The aim of this study was to compare bibliometric indicators of scientific productivity in physical activity (PA) in children and adolescents with Autism spectrum disorder (ASD) and Down syndrome (DS) in the PubMed database. A bibliometric study was conducted for the last 5 years (2017 to 2021). The data collected for each article were: year of publication, language of publication, country, journal name, and type of paper. The results showed that there was higher scientific productivity in the population with DS (20 studies) relative to their counterparts with ASD (31 studies). The language of publication in both cases was English. There were 10 countries that published on PA in ASD and 14 countries that published on DS. Overall, the greatest interest in publishing on PA in children and adolescents with ASD was in North America (6 studies), followed by Asia (5 studies) and Europe (4 studies). In the DS population it was in Europe (13 studies), North America (9 studies) and South America (4 studies). Nineteen journals were identified that published in the ASD population and 29 journals in DS. Six experimental studies were identified in ASD and 7 in DS. There was a higher scientific productivity with original studies. There was a positive trend of increasing scientific productivity over the years in both populations. We suggest the need to promote research on PA in both populations, regardless of the type of study, as it is an indicator of overall health status.

Summary: Background: Surgical care holds significant importance in healthcare, especially in low and middle-income countries, as at least 50% of the 4.2 million deaths within the initial 30 days following surgery take place in these countries. The Lancet Commission on Global Surgery proposed six indicators to enhance surgical care. In Colombia, studies have been made using secondary data. However, strategies to reduce perioperative mortality have not been implemented. This study aims to describe the fourth indicator, perioperative mortality rate (POMR), with primary data in Colombia. Methods: A multicentre prospective cohort study was conducted across 54 centres (hospitals) in Colombia. Each centre selected a 7-day recruitment period between 05/2022 and 01/2023. Inclusion criteria involved patients over 18 years of age undergoing surgical procedures in operating rooms. Data quality was ensured through a verification guideline and statistical analysis using mixed-effects multilevel modelling with a case mix analysis of mortality by procedure-related, patient-related, and hospital-related conditions. Findings: 3807 patients were included with a median age of 48 (IQR 32–64), 80.3% were classified as ASA I or II, and 27% of the procedures had a low-surgical complexity. Leading procedures were Orthopedics (19.2%) and Gynaecology/Obstetrics (17.7%). According to the Clavien–Dindo scale, postoperative complications were distributed in major complications (11.7%, 10.68–12.76) and any complication (31.6%, 30.09–33.07). POMR stood at 1.9% (1.48–2.37), with elective and emergency surgery mortalities at 0.7% (0.40–1.23) and 3% (2.3–3.89) respectively. Interpretation: The POMR was higher than the ratio reported in previous national studies, even when patients had a low–risk profile and low-complexity procedures. The present research represents significant public health progress with valuable insights for national decision-makers to improve the quality of surgical care. BACKGROUND AND OBJECTIVES: Ectopic intracerebral calcifications (EICs) in the basal ganglia, thalamus, cerebellum, or white matter are seen in a variety of disease states or may be found incidentally on brain imaging. The clinical significance and proportion of cases attributable to an underlying genetic cause is unknown. METHODS: This retrospective cohort study details the clinical, imaging, and genomic findings of 44 patients with EICs who had no established diagnosis despite extensive medical workup. RESULTS: In total, 15 of 44 patients received a diagnosis through genomic testing explaining their calcifications, and 2 more received a diagnosis that has not been previously associated with EICs. Six of the 15 were found to have one of the 4 genes (PDGFB, PDGFRB, SLC20A2, and XPR1) conventionally associated with the phenotypic term idiopathic basal ganglia calcifications. DISCUSSION: These findings support the use of genomic testing for symptomatic patients with EICs.

A major mechanism conferring resistance to mTOR inhibitors is activation of a salvage pathway stimulating internal ribosome entry site (IRES)-mediated mRNA translation, driving the synthesis of proteins promoting resistance of glioblastoma (GBM). Previously, we found this pathway is stimulated by the requisite IRES-trans-acting factor (ITAF) hnRNP A1, which itself is subject to phosphorylation and methylation events regulating cyclin D1 and c-myc IRES activity. Here we describe the requirement for m6A-modification of IRES RNAs for efficient translation and resistance to mTOR inhibition. DRACH-motifs within these IRES RNAs upon m6A modification resulted in enhanced IRES activity via increased hnRNP A1-binding following mTOR inhibitor exposure. Inhibitor exposure stimulated the expression of m6A-methylosome components resulting in increased activity in GBM. Silencing of METTL3-14 complexes reduced IRES activity upon inhibitor exposure and sensitized resistant GBM lines. YTHDF3 associates with m6A-modified cyclin D1 or c-myc IRESs, regulating IRES activity, and mTOR inhibitor sensitivity in vitro and in xenograft experiments. YTHDF3 interacted directly with hnRNP A1 and together stimulated hnRNP A1-dependent nucleic acid strand annealing activity. These data demonstrate that m6A-methylation of IRES RNAs regulate GBM responses to this class of inhibitors.

IntroductionHGF/c-MET signaling is a significant driver of glioblastoma (GBM) growth and disease progression. Unfortunately, c-MET targeted therapies have been found to be largely ineffective suggesting additional redundant mechanisms of c-MET activation.MethodsUtilizing RNA-sequencing (RNA-seq) and ribosome profiling analyses of circular RNAs, circ-HGF (hsa_circ_0080914) was identified as markedly upregulated in primary GBM and found to potentially encode an HGF protein variant (C-HGF) 119 amino acids in length. This candidate HGF variant was characterized and evaluated for its ability to mediate c-MET activation and regulate PDX GBM cell growth, motility and invasive potential in vitro and tumor burden in intracranial xenografts in mice.ResultsAn internal ribosome entry site (IRES) was identified within the circ-HGF RNA which mediated translation of the cross-junctional ORF encoding C-HGF and was observed to be highly expressed in GBM relative to normal brain tissue. C-HGF was also found to be secreted from GBM cells and concentrated cell culture supernatants or recombinant C-HGF activated known signaling cascades downstream of c-MET. C-HGF was shown to interact directly with the c-MET receptor resulting in its autophosphorylation and activation in PDX GBM lines. Knockdown of C-HGF resulted in suppression of c-MET signaling and marked inhibition of cell growth, motility and invasiveness, whereas overexpression of C-HGF displayed the opposite effects. Additionally, modulation of C-HGF expression regulated tumor growth in intracranial xenografted PDX GBM models.ConclusionsThese results reveal an alternative mechanism of c-MET activation via a circular RNA encoded HGF protein variant which is relevant in GBM biology. Targeting C-HGF may offer a promising approach for GBM clinical management.

Abstract Climate change brings a range of challenges and opportunities to shrimp fisheries globally. The case of the Colombian Pacific Ocean (CPO) is notable due the crucial role of shrimps in the economy, supporting livelihoods for numerous families. However, the potential impacts of climate change on the distribution of shrimps loom large, making it urgent to scrutinize the prospective alterations that might unfurl across the CPO. Employing the Species Distribution Modeling approach under Global Circulation Model scenarios, we predicted the current and future potential distributions of five commercially important shrimps (Litopenaeus occidentalis, Xiphopenaeus riveti, Solenocera agassizii, Penaeus brevirostris, and Penaeus californiensis) based on an annual cycle, and considering the decades 2030 and 2050 under the Shared Socioeconomic Pathways SSP 2.6, SSP 4.5, SSP 7.0, and SSP 8.5. The Bathymetric Projection Method was utilized to obtain spatiotemporal ocean bottom predictors, giving the models more realism for reliable habitat predictions. Six spatiotemporal attributes were computed to gauge the changes in these distributions: area, depth range, spatial aggregation, percentage suitability change, gain or loss of areas, and seasonality. L. occidentalis and X. riveti exhibited favorable shifts during the initial semester for both decades and all scenarios, but unfavorable changes during the latter half of the year, primarily influenced by projected modifications in bottom salinity and bottom temperature. Conversely, for S. agassizii, P. brevirostris, and P. californiensis, predominantly negative changes surfaced across all months, decades, and scenarios, primarily driven by precipitation. These changes pose both threats and opportunities to shrimp fisheries in the CPO. However, their effects are not uniform across space and time. Instead, they form a mosaic of complex interactions that merit careful consideration when seeking practical solutions. These findings hold potential utility for informed decision-making, climate change mitigation, and adaptive strategies within the context of shrimp fisheries management in the CPO.

Abstract Background COVID-19 vaccines effectively prevent infection and hospitalization. However, few population-based studies have compared the clinical characteristics and outcomes of patients hospitalized for COVID-19 using advanced statistical methods. Our objective is to address this evidence gap by comparing vaccinated and unvaccinated patients hospitalized for COVID-19. Methods This retrospective cohort included adult COVID-19 patients admitted from March 2021 to August 2022 from 27 hospitals. Clinical characteristics, vaccination status, and outcomes were extracted from medical records. Vaccinated and unvaccinated patients were compared using propensity score analyses, calculated based on variables associated with vaccination status and/or outcomes, including waves. The vaccination effect was also assessed by covariate adjustment and feature importance by permutation. Results From the 3,188 patients, 1,963 (61.6%) were unvaccinated and 1,225 (38.4%) were fully vaccinated. Among these, 558 vaccinated individuals were matched with 558 unvaccinated ones. Vaccinated patients had lower rates of mortality (19.4% vs. 33.3%), invasive mechanical ventilation (IMV-18.3% vs. 34.6%), noninvasive mechanical ventilation (NIMV-10.6% vs. 22.0%), intensive care unit admission (ICU-32.0% vs. 44.1%) vasoactive drug use (21.1% vs. 32.6%), dialysis (8.2% vs. 14.7%) hospital length of stay (7.0 vs. 9.0 days), and thromboembolic events (3.9% vs.7.7%), p

Abstract West Nile virus (WNV) and Usutu virus (USUV) are closely related flaviviruses with differing capacities to cause neurological disease in humans. WNV is thought to use a transneural route of neuroinvasion along motor neurons and causes severe motor deficits. The potential for use of transneural routes of neuroinvasion by USUV has not been investigated experimentally, and evidence from the few clinical case reports of USUV-associated neuroinvasive disease is lacking. We hypothesised that, compared with WNV, USUV is less able to infect motor neurons, and therefore determined the susceptibility of human induced pluripotent stem cell (iPSC)-derived spinal cord motor neurons to infection. Both viruses could grow to high titres in iPSC-derived neural cultures. However, USUV could not productively infect motor neurons due to restriction by the antiviral response, which was not induced upon WNV infection. Inhibition of the antiviral response allowed for widespread infection and transportation of USUV along motor neurons within a compartmented culture system. These results show a stark difference in the ability of these two viruses to evade initiation of intrinsic antiviral immunity. Our data suggests that USUV cannot infect motor neurons in healthy individuals but in case of immunodeficiency may pose a risk for motor-related neurological disease and transneural invasion.

In this study, an Artificial Neural Network-Genetic Algorithm (ANN-GA) approach was successfully applied to optimise the physicochemical factors influencing the synthesis of unsaturated fatty acids (UFAs) in the microalgae P. kessleri UCM 001. The optimized model recommended specific cultivation conditions, including glucose at 29 g/L, NaNO3 at 2.4 g/L, K2HPO4 at 0.4 g/L, red LED light, an intensity of 1000 lx, and an 8:16-h light-dark cycle. Through ANN-GA optimisation, a remarkable 66.79% increase in UFAs production in P. kessleri UCM 001 was achieved, compared to previous studies. This underscores the potential of this technology for enhancing valuable lipid production. Sequential variations in the application of physicochemical factors during microalgae culture under mixotrophic conditions, as optimized by ANN-GA, induced alterations in UFAs production and composition in P. kessleri UCM 001. This suggests the feasibility of tailoring the lipid profile of microalgae to obtain specific lipids for diverse industrial applications. The microalgae were isolated from a high-mountain lake in Colombia, highlighting their adaptation to extreme conditions. This underscores their potential for sustainable lipid and biomaterial production. This study demonstrates the effectiveness of using ANN-GA technology to optimise UFAs production in microalgae, offering a promising avenue for obtaining valuable lipids. The microalgae's unique origin in a high-mountain environment in Colombia emphasises the importance of exploring and harnessing microbial resources in distinctive geographical regions for biotechnological applications.

Nosocomial infections are a significant cause of morbidity, mortality, and increased treatment costs in hospitals. This study aimed to analyze the factors determining the implementation of biosafety measures by the nursing staff of four hospitals in Piura via a structural equation modeling (SEM) approach. A total of 215 nurses from various hospitals in the region participated by completing an online survey. The results demonstrated that extrinsic factors (FEX) positively influence the implementation of biosafety measures (BIOM) (β = 0.319 ***), as do intrinsic factors (FINT) (β = 0.520 **). Furthermore, intrinsic factors mediate the relationship between extrinsic factors and the implementation of biosafety measures (β = 0.443 ***). In conclusion, this study provides a deeper understanding of biosafety dynamics in healthcare settings and lays the groundwork for the development of customized interventions and ongoing training programs that ensure the optimal implementation of biosafety measures in hospitals.

Audiovisual materials have been widely investigated in mathematics education for their capacity to improve learning outcomes and mitigate student disengagement. Therefore, this study aims to evaluate the acceptance of audiovisual materials as instructional aids among eighth-grade students. Adopting a mixed-method approach, the research first involved semi-structured interviews with mathematics teachers to identify key teaching and learning challenges. Based on the insights gained, educational videos were developed, adhering to motivational and didactic criteria. In the final phase, 78 students from a public high school in Ibarra, Ecuador, participated in a Likert-scale survey, guided by the Technology Acceptance Model (TAM), to assess their reception of the videos. The findings revealed that teachers highlighted fear of mathematics, insufficient prior knowledge, and the abstract nature of the content as major obstacles to be addressed. Most students responded positively, considering the videos effective for enhancing their understanding of mathematical concepts and boosting their motivation to learn. This study underscores the potential of audiovisual materials as effective tools to address educational challenges and foster student engagement in mathematics.

Abstract Background Dysmetria, the inability to accurately estimate distance in motor tasks, is a characteristic clinical feature of cerebellar injury. Even though subjective dysmetria can be quickly detected during the neurological examination with the finger-to-nose test, objective quantification of reaching accuracy for clinical assessment is still lacking. Emerging VR technology allows for the delivery of rich multisensory environmental stimuli with a high degree of control. Furthermore, recent improvements in the hand-tracking feature offer an opportunity to closely examine the speed, accuracy, and consistency of fine hand movements and proprioceptive function. This study aims to investigate the application of virtual reality (VR) with hand tracking in the rapid quantification of reaching accuracy at the bedside for patients with cerebellar stroke (CS). Methods and results Thirty individuals (10 CS patients and 20 age-matched neurologically healthy controls) performed a simple task that allowed us to measure reaching accuracy using a VR headset (Oculus Quest 2). During this task, the participant was asked to reach for a target placed along a horizontal sixty-degree arc. Once the fingertip passed through the arc, the target immediately extinguished. 50% of the trials displayed a visible, real-time rendering of the hand as the participant reached for the target (visible hand condition), while the remaining 50% only showed the target being extinguished (invisible hand condition). The invisible hand condition isolates proprioception-guided movements by removing the visibility of the participant’s hand. Reaching error was calculated as the difference in degrees between the location of the target, and where the fingertip contacted the arc. Both CS patients and age-matched controls displayed higher average reaching error and took longer to perform a reaching motion in the invisible hand condition than in the visible hand condition. Reaching error was higher in CS than in controls in the invisible hand condition but not in the visible hand condition. Average time taken to perform each trial was higher in CS than in controls in the invisible hand conditions but not in the visible hand condition. Conclusions Reaching accuracy assessed by VR offers a non-invasive and rapid approach to quantifying fine motor functions in clinical settings. Furthermore, this technology enhances our understanding of proprioceptive function in patients with visuomotor disabilities by allowing the isolation of proprioception from vision. Future studies with larger cohorts and longitudinal designs will examine the quantitative changes in reaching accuracy after stroke and explore the long-term benefits of VR in functional recovery.

Abstract Mesoscale eddies influence the distribution of diazotrophic (nitrogen-fixing) cyanobacteria, impacting marine productivity and carbon export. Non-cyanobacterial diazotrophs (NCDs) are emerging as potential contributors to marine nitrogen fixation, relying on organic matter particles for resources, impacting nitrogen and carbon cycling. However, their diversity and biogeochemical importance remain poorly understood. In the subtropical North Atlantic along a single transect, this study explored the horizontal and vertical spatial variability of NCDs associated with suspended, slow-sinking, and fast-sinking particles collected with a marine snow catcher. The investigation combined amplicon sequencing with hydrographic and biogeochemical data. Cyanobacterial diazotrophs and NCDs were equally abundant, and their diversity was explained by the structure of the eddy. The unicellular symbiotic cyanobacterium UCYN-A was widespread across the eddy, whereas Trichodesmium and Crocosphaera accumulated at outer fronts. The diversity of particle-associated NCDs varied more horizontally than vertically. NCDs constituted most reads in the fast-sinking fractions, mainly comprising Alphaproteobacteria, whose abundance significantly differed from the suspended and slow-sinking fractions. Horizontally, Gammaproteobacteria and Betaproteobacteria exhibited inverse distributions, influenced by physicochemical characteristics of water intrusions at the eddy periphery. Niche differentiations across the anticyclonic eddy underscored NCD-particle associations and mesoscale dynamics, deepening our understanding of their ecological role and impact on ocean biogeochemistry.

This empirical study examines the motivational determinants driving the consumption of traditional local food and beverages in Latacunga in Central Ecuador (South America). This geographical area embraces a rich cultural and historical heritage, making it a year-round destination for tourists looking to experience its unique local cuisine and festivals. An online survey was conducted between September 2021 and February 2022, during which 445 valid questionnaires were collected; these were quantitatively analysed using binary logistic regression analysis. Model-based findings revealed that age, taste, health and safety concerns, cultural experience, and expenditure were the most significant factors influencing the consumption of traditional local food and beverages. This information will help understand tourists’ behaviour concerning gastronomic tourism, and help develop optimal positioning, branding, and marketing strategies for sustainable development at top-ranking destinations, particularly cities in Central Ecuador and other international cities featuring similar tourism characteristics.

Abstract The biomedical research community addresses reproducibility challenges in animal studies through standardized nomenclature, improved experimental design, transparent reporting, data sharing, and centralized repositories. The ARRIVE guidelines outline documentation standards for laboratory animals in experiments, but genetic information is often incomplete. To remedy this, we propose the Laboratory Animal Genetic Reporting (LAG-R) framework. LAG-R aims to document animals’ genetic makeup in scientific publications, providing essential details for replication and appropriate model use. While verifying complete genetic compositions may be impractical, better reporting and validation efforts enhance reliability of research. LAG-R standardization will bolster reproducibility, peer review, and overall scientific rigor.

We report development and preparation of synthetic polypeptide based, coacervate core polyelectrolyte complex micelles, PCMs, in aqueous media, which were characterized and evaluated for the encapsulation and in vitro release of a model single-stranded RNA, polyadenylic acid, poly(A). Cationic, α-helical polypeptides pegylated at their N-termini, PEG113-b-5bn and PEG113-b-5cn, were designed to form coacervate core PCMs upon mixing with multivalent anions in aqueous media. Sodium tripolyphosphate (TPP) and poly(A) were used as model multivalent anions that allowed optimization of polypeptide composition and chain length for formation of stable, nanoscale PCMs. PEG113-b-5c27 was selected for preparation of PCMs that were characterized under different environmental conditions using dynamic light scattering, atomic force microscopy and cryoelectron microscopy. The PCMs were found to efficiently encapsulate poly(A), were stable at physiologically relevant pH and solution ionic strength, and were able to release poly(A) in the presence of excess polyvalent anions. These PCMs were found to be a promising model system for further development of polypeptide based therapeutic delivery vehicles.

Abstract. Marine diazotrophs convert dinitrogen (N2) gas intobioavailable nitrogen (N), supporting life in the global ocean. In 2012, thefirst version of the global oceanic diazotroph database (version 1) waspublished. Here, we present an updated version of the database (version 2),significantly increasing the number of in situ diazotrophic measurements from13 565 to 55 286. Data points for N2 fixation rates, diazotrophic cellabundance, and nifH gene copy abundance have increased by 184 %, 86 %, and809 %, respectively. Version 2 includes two new data sheets for the nifH genecopy abundance of non-cyanobacterial diazotrophs and cell-specific N2fixation rates. The measurements of N2 fixation rates approximatelyfollow a log-normal distribution in both version 1 and version 2. However,version 2 considerably extends both the left and right tails of thedistribution. Consequently, when estimating global oceanic N2 fixationrates using the geometric means of different ocean basins, version 1 andversion 2 yield similar rates (43–57 versus 45–63 Tg N yr−1; rangesbased on one geometric standard error). In contrast, when using arithmeticmeans, version 2 suggests a significantly higher rate of 223±30 Tg N yr−1 (mean ± standard error; same hereafter) compared to version 1(74±7 Tg N yr−1). Specifically, substantial rate increases areestimated for the South Pacific Ocean (88±23 versus 20±2 Tg N yr−1), primarily driven by measurements in the southwestern subtropics,and for the North Atlantic Ocean (40±9 versus 10±2 Tg N yr−1). Moreover, version 2 estimates the N2 fixation rate in theIndian Ocean to be 35±14 Tg N yr−1, which could not be estimatedusing version 1 due to limited data availability. Furthermore, a comparisonof N2 fixation rates obtained through different measurement methods atthe same months, locations, and depths reveals that the conventional15N2 bubble method yields lower rates in 69 % cases compared tothe new 15N2 dissolution method. This updated version of thedatabase can facilitate future studies in marine ecology andbiogeochemistry. In addition to many global disruptions due to the COVID-19 pandemic, institutions of higher education within the United States were forced to move classes online and put a pause to many initiatives. Areas that were particularly affected by the pandemic were education abroad and global service-learning (GSL) programs. As GSL programs resume operations with partners around the world, there exists an opportunity to redefine partnerships with the goal of reciprocity at the center (Bartleet et al., 2019; Brown et al., 2016; Grain et al., 2019). In this study, authors conducted interviews with partner organizations in a GSL program at a large, public university in the Midwest. Through this work, the authors developed a set of guiding questions to help GSL scholar-practitioners relaunch their programs with the goal of reciprocal partnerships.

The development of early childhood vocabulary is influenced by both biological and environmental factors, which shape language acquisition. This research investigates the variability in early expressive vocabulary among typically developing children (TD), Late Talkers (LTs), and those at risk for neurodevelopmental conditions like Autism Spectrum Disorder (ASD), and Developmental Language Disorder (DLD). Participants included 132 Mexican Spanish-speaking children: 37 with TD (M = 24.89, SD =4.01), 37 LTs (M = 24.78, SD 3.51), 41 at risk for ASD (M = 24.39, SD = 4.31) and 17 at risk for DLD (M = 37.71, SD = 4.50). The MacArthur-Bates Communicative Development Inventory II was utilized to assess 23 vocabulary categories, which were grouped into six broader categories (nouns, verbs, adjectives-adverbs, functional words, routines and sounds-onomatopoeias). The results indicated differences in vocabulary distribution among the groups. Although TD children generally exhibited the highest performance, there was notable variability within this group. Both LTs and children at risk for ASD showed differences compared to TD children, with LTs demonstrating the most reduced lexical usage. Children at risk for DLD and LTs displayed similar lexical profiles, characterized by reduced use of verbs and functional words. LTs and most children at risk for ASD exhibited low usage across all vocabulary categories. This analysis identified distinct vocabulary profiles among TD, LTs, ASD, and DLD groups, with variability across vocabulary categories reflecting the unique characteristics of each group. These findings enhance our understanding of the heterogeneity in early language development across clinical populations.

Este dossier documenta las tendencias recientes de historia de las misiones con nuevos estudios de caso. Veintitrés investigadores respondieron a nuestra convocatoria con la presentación de manuscritos, diez de los cuales fueron seleccionados para su publicación en este monográfico. De acuerdo con la convocatoria, los ensayos se centran en los siglos XIX y XX, haciendo hincapié en el último tercio del siglo XIX y la primera mitad del XX, en consonancia con la evolución expuesta. Sin embargo, los artículos sobre protestantismo amplían el periodo a gran parte del siglo XX. En cuanto a la localización geográfica, la atención de los manuscritos recibidos se centró —sorprendentemente para nosotros— de manera predominante en Sudamérica, pese a que la convocatoria incluía Mesoamérica y el Caribe. Aparte de Colombia, donde una nueva generación se ha propuesto en la última década investigar de forma consecuente la historia de las regiones fronterizas de este diverso país, escogimos manuscritos sobre y procedentes de Argentina, Brasil y Chile. Si se observan los temas, enfoques y métodos adoptados en las contribuciones presentadas a continuación, destacan ciertas características. Los manuscritos seleccionados para este dossier se centran claramente en la expansión estatal hacia zonas fronterizas, con una división de funciones entre el Estado y las misiones. Los intereses respectivos eran diferentes, pero el Estado y las misiones compartían la idea de que perseguían un proyecto común de “colonización”, “civilización” y “educación”.

ObjectiveThis study aims to describe clinical variables and quality care indicators in pediatric stroke management at a high-complexity pediatric care center in Latin America.MethodsRetrospective study of patients with stroke, aged 2–18 years from 2011 to 2021. The principal outcomes were the mRs and mortality. Differences between groups were assessed using Fisher’s exact test and the Mann–Whitney U test. We used logistic regression to explore the association between characteristics reported as relevant in literature and mortality.ResultsOne hundred thirty six patients included, with a median age of 11 years, 54% were male. 47% were hemorrhagic strokes, followed by ischemic strokes at 39%. One-third of the cases presented in hospital. 51% of the patients had no prior medical history. The most common symptoms were altered consciousness, headache, and hemiparesis. The median door-to-image time was 123 min. The most frequent etiologies in ischemic stroke were arteriopathies and cardiac pathology, while vascular malformation and coagulopathies were predominant in hemorrhagic stroke. No patient received reperfusion therapy. At discharge, 48% of patients had a favorable mRs. The mortality rate was 21%. Patients with in-hospital stroke have approximately 7.37 times the odds of dying compared to those with out-of-hospital stroke. Patients with hemorrhagic stroke have approximately 7.46 times the odds of dying compared to those with ischemic stroke.ConclusionSignificant gaps exist in the epidemiology and quality indicators of pediatric stroke care compared to adult protocols. Implementing a “Pediatric code stroke” protocol and conducting prospective studies are crucial for improving pediatric stroke care and outcomes.

Leptospirosis is a neglected zoonotic disease that is endemic in tropical regions, including Ecuador. It is caused by spirochetes of the genus Leptospira, which can infect humans through animal reservoirs such as rats and dogs, or through contact with contaminated water or soil. In March 2023, public health authorities declared a concerning outbreak of leptospirosis in Durán Cantón, located in the Coastal region of Ecuador. For the first time in the country, a multidisciplinary approach involving physicians and veterinarians was implemented for the surveillance and management of this leptospirosis outbreak. A total of 335 samples were collected, including suspected human cases, household contacts, household dogs, synanthropic rats, and water samples within the area of human cases. Samples were processed by qPCR targeting lipL32, secY, and rrs fragment genes and characterized further for Sanger sequencing. Overall, 26.2 % of human samples, 43.8 % of dog samples, 38.5 % of rat samples, and 39.4 % of water samples tested positive for Leptospira. Further, phylogenetic analysis shows that human, dog, and rat sequences are clustered within the pathogenic subclade P1, within the branch of L. kirschneri and L. interrogans. This study is the first of its kind in Ecuador, where an ongoing outbreak of leptospirosis was managed in real-time by using molecular diagnosis and not serological tools, and where the epidemiological surveillance was done following a One Health approach. This experience should inspire public and animal health authorities in Ecuador to promote a national One Health surveillance and control program for zoonotic diseases.

BackgroundThis meta-analysis investigates the role of specific brain regions in semantic control processes using Transcranial Magnetic Stimulation (TMS). According to the Controlled Semantic Cognition framework, control processes help manage the contextually appropriate retrieval of semantic information by activating a distributed neural network, including the inferior frontal gyrus, the posterior middle temporal gyrus, and inferior parietal lobule. Lesions in these areas can lead to difficulties in manipulating weakly activated or competing semantic information. Researchers have used TMS to simulate such deficits in healthy individuals.MethodBy synthesizing results from TMS studies that targeted these regions, we aimed to evaluate whether neurostimulation over these areas can effectively impair participants’ performance under high semantic control demands.ResultsResults from different meta-analytical approaches consistently showed no significant effects of TMS, especially after correcting for publication bias. Nevertheless, variability in experimental methodologies was evident.ConclusionThese findings raise questions about the effectiveness of TMS in simulating deficits in semantic control and highlight the need for methodological improvements in future studies to enhance reliability and interpretability.

Electric arc furnace slag (EAFS) is a viable alternative in asphalt mixtures due to its favourable mechanical properties. This study examines the impact of EAFS content and bitumen film thickness (TF) on the fatigue performance of asphalt mixtures. Mixtures with varying levels of EAFS replacement were designed, and their mechanical properties were evaluated through indirect tensile strength and stiffness tests, followed by fatigue tests using the four-point bending method and EBADE (Strain Sweep Test). The results indicated that mixtures with EAFS exhibited increased stiffness, but fatigue performance decreased at high strain levels. At low strain levels, EAFS mixtures performed similarly or better than the control. HMA_GL had the highest TF (13.97 μm), followed by HMA_GS (13.60 μm), HMA_SL (12.66 μm), and HMA_SS (11.77 μm), showing that as the EAFS content increases, the TF decreases. This finding was verified through Digital Image Analysis. This decrease in TF is due to the high porosity and roughness of the EAFS, which in turn reduces the effective bitumen (Pbe) in the mixture. HMA_SL*, with a TF equal to the control, demonstrated a 22 % improvement in fatigue performance compared to HMA_SL. In the EBADE tests, HMA_GL achieved 44.69 MJ/m3 of dissipated energy, HMA_GS 31.55 MJ/m3, HMA_SL 34.45 MJ/m3, and HMA_SS 35.54 MJ/m3. The improved HMA_SL* recorded 42.15 MJ/m3, nearly matching the control. EBADE results confirmed that higher EAFS content increased initial stiffness, but the complex modulus (|E*|) decreased more rapidly as deformation increased. These results are consistent with the stiffness tests. These findings suggest that EAFS can successfully replace natural aggregates in asphalt mixtures, with a moderate increase in bitumen content recommended to improve fatigue performance.

Animal venoms are natural products that have served as a source of novel molecules that have inspired novel drugs for several diseases, including for metabolic diseases such as type-2 diabetes and obesity. From venoms, toxins such as exendin-4 (Heloderma suspectum) and crotamine (Crotalus durissus terrificus) have demonstrated their potential as treatments for obesity. Moreover, other toxins such as Phospholipases A2 and Disintegrins have shown their potential to modulate insulin secretion in vitro. This suggests an unexplored diversity of venom peptides with a potential anti-obesogenic in Mexican rattlesnake venoms. For that reason, this study explored the in vitro effect of Crotalus venom peptide-rich fractions on models for insulin resistance, adipocyte lipid accumulation, antioxidant activity, and inflammation process through nitric oxide production inhibition. Our results demonstrated that the peptide-rich fractions of C. aquilus, C. ravus, and C. scutulatus scutulatus were capable of reverting insulin resistance, enhancing glucose consumption to normal control; C. culminatus, C. molossus oaxacus, and C. polystictus diminished the lipid accumulation on adipocytes by 20%; C. aquilus, C. ravus, and C. s. salvini had the most significant cellular antioxidant activity, having nearly 80% of ROS inhibition. C. aquilus, C. pyrrhus, and C. s. salvini inhibited nitric oxide production by about 85%. We demonstrated the potential of these peptides from Crotalus venoms to develop novel drugs to treat type-2 diabetes and obesity. Moreover, we described for the first time that Crotalus venom peptide fractions have antioxidant and inflammatory properties in vitro models.

At the forefront of agricultural innovation, copper-based nanoparticles (Cu-based NPs) have seized global attention in recent years, revolutionizing fields as diverse as electronics, medicine, and, notably, agriculture. Their prowess in combating phytopathogenic microorganisms, boosting plant yield and defenses, and their dual role as pesticides or fertilizers - depending on the dosage - positions them at the epicenter of exciting scientific advancements. However, this is a two-edged sword: the environmental impact of Cu-based NPs is an escalating concern. The release of these particles post-use raises serious questions about the accumulation of toxic copper levels in soil and, consequently, in crops. In a market with rising metal nanoparticles, a deeper exploration is essential to balance the benefits and risks associated with Cu-based NPs in agriculture. This review synthesizes the state-of-the-art Cu-based NP synthesis, their application as antimicrobial agents, pesticides, and fertilizers, and their interactions with soil, highlighting its advantages and disadvantages. Finally, we delve into future perspectives, spotlighting the research gaps in Cu-based NP studies that beckon for attention in the coming years.

AbstractBackground Colombia has a mestizo population and the prevalence of haemoglobin variants varies according to each region, but heterozygous carriers can be found in all of them.Aim To characterise sickle cell disease (SCD) haematologically, biochemically, and molecularly, and detect classic haplotypes by DNA sequencing in a group of samples from Bolívar, Colombia.Subjects and methods Blood samples were collected after informed consent from volunteers from eight communities in the Bolívar department, plus samples from the Pacific region, Providencia Island, and Bogotá were included. Data were obtained from: (1) haematological analyses; (2) biochemical tests: dHPLC was used to determine haemoglobin (Hb); and (3) DNA sequencing data through five SNPs.Results 101 samples were identified by rs334 through Sanger’s Sequencing, structural haemoglobinopathies HbAS (34.65%), HbSS (2.97%) and HbAC (1.98%) were found. When contrasting the Hb identification results between SNP rs334 Vs. dHPLC/Isoelectric Focusing (IEF), a coincidence was found in 39/43 samples analysed, therefore, when comparing these techniques, a significant correlation was found (Pearson’s correlation coefficient r = 0.998). 26 samples previously analysed by rs334 were classified into classical haplotypes CAR (50.0%), BEN (30.76%), CAM (7.69%), SEN (3.84%), and ATP-I (7.69%).Conclusions SCD characterisation and SNPs-based classification through Sanger’s DNA sequencing have not been performed before in Colombia. The results of this work will make it possible to expand the data or records of carriers and those affected, which will benefit patients and their families.
