Research conducted on monensin treatment of a gram-negative bacterium is presented. Experiments demonstrate that the subpopulation of Prevotella bryantii cultures has different membrane characteristics.

The objective of this study was to determine the effects of organic acids and monensin on the in vitro fermentation of cracked corn by mixed ruminal microorganisms. Ruminal fluid was collected from a steer fed 36.3 kg of wheat silage and 4.5 kg of concentrate supplement once daily. Mixed ruminal microorganisms were incubated in anaerobic media that contained 20% (vol/vol) ruminal fluid and .4 g of cracked corn. Incubations were carried out in batch culture for 24 h at 39 [degrees] C. Organic acids (L-aspartate, fumarate, and DL-malate) were added to serum bottles (n = 4) to achieve final concentrations of 0, 4, 8, or 12 mM. Monensin, dissolved in ethanol, was included in serum bottles at a final concentration of 0 or 5 ppm of culture fluid. The addition of 8 and 12 mM organic acids to cracked corn fermentations increased final pH (P < .05), tended to increase total gas production and C[O.sub.2] concentration, and decreased the acetate:propionate ratio (P < .05). Organic acids tended to decrease methane concentrations and hydrogen concentration was not altered. DL-Malate addition at all levels reduced (P < .05) lactate accumulation. Additive effects of monensin and organic acids were observed in some fermentations. In conclusion, organic acid addition to in vitro mixed ruminal microorganism fermentations yielded beneficial results independent of monensin treatment by decreasing the acetate: propionate ratio and increasing final pH. Background: The fecal microbiome of healthy horses may be influenced by signalment, diet, environmental factors, and disease. Objectives: To assess the effects of age, breed, sex, geographic location, season, diet, and colitis caused by antibiotic use (antimicrobial‐associated diarrhea [AAD]) and Salmonella infection on fecal microbiota. Animals: Healthy horses (n = 80) were sampled from nonhospital environments across multiple geographical locations in the United States. Horses with AAD (n = 14) were defined as those that developed diarrhea secondary to antimicrobial use. Horses with Salmonella infection (n = 12) were presented with spontaneous onset of colitis and subsequently tested positive on Salmonella quantitative polymerase chain reaction. All horses were >1 year of age and stratified by a dietary scale that included forages (pasture and hay) and concentrates grouped by percentage of fiber and amount. Methods: Illumina sequencing of 16S rRNA genes was performed on fecal DNA. Results: Healthy horses fed higher amounts of grain clustered separately from those fed lower amounts of grain (analysis of similarities [ANOSIM], R = 0.356‐0.385, Q = 0.002). Horses with AAD and Salmonella had decreased richness and evenness compared to healthy horses (P <.05). Univariable analysis of the 3 groups identified increases in Bacteroidetes (Q = 0.002) and Protebacteria (Q = 0.001) and decreases in Verrucomicrobia (Q = 0.001) in AAD horses whereas Salmonella horses had less Firmicutes (Q = 0.001) when compared to healthy horses. Conclusions and Clinical Importance: Although the amount of grain in the diet had some impact on the fecal microbiome, colitis had a significantly larger influence. Abstract Background The microbiota of an animal's intestinal tract plays important roles in the animal's overall health, productivity and well-being. There is still a scarcity of information on the microbial diversity in the gut of livestock species such as cattle. The primary reason for this lack of data relates to the expense of methods needed to generate such data. Here we have utilized a bacterial tag-encoded FLX 16s rDNA amplicon pyrosequencing (bTEFAP) approach that is able to perform diversity analyses of gastrointestinal populations. bTEFAP is relatively inexpensive in terms of both time and labor due to the implementation of a novel tag priming method and an efficient bioinformatics pipeline. We have evaluated the microbiome from the feces of 20 commercial, lactating dairy cows. Results Ubiquitous bacteria detected from the cattle feces included Clostridium, Bacteroides, Porpyhyromonas, Ruminococcus, Alistipes, Lachnospiraceae, Prevotella, Lachnospira, Enterococcus, Oscillospira, Cytophage, Anaerotruncus, and Acidaminococcus spp. Foodborne pathogenic bacteria were detected in several of the cattle, a total of 4 cows were found to be positive for Salmonella spp (tentative enterica) and 6 cows were positive for Campylobacter spp. (tentative lanienae). Conclusion Using bTEFAP we have examined the microbiota in the feces of cattle. As these methods continue to mature we will better understand the ecology of the major populations of bacteria the lower intestinal tract. This in turn will allow for a better understanding of ways in which the intestinal microbiome contributes to animal health, productivity and wellbeing.

Though much effort has been made to characterize the in vivo (IV) and in situ (IS) dry matter digestibility (DMD) of southeastern forages, there is a lack of information identifying methods that best represent the in vivo digestibility of these forages. Thus, the objective of our study was to compare IV and IS methods to determine which technique best represents the in vivo DMD of four bermudagrass (Cynodon dactylon [L.] Pers.) cul-tivars. In an IV experiment, ruminally fistulated heifers (n = 4) were randomly assigned to one of four bermudagrass cultivars (Coastal [COS], Russell [RUS], Tifton 44 [T44], or Tifton 85 [T85]) for four 30-d in vivo periods (21-d adaptation and 9-d collection) in a Latin square design. On d 28 of each period, rumen fluid was collected 4 h post-feeding for an accompanying IV experiment using a completely randomized design with a 4 x 2 factorial treatment structure. Factors in the IV experiment included bermudagrass cultivar (COS, RUS, T44, and T85) and digestibility method (Tilley and Terry [TT] or Goering and Van Soest [GVS]). On d 31 of in vivo periods 3 and 5, an accompanying IS experiment was conducted as a randomized complete block design with three treatment factors (in vivo diet [previous described], bermudagrass cultivar [previously described], and incubation timepoint [0, 0.25, 0.50, 0.75, 1, 2, 3, 4, 8, 12, 16, 20, 24, 48, 72, 96, 120, 144, and 168 h]). Correlations were computed between in vivo DM disappearance and each of the IV (TT and GVS) and IS (24, 48, and 72 h) methods. Regressions were calculated for each of the individual IV and IS measurements, and a stepwise regression was used to determine the predictive value of linear combinations of the measurements. None of the IV or IS DMD values were correlated with in vivo DMD (P ≥ 0.18). Individual IV and IS DMD values did not yield significant models through linear regression (P ≥ 0.18), though the best model (based on AIC) was TT IVDMD (r² = 0.09; P = 0.26). Stepwise regression revealed that there was no linear combination of IV or IS DMD that improved prediction beyond single predictor models. While both IV and IS experiments remain viable tools to screen forages and make relative comparisons, results from this experiment are interpreted to mean that neither of these methods is suitable for prediction of in vivo performance. A T4-like bacteriophage CEV1 is isolated from sheep that are resistant to colonization by Escherichia coli O157:H7 and is characterized in order to determine its potential to control the levels of the pathogen in sheep. It is observed that CEV1 efficiently infects E.coli O157:H7 grown both aerobically and anaerobically in vitro, and reduces sheep intestinal E.coli levels within 2 days of administration of an oral dose in vivo.

While much effort has been devoted to both the characterization of ruminal fermentation dynamics and the evaluation of bermudagrass production, independently, there is a lack of information regarding ruminal digestive kinetics using beef cattle in southern forage systems. Thus, the objective of our study was to evaluate in situ digestive kinetics of four bermudagrass (Cynodon dactylon [L.] Pers.) cultivars in heifers consuming typical southern forages. (Coastal, Tifton 44, Tifton 85, and Russell). Ruminally fistulated heifers (n = 4) were assigned randomly to one of four bermudagrass cultivars (Coastal [COS], Russell [RUS], Tifton 44 [T44], or Tifton 85 [T85]) for four 30-d in vivo periods (21-d adaptation and 9-d collection). On d 31 of periods 2 and 3, an in-situ experiment was conducted as a randomized complete block design with a 3-factor factorial treatment structure. The first factor (n = 4) was diet consumed by the animal (described above). The second factor (n = 4) was the incubated forage (same cultivars as the in vivo experiment). The third factor was incubation timepoint (n = 19; 0, 0.25, 0.50, 0.75, 1, 2, 3, 4, 8, 12, 16, 20, 24, 48, 72, 96, 120, 144, and 168 h). Four in situ bags were incubated in each animal at each timepoint; two were used for NDF, ADF, and ADL, and two were used for CP determination. Data were analyzed using SAS v. 9.4. There was a decreased (P < 0.05) potentially degradable fraction from COS (30.0%) compared with T85 (55.1%), T44 (48.1%), or RUS (47.1%). Similarly, COS presented the greatest (P < 0.05) undegradable fraction (37.0%) and T85 the least (15.9%), with RUS and T44 intermediate (25.0 and 21.9%, respectively). There was no effect of bermudagrass cultivar, however, on the rate constant of digestion (0.94%/h; P = 0.26) or lag time (3.2 h; P = 0.38). A visual appraisal of the degradation curve suggests that the asymptote of digestion was not reached at the measured 168 h. Results are interpreted to mean that T44 is the preferable bermudagrass cultivar, followed by T85 and RUS, while COS is the least favorable. Further, given the lack of visible asymptote, we speculate that bermudagrass likely passes the rumen before full digestion is realized. [ABSTRACT FROM AUTHOR]

The disinfectant and antimicrobial susceptibility profiles of 138 non-0157 Shiga toxin-producing Escherichia coli strains (STECs) from food animals and humans were determined. Antimicrobial resistance (AMR) was moderate (39.1% of strains) in response to 15 antimicrobial agents. Animal strains had a lower AMR prevalence (35.6%) than did human strains (43.9%) but a higher prevalence of the resistance profile GEN-KAN-TET. A decreasing prevalence of AMR was found among animal strains from serogroups 045 > 0145 > 0121 > 01 11 > 026 > 0103 and among human strains from serogroups 0145 > 0103 > 026 > 01 11 > 0121 > 045. One animal strain from serogroups 0121 and 0145 and one human strain from serogroup 026 had extensive drug resistance. A high prevalence of AMR in animal 045 and 0121 strains and no resistance or a low prevalence of resistance in human strains from these serogroups suggests a source other than food animals for human exposure to these strains. Among the 24 disinfectants evaluated, all strains were susceptible to triclosan. Animal strains had a higher prevalence of resistance to chlorhexidine than did human strains. Both animal and human strains had a similar low prevalence of low-level benzalkonium chloride resistance, and animal and human strains had similar susceptibility profiles for most other disinfectants. Benzyldimethylammonium chlorides and C10AC were the primary active components in disinfectants DC&R and P-128, respectively, against non-0157 STECs. A disinfectant FS512 MIC > 8 ug/ml was more prevalent among animal 0121 strains (61.5%) than among human 0121 strains (25%), which may also suggest a source of human exposure to STEC 0121 other than food animals. Bacterial inhibition was not dependent solely on pH but was correlated with the presence of dissociated organic acid species and some undissociated acids. [ABSTRACT FROM AUTHOR]
