Category: Trauma; Ankle Introduction/Purpose: Overtightening of the distal tibiofibular syndesmosis during open reduction and internal fixation (ORIF) of ankle fractures remains a clinical concern, and there are limited data delineating the effect of syndesmosis overcompression on subsequent range of motion of the ankle joint bones. The primary objective of the current study was to determine the effect of syndesmotic overcompression on the relative range of motion (ROM) of the tibia, fibula, and talus, and to determine the syndesmotic fixation construct that most accurately restores native motion. Methods: Ten cadaveric lower limbs were utilized (78.3±13.0 yrs, 4F; 6M). A musculoskeletal simulator at ±7.5 Nm was used to test flexibility in dorsiflexion-plantarflexion, inversion-eversion, and internal-external rotation. After intact testing, syndesmotic probes were used to arthroscopically measure the intact position of the distal tibiofibular syndesmosis. The force needed to compress the syndesmosis just beyond the intact position represented the intact force (100%), and overcompression was defined as 150% of the intact force. The anterior inferior tibiofibular ligament (AITFL), interosseus membrane (IOM), and posterior inferior tibiofibular ligament (PITFL) were then sectioned. Testing was repeated for destabilized, IOM reconstruction at 100% compression and 150% overcompression, IOM reconstruction plus AITFL fixation at 100% compression and 150% overcompression, and AITFL fixation alone. ROM data was reported as a percentage of the intact condition. Repeated measures ANOVA and Bonferroni post hoc test were performed to assess differences between conditions with p< 0.05. Results: Average force needed to compress the syndesmosis was 71.0 ± 22.2 N. There was a significant increase in distal tibiofibular ROM in internal and external rotation for all conditions compared to intact (Figure 1) (p < 0.05). In the axial plane, reconstruction of the IOM membrane did not restore the intact condition and was similar to the destabilized treatment, regardless of compression force. The addition of AITFL fixation significantly reduced distal tibiofibular motion compared to IOM reconstructions alone, but still did not approach the intact condition. Overcompression of the syndesmosis did not significantly affect relative ROM of the ankle bones in any plane (p>0.05). Changes in the proximal tibiofibular ROM were similar in magnitude and direction as the distal tibiofibular ROM for all conditions. Conclusion: In the current study, overcompression of the distal tibiofibular syndesmosis did not restrict relative ROM of the tibia, fibula, or talus. Reconstructing the AITFL in addition to the IOM decreased tibiofibular ROM, particularly in the axial plane. In the clinical setting, surgical reconstruction with dynamic fixation of the distal tibiofibular syndesmosis may result in increased ROM and mechanical stress at the proximal tibiofibular articulation.

The formation of discrete regions in a water distribution system, referred to as district metered areas (DMAs), can be a pragmatic approach to diagnose both system and leakage characteristics. Their application, however, has historically been limited in the North American context owing in part to their costly implementation and operational challenges. Both to overcome these barriers and to demonstrate the benefits of DMAs, a leakage testing programme was undertaken in Ontario, Canada. Novelty arises from the development and deployment of a mobile testing unit specifically designed to collect minimum night flow (MNF) and pressure data into temporarily configured DMAs. Moreover, activation of a pressure reducing valve facilitated the direct testing of pressure modulation on leakage reduction. The mobile unit was deployed in 22 DMAs across eight water systems with results indicating a clear relationship between MNFs and system characteristics for well-performing DMAs. MNF benchmarks were quantified to enable an evidence-based assessment of leakage performance at the DMA level in typical Canadian water systems. This project established the proof-of-concept of the mobile unit for providing both accurate and reliable measurements of key leakage performance characteristics and for predicting leakage reduction following system interventions. Abstract Background Despite aggressive treatment, more than 90% of glioblastoma (GBM) patients experience recurrences. GBM response to therapy is currently assessed by imaging techniques and tissue biopsy. However, difficulties with these methods may cause misinterpretation of treatment outcomes. Currently, no validated therapy response biomarkers are available for monitoring GBM progression. Metabolomics holds potential as a complementary tool to improve the interpretation of therapy responses to help in clinical interventions for GBM patients. Methods Saliva and blood from GBM patients were collected pre and postoperatively. Patients were stratified conforming their progression‐free survival (PFS) into favourable or unfavourable clinical outcomes (>9 months or PFS ≤ 9 months, respectively). Analysis of saliva (whole‐mouth and oral rinse) and plasma samples was conducted utilising LC‐QqQ‐MS and LC‐QTOF‐MS to determine the metabolomic and lipidomic profiles. The data were investigated using univariate and multivariate statistical analyses and graphical LASSO‐based graphic network analyses. Results Altogether, 151 metabolites and 197 lipids were detected within all saliva and plasma samples. Among the patients with unfavourable outcomes, metabolites such as cyclic‐AMP, 3‐hydroxy‐kynurenine, dihydroorotate, UDP and cis‐aconitate were elevated, compared to patients with favourable outcomes during pre‐and post‐surgery. These metabolites showed to impact the pentose phosphate and Warburg effect pathways. The lipid profile of patients who experienced unfavourable outcomes revealed a higher heterogeneity in the abundance of lipids and fewer associations between markers in contrast to the favourable outcome group. Conclusion Our findings indicate that changes in salivary and plasma metabolites in GBM patients can potentially be employed as less invasive prognostic biomarkers/biomarker panel but validation with larger cohorts is required.

Abstract A growing number of studies have investigated the feasibility of utilizing hemp by-products as livestock feedstuffs; however, their impact on livestock microbiomes remains unexplored. Here, we evaluated the effects of feeding hempseed cake on the gastrointestinal, respiratory, and reproductive microbiota in beef heifers. Angus-crossbred heifers (19-months old, initial body weight = 494 ± 10 kg [SE]) were fed a corn-based finishing diet containing 20% hempseed cake as a substitute for 20% corn dried distillers’ grains with solubles (DM basis; Control; n = 16/group) for 111 days until slaughter. Ruminal fluid and deep nasopharyngeal swabs (days 0, 7, 42, 70 and 98), and vaginal and uterine swabs (at slaughter) were collected, and the microbiota assessed using 16S rRNA gene sequencing. Nutritional and contaminant stressors influence organismal physiology, trophic interactions, community structure, and ecosystem-level processes; however, the interactions between toxicity and elemental imbalance in food resources have been examined in only a few ecotoxicity studies. Integrating well-developed ecological theories that cross all levels of biological organization can enhance our understanding of ecotoxicology. In the present article, we underline the opportunity to couple concepts and approaches used in the theory of ecological stoichiometry (ES) to ask ecotoxicological questions and introduce stoichiometric ecotoxicology, a subfield in ecology that examines how contaminant stress, nutrient supply, and elemental constraints interact throughout all levels of biological organization. This conceptual framework unifying ecotoxicology with ES offers potential for both empirical and theoretical studies to deepen our mechanistic understanding of the adverse outcomes of chemicals across ecological scales and improve the predictive powers of ecotoxicology.

Nucleation in atomic crystallization remains poorly understood, despite advances in classical nucleation theory. The nucleation process has been described to involve a nonclassical mechanism that includes a spontaneous transition from disordered to crystalline states, but a detailed understanding of dynamics requires further investigation. In situ electron microscopy of heterogeneous nucleation of individual gold nanocrystals with millisecond temporal resolution shows that the early stage of atomic crystallization proceeds through dynamic structural fluctuations between disordered and crystalline states, rather than through a single irreversible transition. Our experimental and theoretical analyses support the idea that structural fluctuations originate from size-dependent thermodynamic stability of the two states in atomic clusters. These findings, based on dynamics in a real atomic system, reshape and improve our understanding of nucleation mechanisms in atomic crystallization.

In operando, ambient-pressure x-ray photoelectron spectroscopy (AP-XPS) has been used to evaluate surface states of gadolinia-doped ceria (GDC) thin-film electrodes during H2 oxidation and H2O electrolysis, on yttria-stabilized zirconia (YSZ)-supported solid oxide electrochemical cells (SOCs). Porous nickel (Ni) and gold (Au) overlayers deposited on separate GDC thin films served as current collectors and potential electrocatalysts to facilitate heterogeneous chemistry for H2 oxidation or H2O electrolysis. Electrochemical characterization of the GDC thin-film electrodes complemented in operando XPS measurements of the O 1s spectra to correlate electrochemical overpotentials with surface chemistry near the Ni/GDC and Au/GDC interfaces. Shifts in O 1s binding energies across the metal/GDC/YSZ interfaces signified changes of local surface potential and provided a means of estimating kinetic parameters associated with charge transfer reactions. Effective oxygen partial pressure and surface potential impacted oxide vacancy and ceria polaron concentrations in the GDC, resulting in different reactivities of the GDC under the tested conditions. Both the Ni/GDC and Au/GDC demonstrated much higher currents for H2O electrolysis vs. H2 oxidation for comparable metal/GDC overpotentials due to increased electronic conductivity of the GDC under positive potentials and associated spreading of the electrochemically active region away from the triple-phase boundary. Higher electrochemical activity of the Ni/GDC electrode is attributed to the increased H2 activation on Ni in promoting charge transfer reactions (particularly for H2 oxidation). These results provide a basis for developing more informed reaction mechanisms for both H2 oxidation and H2O electrolysis of GDC-based composite electrodes in SOCs.

This quantitative study utilized 1,640 P-5 students' learning outcomes as a result of unit instructions that pre-service teachers gave to P-5 students in the field. The study investigated the difference in P-5 student learning outcomes after a unit instruction by three pre-service teaching practicum course tiers, considering socioeconomic statuses, student grade levels, and subject areas of the content taught by the pre-service teacher. Using normalized gain scores, we used a t test and regression analysis to analyze the data. Based on the findings, recommendations for pre-service teacher education include three items: a) to provide differentiated supervision based on pre-service teachers' experiences and needs, b) to require a proportionate and incremental assignment responding to the amount of pre-service teachers' experiences and hours in the classroom, and c) to incorporate co-teaching opportunities to facilitate peer learning and support in early field experiences.

IntroductionThe use of microorganisms as drug delivery systems to treat cancer has expanded recently, including FDA approval of certain viruses as oncolytics. Microorganisms have several unique benefits compared to traditional pharmacologic agents including dose independence, the ability to produce therapeutic proteins locally within the tumor, and simplicity of administration. However, current microbial delivery systems such as AAV9 and herpes virus have limited cassette sizes, minimal cancer cell selectivity, and low innate cytotoxicity. To address these issues, we sought to generate a strain of Shigella flexneri to selectively internalize into glioblastoma (GBM) brain tumor cells as an initial step to generating a bacterial-based drug delivery system.MethodsWe generated S. flexneri that selectively internalize into GBM cells using iterative co-cultured assays.ResultsAfter 50 rounds of co-culture, the new strain infected 95 percent of GBM cells in 2 hours. GBM-infecting Shigella demonstrate a 124-fold preference for internalizing in nine different GBM cell lines compared to Normal Astrocytes (NA) controls. Additionally, we developed an in-cell western to identify GBM-infecting Shigella clones that preferentially internalize in patient samples without iterative co-culture. Finally, we demonstrate internalization into GBM cells is mediated via a factor modified by myristoylation.DiscussionIn conclusion, here we present a novel bacterial platform that preferentially internalizes in brain tumor cells. This system provides numerous potential benefits over current interventions and other microbial strategies for treating brain tumors.

Objectives Non-compressible torso hemorrhage remains a high mortality injury, with difficulty mobilizing resources before exsanguination. Previous studies reported on a retrievable stent graft for damage control and morphometric algorithms for rapid placement, yet fluoroscopy is impractical for the austere environment. We hypothesized that magnetic sensors could be used to position stents relative to an external magnet placed on an anatomic landmark, whereas an electromagnet would allow self-calibration to account for environmental noise.Methods A magnetic sensor alone (MSA) and with integrated stent (MSIS) were examined in a porcine model under anesthesia. A target electromagnet was placed on the xiphoid process (position 0 cm). Sensors were placed in the aorta and measurements obtained at positions 0 cm, +4 cm, and +12 cm from the magnet and compared with fluoroscopy. Sensors were examined under conditions of tachycardia/hypertension, hypotension, vibration, and metal shrapnel to simulate environmental factors that might impact accuracy. General linear models compared mean differences between fluoroscopy and sensor readings.Results Both sensors were compatible with a 10 French catheter system and provided real-time assessment of the distance between the sensor and magnetic target in centimeters. Mean differences between fluoroscopy and both magnetic sensor readings demonstrated accuracy within ±0.5 cm for all but one condition at 0 cm and +4 cm, whereas accuracy decreased at +12 cm from the target. Using the control as a reference, there was no significant difference in mean differences between fluoroscopy and both MSA or MSIS readings at 0 cm and +4 cm for all conditions. The system retained effectiveness if the target was overshot.Conclusion Magnetic sensors achieved the highest accuracy as sensors approached the target. Oscillation of the electromagnet on and off effectively accounts for environmental noise.This approach is promising for rapid and accurate placement of damage control retrievable stent grafts when fluoroscopy is impractical.Level of evidence Not applicable.

The lens proteome undergoes dramatic composition changes during development and maturation. A defective developmental process leads to congenital cataracts that account for about 30% of cases of childhood blindness. Gene mutations are associated with approximately 50% of early-onset forms of lens opacity, with the remainder being of unknown etiology. To gain a better understanding of cataractogenesis, we utilized a transgenic mouse model expressing a mutant ubiquitin protein in the lens (K6W-Ub) that recapitulates most of the early pathological changes seen in human congenital cataracts. We performed mass spectrometry-based tandem-mass-tag quantitative proteomics in E15, P1, and P30 control or K6W-Ub lenses. Our analysis identified targets that are required for early normal differentiation steps and altered in cataractous lenses, particularly metabolic pathways involving glutathione and amino acids. Computational molecular phenotyping revealed that glutathione and taurine were spatially altered in the K6W-Ub cataractous lens. High-performance liquid chromatography revealed that both taurine and the ratio of reduced glutathione to oxidized glutathione, two indicators of redox status, were differentially compromised in lens biology. In sum, our research documents that dynamic proteome changes in a mouse model of congenital cataracts impact redox biology in lens. Our findings shed light on the molecular mechanisms associated with congenital cataracts and point out that unbalanced redox status due to reduced levels of taurine and glutathione, metabolites already linked to age-related cataract, could be a major underlying mechanism behind lens opacities that appear early in life.

Cerium oxide (ceria, CeO2) is a technologically important material for energy conversion applications. Its activities strongly depend on redox states and oxygen vacancy concentration. Understanding the functionality of chemical active species and behavior of oxygen vacancy during operation, especially in high-temperature solid-state electrochemical cells, is the key to advance future material design. Herein, the structure evolution of ceria is spatially resolved using bulk-sensitive operando X-ray diffraction and spectroscopy techniques. During water electrolysis, ceria undergoes reduction, and its oxygen non-stoichiometry shows a dependence on the electrochemical current. Cerium local bonding environments vary concurrently to accommodate oxygen vacancy formation, resulting in changes in Ce-O coordination number and Ce3+/Ce4+ redox couple. When reduced enough, a crystallographic phase transition occurs from α to an α' phase with more oxygen vacancies. Nevertheless, the transition behavior is intriguingly different from the one predicted in the standard phase diagram of ceria. This paper demonstrates a feasible means to control oxygen non-stoichiometry in ceria via electrochemical potential. It also sheds light on the mechanism of phase transitions induced by electrochemical potential. For electrochemical systems, effects from a large-scale electrical environment should be taken into consideration, besides effective oxygen partial pressure and temperature.

This special issue of Foundations of Physics collects together articles representing some recent new perspectives on the hole argument in the history and philosophy of physics. Our task here is to introduce those new perspectives.

Abstract Freshwater ecosystems are experiencing increased salinization. Adaptive management of harmful algal blooms (HABs) contributes to eutrophication/salinization interactions through the hydrologic transport of blooms to coastal environments. We examined how nutrients and salinity interact to affect growth, elemental composition, and cyanotoxin production/release in two common HAB genera. Microcystis aeruginosa (non‐nitrogen [N]‐fixer and microcystin‐LR producer [MC‐LR]) and Aphanizomenon flos‐aquae (N‐fixer and cylindrospermopsin producer [CYN]) were grown in N : phosphorus (N : P) 4 and 50 (by atom) for 21 and 33 d, respectively, then dosed with a salinity gradient (0–10.5 g L−1). Both total MC‐LR and CYN were correlated with particulate N. We found Microcystis MC‐LR production and release was affected by salinity only in the N : P 50 treatment. However, Aphanizomenon CYN production and release was affected by salinity regardless of N availability. Our results highlight how cyanotoxin production and release across the freshwater–marine continuum are controlled by ecophysiological differences between N‐acquisition traits.

In retinal degenerative diseases, such as retinitis pigmentosa (RP) and age-related macular degeneration (AMD), the photoreceptors become stressed and start to degenerate in the early stages of the disease. Retinal prosthetic devices have been developed to restore vision in patients by applying electrical stimulation to the surviving retinal cells. However, these devices provide limited visual perception as the therapeutic interventions are generally considered in the later stages of the disease when only inner retinal layer cells are left. A potential treatment option for retinal degenerative diseases in the early stages can be stimulating bipolar cells, which receive presynaptic signals from photoreceptors. In this work, we constructed computational models of healthy and degenerated (both ON and OFF-type) cone bipolar cells (CBCs) with realistic morphologies extracted from connectomes of the healthy and early-stage degenerated rabbit retina. We examined these cells’ membrane potential and axon terminal calcium current differences when subjected to electrical stimulation. In addition, we investigated how differently healthy and degenerated cells behave with respect to various stimulation parameters, including pulse duration and cells’ distance from the stimulating electrode. The results suggested that regardless of the position of the OFF CBCs in the retina model, there is not a significant difference between the membrane potential of healthy and degenerate cells when electrically stimulated. However, the healthy ON CBC axon terminal membrane potential rising time-constant is shorter (0.29 ± 0.03 ms) than the degenerated cells (0.8 ± 0.07 ms). Moreover, the ionic calcium channels at the axon terminals of the cells have a higher concentration and higher current in degenerated cells (32.24 ± 6.12 pA) than the healthy cells (13.64 ± 2.88 pA) independently of the cell’s position.

The systems for multiphoton 3D nanoprinting are rapidly increasing in print speed for larger throughput and scale, unfortunately without also improvement in resolution. Separately, the process of photoinhibition lithography has been demonstrated to enhance the resolution of multiphoton printing through the introduction of a secondary laser source. The photo-chemical dynamics and interactions for achieving photoinhibition in the various multiphoton photoinitiator systems are complex and still not well understood. Here, we examine the photoinhibition process of the common photoinitiator 7-diethylamino 3-thenoylcoumarin (DETC) with inhibition lasers near or at the multiphoton printing laser wavelength in typical low peak intensity, high repetition rate 3D nanoprinting processes. We demonstrate the clear inhibition of the polymerization process consistent with a triplet absorption deactivation mechanism for a DETC photoresist as well as show inhibition for several other photoresist systems. Additionally, we explore options to recover the photoinhibition process when printing with high intensity, low repetition rate lasers. Finally, we demonstrate photoinhibition in a projection multiphoton printing system. This investigation of photoinhibition lithography with common photoinitiators elucidates the possibility for photoinhibition occurring in many resist systems with typical high repetition rate multiphoton printing lasers as well as for high-speed projection multiphoton printing.

The aim of this project was to determine research priorities, barriers, and enablers for adult primary brain tumour research in Australia and New Zealand. Consumers, health professionals, and researchers were invited to participate in a two-phase modified Delphi study. Phase 1 comprised an initial online survey (n = 91) and then focus groups (n = 29) which identified 60 key research topics, 26 barriers, and 32 enablers. Phase 2 comprised two online surveys to (1) reduce the list to 37 research priorities which achieved consensus (>75% 2-point agreement) and had high mean importance ratings (n = 116 participants) and (2) determine the most important priorities, barriers, and enablers (n = 90 participants). The top ten ranked research priorities for the overall sample and sub-groups (consumers, health professionals, and researchers) were identified. Priorities focused on: tumour biology, pre-clinical research, clinical and translational research, and supportive care. Variations were seen between sub-groups. The top ten barriers to conducting brain tumour research related to funding and resources, accessibility and awareness of research, collaboration, and process. The top ten research enablers were funding and resources, collaboration, and workforce. The broad list of research priorities identified by this Delphi study, together with how consumers, health professionals, and researchers prioritised items differently, and provides an evidence-based research agenda for brain tumour research that is needed across a wide range of areas.
