The effects of naltrexone injected intravenously (i.v.) on the pharmacological actions and distribution of i.v. injected morphine in brain regions and spinal cord of male Sprague-Dawley rats were determined. Naltrexone (0.625- and 2.5-mg/kg doses) antagonized the analgesic and hyperthermic effects of morphine (10-mg/kg dose). For distribution studies, naltrexone (0.625- and 2.5-mg/kg doses) was co-administered with morphine via indwelling catheters. Rats were sacrificed at various times after drug injection and the concentration of morphine in brain regions (hypothalamus, hippocampus, cortex, pons and medulla, amygdala, midbrain and corpus striatum), spinal cord and serum was determined by radioimmunoassay. The concentration of morphine in various brain regions was found to be time dependent. Initially, at 5 min, the highest concentration of morphine was found in the hypothalamus and the lowest in the striatum. In cortex and spinal cord, the concentration of morphine was significantly higher in comparison to the other brain regions at 30- and 60-min time points. Co-administration of lower dose of naltrexone (0.625 mg/kg) did not significantly alter the distribution of morphine in brain regions and spinal cord with some exceptions. The higher dose of naltrexone (2.5 mg/kg) increased the concentration of morphine in several brain regions and spinal cord. The ratio of the concentration of morphine in brain region or spinal cord to serum was decreased by naltrexone. It is concluded that naltrexone also alters the distribution of morphine in the central nervous system.

The effect of morphine tolerance-dependence and abstinence on the characteristics of dopamine D2 receptors in brain regions and spinal cord was determined in the rat. Male Sprague-Dawley rats were implanted s.c. under light ether anesthesia with 6 morphine pellets for a 7-day period, each containing 75 mg of morphine free base. Rats implanted with placebo pellets served as controls. This procedure resulted in the development of tolerance to morphine as evidenced by decreased analgesic response to a challenge dose of morphine. Similarly, the development of physical dependence was evidenced by a decrease in body weight and colonic temperature after morphine pellet removal (withdrawal). The binding characteristics (Bmax and Kd values) of [3H]spiroperidol to dopamine D2 receptors were determined in the tissues of morphine-tolerant and morphine-abstinent rats. In the tolerant rats, the pellets were left intact at the time of sacrificing, whereas, in the abstinent rats the pellets were removed 18 h prior to sacrificing. The binding of [3H]spiroperidol was determined in membranes prepared from brain regions (hypothalamus, hippocampus, cortex, pons and medulla, midbrain, corpus striatum and amygdala) and spinal cord of rats from various treatment groups. [3H]Spiroperidol bound to brain regions and spinal cord at a single high affinity site. The Bmax or the Kd values in brain regions and spinal cord of morphine-tolerant and -abstinent rats did not differ from their respective placebo controls. The behavioral responses to a selective dopamine D2 receptor agonist, 2-bromo-alpha-ergocryptine were also determine in the morphine-abstinent rats.(ABSTRACT TRUNCATED AT 250 WORDS)

Male Sprague-Dawley rats were rendered tolerant to and physically dependent on U-50,488H, a kappa-opiate agonist, by injecting 25 mg/kg of the drug intraperitoneally twice a day for 4 days. Two sets of rats were used. Rats labeled as tolerant-dependent were injected with U-50,488H (25 mg/kg) 1 h before sacrificing on day 5, whereas the abstinent rats were sacrificed on day 5 without the injection of U-50,488H. Of all the tissues on day 5 without the injection of U-50,488H. Of all the tissues examined, the pituitary gland had the highest level of dynorphin (1-13), whereas the heart had the lowest level. The levels of dynorphin (1-13) increased in the hypothalamus, hippocampus and pons/medulla of U-50,488H tolerant-dependent rats, whereas in abstinent rats the levels of dynorphin (1-13) were elevated only in the midbrain. The levels of dynorphin (1-13) in the pituitary gland of U-50,488H tolerant-dependent or abstinent rats were unchanged. In peripheral tissues, the levels of dynorphin (1-13) in the heart of U-50,488H tolerant-dependent rats were increased. In the abstinent rats they were elevated in the adrenals, spleen, and the heart but were decreased in the kidneys. Compared to morphine tolerant-dependent and abstinent rats, significant differences in the levels of dynorphin (1-13) in tissues of 50,488H tolerant-dependent and abstinent rats were observed and may explain many pharmacological differences in the mu- and kappa-opiate induced tolerance-dependence and abstinence processes.

Phencyclidine hydrochloride (PCP) was tested for its ability to alter a variety of immune effector and regulatory functions in vitro. B6C3F1 murine splenic lymphocytes or elicited peritoneal macrophages were cultured in vitro with medium only or medium containing 10(-10)-10(-4) M PCP. Macrophages cultured with or without PCP were stimulated with lipopolysaccharide, and production of interleukin 6 (IL-6) and tumor necrosis factor (TNF) was assessed by bioassay. Cytotoxic T-cell effector function was determined following 5-day lymphocyte co-culture with tumor stimulator cells in the presence of PCP. In addition, the ability of T-lymphocytes to produce specific immunoregulatory cytokines IL-2 and IL-4 in the presence of PCP was quantitated by bioassay. B-lymphocyte function was determined by quantitating lymphocyte proliferation following stimulation with anti-IgM antibody and murine IL-4. Natural immunity was assessed by culturing lymphocytes with or without PCP for 24 h, then quantitating basal and IL-2 augmented natural killer (NK) cell activity. In the absence of effects on cell viability, significant suppression of IL-2 production by T-cells was noted at pharmacologically relevant PCP concentrations (1 microM). In vitro concentrations of 10 microM suppressed the generation of specifically sensitized cytotoxic T-cells. In addition, PCP significantly suppressed both IL-2-augmented NK function as well as B-lymphocyte proliferation. By comparison, macrophage IL-6 production was not affected by any concentration of PCP examined in this study.

Effects of naltrexone administered intravenously on the pharmacological actions and kinetics of morphine in serum following intravenous administration of morphine were determined in male Sprague-Dawley rats. A 10 mg/kg dose of morphine produced an analgesic response as measured by the tail flick test. Morphine also produced a hyperthermic effect. Naltrexone dose (0.625-2.5 mg/kg)-dependently antagonized the analgesic and hyperthermic effects of morphine. The effect of naltrexone (0.625 and 2.5 mg/kg) on the pharmacokinetic parameters area under the serum morphine concentration time curve (AUC0-->infinity), serum levels of morphine extrapolated to zero time (Cmax), half-life (t1/2), mean residence time (MRT), total body clearance (Clt), and volume of distribution at steady state (Vss) of morphine in serum was determined. Naltrexone (0.625 mg/kg) significantly increased AUC0-->infinity, Cmax, t1/2 MRT, but decreased the Vss, elimination rate constant (k) and Clt. The higher dose of naltrexone (2.5 mg/kg) produced an increase in the Cmax value of morphine in the serum, but the other pharmacokinetic parameters were unaffected. Since increased morphine concentrations in serum produced by naltrexone cannot explain its antagonistic effects on analgesia and hyperthermia, it is concluded that naltrexone produces its effects by blocking opiate receptors at the appropriate sites. The increases in serum morphine levels by naltrexone may be related to displacement of morphine from protein binding sites and inhibition of morphine metabolism by glucuronyl transferase. This study for the first time demonstrates that in the rat, when morphine and naltrexone are given concurrently, although serum levels of morphine increase, pharmacological effects of morphine are antagonized.(ABSTRACT TRUNCATED AT 250 WORDS)

The binding of 3H-D-Pen2-D-Pen5-enkephalin (DPDPE), a highly selective delta-opiate receptor agonist, to membranes of discrete brain regions and spinal cord of 10-week-old spontaneously hypertensive (SHR) and normotensive Wistar-Kyoto (WKY) rats was determined. The brain regions examined were amygdala, hippocampus, hypothalamus, pons and medulla, corpus striatum, midbrain and cortex. 3H-DPDPE bound to membranes of brain regions and spinal cord at a single high affinity site with apparent dissociation constant value of 4 nmol/l, except in cortex where the values were higher. The highest density of 3H-DPDPE binding sites were in hypothalamus and lowest in pons and medulla. The receptor density (Bmax value) and apparent dissociation constant (Kd value) of 3H-DPDPE to bind to delta-opiate receptors on the membranes of hippocampus, hypothalamus, corpus striatum, midbrain, cortex, pons and medulla and spinal cord of WKY and SHR rats did not differ. The Bmax value of 3H-DPDPE in amygdala of SHR rats was higher than in WKY rats, but the Kd values in the two strains did not differ. It is concluded that SHR rats have a higher density of delta-opiate receptors, labeled with 3H-DPDPE, in amygdala in comparison with WKY rats. Whether such a difference in the density of delta-opiate receptors is related to the elevated blood pressure of SHR rats is not clear.

The effect of trans-(+/-)-3,4-dichloro-N-methyl-N-[2-(1-pyrrolidinyl) cyclohexyl]-benzene-acetamide methane sulfonate (U-50,488H), a kappa-opiate agonist-induced tolerance and abstinence on 5-HT1A receptors was determined in regions of the brain and spinal cord of the rat. The administration of U-50,488H (25 mg/kg, i.p., twice daily) to male Sprague-Dawley rats for 4 days resulted in the development of almost complete tolerance to its analgesic and hypothermic effects. On day 5, the animals were divided into tolerant and abstinent groups and sacrificed. The brain and spinal cord were excised from all groups of rats and the brain was dissected into 6 regions, namely, amygdala, hypothalamus, striatum, midbrain, pons+medulla and cortex. The 5-HT1A receptors were characterized by using [3H]8-hydroxy-2-(di-n-propylamino)tetralin ([3H]DPAT) as the ligand. The binding constants (Bmax and Kd values) of [3H]DPAT in regions of the brain and spinal cord of rats tolerant to U-50,488H and vehicle did not differ. However, the Bmax value of [3H]DPAT in the hypothalamus of U-50,488H-abstinent rats was decreased but the Kd value did not change. In the other regions of the brain and spinal cord of U-50,488H-abstinent rats, the Bmax and Kd values of [3H]DPAT were unaffected. Subcutaneous administration of DPAT produced hypothermic response in vehicle- and U-50,488H-treated rats. The intensity of this effect was more marked in U-50,488H-abstinent group. It is concluded that 5-HT1A receptors are down-regulated in the hypothalamus of U-50,488H-abstinent rats but the hypothermic response to 5-HT1A agonist is intensified.

In order to determine the possible contribution of altered distribution of morphine in the morphine tolerance process, the distribution of morphine was studied in brain regions and spinal cord, following its intravenous administration. Male Sprague-Dawley rats were made tolerant to morphine by implanting 6 morphine pellets, each containing 75 mg of morphine base, for 7 days. Seventy-two hours after the removal of the pellets, a time when serum morphine levels were negligible or absent and yet tolerance to the pharmacological effects of morphine was present, morphine (10 mg/kg, i.v.) was injected in placebo and morphine pellet implanted rats. At various times (5, 30, 60, 120 and 360 min) after the injection of morphine, brain regions (hypothalamus, cortex, hippocampus, midbrain, pons and medulla, striatum and amygdala), spinal cord and serum were collected. The level of morphine in the tissues was determined by using a highly sensitive and specific radioimmunoassay (RIA) method. Five minutes after morphine injection, the concentration of morphine was the highest in the hypothalamus and the lowest in amygdala. The concentration of morphine in hypothalamus, pons and medulla, hippocampus and midbrain of morphine tolerant rats was smaller than in placebo pellet implanted rats. The tissue to serum ratio of morphine in the hypothalamus, hippocampus, striatum, midbrain and cortex were also smaller in morphine tolerant than in non-tolerant rats. The concentration of morphine in brain regions with time did not exhibit linearity. At other time intervals like 30 and 60 min, the concentration of morphine in several brain regions and spinal cord was significantly higher in morphine tolerant than in non-tolerant rats.(ABSTRACT TRUNCATED AT 250 WORDS)

1. The possible role of pharmacokinetics of morphine in the development of tolerance to the analgesic and hyperthermic effects of morphine was studied in the rat. 2. Male Sprague-Dawley rats were made tolerant to morphine by implanting 6 morphine pellets each containing 75 mg of morphine base for 7 days. The assessment of the degree of tolerance to morphine and pharmacokinetic parameters were done 72 hr after pellet removal. 3. Tolerance developed to both the analgesic and hyperthermic effects of morphine as evidenced by decreased responses to morphine in morphine pellet implanted rats compared with placebo pellet implanted rats. 4. The pharmacokinetic parameters, AUC0-->infinity, Cmax, t1/2, k, MRT, Vss and Clt were determined after injecting 5 and 10 mg/kg doses of morphine intravenously to placebo and morphine pellet implanted rats and using a highly sensitive and specific RIA method to quantitate serum levels of morphine. For a 5 mg/kg dose of morphine, the AUC0-->infinity and t1/2 in morphine pellet implanted rats were significantly higher than in placebo pellet implanted rats, but the k value was lower. The other pharmacokinetic parameters for morphine in the two treatment groups did not differ. For 10 mg/kg dose, the only change was an increase in the MRT in morphine tolerant rats when compared to nontolerant rats. 5. The results establish that the development of tolerance to the analgesic and hyperthermic effects of morphine is not related to pharmacokinetics of morphine in serum but may be related to modification of receptor systems in the central nervous system.

beta-Endorphin, an opioid peptide that contains 31-amino acids derived from proopiomelanocortin, has been implicated in a variety of disorders. To understand its role in pathophysiological states, its levels have been determined in body fluids, particularly in serum or plasma, with the use of commercially available radiometric assay kits. Because the circulating levels of this endogenous opioid peptide are small and antibodies can cross-react with chemically related peptides to different degrees and give rise to faulty interpretation of the data, the performance characteristics of the available radioimmunoassay kits for beta-endorphin from the Immuno Nuclear Corporation, Stillwater, Minn, the New England Nuclear Corporation, Boston, Mass, and the Nichols Institute, San Juan Capistrano, Calif, were evaluated. In overall performance, the Nichols kit was found to be the most reliable in this laboratory, even though the sensitivity of the Immuno Nuclear kit was better at lower concentrations of beta-endorphin. Serum was better than plasma in terms of recovery of beta-endorphin. When collected on ice, no significant loss in beta-endorphin immunoreactivity was observed at 1 hour. The use of edetic acid (EDTA), siliconized edetic acid, and aprotinin (Trasylol)-added siliconized edetic acid tubes was not helpful in improving the performance of the assay. The optimal condition to collect the specimen was to use glass tubes, place the glass tubes on ice immediately, separate the serum, and freeze the sample within 1 hour of collection.

The effect of morphine tolerance dependence and protracted abstinence on the levels of dynorphin (1-13) in discrete brain regions, spinal cord, pituitary gland and peripheral tissues was determined in male Sprague-Dawley rats. Of all the tissues examined, the highest level of dynorphin (1-13) was found to be in the pituitary gland. Among the brain regions and spinal cord examined, the levels of dynorphin (1-13) in descending order were: hypothalamus, spinal cord, midbrain, pons and medulla, hippocampus, cortex, amygdala and striatum. The descending order for the levels of dynorphin (1-13) in peripheral tissues was: adrenals, heart and kidneys. In morphine tolerant rats, the levels of dynorphin (1-13) increased in amygdala but were decreased in pons and medulla. In morphine abstinent rats, the levels of dynorphin (1-13) were increased in amygdala, hypothalamus and hippocampus. The levels of dynorphin (1-13) were increased in pituitary but decreased in spinal cord and remained so even during protracted abstinence. The levels of dynorphin (1-13) in the peripheral tissues of morphine tolerant rats were unaffected. However, in the heart and kidneys of morphine abstinent rats, the levels of dynorphin (1-13) were increased significantly. It is concluded that both morphine tolerance and abstinence modify the levels of dynorphin (1-13) in pituitary, central and peripheral tissues. Morphine abstinence differed from non-abstinence process in that there were additional changes (increases) in the levels of dynorphin (1-13) in brain regions (hypothalamus and hippocampus) and peripheral tissues (heart and kidneys) and may contribute to the symptoms of the morphine abstinence syndrome.(ABSTRACT TRUNCATED AT 250 WORDS)

Previous studies from this laboratory have demonstrated that the analgesic and hyperthermic effects of morphine were found to be greater in spontaneously hypertensive (SHR) rats than in normotensive Wistar-Kyoto (WKY) rats. The enhanced response to morphine could not be explained on the basis of any of the pharmacokinetic parameters of morphine in the serum. In order to determine the possible contribution of altered distribution of morphine in the central nervous system in the differences in the pharmacological response to morphine in the two strains, the time course of the distribution of morphine was determined in brain regions and spinal cord after its i.v. administration. SHR and WKY rats were injected with morphine (10 mg/kg). At various times (5, 30, 60, 120 and 360 min) after the injection of morphine, brain regions (hypothalamus, cortex, hippocampus, midbrain, pons and medulla, striatum and amygdala) and spinal cord were collected. The level of morphine in the tissues was determined by using a highly sensitive and specific radioimmunoassay method. Five minutes after morphine injection, the concentration of morphine was the highest in the spinal cord. Among the brain regions, the highest concentration of morphine was in the hypothalamus and the lowest in the amygdala. In all the brain regions and spinal cord, the concentration of morphine was significantly higher in the SHR than in the WKY rats. Similar effects were observed at 30, 60 and 120 min after morphine injection. The analgesic and hyperthermic effects of i.v. administered morphine were determined in age-matched male spontaneously hypertensive (SHR) and normotensive Wistar-Kyoto (WKY) rats. Two doses of morphine (5 and 10 mg/kg) were used. In the doses used, morphine produced analgesia in WKY rats as measured by the tail-flick test and expressed as area under the time-response curve. The intensity and duration of the analgesic area under the time-response curve was significantly greater in SHR rats in comparison to WKY rats. For example, whereas the effect of a 10-mg/kg dose lasted for 4 hr in WKY rats; in SHR rats the effect lasted for 10 hr. Similarly, the intensity and duration of morphine-induced hyperthermic response was greater in SHR rats in comparison to WKY rats. In order to determine if the differences in responses to morphine in SHR and WKY were related to its kinetics, the pharmacokinetic parameters of morphine in serum were determined. The area under the serum morphine concentration-time curve, serum levels of morphine extrapolated to zero time, T1/2, apparent volume of distribution at steady state, terminal elimination rate constant and total body clearance values for morphine in SHR and WKY rats did not differ. Previous studies from this laboratory indicate that the density of mu opiate receptors labeled with [3H]D-Ala2.MePhe4,Gly-ol5-enkephalin and [3H]naltrexone and of kappa-opiate receptors labeled with [3H]ethylketocyclazocine in the brain of SHR rats is higher than in WKY rats.(ABSTRACT TRUNCATED AT 250 WORDS)

The effect of U-50,488H, a selective kappa opiate agonist, on tolerance-dependence and abstinence on the TRH receptors of the spinal cord and discrete regions of the brain of male Sprague-Dawley rats was determined. Rats were injected intraperitoneally twice daily with 25 mg/kg of U-50,488H for 4 days. Rats serving as controls were injected with the vehicle. On day 5, rats which were labeled as tolerant to U-50,488H were injected with U-50,488H (25 mg/kg) and sacrificed 1 hr later, whereas those labeled as abstinent were sacrificed without any injection. The above procedure has been previously shown to produce a high degree of tolerance to the analgesic and hypothermic effects of U-50,488H. The spinal cord and regions of the brain (hippocampus, cortex, midbrain, hypothalamus, corpus striatum, pons and medulla, and amygdala) were isolated for binding studies. The ligand [3H]MeTRH was used for TRH receptors. The binding constants, Bmax and Kd values, of [3H]MeTRH to bind to membranes prepared from various regions of the brain and spinal cord of rats tolerant-dependent on U-50,488H were unaffected. However, in rats abstinent to U-50,488H, the binding of [3H]MeTRH to membranes of the hypothalamus, and pons and medulla, was decreased. The decreased binding of [3H]MeTRH to hypothalamic membranes was due to changes in Bmax value, while in pons and medulla it was due to an increase in the Kd value.(ABSTRACT TRUNCATED AT 250 WORDS)

The decay characteristics of tolerance and physical dependence on morphine induced by a pellet implantation procedure were determined in male Sprague-Dawley rats. Rats were implanted subcutaneously with 6 morphine pellets during a 7-day period. The pellets were removed, and at various times thereafter tolerance to the analgesic and hyperthermic effects of morphine was measured by determining the response in rats implanted with morphine and placebo pellets. Similarly, the physical dependence was assessed by monitoring withdrawal signs following an injection of naloxone. A high degree of tolerance developed to the analgesic and hyperthermic effects of morphine. Similarly, a high degree of physical dependence also developed as evidenced by a high incidence of jumping response, teeth chattering and production of fecal boli induced by injections of naloxone. In addition, loss of body weight and body temperature also occurred. The analgesic and hyperthermic response to morphine recovered very gradually. There was no significant difference in the analgesic and hyperthermic responses to morphine on day 4 after the pellet removal in rats implanted with morphine and placebo pellets. The decay of tolerance was linear with time for the analgesic effect (r = 0.98) and for the hyperthermic effect (r = 0.93). The change in symptoms of physical dependence on morphine with time depended on the specific symptom monitored. The average number of jumps and teeth chattering decreased with time in a linear fashion with r values of 0.98 and 0.99, respectively. However, the number of fecal boli and wet dog shakes increased linearly with time (r = 0.97). The recovery of loss of body weight was also linear with time. Thus, it is clear that fecal boli and wet dog shakes, which increase in number as the dependence decays, are signs of a low degree of dependence. The results suggest that different central or peripheral mechanisms may be operating in different withdrawal symptoms. These studies may prove to be useful when studying the mechanisms involved in the induction and reversibility of tolerance and dependence processes, and in long-term effects of opiates at a time when the tolerance and physical dependence is no longer evident.

The effect of morphine tolerance-dependence and abstinence on the characteristics of delta-opiate receptors was determined in male Sprague-Dawley rats. Two ligands used for characterizing the receptors were [3H][D-Ser2,Thr6]leucine-enkephalin ([3H]DSTLE) and [3H][D-Pen2,D-Pen5]enkephalin ([3H]DPDPE). Rats were implanted s.c. under light ether anesthesia with six morphine pellets (each containing 75 mg of morphine free base). Rats which served as controls were implanted similarly with placebo pellets. Two sets of rats were used. In one group of rats, the pellets were left intact (tolerant-dependent) at the time of sacrificing and in the other the pellets had been removed 18 h earlier (abstinent). The spinal cord and brain regions (amygdala, hippocampus, hypothalamus, corpus striatum, mid-brain, pons and medulla and cortex) were dissected for binding studies. The binding of [3H]DSTLE to membranes of cerebral cortex of morphine-tolerant-dependent rats was decreased in comparison to control rats, and was due to a decrease in Bmax rather than Kd value. The binding of [3H]DSTLE to other brain regions or spinal cord of morphine-tolerant-dependent and abstinent rats did not differ from their respective controls. On the other hand, the binding of [3H]DPDPE was unaffected in any brain region or the spinal cord of morphine-tolerant-dependent and abstinent rats when compared to their controls. The decrease in binding of [3H]DSTLE to cortical membranes of morphine-tolerant-dependent rats amounted to 15%. Since DSTLE also binds to mu-opiate receptors, which have earlier been shown to be decreased in cortex of morphine-tolerant-dependent rats, and the binding of a more selective delta-opiate ligand [3H]DPDPE was unaffected, it is concluded that central delta-opiate receptors do not play a role in the development of morphine-induced tolerance-dependence or abstinence processes in the rat.

The analgesic response, colonic temperature changes and the pharmacokinetic parameters of i.v. administered morphine sulfate (2.5, 5.0 and 10.0 mg/kg) through indwelling cannulas in the jugular vein were determined in male Sprague-Dawley rats. The analgesic and hyperthermic responses were determined before and at 30-min intervals for a period of 360 min after the morphine injection and were expressed as area under the time-response curve. Blood samples were collected periodically for a period of 24 h. Serum was separated and was used for the estimation of morphine by the highly sensitive radioimmunoassay. The pharmacokinetic parameters, half life, the terminal elimination rate constant, the mean residence time, the apparent volume of distribution at steady state and the total clearance were determined by using noncompartmental analysis. The area under the serum concentration-time curve from time zero to infinity was related to the dose of morphine. The other parameters were the same for all the doses of morphine, except the serum levels of morphine extrapolated to zero time which increase gradually with the dose. Dose-dependent analgesic and hyperthermic responses were related to the area under the serum concentration-time curve. It is concluded that the dose-dependent pharmacological effects of morphine were related to the area under the serum concentration-time curve from time zero to infinity for serum levels of morphine but not to the other pharmacokinetic parameters.

The effect of morphine tolerance-dependence, protracted and naloxone-precipitated abstinence on the levels of beta-endorphin and methionine-enkephalin in discrete brain regions, spinal cord, pituitary gland and plasma was determined in the male Sprague-Dawley rats. Among the brain regions examined, the levels of beta-endorphin in descending order were: hypothalamus, amygdala, midbrain, hippocampus corpus striatum, pons and medulla and cortex. The levels of beta-endorphin in midbrain, hypothalamus, and pituitary of morphine tolerant-dependent rats were decreased significantly. During protracted withdrawal beta-endorphin levels were decreased in amygdala, spinal cord and pituitary. During naloxone-precipitated abstinence beta-endorphin levels were increased in corpus striatum, midbrain and cortex. In addition, in naloxone-precipitated abstinence beta-endorphin levels were decreased in pituitary gland and hippocampus but increased in plasma. The levels of methionine-enkephalin in brain regions in decreasing order were: corpus striatum, pons and medulla, amygdala, hypothalamus, midbrain, hippocampus and cortex. The levels of methionine-enkephalin in pons and medulla, amygdala, hippocampus and pituitary gland were decreased in morphine tolerant-dependent rats. During protracted abstinence from morphine, methionine-enkephalin levels in spinal cord, amygdala, pons and medulla, midbrain, cortex, corpus striatum and pituitary gland were decreased. The levels of methionine-enkephalin in hypothalamus and corpus striatum of naloxone-precipitated abstinent rats were increased but were decreased in amygdala and pituitary gland. These results suggest that during morphine tolerance-dependence and during protracted abstinence beta-endorphin and methionine-enkephalin levels in discrete brain regions and pituitary gland are decreased.(ABSTRACT TRUNCATED AT 250 WORDS)

The effects of chronic administration of U-50,488H, a K-opiate receptor agonist, on the binding of [3H]ethylketocyclazocine ([3H]EKC) to K-opiate receptors on the cerebral cortical and spinal cord membranes of spontaneously hypertensive (SHR), Wistar-Kyoto normotensive (WKY), and Sprague-Dawley (SD) rats were determined. Age-matched (10 weeks old) male rats of each strain were injected twice daily for 7 days with either U-50,488H (25 mg/kg, i.p.) or its vehicle. On day 8, the rats were killed. The cerebral cortex and the spinal cord were isolated for binding studies. The systolic blood pressure and heart rate of SD and WKY rats did not differ but the blood pressure of SHR rats were higher than that of SD and WKY rats. The receptor density (Bmax) and apparent dissociation constant (Kd) values of [3H]EKC binding to the spinal cord of WKY and SHR rats did not differ. However, the spinal cord of SD rats had higher Bmax and Kd values than WKY or SHR rats. The cortex of the SD rats had a lower Bmax value than the other two strains. Treatment with U-50,488H decreased the Bmax value of [3H]EKC in spinal cord of SD rats, increased the Kd value in SHR rats, and had no effect in WKY rats. Decreases in the Bmax value were produced in the cortex of all strains of rats, but a greater effect was observed in WKY and SHR rats than in SD rats.

A novel coating process, leading to formation of uniform, defect-free coating on solid dosage forms, is proposed. The coating process, termed "diffusion-controlled interfacial complexation," involves a chemical reaction between a reactant incorporated in the solid unit to be coated and a polymer solution, forming the coating medium. The reaction results in the formation of an insoluble reactant-polymer film around the solid. The rate of film/membrane formation is controlled by the rate of diffusion of reactant through the reactant-polymer film. In the model system, calcium acetate was selected as the reactant and algin as the polymer. The coating process was mathematically characterized in terms of rate of increase in film thickness, film weight, and depletion of reactant. Compressed tablets coated using the above process provided zero-order release in distilled water.

1. In order to determine whether the degree of tolerance to and physical dependence on morphine induced by pellet implantation procedure in the rat depends on the dose used and the kinetic parameters, the effect of implantation of different number of pellets on tolerance-dependence and elimination kinetics of morphine from serum was determined. 2. Male Sprague-Dawley rats were implanted subcutaneously with pellets. Each pellet contained 75 mg of morphine free base. Three schedules of implantation were used. They included 2 pellets during a 3-day period (2/3), 4 pellets during a 3-day period (4/3) and 6 pellets during a 7-day period (6/7). Placebo pellets which did not contain morphine were implanted in rats which served as controls. 3. The degree of tolerance to and physical dependence on morphine increased as the number of morphine pellets implanted increased. 4. In separate groups of rats implanted with pellets, elimination kinetics of morphine was studied using radioimmunoassay. The kinetic parameters were: area under serum morphine concentration time curve (AUC0----infinity), serum concentration of morphine extrapolated to time zero (Cmax), half-life (t1/2), elimination rate constant (k), mean residence time (MRT) and total body clearance (Clt). 5. The AUC0----infinity and Cmax increased in proportion to the number of pellets implanted. The t1/2, k, MRT and Clt values for 2/3 and 4/3 schedules did not differ, but for 6/7 schedule were significantly different from the other two schedules. The degree of tolerance to and physical dependence on morphine was directly related to the AUC0----infinity and Cmax. The longer t1/2 and MRT and lower Clt and k values in 6/7 schedule may reflect a saturation of glucuronic acid transferase, the main enzyme metabolizing morphine in the liver, and may account for the greater degree of tolerance and physical dependence.

The binding of 3H-naltrexone, an opiate receptor antagonist, to membranes of discrete brain regions and spinal cord of 10 week old spontaneously hypertensive (SHR) and normotensive Wistar-Kyoto (WKY) rats was determined. The brain regions examined were hypothalamus, amygdala, hippocampus, corpus striatum, pons and medulla, midbrain and cortex. 3H-Naltrexone bound to membranes of brain regions and spinal cord at a single high affinity site with an apparent dissociation constant value of 3 nM. The highest density of 3H-naltrexone binding sites were in hippocampus and lowest in the cerebral cortex. The receptor density (Bmax value) and apparent dissociation constant (Kd value) values of 3H-naltrexone to bind to opiate receptors on the membranes of amygdala, hippocampus, corpus striatum, pons and medulla, midbrain, cortex and spinal cord of WKY and SHR rats did not differ. The Bmax value of 3H-naltrexone binding to membranes of hypothalamus of SHR rats was 518% higher than WKY rats but the Kd values in the two strains did not differ. It is concluded that SHR rats have higher density of opiate receptors labeled with 3H-naltrexone in the hypothalamus only, in comparison with WKY rats, and that such a difference in the density of opiate receptors may be related to the elevated blood pressure in SHR rats.

The analgesic and thermic responses to morphine (5 and 10 mg/kg) injected intravenously to 8- and 24-week-old male Sprague-Dawley rats were determined. Greater analgesic and lower hyperthermic responses to morphine in 24-week-old rats in comparison to 8-week-old rats were observed. The pharmacokinetic parameters of morphine administered intravenously were also determined. Cmax for 5 and 10 mg/kg doses of morphine were smaller in 24-week-old rats in comparison to 8-week-old rats; however, AUC0----infinity was smaller only for 5 mg/kg dose. For 10 mg/kg dose, mean residence time (MRT) and the apparent steady state volume of distribution (Vss) for the older rats were higher than for the younger ones, but for 5 mg/kg dose the values did not differ. The enhanced responses to morphine in older age group of rats for 5 mg/kg dose cannot be explained solely on the basis of pharmacokinetics. However, for 10 mg/kg dose of morphine, the greater responses in 24-week-old rats could probably be related to increases in MRT and Vss. Factors other than serum kinetics, like kinetics of morphine in the brain as well as the brain opiate receptors, may also be involved in the differential effects of morphine in rats of different ages.

1. Chronic administration of opiates to rodents results in the development of tolerance to their pharmacological effects. Physical dependence also develops and is shown by the appearance of abstinence syndrome. 2. Opiates produce their effects by acting on three types of opiate receptors, namely mu, delta and kappa. The qualitative and quantitative aspects of the tolerance-dependence and abstinence symptoms observed after chronic administration of agonists acting at mu-, delta- and kappa-opiate receptors appear to differ. 3. Tolerance-dependence on mu-opiate agonists, such as morphine, is associated with down-regulation of mu-opiate receptors in spinal cord and specific areas of the brain but delta- and kappa-opiate receptors are unchanged. During abstinence from mu-opiate agonists, brain and spinal cord mu-, delta- and kappa-opiate receptors are unaffected. 4. Chronic administration of kappa-opiate agonists, such as U-50,488H, results in the development of tolerance to its pharmacological effects and a mild degree of physical dependence. Such changes are associated not only with alterations of delta and kappa opiate receptors in brain and spinal cord, but also primarily with a down-regulation of kappa-opiate receptors in spinal cord and specific brain regions. mu-Opiate receptors are unaffected. 5. Chronic administration of delta-opiate agonists results in down-regulation of brain delta-opiate receptors. 6. It is concluded that tolerance-dependence on mu-, delta- and kappa-opiate receptors is associated with down-regulation of their own type of receptors in the spinal and supraspinal areas. Abstinence, on the other hand, does not alter brain and spinal cord opiate receptors.(ABSTRACT TRUNCATED AT 250 WORDS)

1. The effect of intraperitoneal administration of Panax ginseng on the development of tolerance to the analgesic and hyperthermic actions of morphine was determined in male Sprague-Dawley rats. Rats were rendered tolerant to morphine to different degrees by the subcutaneous implantation of either four pellets of morphine over a 3-day period or six pellets over a 7-day period. Each pellet contained 75 mg of morphine free base. Rats serving as controls were implanted with placebo pellets. 2. Daily administration of ginseng extract (6.25-50.0 mg/kg) for 3 days inhibited the development of tolerance to the analgesic effect but not to the hyperthermic effect of morphine in the four pellet schedule. 3. In six pellet schedule, daily administration of ginseng extract (25 and 50 mg/kg) for 7 days also inhibited the development of tolerance to the analgesic effect of morphine, but the 100 mg/kg dose had no effect. On the other hand, in six pellet schedule, the administration of ginseng extract (50 and 100 mg/kg) once daily for 7 days inhibited the development of tolerance to the hyperthermic effect of morphine. 4. It is concluded that in appropriate doses, ginseng extract has inhibitory activity on the development of tolerance to the pharmacological actions of morphine.

The effect of delta- and kappa-opiate receptor agonists on the binding of 3H-(3-MeHis2) thyrotropin-releasing hormone (3H-MeTRH) to membranes of the spinal cord and amygdala of male Sprague-Dawley rats was determined in an effort to further understand interactions between opiates and TRH receptors. The agonists used were D-Ala2-MePhe4-Gly-ol5-enkephalin (DAMGO, mu-receptor), cyclic D-penicillamine2-D-penicillamine5-enkephalin (DPDPE, delta-receptor), cyclic D-penicillamine2-L-penicillamine5-enkephalin (DPLPE, delta-receptor), D-Ala2-D-Leu5-enkephalin (delta-receptor), U-50,488H and U-69,593 (kappa-receptor). 3H-MeTRH bound to amygdala and spinal cord membranes at a single site with Bmax values of 35.7 +/- 5.4 and 15.8 +/- 2.6 fmol/mg protein, and Kd values of 6.3 +/- 1.1 and 5.2 +/- 0.7 nmol/l, respectively. The competition experiments were carried out at a concentration of 2 nmol/l 3H-MeTRH. The concentration of opiate ranged from 10(-9) to 10(-4) mol/l. DAMGO, DPDPE and DPLPE had no effect on the binding of 3H-MeTRH to amygdala or spinal cord membranes. The two highly selective kappa-agonists differed in their interaction with TRH receptors. Whereas U-69,593 did not modify the binding of 3H-MeTRH, U-50,488H significantly inhibited the binding of 3H-MeTRH to both spinal cord and amygdala membranes. U-50,488H has been found to be 10 times more potent than U-69,593 at the central kappa-opiate receptors and may explain their differential action at the TRH receptors. It is concluded that mu- and delta-opiate agonists do not interact with brain and spinal cord TRH receptors.(ABSTRACT TRUNCATED AT 250 WORDS)

The effects of chronic administration of U-50,488H, a highly selective kappa-opiate receptor agonist, and its subsequent withdrawal were determined. The binding characteristics of [3H]SCH 23390 to dopamine D1 receptors were investigated on membranes of rat spinal cord and in discrete brain regions. Male Sprague-Dawley rats received U-50,488H (25 mg/kg) intraperitoneally twice a day for 4 days. Animals serving as controls received only vehicle. Two different treatment schedules were used. In one, rats were injected with U-40,588H on day 5 and were sacrificed 1 h later (tolerant-dependent rats). In another, the rats were sacrificed 16 h after the last injection of U-50,488H (abstinent rats). Chronic administration of U-50,488H resulted in the development of tolerance to its analgesic effect, as demonstrated by the decreased response to a challenge dose of U-50,488H compared to vehicle-injected rats. The binding characteristics (Bmax and Kd values) of [3H]SCH 23390 were unaffected in spinal cord and other regions of brain of tolerant-dependent rats. In U-50,488H-abstinent rats, Bmax values of [3H]SCH 23390 were increased in hypothalamic and striatal membranes, but Kd values were unaffected. These results suggest that, in U-50,488H-abstinent rats, dopamine D1 receptors are up-regulated in hypothalamus and striatum.

It is concluded that activation of kappa-opiate receptors does not modify the development of tolerance to morphine in the rat.

The effect of chronic administration of morphine and its withdrawal on the characteristics of mu-opiate receptors was determined in male Sprague-Dawley rats. The ligand used for characterizing the receptors was [3H][D-Ala2,MePhe4,Gly5-ol]enkephalin ([3H]DAMGO). Rats were implanted s.c. under light ether anesthesia with six morphine pellets (each containing 75 mg of morphine free base). Rats serving as controls were implanted similarly with placebo pellets. Two sets of animals were used. In one group of rats, the pellets were left intact (tolerant-dependent) at the time of killing and in the other the pellets had been removed 18 h earlier (abstinent). The spinal cord and brain regions (amygdala, hippocampus, hypothalamus, corpus striatum, midbrain, pons and medulla, and cortex) were dissected. In morphine-abstinent rats, the binding of ligands of mu-opiate receptors to membranes of spinal cord and brain regions did not change. In non-abstinent morphine-tolerant-dependent rats, the binding of [3H]DAMGO to membranes of spinal cord, pons and medulla, and cerebral cortex was found to be decreased. These changes were due to decreases in the Bmax values rather than Kd values for the binding of [3H]DAMGO. The results clearly indicate that morphine-induced tolerance-dependence in the rat is associated with changes in the selected brain regions and spinal cord with mu-opiate receptors being down-regulated in spinal cord, pons and medulla, and cerebral cortex. It is concluded that tolerance to morphine in the rats may be due to down-regulation of central mu-opiate receptors. However, mu-opiate receptors are unaffected in morphine abstinence.

The effects of morphine tolerance-dependence and abstinence on 5-HT1A receptors in brain regions and spinal cord of the rat were determined. Tolerance to and physical dependence on morphine was induced in male Sprague-Dawley rats by implanting six morphine pellets (each containing 75 mg of morphine free base) during a seven day period. Two groups of rats were used for binding studies. In one group the pellets were left intact (tolerant-dependent) and in the other they were removed (abstinent). Rats were killed, and spinal cords and brains were excised. Brain was dissected into seven regions (amygdala, hippocampus, hypothalamus, striatum, midbrain, pons + medulla and cortex). 5-HT1A receptors were characterized by using [3H]8-hydroxy-di-n-propylaminotetralin [( 3H]DPAT) as the ligand and unlabelled 5-HT to determine the non-specific binding. In morphine and placebo tolerant-dependent rats the binding of [3H]DPAT to 5-HT1A receptors in brain regions and spinal cord did not differ. The Bmax value of [3H]DPAT in the hypothalamus of morphine abstinent rats was decreased by 61.9%. No change in Bmax value was observed in other brain regions and spinal cord. The Kd values were unaffected. Subcutaneous administration of DPAT produced hypothermia in rats from which pellets had been removed. The intensity of DPAT-induced hypothermic response was greater in morphine abstinent rats as compared to placebo abstinent rats. Since DPAT is believed to have a major action on the presynaptic 5-HT neurons, it is concluded that in morphine abstinent rats 5-HT1A receptors are down-regulated in hypothalamus, but in morphine tolerant-dependent rats they are unaffected.

The effect of chronic administration of morphine and its withdrawal on the binding of 3H-[3-MeHis2]thyrotropin releasing hormone (3H-MeTRH) to membranes of the spinal cord of the rat was determined. Male Sprague-Dawley rats were implanted with either 6 placebo or 6 morphine pellets (each containing 75-mg morphine base) during a 7-day period. Two sets of animals were used. In one, the pellets were left intact at the time of sacrificing (tolerant-dependent) and in the other, the pellets were removed 16 hours prior to sacrificing (abstinent rats). In placebo-pellet-implanted rats, 3H-MeTRH bound to the spinal cord membranes at a single high affinity binding site with a Bmax of 21.3 +/- 1.6 fmol/mg protein, and an apparent dissociation constant Kd of 4.7 +/- 0.8 nM. In morphine tolerant-dependent or abstinent rats, the binding constants of 3H-MeTRH to spinal cord membranes were unaffected. Previous studies from this laboratory indicate that TRH can inhibit morphine tolerance-dependence and abstinence processes without modifying brain TRH receptors. Together with the present results, it appears that the inhibitory effect of TRH on morphine tolerance-dependence and abstinence is probably not mediated via central TRH receptors but may be due to its interaction with other neurotransmitter systems.

The effect of thyrotropin releasing hormone (TRH) on the development of tolerance to the analgesic and hyperthermic actions of morphine was determined in male Sprague-Dawley rats. The tolerance to morphine was induced by subcutaneous implantation of 6 morphine pellets, each containing 75 mg of morphine free base. Rats serving as controls were implanted with 6 placebo pellets. Subcutaneous injections of TRH (1, 3 or 10 mg/kg) twice a day for 7 days inhibited the development of tolerance to the analgesic, but not to the hyperthermic action of morphine. Doses lower than 1 mg/kg or greater than 10 mg/kg administered twice a day for 7 days did not modify tolerance development. The inability of higher doses of TRH to inhibit tolerance to morphine may be related to rapid down-regulation of TRH receptors. The results suggest that in appropriate doses, TRH can selectively inhibit the development of tolerance to the analgesic action of morphine.

The effect of morphine tolerance-dependence and abstinence on dopamine D1 receptors in brain regions and spinal cord was determined in rats. Male Sprague-Dawley rats were implanted s.c. under light ether anesthesia with 6 morphine pellets, each containing 75 mg of morphine free base. Rats serving as controls were implanted with placebo pellets. This procedure resulted in the development of tolerance to morphine as evidenced by decreased analgesic response to a challenge dose of morphine. Similarly, the development of physical dependence was evidenced by decreased body weight and colonic temperature after morphine pellet removal (withdrawal). Two sets of animals were used for receptor binding studies. In one, the pellets were left intact and in the other, the pellets were removed. Eighteen hours after pellet removal, the rats were sacrificed. [3H]SCH 23390 [( R-(+)-8-chloro-2,3,4,5-tetrahydro-3-methyl-5-phenyl-1-3-benzapine- 7-ol]) bound to brain region and spinal cord membranes of placebo pellet-implanted rats at a single high affinity site. In rats treated chronically with morphine and then withdrawn, the binding of [3H]SCH 23390 to membranes of spinal cord, hypothalamus and striatum was increased but the binding to amygdalar membranes was decreased in comparison with placebo-treated rats. The changes in binding were due to the changes in Bmax values; the Kd values were unaffected. The behavioral responses to a selective dopamine D1 receptor agonist, SKF 38393 [( 1-phenyl-2,3,4,5-tetrahydro (1H)-3-benzapine-7,8-diol hydrochloride]), were also enhanced in morphine-withdrawn rats when compared to placebo controls.(ABSTRACT TRUNCATED AT 250 WORDS)

The studies have clearly demonstrated that binding sites for opioid peptides, beta-endorphin, and methionine-enkephalin exist on T lymphocytes. beta-Endorphin appears to be immunodepressant, whereas methionine-enkephalin is immunostimulant. Both in vitro and in vivo studies have shown that methionine-enkephalin can influence some immune functions. Since in vitro modification of immune function requires very low concentrations, it is reasonable to believe that methionine-enkephalin plays a physiological role in the immune system. Although not well established, methionine-enkephalin appears to activate T lymphocytes via opioid receptors and triggers a series of intracellular signals leading to the activation of receptors for interleukin-2 (IL-2), OKT10, and active sheep T red blood cell receptors. Methionine-enkephalin enhances the activity of NK cells and induces the production of IL-2, which in turn may recruit and activate other T-cell subsets like CD3 and CD4. Methionine-enkephalin also enhances mitogen-induced proliferation of lymphocytes. Since preliminary studies with methionine-enkephalin in ARC patients have provided beneficial effects by the improvements in their symptoms, it will be worthwhile to extend these observations to a larger number of patients with ARC and AIDS. Finally, it appears that some endogenous opioid peptides and their analogs, in addition to methionine-enkephalin, may provide therapeutic benefits not only in ARC and AIDS but also in other immunodeficient states.

The binding of [3H] DAMGO, a highly selective ligand for mu-opiate receptors, to membranes of discrete brain regions and spinal cord of 10 week old spontaneously hypertensive (SHR) and normotensive Wistar-Kyoto (WKY) rats was determined. On the other hand, the Bmax value of [3H] DAMGO in membranes of amygdala of SHR rats was lower than that of WKY rats but the Kd values in the two strains were similar. It is concluded that SHR rats have higher density of mu-opiate receptors in hypothalamus and midbrain but lower density in amygdala in comparison with WKY rats, and that such differences in the distribution of mu-opiate receptors may be related to the elevated blood pressure in SHR rats.

The effect of melanotropin release inhibiting factor (Pro-Leu-Gly-NH2, MIF) was determined on changes induced by two neuroleptics, haloperidol and centbutindole, in cerebral cortical 5-hydroxytryptamine (5-HT) receptors. Male Sprague-Dawley rats were injected daily i.p. with vehicle, haloperidol (1.0 mg/kg) or centbutindole (0.5 mg/kg), respectively, for 21 days. On day 22, these 3 groups were further divided into 2 subgroups and injected with either vehicle or MIF (2.0 mg/kg, i.p.) daily for 3 days. 3H-5-HT was used to study 5-HT1 receptors, and 3H-spiroperidol to label 5-HT2 receptors in the cerebral cortex. Chronic administration of haloperidol significantly increased (39.7%) the maximal binding capacity (Bmax) of 3H-5-HT binding to 5-HT1 receptors. Dissociation constant (Kd) values did not change. Centbutindole had no effect on 5-HT1 receptors. MIF had no effect on 5-HT1 receptors, nor did it alter haloperidol-induced increases in the Bmax of 3H-5-HT binding to 5-HT1 receptors. Chronic administration of centbutindole significantly increased (61.1%) the Bmax of 3H-spiroperidol binding to 5-HT2 receptors. No change occurred in the Kd values. Chronic treatment with haloperidol had no effect on 5-HT2 receptor characteristics. MIF had no effect on 5-HT2 receptors or on the increase in 5-HT2 receptor density induced by centbutindole. The behavioral syndrome induced by 5-hydroxytryptophan (5-HTP) (50, 100 and 200 mg/kg, i.p.) was also measured in rats treated chronically with haloperidol or centbutindole. Haloperidol had no effect on the 5-HTP syndrome, whereas centbutindole stimulated by 24-45% the intensity of the syndrome.(ABSTRACT TRUNCATED AT 250 WORDS)

1. The effects of intraperitoneal administration of a standard extract of Panax ginseng alone and in combination with morphine were determined in male Sprague-Dawley rats. 2. Ginseng extract at 200 mg/kg produced analgesia and hypothermia. These effects of ginseng were not reversed by naltrexone. 3. A dose of morphine (8 mg/kg) produced analgesia and hyperthermia. The analgesic response to morphine was antagonized by 25 and 50 mg/kg doses of ginseng but not by 12.5, 100 and 200 mg/kg doses. 4. Morphine-induced hyperthermia was antagonized by 12.5-200 mg/kg doses of ginseng. 5. Administration of morphine (50 mg/kg) produced cataleptic effect which was antagonized by 25 mg/kg of ginseng. 6. The results suggest that ginseng extract at high doses produces analgesia and hypothermia in the rat by a non-opiate mechanism, and antagonizes the acute pharmacological effects of morphine.

Studies in these laboratories have shown that morphine and thyrotropin releasing hormone (TRH) inhibit gastrointestinal transit in the mouse. Administration of morphine sulfate (5 mg/kg, s.c.) or TRH (10 microgram i.c.v.) to mice inhibited gastrointestinal transit as measured by the charcoal meal test. In order to determine whether the effects of TRH and morphine were mediated via stereospecific opiate receptors, the effects of two stereoisomers of an antagonist, (-) alpha -5,9-diethyl-2'-hydroxy-2-(3-furylmethyl)6,7-benzomorphan (MR2266), the active isomer and (+) alpha-5,9-diethyl-2'-hydroxy-2-(3-furylmethyl)6,7-benzomorphan (MR 2267), the inactive isomer, on morphine and TRH induced changes in gastrointestinal transit were determined. Morphine and THR induced inhibition of gastrointestinal transit was antagonized by MR 2266 (1 and 3 mg/kg, s.c.) but was unaffected by MR 2267. These studies provide evidence for the involvement of opiate receptors in the actions of morphine and TRH on gastrointestinal transit, and further suggest that the receptors are stereospecific in nature.

The influence of thyrotropin releasing hormone (TRH) and its postulated metabolite histidyl-proline diketopiperazine (HPD) on sedation and analgesia produced by delta 9-tetrahydrocannabinol (THC) was investigated in mice. Intracerebral administration of HPD (10 microgram) by itself decreased motor activity, whereas, TRH (10 microgram) was without any effect. THC (10 and 20 mg/kg) given i.p. decreased the motor activity and this effect was antagonised by prior treatment with TRH or HPD. Administration of THC produced analgesia as evidenced by delay in the tail-flick reaction time to a thermal stimulus. Pretreatment with TRH or HPD also antagonized the THC-induced analgesia. THC-induced analgesia was not modified by s.c. pretreatment with naloxone, an opiate antagonist. TRH or HPD pretreatment had no effect on the distribution of total radioactivity representing THC and its metabolites in brain and plasma. These studies indicate that TRH and HPD can antagonize THC-induced sedation and analgesia and that HPD may be an active metabolite of TRH. These effects were observed without altering the distribution of the drug and metabolite in brain and plasma, and therefore their effect may involve their direct actions on the central nervous system. Finally THC induced analgesia does not seem to involve opiate receptors and their involvement in the mechanism by which TRH or HPD antagonize THC-induced analgesia is unlikely.

The effect of U-50,488H, a selective kappa opioid receptor agonist on the intake of food in food-deprived and non-deprived spontaneously hypertensive and normotensive Wistar-Kyoto rats was determined. In food deprived Wistar-Kyoto rats, intraperitoneal administration of U-50,488H, produced a bell-shaped curve on the intake of food, consistent with earlier reports in the literature. Thus, at a dose of 0.1 mg/kg there was a significant increase in the intake of food and at 10 mg/kg it caused a decrease in the intake of food, at 2, 4 and 6 hr after the treatment with drug. The amount of food consumed at each dose and the time interval was greater in hypertensive rats than in Wistar-Kyoto rats. Similar effects were produced when the animals were not deprived of food. The basal intake of food in the two strains of rats did not differ. The results indicate a greater sensitivity of spontaneously hypertensive rats to kappa opioid receptor agonists and further the previous observations that compared to Wistar-Kyoto rats, the hypertensive rats have a greater density of central kappa opioid receptors.
