- Abstract
The COVID-19 pandemic has raised even greater concerns about a growing teacher shortage. In this study, we use administrative data on more than 140,000 Michigan traditional public and charter school teachers in an interrupted time series framework to understand how teacher attrition and supply may have shifted after the onset of the COVID-19 pandemic. Our results suggest that Michigan teachers were more likely to leave the profession, less likely to leave their districts, and more likely to switch schools within their district after the 2020-21 and 2021-22 school years relative to pre-pandemic trends. Additionally, teachers in low COVID-19 rate communities, as well as those in districts that offered fully in-person instruction at the start of the 2020-21 school year, were more likely to leave the teaching profession or switch districts following the 2019-20 school year compared to teachers in fully remote districts.

- Abstract
Teacher-family partnerships are crucial elements of high-quality early childhood education. The factors influencing these partnerships, particularly for children under age 3, are not well known. This study compares teachers' and mothers' perspectives on their partnership and the ideal practices they would like to see implemented. Additionally, it investigates child, family, and program-level factors predictive of partnership practices. Participants were mothers and teachers of 90 infants who completed the Real-Ideal Teacher-parents Partnership Scale; mothers rated child's temperament, and classroom quality was observed 6-months after infants entered childcare. Home environment was assessed before infants entered childcare. Both mothers and teachers reported a medium-high number of practices being implemented, but ideally, would like more practices to be implemented. The frequency of mother and teacher reports of real and ideal practices were significantly associated, although teacher reports were higher. Findings from the regression analyses showed that teacher level of education (having a Master) was uniquely and positively associated with both teachers' and mothers' reports of real partnership practices, and was significantly associated with teachers' reports of ideal partnership practices. Mothers' education level predicted their ideal partnership practices. Findings highlight the importance of teacher education for partnerships, especially considering the variability in legal requirements regarding qualifications for teaching infants across Europe.

In the realm of chest X-ray (CXR) image analysis, radiologists meticulously examine various regions, documenting their observations in reports. The prevalence of errors in CXR diagnoses, particularly among inexperienced radiologists and hospital residents, underscores the importance of understanding radiologists' intentions and the corresponding regions of interest. This understanding is crucial for correcting mistakes by guiding radiologists to the accurate regions of interest, especially in the diagnosis of chest radiograph abnormalities. In response to this imperative, we propose a novel system designed to identify the primary intentions articulated by radiologists in their reports and the corresponding regions of interest in CXR images. This system seeks to elucidate the visual context underlying radiologists' textual findings, with the potential to rectify errors made by less experienced practitioners and direct them to precise regions of interest. Importantly, the proposed system can be instrumental in providing constructive feedback to inexperienced radiologists or junior residents in the hospital, bridging the gap in face-to-face communication. The system represents a valuable tool for enhancing diagnostic accuracy and fostering continuous learning within the medical community., Comment: Accepted in ISBI 2024

Astrophysics - Earth and Planetary Astrophysics - Abstract
We present the results from the first two years of the Planet Hunters NGTS citizen science project, which searches for transiting planet candidates in data from the Next Generation Transit Survey (NGTS) by enlisting the help of members of the general public. Over 8,000 registered volunteers reviewed 138,198 light curves from the NGTS Public Data Releases 1 and 2. We utilize a user weighting scheme to combine the classifications of multiple users to identify the most promising planet candidates not initially discovered by the NGTS team. We highlight the five most interesting planet candidates detected through this search, which are all candidate short-period giant planets. This includes the TIC-165227846 system that, if confirmed, would be the lowest-mass star to host a close-in giant planet. We assess the detection efficiency of the project by determining the number of confirmed planets from the NASA Exoplanet Archive and TESS Objects of Interest (TOIs) successfully recovered by this search and find that 74% of confirmed planets and 63% of TOIs detected by NGTS are recovered by the Planet Hunters NGTS project. The identification of new planet candidates shows that the citizen science approach can provide a complementary method to the detection of exoplanets with ground-based surveys such as NGTS., Comment: 42 pages, 20 figures, 17 tables. Precision measurements of space and time, like those made by the detectors of the Laser Interferometer Gravitational-wave Observatory (LIGO), are often confronted with fundamental limitations imposed by quantum mechanics. The Heisenberg uncertainty principle dictates that the position and momentum of an object cannot both be precisely measured, giving rise to an apparent limitation called the Standard Quantum Limit (SQL). Reducing quantum noise below the SQL in gravitational-wave detectors, where photons are used to continuously measure the positions of freely falling mirrors, has been an active area of research for decades. Here we show how the LIGO A+ upgrade reduced the detectors' quantum noise below the SQL by up to 3 dB while achieving a broadband sensitivity improvement, more than two decades after this possibility was first presented.

Computer Science - Computation and Language - Abstract
Code-switching (CSW) is a common phenomenon among multilingual speakers where multiple languages are used in a single discourse or utterance. Mixed language utterances may still contain grammatical errors however, yet most existing Grammar Error Correction (GEC) systems have been trained on monolingual data and not developed with CSW in mind. In this work, we conduct the first exploration into the use of GEC systems on CSW text. Through this exploration, we propose a novel method of generating synthetic CSW GEC datasets by translating different spans of text within existing GEC corpora. We then investigate different methods of selecting these spans based on CSW ratio, switch-point factor and linguistic constraints, and identify how they affect the performance of GEC systems on CSW text. Our best model achieves an average increase of 1.57 $F_{0.5}$ across 3 CSW test sets (English-Chinese, English-Korean and English-Japanese) without affecting the model's performance on a monolingual dataset. We furthermore discovered that models trained on one CSW language generalise relatively well to other typologically similar CSW languages.

Nagaoka ferromagnetism (NF) is a long-predicted example of itinerant ferromagnetism (IF) in the Hubbard model that has been studied theoretically for many years. The condition for NF, an infinite on-site Coulomb repulsion and a single hole in a half-filled band, does not arise naturally in materials. NF was only realized recently for the first time in experiments on a $2\times 2$ array of gated quantum dots. Dopant arrays and gated quantum dots in Si allow for engineering controllable systems with complex geometries. This makes quantum dot arrays good candidates to study NF in different geometries through analog quantum simulation. Here we present theoretical simulations done for $3\times 3$ arrays and larger $N\times N$ arrays, and predict the emergence of different forms of ferromagnetism in different geometries. We find NF in perfect $3\times 3$ arrays, and in $N\times N$ arrays for one hole doping of a half-filled band. The ratio of the hopping $t$ to Hubbard on-site repulsion $U$ that defines the onset of NF scales as $1/N^{4}$ as $N$ increases, approaching the bulk limit of infinite $U$ for large $N$. Additional simulations are done for geometries made by removing sites from $N\times N$ arrays. Different forms of ferromagnetism are found for different geometries. Loops show ferromagnetism, but only for three electrons. For loops, the critical $t/U$ for the onset of ferromagnetism scales as $N$ as the loop length increases. We show that the different dependences on size for loops and $N\times N$ arrays can be understood by scaling arguments that highlight the different energy contributions to each form of ferromagnetism. Our results show how analog quantum simulation with small arrays can elucidate the role of effects including wavefunction connectivity; system geometry, size and symmetry; bulk and edge sites; and kinetic energy in determining quantum magnetism of small systems.

Astrophysics - High Energy Astrophysical Phenomena ,General Relativity and Quantum Cosmology - Abstract
We report the observation of a coalescing compact binary with component masses $2.5\text{-}4.5~M_\odot$ and $1.2\text{-}2.0~M_\odot$ (all measurements quoted at the 90% credible level). The gravitational-wave signal GW230529_181500 was observed during the fourth observing run of the LIGO-Virgo-KAGRA detector network on 2023 May 29 by the LIGO Livingston Observatory. The primary component of the source has a mass less than $5~M_\odot$ at 99% credibility. We cannot definitively determine from gravitational-wave data alone whether either component of the source is a neutron star or a black hole. However, given existing estimates of the maximum neutron star mass, we find the most probable interpretation of the source to be the coalescence of a neutron star with a black hole that has a mass between the most massive neutron stars and the least massive black holes observed in the Galaxy. We provisionally estimate a merger rate density of $55^{+127}_{-47}~\text{Gpc}^{-3}\,\text{yr}^{-1}$ for compact binary coalescences with properties similar to the source of GW230529_181500; assuming that the source is a neutron star-black hole merger, GW230529_181500-like sources constitute about 60% of the total merger rate inferred for neutron star-black hole coalescences. The discovery of this system implies an increase in the expected rate of neutron star-black hole mergers with electromagnetic counterparts and provides further evidence for compact objects existing within the purported lower mass gap., Comment: 45 pages (10 pages author list, 13 pages main text, 1 page acknowledgements, 13 pages appendices, 8 pages bibliography), 17 figures, 16 tables. Astrophysics - Cosmology and Nongalactic Astrophysics - Abstract
We measure the stacked lensing signal in the direction of galaxy clusters in the Dark Energy Survey Year 3 (DES Y3) redMaPPer sample, using cosmic microwave background (CMB) temperature data from SPT-3G, the third-generation CMB camera on the South Pole Telescope (SPT). We estimate the lensing signal using temperature maps constructed from the initial 2 years of data from the SPT-3G 'Main' survey, covering 1500 deg$^2$ of the Southern sky. We then use this signal as a proxy for the mean cluster mass of the DES sample. In this work, we employ three versions of the redMaPPer catalogue: a Flux-Limited sample containing 8865 clusters, a Volume-Limited sample with 5391 clusters, and a Volume&Redshift-Limited sample with 4450 clusters. For the three samples, we find the mean cluster masses to be ${M}_{200{\rm{m}}}=1.66\pm0.13$ [stat.]$\pm0.03$ [sys.], $1.97\pm0.18$ [stat.]$\pm0.05$ [sys.], and $2.11\pm0.20$ [stat.]$\pm0.05$ [sys.]$\times{10}^{14}\ {\rm{M}}_{\odot }$, respectively. This is a factor of $\sim2$ improvement relative to the precision of measurements with previous generations of SPT surveys and the most constraining cluster mass measurements using CMB cluster lensing to date. Overall, we find no significant tensions between our results and masses given by redMaPPer mass-richness scaling relations of previous works, which were calibrated using CMB cluster lensing, optical weak lensing, and velocity dispersion measurements from various combinations of DES, SDSS and Planck data. We then divide our sample into 3 redshift and 3 richness bins, finding no significant tensions with optical weak-lensing calibrated masses in these bins. We forecast a $5.7\%$ constraint on the mean cluster mass of the DES Y3 sample with the complete SPT-3G surveys when using both temperature and polarization data and including an additional $\sim1400$ deg$^2$ of observations from the 'Extended' SPT-3G survey., Comment: 23 pages, 9 figures, accepted for publication in JCAP. We classify curvature homogeneous hypersurfaces in S^4 and H^4. In higher dimesnsion one only has the FKM examples and an isolate one by Tsukada of a hypersurface in H^5. Besides some simple examples, we show that there exists an isolated hypersurface with a circle of symmetries and and a one parameter family admitting no continuous symmetries. Outside the set of minimal points, which only exists in the case of S^4, every example is locally and up to covers of this form.

Astrophysics - Cosmology and Nongalactic Astrophysics - Abstract
We forecast constraints on cosmological parameters enabled by three surveys conducted with SPT-3G, the third-generation camera on the South Pole Telescope. The surveys cover separate regions of 1500, 2650, and 6000 ${\rm deg}^{2}$ to different depths, in total observing 25% of the sky. These regions will be measured to white noise levels of roughly 2.5, 9, and 12 $\mu{\rm K-arcmin}$, respectively, in CMB temperature units at 150 GHz by the end of 2024. The survey also includes measurements at 95 and 220 GHz, which have noise levels a factor of ~1.2 and 3.5 times higher than 150 GHz, respectively, with each band having a polarization noise level ~$\sqrt{\text{2}}$ times higher than the temperature noise. We use a novel approach to obtain the covariance matrices for jointly and optimally estimated gravitational lensing potential bandpowers and unlensed CMB temperature and polarization bandpowers. We demonstrate the ability to test the $\Lambda{\rm CDM}$ model via the consistency of cosmological parameters constrained independently from SPT-3G and Planck data, and consider the improvement in constraints on $\Lambda{\rm CDM}$ extension parameters from a joint analysis of SPT-3G and Planck data. The $\Lambda{\rm CDM}$ cosmological parameters are typically constrained with uncertainties up to ~2 times smaller with SPT-3G data, compared to Planck, with the two data sets measuring significantly different angular scales and polarization levels, providing additional tests of the standard cosmological model., Comment: 26 pages; 13 figures; Accepted for publication in ApJ; Minor edits have been made

The Wide-field Spectroscopic Telescope (WST) is proposed as a new facility dedicated to the efficient delivery of spectroscopic surveys. This white paper summarises the initial concept as well as the corresponding science cases. WST will feature simultaneous operation of a large field-of-view (3 sq. degree), a high multiplex (20,000) multi-object spectrograph (MOS) and a giant 3x3 sq. arcmin integral field spectrograph (IFS). In scientific capability these requirements place WST far ahead of existing and planned facilities. Given the current investment in deep imaging surveys and noting the diagnostic power of spectroscopy, WST will fill a crucial gap in astronomical capability and work synergistically with future ground and space-based facilities. This white paper shows that WST can address outstanding scientific questions in the areas of cosmology; galaxy assembly, evolution, and enrichment, including our own Milky Way; origin of stars and planets; time domain and multi-messenger astrophysics. WST's uniquely rich dataset will deliver unforeseen discoveries in many of these areas. The WST Science Team (already including more than 500 scientists worldwide) is open to the all astronomical community. Among the various candidates for dark matter (DM), ultralight vector DM can be probed by laser interferometric gravitational wave detectors through the measurement of oscillating length changes in the arm cavities. In this context, KAGRA has a unique feature due to differing compositions of its mirrors, enhancing the signal of vector DM in the length change in the auxiliary channels. Here we present the result of a search for $U(1)_{B-L}$ gauge boson DM using the KAGRA data from auxiliary length channels during the first joint observation run together with GEO600. By applying our search pipeline, which takes into account the stochastic nature of ultralight DM, upper bounds on the coupling strength between the $U(1)_{B-L}$ gauge boson and ordinary matter are obtained for a range of DM masses. While our constraints are less stringent than those derived from previous experiments, this study demonstrates the applicability of our method to the lower-mass vector DM search, which is made difficult in this measurement by the short observation time compared to the auto-correlation time scale of DM., Comment: 20 pages, 5 figures

We report results from an analysis aimed at detecting the trispectrum of the kinematic Sunyaev-Zel{'}dovich (kSZ) effect by combining data from the South Pole Telescope (SPT) and {\it Herschel}-SPIRE experiments over a 100 ${\rm deg}^{2}$ field. The SPT observations combine data from the previous and current surveys, namely SPTpol and SPT-3G, to achieve depths of 4.5, 3, and 16 $\mu {\rm K-arcmin}$ in bands centered at 95, 150, and 220 GHz. For SPIRE, we include data from the 600 and 857 GHz bands. We reconstruct the velocity-induced large-scale correlation of the small-scale kSZ signal with a quadratic estimator that uses two cosmic microwave background (CMB) temperature maps, constructed by optimally combining data from all the frequency bands. We reject the null hypothesis of a zero trispectrum at $10.3\sigma$ level. However, the measured trispectrum contains contributions from both the kSZ and other undesired components, such as CMB lensing and astrophysical foregrounds, with kSZ being sub-dominant. We use the \textsc{Agora} simulations to estimate the expected signal from CMB lensing and astrophysical foregrounds. After accounting for the contributions from CMB lensing and foreground signals, we do not detect an excess kSZ-only trispectrum and use this non-detection to set constraints on reionization. By applying a prior based on observations of the Gunn-Peterson trough, we obtain an upper limit on the duration of reionization of $\Delta z_{\rm re, 50} < 4.5$ (95\% C.L). We find these constraints are fairly robust to foregrounds assumptions. This trispectrum measurement is independent of, but consistent with, {\it Planck}'s optical depth measurement. This result is the first constraint on the epoch of reionization using the non-Gaussian nature of the kSZ signal., Comment: 15 pages, 5 figures (3 in main text and 2 in Appendix); Accepted for publication in PRL; Some texts have been moved to Appendix; Minor change in Fig. 2 to include nomalization; Data products and plotting scripts can be downloaded from https://github.com/sriniraghunathan/kSZ_4pt_SPT_SPIRE

A set of vertices of a graph $G$ such that each vertex of $G$ is either in the set or is adjacent to a vertex in the set is called a dominating set of $G$. If additionally, the set of vertices induces a connected subgraph of $G$ then the set is a connected dominating set of $G$. The domination number $\gamma(G)$ of $G$ is the smallest number of vertices in a dominating set of $G$, and the connected domination number $\gamma_c(G)$ of $G$ is the smallest number of vertices in a connected dominating set of $G$. We find the connected domination numbers for all triangulations of up to thirteen vertices. For $n\ge 15$, $n\equiv 0$ (mod 3), we find graphs of order $n$ and $\gamma_c=\frac{n}{3}$. We also show that the difference $\gamma_c(G)-\gamma(G)$ can be arbitrarily large., Comment: 12 pages, 10 figures, 1 table

We characterize the single-electron energies and the wavefunction structure of arrays with two, three, and four phosphorus atoms in silicon by implementing atomistic tight-binding calculations and analyzing wavefunction overlaps to identify the single-dopant states that hybridize to make the array states. The energy spectrum and wavefunction overlap variation as a function of dopant separation for these arrays shows that hybridization mostly occurs between single-dopant states of the same type, with some cross-hybridization between $A_1$ and $E$ states occurring at short separations. We also observe energy crossings between hybrid states of different types as a function of impurity separation. We then extract tunneling rates for electrons in different dopants by mapping the state energies into hopping Hamiltonians in the site representation. Significantly, we find that diagonal and nearest neighbor tunneling rates are similar in magnitude in a square array. Our analysis also accounts for the shift of the on-site energy at each phosphorus atom resulting from the nuclear potential of the other dopants. This approach constitutes a solid protocol to map the electron energies and wavefunction structure into Fermi-Hubbard Hamiltonians needed to implement and validate analog quantum simulations in these devices.

We report the discovery of a brown dwarf orbiting a M1 host star. We first identified the brown dwarf within the Next Generation Transit Survey data, with supporting observations found in TESS sectors 11 and 38. We confirmed the discovery with follow-up photometry from the South African Astronomical Observatory, SPECULOOS-S, and TRAPPIST-S, and radial velocity measurements from HARPS, which allowed us to characterise the system. We find an orbital period of ~1.25 d, a mass of 69.0+5.3-4.8 MJ, close to the Hydrogen burning limit, and a radius of 0.95 +- 0.05 RJ. We determine the age to be >0.5 Gyr, using model isochrones, which is found to be in agreement with SED fitting within errors. NGTS-28Ab is one of the shortest period systems found within the brown dwarf desert, as well as one of the highest mass brown dwarfs that transits an M dwarf. This makes NGTS-28Ab another important discovery within this scarcely populated region., Comment: 20 pages (inc. appendices), 16 figures, accepted for publication in MNRAS

Bulk high-temperature superconductors (HTS) are capable of generating very strong magnetic fields while maintaining a relatively compact form factor. Solenoids constructed using stacks of ring-shaped bulk HTS have been demonstrated to be capable of nuclear magnetic resonance (NMR) spectroscopy and magnetic resonance imaging (MRI). However, these stacks were magnetised via field cooling (FC), which typically requires a secondary superconducting charging magnet capable of sustaining a high magnetic field for a long period. A more economical alternative to FC is pulsed field magnetisation, which can be carried out with a magnet wound from a normal conductor, such as copper. In this work, we present a technique we have developed for iteratively homogenising the magnetic field within a stack of ring-shaped bulk HTS by manipulating the spatial profile of the applied pulsed field., Comment: 4 pages, 2 figures

Astrophysics - Astrophysics of Galaxies - Abstract
We use the SAMI Galaxy Survey to examine the drivers of galaxy spin, $\lambda_{R_e}$, in a multi-dimensional parameter space including stellar mass, stellar population age (or specific star formation rate) and various environmental metrics (local density, halo mass, satellite vs. central). Using a partial correlation analysis we consistently find that age or specific star formation rate is the primary parameter correlating with spin. Light-weighted age and specific star formation rate are more strongly correlated with spin than mass-weighted age. In fact, across our sample, once the relation between light-weighted age and spin is accounted for, there is no significant residual correlation between spin and mass, or spin and environment. This result is strongly suggestive that present-day environment only indirectly influences spin, via the removal of gas and star formation quenching. That is, environment affects age, then age affects spin. Older galaxies then have lower spin, either due to stars being born dynamically hotter at high redshift, or due to secular heating. Our results appear to rule out environmentally dependent dynamical heating (e.g. galaxy-galaxy interactions) being important, at least within $1R_e$ where our kinematic measurements are made. The picture is more complex when we only consider high-mass galaxies ($M_*\gtrsim 10^{11}$M$_{\odot}$). While the age-spin relation is still strong for these high-mass galaxies, there is a residual environmental trend with central galaxies preferentially having lower spin, compared to satellites of the same age and mass. We argue that this trend is likely due to central galaxies being a preferred location for mergers., Comment: 24 pages, 9 figures. Accepted for publication in MNRAS

Astrophysics - Astrophysics of Galaxies - Abstract
Slow rotator galaxies are distinct amongst galaxy populations, with simulations suggesting that a mix of minor and major mergers are responsible for their formation. A promising path to resolve outstanding questions on the type of merger responsible, is by investigating deep imaging of massive galaxies for signs of potential merger remnants. We utilise deep imaging from the Subaru-Hyper Suprime Cam Wide data to search for tidal features in massive ($\log_{10}(M_*/M_{\odot}) > 10$) early-type galaxies (ETGs) in the SAMI Galaxy Survey. We perform a visual check for tidal features on images where the galaxy has been subtracted using a Multi-Gauss Expansion (MGE) model. We find that $31\pm 2$ percent of our sample show tidal features. When comparing galaxies with and without features, we find that the distributions in stellar mass, light-weighted mean stellar population age and H$\alpha$ equivalent width are significantly different, whereas spin ($\lambda_{R_e}$), ellipticity and bulge to total ratio have similar distributions. When splitting our sample in age, we find that galaxies below the median age (10.8 Gyr) show a correlation between the presence of shells and lower $\lambda_{R_e}$, as expected from simulations. We also find these younger galaxies which are classified as having "strong" shells have lower $\lambda_{R_e}$. However, simulations suggest that merger features become undetectable within $\sim 2-4$ Gyr post-merger. This implies that the relationship between tidal features and merger history disappears for galaxies with older stellar ages, i.e. those that are more likely to have merged long ago., Comment: Accepted for publication in MNRAS. 22 pages, 14 figures

Astrophysics - Astrophysics of Galaxies - Abstract
We use the SAMI galaxy survey to study the the kinematic morphology-density relation: the observation that the fraction of slow rotator galaxies increases towards dense environments. We build a logistic regression model to quantitatively study the dependence of kinematic morphology (whether a galaxy is a fast rotator or slow rotator) on a wide range of parameters, without resorting to binning the data. Our model uses a combination of stellar mass, star-formation rate (SFR), $r$-band half-light radius and a binary variable based on whether the galaxy's observed ellipticity ($\epsilon$) is less than 0.4. We show that, at fixed mass, size, SFR and $\epsilon$, a galaxy's local environmental surface density ($\log_{10}(\Sigma_5/\mathrm{Mpc}^{-2})$) gives no further information about whether a galaxy is a slow rotator, i.e. the observed kinematic-morphology density relation can be entirely explained by the well-known correlations between environment and other quantities. We show how our model can be applied to different galaxy surveys to predict the fraction of slow rotators which would be observed and discuss its implications for the formation pathways of slow rotators., Comment: 12 pages, 6 figures. Statistics - Methodology - Abstract
Data on historical populations often extends no further than numbers of people by broad age-sex group, with nothing on numbers of births or deaths. Demographers studying these populations have experimented with methods that use the data on numbers of people to infer birth and death rates. These methods have, however, received little attention since they were first developed in the 1960s. We revisit the problem of inferring demographic rates from population structure, spelling out the assumptions needed, and specialising the methods to the case where only child-woman ratios are available. We apply the methods to the case of Maori populations in nineteenth-century Aotearoa New Zealand. We find that, in this particular case, the methods reveal as much about the nature of the data as they do about historical demographic conditions., Comment: 21 pages, 11 figures

Bacteroides fragilis is a Gram-negative commensal bacterium commonly found in the human colon, which differentiates into two genomospecies termed divisions I and II. Through a comprehensive collection of 694 B. fragilis whole genome sequences, we identify novel features distinguishing these divisions. Our study reveals a distinct geographic distribution with division I strains predominantly found in North America and division II strains in Asia. Additionally, division II strains are more frequently associated with bloodstream infections, suggesting a distinct pathogenic potential. We report differences between the two divisions in gene abundance related to metabolism, virulence, stress response, and colonization strategies. Notably, division II strains harbor more antimicrobial resistance (AMR) genes than division I strains. These findings offer new insights into the functional roles of division I and II strains, indicating specialized niches within the intestine and potential pathogenic roles in extraintestinal sites.ImportanceUnderstanding the distinct functions of microbial species in the gut microbiome is crucial for deciphering their impact on human health. Classifying division II strains as Bacteroides fragilis can lead to erroneous associations, as researchers may mistakenly attribute characteristics observed in division II strains to the more extensively studied division I B. fragilis. Our findings underscore the necessity of recognizing these divisions as separate species with distinct functions. We unveil new findings of differential gene prevalence between division I and II strains in genes associated with intestinal colonization and survival strategies, potentially influencing their role as gut commensals and their pathogenicity in extraintestinal sites. Despite the significant niche overlap and colonization patterns between these groups, our study highlights the complex dynamics that govern strain distribution and behavior, emphasizing the need for a nuanced understanding of these microorganisms.

PURPOSE: Minimal residual disease (MRD) detection can identify the recurrence in patients with colorectal cancer (CRC) following definitive treatment. We evaluated a plasma-only MRD assay to predict recurrence and survival in patients with metastatic CRC who underwent curative intent procedures (surgery and/or radiotherapy), with or without (neo)adjuvant chemotherapy. The primary objective of this study was to assess the correlation of postprocedure tumor cell-free DNA detection status with radiographic disease recurrence. EXPERIMENTAL DESIGN: Preprocedure and postprocedure longitudinal samples were collected from 53 patients and analyzed with a multiomic MRD assay detecting circulating tumor DNA (ctDNA) from genomic and epigenomic signals. Preprocedure and postprocedure ctDNA detection correlated with recurrence-free and overall survival (OS). RESULTS: From 52 patients, 230/233 samples were successfully analyzed. At the time of data cutoff, 36 (69.2%) patients recurred with median follow-up of 31 months. Detectable ctDNA was observed in 19/42 patients (45.2%) with ctDNA analyzed 3 weeks postprocedure. ctDNA detection 3 weeks postprocedure was associated with shorter median recurrence-free survival (RFS; HR, 5.27; 95% CI, 2.31-12.0; P < 0.0001) and OS (HR, 12.83; 95% CI, 3.6-45.9; P < 0.0001). Preprocedure ctDNA detection status was not associated with RFS but was associated with improved OS (HR, 4.65; 95% CI, 1.4-15.2; P = 0.0111). Undetectable ctDNA preprocedure had notable long-term OS, >90% 3 years postprocedure. CONCLUSIONS: In this cohort of oligometastatic CRC, detection of ctDNA preprocedure or postprocedure was associated with inferior outcomes even after accounting for known prognostic clinicopathologic variables. This suggests ctDNA may enhance current risk stratification methods helping the evaluation of novel treatments and surveillance strategies toward improving patient outcomes.

The phytocannabinoid cannabinol (CBN) has a potential mechanism of action as an alternative sleep aid but there is minimal evidence to support its effectiveness. The aim of this randomized, double-blind, placebo-controlled study was to assess the safety and effects of three formulations of a hemp-derived CBN sleep aid, TruCBN™ [25 mg (n = 206), 50 mg (n = 205), 100 mg (n = 203)], on sleep quality (PROMIS Sleep Disturbance 8A), relative to placebo (n = 204). The effectiveness and safety of these formulations relative to 4 mg of melatonin (n = 202) was assessed. Exploratory measures were stress (PROMIS Stress 4A), anxiety (Anxiety 4A), pain (PROMIS™ PEG), and well-being (WHO 5). All groups and the 4 mg melatonin group experienced significant improvement in sleep quality relative to the placebo group with no significant differences between any group and the melatonin group. Participants taking 100 mg showed a larger decrease in stress compared to the placebo group. There were no significant differences in anxiety, pain, well-being, or the frequency of side effects between any group and the placebo group. There was no significant difference in improvements in sleep quality between any of the treatment groups and the 4 mg melatonin group. Orally ingested CBN, at 25 mg, 50 mg, and 100 mg, is a safe and effective alternative for the improvement of sleep.
