency from plant isolates were found in our strain collection. In silico detection revealed 297 incompatibility (Inc) groups, but the IncFIB K  (216/297) predominated in plasmids from human and environmental samples, followed by IncFII K  (89/297) and IncFIA/FIA(HI1) (75/297). These Inc groups were associated with clinically important ESBL (CTX-M-15), carbapenemases (KPC-2 and NDM-1), and colistin-resistant genes which were associated with major sequence types (ST): ST60, ST20, and ST10. In silico MOB typing showed 76% (311/404) of the genomes contained one or more of the six relaxase families with MOB F  being most abundant. We identified untypeable plasmids carrying bla KPC-2 , bla IMP-1 , and bla SHV-187 but for which a relaxase was found; this may suggest that novel plasmid structures could be emerging in this bacterial species. The plasmid content in K. variicola has limited diversity, predominantly composed of IncFIB K plasmids dispersed in different STs. Plasmid detection using the replicon and MOB typing scheme provide a broader context of the plasmids in K. variicola. This study showed that whole-sequence-based typing provides current insights of the prevalence of plasmid types and their association with antimicrobial resistant genes in K. variicola obtained from humans and environmental niches., (© 2023. Introduction: Hypervirulent strains of Klebsiella pneumoniae have gained clinical and epidemiological interest because of their capacity to cause severe and life-threatening infections., Methodology: We report a case involving infection with a hypervirulent K. pneumoniae K2 strain that caused liver abscess in a young woman with type 1 diabetes in Mexico., Results: The infection was found to be associated with biliary tract communication. The virulence factors and capsular serotypes were identified by polymerase chain reaction analysis. After guided drainage and directed antibiotic treatment, the infection resolved and the patient recovered. Abstract

Accurate recognition of the closely related species Klebsiella pneumoniae, Klebsiella quasipneumoniae and Klebsiella variicola by phenotypic, biochemical and automated tests is notoriously unreliable in hospitals' diagnostic laboratories. A comparative genomics approach was conducted for the correct differentiation of the main bacterial species in the K. pneumoniae complex. Analysis of the deduced proteomes of 87 unique genomes of the Klebsiella in public databases, was used for the identification of unique protein family members. This allowed the design of a multiplex-PCR assay for the correct differentiation of these three species from different origins. This system allowed us to determine the prevalence of K. pneumoniae, K. quasipneumoniae and K. variicola among a collection of 552 clinical isolates. Of these, 87.3% (482/552) isolates corresponded to K. pneumoniae, 6.7% (33/552) to K. quasipneumoniae and 5.9% (33/552) to K. variicola. Abstract

Growth Hormone-Releasing Peptide 6 (GHRP-6) (His-(D-Trp)-Ala-Trp-(D-Phe)-Lys-NH2) is an agonist of the growth hormone secretagogue receptor. GHRP-6 mimics the effect of ghrelin. The present study focuses on the immunomodulatory effects of GHRP-6 in tilapia with and without the presence of Pseudomonas aeruginosa infection. GHRP-6 up-regulated the transcription levels of three piscidin-like antimicrobial peptides (Oreochromicins I, II, and III) and granzyme in a tissue-dependent manner. Antimicrobial activity stimulation in serum (lysozyme and anti-protease activity) was also confirmed. Besides, GHRP-6 enhanced the in vitro antimicrobial activity against P. aeruginosa in tilapia gills mucus and serum samples and decreased the bacterial load in vivo after infection with this Gram-negative bacterium. Our results evidenced, for the first time, a direct link between a growth hormone secretagogue ghrelin mimetic in fish and the enhancement of antimicrobial peptides transcription, which suggests that this secretagogue is capable to lead the activation of microbicidal activity in tilapia. Thus, these results open new possibilities for GHRP-6 application in aquaculture to stimulate the teleost immune system as an alternative treatment against opportunistic bacteria., (Copyright © 2021. Abstract

Klebsiella variicola has been found in various natural niches, alone or in association with other bacteria, and causes diseases in animals and plants with important economic and environmental impacts. K. variicola has the capacity to fix nitrogen in the rhizosphere and soil; produces indole acetic acid, acetoin, and ammonia; and dissolves phosphorus and potassium, which play an important role in plant growth promotion and nutrition. Some members of K. variicola have properties such as halotolerance and alkalotolerance, conferring an evolutionary advantage. In the environmental protection, K. variicola can be used in the wastewater treatment, biodegradation, and bioremediation of polluted soil, either alone or in association with other organisms. In addition, it has the potential to carry out industrial processes in the food and pharmaceutical industries, like the production of maltose and glucose by the catalysis of debranching unmodified oligosaccharides by the pullulanase enzyme. Finally, this bacterium has the ability to transform chemical energy into electrical energy, such as a biocatalyst, which could be useful in the near future. These properties show that K. variicola should be considered an eco-friendly bacterium with hopeful technological promise. Abstract

Purpose: The pathogenesis of ocular surface squamous neoplasia is not fully understood. Therefore, we evaluated the role of oncogenic viruses in the pathogenesis of ocular surface squamous neoplasia in Israel., Methods: Patients with ocular surface squamous neoplasia were enrolled in this retrospective study. The specimens were taken during 2004-2015 from two big centers: Emek Medical Center, Afula and the Hadassah Medical Center, Jerusalem. All the specimens (totally 26) were analyzed by immunohistochemistry for evidence of oncogenic viruses that included Human Papilloma virus, Herpes Simplex virus and Cytomegalovirus infection and 14 samples were examined by polymerase chain reaction. In addition, all the samples were examined for Epstein Barr virus infection by immunohistochemistry and Epstein Barr encoding region test., Results: Twenty-six patients were included with a mean age of 61.81 [Formula: see text] 3.83 years (mean [Formula: see text]. Immunohistochemistry staining and Epstein Barr encoding region test did not detect any of the oncogenic viruses in the 26 samples. Human Papilloma virus-16 and -18, and Herpes Simplex virus were detected by polymerase chain reaction in 14.2%, 7% and 7%, respectively., Conclusion: We conclude from our study that oncogenic viruses may play a role in the pathogenesis of ocular surface squamous neoplasia in Israel., (© 2021.
