Abstract

International audience; Diaphragm repair after congenital diaphragmatic hernia is associated with hernia recurrence due to prosthesis failure. Expanded polytetrafluoroethylene (e-PTFE), a synthetic non-degradable biomaterial, is currently used for those diaphragmatic defect repairs. The drawback of e-PTFE is its poor wettability that leads to coating difficulties, bonding that could favor implant integration. However, polydopa-mine (PDA) can be deposited as well on organic as on inorganic substrates. Therefore, we assessed the biological responses of a clinically used e-PTFE bio-material treated with PDA in two different manners: one impregnated with PDA and the other coated with a one side PDA film. Mechanical properties of the raw e-PTFE, the PDA soaked biomaterial and the PDA coated surface were characterized by colloidal probe atomic force microscopy. Behaviors of primary human fibroblasts and Wharton's jelly stem cells were investigated by electron micro-scopy. Findings reveal that the mechanical properties at the microscopic scale are not modified by the PDA treatments. Cells spread onto both PDA functionalized substrates. In addition, microscopic observations disclose numerous focal cell contacts, evidencing cell attachment, and cytoplasmic projections particularly with the nanoscale PDA coating. Results clearly suggest that PDA in general but above all the PDA coating enhance cellular colonization of the implant material.

Abstract

Polydopamine can form biocompatible particles that convert light into heat. Recently, a protocol has been optimized to synthesize polydopamine/protein hybrid nanoparticles that retain the biological function of proteins, and combine it with the stimuli-induced heat generation of polydopamine. We have utilized this novel system to form polydopamine particles, containing transferrin (PDA/Tf). Mouse melanoma cells, which strongly express the transferrin receptor, were exposed to PDA/Tf nanoparticles (NPs) and, subsequently, were irradiated with a UV laser. The cell death rate was monitored in real-time. When irradiated, the melanoma cells exposed to PDA/Tf NPs underwent apoptosis, faster than the control cells, pointing towards the ability of PDA/Tf to mediate UV-light-induced cell death. The system was also validated in an organotypic, 3D-printed tumor spheroid model, comprising mouse melanoma cells, and the exposure and subsequent irradiation with UV-light, yielded similar results to the 2D cell culture. The process of apoptosis was found to be targeted and mediated by the lysosomal membrane permeabilization. Therefore, the herein presented polydopamine/protein NPs constitute a versatile and stable system for cancer cell-targeting and photothermal apoptosis induction.

Abstract

Polyelectrolyte multilayer films made from the alternated deposition of poly(allylamine) as the polycation (PAH) and from poly(styrene-4-sulfonic acid) as the polyanion are known to be very compact and impermeable to multivalent ions and to display a strong selectivity for monovalent cations with respect to multilvalent ones. Herein, the particular case of the diffusion of silver cations at pH 5.0 through (PAH–PSS)m films of variable thickness is investigated using an original detection method of the silver cations. In this method, the (PAH–PSS)m multilayer films are deposited on films made from the alternated adsorption of PAH and tannic acid (TA). When those (PAH–TA)n films are put in contact with Ag+ cations, the TA allows for the reduction of Ag+ cations in silver particles easily detectable by means of UV–vis spectroscopy. The deposition of (PAH–PSS)m multilayers on top of (PAH–TA)6 films does not imped the deposition of silver up to m = 10 (PAH–PSS) bilayers, corresponding to a thickness of 25 nm. The high permeability of (PAH–PSS)m films is not due to a lack of homogeinity because hexacyanoferrate anions are not able to react with Fe3+ cations encapsulated in (PAH–TA)6 films when those films are capped with a (PAH–PSS)5 film known to be impermeable to Fe(CN)64− in the absence of defects in the film. It is also found that the reduction of Ag+ cations on a (PAH–TA)4 film is a slow process needing more than 1 h to be achieved.

Abstract

The oxidation of dopamine and of other catecholamines leads to the formation of conformal films on the surface of all known materials and to the formation of a precipitate in solution. In some cases, it has been shown that the addition of additives in the dopamine solution, like certain surfactants or polymers, polyelectrolytes, and certain proteins, allows to get polydopamine nanoparticles of controlled size and the concomitant decrease, in an additive/dopamine dependent manner, in film formation on the surface of the reaction beaker. However, the mechanism behind this controlled oxidation and self-assembly of catecholamines is not known. In this article, it is shown that a specific diad of amino acids in proteins, namely KE, allows for specific control in the oxidation-self-assembly of dopamine to obtain polydopamine@protein core–shell nanoparticles which are biocompatible. The interactions between dopamine and the adjacent KE amino acids potentially responsible for the size control of polydopamine aggregates was investigated by molecular dynamics simulations. The obtained core– shell nanoparticles display the biological activity of the protein used to control the self- assembly of PDA. The photon to heat conversion ability of PDA is conserved in the PDA@protein particles.

Abstract

An unusual case of self-patterning layer-by-layer (LBL) films made from poly(allylamine hydrochloride) and poly(sodium phosphate) is described. The studies have shown that the adsorption of individual elayersE of polyanions and polycations has proceeded without a regular inversion of the [zeta] potential and with the occurrence of a growth instability at 75 layers.

Abstract

When catecholamine solutions are put in the presence of an oxidant, a spontaneous oxidation process allows one to coat the surface of all known classes of materials with a brown-black insoluble material. This material is related to eumelanins from both compositional and structural points of view. Simultaneously with the film deposition at solid/liquid interfaces, an insoluble material is obtained in solution. Under particular experimental conditions, the chemistry of catecholamines, mostly dopamine, can be extended to the water/air interface to produce films which can be handled and transferred on solid substrates. The mechanical properties of these ‘polydopamine’ films can be improved in a way allowing for the realization of micrometre-thick free-standing Janus-like films. Polydopamine films at liquid/liquid interfaces have also been found to stabilize microemulsions. The aim of this mini-review is to summarize these recent advances of catecholamine chemistry to obtain functional coatings at fluid interfaces

Abstract

A study was conducted to establish a correlation between the nature of the growth process and the heat of complexation between the polyanions and the polycations constituting the polyelectrolyte multilayer films (PEM). Weakly exothermic processes seem to be associated with weakly exponentially growing films which show that exponentially growing processes are mainly driven by entropy.
