- Abstract
OBJECTIVE: Atherosclerosis develops near branches and bends of arteries that are exposed to low shear stress (mechanical drag). These sites are characterized by excessive endothelial cell (EC) proliferation and inflammation that promote lesion initiation. The transcription factor HIF1α (hypoxia-inducible factor 1α) is canonically activated by hypoxia and has a role in plaque neovascularization. We studied the influence of shear stress on HIF1α activation and the contribution of this noncanonical pathway to lesion initiation. APPROACH AND RESULTS: Quantitative polymerase chain reaction and en face staining revealed that HIF1α was expressed preferentially at low shear stress regions of porcine and murine arteries. Low shear stress induced HIF1α in cultured EC in the presence of atmospheric oxygen. The mechanism involves the transcription factor nuclear factor-κB that induced HIF1α transcripts and induction of the deubiquitinating enzyme Cezanne that stabilized HIF1α protein. Gene silencing revealed that HIF1α enhanced proliferation and inflammatory activation in EC exposed to low shear stress via induction of glycolysis enzymes. We validated this observation by imposing low shear stress in murine carotid arteries (partial ligation) that upregulated the expression of HIF1α, glycolysis enzymes, and inflammatory genes and enhanced EC proliferation. EC-specific genetic deletion of HIF1α in hypercholesterolemic apolipoprotein E-defecient mice reduced inflammation and endothelial proliferation in partially ligated arteries, indicating that HIF1α drives inflammation and vascular dysfunction at low shear stress regions. CONCLUSIONS: Mechanical low shear stress activates HIF1α at atheroprone regions of arteries via nuclear factor-κB and Cezanne. HIF1α promotes atherosclerosis initiation at these sites by inducing excessive EC proliferation and inflammation via the induction of glycolysis enzymes.

By operating an antineutrino detector of simple design during several fuel cycles, we observed long term changes in antineutrino flux that result from the isotopic evolution of a commercial pressurized water reactor. Measurements made with simple antineutrino detectors of this kind offer an alternative means for verifying fissile inventories at reactors, as part of International Atomic Energy Agency and other reactor safeguards regimes. - Abstract
The US neutrino community gathered at the Workshop on the Intermediate Neutrino Program (WINP) at Brookhaven National Laboratory February 4-6, 2015 to explore opportunities in neutrino physics over the next five to ten years. Scientists from particle, astroparticle and nuclear physics participated in the workshop. The workshop examined promising opportunities for neutrino physics in the intermediate term, including possible new small to mid-scale experiments, US contributions to large experiments, upgrades to existing experiments, R&D plans and theory. The workshop was organized into two sets of parallel working group sessions, divided by physics topics and technology. Physics working groups covered topics on Sterile Neutrinos, Neutrino Mixing, Neutrino Interactions, Neutrino Properties and Astrophysical Neutrinos. Technology sessions were organized into Theory, Short-Baseline Accelerator Neutrinos, Reactor Neutrinos, Detector R&D and Source, Cyclotron and Meson Decay at Rest sessions.This report summarizes discussion and conclusions from the workshop.

- Abstract
Aims: Stent deployment causes endothelial cells (EC) denudation, which promotes in-stent restenosis and thrombosis. Thus endothelial regrowth in stented arteries is an important therapeutic goal. Stent struts modify local hemodynamics, however the effects of flow perturbation on EC injury and repair are incompletely understood. By studying the effects of stent struts on flow and EC migration, we identified an intervention that promotes endothelial repair in stented arteries. Methods and Results: In vitro and in vivo models were developed to monitor endothelialization under flow and the influence of stent struts. A 2D parallel-plate flow chamber with 100lm ridges arranged perpendicular to the flow was used. Live cell imaging coupled to computational fluid dynamic simulations revealed that EC migrate in the direction of flow upstream from the ridges but subsequently accumulate downstream from ridges at sites of bidirectional flow. The mechanism of EC trapping by bidirectional flow involved reduced migratory polarity associated with altered actin dynamics. Inhibition of Rho-associated protein kinase (ROCK) enhanced endothelialization of ridged surfaces by promoting migratory polarity under bidirectional flow (P < 0.01). To more closely mimic the in vivo situation, we cultured EC on the inner surface of polydimethylsiloxane tubing containing Coroflex Blue stents (65 lm struts) and monitored migration. ROCK inhibition significantly enhanced EC accumulation downstream from struts under flow (P < 0.05). We investigated the effects of ROCK inhibition on re-endothelialization in vivo using a porcine model of EC denudation and stent placement. En face staining and confocal microscopy revealed that inhibition of ROCK using fasudil (30 mg/day via osmotic minipump) significantly increased re-endothelialization of stented carotid arteries (P < 0.05). The Double Chooz experiment has determined the value of the neutrino oscillation parameter $��_{13}$ from an analysis of inverse beta decay interactions with neutron capture on hydrogen. This analysis uses a three times larger fiducial volume than the standard Double Chooz assessment, which is restricted to a region doped with gadolinium (Gd), yielding an exposure of 113.1 GW-ton-years. The data sample used in this analysis is distinct from that of the Gd analysis, and the systematic uncertainties are also largely independent, with some exceptions, such as the reactor neutrino flux prediction. A combined rate- and energy-dependent fit finds $\sin^2 2��_{13}=0.097\pm 0.034(stat.) \pm 0.034 (syst.)$, excluding the no-oscillation hypothesis at 2.0 ��. An observed-to-predicted ratio of events of 0.944±0.016(stat)±0.040(syst) was obtained in 101 days of running at the Chooz nuclear power plant in France, with two 4.25GWth reactors. The results were obtained from a single 10m3 fiducial volume detector located 1050 m from the two reactor cores. The reactor antineutrino flux prediction used the Bugey4 flux measurement after correction for differences in core composition. The deficit can be interpreted as an indication of a nonzero value of the still unmeasured neutrino mixing parameter sin22θ13. Analyzing both the rate of the prompt positrons and their energy spectrum, we find sin22θ13=0.086±0.041(stat)±0.030(syst), or, at 90% C.L., 0.017

We report on several features present in the energy spectrum from an ultra low-noise germanium detector operated at 2,100 m.w.e. By implementing a new technique able to reject surface events, a number of cosmogenic peaks can be observed for the first time. We discuss several possible causes for an irreducible excess of bulk-like events below 3 keVee, including a dark matter candidate common to the DAMA/LIBRA annual modulation effect, the hint of a signal in CDMS, and phenomenological predictions. - Abstract
Double Chooz is unique among modern reactor-based neutrino experiments studying (nu) over bar (e) disappearance in that data can be collected with all reactors off. In this paper, we present data from 7.53 days of reactor-off running. Applying the same selection criteria as used in the Double Chooz reactor-on oscillation analysis, a measured background rate of 1.0 +/- 0.4 events/day is obtained. The background model for accidentals, cosmogenic beta-n-emitting isotopes, fast neutrons from cosmic muons, and stopped-mu decays used in the oscillation analysis is demonstrated to be correct within the uncertainties. Kinematic distributions of the events, which are dominantly cosmic-ray-produced correlated-background events, are provided. The background rates are scaled to the shielding depths of two other reactor-based oscillation experiments, Daya Bay and RENO.

- Abstract
We present a search for Lorentz violation with 8249 candidate electron antineutrino events taken by the Double Chooz experiment in 227.9 live days of running. This analysis, featuring a search for a sidereal time dependence of the events, is the first test of Lorentz invariance using a reactor-based antineutrino source. No sidereal variation is present in the data and the disappearance results are consistent with sidereal time independent oscillations. Under the Standard-Model Extension, we set the first limits on 14 Lorentz violating coefficients associated with transitions between electron and tau flavor, and set two competitive limits associated with transitions between electron and muon flavor.

- Abstract
The Double Chooz experiment has observed 8249 candidate electron antineutrino events in 227.93 live days with 33.71 GW-ton-years (reactor power X detector mass X live time) exposure using a 10.3 m(3) fiducial volume detector located at 1050 m from the reactor cores of the Chooz nuclear power plant in France. The expectation in case of theta(13) = 0 is 8937 events. The deficit is interpreted as evidence of electron antineutrino disappearance. From a rate plus spectral shape analysis we find sin(2)2 theta(13) = 0.109 +/- 0.030(stat) +/- 0.025(syst). The data exclude the no-oscillation hypothesis at 99.8% CL (2.9 sigma).

- Abstract
Analysis of global microRNA (miRNA) expression in postmortem cortical grey matter from the superior temporal gyrus, revealed significant up-regulation of miR-181b expression in schizophrenia. This finding was supported by quantitative real-time RT-PCR analysis of miRNA expression in a cohort of 21 matched pairs of schizophrenia and non-psychiatric controls. The implications of this finding are substantial, as this miRNA is predicted to regulate many target genes with potential significance to the development of schizophrenia. They include the calcium sensor gene visinin-like 1 (VSNL1) and the ionotropic AMPA glutamate receptor subunit (GRIA2), which were found to be down-regulated in the same cortical tissue from the schizophrenia group. Both of these genes were also suppressed in miR-181b transfected cells and shown to contain functional miR-181b miRNA recognition elements by reporter gene assay. This study suggests altered miRNA levels could be a significant factor in the dysregulation of cortical gene expression in schizophrenia. © 2008 The Author(s).

Only our ATRAP Collaboration is yet able to accumulate and store 4.2 K antiprotons and positrons. The antiprotons come initially from the new Antiproton Decel- erator facility at CERN. Good control of such cold antimatter plasmas is key to aspirations to produce and study antihydrogen atoms that are cold enough to confine by their magnetic moments. In the closest approach to cold antihydrogen realized to date the cold positrons have been used to cool antiprotons the first time that positron cooling has ever been oh- served. - Abstract
This paper describes the spontaneous formation of patterns of aligned buckles in a thin film of gold deposited on the surface of an elastomer [poly(dimethylsiloxane), PDMS]. The surface of the elastomer is patterned photochemically into areas differing in stiffness and coefficient of thermal expansion. The gold is deposited while the surface of the patterned elastomer is warm (T ∼ 100 °C). On cooling, shrinkage in the elastomer places the gold film under compressive stress. The buckles relieve this compressive stress. The distribution of stresses and buckle patterns is described during the pre- and postbuckling regimes using solutions from calculations describing a model comprising a thin stiff plate resting on a thick elastic foundation.
